L'objet de la présente invention est la préparation de la 17c(-hydroxy 2, 4, 21-tribromo pregnane 3, 11, 20-trione de for- mule I, inconnue jusqu'ici. L'intérêt de ce composé, I, réside
<Desc/Clms Page number 2>
<Desc/Clms Page number 3>

<Desc/Clms Page number 4>

The object of the present invention is the preparation of 17c (-hydroxy 2, 4, 21-tribromo pregnane 3, 11, 20-trione of formula I, hitherto unknown. The interest of this compound, I , resides
<Desc / Clms Page number 2>
in that it allows access to 17Ó-hydroxy 21-bromo ¯1,4 -pregnadiene 3, 11, 20-trione, II. By proceeding, according to known methods, dehydration is carried out without attacking the bromine by
21, which could not be expected,
EMI2.1
Saponification of the novel 21-bromodiene-1,4,11 thus obtained leads to ¯1 -dehydrocortisone, III, or metacortandracin, while by acetoxylation, the corresponding acetate, IV.
EMI2.2
17O-hydroxy 2, 4, 21-tribromo pregnane 3, 11, 20-tri-one, I, can exist in two stereoisomeric forms. These two forms, which depend on the position occupied in space by, bromine in 2, are clearly differentiated by their rotary power.
One of them, 17Ó-hydroxy 2Ó, 4ss, 21-tribromo pre- gnane 3,11, 20-trione is levorotatory, while the other, 17Ó-hydroxy 2ss, 4ss, 21-tribromo pregnan @ e 3,11,20-trione is dextrorotatory. / v.
<Desc / Clms Page number 3>
Obtaining one or the other of these forms depends essentially on the method of bromination used at the time of the fixation of the bromine on the 2 carbon.

known.t1; pplicated to one or other of the forms stere.oi, somers of 1 $ .- cc4gduit directly to the sought diene, II. One can for this, set operate with lithium chloride within dimethylformamide (J. Am.dém.Soc., 1953, 75, 4432), or go through the interim of a dinitrophenyl hydrazone according to Demaecker and Martin, (Nature 1954, 173 p.266).
By saponification of the bromine at 21 of diene, II, one accesses directly metacortandracin or Ô 1-dehydrocortisone, III.
If, instead of saponifying the bromine, one reacts with the brominated diene, II, sodium, potassium, lithium or ar-

gent of organic acids, the result is esters of 1-dehydro-cortisone. According to a variant, 'to obtain, for example, metacortandracin acetate, IV, sodium iodide can first be reacted with compound II to replace bromine with iodine.
<Desc / Clms Page number 4>
The potassium acetate then leads, by formation of iodide of

potassium, β-1-Dehydrocortisone acetate, IV.
Each of the following examples illustrates one of the steps of the invention without, however, limiting it. It is in particular possible to change the starting material, or to vary the temperature or the nature of the oxidizable alcohol employed or that of the solvent, to use inorganic salts other than lithium chloride for debromhydration, without s' for this to be removed from the scope of the invention.

2.65 g of levorotatory compound 1, obtained according to Example 2, are treated with lithium chloride in the presence of dimethylformamide. 1.4 g of crude diene are obtained which is purified by washing with acetone and then by recrystallization from aqueous dimethylformamide. After draining and drying, the 21-bromo 17Ó-hydroxy ¯1,4-pregnaidene 3, 11, 20-trione is obtained which is identical to the compound separated in Example 3.
Example 5: Preparation of ¯ 1-dehydrocortiosone acetate. -
3 g of 21-bromo 17Ó-hydroxy ¯1,4-pregnadiene 3, 11, 20-trione obtained according to Example 3 or 1 are mixed with 1.3 g of dry sodium iodide and 45 cm3 are added. acetone and 21 cm3 of absolute alcohol at room temperature. The diene dissolves very quickly and the sodium bromide precipitates. The mixture is left in contact for fifteen minutes, the sodium bromide is filtered off and washed with a little anhydrous acetone.
The washing acetone is combined with the filter of sodium bromide, 3.55 g of dry potassium acetate are added.

and 0.3 cm3 of water and refluxed for one and a quarter hours under a stream of nitrogen. The cooled mixture is poured into ice-cold water, filtered off and washed with water until the latter no longer contains potassium iodide. After spinning and drying,
