Abstract

GLICOENGENHARIA QUIMIOENZIMÁTICA DE ANTICORPOS E FRAGMENTOS FC DOS MESMOS. A presente invenção proporciona mutantes Endo-S recombinantes que exibem atividade de hidrólise reduzida e atividade de transglicosilação aumentada para a síntese das glicoproteínas, em que uma desejada oxazolina sialilada ou oxazolina oligossacarideo sintética é adicionada a um aceptor G1cNAc-proteína de núcleo fucosilado ou não fucosilado. Tais mutantes Endo-S recombinantes são úteis para a remodelação de glicosilação eficiente domínio IgG1-Fc para fornecer diferentes glicoformas de anticorpos que portam estruturalmente Fc N-glicanos bem definidos.
