L'objet de la présente invention est un procédé de préparation d'a-glutamyl-peptides dérivant de l'acide glutamique de la formule suivante 1 :

On sait que différents polypeptides naturels renferment parmi l'enchaînement des peptides qui les constituent un ou plusieurs groupes glutamyl-peptide et le procédé selon l'invention fournit un moyen d'accès aux L-glutamyl-peptides de formule VII à partir desquels on peut édifier des chaînes polypepti- diques ou qu'on peut introduire dans la molécule à synthétiser. Certains de ces glutamyl-peptides peuvent également présenter un intérêt biologique ou industriel par eux-mêmes.
Dans son brevet suisse Nu 338197, la titulaire a décrit la préparation de N-trityl-aminoacides par réaction des esters des aminoacides correspondants employés à l'état libre ou de chlorhydrate de la fonc- tion amine, avec le chlorure de trityle en présence d'une base tertiaire. Si 1'ester est employé à l'état de chlorhydrate, on utilise un excès de base susceptible de le libérer extemporanément et de neutraliser en même temps l'acide chlorhydrique formé au cours de la réaction. La saponification de l'aminoester N-tri tylé ainsi formé conduit au N-trityl-aminoacide cherché.
En outre, le brevet susmentionné décrit un procédé d'obtention de N-peptides en transformant lesdits N-trityl-aminoacides en anhydrides mixtes selon les méthodes connues et en faisant réagir ces derniers avec un amino ester. La sponification de l'ester du N-trityl-peptide obtenu suivie d'une détrityla- tion par chauffage avec l'acide acétique aqueux conduit selon une caractéristique particulière du procédé aux peptides cherchés sans racémisation. Certains aminoacides, N-tritylés ne se prêtant pas à la transformation en peptides par la méthode aux anhydrides mixtes,
la titulaire a montré dans son brevet suisse ? 338197 qu'on peut les transformer aisément en N-trityl-peptides correspondants en les faisant réagir sur un aminoester en présence de dicyclohexyl- carbodiimide. La saponification et la détritylation à l'acide acétique aqueux fournissent le N-peptide cherché.
On peut accéder aux a-glutamyl-peptides en mettant à profit certaines propriétés particulières de dif férents diesters N-tritylés de l'acide L-glutamique III, ainsi que des monoesters de formule IV, ou de certains diesters mixtes de formule VIII où R'est différent de R.
Les diesters glutamiques III excellent par une stabilité très grande du groupe ester en a vis-à-vis des alcalis de sorte que leur monosaponification en y est facile à réaliser. On peut aboutir ainsi à l'a- monoester N-tritylé de formule IV.
Cette même stabilité peut être mise à profit, comme la titulaire l'a déjà montré dans le brevet suisse No 344714, pour préparer le N-trityl-glutamate de y-méthyle et d'abenzyle de formule VIII, où R'=CH2-C6H5 et R= CH3 et qui consiste à soumettre le N-tritylglutamate de dibenzyle de formule III
(R = R'=-CH2-C6H5 à une méthanolyse en présence d'un agent alcalin.
Par hydrogénolyse du N-trityl-glutamate de y-méfhyle et d'a-benzyle, on peut aboutir au N-trityl-glutamate de y-monométhyle de formule IX.
Le procédé suivant l'invention consiste à condenser un a-ou y-monoester glutamique N-tritylé (formules IV et IX) avec un ester d'aminoacide, à saponifier par des alcalis le diester du N-trityl glutamyl-peptide obtenu et à détrityler le N-trityl-a- glutamyl-peptide résultant.
Il est avantageux d'effectuer la condensation, en présence de dicyclohexylcarbodiimide. La détdtyla- tion est effectuée, de préférence, à l'aide d'acide acétique aqueux chaud.
On aurait pu s'attendre à ce que la condensation des a-monoesters de formule IV avec les esters d'aminoacides conduise aux y-peptides. En fait, l'application de la méthode du dicyclohexylcarbodiimide ne conduit pas aux y-peptides attendus. Il se forme cependant le diester du N-trityl-Ly-utamyl-peptide de formule V. Mais la fonction ester en a de ce composé est très labile tandis qu'elle est très stable dans le diester glutamique N-tritylé. En présence d'alcalis, le composé de formule V engendre vraisemblablement le composé intermédiaire de formule VI.
La détritylation de ce composé fournit essentiellement

<tb> l'a-glutamyl-peptide de formule VII, à côté d'un faible pourcentage de y-glutamyl-peptide.
Il est à remarquer qu'en partant aussi bien d'un y-monoester glutamique N-tritylé que d'un a-moester glutamique N-tritylé, on obtient des a-glu- tamyl-peptides avec d'excellents rendements et qu'il n'y a pas de racémisation. A fortiori, peut-on préparer selon les mêmes modes d'exécution les a- peptides de l'acide DL-glutamique et il est évident que le procédé est également applicable à l'acide Dglutamique.
Le schéma (page 2) explique les modalités d'ac cès aux différents composés illustrés par les exemples suivants. Les points de fusion sont des points de fusion instantanée déterminés sur bloc de Maquenne.
Exemple 1
Preparation de l'a-L-glutamyl-glycine a) Condensation du N-trityl-L-glutamate d'a-mono
éthyle avec le glycinate d'éthyle
On dissout 1, 1 g de glycinate d'éthyle libre (obtenu à partir de son chlorhydrate d'après le procédé décrit dans le brevet suisse No 338197) dans 3 cm3 de chlorure de méthylène et on ajoute la solution de 2, 5 g de, dicyclohexylcarbodiimide dans 5 cm3 de chlorure de méthylène. On refroidit à 0 et introduit 4, 17 g de N-trityl-L-glutamate d'a-monoéthyle.
On abandonne une nuit à la température ambiante, décompose l'excès de dicyclohexylcarbodiimide par addition de 0, 5 cm3 d'acide acétique, abandonne trente minutes, essore la dicyclohexylurée formée, lave la solution en ampoule avec 3 cm3 d'acide chlorhydrique normal, puis à l'eau, à l'ammoniaque diluée et à l'eau, sèche sur sulfate de ma gnésium, filtre et évapore à sec.
On obtient ainsi le diester diéthylique brut (formule V, R' et R" =-CBH,) qu'on utilise sans purification. b) Saponification du diester diéthylique brut de la N-trityl-{-glutamyl-glycine
Le diester brut obtenu selon a) est saponifié par cinq minutes de reflux avec 10 cm3 de potasse méthanolique à 20% et 2 cm3 d'eau. On dilue par 50 cm3 d'eau, essore un léger insoluble et acidifie par l'acide acétique. Après extraction au chloroforme et lavage à l'eau, la solution chloroformique est séchée sur sulfate de magnésium puis évaporée à sec.
A la suite de la transposition provoquée par la sapo nification, le produit obtenu est constitué essentiellement par la N-trityl-a-glutamyl-glycine. Le produit brut est chauffé cinq minutes au bain-marie avec 20 cm d'acide acétique à 50 /n. Le triphényl-car- binol libéré cristallise. On refroidit, ajoute 10 cm3 d'eau et essore le triphényl-carbinol formé. Le filtrat est concentré à faible volume. On ajoute de l'alcool absolu et laisse cristalliser.
Après glaçage et essorage, lavage à l'alcool et séchage, on obtient 1, 5 g (soit un rendement de 75%) d'a-glutamyl-glycine de formule VII (RI = H) que l'on recristallise dans l'al- cool à 50s /o. On obtient ainsi, après séchage, 1, 1 g d'a-glutamyl-glycine exempte de y-glutamyl-glycine (le produit ne donne pas de réaction à la ninhydrine),
F = 220 C (déc.), [oj = + 79"5 1 (c = 2 /o, eau). Le produit est soluble dans l'eau, les acides et alcalis dilués aqueux, insolubles dans l'alcool et l'Úther. La littérature indique un pouvoir rotatoire de + 800.

Le N-trityl-L-glutamate d'a-monoéthyle utilisé comme composé de départ dans cet exemple peut être obtenu de la manière suivante :
A une solution refroidie à 00 de 2, 4 g de chlor- hydrate de L-glutamate de diéthyle dans 25 cm3 de chloroforme et 3 cm3 de triéthylamine, on ajoute 2, 8 g de chlorure de trityle. On abandonne une heure à la température ordinaire.
La solution chlorofor- mique du produit réactionnel est lavée à 1'eau et séchée sur sulfate de magnésium. On chasse le solvant et reprend le produit brut huileux par 40 cm3 d'alcool. On neutralise la solution par quelques gout- tes de soude normale, porte au reflux et ajoute par fractions en vingt minutes environ, au fur et à mesure de la neutralisation, qu'on suit à la phénolphtaléine, 10 cm3 de soude normale.
A la fin de la réaction, la solution reste légèrement alcaline. On refroidit, acidifie à l'acide acétique, essore le précipité et le lave à l'eau. Après séchage, on obtient 3, 3 g de N-trityl-L-glutamate d'a-mono- éthyle, soit un rendement de 80 /0. Le produit gom- meux n'a pas pu être cristallisé. Il fournit par contre dans l'alcool à 50"/o, avec le D (-)-thréo-l-p-nitro- phényl-2-amino-propane-1, 3-diol un sel bien cristallisé en feuillets hydratés (environ une molécule d'eau de cristallisation).
F = 100¯ C (déc.), [α]20 D = + 34¯, 5 1 (c = 2 /0, méthanol). Le produit est jaunâtre, soluble en alcool et acétone, peu soluble en éther, très peu soluble dans l'eau, insoluble dans le benzène. Les acides et alcalis dilués le décomposent.

On arrive au même produit au départ d'autres a-monoesters de l'acide N-trityl-glutamique comme l'ester a-méthylique en suivant le même mode opératoire. Les radicaux esters de la glycine peuvent être différents de ceux de l'acide glutamique. Ainsi, à partir du N-trityl-glutamate d'a-méthyle et du glycinate d'éthyle, on obtient le diester mixte de formule
V, (R'= CH3, R"= CJHÏ) fournissant l'a-gluta- myl-glycine.
Le N-trityl-L-glutamate d'a-monométhyle peut être préparé de la manière suivante :
Ce composé est obtenu au départ du chlorhydrate du L-glutamate de diméthyle en opérant, pour la Ntritylation et la saponification, de la manière décrite ci-dessus. Le produit brut peut être cristallisé en cyclohexane. Le rendement en produit pur,
F = 140-141o C, [a] D = + 450 + 1 (c = 2 11/o, m6- thanol), est de 50 /0. I1 se présente en aiguilles incolores, solubles en acétone, chloroforme, éther et les alcalis dilués, peu solubles en alcool, insolubles dans 1'eau et les acides dilués. Ce produit est nou- veau.
Il fournit, avec le D (-)-thréo-l-p-nitro-phényl- 2-amino-propane-1, 3-diol, un sel hydraté,
F = 100-C (déc.), [a] = + 32 , 5 l (c = 2 /o, méthanol) qui est également nouveau.
Le N. trityl-L-glutamate d'-a-monométhyle, F = 140 C, a été soumis à l'analyse :

avec le glycinate d'ethyle
On opère comme dans l'exemple 1 a). b) Saponification du diester de la N-trityl-a-gluta
myl-glycine et dÚtritylation
On opère comme dans l'exemple 1 b) et obtient l'a-L-glutamyl-glycine avec un rendement de 55 /o.
Exemple 3
Préparation de la L-a-glutamyl-glycine a) Condensation du N-trityl-L-glutamate d'a-benzyle
avec le glycinate de benzyle
On prépare 3, 6 g de glycinate de benzyle libre en traitant une solution du chlorhydrate de glycinate de benzyle avec la quantité théorique de potasse 5 N, relargue au sel, extrait à l'éther et évapore. Le glycinat de benzyle ainsi obtenu est dissous dans 6 cm3 de chlorure de méthylène, on ajoute la solu- tion de 5 g de dicyclohexylcarbodiimide dans 10 cm3 de chlorure de méthylène, refroidit à 00 et introduit 9, 6 g de N-trityl-L-glutamate d'a-benzyle brut.
Après un traitement identique à celui de l'exemple 1, on obtient le diester dibenzylique brut de formule V (R'et R"=-CH -C6HÏ Rj = H) avec un rendement quantitatif. b) Saponification du diester dibenzylique brut de la
N-trityl-y-glutamyl-glycine et detritylation
On saponifie le produit obtenu selon a) par dix minutes d'ébullition avec la potasse méthanolique comme dans l'exemple précédent. Après distillation d'une partie du méthanol et dilution par l'eau, on
extrait, après acidification à l'acide acétique, au chloroforme.
Le produit constitué par un mélange de N-trityl-L-a-glutamyl-glycine et de N-trityl-L-y- glutamyl-glycine obtenu après évaporation du chloroforme est détritylé par chauffage avec l'acide aqueux à 50 /0. Après séparation du triphényl-car- binol, on concentre et fait cristalliser comme précé- demment par addition d'alcool. Après recristallisation en alcool aqueux, on obtient finalement, avec un rendement de 50 /o, l'a-glutamyl-glycine pure, identique au produit obtenu dans l'exemple 1.
Le N-trityl-L-glutamate d'a-benzyle utilisé comme composé de départ peut être obtenu de la manière suivante :
Ce composé est préparé au départ du N-trityl-Lglutamate d'a-benzyle et de y-méthyle décrit dans l'exemple 2 du brevet suisse No 344714.
A une solution bouillant à reflux de 9, 87 g de ce diester mixte dans 100 cm3 de méthanol, on ajoute petit à petit, en quinze à vingt minutes, environ 21 cm3 de soude normale et maintient l'ébullition encore quinze minutes. On concentre la solution de moitié, la dilue par 40 cm3 d'eau, passe, au noir végétal puis acidifie par l'acide chlorhydrique jusqu'à pH 2 à 3. Le produit précipite à l'état pulvérulent.
On l'essore, lave à 1'eau et sèche en exsiccateur. Le e
N-trityl-L-glutamate d'a-benzyle ainsi obtenu peut être utilisé directement pour la réaction suivante.
Exemple 4
Préparation de la L-a-glutamyl-L-leucine a) Condensation du N-trityl-glutamate de y-methyle avec le L-leucinate de methyle
On prépare 3, 2g de leucinate de méthyle libre à partir de son chlorhydrate selon la technique employée pour la préparation du glycinate d'éthyle citée dans l'exemple 1. Le produit est dissous dans 20 cm3 de chlorure de méthylène, on ajoute 5 g de dicyclohexylcarbodiimide, refroidit à 0 C, ajoute 8, 1 g de N-trityl-glutamate de y-méthyle dont la préparation est décrite dans le brevet suisse No 344714, agite pour dissoudre et abandonne 65 heures en glacière.
On ajoute alors 1 cm3 d'acide acétique, laisse trente minutes au repos, essore la dicyclohexylurée qu'on lave au chlorure de méthylène que l'on joint au filtrat. Ce filtrat est lavé à l'acide chlorhydrique dilué, à l'ammoniaque diluée puis à l'eau, filtre et évapore à sec sous vide. Le résidu huileux est repris par 25 cm3 de méthanol, on ajoute 4 cm3 d'eau, laisse cristalliser en glaçant, essore, lave au méthanol aqueux et sèche. Le produit directement utilisable pèse 6, 8 g.
Les eaux-mères abandonnent par cris tallisation prolongée encore 0, 6 g du même produit de formule X

soit un rendement de 70 /o. Pour l'analyse on le recristallise en méthanol aqueux
F = 121-133o C, [a32D = + 10o + 1 (c = 2 /0, méthanol). Le diester méthylique de la L-N-trityl-a- glutamyl-L-leucine se présente sous forme d'aiguilles solubles dans l'alcool et l'éther, les alcools aqueux, insolubles dans 1'eau et les alcalis dilués aqueux. Les acides dilués aqueux le décomposent.

Ce composé est nouveau. b) Saponification du diester mithylique de la L-N trityl-α-glutamyl-L-leucine et dÚtritylation
6, 3 g de diester brut préparés selon a) sont repris par 20 cm3 de méthanol.
On ajoute 10 cm3 de potasse méthanolique à 20 /o et 5 cm3 d'eau. On porte deux minutes au reflux et ajoute au liquide bouillant 25 cm3 d'eau, chasse le méthanol sous vide partiel, refroidit, acidifie à pH 2 par addition d'environ 35 cms d'acide chlorhydrique normal et extrait le trityl-peptide au chloroforme. La solution chloroformique est séchée sur sulfate de magnésium, filtrée et évaporée à sec sous vide. Le résidu est repris par 20 cm d'acide acétique aqueux à 50 ID/o. On fait bouillir une minute, ajoute 20 cm3 d'eau, filtre le triphényl-carbinol à chaud et le lave à 1'eau bouillante. Les solutions acétiques réunies sont concentrées à faible volume. Le peptide cristallise.
On ajoute de l'alcool absolu, glace, essore, sèche et obtient 2, 2 g de L-a-glutamyl-L-leucine, soit un rendement de 71 ID/o. Le produit se présente en feuillets incolores, F 2500 C. En tube capillaire il fond à 195¯ C environ. [a] 2D = + 10 + 1 (c = 2 /o, eau et une molécule d'acide chlorhydrique). Il est soluble dans les acides et alcalis aqueux dilués, peu soluble dans l'eau, insoluble dans l'alcool, l'éther, le benzène et l'acétone. Il ne donne pas de réaction à la ninhydrine. Le même composé a été obtenu par une autre voie par Le Quesne et Young (J. Chem.
Soc., 1950, p. 1954).
Exemple 5
Préparation de la L-a-glutamyl-L-tyrosine a) Condensation du N-trityl-L-glutamate de y-me
thyle avec le tyrosinate d'éthyle
Le tyrosinate d'éthyle libre est préparé par reprise du chlorhydrate dans l'ammoniaque 5 N en excès. Le produit cristallise. On essore, lave à l'eau et sèche en exsiccateur sulfurique.
En opérant comme dans l'exemple 1 a), on obtient au départ de 4, 3 g de tyrosinate d'éthyle, 20 cm3 de chlorure de méthy- lène et 8, 1 g de L-N-trityl-glutamate de y-monométhyle, 14 g de produit de condensation brut de formule X

On saponifie comme dans l'exemple 1 b) et en présence de 20 cm3 d'éther, acidifie le produit sapo nifié filtré par l'acide chlorhydrique jusqu'à pH 2.
Le composé se résout en une masse cristalline puis se délite peu à peu. Après empâtage à l'éther et essorage, on obtient 7, 8 g de N-trityl-a-glutamyl-peptide qu'on peut détrityler directement. Pour l'analyse, on recristallise en méthanol aqueux. Le produit se présente en aiguilles. [a] 25 D = + 19¯ ¯ 1 (c = 1 /0, méthanol à 75"/e + 1 NaOH). Il est soluble dans l'éther, le chloroforme et l'alcool, peu soluble dans les alcalis dilués aqueux, insoluble dans l'eau. Les acides dilués le décomposent.

Ce composé est nouveau.
En détritylant le produit brut par l'acide acétique à 50 < Vo on obtient après le traitement habituel la L-a-glutamyl-L-tyrosine avec un rendement de 75 %.
F (déc.) = 235¯ C [α] D = + 29¯ ¯ 1 (c = 2 ID/o, eau + 1 HC1). Le peptide se présente en aiguilles incolores. Il est soluble dans les acides et alcalis aqueux, très peu soluble dans l'eau, insoluble dans l'alcool, l'acétone, le benzène. Il est identique au produit décrit par Bergmann et Coll. (Ztschr. f. physiol. Chem., 1934, 224, 17).

The object of the present invention is a process for the preparation of α-glutamyl-peptides derived from glutamic acid of the following formula 1:

It is known that various natural polypeptides contain, among the chain of peptides which constitute them, one or more glutamyl-peptide groups and the method according to the invention provides a means of access to the L-glutamyl-peptides of formula VII from which it is possible build polypeptide chains or which can be introduced into the molecule to be synthesized. Some of these glutamyl peptides can also be of biological or industrial interest on their own.
In its Swiss patent Nu 338197, the patentee described the preparation of N-trityl-amino acids by reaction of the esters of the corresponding amino acids employed in the free state or of the hydrochloride of the amine function, with trityl chloride in the presence of 'a tertiary base. If the ester is used in the hydrochloride form, an excess of base is used which is capable of releasing it extemporaneously and at the same time neutralizing the hydrochloric acid formed during the reaction. Saponification of the N-tri tylated amino ester thus formed leads to the desired N-trityl-amino acid.
In addition, the aforementioned patent describes a process for obtaining N-peptides by transforming said N-trityl-amino acids into mixed anhydrides according to known methods and by reacting the latter with an amino ester. The sponification of the ester of the N-trityl-peptide obtained followed by a detritylation by heating with aqueous acetic acid leads, according to a particular characteristic of the process, to the peptides sought without racemization. Certain amino acids, N-tritylated, which do not lend themselves to transformation into peptides by the mixed anhydride method,
has the holder shown in his Swiss patent? 338197 that they can easily be converted into the corresponding N-trityl-peptides by reacting them with an amino ester in the presence of dicyclohexylcarbodiimide. Saponification and detritylation with aqueous acetic acid provides the desired N-peptide.
The α-glutamyl-peptides can be accessed by taking advantage of certain particular properties of different N-tritylated diesters of L-glutamic acid III, as well as monoesters of formula IV, or of certain mixed diesters of formula VIII where R 'is different from R.
Glutamic III diesters excel with a very great stability of the ester group in a vis-à-vis alkalis so that their monosaponification is easy to achieve therein. This gives the N-trityl alpha-ester of formula IV.
This same stability can be taken advantage of, as the proprietor has already shown in Swiss Patent No. 344714, to prepare γ-methyl-abenzyl N-trityl-glutamate of formula VIII, where R '= CH2- C6H5 and R = CH3 and which consists in subjecting the dibenzyl N-tritylglutamate of formula III
(R = R '= - CH2-C6H5 to methanolysis in the presence of an alkaline agent.
By hydrogenolysis of γ-methyl and α-benzyl N-trityl-glutamate, the γ-monomethyl N-trityl-glutamate of formula IX can be obtained.
The process according to the invention consists in condensing an N-trityl α- or γ-glutamic monoester (formulas IV and IX) with an amino acid ester, in saponifying the N-trityl glutamyl-peptide diester obtained with alkalis and in detritylate the resulting N-trityl-α-glutamyl-peptide.
It is advantageous to carry out the condensation in the presence of dicyclohexylcarbodiimide. The detoxification is preferably carried out using hot aqueous acetic acid.
One would have expected that the condensation of the α-monoesters of formula IV with the amino acid esters would lead to the γ-peptides. In fact, the application of the dicyclohexylcarbodiimide method does not lead to the expected γ-peptides. However, the diester of N-trityl-Ly-utamyl-peptide of formula V is formed. However, the ester function at a of this compound is very labile while it is very stable in the glutamic N-trityl diester. In the presence of alkalis, the compound of formula V presumably generates the intermediate compound of formula VI.

It should be noted that starting from both an N-tritylated glutamic γ-monoester and an N-tritylated glutamic α-moester, α-glutamyl-peptides are obtained in excellent yields and that there is no racemization. A fortiori, can the β-peptides of DL-glutamic acid be prepared according to the same methods and it is obvious that the method is also applicable to Dglutamic acid.
The diagram (page 2) explains the modalities of access to the various compounds illustrated by the following examples. The melting points are instantaneous melting points determined on a Maquenne block.
Example 1
Preparation of α-L-glutamyl-glycine a) Condensation of α-mono N-trityl-L-glutamate
ethyl with ethyl glycinate
1.1 g of free ethyl glycinate (obtained from its hydrochloride according to the process described in Swiss Patent No. 338197) is dissolved in 3 cm3 of methylene chloride and the solution of 2.5 g of , dicyclohexylcarbodiimide in 5 cm3 of methylene chloride. Cooled to 0 and introduced 4.17 g of α-monoethyl N-trityl-L-glutamate.
It is left overnight at room temperature, the excess of dicyclohexylcarbodiimide is decomposed by adding 0.5 cm3 of acetic acid, left for thirty minutes, the dicyclohexylurea formed is filtered off, the solution in an ampoule is washed with 3 cm3 of normal hydrochloric acid, then with water, diluted ammonia and water, dried over magnesium sulfate, filtered and evaporated to dryness.
The crude diethyl diester (formula V, R 'and R "= -CBH,) is thus obtained, which is used without purification. B) Saponification of the crude diethyl diester of N-trityl - {- glutamyl-glycine
The crude diester obtained according to a) is saponified by refluxing for five minutes with 10 cm3 of 20% methanolic potassium hydroxide and 2 cm3 of water. Diluted with 50 cm3 of water, filtered off with a slight insoluble material and acidified with acetic acid. After extraction with chloroform and washing with water, the chloroform solution is dried over magnesium sulphate and then evaporated to dryness.
Following the transposition caused by saponification, the product obtained consists essentially of N-trityl-α-glutamyl-glycine. The crude product is heated for five minutes in a water bath with 20 cm of acetic acid at 50 / n. The liberated triphenyl-carbinol crystallizes. Cooled, 10 cm3 of water are added and the triphenyl-carbinol formed is filtered off. The filtrate is concentrated to low volume. Absolute alcohol is added and left to crystallize.
After icing and draining, washing with alcohol and drying, 1.5 g (i.e. a yield of 75%) of α-glutamyl-glycine of formula VII (RI = H) are obtained which are recrystallized from alcohol at 50s / o. In this way, after drying, 1.1 g of α-glutamyl-glycine free of γ-glutamyl-glycine are obtained (the product does not give reaction to ninhydrin),
Mp = 220 C (dec.), [Oj = + 79 "5 1 (c = 2 / o, water). The product is soluble in water, dilute aqueous acids and alkalis, insoluble in alcohol and 'Úther Literature indicates a rotational power of +800.

The α-monoethyl N-trityl-L-glutamate used as the starting compound in this example can be obtained as follows:
To a solution cooled to 00 of 2.4 g of diethyl L-glutamate hydrochloride in 25 cm3 of chloroform and 3 cm3 of triethylamine is added 2.8 g of trityl chloride. We leave for an hour at ordinary temperature.
The chloroform solution of the reaction product is washed with water and dried over magnesium sulfate. The solvent is removed and the crude oily product is taken up in 40 cm3 of alcohol. The solution is neutralized with a few drops of normal sodium hydroxide, brought to reflux and added in fractions over about twenty minutes, as the neutralization progresses, which is followed with phenolphthalein, 10 cm3 of normal sodium hydroxide.
At the end of the reaction, the solution remains slightly alkaline. Cooled, acidified with acetic acid, the precipitate is filtered off and washed with water. After drying, 3.3 g of α-monoethyl N-trityl-L-glutamate are obtained, ie a yield of 80/0. The gummy product could not be crystallized. On the other hand, it provides in alcohol at 50 "/ o, with the D (-) - threo-lp-nitro-phenyl-2-amino-propane-1, 3-diol, a salt which is well crystallized in hydrated layers (approximately one water molecule of crystallization).
F = 100¯ C (dec.), [Α] 20 D = + 34¯, 5 1 (c = 2/0, methanol). The product is yellowish, soluble in alcohol and acetone, slightly soluble in ether, very slightly soluble in water, insoluble in benzene. Dilute acids and alkalis break it down.

The same product is obtained from other α-monoesters of N-trityl-glutamic acid such as α-methyl ester by following the same procedure. The ester radicals of glycine may be different from those of glutamic acid. Thus, from α-methyl N-trityl-glutamate and ethyl glycinate, the mixed diester of formula is obtained

Α-Monomethyl N-trityl-L-glutamate can be prepared as follows:
This compound is obtained starting from the hydrochloride of dimethyl L-glutamate by operating, for Ntritylation and saponification, as described above. The crude product can be crystallized from cyclohexane. The yield of pure product,
F = 140-141o C, [a] D = + 450 + 1 (c = 2 11 / o, methanol), is 50/0. It comes in colorless needles, soluble in acetone, chloroform, ether and dilute alkalis, sparingly soluble in alcohol, insoluble in water and dilute acids. This product is new.
It provides, with D (-) - threo-1-p-nitro-phenyl-2-amino-propane-1, 3-diol, a hydrated salt,

Example 3
Preparation of L-a-glutamyl-glycine a) Condensation of a-benzyl N-trityl-L-glutamate
with benzyl glycinate
3.6 g of free benzyl glycinate are prepared by treating a solution of benzyl glycinate hydrochloride with the theoretical amount of 5 N potassium hydroxide, salted, extracted with ether and evaporated. The benzyl glycinate thus obtained is dissolved in 6 cm3 of methylene chloride, the solution of 5 g of dicyclohexylcarbodiimide in 10 cm3 of methylene chloride is added, cooled to 00 and 9.6 g of N-trityl-L is added. -crude α-benzyl glutamate.
After a treatment identical to that of Example 1, the crude dibenzyl diester of formula V (R ′ and R "= - CH -C6H Rj = H) is obtained with a quantitative yield. B) Saponification of the crude dibenzyl diester of
N-trityl-y-glutamyl-glycine and detritylation
The product obtained according to a) is saponified by boiling for ten minutes with methanolic potassium hydroxide as in the previous example. After distillation of part of the methanol and dilution with water,
extract, after acidification with acetic acid, with chloroform.
The product consisting of a mixture of N-trityl-L-a-glutamyl-glycine and of N-trityl-L-y-glutamyl-glycine obtained after evaporation of the chloroform is detritylated by heating with 50/0 aqueous acid. After separation of the triphenyl-carbinol, the mixture is concentrated and crystallized as before by addition of alcohol. After recrystallization from aqueous alcohol, one finally obtains, with a yield of 50 / o, pure α-glutamyl-glycine, identical to the product obtained in Example 1.
The α-benzyl N-trityl-L-glutamate used as the starting compound can be obtained as follows:
This compound is prepared starting from α-benzyl and γ-methyl N-trityl-Lglutamate described in Example 2 of Swiss Patent No. 344714.
To a solution boiling at reflux of 9, 87 g of this mixed diester in 100 cm3 of methanol, is added little by little, over fifteen to twenty minutes, about 21 cm3 of normal sodium hydroxide and the boiling point is maintained for another fifteen minutes. The solution is concentrated by half, diluted with 40 cm3 of water, passed to vegetable black and then acidified with hydrochloric acid to pH 2 to 3. The product precipitates in the pulverulent state.

Example 4
Preparation of L-a-glutamyl-L-leucine a) Condensation of γ-methyl N-trityl-glutamate with methyl L-leucinate
3.2g of free methyl leucinate is prepared from its hydrochloride according to the technique used for the preparation of ethyl glycinate cited in Example 1. The product is dissolved in 20 cm3 of methylene chloride, 5 g is added. of dicyclohexylcarbodiimide, cooled to 0 C, add 8.1 g of γ-methyl N-trityl-glutamate, the preparation of which is described in Swiss patent No. 344714, stir to dissolve and leave for 65 hours in an icebox.
Then added 1 cm3 of acetic acid, left to stand for thirty minutes, drained the dicyclohexylurea which was washed with methylene chloride which was added to the filtrate. This filtrate is washed with dilute hydrochloric acid, with dilute ammonia and then with water, filtered and evaporated to dryness under vacuum. The oily residue is taken up in 25 cm3 of methanol, 4 cm3 of water are added, left to crystallize while ice-washing, filtered off, washed with aqueous methanol and dried. The directly usable product weighs 6.8 g.
The mother liquors give up by prolonged crystallization another 0.6 g of the same product of formula X

This compound is new. b) Saponification of the mithyl diester of L-N trityl- α -glutamyl-L-leucine and dÚtritylation
6.3 g of crude diester prepared according to a) are taken up in 20 cm3 of methanol.
10 cm3 of 20 / o methanolic potassium hydroxide and 5 cm3 of water are added. It is brought to reflux for two minutes and 25 cm3 of water are added to the boiling liquid, the methanol is driven off under partial vacuum, cooled, acidified to pH 2 by adding about 35 cms of normal hydrochloric acid and the trityl-peptide is extracted with chloroform . The chloroform solution is dried over magnesium sulfate, filtered and evaporated to dryness under vacuum. The residue is taken up in 20 cm of aqueous acetic acid at 50 ID / o. Boil for one minute, add 20 cm3 of water, filter the hot triphenyl-carbinol and wash it with boiling water. The combined acetic solutions are concentrated at low volume. The peptide crystallizes.
Absolute alcohol is added, ice, filtered off, dried and 2.2 g of L-a-glutamyl-L-leucine is obtained, ie a yield of 71 ID / o. The product is in colorless sheets, F 2500 C. In a capillary tube it melts at approximately 195¯ C. [a] 2D = + 10 + 1 (c = 2 / o, water and a molecule of hydrochloric acid). It is soluble in dilute aqueous acids and alkalis, sparingly soluble in water, insoluble in alcohol, ether, benzene and acetone. It does not give ninhydrin reaction. The same compound was obtained by another route by Le Quesne and Young (J. Chem.
Soc., 1950, p. 1954).
Example 5
Preparation of L-a-glutamyl-L-tyrosine a) Condensation of y-me N-trityl-L-glutamate
thyl with ethyl tyrosinate
The free ethyl tyrosinate is prepared by taking up the hydrochloride in excess 5N ammonia. The product crystallizes. It is filtered off, washed with water and dried in a sulfuric exsiccator.
By operating as in Example 1 a), starting 4.3 g of ethyl tyrosinate, 20 cm3 of methylene chloride and 8.1 g of γ-monomethyl LN-trityl-glutamate are obtained, 14 g of crude condensation product of formula X

Saponified as in Example 1 b) and in the presence of 20 cm3 of ether, the saponified product filtered off with hydrochloric acid is acidified to pH 2.
The compound resolves into a crystalline mass and then gradually disintegrates. After pasting with ether and filtering off, 7.8 g of N-trityl-α-glutamyl-peptide are obtained, which can be detritylated directly. For analysis, it is recrystallized from aqueous methanol. The product is presented in needles. [a] 25 D = + 19¯ ¯ 1 (c = 1/0, 75 "methanol / e + 1 NaOH). It is soluble in ether, chloroform and alcohol, sparingly soluble in dilute alkalis aqueous, insoluble in water, dilute acids decompose it.

F (dec.) = 235¯ C [α] D = + 29¯ ¯ 1 (c = 2 ID / o, water + 1 HC1). The peptide comes in colorless needles. It is soluble in aqueous acids and alkalis, very slightly soluble in water, insoluble in alcohol, acetone, benzene. It is identical to the product described by Bergmann et al. (Ztschr. F. Physiol. Chem., 1934, 224, 17).
