Procédé pour la préparation d'un dérivé furannique La présente invention a pour objet un procédé de préparation d'un dérivé furanniqu@e, la 2,5-diméthyl-4,5- dihyd,rofu.ranne-ol-(3)-one-(4) qui est douée de propriétés organoleptiques et est utilisée comme agent aromatisant de produits alimentaires et de boissons.
La 2,5-diméthyl-4,5-dihydro-furanne-ol-(3)-one-(4) a déjà été préparée soit a), par chauffage du rhamnose en présence d'acétate de pipéridine (JE. Hodge, B.E. Fisher et E.C. Nel.son, Proc. Am.
Soc. Brewing Chemists, p. 84 [1963]), soit b), par hydrogénation de l'acétylformoïne (A. Hofmann et C.H. Eugster, Helv. Chim. Acta 49, 53 [1966]), soit c), par hydrogénation catalytique du di- benzoate du 2,5-dihydroxyméthyl-3,4-dûbenzoxy-furanne (D. W. Henry et R.M. Silverstein, J. Org. Chem. 31, 2391 [1966]).
Ces .trois méthodes présentent les inconvénients suivants a) le rhamnose est un produit naturel rare et difficile à obtenir.
b) L'acétylformoïne n'est pas un produit commer- cial ; le rendement de l'hydrogénolyse n'est que 10 % environ.
c) La préparation du produit de départ comporte 4 étapes avec un rendement global d'environ 6,5 0/0. Le procédé suivant l'invention présente vis-à-vis des mé thodes d'obtention ci-dessus des avantages en ce qui concerne la disponibilité des matières premières, le ren dement, la commodité et le coût de la fabrication.
Le procédé selon l'invention consiste en la cyclisa- tion, en présence d'un catalyseur de cyclisation, de l'hexanediol-(3,4)-d.ione-(2,5) de formule CH3-CO-CHOH-CHOH-CO-CH 3 I Lorsqu'on cyclise I on obtient directement la 2,5- diméthyl-4,5-dihydro-furanne-ol(3)-one-(4), appelée éga lement dérivé furannique dans ce qui suit.
Pour la cyclisation, au cours de laquelle i1 y a perte d'une molécule d'eau, les agents déshydratants usuels (tel le P20,) ne sont pas efficaces. Comme catalyseur, on peut employer un sel formé par un acide et une amine organiques, par exemple l'acétate de pipéridine, ou un sel métallique d'un acide mono ou polybasique, par exemple le carbonate de soude ou un mélange de phos phates mono-, di- et trisodiques.
L'élimination de l'eau formée lors de la cyclisation n'est nullement indispensable à l'obtention d'un rende ment favorable en produit cyclisé et c'est là un des as pects surprenants et avantageux de l'invention : on a dé couvert en effet que la cyclisation peut s'effectuer en milieu protique et même aqueux.
L'hexane-diol-(3,4)-d.ione-(2,5) I, un corps nouveau, peut "être obtenue, soit à partir d'aldéhyde pyruvique par réduction par un métal en milieu acide ou encore par un sel dérivé de l'acide sulfoxylique, soit à partir du 2,4- dim6thyl-furanne qui est d'abord méthoxylé en -2,4- puis oxydé catalytiquement dans un milieu hydrolysant, soit encore par l'oxydation par l'air de l'acétol.
Selon un mode opératoire, on peut cycliser l'hexane- diol-(3,4)-dione-(2,5) I en présence d'un catalyseur salin. On peut faire agir par exemple le bicarbonate, le car bonate ou le phosphate de sodium, ou l'acétate de pi- péridine, en milieu aqueux ou alcoolique à des tem pératures comprises entre 20 et 150 C.
La vitesse de la cyclisation est en rapport direct avec la température et le pH du milieu cyclisant qui peut être compris entre 5 et 9 ; cependant, le produit cyclisé n'est pas très stable vis-à-vis des alcalis et pour cette raison, on préfère opé rer par exemple, en milieu presque neutre, par exemple dont le pH est compris entre 7 et 8, et avec des temps réactionnels longs à une température modérée, par exemple comprise entre 60 et 80 C ou alors, dès le début de la cyclisation adopter une technique d'extrac tion en continu qui permette de séparer le produit au fur et à mesure de sa formation.
Les exemples qui suivent illustrent la mise en oeuvre de l'invention. Les températures sont indiquées en degrés centigrades.
Exemple 1 <I>Préparation de la</I> 2,5-diméthyl-4,5-dihydro-furanne-ol- (3)-one-(4) Une solution contenant 5,4g d'hexane-diol-(3,4)- dione-(2,5), préparée selon le procédé décrit ci-dessous, dans l00 ml d'une solution saturée de bicarbonate de soude est soumise à une extraction au n-pentane pen dant une semaine, puis à l'éther contenant 10-151)/o de pentane (v/v) pendant également une semaine.
Les ex traits combinés fournissent après le traitement habituel 2,42 g (510/0) de dérivé furannique qui peut être puri fié par sublimation.
Spectre RMN (CDC13) s = 1,45 ppm, 3H, d ; 2,30 ppm, 3H, s ; 4,55 ppm, 1H, quarter, 7,65 ppm ; 1H, s. L'hexanediol-(3,4)-dione-(2,5) utilisée dans le présent exemple peut être préparée comme suit On chauffe 18 heures à 45-500 une solution conte nant 7,16g (45,2 m Mole) de 2,5-diméthyl-2,5-diméthoxy- dihydrofuranne, 7,9 g (64,5 m Mole) de chlorate de po tasse, 0,25 g (1,0 m Mole) de tétroxyde d'osmium, 50 ml de tétrahydrofuranne et 100 ,ml d'eau.
La solution est évaporée à sec sous vide et le .résidu épuisé à l'acétate d'éthyle. Les extraits organiques .sont filtrés, séchés suer NazS04 et par concentration fournissent 6,9 g de produit brut qui peut être utilisé directement pour la cyclisa- tion. Par cristallisation dans un mélange de CHC13 et CCl4 ou dans le chlorure de méthylène, on obtient l'échantillon analytique dont les propriétés spectrales sont les suivantes IR,
vC0 1710 cm-' (CHC13), RMN, & = 2,25 ppm, 6H, s ; 4,25 ppm, 2H, s ; 4,37 ppm, 2H, s. F. 58-60.
Exemple 2 <I>Préparation de la</I> 2,5-diméthyl-4,5-dihydrofuranne-ol-(3)- one-(4) On prépare une solution d'acétate de pipéridine en ajoutant à 17 ml d'eau 1,49 g d'acide acétique puis, goutte à goutte, 1,2 ml de pipéridine, la température étant maintenue au-dessous ou égale à 200.
Sous protec- tion d'azote on ajoute alors 2,85g d'hexane-diol-(3,4)- dione-(2,5) préparée selon le procédé décrit ci-dessous. On porte une semaine à reflux sous azote puis, entre 0 et 1011,
neutralise au moyen d'une solution aqueuse à 4% de NaOH. On extrait à l'éther en continu pendant la nuit. L'extrait éthéré, fournit, selon la technique ha bituelle, 1,85 g de dérivé furannique brut.
Par sublima- tion sous vide poussé (température du bain 800) on ob tient 1,34 g de produit sublimé que l'on lave deux fois avec un peu d'éther de pétrole.
On obtient ainsi <B>1,27</B> g (510/0) de dérivé furannique dont la pureté est attestée par l'analyse par chromatographie en couche mince ou phase gazeuse F. 77-79 .
L'hexanediol-(3,4)-dione-.(2,5) utilisée dans le présent exemple peut être préparée comme suit Sous vigoureuse agitation et protection d'azote, on chauffe un mélange de 680 g d'aldéhyde pyruvique sous forme d'une solution aqueuse à 50 0/0, 680 g d'éthanol et 500 g d'acide acétique glacial à 450.
On ajoute ensuite par portions, durant<B>1</B>1/2 heure, 600 g de zinc en poudre, la température étant maintenue entre 45 ,et 60o. On filtre la solution, la concentre sous vide et reprend le résidu par 500 ml d'acétate d'éthyle chaud.
On filtre la solu tion organique et par concentration .sous vide on pro voque l'a cristallisation de 336g de produit brut qu'on purifie ensuite par recristallisation dans l'acétate d'éthyle ou par sublimation. L'hexane-diol-(3,4)-dione-(2,5) pure fond à 89-90.
Exemple 3 <I>Préparation de la</I> 2,5-diméthyl-4,5-dihydrofuranne-ol- (3)-one-(4) On prépare une solution contenant 10g d'hexane- diol-(3,4)-dione=(2,5), préparée selon la méthode décrite ci-dessous, dissoute dans 120 ml d'une solution molaire aqueuse de phosphate trisodique. On ajuste :le pH de cette solution à pH 8-8,2 par adjonction d'une solution molaire d'acide orthophosphorique (70-80 ml environ) et chauffe le .tout à 750 pendant 24 heures.
On extrait en suite en continu pendant 24 heures avec un mélange 1 : 1 (v/v) d'éther et d'éther de pétrole {Eb. 30-50 ). L'extrait fournit, selon la technique habituelle, le dérivé furannique pratiquement pur selon l'analyse en RMN (rendement 55-60'0/a). L'hexane-diol-(3,4)-dnone-(2,5)
uti lisée dans le présent exemple peut âtre préparée comme suit Sous vigoureuse agitation et protection d'azote, on chauffe entre 40 et 500 un mélange de 625 g d'une so- lution aqueuse d'aldéhyde pyruvique à 40 %, 815 ml d'eau et 185 g d'acide acétique pur.
On introduit alors par portions, en maintenant la température entre 40 et 500, 220 g de zinc en poudre. L'addition demande en viron 3 heures, après quoi on maintient le mélange en agitation pendant 2 heures à température ordinaire. On filtre le zinc métallique non utilisé, concentre le filtrat sous vide à 35-400,
sépare l'acétate de zinc par filtra- tion et dilue le filtrat par 2 litres d'eau. On procède alors à une extraction en continu avec un mélange éther- éther de pétrole (Eb. 30-50 ), 2: 1 (v/v) pendant une semaine, puis avec un mélange 9 : 1 (v/v) pendant une autre semaine.
Après élimination des solvants volatils, l'extrait fournit 198,5 g (environ 80 0/0) d'hexane-diol- (3,4)-dione-(2,5) brute, cristallisant au froid, nr itdisable sans autre pour la préparation du dérivé fumannique. On peut également obtenir l'hexanediol@(3,4)-dione-(2,5) comme suit On chauffe à 900 une solution contenant 22g d'acé- tate de soude hydraté, 0,67 g d'acide sulfurique,
200 ml d'eau et 72,6 g d'acétol et y fait passer un courant d'air divisé en fines bulles pendant 70 heures. On évapore à sec ,sous vide, reprend par 200 ml d'eau et extrait en continu pendant 50 heures pair de l'éther contenant 10% (v/v)
d'éther de pétrole (Eb. 30-500. Le traitement usuel de l'extrait fournit 13,9 g de produit brut qui peut être employé directement pour la cyclisation.

These three methods have the following drawbacks a) rhamnose is a rare natural product and difficult to obtain.
b) Acetylformoin is not a commercial product; the yield of hydrogenolysis is only about 10%.
c) The preparation of the starting product comprises 4 stages with an overall yield of approximately 6.5%. The process according to the invention has, over the above methods of obtaining, advantages as regards the availability of the raw materials, the yield, the convenience and the cost of manufacture.
The process according to the invention consists of the cyclization, in the presence of a cyclization catalyst, of hexanediol- (3,4) -d.ione- (2,5) of formula CH3-CO-CHOH- CHOH-CO-CH 3 I When cyclized I one obtains directly 2,5-dimethyl-4,5-dihydro-furan-ol (3) -one- (4), also called furan derivative in what follows .
For cyclization, during which there is loss of a water molecule, the usual dehydrating agents (such as P20,) are not effective. As a catalyst, a salt formed by an organic acid and an organic amine, for example piperidine acetate, or a metal salt of a mono or polybasic acid, for example sodium carbonate or a mixture of mono phos phates, can be employed. -, di- and trisodium.
The elimination of the water formed during the cyclization is in no way essential to obtaining a favorable yield of cyclized product and this is one of the surprising and advantageous aspects of the invention: covered in fact that the cyclization can be carried out in a protic and even aqueous medium.
Hexanediol- (3,4) -d.ione- (2,5) I, a new body, can "be obtained either from pyruvic aldehyde by reduction with a metal in an acidic medium or by a salt derived from sulfoxylic acid, either from 2,4-dim6thyl-furan which is first methoxylated in -2,4- then oxidized catalytically in a hydrolysing medium, or again by oxidation by air acetol.
According to one procedure, hexanediol- (3,4) -dione- (2,5) I can be cyclized in the presence of a salt catalyst. For example, bicarbonate, sodium carbonate or phosphate, or peridin acetate, can be made to act in an aqueous or alcoholic medium at temperatures between 20 and 150 C.
The rate of cyclization is directly related to the temperature and the pH of the cyclizing medium, which may be between 5 and 9; however, the cyclized product is not very stable vis-à-vis alkalis and for this reason, it is preferred to operate, for example, in an almost neutral medium, for example whose pH is between 7 and 8, and with long reaction times at a moderate temperature, for example between 60 and 80 ° C. or else, from the start of cyclization, adopt a continuous extraction technique which makes it possible to separate the product as it is formed.
The examples which follow illustrate the implementation of the invention. Temperatures are given in degrees centigrade.
Example 1 <I> Preparation of </I> 2,5-dimethyl-4,5-dihydro-furan-ol- (3) -one- (4) A solution containing 5.4 g of hexane-diol- ( 3,4) - dione- (2,5), prepared according to the process described below, in 100 ml of a saturated solution of sodium bicarbonate is subjected to extraction with n-pentane for a week, then to ether containing 10-151) / o pentane (v / v) for also a week.
The combined extracts after the usual treatment provide 2.42 g (510/0) of furan derivative which can be purified by sublimation.
NMR spectrum (CDCl3) s = 1.45 ppm, 3H, d; 2.30 ppm, 3H, s; 4.55 ppm, 1H, quarter, 7.65 ppm; 1H, s. The hexanediol- (3,4) -dione- (2,5) used in the present example can be prepared as follows. A solution containing 7.16 g (45.2 m Mole) is heated for 18 hours at 45-500. 2,5-dimethyl-2,5-dimethoxy-dihydrofuran, 7.9 g (64.5 m Mole) of po cup chlorate, 0.25 g (1.0 m Mole) of osmium tetroxide, 50 ml of tetrahydrofuran and 100 ml of water.
The solution is evaporated to dryness in vacuo and the residue taken up with ethyl acetate. The organic extracts are filtered, dried over NazSO4 and on concentration provide 6.9 g of crude product which can be used directly for cyclization. By crystallization in a mixture of CHCl3 and CCl4 or in methylene chloride, the analytical sample is obtained, the spectral properties of which are as follows IR,
vCO 1710 cm- '(CHCl3), NMR, = 2.25 ppm, 6H, s; 4.25 ppm, 2H, s; 4.37 ppm, 2H, s. F. 58-60.
Example 2 <I> Preparation of </I> 2,5-dimethyl-4,5-dihydrofuran-ol- (3) - one- (4) A solution of piperidine acetate is prepared by adding to 17 ml of water 1.49 g of acetic acid and then, dropwise, 1.2 ml of piperidine, the temperature being kept below or equal to 200.
Under nitrogen protection, 2.85 g of hexane-diol- (3,4) - dione- (2,5) prepared according to the process described below are then added. It is brought to reflux for one week under nitrogen then, between 0 and 1011,
neutralized with a 4% aqueous solution of NaOH. It is extracted with ether continuously overnight. The ethereal extract provides, according to the usual technique, 1.85 g of crude furan derivative.
By sublimation under high vacuum (bath temperature 800), 1.34 g of sublimated product are obtained, which are washed twice with a little petroleum ether.
In this way <B> 1.27 </B> g (510/0) of furan derivative is obtained, the purity of which is attested by analysis by thin layer chromatography or gas phase F. 77-79.
The hexanediol- (3,4) -dione -. (2,5) used in the present example can be prepared as follows Under vigorous stirring and protection of nitrogen, a mixture of 680 g of pyruvic aldehyde is heated in the form of of a 50% aqueous solution, 680 g of ethanol and 500 g of glacial acetic acid at 450.
Then added in portions, during <B> 1 </B> 1/2 hour, 600 g of powdered zinc, the temperature being maintained between 45 and 60o. The solution is filtered, concentrated under vacuum and the residue is taken up in 500 ml of hot ethyl acetate.
The organic solution is filtered and by concentration in vacuo the crystallization of 336 g of crude product is caused, which is then purified by recrystallization from ethyl acetate or by sublimation. Pure hexane-diol- (3,4) -dione- (2,5) melts at 89-90.
Example 3 <I> Preparation of </I> 2,5-dimethyl-4,5-dihydrofuran-ol- (3) -one- (4) A solution is prepared containing 10 g of hexanediol- (3, 4) -dione = (2,5), prepared according to the method described below, dissolved in 120 ml of an aqueous molar solution of trisodium phosphate. The pH of this solution is adjusted to pH 8-8.2 by adding a molar solution of orthophosphoric acid (approximately 70-80 ml) and the whole is heated to 750 for 24 hours.
Extraction is carried out continuously for 24 hours with a 1: 1 (v / v) mixture of ether and petroleum ether (Eb. 30-50). The extract provides, according to the usual technique, the substantially pure furan derivative according to NMR analysis (yield 55-60 ° / a). Hexanediol- (3,4) -dnone- (2,5)
