Description

異種タンパク質の分泌発現用の遺伝子構築物を有するコリネ型細菌を培養すること、および分泌生産された異種タンパク質を回収することを含む、異種タンパク質の製造方法であって、
前記コリネ型細菌が、HrrSAシステムの細胞当たりの分子数が非改変株と比較して低下するように改変されており、
前記遺伝子構築物が、5’から3’方向に、コリネ型細菌で機能するプロモーター配列、コリネ型細菌で機能するシグナルペプチドをコードする核酸配列、および異種タンパク質をコードする核酸配列を含み、
前記異種タンパク質が、前記シグナルペプチドとの融合タンパク質として発現する、方法。

本発明の細菌は、HrrSAシステムの活性が低下するように改変されている。本発明の細菌は、具体的には、HrrSAシステムの活性が非改変株と比較して低下するように改変されている。HrrSAシステムの活性が低下するようにコリネ型細菌を改変することにより、同細菌の異種タンパク質を分泌生産する能力を向上させることができる、すなわち、同細菌による異種タンパク質の分泌生産を増大させることができる。
本発明の細菌は、異種タンパク質を分泌生産できる限り、所望の性質を有していてよい。例えば、本発明の細菌は、細胞表層タンパク質の活性が低下していてよい(WO2013/065869、WO2013/065772、WO2013/118544、WO2013/062029)。また、本発明の細菌は、ペニシリン結合タンパク質の活性が低下するように改変されていてよい(WO2013/065869)。また、本発明の細菌は、メタロペプチダーゼをコードする遺伝子の発現が上昇するように改変されていてよい(WO2013/065772)。また、本発明の細菌は、変異型リボソームタンパク質S1遺伝子(変異型rpsA遺伝子)を有するように改変されていてよい(WO2013/118544)。また、本発明の細菌は、変異型phoS遺伝子を有するように改変されていてよい(WO2016/171224)。また、本発明の細菌は、Tat系分泌装置が増強されていてよい。これらの性質または改変は、単独で、あるいは適宜組み合わせて、利用することができる。
本発明の細菌は、変異型phoS遺伝子を保持するように改変されていてよい。「変異型phoS遺伝子を保持する」ことを、「変異型phoS遺伝子を有する」または「phoS遺伝子に変異を有する」ともいう。また、「変異型phoS遺伝子を保持する」ことを、「変異型PhoSタンパク質を有する」または「PhoSタンパク質に変異を有する」ともいう。
本発明の細菌は、細胞表層タンパク質の活性が低下しているものであってよい。本発明の細菌は、具体的には、細胞表層タンパク質の活性が非改変株と比較して低下しているものであってよい。「細胞表層タンパク質の活性が低下する」とは、特に、細胞表層タンパク質の細胞当たりの分子数が低下することを意味してもよい。以下に、細胞表層タンパク質およびそれをコードする遺伝子について説明する。

本発明の細菌は、タンパク質の分泌系を有する。タンパク質の分泌系は、目的の異種タンパク質を分泌できるものであれば、特に制限されない。タンパク質の分泌系としては、Sec系(Sec系分泌装置)やTat系(Tat系分泌装置)が挙げられる。本発明の細菌は、タンパク質の分泌系が増強されていてもよい。例えば、本発明の細菌は、Tat系分泌装置をコードする遺伝子から選択される1またはそれ以上の遺伝子の発現が上昇するように改変されていてもよい。本発明において、このような改変を「Tat系分泌装置の増強」ともいう。Tat系分泌装置の増強は、特に、Tat系依存シグナルペプチドを利用して異種タンパク質を分泌生産する場合に好適である。Tat系分泌装置をコードする遺伝子の発現を上昇させる手法については特許第4730302号に記載されている。
以下に、HrrSAタンパク質等のタンパク質の活性を低下させる手法について説明する。なお、以下に記載するタンパク質の活性を低下させる手法は、野生型PhoSタンパク質の破壊にも利用できる。
以下に、Tat系分泌装置をコードする遺伝子等の遺伝子の発現を上昇させる手法について説明する。
分泌性タンパク質(secretory protein)は、一般に、プレタンパク質(プレペプチドともいう)またはプレプロタンパク質(プレプロペプチドともいう)として翻訳され、その後、プロセッシングにより成熟タンパク質(mature protein)になることが知られている。具体的には、分泌性タンパク質は、一般に、プレタンパク質またはプレプロタンパク質として翻訳された後、プレ部分であるシグナルペプチドがプロテアーゼ(一般にシグナルペプチダーゼと呼ばれる)によって切断されて成熟タンパク質またはプロタンパク質に変換され、プロタンパク質はプロテアーゼによってさらにプロ部分が切断されて成熟タンパク質になる。よって、本発明の方法においては、異種タンパク質の分泌生産にシグナルペプチドを利用する。なお、本発明において、分泌型タンパク質のプレタンパク質およびプレプロタンパク質を総称して「分泌型タンパク質前駆体」という場合がある。本発明において、「シグナルペプチド」(「シグナル配列」ともいう)とは、分泌性タンパク質前駆体のN末端に存在し、かつ、通常、天然の成熟タンパク質には存在しないアミノ酸配列をいう。

A~Hのアミノ酸配列において、Xaa1はAsn、Gly、Thr、Pro、またはAlaであり、Xaa2はPro、Thr、またはValであり、Xaa3はThrまたはTyrである。また、A~Hのアミノ酸配列において、「Gln-Glu-Thrに成熟CspBの4~X位のアミノ酸残基が付加された」とは、Gln-Glu-ThrのThrに成熟CspBのN末端の4位からX位までのアミノ酸残基が付加されていることを意味する。なお、通常、成熟CspBのN末端の1~3番目のアミノ酸残基はGln-Glu-Thrであり、その場合、「Gln-Glu-Thrに成熟CspBの4~X位のアミノ酸残基が付加されたアミノ酸配列」とは、成熟CspBの1~X位のアミノ酸残基からなるアミノ酸配列と同義である。
上記のようにして得られる本発明の細菌を培養し、異種タンパク質を発現させることにより、菌体外に分泌された多量の異種タンパク質が得られる。
(1)phoS遺伝子に変異を有する自然変異株の取得
WO2014/126260に記載の、生理活性ペプチドであるTeriparatideの分泌発現プラスミドpPKK50TEV-Teriを用いて、WO2002/081694に記載のC. glutamicum YDK010株を形質転換した。なお、pPKK50TEV-Teriは生理活性ペプチドであるTeriparatideの分泌発現用ベクターであって、C. glutamicum ATCC13869株のcspB遺伝子のプロモーター領域、同プロモーターの下流に発現可能に連結された同株のCspBシグナルペプチド、同株の成熟CspBのN末端50アミノ酸残基、ProTEVプロテアーゼの認識配列ENLYFQ、およびTeriparatideの融合タンパク質(以下、CspB50TEV-Teriと表記する)をコードする塩基配列を有するプラスミドである(WO2014/126260)。C. glutamicum YDK010株は、C. glutamicum AJ12036(FERM BP-734)の細胞表層タンパク質CspBの欠損株である(WO2002/081694)。得られた形質転換体を、25 mg/lのカナマイシンを含むCM-Dex寒天培地(グルコース 5 g、硫酸マグネシウム七水和物 0.4 g、硫酸鉄七水和物 0.01 g、硫酸マンガン五水和物 0.01 g、リン酸二水素カリウム 1 g、ビオチン 10 μg、DifcoTM Select Soytone(Becton Dickinson)10 g、BactoTM Yeast Extract(Becton Dickinson)10 g、尿素 3 g、大豆塩酸加水分解液(全窒素量 1.2 g)、寒天抹 20 g、水で1 LにしてpH6.5に調整)上で30℃で培養してコロニーを形成させた。
<配列番号05>および<配列番号06>に記載のプライマーを用いて、PurEluteTM Genomic DNA Kit(EdgeBio)を用いて調製したC. glutamicum YDK0107株の染色体DNAを鋳型として、変異型PhoS(W302C)をコードするphoS遺伝子(変異型phoS遺伝子または変異型phoS(W302C)遺伝子ともいう)を含む約1.5 kbpの領域をPCR法によって増幅した。PCR反応にはPyrobest(R) DNA polymerase(Takara Bio)を用い、反応条件は業者の推奨するプロトコルに従った。
参考例1(2)で構築したプラスミドpBS5T-phoS(W302C)でWO2002/081694に記載のC. glutamicum YDK010株を形質転換した。得られた形質転換体からWO2006/057450に記載の方法に従って菌株の選択を行い、染色体上の野生型phoS遺伝子が変異型phoS遺伝子に置換されたYDK010::phoS(W302C)株を得た。なお、YDK010::phoS(W302C)株は、YDK0107株の染色体DNAを利用せずとも、例えば遺伝子工学的に取得した変異型phoS遺伝子断片等を利用して再現的に構築できる。
(1)hrrA遺伝子欠損用ベクターpBS5TΔhrrAの構築
C. glutamicum ATCC13869株のゲノム配列および二成分制御系HrrSAのレスポンスレギュレーターHrrAをコードするhrrA遺伝子の塩基配列は既に決定されている(GenBank Accession No. AP017557, NCBI locus_tag CGBL_0128750)。
実施例1(1)で構築したpBS5TΔhrrAで、WO2002/081694に記載のC. glutamicum YDK010株および参考例1(3)で構築したYDK010::phoS(W302C)株のそれぞれを形質転換した。得られた形質転換体からWO2006/057450に記載の方法に従って菌株の選択を行い、hrrA遺伝子が欠損したYDK010ΔhrrA株およびYDK010::phoS(W302C)ΔhrrA株を得た。
(1)Protein Lの分泌発現プラスミドの構築
Finegoldia magna由来イムノグロブリン結合タンパク質であるProtein Lのアミノ配列は既に決定されている(GenBank Accession No. AAA25612)。Protein Lは、シグナルペプチド(N末端側1残基目から18残基目)と成熟ペプチド(19残基目から719残基目)から成り、成熟ペプチドのN末端側には5つの抗体結合ドメインが存在する。Protein Lの成熟ペプチド中の5つの抗体結合ドメイン(19残基目から463残基目)のアミノ酸配列を<配列番号11>に示す。C. glutamicumのコドン使用頻度を考慮し、Protein Lの抗体結合ドメインをコードする塩基配列をデザインした。デザインした塩基配列を<配列番号12>に示す。
実施例2(1)で得られたProtein Lの分泌発現プラスミドpPK4_CspAss_ProteinLを用いて、WO2002/081694に記載のC. glutamicum YDK010株および実施例1(2)で得られたYDK010ΔhrrA株のそれぞれを形質転換し、YDK010/pPK4_CspAss_ProteinL株およびYDK010ΔhrrA/pPK4_CspAss_ProteinL株を得た。得られた各形質転換体を、25 mg/lのカナマイシンを含むMMTG液体培地(グルコース 120 g、硫酸マグネシウム七水和物 3 g、硫酸アンモニウム 30 g、リン酸二水素カリウム 1.5 g、硫酸鉄七水和物 0.03 g、硫酸マンガン五水和物 0.03 g、チアミン塩酸塩0.45 mg、ビオチン 0.45 mg、DL-メチオニン 0.15 g、大豆塩酸加水分解液(全窒素量 0.2 g)、炭酸カルシウム 50 g、水で1 LにしてpH7.0に調整)でそれぞれ30℃、72時間培養した。培養終了後、各培養液を遠心分離して得られた培養上清3.0 μlを還元SDS-PAGEに供してからSYPRO Ruby(Life technologies)にて染色を行った。その結果、YDK010ΔhrrA株では、YDK010株と比較して、Protein L分泌量が顕著に向上していた(図1)。染色後、画像解析ソフトMulti Gauge(FUJIFILM)を用いてProtein Lのバンド強度の数値化を行い、YDK010ΔhrrA株でProtein Lを発現させた際のバンド強度の平均値を、YDK010株でProtein Lを発現させた際のバンド強度の平均値を1とした時の相対値として算出した。その結果、YDK010ΔhrrA株では、YDK010株と比較して、Protein L分泌量が約2.0倍に向上していることが確認された(表1)。このことから、ΔhrrA変異(hrrA遺伝子の欠損)は、Sec系のCspA分泌シグナルを用いたProtein L分泌生産において、分泌量向上をもたらす有効変異であることが明らかとなった。
(1)CspBの成熟タンパク質のN末端6アミノ酸残基を融合したLiver-type Fatty acid-binding protein(LFABP)の分泌発現プラスミドの構築
ヒトのLiver-type fatty acid-binding protein(以下、LFABPと表記する)のアミノ酸配列は既に決定されている(RefSeq Accession No. NP_001434)。このアミノ酸配列を<配列番号13>に示した。C. glutamicumのコドン使用頻度を考慮し、LFABPをコードする塩基配列をデザインした。さらに、C. glutamicum ATCC13869株由来のCspBのシグナルペプチド30アミノ酸残基、同株由来のCspB成熟タンパク質のN末端6アミノ酸残基、Factor Xaプロテアーゼの認識配列IEGR、およびLFABPの融合タンパク質(以下、CspB6Xa-LFABPと表記する)と、その融合タンパク質をコードする塩基配列をデザインした。デザインした融合タンパク質をコードする塩基配列を<配列番号14>に、融合タンパク質のアミノ酸配列を<配列番号15>に示した。
実施例3(1)で得られたCspB6Xa-LFABPの分泌発現プラスミドpPK4_CspB6Xa-LFABPを用いて、参考例1(3)で得られたYDK010::phoS(W302C)株および実施例1(2)で得られたYDK010::phoS(W302C)ΔhrrA株のそれぞれを形質転換し、YDK010::phoS(W302C)/pPK4_CspB6Xa-LFABP株およびYDK010::phoS(W302C)ΔhrrA/pPK4_CspB6Xa-LFABP株を得た。得られた各形質転換体を、25 mg/lのカナマイシンを含むMMTG液体培地(グルコース 120 g、硫酸マグネシウム七水和物 3 g、硫酸アンモニウム 30 g、リン酸二水素カリウム 1.5 g、硫酸鉄七水和物 0.03 g、硫酸マンガン五水和物 0.03 g、チアミン塩酸塩0.45 mg、ビオチン 0.45 mg、DL-メチオニン 0.15 g、大豆塩酸加水分解液(全窒素量 0.2 g)、炭酸カルシウム 50 g、水で1 LにしてpH7.0に調整)でそれぞれ30℃、72時間培養した。培養終了後、各培養液を遠心分離して得られた培養上清2.0 μlを還元SDS-PAGEに供してからSYPRO Ruby(Life technologies)にて染色を行った。その結果、YDK010::phoS(W302C)ΔhrrA株では、YDK010::phoS(W302C)株と比較して、CspB6Xa-LFABP分泌量が顕著に向上していた(図2)。染色後、画像解析ソフトMulti Gauge(FUJIFILM)を用いてCspB6Xa-LFABPのバンド強度の数値化を行い、YDK010::phoS(W302C)ΔhrrA株でCspB6Xa-LFABPを発現させた際のバンド強度の平均値を、YDK010::phoS(W302C)株でCspB6Xa-LFABPを発現させた際のバンド強度の平均値を1とした時の相対値として算出した。その結果、YDK010::phoS(W302C)ΔhrrA株では、YDK010::phoS(W302C)株と比較して、CspB6Xa-LFABP分泌量が約1.3倍に向上していることが確認された(表2)。このことから、ΔhrrA変異(hrrA遺伝子の欠損)は、Sec系のCspB分泌シグナルを用いた、YDK010::phoS(W302C)株におけるCspB6Xa-LFABP分泌生産においても、分泌量向上をもたらす有効変異であることが明らかとなった。
(1)Tat系分泌装置をコードするtatABC遺伝子とTorAシグナル配列が付加されたGreen Fluorescent Protein(GFP)をコードする遺伝子の共発現プラスミドの構築
(a)pPK4ベクター中のNaeI認識サイトを改変したベクター(pPK5)の構築
特開平9-322774に記載のpPK4の中には、制限酵素NaeIの認識配列が一ヵ所存在する。この配列を改変するため、NaeI認識配列gccggcをgcaggcに改変した配列とpPK4におけるその周辺配列を含む<配列番号16>および<配列番号17>に記載のプライマーを合成した。次に、pPK4を鋳型として、<配列番号16>と<配列番号17>のプライマーを用いて、約5.6 kbpのプラスミド全長をPCR法によって増幅した。PCR反応にはPyrobest(R) DNA polymerase(Takara Bio)を用い、反応条件は95℃ 5分、(95℃ 30秒、55℃ 1分、72℃ 12分)x 12cycleで行った。
次に、WO2005/103278に記載のTat系分泌装置の増幅プラスミドであるpVtatABCを鋳型にして、<配列番号18>と<配列番号19>のプライマーを用いて、tatABC遺伝子をコードする配列を含む約3.7 kbpのDNA断片をPCR法によって増幅した。<配列番号19>のプライマーには制限酵素KpnIとApaIの認識配列がデザインしてある。PCRにはPyrobest(R) DNA polymerase(Takara Bio)を用い、反応条件は業者の推奨するプロトコルに従った。このDNA断片の末端をBKL Kit(Takara Bio)を用いてリン酸化し、別途KpnI処理し、さらにBKL Kit(Takara Bio)を用いて平滑末端化し、さらにCIAP(Takara Bio)を用いて末端を脱リン酸化処理したpPK5ベクターに挿入することによって、tatABC遺伝子搭載ベクターであるpPK5-tatABCを構築した。ライゲーション反応にはDNA Ligation Kit Ver.2.1(Takara Bio)を用い、反応条件は業者の推奨するプロトコルに従った。挿入断片の塩基配列決定の結果、予想通りの遺伝子が挿入されていることを確認した。塩基配列の決定はBigDye(R) Terminator v3.1 Cycle Sequencing Kit(Applied Biosystems)と3130 Genetic Analyzer(Applied Biosystems)を用いて行った。
(b)で構築したpPK5-tatABCプラスミド中のtatABC遺伝子領域の中には、制限酵素KpnIおよびXbaIの認識配列が1ヵ所ずつ存在する。これらの配列を改変するため、KpnI認識配列ggtaccをggaaccに改変した配列とpPK5-tatABCにおけるその周辺配列を含む<配列番号20>および<配列番号21>に記載のプライマーと、XbaI認識配列tctagaをtgtagaに改変した配列とpPK5-tatABCにおけるその周辺配列を含む<配列番号22>および<配列番号23>に記載のプライマーを合成した。
次に、得られたPCR産物を制限酵素DpnIで処理し、メチル化されている鋳型DNAを消化した。DpnI消化後に得られた非メチル化プラスミドを、E. coli JM109(Takara Bio)のコンピテントセルに導入し、プラスミドを取得した。こうしてtatABC遺伝子領域内のXbaI認識配列を改変したベクターであるpPK5-tatABCΔKpnIΔXbaIを取得した。塩基配列決定の結果、予想通りの遺伝子が構築されていることを確認した。塩基配列の決定はBigDye(R) Terminator v3.1 Cycle Sequencing Kit(Applied Biosystems)と3130 Genetic Analyzer(Applied Biosystems)を用いて行った。
Appl. Environ. Microbiol., 72, 7183-7192(2006)に記載のpPTGFPを鋳型として、<配列番号24>と<配列番号25>のプライマーを用いて、C. glutamicum ATCC13869株由来のcspB遺伝子のプロモーター領域、E. coli由来のTorAシグナル配列をコードする塩基配列、GFPをコードする塩基配列を含む約1.4 kbpのDNA断片をPCR法によって増幅した。PCRにはPyrobest(R) DNA polymerase(Takara Bio)を用い、反応条件は業者の推奨するプロトコルに従った。増幅したDNA断片をアガロースゲル電気泳動後、目的のバンドを切り出し、Wizard(R) SV Gel and PCR Clean-Up System(Promega)を用いてゲルより回収した。回収したDNA断片を、インフュージョン反応により実施例3(1)(c)に記載のpPK6のKpnI部位に挿入することによってGreen Fluorescent Protein(GFP)の分泌発現プラスミドpPK6_T_GFPを得た。インフュージョン反応にはIn-Fusion(R) HD Cloning Kit(Takara Bio)を用い、反応条件は業者の推奨するプロトコルに従った。挿入断片の塩基配列決定の結果、予想通りのGFPをコードする遺伝子が構築されていることを確認した。塩基配列の決定はBigDye(R) Terminator v3.1 Cycle Sequencing Kit(Applied Biosystems)と3130 Genetic Analyzer(Applied Biosystems)を用いて行った。
実施例4(1)で得られたGFPの分泌発現プラスミドpPK6_T_GFPを用いて、参考例1(3)で得られたYDK010::phoS(W302C)株および実施例1(2)で得られたYDK010::phoS(W302C)ΔhrrA株のそれぞれを形質転換し、YDK010::phoS(W302C)/pPK6_T_GFP株およびYDK010::phoS(W302C)ΔhrrA/pPK6_T_GFP株を得た。得られた各形質転換体を、25 mg/lのカナマイシンを含むMMTG液体培地(グルコース 120 g、硫酸マグネシウム七水和物 3 g、硫酸アンモニウム 30 g、リン酸二水素カリウム 1.5 g、硫酸鉄七水和物 0.03 g、硫酸マンガン五水和物 0.03 g、チアミン塩酸塩0.45 mg、ビオチン 0.45 mg、DL-メチオニン 0.15 g、大豆塩酸加水分解液(全窒素量 0.2 g)、炭酸カルシウム 50 g、水で1 LにしてpH7.0に調整)でそれぞれ30℃、72時間培養した。培養終了後、各培養液を遠心分離して得られた培養上清5.0 μlを還元SDS-PAGEに供してからSYPRO Orange(Life technologies)にて染色を行った。その結果、YDK010::phoS(W302C)ΔhrrA株では、YDK010::phoS(W302C)株と比較して、GFP分泌量が向上していた(図3)。染色後、画像解析ソフトMulti Gauge(FUJIFILM)を用いてGFPのバンド強度の数値化を行い、YDK010::phoS(W302C)ΔhrrA株でGFPを発現させた際のバンド強度の平均値を、YDK010::phoS(W302C)株でGFPを発現させた際のバンド強度の平均値を1とした時の相対値として算出した。その結果、YDK010::phoS(W302C)ΔhrrA株では、YDK010::phoS(W302C)株と比較して、GFP分泌量が約1.6倍に向上していることが確認された(表3)。このことから、ΔhrrA変異(hrrA遺伝子の欠損)は、Tat系のTorA分泌シグナルを用いた、YDK010::phoS(W302C)株におけるGFP分泌生産においても、分泌量向上をもたらす有効変異であることが明らかとなった。

Claims (18)
- 異種タンパク質の分泌発現用の遺伝子構築物を有するコリネ型細菌を培養すること、および分泌生産された異種タンパク質を回収することを含む、異種タンパク質の製造方法であって、
前記コリネ型細菌が、HrrSAシステムの細胞当たりの分子数が非改変株と比較して低下
するように改変されており、
前記コリネ型細菌が、コリネバクテリウム属細菌であり、
前記HrrSAシステムが、HrrSタンパク質およびHrrAタンパク質からなり、
前記遺伝子構築物が、5’から3’方向に、コリネ型細菌で機能するプロモーター配列、コリネ型細菌で機能するシグナルペプチドをコードする核酸配列、および異種タンパク質をコードする核酸配列を含み、
前記異種タンパク質が、前記シグナルペプチドとの融合タンパク質として発現し、
前記HrrSタンパク質が、下記(1a)、(1b)、または(1c)に記載のタンパク質であり:
(1a)配列番号63に示すアミノ酸配列を含むタンパク質;
(1b)配列番号63に示すアミノ酸配列において、1~10個のアミノ酸残基の置換、欠失、挿入、および/または付加を含むアミノ酸配列を含み、且つ、HrrSAシステムのセ
ンサーキナーゼとしての機能を有するタンパク質;
(1c)配列番号63に示すアミノ酸配列に対して90%以上の同一性を有するアミノ酸配列を含み、且つ、HrrSAシステムのセンサーキナーゼとしての機能を有するタンパク質
、
前記HrrAタンパク質が、下記(2a)、(2b)、または(2c)に記載のタンパク質である、方法:
(2a)配列番号65に示すアミノ酸配列を含むタンパク質;
(2b)配列番号65に示すアミノ酸配列において、1~10個のアミノ酸残基の置換、欠失、挿入、および/または付加を含むアミノ酸配列を含み、且つ、HrrSAシステムのレ
スポンスレギュレーターとしての機能を有するタンパク質;
(2c)配列番号65に示すアミノ酸配列に対して90%以上の同一性を有するアミノ酸配列を含み、且つ、HrrSAシステムのレスポンスレギュレーターとしての機能を有するタ
ンパク質。 - HrrSAシステムの細胞当たりの分子数が、HrrSタンパク質およびHrrAタンパク質の一方
または両方の細胞当たりの分子数を低下させることにより、低下した、請求項1に記載の方法。 - 少なくともHrrAタンパク質の細胞当たりの分子数が低下した、請求項2に記載の方法。
- hrrS遺伝子および/またはhrrA遺伝子の発現を低下させることにより、またはhrrS遺伝子および/またはhrrA遺伝子を破壊することにより、HrrSタンパク質および/またはHrrAタンパク質の細胞当たりの分子数が低下した、請求項2または3に記載の方法。
- hrrS遺伝子および/またはhrrA遺伝子を欠損させることにより、HrrSタンパク質および/またはHrrAタンパク質の細胞当たりの分子数が低下した、請求項2~4のいずれか一項に記載の方法。
- 前記コリネ型細菌が、さらに、変異型PhoSタンパク質をコードするphoS遺伝子を保持するように改変されており、
前記変異が、野生型PhoSタンパク質において、配列番号4の302位のトリプトファン残基に相当するアミノ酸残基が芳香族アミノ酸およびヒスチジン以外のアミノ酸残基に置換される変異であり、
前記野生型PhoSタンパク質が、下記(a)、(b)、又は(c)に記載のタンパク質である、請求項1~5のいずれか一項に記載の方法:
(a)配列番号4、26、27、28、29、または30に示すアミノ酸配列を含むタンパク質;
(b)配列番号4、26、27、28、29、または30に示すアミノ酸配列において、1~10個のアミノ酸残基の置換、欠失、挿入、または付加を含むアミノ酸配列を含み、且つ、PhoRSシステムのセンサーキナーゼとしての機能を有するタンパク質;
(c)配列番号4、26、27、28、29、または30に示すアミノ酸配列に対し90%以上の同一性を有するアミノ酸配列を含み、且つ、PhoRSシステムのセンサーキナーゼ
としての機能を有するタンパク質。 - 前記芳香族アミノ酸およびヒスチジン以外のアミノ酸残基が、リジン残基、アラニン残基、バリン残基、セリン残基、システイン残基、メチオニン残基、アスパラギン酸残基、またはアスパラギン残基である、請求項6に記載の方法。
- 前記シグナルペプチドがTat系依存シグナルペプチドである、請求項1~7のいずれか
1項に記載の方法。 - 前記Tat系依存シグナルペプチドが、TorAシグナルペプチド、SufIシグナルペプチド、PhoDシグナルペプチド、LipAシグナルペプチド、およびIMDシグナルペプチドからなる群より選択されるいずれか1つのシグナルペプチドである、請求項8に記載の方法。
- 前記コリネ型細菌が、さらに、Tat系分泌装置をコードする遺伝子から選択される1ま
たはそれ以上の遺伝子の発現が非改変株と比較して上昇するように改変されている、請求項8または9に記載の方法。 - 前記Tat系分泌装置をコードする遺伝子が、tatA遺伝子、tatB遺伝子、tatC遺伝子、お
よびtatE遺伝子からなる、請求項10に記載の方法。 - 前記シグナルペプチドがSec系依存シグナルペプチドである、請求項1~11のいずれ
か1項に記載の方法。 - 前記Sec系依存シグナルペプチドが、PS1シグナルペプチド、PS2シグナルペプチド、お
よびSlpAシグナルペプチドからなる群より選択されるいずれか1つのシグナルペプチドである、請求項12に記載の方法。 - 前記遺伝子構築物が、コリネ型細菌で機能するシグナルペプチドをコードする核酸配列と異種タンパク質をコードする核酸配列との間に、さらに、Gln-Glu-Thrを含むアミノ酸
配列をコードする核酸配列を含む、請求項1~13のいずれか1項に記載の方法。 - 前記遺伝子構築物が、Gln-Glu-Thrを含むアミノ酸配列をコードする核酸配列と異種タ
ンパク質をコードする核酸配列との間に、さらに、酵素的切断に使用されるアミノ酸配列をコードする核酸配列を含む、請求項14に記載の方法。 - 前記コリネ型細菌が、コリネバクテリウム・グルタミカムである、請求項1~15のいずれか1項に記載の方法。
- 前記コリネ型細菌が、コリネバクテリウム・グルタミカムAJ12036(FERM BP-734)に由来する改変株またはコリネバクテリウム・グルタミカムATCC 13869に由来する改変株である、請求項16に記載の方法。
- 前記コリネ型細菌が、細胞表層タンパク質CspBをもともと有しないコリネ型細菌であるか、cspB遺伝子の発現を低下させることにより、またはcspB遺伝子を破壊することにより、細胞表層タンパク質CspBの細胞当たりの分子数が非改変株と比較して低下するように改変されたコリネ型細菌である、請求項1~17のいずれか1項に記載の方法。
