Procédé de préparation de peptides
Dans son brevet suisse No 338197 la titulaire a décrit un procédé d'obtention de peptides, par l'in- termédiaire des N-trityl-peptides correspondants, ledit procédé consistant en une transformation d'un Ntrityl-aminoacide en son anhydride mixte que l'on fait réagir avec 1'ester d'un aminoacide ou peptide, le produit de condensation étant ensuite saponifié pour libérer le groupement carboxyle, puis détritylé par chauffage avec l'acide acétique aqueux.
Dans son brevet suisse No 343407 la titulaire a démontré que la réaction de condensation d'un N-trityl-aminoacide sur 1'ester d'un aminoacide s'effectue avantageusement si l'on opère en présence de dicyclohexylcarbodiimide, agent de condensation préconisé en synthèse peptidique par Sheehan et Hess (J. Am.
Chem. Soc., 1955, 77, 1066) pour d'autres dérivés d'aminoacides, 1'ester du peptide N-tritylé ainsi obtenu étant ensuite saponifié puis détritylé par l'acide acétique aqueux chaud.
Or, il a maintenant été trouvé que la condensation de N-trityl-aminoacides dibasiques comportant une fonction amide primaire avec les esters d'aminoacides ou de peptides conduit de façon surprenante à de bons rendements si on 1'effectue à basse température en présence de dicyclohexylcarbodiimide.
Une telle réaction, effectuée à température ordinaire et, à plus forte raison, à température élevée, se solde par des rendements inférieurs dus à des réactions parasites de la fonction amide primaire.
La présente invention a pour objet un procédé de préparation des peptides amidés de formule gé- nérale :

dans laquelle n est un multiple de 1, compris entre 1 et 3, R'et R"représentant l'un le groupe-NHS et l'autre le groupe-OH, en solution dans un solvant, avec un ester d'aminoacide ou de peptide à fonction amine non salifiée, en présence de dicyclo- : hexylcarbodiimide, et détrityle, par chauffage avec l'acide acétique aqueux, le N-trityl-peptide. Avant d'effectuer la détritylation, on peut procéder à une hydrolyse ou hydrogénolyse du groupe cité du tritylpeptide.
L'importance du rôle physiologique d'aminoacides dibasiques monoamidés tels que la glutamine, l'isoglutamine et l'asparagine, a suggéré l'étude du comportement de leurs peptides (cf. H. K. Miller et
H. Welsch, Archiv. biochem. biophysics, 1952, 35, 176). L'excellente utilisation des asparaginyl-peptides dans certaines expériences sur la croissance de microorganismes est particulièrement utile.
Ainsi, Miller,
Neidle et Waelsch (Archiv. biochem. biophysics, 1955, 56, 11) ont étudié Faction de l'asparaginyl- leucine sur des cultures de Leuconostoc Mesenteroides et Miller et Waelsch (Archiv. biochem. biophysics, 1952, 35, 184), celle de la glutaminyl-glycine et de l'asparaginyl-glycine sur Lactobacillus Arabinosus et Leuconostoc Mesenteroides.
Les hormones post-hypophysaires : l'ocytocine et les vasopressines dont l'importance biologique est considérable, sont des peptides comportant de l'aspa- ragine et de la glutamine (cf. J. M. Swan et V. du
Vigneaud, J. Am. Chem. Soc., 1954, 76, 3110).
Dans un mode préféré de mise en oeuvre du procédé de l'invention, la réaction de condensation peptidique est effectuée à une température comprise entre-10 et 00 C, le solvant étant le chlorure de méthylène.
Les exemples suivants illustrent l'invention.
La préparation de quelques matières premières telles que la N-trityl-glutamine et la N-trityl-asparagine est donnée ici à titre indicatif.
Les points de fusion sont des points de fusion instantanée et ont été déterminés sur bloc de Maquenne.
Exemple 1
On part de N-trityl-L-asparagine, qui a été obtenue comme suit :
A. Préparation de N-trityl-L-asparaginate de trityle
[formule I :
n = 1, R' = - NH2, R" = - O - C(C6H5)3]
A une solution de 1, 5 g de monohydrate de Lasparagine dans 25 cm3 d'eau et 7 cm3 de triéthyl- amine, on ajoute 12 g de chlorure de trityle dissous dans 70 cm3 d'éther. On agite trois heures et demie à 00 C puis ajoute de l'acide chlorhydrique dilué jusqu'à légère acidité. Le précipité est séparé, lavé à l'eau puis à l'Úther. On recristallise en benzène-éther.
Rendement : 83 O/o ; F. 218-222 C ; [a] 2D0 =-64
+ 2 (c = 2 11/o, chloroforme). Le produit est insoluble dans 1'eau et l'Úther, soluble dans l'alcool, l'acétone, le benzène, le chloroforme et l'acétate d'éthyle.

A'. Préparation de
N-trityl-L-asparaginate de benzyle
[formule I :
n = 1, R' = - NH2, R" = - O - CH2 - C6H5]
A une solution bouillante de 33, 3 g de N-trityl
L-aspartate de dibenzyle (obtenus selon Velluz et coll., Bull. Soc. Chim., 1956, p. 1464) dans 180 cnr de dioxane et 30 cm3 d'eau, on ajoute peu à peu, en trente minutes, 66 cm3 de potasse normale.
Quinze minutes après l'introduction, on élimine le dioxane sous vide, dilue à l'eau, acidifie par 70 cm3 d'acide chlorhydrique N et extrait au chlorure de méthylène. La solution organique est lavée à l'eau, séchée sur sulfate de sodium et concentrée à 100 cm3. On y envoie un courant d'ammoniac jusqu'à légère alcalinité (pH 8-9), ajoute 16 g de di cyclohexylcarbodiimide et maintient le pH vers 8-9 par un léger courant d'ammoniac. Après deux heures, on sépare la dicyclohexylurée précipitée (12, 4 g, rendement : 71 % ) et chasse le solvant sous vide à 300C. Le résidu, repris au méthanol, fournit 12 g (43 % ) de N-trityl-L-asparaginate de benzyle. On recristallise en alcool absolu.
F. 166-167o C, [a] 2D0 =-50 ¯ 1 (c = 2l /o, chloroforme). Le produit est insoluble dans l'eau, peu soluble dans l'alcool et l'Úther, soluble dans le chloroforme.
Analyse : CnoH2803N2 = 464, 5

B. Préparation de la N-trityl-L-asparagine
[formule I : n= 1, R'=-NH2, R"=-OH] a) par hydrolyse du N-trityl-L-asparaginate
de trityle
A une solution de 10 g de N-trityl-L-aspa-
raginate de trityle, obtenu selon A, dans 200 cm3
d'acétone, on ajoute 10 cm3 d'eau et 5 cm3 de
pyridine et laisse reposer une nuit à température
ambiante. On distille l'acétone sous vide à 400 C,
reprend le résidu huileux par 50 cm3 d'eau et
50 cm3 d'éther et ajoute de l'acide chlorhydrique
N jusqu'à légère acidité.
On sépare la N-trityl
L-asparagine hydratée qui précipite (5, 2 g, ren
dement : 85 /o). On recristallise en méthyl-
ÚthylcÚtone, F. 194-196 C, [a] % 20 =-7O (c = 2 /o,
méthanol). Zervas et Theodoropoulos (J. Am.
Chem. Soc., 1956, 78, 1359) indiquent F. 173
174 C et [a] 2D0 = 6, 1 ¯ (c = 3, 5"/o, méthanol).
Le produit est insoluble dans l'éther, 1'eau et le
benzène, peu soluble dans le chloroforme, solu
ble dans l'alcool et l'acétone. b) par hydrogenolyse du N-trityl-L-asparaginate
de benzyle
On hydrogène à pression atmosphérique et
température ambiante 2 g de N-trityl-L-aspara
ginate de benzyle, obtenu selon A', dans 40 cm3
d'acétate d'éthyle en présence de 0, 5 g de noir
palladié et 0, 6 cm3 de triéthylamine. Après sépa-
ration du catalyseur, la solution est agitée avec
15 cm3 d'acide chlorhydrique 0, 3 N. On recueille
1, 3 g (80 /o) de N-trityl-L-asparagine hydratée.
C. Préparation de la
N-trityl-L-asparagine (sel de diéthylamine)
[formule I :
n = 1, R'NH2, R"OH, NH (C2H5) 2]
On hydrogène à la température ambiante et à la pression atmosphérique en présence de noir palladié 15 g de N-trityl-L-asparaginate de trityle obtenu selon A., dans 150 cm3 d'alcool absolu et 3 cm3 de diéthylamine. Après séparation du catalyseur par filtration, on concentre la solution à 10 cm3, ajoute 0, 5 cm3 de diéthylamine puis précipite par 200 cm3 d'éther.
On recueille 9, 85 g (90 /o) de sel de diéthyl- amine de la N-trityl-L-asparagine, F. 152-155 C, [a] 20 290 2 (c = 1 O/o, chloroforme). Zervas et Theodoropoulos (J. Am. Chem. Soc., 1956, 78, 1359) indiquent F. 150-151 C. Le produit est insoluble dans l'éther, peu soluble dans l'acétone, soluble dans l'eau, l'alcool, le chloroforme et le benzène chaud.
D. Préparation du
N-trityl-L-asparaginyl-glycinate de benzyle
[formule I : n = 1, R' = - NH2, R"=-HN-CH2-C02-CH2-CGH5]
On refroidit Ó - 10¯ C un mélange de 2, 25 g de
N-trityl-L-asparaginate de diéthylamine, obtenu selon
C., de 1, 01 g de chlorhydrate de glycinate de benzyle et de 0, 925 g de N-trityl-L-asparagine, obtenue selon
B., dans 40 cm3 de chlorure de méthylène et ajoute 1, 6 g de dicyclohexylcarbodiimide, le milieu réactionnel étant suffisamment basique pour que la fonction amine soit libérée. On agite pendant cinq heures en laissant remonter lentement la température à 0O, puis sépare la dicyclohexylurée précipitée. Le solvant est distillé sous vide et le résidu est dissous dans 5 cm3 d'alcool et 50 cm3 d'éther.
La solution obtenue est lavée par 10 cm3 d'alcool à 15"/o légèrement chlorhydrique, par 10 cm3 d'alcool à 15 /o, par 10 cm3 d'alcool à 15 /o légèrement ammoniacal, on termine par quatre lavages avec 10 cm3 d'alcool à 15 lo à chaque fois. La solution éthérée est séchée sur sulfate de sodium puis concentrée. Le résidu est repris par 20 cm3 d'acétate d'éthyle, on lave à l'eau, sèche et concentre à sec. Le résidu est cris tallisé dans l'alcool-cyclohexane, rendement : 82 o/o.
On recristallise dans l'alcool-éther de pétrole. F. vers 160¯ C, [a] 20 = - 71¯ ¯ 1 (c = 1%, chloroforme).
Le produit est insoluble dans l'eau, peu soluble dans l'alcool, soluble dans l'éther, l'acétone et le chloroforme.

E. Préparation du
N-trityl-L-asparaginyl-glycinate de diethylamine
[formule I : n = 1, R'=-NH2, R"= HN-CH2-CO2H, NH (C2H5) 2]
On hydrogène 1 g de N-trityl-L-asparaginyl-glycinate de benzyle obtenu selon D. en présence de 0, 5 g de noir palladié dans 20 cm3 d'alcool à 95 O/o contenant 0, 3 cm3 de diéthylamine. Après traitement usuel, on isole par reprise à l'éther 0, 86 g (90 /o) de N-trityl-L-asparaginyl-glycinate de diéthylamine qu'on recristallise dans l'alcool-acétate d'éthyle, F.
174-176 C, [a] 20/D = - 48¯ ¯ 1 (c = 1 ouzo alcool absolu). Le produit est insoluble dans l'éther et le benzène, peu soluble dans l'acétone, soluble dans l'eau, l'alcool et le chloroforme.

F. Preparation de la L-asparaginyl-glycine
[formule II : n = 1, Rl =-NH2, R2 =-NH-CH2-CO2H]
On chauffe cinq minutes à 50O C 0, 6 g de Ntrityl-L-asparaginyl-glycinate de diéthylamine, obtenu selon E., dans 1 cm3 d'eau et 1 cm3 d'acide acétique.
On dilue par 5 cm3 d'eau et sépare le triphénylcar- binol formé. Le filtrat est concentré sous vide jusqu'à début de cristallisation. Par addition de 10 cm3 d'alcool chaud et refroidissement, on provoque la séparation de 0, 19g (85 /o) de L-asparaginyl-glycine, [α]20/D = + 55¯ ¯ 1 (c = 1 %, eau). Miller et
Waelsch (Archiv. biochem. biophysics, 1952, 35, 176) indiquent [a] 20/D = + 53¯ (c = 4,6 %, dans l'eau). Les cristaux sont insolubles dans la plupart des solvants organiques, solubles dans l'eau.

On condense 2, 25 g de N-trityl-L-asparaginate de dióthylamine, obtenus selon l'exemple 1C, et 1, 3 g de chlorhydrate de L-leucinate de benzyle en présence d'un excès de 0, 925 g de N-trityl-L-aspara gine, obtenue selon l'exemple 1B, dans 50cm3 de chlorure de méthylène à-10 avec 1, 6 g de dicyclohexylcarbodiimide.
Après traitement identique aux précédents, le N-trityl-L-asparaginyl-L-leucinate de benzyle brut obtenu est hydrogénolysé dans 50 cm3 d'alcool absolu en présence de 0, 75 g de noir palladié et 0, 55 cm3 de diéthylamine. Après filtration et évaporation de l'alcool, on dissout le résidu dans 50 cm3 d'eau et on ajoute une solution de 1, 1 g de chlorhydrate de benzhydrylamine dans 10 cm3 d'eau chaude. On obtient 85 % de sels de benzhydrylamine que l'on recristallise dans le méthanol.
Le produit se présente sous forme de feuillets solvatés,
F. vers 120 C, [a] 2D0 =-18 1 (c = 2 /o, chloroforme), insolubles dans 1'eau et l'éther, peu solubles dans le benzène, solubles dans l'alcool et le chloroforme.

On libère la N-trityl-L-asparaginyl-L-leucine de 1, 8 g de son sel de benzhydrylamine obtenu ci-dessus par lavage de la solution en chlorure de méthylène par l'acide chlorhydrique dilué. Après séchage et évaporation de la phase organique, on reprend le résidu par 2 cm3 d'acide acétique aqueux à 50 ouzo et agite dix minutes a 40 C. On ajoute de l'acétone et sépare 73 zozo de L-asparaginyl-L-leucine.
On recristallise en alcool dilué [a] 2D0 =-10 ¯ 1 (c = 1 /o, eau). Miller, Weidle et Waelsch (Archiv. biochem. biophysics, 1955, 56, 11) indiquent [α] 20/D= -9, 80 (c = 5 /o, eau). Le produit est insoluble dans les solvants organiques usuels, soluble dans l'eau.

On opère comme dans le cas du N-trityl-L-asparaginyl-glycinate de benzyle à partir de 10 g de chlorhydrate de S-trityl-L-cystéinyl-L-prolyl-L- leucyl-glycinate de méthyle obtenus selon Velluz et coll. (Bull. Soc.
Chim., 1956, p. 1464), 6, 8 g de
N-trityl-L-asparaginate de diéthylamine, obtenus selon l'exemple 1C, 2, 85 g de N-trityl-L-asparagine, obtenus selon l'exemple 1B, et 4, 8 g de dicyclohexylcarbodiimide dans 100 cm de chlorure de méthyl- ène.
Le tétrapeptide-ester qui n'a pas réagi est Úliminé sous forme de chlorhydrate par des lavages acides et la N-trityl-L-asparagine par des lavages alcalins comme indiqué précédemment.
Le résidu de la distillation de la solution éthéro- alcoolique est repris au chlorure de méthylène. On lave à l'eau, sèche, distille le solvant et reprend à l'Úther. On obtient 14, 1 g (96 %), [α]20/D = - 77¯ ¯ 2 (c = 1 /o, chloroforme) de produit insoluble dans 1'eau et l'éther, peu soluble dans le benzène, soluble dans l'alcool, l'acétone et le chloroforme.

L-prolyl-L-leucyl-glycinate de mÚthyle]
On traite 13, 9 g de N, S-ditrityl-pentapeptide obtenu selon A par 14 cm3 d'acide acétique et 14 cm2 d'acide chlorhydrique N à 400 C pendant dix minutes. On dilue par 140 cm3 d'eau et sépare le triphénylcarbinol précipité. Au filtrat on ajoute 20 g de chlorure d'ammonium et extrait au chlorure de méthylène.
Le produit brut est lavé à l'éther et purifié par dissolution dans 10cm3 de chlorure de méthylène et précipitation par 100cmss d'éther. On obtient 10, 6 g (96 /o), [a] 2D0 =-230 2 (c = 1 /o, chloroforme), de produit insoluble dans l'éther et le benzène, soluble dans l'eau, l'alcool, l'acétone et le chloroforme.
Ce produit est nouveau.
Exemple 4
A. Préparation du
N-trityl-L-glutaminate de diethylamine
[formule I :
n = 2, R"= OH, NH(C2H5)2, R' = NH2]
On abandonne sous pression pendant soixantecinq heures, à température ambiante, une solution préalablement saturée à-10 par un courant d'ammoniac, de 60 g de N-trityl-L-glutamate de y-mono- méthyle préparé selon Amiard et coll. (Bull. Soc.
Chim., 1956, p. 97), dans 200 cm3 d'ammoniaque et 110 cm3 de méthanol. On chasse l'ammoniaque et le méthanol sous vide à température inférieure à 400 C. Le résidu huileux est dissous dans 200 cm3 de chloroforme et agité à 00 avec 13 cm3 d'acide
chlorhydrique concentré puis 150 cm3 d'acide chlor
hydrique N. La solution chloroformique est ensuite
lavée à l'eau, séchée et concentrée à température
inférieure à 300 C. Le résidu est dissous dans
200 cm3 d'acétate d'éthyle. L'addition de 15 cm8
de diéthylamine provoque la séparation de 67 g
(89 : 0/o) de sel de diéthylamine de la N-trityl-L-glutamine. On recristallise en acétate d'éthyle.
On obtient des aiguilles solvates, F. 112-114¯ C, [α]20/D =
+ 12" 2 (c = 2 /o, chloroforme), insolubles dans l'éther, peu solubles dans l'alcool froid, solubles dans l'eau, l'alcool chaud, le chloroforme, l'acétate d'éthyle et le benzène.

B. Préparation de la N-trityl-L-glutaminyl-glycine
[formule I :
n = 2, R'=-NH2, R" = - NH-CH2-CO2H]
On condense 1, 95 g de N-trityl-L-glutamine et 2, 4 g de N-trityl-L-glutaminate de diéthylamine préparé selon A avec 1 g de chlorhydrate du glycinate de benzyle Ó - 10¯, en quinze heures, dans 30 cm3 de chlorure de méthylène par 2, 2g de dicyclohexylcarbodiimide. Après traitement habituel, le produit brut est purifié comme précédemment par lavages de sa solution éthéro-alcoolique par l'alcool à 15 % d'abord acidifié à l'acide chlorhydrique puis alcalinisé à l'ammoniaque.
L'hydrogénolyse de la fonction ester benzylique est effectuée sur le produit brut dans 25 cm3 d'alcool à 95 /0 en présence de noir palladié et de 0, 5 cm3 de diéthylamine. Après traitement usuel, le résidu est dissous dans 40 cm3 d'eau et lavé à l'éther pour éliminer les fractions neutres. La solution aqueuse est acidifiée à l'acide chlorhydrique en présence de chlorure de méthylène. Celui-ci, séparé, laisse cristalliser 1, 6 g (70 O/o par rapport au glycinate de benzyle) de N-trityl-L-glutaminyl-glycine. On recristallise dans l'alcool-chlorure de méthylène-éther de pétrole.
On obtient des aiguilles incolores, F. vers 1300 C, [a] 20/D = + 25¯ ¯ 1 (c = 1 /o, alcool absolu), insolubles dans 1'eau et l'éther, peu solubles dans le chloroforme, l'acétone et le benzène, solubles dans l'alcool.

C. Préparation de la L-glutaminyl-glycine

On détrityle en dix minutes, à 35¯, 0, 3 g de la N-trityl-L-glutaminyl-glycine obtenue ci-dessus dans 0, 6 cm3 d'acide acétique aqueux à 50 /o. Par addition de 10 cm3 d'alcool puis recristallisation dans l'alcool aqueux, on obtient 0, 11 g (80 /o) d'aiguilles, [a] 21 = + 810 2 (c = 1'0/o, eau), insolubles dans l'alcool, l'éther, l'acétone, le benzène et le chloroforme, solubles dans l'eau.
Melville (Biochem. J., 1935, 29, 179) indique [a] 20 + 760 et Miller et
Waelsch (Archiv. biochem. biophysics, 1952, 35, 176) : [a] 20/D = + 77¯.

On condense comme précédemment 2 g de N trityl-L-glutamine et 2, 4 g de N-trityl-L-glutaminate de diéthylamine avec 1, 3 g de chlorhydrate de Lleucinate de benzyle à-10 dans 30 cm3 de chlorure de méthylène par 2, 2 g de dicyclohexylcarbodiimide.
Après traitement habituel, le produit brut est hydro génolysé dans 25 cm3 d'alcool a 95'0/o avec 0, 5 cm3 de diéthylamine en présence de noir palladié. Après filtration et évaporation de l'alcool, le résidu est dissous dans l'eau. On déplace la N-trityl-L-glutaminyl-
L-leucine de son sel de diéthylamine par l'acide chlorhydrique dilué et 1'extrait au chlorure de méthylène. Après purification dans l'éther-éther de pétrole, on obtient 1, 75 g (70 /o).
On détrityle 1 g du produit précédent dans 2 cm3 d'acide acétique aqueux à 50'0/o, à 40 , en dix minutes. On précipite le peptide par l'alcool absolu.
On obtient 0, 3 g (70 /o). On purifie par dissolution dans la soude hydroalcoolique puis acidification ace- tique. On obtient des feuilles brillantes [a] 2D0= + 4, 5 1 (c == 2 /o, acide chlorhydrique N).
Sondheimer et Holley (J. Am. Chem. Soc., 1954, 76, 2816) indiquent [a] 22/D = + 11,5¯ (c = 1 %, acide chlorhydrique N). Le produit est insoluble dans l'alcool, l'éther, l'acétone, le benzène et le chloroforme, peu soluble dans l'eau, soluble dans les alcalis et acides dilués aqueux.

On fait passer un rapide courant d'acide chlorhydrique gazeux dans une suspension de 14 g de
N-trityl-L-asparaginate de benzyle dans 42 cm3 de chloroforme. Il y a dissolution puis reprecipitation.
Après cinq minutes à température ambiante, on ajoute 150 cm3 d'éther et sépare le précipité. On recristallise en méthanol-acétate d'éthyle. Rendement : 92 /o, F. 125 C, [a] 2D5 = 0" 1 (c = 2 O/o, eau). Le produit est insoluble dans l'éther, peu soluble dans l'acétone, le benzène et le chloroforme, soluble dans 1'eau et l'alcool.

On hydrogénolyse 1 g de N-trityl-L-glutaminyl
L-asparaginate de benzyle obtenu selon B dans 20 cm3 d'alcool à 95"/e en présence de 0, 5 g de noir palladié et 0, 2 ce de diéthylamine. Après séparation du catalyseur et évaporation du solvant, on déplace la N-trityl-L-glutaminyl-L-asparagine de son sel de diéthylamine par reprise à l'eau, acidification acétique et extraction au chlorure de méthylène.
Le produit brut est détritylé en dix minutes à 35"par 2 ce d'acide acétique aqueux à 50 /o. L'addition d'acétone permet de séparer le dipeptide, rendement : 60 /o. On recristallise dans l'alcool dilué, [a] 25/D = + 24¯ ¯ 2 (c = 1%, eau), Swan et du

L-cystÚinyl-L-prolyl-L-leucyl-glycinate de mÚthyle]
On condense 10, 6 g de chlorhydrate du S-tritylpentapeptide-ester décrit à l'exemple 3B, avec 6 g de N-trityl-L-glutaminate de diéthylamine et 5, 1 g de N-trityl-glutamine par 5, 3 g de dicyclohexylcarbodiimide dans 100cm3 de chlorure de méthylène en quarante-cinq heures à-10 .
Après traitement habituel, le produit brut est fractionné comme dans les exemples précédents par lavages de sa solution éthéro-alcoolique à l'alcool à 15 O/o d'abord acidifié à l'acide chlorhydrique, puis alcalinisé à l'ammo- niaque. On obtient finalement 12, 1 g de substance qu'on purifie dans le méthanol-éther 10, 8 g (72 /o), [a] 20/D = - 35¯ ¯ 2 (c = 1"/o, chloroforme). On peut récupérer 10 /o du chlorhydrate du pentapeptide initial des solutions hydroalcooliques de lavage acide. ce qui porte le rendement à 82 /o.
Le produit est insoluble dans l'eau et l'éther, peu soluble dans le benzène, soluble dans l'alcool, l'acétone et le chloroforme.

N-trityl-L-isoglutaminyl-glycinate de benzyle

On effectue la condensation en quinze heures, à -I Ol, dans 20 cm3 de chlorure de méthylène de 3, 9 g de N-trityl-L-isoglutamine avec 1 g de chlorhydrate de glycinate de benzyle en présence dle 0, 5 cm3 de diéthylamine et 2, 2 g de dicyclohexylcarbodiimide. Après traitement habituel et lavages à l'alcool à 15 /o d'abord alcalinisé à la soude, puis acidifie à l'acide chlorhydrique, on obtient dans l'éther, 1, 95 g (72 O/o par rapport au glycinate de benzyle).
On recristallise dans le chlorure de méthyl- ène-éther. On obtient des aiguilles incolores, F. vers 160¯, [a] = + 27¯ ¯ 1 (c = 1'"/o, chloroforme), insolubles dans l'eau, peu solubles dans l'éther et le benzène, solubles dans l'alcool, l'acétone et le chlo roforme.

On hydrogénolyse 1 g de N-trityl-L-isoglutami- nyl-glycinate de benzyle dans 20 cm3 d'alcool à 90 ouzo contenant 0, 2 cm3 de diéthylamine en présence de 0, 5 g de noir palladié. Après filtration et évaporation de l'alcool, le résidu est repris par 10 cm3 de chloroforme et 2 cm3 d'acide chlorhydrique normal.
On obtient 0, 8 g (95 ID/o) de produit légèrement solvaté après séchage à 800 sous vide, F. 175 C, [α]20/D = + 55¯ ¯ 1 (c = 1 /o, alcool absolu). Le produit est insoluble dans 1'eau et l'éther, peu soluble dans l'acétone, le benzène et le chloroforme, soluble dans l'alcool.

On agite quinze minutes à 30O dans 1 cm3 d'acide acétique aqueux à 50 % et 0, 5 cm3 d'acétone 0, 5 g de N-trityl-L-isoglutaminyl-glycine. On ajoute de l'acétone, sépare le précipité et le lave à l'acétone.
On obtient 0, 2g (90 /o) de peptide qu'on recristallise dans l'alcool dilué, [α]20/D = + 17¯ ¯ 2 (c = 1 /o, eau). Ce produit renferme une faible proportion de glutamyl-glycine. Il est insoluble dans s l'alcool, l'éther, l'acétone, le benzène et le chloro- forme, soluble dans 1'eau et les acides et alcalis dilués aqueux.

On condense en quinze heures à-10 dans 20 cm3 de chlorure de méthylène 3, 9 g de N-trityl
L-isoglutamine avec 1, 3-g de chlorhydrate de Lleucinate de benzyle en présence de 0, 5 cm3 de di éthylamine et 2, 2 g de dicyclohexylcarbodiimide.
Après traitement identique à celui de l'exemple précédent, le produit brut est hydrogénolysé dans 20 cm3 d'alcool à 95% avec 0, 5 cm3 de diéthyl- amine et 1 g de noir palladié. Après séparation du catalyseur, on évapore à sec sous vide, reprend par 100 cm3 d'eau, acidifie à pH 3-4 par l'acide acéti- que et extrait au chlorure de méthylène.
Par reprise à l'éther de pétrole, on sépare 2 g (80 /o) de N trityl-L-isog, lutaminyl-L-leucine amorphe qu'on dé- trityle en quarante-cinq minutes à 40-50¯ dans 4 cm3 d'acide acétique à 50 /o. Par addition de 200 cm3 d'acétone, on sépare 0, 35 g (34 O/o) de L isoglutaminyl-L-leucine. On recristallise dans 1'eauacétone ; F vers 180 C, [a] 2Dw = + 20 + 1 (c= 2 /o, alcool à 50 fl/o). Le produit est insoluble dans l'al- cool, l'éther, l'acétone et le benzène, soluble dans l'eau, les acides et alcalis dilués aqueux.

Process for preparing peptides
In its Swiss patent No. 338197 the patentee described a process for obtaining peptides, via the corresponding N-trityl-peptides, said process consisting of a transformation of an Ntrityl-amino acid into its mixed anhydride which Reacting with the ester of an amino acid or peptide, the condensation product then being saponified to liberate the carboxyl group, then detritylated by heating with aqueous acetic acid.
In its Swiss patent No. 343407 the holder demonstrated that the condensation reaction of an N-trityl-amino acid on the ester of an amino acid is advantageously carried out if the operation is carried out in the presence of dicyclohexylcarbodiimide, the condensation agent recommended in peptide synthesis by Sheehan and Hess (J. Am.
Chem. Soc., 1955, 77, 1066) for other amino acid derivatives, the ester of the N-trityl peptide thus obtained being then saponified and then detritylated with hot aqueous acetic acid.
Now, it has now been found that the condensation of dibasic N-trityl-amino acids comprising a primary amide function with esters of amino acids or peptides surprisingly leads to good yields if it is carried out at low temperature in the presence of dicyclohexylcarbodiimide.
Such a reaction, carried out at ordinary temperature and, even more so, at high temperature, results in lower yields due to side reactions of the primary amide function.
The subject of the present invention is a process for the preparation of amidated peptides of general formula:

The importance of the physiological role of monoamid dibasic amino acids such as glutamine, isoglutamine and asparagine, has suggested the study of the behavior of their peptides (cf. H. K. Miller et al.
H. Welsch, Archiv. biochem. biophysics, 1952, 35, 176). The excellent use of asparaginyl peptides in certain experiments on the growth of microorganisms is particularly useful.
So, Miller,
Neidle and Waelsch (Archiv. Biochem. Biophysics, 1955, 56, 11) studied the action of asparaginyl leucine on cultures of Leuconostoc Mesenteroides and Miller and Waelsch (Archiv. Biochem. Biophysics, 1952, 35, 184), that glutaminyl-glycine and asparaginyl-glycine on Lactobacillus Arabinosus and Leuconostoc Mesenteroides.
Post-pituitary hormones: oxytocin and vasopressins, which are of considerable biological importance, are peptides comprising asparagin and glutamine (cf. J. M. Swan and V. du
Vigneaud, J. Am. Chem. Soc., 1954, 76, 3110).
In a preferred embodiment of the process of the invention, the peptide condensation reaction is carried out at a temperature of between -10 and 00 ° C., the solvent being methylene chloride.
The following examples illustrate the invention.
The preparation of some raw materials such as N-trityl-glutamine and N-trityl-asparagine is given here as an indication.
The melting points are instantaneous melting points and were determined on a Maquenne block.
Example 1
We start from N-trityl-L-asparagine, which was obtained as follows:
A. Preparation of trityl N-trityl-L-asparaginate

To a solution of 1.5 g of Lasparagine monohydrate in 25 cm3 of water and 7 cm3 of triethylamine, 12 g of trityl chloride dissolved in 70 cm3 of ether are added. Stirred three and a half hours at 00 C then added dilute hydrochloric acid until slight acidity. The precipitate is separated, washed with water and then with Úther. Recrystallized from benzene-ether.
Yield: 83 O / o; Mp 218-222 C; [a] 2D0 = -64

Benzyl N-trityl-L-asparaginate

Has a boiling solution of 33.3 g of N-trityl
Dibenzyl L-aspartate (obtained according to Velluz et al., Bull. Soc. Chim., 1956, p. 1464) in 180 cnr of dioxane and 30 cm3 of water, one adds little by little, in thirty minutes, 66 cm3 of normal potash.
Fifteen minutes after the introduction, the dioxane is removed under vacuum, diluted with water, acidified with 70 cm3 of N hydrochloric acid and extracted with methylene chloride. The organic solution is washed with water, dried over sodium sulfate and concentrated to 100 cm3. A stream of ammonia is sent to it until slight alkalinity (pH 8-9), 16 g of di cyclohexylcarbodiimide are added and the pH is maintained at around 8-9 by a slight stream of ammonia. After two hours, the precipitated dicyclohexylurea is separated (12.4 g, yield: 71%) and the solvent is removed in vacuo at 300C. The residue, taken up in methanol, gives 12 g (43%) of benzyl N-trityl-L-asparaginate. Recrystallized from absolute alcohol.
Mp 166-167o C, [a] 2D0 = -50 ¯ 1 (c = 2l / o, chloroform). The product is insoluble in water, sparingly soluble in alcohol and Úther, soluble in chloroform.

15 g of trityl N-trityl-L-asparaginate obtained according to A., in 150 cm3 of absolute alcohol and 3 cm3 of diethylamine, are hydrogenated at room temperature and at atmospheric pressure in the presence of palladium black. After separation of the catalyst by filtration, the solution is concentrated to 10 cm3, 0.5 cm3 of diethylamine is added and then precipitated with 200 cm3 of ether.
9. 85 g (90%) of diethylamine salt of N-trityl-L-asparagine, mp 152-155 C, [a] 20 290 2 (c = 1 O / o, chloroform) are collected. . Zervas and Theodoropoulos (J. Am. Chem. Soc., 1956, 78, 1359) indicate F. 150-151 C. The product is insoluble in ether, sparingly soluble in acetone, soluble in water. alcohol, chloroform and hot benzene.
D. Preparation of
Benzyl N-trityl-L-asparaginyl-glycinate

Diethylamine N-trityl-L-asparaginate, obtained according to
C., 1.01 g of benzyl glycinate hydrochloride and 0.925 g of N-trityl-L-asparagine, obtained according to
B., in 40 cm3 of methylene chloride and add 1.6 g of dicyclohexylcarbodiimide, the reaction medium being sufficiently basic for the amine function to be released. Stirred for five hours while allowing the temperature to rise slowly to 0O, then separating the precipitated dicyclohexylurea. The solvent is distilled off in vacuo and the residue is dissolved in 5 cm3 of alcohol and 50 cm3 of ether.
