Abstract
Согласно изобретению предложена эндогликозидаза, обозначаемая EndoS49 и имеющая аминокислотную последовательность SEQ ID NO: 1. EndoS49 выделяли из штамма NZ131 Streptococcus pyogenesона представляет собой гомолог EndoS. EndoS49 обладает специфичной эндогликозидазной активностью в отношении нативного IgG и расщепляет более широкий спектр гликанов Fc, чем EndoS. Получен ее мутант с заменой глутаминовой кислоты в положении 186 SEQ ID NO: 1: указанный мутант не обладает эндогликозидазной активностью, но способен связываться с IgG. Раскрыты способы с применением EndoS49, ее делеций и указанного мутанта, особенно для оценки гликозилирования IgG или выделения IgG.
