Abstract

"PROCESSOS PARA PRODUZIR UMA PROTEìNA SECRETORA HETERóLOGA, E PARA A PRODUçãO SECRETORA DE UMA TRANSGLUTAMINASE MADURA, PEPTIDASE ESPECìFICA PARA PROLINA, POLIPEPTìDEO, E, MOLéCULA DE áCIDO NUCLEICO". A presente invenção refere-se a um processo para a produção secretora de uma proteína estranha, em particular transglutaminase, por meio de uma bactéria coryneform. De acordo com a presente invenção, proporciona-se um processo para a produção secretora de uma proteína estranha, em particular transglutaminase, fazendo-se com que uma bactéria coryneform produza uma proteína estranha industrialmente útil, em particular transglutaminase, e libere eficientemente o produto extracelularmente (isto é, produção secretora). Uma proteína estranha desejada, em particular transglutaminase, é produzida utilizando-se uma construção de expressão em que a seq³ência de genes da proteína estranha desejada contendo a parte pró-estrutura, em particular seq³ência de genes de pró-transglutaminase, é ligada a jusante de uma seq³ência codificando a região de peptídeo sinal de uma bactéria coryneform introduzindo esta construção genética expressional numa bactéria coryneform, cultivando a assim transformada bactéria coryneform, e tratando a proteína liberada extracelularmente com uma protease, etc. para clivar e eliminar a parte pró.
