Dans sa demande de brevet/Il' 430.722 du 14 avril 1956, la Société demanderesse a décrit un procédé d'obtention de la 2,4-dibromo 17[alpha]-hydroxy 21-acôtoxy pregnane 3,11,20-trione et la transformation de ce composé en #1-déhydrocortisone ou predni- sone.
Ce procédé consiste en une bromuration au sein du dioxane de la 4-bromo 17-hydroxy 21-acétoxy pregnane 3,11,20-trione qui conduit au dérivé dibromé cherché dont la double débromhydra-

Exemple 1 : Préparation de la 3,20-disemicarbazone 17[alpha]-hydroxy 21-acétoxy pregnane 3,11,20- trione, II.
Dans un ballon à trois tubulures muni d'un agitateur, d'un thermomètre et d'un barbotage d'azote, on introduit 325 cm3 d'acide acétique à 1 % d'eau, 50g de 17[alpha]-hydroxy 21-acétoxy pro- gnane 3,11,20-trione, I, et 60g de chlorhydrate de semicarbazide.
On chauffe à 35 puis ajoute rapidement 120g de phosphate diso- dique sec. Tout en -continuant l'agitation, on chauffe pendant six heures à 45 . On verse ensuite dans l'eau glacée et neutra- lise en refroidissant, par agitation avec 500 cm3 environ d'am- moniaque à 2 Bé jusqu'à ce que le pH soit de 8. On abandonne la disemicarbazone II brute pendant trente minutes environ, es- sore et lave à l'eau. Après séchage, on obtient environ 60g soit un rendement de 100 % en disemicarbazone brute pouvant être utilisée directement pour la réduction. Le produit retient en- viron 10,; d'eau, [[alpha]]D20 = + 45 à +50 (c= 1 %, diméthylforma-. mide à 10 % d'eau).
Pour la purification,on lave le produit à chaud à l'éther isopropylique, refroidit, essore et sèche, lave ensuite à froid d'abord à l'acétone (5 volumes) puis à l'éther isopro- pylique, sèche, dissout dans 3,5 volumes de méthanol, passe noir et ajoute 20 volumes d'acétone. Après avoir glacé, on @@re le produit et sèche après lavage au méthanol et à l'acé- @ composé II pur est ainsi obtenu avec un rendement fenferme 4 % d'eau. [ ]D20= + 55 (c= 1 @, diméthyl- ' , d'eau).

Après sécha- ge, on obtient 3,25g, soit un rendement de 40%, de produit V, F. = 199 et 218 , [[alpha]]D20= + 86 + 2 (c= 1%, acétone) que l'on utilise tel quel pour le stade suivant.
Après purification par recristallisation dans l'iso- propanol, on obtient le composé V pur présentant les constantes de la littérature. F= 199 puis 218 , 2[[alpha]]D20= + 85 + 1,5 (c= 1%, acétone).
Exemple 5 : Préparation de la 2,4-dibromo 11 @, 17 '- dihydroxy 21-acétoxy prégnane 3,20-dione, VI.-
3g de produit V obtenu selon l'exemple 4 sont dissous dans 45cm3 de dioxane'. On fait barboter un léger courant d'azo- te et ajoute, sous agitation, 0,2 cm3 d'une solution d'acide brom- hydrique anhydre(à 0,956 mol. d'acide bromhydrique par litre), puis, à 20 environ, on introduit en quelques minutes 35,5 cm3 d'une solution acétique de. brome à 7,7g %. Un verse dans un mélange d'eau et de glace renfermant 2,4g d'acétate de sodium cristallisé. On essore et lave le produit sur filtre à l'eau jusqu'à ce que les eaux de lavage soient exemptes de bromure, puis sèche sous vide.
On obtient 1,1g de composé VI brut (soit un rendement de 98,5%). Le produit titre 27% de brome (théorie et renferme 1,5 % d'eau. On le purifie par empâtage dans l'acide acétique d'abord à 50 % et ensuite dans l'acide acéti- que pur, on le dissout ensuite dans 10 volumes d'acétone,filtre et ajoute 10 volumes d'eau. Le composé VI cristallise en pail- lettes. On essore, lave à l'eau et sèche. Le rendement de la purification est de 20 %. F.(déc.) 195-200 , [ ]D20=0 ¯2 (c = 0,5 @, acétone).
