Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer substituierter Fluoracylresorcine der Formel I

In der obigen Formel I bedeuten
Rj eine perfluorierte Alkylgruppe mit 1 bis 8 Kohlenstoffatomen oder die 2,2,3,3-Tetrafluorcyclobutylgruppe;
R2 und R4, die gleich oder voneinander verschieden sein können, ein Wasserstoffatom, eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 10 Kohlenstoffatomen, eine aliphatische Acylgruppe mit 2 bis 18 Kohlenstoffatomen, die Benzoyl-, Salicyloyl- oder Phenylacetylgruppe, wobei jeweils mindestens einer der Reste R2 und R4 ein Wasserstoffatom darstellt;
R3 und R5, die gleich oder voneinander verschieden sein können, Alkylgruppen mit 3 bis 18 Kohlenstoffatomen, Halogenatome, die Nitrogruppe oder die p-Toluolsulfonyl-gruppe, die Cyclopentyl-, Cyclohexyl-, Cycloheptyl-, Cyclo-dodecyl-, Methylcyclohexyl-, Dimethylcyclohexyl-, Benzyl-oder Methylthiogruppe; oder eine Gruppe der Formel

Rs auch eine Methylgruppe, es kann aber auch einer der beiden Reste R3 und R5 ein Wasserstoffatom oder die Äthylgruppe bedeuten, sofern dann der andere dieser Reste R3 und Rs die übrigen oben angegebenen Bedeutungen mit Ausnahme der eines Wasserstoffatoms innehat oder sofern Ri die oben angegebenen Bedeutungen mit Ausnahme der einer Trifluormethylgruppe besitzt oder sofern R2 und R4 die oben angegebenen Bedeutungen mit Ausnahme der eines Wasserstoffatoms oder einer Methylgruppe besitzen.
Die substituierten Fluoracylresorcine der Formel I werden durch Umlagerung von Perfluoracylresorcinen der For-

in der die Reste R3 und R5 wie oben definiert sind und D die Gruppe 0R4 bedeutet, sofern E die Gruppe -0-C0Rx darstellt, oder D die Gruppe -O-CORj ist, sofem dann E für die Gruppe -OR2 steht, wobei in diesen Gruppen R1; R2 und R4 wie oben beschrieben definiert sind oder gegebenenfalls beide Reste D und E eine Gruppe -O-CORi bedeuten, hergestellt.
Die Umlagerung erfolgt in Gegenwart einer Lewis-Säure als Katalysator, gegebenenfalls in Gegenwart eines Lösungsmittels, bei Temperaturen zwischen 0 und 150 °C.
Als Lewis-Säuren eignen sich unter anderen insbesondere wasserfreies Zinkchlorid, wasserfreies Aluminiumchlorid, Zinkchlorid in Anwesenheit einer Halogenwasserstoffsäure, des weiteren beispielsweise Eisen(III)chlorid und Zinn-(IV)chlorid. Als Lösungsmittel kommen beispielsweise Äther oder aromatische Kohlenwasserstoffe, wie Chlorbenzol, Nitrobenzol, Toluol, Dichlorbenzol oder Xylol, und Phosphoroxychlorid in Frage.
Durch die Umlagerung einer Verbindung der Formel II entsteht eine Verbindung der Formel I, in der mindestens einer der Reste R2 und R4 ein Wasserstoffatom darstellt.
Gegenstand der Erfindung sind weiterhin Verfahren zur Herstellung von Verbindungen der Formeln I' und I", wie sie in den Patentansprüchen 4 und 7 angegeben sind. Bevorzugte Ausführungsformen sind in den Patentansprüchen 5, 6, 8 und 9 angegeben.
Die nach dem vorstehend genannten Verfahren gewonnenen Verbindungen der Formel I, in der R2 und/oder R4 Wasserstoffatome sind, lassen sich, wie in den Patentansprüchen 4 und 7 angegeben, anschliessend durch Veräthern, z.B. mit Alkylhalogeniden, oder durch Verestern, z.B. mit Säurehalogeniden oder Säureanhydriden, in Verbindungen der Formel I' bzw. I" überführen.
Die Ausgangsverbindungen der Formel II, worin D oder E die Gruppe -O-CORj bedeutet, erhält man vorteilhaft durch Veresterung der entsprechenden Verbindungen, in welchen D oder E Hydroxygruppen darstellen, nach an sich bekannten Methoden.
Es wurde überraschenderweise gefunden, dass die Verbindungen der Formel I bzw. I' und I" wertvolle pharmakologische und/oder pestizide Eigenschaften besitzen: sie sind insbesondere wirksam gegen Bakterien, Dermatophyten, Hefen, Schimmelpilze und phytopathogene Pilze; sie wirken hemmend auf verschiedene Schlüsselenzyme des Kohlenhydratstoffwechsels und auf Zellkulturen, verlangsamen damit beschleunigte Zellteilungsvorgänge in und auf der Haut. Sie eignen sich deshalb zur Behandlung von Akne, Kopfschuppen, bakteriellen Hautinfektionen, Mykosen, Psoriasis, Ichthyosis, hyperkeratotischen Hautzuständen, zur Bekämpfung boden- und samenbürtiger Pilze im Pflanzenanbau sowie als Herbizide, z.B. selektiv gegen Flughafer. Verschiedene Verbindungen der Formel I bzw. I' und 1" sind auch an-thelmintisch wirksam.

auf ihre Hemmwirkungen gegen Bakterien und Pilze, auf ihre Hemmwirkungen auf Zellkulturen und ihre Hemmwirkungen auf die Enzymaktivitäten vergleichend getestet.
Die Hemmwirkung auf Bakterien und Pilze wurde nach dem Reihenverdünnungstest und dem Agardiffusionstest (Loch-Test) geprüft. Als Bakterien wurden eingesetzt: Sta-phylococcus aureus SG 511, Streptococcus Aronson, Streptococcus pyogenes AT CC 86 68; als Pilze: Candida albicans AT CC 10231, Trichophyton mentagrophytes AT CC 9129 und Aspergillus niger.
Reihenverdünnungstest Nährmedien
1. Fleischextraktbouillon: für St.aureus SG 511
Rezept: Pepton 10 g
Fleischextrakt Oxoid 8 g
Kochsalz 3 g
Sek. Na-Phosphat (Na2HP04) 2 g ad 1000 ml Aqua dest. (pH 7.2-7.4)
Sterilisation: 15 Min. bei 120 °C im Autoklav
2. Glucosebouillon: für Sc. Aronson und Streptococcus pyogenes Rezept wie bei Fleischextraktbouillon. Nach dem Sterilisieren 1% der Glucose als sterile 50%ige Lösung zusetzen.
3. Sabouraudbouillon: für C. alb., Trich. mnt., A. niger
Rezept: Pepton aus Casein 10 g
Glucose 40 g
Kochsalz 1 g
Sek. Na-Phosphat (Na2HP04) 1 g ad 1000 ml Aqua dest.
Sterilisation: 5-10 Min. bei 120 °C, ein pH wird nicht eingestellt.
Einstellung der Keimdichte
Das Alter der Primärkulturen beträgt bei Bakterien 24 h und bei Pilzen 14 Tage. Die Einstellung der Keimsuspension erfolgt am Photometer «Eppendorf» (Reagenzglas 0 14 mm, Filter 546 nm) nach Bariumsulfat-Vergleichssuspension (= Trübung einer Bariumsulfat-Aufschwemmung von 3,0 ml l%iger Bariumchloridlösung und 97 ml l%iger Schwefelsäure). Nach der Einstellung werden die Bakterien 1 : 1000 mit Kochsalzlösung weiter verdünnt, die Pilze werden unverdünnt eingesetzt.
Vorbereitung der Substanzkonzentration
40 mg der Substanz werden in 10 ml-Messkolben eingewogen und mit dem Lösungsmittel bis zur Marke aufgefüllt (entspricht einer Verdünnung von 1 : 250 = 4000 ng/ml). Die weitere Verdünnungsreihe wird mit Aqua dest. oder dem jeweiligen Lösungsmittel eingestellt und folgende Substanzkonzentrationen hergestellt: 1000; 250; 62,5 (ig/ml.
Durchführung des Testes
Die Röhrchen werden mit 4,9 ml des entsprechenden flüssigen Nährmediums beschickt. Jedem Röhrchen werden nun 0,1 ml der oben hergestellten Substanzverdünnung zugegeben, so dass die erwähnten Endkonzentrationen vorliegen. Schliesslich wird jedes Röhrchen mit 0,1 ml der eingestellten Keimsuspension beimpft. Eine Lösungsmittelkontrolle ist immer mitzuführen.
Bebrütung
Bakterien werden 18-20 Stunden bei 37 °C und Pilze 7 Tage bei 27 °C bebrütet.
Auswertung
Die Ablesung erfolgt makroskopisch unter Angabe der Grenzkonzentration (= niedrigste noch mikrobiostatisch wirksame Konzentration).

Pronagar 15 g ad 1000 ml Aqua dest.
Sterilisation: 5-10 Minuten bei 120 °C. Ein pH wird nicht eingestellt.
Einstellung der Keimdichte
Das Alter der Primärkulturen beträgt bei Bakterien 24 Stunden und bei Pilzen 14 Tage.
Die Einstellung der Keimsuspension erfolgt am Photometer «Eppendorf» (Reagenzglas 0 14 mm, Filter 546 nm) nach Bariumsulfat-Vergleichssuspension (= Trübung einer Bariumsulfat-Aufschwemmung von 3,0 ml 1 %iger Bariumchloridlösung und 97 ml 1 %iger Schwefelsäure). Nach der Einstellung werden St. aureus SG 511, 1: 100 und Sc. pyogenes und Aronson 1 : 100 mit Kochsalzlösung weiter verdünnt. Die Pilze werden unverdünnt eingesetzt.
Vorbereitung der Substanzkonzentrationen
40 mg der Substanz werden in 10 ml Messkolben eingewogen und mit dem Lösungsmittel bis zur Marke aufgefüllt (entspricht einer Verdünnung von 1 : 250 = 4000 jxg/ml).
Die Verdünnungen auf die zu prüfenden Konzentrationen erfolgen mit Aqua dest. oder dem jeweiligen Lösungsmittel.
Durchführung des Testes
In sterile Petrischalen von 8 cm Durchmesser werden 19 ml Nährmedium ausgegossen und vorgetrocknet. Anschliessend werden die Agarplatten mit 4 ml Saatagar be5

schickt. 100 ml Saatagar enthalten 1,25 ml Keimsuspension,
eine Agarplatte enthält somit 0,05 ml Keimsuspension. Nach Erstarren des Agars werden 5 Löcher von 5 mm Durchmesser in die Platten gestanzt und mit 0,05 ml der entsprechend konzentrierten Substanzlösung gefüllt. Eine Lösungsmittel- 5 kontrolle ist immer mitzuführen.
Bebrütung
Bakterien werden 18-20 Stunden bei 37 °C und Pilze 7 Tage bei 27 C bebrütet. 10
Auswertung
Gemessen wird der Durchmesser des Hemmhofes in mm (abzüglich des Lochdurchmessers). Wenn statt einer wachstumsfreien Zone nur deutlich vermindertes Wachstum zu 15 verzeichnen ist, werden diese Werte in Klammern gesetzt.

Die Suspension mit Corynebacterien acnes wurde 48 Stunden bei 37 °C, die Suspension von Pityrosporum ovale 7 Tage bei 27 °C bebrütet. Die Ablesung erfolgte durch makroskopische Beurteilung des Keimwachstums und Registrierung der Grenzkonzentrationen.
Agardiffusionstest bei Pityrosporum ovale CBS 1878 Nährboden
Littmann's Agar, 23 ml pro Petrischale, Schalendurchmesser 100 mm.
Keimdichte
Keimsuspension in 0,9%iger Kochsalzlösung, eingestellt im Photometer «Eppendorf» anhand einer Bariumsulfat-Vergleichssuspension, hiervon wurden jeweils 0,05 ml pro Platte verwendet. Die Testsubstanzen waren in Dimethyl-sulfoxid gelöst. Die Bebrütungszeit betrug 7 Tage bei 27 °C; gemessen wurde der Hemmhofdurchmesser in mm, es wurden 0,05 ml Substanzlösung pro Stanzloch von 6 mm Durchmesser verwendet.
Die bei diesen Vergleichsversuchen gefundenen Werte sind in den nachfolgenden Tabellen 1 und 2 enthalten:

Beobachtet wird das Gleichgewicht:

(NADP = Nikotinamid-adenin-dinukleotid-phosphat, G6P-DH = Gluconsäure-6-phosphat-dehydrogenase).
Die Bildungsgeschwindigkeit von NADPH ist ein Mass für die Enzymaktivität; sie kann anhand der Extinktionszunahme bei 340,334 oder 366 nm pro Zeiteinheit verfolgt werden.
Methodik
0,025 ml Glucose-6-phosphat-dehydrogenase (Fa. Boeh-ringer Mannheim) werden mit destilliertem Wasser auf 10 ml aufgefüllt (Lösung I). 100 mg Nikotinamid-adenin-dinukleotidphosphat werden in 13 ml dest. Wasser gelöst (Lösung II). 47,2 mg Glucose-6-phosphat löst man in weiteren 10 ml dest. Wasser (Lösung III). Nebenher wird eine Pufferlösung (Lösung IV) wie folgt bereitet:
0,28 g Triäthanolamin-hydrochlorid und 1,461 g Äthylendiamintetraessigsäure-dinatriumsalz werden in 1 Liter dest. Wasser gelöst und mit Natronlauge auf pH 7,6 eingestellt. Die zu untersuchende Substanz wird in Dimethyl-formamid oder Äthanol gelöst (Lösung V). Geprüfte Konzentrationen: 50; 25; 12,5; 6,25; 3,125; 1,56 und 0,78 Hg/ml.
Bestimmung der Soforthemmung
0,1 ml Lösung 1,0,1 ml Lösung II, 2,67 ml Lösung IV und 0,03 ml Lösung V werden vermischt und 5 Minuten auf 25 °C gehalten. Dann wird 0,1 ml Lösung III zugegeben, durchmischt und die Extinktionsänderung bei 366 nm über 3 Minuten spektralphotometrisch bestimmt.
Bestimmung der Inkubationshemmung
0,1 ml Lösung 1,0,1 ml Lösung II, 2,67 ml Lösung IV und 0,03 ml Lösung V werden vermischt und 60 Minuten bei 37 °C gehalten. Dann wird 0,1 ml Lösung III zugegeben, durchmischt und die Extinktionsänderung bei 366 nm über 3 Minuten spektralphotometrisch gemessen.
Die Hemmwerte werden aus den Mittelwerten dreier Messungen (als Extinktionsänderung pro Minute) im Vergleich zu Kontrollen, bei denen als Inhibitorlösung das reine Lösungsmittel zugesetzt wird, berechnet. Dann wird aus den
Hemmwerten für die unterschiedlichen Konzentrationen die ED50 nach der Methode von Reed und Muench berechnet.

Messung der Hemmung von Zellkulturen Methodik
HeLa-Zellkultur wird abtrypsiniert und in frischem Medium auf eine Zellzahl von 150 000 Zellen/ml eingestellt. Die Substanz wird stets in der gleichen Menge Dimethylsulfoxid angelöst und dann mit Wachstumsmedium weiter verdünnt. In Mikrotiterplatten werden je 0,1 ml der Substanzverdünnungen pro Vertiefung eingefüllt und dann 0,2 ml Zellsuspension dazugegeben (pro Verdünnung 4 Vertiefungen). Es werden mehrere Wachstumskontrollen, die statt 0,1 ml Substanzverdünnung 0,1 ml Wachstumsmedium enthalten, angesetzt. Nach sorgfaltigem Durchmischen werden die Kulturen 3 Tage bei 37 °C unter 5% C02-Begasung bebrütet. Die Ablesung erfolgt im Vergleich zur dicht gewachsenen Wachstumskontrolle. Die Ableseergebnisse werden als Prozentsatz von Wachstumsausfall und Degenerationserscheinungen im Vergleich zur Wachstumskontrolle angegeben. Daraus wird die Grenzkonzentration ermittelt und die EDS0 nach Reed und Muench errechnet. Die Angaben beziehen sich auf ng Substanz pro ml Gesamtmedium.

Die genannten Verbindungen sind chemisch stabil, zeigen hohe Lipophilie (Verteilungs-Koeffizient n-Octanol/ Wasser > 1000) und lassen sich gut in Salben, Cremes, Tinkturen, Sprays, Puder usw., die für topische Anwendung geeignet sind, einarbeiten.
Von besonderem Vorteil sind die gute Hautverträglichkeit (eine Creme, die 10% Verbindung E enthielt, wurde über 24 Stunden unter Okklusion reizlos vertragen) und geringe Toxizität:
Die akute Toxizität wurde an der Maus ermittelt. Bestimmt wurde die LD50, d.h. die Dosis, nach deren Verabreichung innerhalb von 14 Tagen 50% der Tiere verstarben.

Im allgemeinen pharmakologischen Screening, das auf eine Beeinflussung wesentlicher Körperfunktionen, wie beispielsweise Herz/Kreislauf oder Zentralnervensystem, schliessen lässt, zeigten sich keine nennenswerten Wirkungen. Systemeische Nebenwirkungen sind deshalb bei lokaler Anwendung nicht zu erwarten.
Wegen der hohen Lipophilie bei gleichzeitiger Anwesenheit polarer Gruppen penetrieren die Verbindungen gut in die Haut, werden aber, wie durch Untersuchung der Ausscheidung gezeigt werden konnte, nur zum geringen Teil resorbiert.
Bei Untersuchung auf Hautverträglichkeit und Sensibilisierung, die an Meerschweinchen durchgeführt wurden, zeigte sich, dass die schwach sensibilisierenden Eigenschaften mancher Resorcine durch die Einführung der Trifluoracetyl-Gruppe verschwindet. Da Resorcine, wie z.B. Hexylresorcin beim Menschen, in manchen Fällen Allergien hervorrufen, ist dies ein wesentlicher Vorteil.
Eine wirkungsvolle Therapie der Akne ist derzeit nur systemisch mit starken Antibiotika (Tetracycline, Erythromycin), lokal mit Schälmitteln, wie Vitamin-A-Säure und Benzoylperoxyd, möglich. Die Anwendung von Antibiotika bei einer keinesfalls lebensbedrohenden Krankheit ist wegen der Resistenzbildung prinzipiell problematisch, bei Anwendung von Schälmitteln ist stets mit erheblicher Hautreizung zu rechnen.
Bei der Akne-Therapie mit Antibiotika werden die bei der Akne wichtigen gram-positiven Bakterien, vor allem Corynebacterium acnes, vermindert, was eine Reduktion des
Gehaltes an freien Fettsäuren, die durch diese Bakterien aus Triglyceriden abgespalten werden, im Sebum zur Folge hat.
Wie Tabelle 1 zeigt, sind die oben genannten Verbindungen stark gegen Corynebacterium acnes wirksam. Ausserdem konnte gezeigt werden, dass nach lokaler Anwendung eine erhebliche Reduktion des Gehaltes an freien Fettsäuren möglich ist. Es ist deshalb eine lokale Therapie, die in ihrer Wirkung der oralen Therapie mit Antibiotika vergleichbar ist, möglich.
Die genaue Ursache der Schuppenbildung ist bisher noch unbekannt. Man findet jedoch bei Schuppen eine Hyper-keratose, d.h. die Zellteilungsvorgänge in der Epidermis laufen beschleunigt ab; ausserdem ist die Verhornung gestört. Nach Aussage einiger Autoren, z.B. R.A. Gosse, R.W. Van-derWyck, J. Soc. Cosmet. Chem. 20 (1969) 603, spielt die Hefe Pityrosporum ovale bei der Genese der Schuppen eine Rolle.
Tabelle 2 zeigt, dass einige der oben genannten Verbindungen eine starke Wirkung gegen Pit. ovale zeigen.
Aus Tabellen 3 und 4 ist ersichtlich, dass diese und andere Verbindungen beschleunigt ablaufende Zellteilungsvorgänge verlangsamen können. Mit Verbindungen, die in den Tabellen 2, 3 und 4 gute Wirksamkeit zeigen, ist deshalb eine Therapie der Kopfschuppen möglich.
Eine wirkungsvolle Therapie der Psoriasis ist derzeit topisch nur mit Dithranol, Teerpräparaten und hochwirksamen Corticoiden, systemisch mit Antimetaboliten, wie Methothrexat, Cortocosteroiden und Cytostatika, möglich. Ausserdem werden noch die physikalische Behandlung mit UV-Licht, Röntgen-Bestrahlung und die kombinierte Anwendung von Psoralenen (systemisch oder lokal) und UV-Licht angewendet. Alle diese Behandlungsmethoden sind entweder umständlich oder von erheblichen Nebenwirkungen begleitet. Eine einfache wirkungsvolle lokale Therapie ist deshalb von besonderem Vorteil. Die Tabellen 3 und 4 zeigen, dass einige der obengenannten Verbindungen zur Psoriasis-Therapie eingesetzt werden können.
Mykosen der Haut gewinnen zunehmend an Bedeutung. Da ein Erreger-Nachweis oft nicht durchgeführt werden 40 kann, ist die Anwendung von breit gegen Dermatophyten, Hefen und Bakterien wirksamen Antimykotica von besonderem Vorteil.
Die Tabellen 1 und 2 zeigen, dass die obengenannten Verbindungen gegen diese Erreger stark wirksam sind. Sie 45 können deshalb zur Therapie von Mykosen und bakteriellen Hautinfektionen eingesetzt werden.
Die folgenden Beispiele sollen die Erfindung noch näher erläutern.
Beispiel 1
so 2,4-Dihydroxy-5-n-hexyl-trifluoracetophenon
2 g 4-n-Hexylresorcin-mono-trifiuoressigsäureester werden mit 3 g wasserfreiem Aluminiumchlorid in 20 ml Nitrobenzol 30 Minuten auf 120 °C erhitzt. Zum Zersetzen des Reaktionsgemisches wird dasselbe unter Rühren auf 100 g 55 Eis gegossen, die organische Phase abgetrennt, die wässrige Phase dreimal mit jeweils 50 ml Äthylenchlorid nachgewaschen. Die vereinigten organischen Phasen werden mit 100 ml Wasser gewaschen, mit Calciumchlorid getrocknet und der nach dem Einengen verbleibende Rückstand aus 60 Heptan oder Petroläther umkristallisiert. F. 90 °C, Ausbeute: 1,4 g (47% der Theorie). Ohne Verwendung eines Lösungsmittels erhält man nach diesem Verfahren eine Ausbeute von 1,1g (38% der Theorie).
Analog dem Verfahren dieses Beispiels wurden noch fol-65 gende Verbindungen hergestellt:

Beispiel 2
2,4-Dihydroxy-3-methyl-trifluoracetophenon-monoacetat 4,4 g (0,02 Mol) 2,4-Dihydroxy-3-methyl-trifluoraceto-phenon werden in 25 ml Benzol gelöst und unter Rühren 3,2 g (0,04 Mol) Acetylchlorid und 3,6 g (0,045 Mol) Pyridin zugefügt. Nach 2 Stunden Rühren bei Zimmertemperatur wird in Wasser gegossen und die Benzollösung abgetrennt. Diese wird mit 50 ml Wasser ausgewaschen und mit Natriumsulfat getrocknet. Nach dem Einengen am Rotationsverdampfer wird aus n-Hexan umkristallisiert. F. 49-50 °C, Ausbeute: 4,5 g (86,5% der Theorie) In analoger Weise wurden hergestellt: 2,4-Dihydroxy-5-chlor-trifluoracetophenon-monoacetat aus 2,4-Dihydroxy-5-chlor-trifluoracetophenon und Acetylchlorid. F. 80-83 °C, Ausbeute: 71,5% der Theorie 2,4-Dihydroxy-5-chlor-trifluoracetophenon-monostearat aua 2,4-Dihydroxy-5-chlor-trifluoracetophenon und Stea-roylchlorid. F. 51 °C, Ausbeute: 40% der Theorie 2,4-Dihydroxy-3-methyl-trifluoracetophenon-
monoundecylenat aus 2,4-Dihydroxy-3-methyl-trifluracetophenon und 11-Undecylensäurechlorid. Diese Verbindung wurde durch Destillation gereinigt. Kp0 07:165 °C, Ausbeute: 64% der Theorie
2,4-Dihydroxy-5-chlor-trifluoracetophenon-monoundecyle-nat aus 2,4-Dihydroxy-5-chlor-trifluoracetophenon und 11-Undecylensäurechlorid. Diese Verbindung wurde durch Destillation gereinigt. Kp 0 07: 168 °C, Ausbeute: 65% der Theorie

Fp.: 30 °C, Ausbeute: 83% der Theorie 2,4-Dihydroxy-5-hexyl-trifluoracetophenon-monostearat aus 2,4-Dihydroxy-5-hexyl-trifluoracetophenon und Stea-roylchlorid. Ausbeute: 77% der Theorie 2,4-Dihydroxy-3-methyl-trifluoracetophenon-di-phenyl-acetat aus Phenylacetylchlorid und 2,4-Dihydroxy-3-methyl-tri-fluoracetophenon. Fp. 65 °C, Ausbeute: 90% der Theorie.
Beispiel 3
Gemisch eines Mono- und Disalicylates von 2,4-Dihydroxy-3- methyl-trifluor-acetophenon 32 g Natriumsalicylat und 2,2 g 2,4-Dihydroxy-3-methyl-trifluoracetophenon werden in 20 ml Benzol gelöst, 3,2 g Phosphoroxychlorid zugegeben und die Mischung zwei Stunden zum Rückfluss erhitzt. Zum Zersetzen des Phos-phoroxychlorids wird dann auf 150 g Eis gegossen, die benzolische Lösung abgetrennt und nochmals mit 50 ml Benzol die wässrige Phase extrahiert. Die vereinigten Benzollösungen werden mit 100 ml Wasser ausgewaschen, mit

Natriumsulfat getrocknet und eingeengt. Der Rückstand wird aus 75%igem Methanol umkristallisiert.
Fp.: 94-98 °C, Ausbeute: 40% der Theorie Laut UV-, IR- und NMR-Spektren handelt es sich um eine Mischung von Mono- und Disalicylat.
Beispiel 4
a) 2-Hydroxy-4-decyloxy-trifluoracetophenon und 2,4-Didecyloxy-trifluoracetophenon 10,3 g (0,05 Mol) 2,4-Dihydroxy-trifluoracetophenon werden mit 7 g (0,05 Mol) Kaliumcarbonat (getrocknet), 26,8 g (0,1 Mol) Decyljodid und 100 ml Aceton 5 Stunden am Rückfluss erhitzt. Das Aceton wird danach am Rotationsverdampfer eingeengt und zu dem Rückstand 100 ml Wasser gegeben. Mit 100 ml Essigester wird ausgeschüttelt und diese Lösung mit Natriumsulfat getrocknet. Nach dem Einengen am Rotationsverdampfer wird das Gemisch aus Mono- und Diäther destilliert. Im Vorlauf wird bei Kp0-1 = 80 °C das nicht umgesetzte Decyljodid erhalten. Die Äther werden danach in Methanol umkristallisiert.
Monoäther: Kp0 I = 150 °C, Fp.: 27-28 °C,
Ausbeute: 9,2 g (53,2% der Theorie)
Diäther: Kp0>1 = 200 °C, Fp.: 37-38 °C,
Ausbeute: 5,6 g (23,0% der Theorie)
In analoger Weise wurden hergestellt: 2-Hydroxy-4-butoxy-trifluoracetophenon aus 2,4-Dihy-droxy-trifluoracetophenon
F. 66°, Ausbeute: 28,5% der Theorie 2,4-Dimethoxy-5-n-hexyl-trifluoracetophenon aus 2,4-Dihydroxy-5-n-hexyl-trifluoracetophenon F. 52°, Ausbeute: 30,5% der Theorie b) 2,4-Dimethoxy-trifluoracetophenon
3 (0,015 Mol) 2,4-Dihydroxy-trifluoracetophenon werden in 75 ml Methylenchlorid gelöst, dazu gibt man 0,6 g (0,002 Mol) Tetrabutylammoniumbromid, eine Lösung von 2 g (0,05 Mol) Natriumhydroxyd in 75 ml Wasser sowie 5 g (0,04 Mol) Dimethylsulfat. Nach 5 Stunden sehr kräftigem Rühren wird in 100 ml Wasser gegossen, mit 15%iger Salzsäure angesäuert, die organische Phase abgetrennt und letztere nach Trocknen mit Natriumsulfat eingeengt. Der Rückstand wird aus n-Pentan umkristallisiert.
Fp. 49 °C; Ausbeute: 2 g (57% der Theorie). Die Verbindungen der Formel I bzw. I' und I" lassen sich in die üblichen pharmazeutischen Zubereitungsformen einarbeiten. Als solche kommen beispielsweise Schaumaerosole, Puder-Sprays, Puder, Rachensprays, Shampoos, Cremes, Salben, Tinkturen, Pasten oder Gelees in Betracht. Die Dosierung der Wirkstoffe liegt zwischen 0,05 und 1%, vorzugsweise 0,1 bis 0,8 Gew.-%.
Für den Einsatz im Pflanzenschutz werden die erfin-dungsgemäss hergestellten Verbindungen zu üblichen Formulierungen verarbeitet, insbesondere zu Lösungs- oder Emulsionskonzentraten, Stäubemitteln, Granulaten, Spritzpulvern, Beizpulvern und Beizlösungen. Der Wirkstoffgehalt in den Spritzbrühen und Stäubemitteln beträgt zwischen 0,01 und etwa 3 Gew.-%. Höhere Wirkstoffkonzentrationen weisen die Beizlösungen (etwa 10 bis 50 Gew.-%) und Beizpulver (etwa 20 bis 90 Gew.-%) sowie die Konzentrate (bis etwa 95 Gew.-%) auf.
Die Beizmittel werden aufgesprüht (Beizlösung) oder mit dem Saatgut vermischt. Sie dienen zur Bekämpfung vor allem der Pilzgattungen Tiletia, Helmintosporium, Ustilago und Fusarium. Die anderen Verbindungen der Formel I bzw. I' und I" können entsprechend angewendet werden.

Verfahren nach Anspruch I, dadurch gekennzeichnet, dass man als Lewis-Säuren wasserfreies Aluminiumchlorid oder Zinkchlorid in Anwesenheit einer Halogenwasserstoffsäure, Eisen(III)chlorid oder Zinn(IV)chlorid, verwendet.2PATENTANSPRÜCHE 1. Verfahren zur Herstellung neuer substituierter Fluor-acylresorcine der Formel IC-RRORRworinRt eine perfluorierte Alkylgruppe mit 1 bis 8 Kohlenstoffatomen oder die 2,2,3,3-Tetrafluorcyclobutylgruppe;R2 und R4, die gleich oder voneinander verschieden sein können, ein Wasserstoffatom, eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 10 Kohlenstoffatomen, eine aliphatische Acylgruppe mit 2 bis 18 Kohlenstoffatomen, die Benzoyl-, Salicyloyl- oder Phenylacetylgruppe, wobei jeweils mindestens einer der Reste R2 und R4 ein Wasserstoffatom darstellt;R3 und Rs, die gleich oder voneinander verschieden sein können, Alkylgruppen mit 3 bis 18 Kohlenstoffatomen, Halogenatome, die Nitrogruppe oder die p-Toluolsulfonyl-gruppe, die Cyclopentyl-, Cyclohexyl-, Cyclohepty-, Cyclo-dodecyl-, Methylcyclohexyl-, Dimethylcyclohexyl-, Benzyl-oder Methylthiogruppe; oder eine Gruppe der FormelORFCORORin der entweder F die Gruppe -CH2- oder -S- bedeutet, sofern G die Gruppe R5 darstellt oder F die Gruppe R3 ist, sofern G die Gruppe -CH2- oder -S- darstellt und Rt, R2 und R4 wie oben definiert sind;R3 ausserdem die Hydroxy-, Methoxy-, Methyl- oder Cyangruppe;R5 auch eine Methylgruppe bedeuten,es kann aber auch einer der beiden Reste R3 und R5 ein Wasserstoffatom oder die Äthylgruppe bedeuten, sofern dann der andere dieser Reste R3 und Rs die übrigen oben angegebenen Bedeutungen mit Ausnahme der eines Wasserstoffatoms innehat oder sofern Rj die oben angegebenen Bedeutungen mit Ausnahme der einer Trifluormethyl-gruppe besitzt oder sofern R2 und R4 die oben angegebenen Bedeutungen mit Ausnahme der eines Wasserstoffatoms oder einer Methylgruppe besitzen,dadurch gekennzeichnet, dass man Perfluoracylresorcine der Formel IIRin der R3 und Rs wie oben definiert sind und D die Gruppe OR4 bedeutet, sofern E die Gruppe -O-CORj darstellt,oder D die Gruppe -O-CORj bedeutet, sofern dann E die Gruppe -OR2 darstellt oder beide Reste D und E eine Gruppe -O-CORj bedeuten, wobei in diesen Gruppen Ri, R2 und R4 wie oben angegeben definiert sind, in Gegenwart einer Lewis-Säure als Katalysator auf Temperaturen zwischen 0 und 150 °C erhitzt.

Verfahren nach Anspruch 4, dadurch gekennzeichnet, dass man als Lewis-Säuren wasserfreies Aluminiumchlorid oder Zinkchlorid in Anwesenheit einer Halogenwasserstoffsäure, Eisen(III)chlorid oder Zinn(IV)chlorid, verwendet.6. Verfahren nach Anspruch 4 oder 5, dadurch gekennzeichnet, dass man als Lösungsmittel Äther, aromatische Kohlenwasserstoffe, wie Chlorbenzol, Nitrobenzol, Toluol, Dichlorbenzol, Xylol, oder Phosphoroxychlorid verwendet und die Umsetzung bei Temperaturen zwischen 0 und 50 °C durchführt.7. Verfahren zur Herstellung neuer substituierter Fluor-acylresorcine der Formel I'C-RORRworinRi eine perfluorierte Alkylgruppe mit 1 bis 8 Kohlenstoffatomen oder die 2,2,3,3-Tetrafluorcyclobutylgruppe;R2 und R4, die gleich oder voneinander verschieden sein können, eine aliphatische Acylgruppe mit 2 bis 18 Kohlenstoffatomen, die Benzoyl-, Salicyloyl- oder Phenylacetylgruppe bedeuten und einer der beiden Reste R2 und R4 zusätzlich eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 10 Kohlenstoffatomen bedeuten kann;R3 und R5, die gleich oder voneinander verschieden sein können, Alkylgruppen mit 3 bis 18 Kohlenstoffatomen, Halogenatomen, die Nitrogruppe oder die p-Toluolsulfonyl-gruppe, die Cyclopentyl-, Cyclohexyl-, Cycloheptyl-, Cyclo-636 835dodecyl-, Methylcyclohexyl-, Dimethylcyclohexyl-, Benzyl-oder Methylthiogruppe oder eine Gruppe der FormelORFORCORin der entweder F die Gruppe -CH2- oder -S- bedeutet, sofern G die Gruppe R5 darstellt oder F die Gruppe R3 ist, sofern G die Gruppe -CH2- oder -S- darstellt und Rj, R2 und R4 wie oben definiert sind;R3 ausserdem die Hydroxy-, Methoxy-, Methyl- oder Cyangruppe;R5 auch eine Methylgruppe bedeuten, es kann aber auch einer der beiden Reste R3 und R5 ein Wasserstoffatom oder die Äthylgruppe bedeuten, sofern dann der andere dieser Reste R3 und R5 die übrigen oben angegebenen Bedeutungen mit Ausnahme der eines Wasserstoffatoms innehat oder sofern Ri die oben angegebenen Bedeutungen mit Ausnahme der einer Trifluormethylgruppe besitzt oder sofern R2 und R4 die oben angegebenen Bedeutungen mit Ausnahme der einer Methylgruppe besitzen, dadurch gekennzeichnet, dass man Perfluoracylresorcine der Formel IIRin der R3 und R5 wie oben definiert sind, und D die Gruppe OR4 bedeutet, sofern E die Gruppe -O-CORi darstellt oder D die Gruppe -O-CORi bedeutet, sofern dann E die Gruppe -OR2 darstellt oder beide Reste D und E eine Gruppe -O-CORi bedeuten, wobei in diesen Gruppen Rl5 R2 und R4 wie oben angegeben definiert sind und R2 und R4 zusätzlich Wasserstoff bedeuten können, in Gegenwart einer Lewis-Säure als Katalysator auf Temperaturen zwischen 0 und 150 °C erhitzt und die erhaltene Verbindungen der Formel I, in denen R2 und/oder R4 Wasserstoff bedeuten, zu Verbindungen der Formel 1" verestert, worin die entsprechenden Reste R2 und/oder R4 eine aliphatische Acylgruppe mit 2 bis 18 Kohlenstoffatomen, die Benzoyl-, Salicyloyl- oder Phenylacetylgruppe bedeuten.8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, dass man als Lewis-Säuren wasserfreies Aluminiumchlorid oder Zinkchlorid in Anwesenheit einer Halogenwasserstoffsäure, Eisen(III)chlorid oder Zinn(IV)chlorid, verwendet.9. Verfahren nach Anspruch 7 oder 8, dadurch gekennzeichnet, dass man als Lösungsmittel Äther, aromatische Kohlenwasserstoffe, wie Chlorbenzol, Nitrobenzol, Toluol, Dichlorbenzol, Xylol, oder Phosphoroxychlorid verwendet und die Umsetzung bei Temperaturen zwischen 0 und 50 °C durchführt.
