Procédé de préparation de peptides Dans son brevet No 338197, la société titulaire a décrit un procédé d'obtention de peptides, par l'intermédiaire des N-trityl-peptides correspondants, ledit procédé consistant en une transformation d'un N-tri- tyl-amino-acide en son anhydride mixte que l'on fait réagir avec l'ester d'un' aminoacide ou peptide, le produit de condensation étant ensuite saponifié pour libérer le carboxyle, puis détritylé par chauffage avec l'acide acétique aqueux.
Ce procédé ne s'applique toutefois pas à certains N-trityl-amino-acides comme la N-trityl-méthionine, la N,N'-ditrityl-histidine et le N-trityl-tryptophane que l'on n'arrive pas à condenser avec des esters d'amino-acides ou peptides par la méthode mettant en oeuvre les anhydrides mixtes.
On a maintenant trouvé que l'on parvient facilement aux peptides correspondant à ces trois amino- acides N-tritylés ainsi que d'ailleurs à ceux des autres aminoacides N-tritylés, en faisant réagir un N-trityl- amino-acide en présence de dicyclohexylcarbodiimide sur un ester d'amino-acide ou de peptide.
La présente invention a pour objet un procédé de préparation de peptides, caractérisé en ce qu'on fait réagir un N-trityl-amino-acide en présence de di- cyclohexylcarbodiimide sur un ester d'amino-acide ou de peptide à fonction amine non salifiée, on élimine la dicyclohexylurée formée, on saponifie l'ester du trityl-peptide obtenu, on isole le N-trityl-peptide libre par acidification et on le détrityle par chauffage avec l'acide acétique aqueux.
Le dicyclohexylcarbodiimide a déjà été appliqué à la condensation des dérivés N-phtaloylés ou N-benzyloxycarbonylés des aminoacides avec des aminoesters en vue de l'obtention des peptides correspondants, mais on ne pouvait pas prévoir qu'il fournirait d'excellents résultats avec des N-trityl- amino-acides ne donnant pas de peptides correspondants par la méthode mettant en oeuvre des anhydrides mixtes.
Le procédé selon l'invention présente un intérêt particulier pour la préparation des peptides de l'histidine et de la méthionine dont on ne connaît que très peu de représentants. Pour l'accès aux peptides du tryptophane, il constitue une méthode de choix au point de vue économique, le chlorure de trityle ayant servi à préparer le N-trityl-tryptophane pouvant être récupéré avec un rendement pratiquement quantitatif, sous forme de triphénylcarbinol grâce à la détritylation.
Pour la mise en pratique du procédé selon l'invention, on opère avantageusement comme suit: on dissout un ester d'amino-acide ou de peptide à fonction amine non salifiée dans un solvant tel que le chlorure dé méthylène ou le chloroforme, ajoute ensuite une solution de dicyclohexylcarbodiimide dans le même solvant et introduit finalement l'amino- acide N-tritylé. Après avoir abandonné une nuit ou même plus longtemps,
on constate une précipitation presque quantitative de dicyclohexylurée que l'on complète par addition d'un peu d'acide acétique destiné à transformer l'excès de dicyclohexylcarbo- diimide en urée correspondante. Après filtration de la dicyclohexylurée, la solution renfermant l'ester du trityl-peptide est évaporée à sec et le résidu fournit après saponification et détritylation par l'acide acétique aqueux chaud le peptide cherché.
Les exemples suivants illustrent l'invention. Les points de fusion sont des points de fusion instantanée déterminés sur bloc de Maquenne.

Exemple 1 Préparation de la DL-tnéthioi2yl-glycine

a) Glycinate d'éthyle 3 g de chlorhydrate de glycinate d'éthyle obtenu par estérification à l'alcool chlorhydrique de la glycine sont mis en suspension dans 6 ce de chloroforme et on ajoute 2 ce de diéthyl-amine. Il y a dissolution. On ajoute 10 ce d'éther pour précipiter le chlorhydrate de diéthyl-amine et abandonne un quart d'heure.
On filtre le chlorhydrate de diéthyl- amine et évapore à sec la solution éthérochloro- formique de glycinate d'éthyle. b) Condensation de la N-trityl-DL-méthiotiirie avec le glycinate d'éthyle en présence de dicyclohexyl- carbodiimide On dissout 1,1 g de glycinate d'éthyle obtenu comme indiqué sous a) dans 3 ce de chlorure de méthylène et y ajoute la solution de 2,5 g de dicyclo- hexylcarbodiimide dans 5 cm-3 de chlorure de méthylène.
Après avoir refroidi à 0 C, on introduit 3,9 de N-trityl-DL-méthionine dont la préparation a été décrite par G. Amiard, R. Heymes et L. Velluz (Bull. Soc. Chim. No 2 (1955) 191) et abandonne une nuit à la température ambiante. Le lendemain, on ajoute 0,5 cms d'acide acétique, abandonne quelques minutes et filtre la dicyclohexylurée qui a précipité. La solution filtrée est lavée à l'acide chlorhydrique normal, à l'eau, à l'ammoniaque, à l'eau jusqu'à neutralité, puis on la sèche sur sulfate de magnésium et évapore à sec.
Le résidu est constitué par du N-trityl-DL-méthionyl-glycinate d'éthyle brut renfermant un peu de chlorure de méthylène. Le rendement est quantitatif. c) Saponification du N-trityl-DL-méthionyl-glycinate d'éthyle Le produit brut obtenu selon b) est débarrassé du chlorure de méthylène qu'il renferme par reprise à l'alcool et distillation d'une partie de celui-ci. On ajoute à la solution alcoolique 8 ce de potasse méthanolique à 20% et 2 ce d'eau et fait bouillir 5 minutes. On dilue par l'eau, refroidit et acidifie par l'acide acétique.
Le trityl-peptide brut cherché se présente sous forme d'un précipité huileux qu'on purifie par reprise en chloroforme et lavage de la solution chloroformique à l'eau. Après séchage de la solution sur sulfate de sodium et évaporation du solvant, on obtient la N-trityl-DL-méthionyl-glycine sous forme d'huile épaisse.
d) Détritylation de la N-trityl-DL-méthiottyl-glycine Le N-trityl-peptide brut obtenu selon c) est repris par 10 ce d'acide acétique. On ajoute 10 ce d'eau et chauffe au bain-marie bouillant. Après quelques minutes de chauffage, le triphénylcarbinol cristallise, le peptide restant dissous. On arrête le chauffage, ajoute de l'eau, essore et lave le triphénylcarbinol à l'eau et concentre le filtrat à petit volume, passe au noir végétal et concentre jusqu'à cristallisation com- mençante. On introduit alors 20 ce d'alcool absolu, essore, lave à l'alcool et sèche.
On obtient 1,5 g de DL-méthionyl-glycine, F. (déc.) = 240-245o C, identique à la DL-méthionyl-glycine décrite dans la littérature. Le produit est soluble dans l'eau, les acides dilués aqueux, insoluble dans l'alcool, l'éther et l'acétone.
Exemple 2 Préparation de la DL-tryptophyl-glycine

La N-trityl- DL-tryptophyl-glycine précipite à l'état pulvérulent. d) Détritylation de la N-trityl-DL-tryptophyl-glycine On opère comme dans.l'exemple 1d) en utilisant 10 cm\; d'acide acétique à 50 %. Après concentra- tion du filtrat du triphénylcarbinol et addition d'al-

La N,N'- ditrityl-L-histidyl-glycine cristallise sous forme de gros prismes hydratés. Après lavage au toluène et séchage à 1100C, on obtient 5,6 g (soit un rendement de 80%) de N,N'-ditrityl-L-histidyl-glycine pure pour l'analyse, F. = 155-160C (avec effer- vescence), [a] ri = -I- 171, 1 (c = 2 %, chloro- forme). Le produit est soluble dans l'acétone et le chloroforme, peu soluble dans l'alcool, l'éther et les alcalis dilués aqueux et insoluble dans l'eau.

c) Saponification du N,N'-ditrityl-L-hi,stidyl- L-leucinate de méthyle On opère dans les conditions de l'exemple le) en utilisant pour la totalité de l'ester brut obtenu 10 ce de potasse méthanolique à 20 % et 2 ce d'eau ut obtient finalement, après acidification, 6,5 g (soit un rendement de 86 0/0) de N,N'-ditrityl-L- histidyl-L-leucine pulvérulente, laquelle, après reprise au chloroforme et lavage à l'eau, devient huileuse sans tendance à la cristallisation.

Process for the preparation of peptides In its patent No. 338197, the proprietor described a process for obtaining peptides, by means of the corresponding N-trityl-peptides, said process consisting of a transformation of an N-tri- tyl -amino acid to its mixed anhydride which is reacted with the ester of an amino acid or peptide, the condensation product then being saponified to liberate the carboxyl, then detritylated by heating with aqueous acetic acid.
However, this method does not apply to certain N-trityl-amino acids such as N-trityl-methionine, N, N'-ditrityl-histidine and N-trityl-tryptophan which cannot be obtained. condense with esters of amino acids or peptides by the method using mixed anhydrides.
It has now been found that peptides corresponding to these three N-trityl amino acids, as well as those of other N-trityl amino acids, are easily obtained by reacting an N-trityl amino acid in the presence of dicyclohexylcarbodiimide on an amino acid or peptide ester.
The present invention relates to a process for the preparation of peptides, characterized in that an N-trityl-amino-acid is reacted in the presence of di-cyclohexylcarbodiimide with an amino-acid ester or of a peptide with a non-amine function. When salified, the dicyclohexylurea formed is removed, the ester of the trityl-peptide obtained is saponified, the free N-trityl-peptide is isolated by acidification and it is detritylated by heating with aqueous acetic acid.
Dicyclohexylcarbodiimide has already been applied to the condensation of N-phthaloyl or N-benzyloxycarbonyl derivatives of amino acids with amino esters in order to obtain the corresponding peptides, but it could not be predicted that it would provide excellent results with N -trityl- amino acids not giving corresponding peptides by the method using mixed anhydrides.
The process according to the invention is of particular interest for the preparation of histidine and methionine peptides, of which very few representatives are known. For access to tryptophan peptides, it constitutes a method of choice from an economic point of view, the trityl chloride having been used to prepare N-trityl-tryptophan being able to be recovered with a practically quantitative yield, in the form of triphenylcarbinol thanks to detritylation.
For the practice of the process according to the invention, the procedure is advantageously as follows: an amino acid ester or peptide containing an unsalted amine function is dissolved in a solvent such as methylene chloride or chloroform, then added a solution of dicyclohexylcarbodiimide in the same solvent and finally introduces the N-trityl amino acid. After giving up overnight or even longer,
there is an almost quantitative precipitation of dicyclohexylurea which is completed by adding a little acetic acid intended to convert the excess of dicyclohexylcarbodiimide into the corresponding urea. After filtration of the dicyclohexylurea, the solution containing the ester of the trityl-peptide is evaporated to dryness and the residue provides, after saponification and detritylation with hot aqueous acetic acid, the desired peptide.

Example 1 Preparation of DL-tnethioi2yl-glycine

The crude trityl-peptide sought is in the form of an oily precipitate which is purified by taking up in chloroform and washing the chloroform solution with water. After drying the solution over sodium sulfate and evaporating the solvent, N-trityl-DL-methionyl-glycine is obtained in the form of a thick oil.
d) Detritylation of N-trityl-DL-methiottyl-glycine The crude N-trityl-peptide obtained according to c) is taken up in 10 cc of acetic acid. Add 10 cc of water and heat in a boiling water bath. After a few minutes of heating, the triphenylcarbinol crystallizes, the peptide remaining dissolved. The heating is stopped, water is added, the triphenylcarbinol is filtered off and washed with water and the filtrate is concentrated to a small volume, passes to vegetable black and concentrates until crystallization begins. 20 cc of absolute alcohol are then introduced, filtered off, washed with alcohol and dried.

c) Saponification of methyl N, N'-ditrityl-L-hi, stidyl-L-leucinate The operation is carried out under the conditions of example 1c) using all of the crude ester obtained 10 cc of methanolic potassium hydroxide at 20% and 2 cc of water ut finally obtains, after acidification, 6.5 g (i.e. a yield of 86%) of powdered N, N'-ditrityl-L-histidyl-L-leucine, which, after recovery with chloroform and washing with water, becomes oily without tendency to crystallize.
d) Detritylation of N, N'-ditrityl-L-histidyl-L-leucine The procedure is as in Example I d) using for 6 g of crude N, N'-ditrityl peptide, 20 cm, 'd' 50% aqueous acetic acid and finally obtains
