Les deux composés ne diffèrent que par la posi tion spatiale du méthyle sur le carbone 5 et l'on dési gnera dans<B>ce</B> qui suit l'isomère dont le pouvoir rotatoire est [a]D = -108() (c = 0,5 %, chloroforme) par 5a-méthyl-réserpine (formule Xla) et par 5b-mé- thyl-réserpine (formule XIb), l'isomère dont [a]2 = -136o (c = 0,25 %,
chloroforme).
De la même façon on fera la distinction des inter médiaires de cette synthèse en les divisant en série a et série b, suivant qu'ils mènent vers la 5a- ou la 5b- méthyl-réserpine.
Le procédé objet de l'invention est caractérisé en ce qu'on condense la 1-(6'-méthoxy-3'-indolyl)-2- propylamine racémique avec l'ester méthylique du 1p-carboxy-méthyl 25-méthoxycarbonyl-3a-méthoxy- 4p-acétoxy-6p-formyl-cyclohexane dextrogyre, traite le mélange d'isomères en C5 de l'ester méthylique de l'acide 18p-ac6toxyi-11,17a-diméthoxy-16p-méthoxy- carbonyl-5-méthyl-2-3,3-4-diséco-A4(z1)
-20a-yohim- bène-3-oïque ainsi obtenu par un borohydrure alcalin pour obtenir un mélange isomère en C5 de 18[P-hy droxy-11,17a-diméthoxy-16p-méthoxycarbonyl-5-mé- thyl-3-oxo-2-3-séco-20a-yohimbanes, saponifie le mé lange isomère obtenu en un mélange isomère en C5 de 16a-carboxy-18µ-hydroxy-11,17a-diméthoxy- 5 -mé- thyl-3-oxo-2-3-séco-2.Oa-yohimbanes,
transforme ce mélange de yohimbanes en mélange de lactones cor respondantes, sépare les deux stéréoisomères eu C5, et continue à soumettre chaque isomère séparément aux opérations suivantes, c'est-à-dire cyclise au moyen d'oxy-chlorure de phosphore en lactones du 16[i-carbo xy-180-hydroxy-11,7a-diméthoxy-5-méthyl-03-20a- yohimbène correspondantes,
réduit ces lactones par un borohydrure alcalin, transforme les lactones des 16p- carboxy-18(3-hydroxy-11,17a-diméthoxy- 5 -méthyl- 20a-yohimbanes formées en leurs isomères 3p par action d'acide formique, soumet à la méthanolyse ces derniers composés et estérifie par le chlorure de 3,4,
5-triméthoxy-benzoyle les 18(3-hydroxy-11,17a-dimé thoxy-16p-méthoxycarbonyl-5-méthyl-3p,20a-yohim- banes obtenus en 5-méthyl-r-serpmes.
La 1-(6'méthoxy-3'-indolyl)-2-propylamine racé mique, l'un des composés de départ utilisés dans le présent procédé, peut être obtenue selon le procédé décrit dans le brevet suisse NI, 369761.
L'autre com posé de départ, l'ester méthylique du 1(3-carboxy-mé thyl-2p-méthoxycarbonyl-3a-méthoxy-40-acétoxy-6a- formyl-cyclohexane racémique, [6120 = -I- 42,50 (c = 1 %, pyridine), peut être obtenu selon le procédé décrit dans le brevet suisse No 372290. Il convient d'effectuer la condensation de ces deux composés au sein d'un solvant neutre tel que le chlorure de mé thylène.
La transformation du produit de condensa tion en composé IV, par réduction de la double liai son en 4(21) et cyclisation consécutive accompagnée de saponification partielle est effectuée de préférence en solution alcoolique à l'aide de borohydrure de potassium.
La séparation des lactones isomères du 16p-carboxy-18a-hydroxy-11,17a-diméthoxy- 5 -mé- thyl-3-oxo-2-3-séco-20a-yohimbane peut se faire grâce à l'insolubilité dans l'acétate d'éthyle de Piso- mère fondant à 2300 C, [ ]D = - 480 (c = 0,5 %, diméthylformamide). La réduction des lactones iso mères du 165-carboxy-18a-hydroxy-11,
17a-dimétho- xy-5-méthyl-A3-20a-yohimbène (base quaternaire) VII est également effectuée de préférence en solution alcoolique à l'aide de borohydrure de potassium.
Les deux stéréoisomères de la 5-méthyl-réserpine lévogyre appartiennent aux composés physiologique- ment actifs de la famille de la réserpine et possèdent des propriétés pharmacodynamiques intéressantes. Dans les exemples suivants, les points de fusion sont des points de fusion instantanée, déterminés sur bloc de Maquenne. Les températures sont exprimées en degrés centigrades.
<I>Exemple 1</I> <I>a) Préparation de Lester méthylique de l'acide 18(p-</I> acétoxy-11,17a-diméthoxy-16p-méthoxy-carbonyl- 5-méthyl-2-3,3-4-diséco-A4(2r) <I>_</I> 20a-yohimbène-3- oïque <I>(III)</I> <I>A) Libération de la</I> tryptamine <I>à partir de son picrate</I> On ajoute à 10 g de picrate de la 1-(6'méthoxy- 3'-indolyl)-2-propylamine, 1, obtenue selon le procédé décrit dans le brevet suisse No. 369761, 200 ce de chlorure de méthylène et 100 cms de lithine aqueuse à 10% et agite jusqu'à dissolution complète.
Après décantation de la phase organique, on extrait la phase aqueuse deux fois au chlorure de méthylène, lave les liqueurs organiques réunies avec la lithine et à l'eau et sèche sur potasse. On filtre, distille à sec et re- cùeille 6 g d'un résidu fortement solvaté, constitué par l'amine libre.
<I>B) Estérification de (acide du composé II</I> 7,5 g de lp-carboxyméthyl-2S-méthoxy-carbonyl- 3a-m6thoxy-4S-acétoxy-6p-formyl-cyclohexane- dex- trogyre (pyridine), II, en solution dans le chlorure de méthylène, sont méthylés au diazométhane à 0,) C.
La solution est ensuite concentrée sous vide à 50 cm3. <I>C) Condensation</I> On ajoute à la solution du composé II, obtenu selon B) la tryptamine I, obtenue selon A), et aban donne le mélange réactionnel pendant une demi-heure à l'abri de lumière à 00 C.
On laisse ensuite remonter lentement la température à 18 C et après une demi- heure refroidit de nouveau à 50 C. Le composé III ainsi formé est utilisé sans autre purification pour la réduction qu'on effectue au stade suivant de la syn thèse. Ce produit est nouveau.
<I>b) Préparation du</I> 18S-hydroxy-11,17a-diméthoxy- 16p-méthoxycarbonyl-5-méthyl-3-oxo-2 <I>-3</I> -séco- 20a-yohimbane <I>(IV)</I> A la base de Sehiff, <B>111,</B> obtenue à partir de 10 g de picrate de l'amine I et contenue dans<B>100</B> cm3 de chlorure de méthylène, on ajoute 70 cm3 de mé thanol, puis, par petites fractions, 2.,5 g de borohy- drure de potassium.
Le mélange réactionnel est laissé en bain de glace pendant une heure, abandonné à la température ambiante pendant l'heure suivante et chauffé ensuite au reflux pendant 65 minutes. On glace, ajoute de l'acide acétique jusqu'à pH 6, puis de l'acide chlorhydrique jusqu'à pH 1 et extrait au chlorure de méthylène. Les extraits sont lavés à l'eau et distillés à sec. Le résidu, constitué par le composé IV fortement solvaté, pèse 13 g. Ce composé n'est pas décrit dans la littérature.
<I>c) Préparation du</I> 16p-carboxy-18p-hydroxy-11,17a- diméthoxy-5-méthyl-3-oxo-2-3 -séco-20a-yohim- bane <I>(V).</I>
On introduit 13 g de composé IV, obtenu sous forme de résine solvatée, dans une solution contenant 8 cm3 de soude 10 N, 35 ce de méthanol et 18 cm3 d'eau, et chauffe au reflux pendant une heure. Après refroidissement et addition d'eau, on extrait deux fois au chlorure de méthylène, ajoute, sous agitation à la liqueur aqueuse, de l'acide chlorhydrique jusqu'à pH 7, puis de l'acétate d'éthyle et encore de l'acide chlor hydrique jusqu'à pH 1. On extrait à l'acétate d'éthyle, lave les extraits à l'eau salée, filtre et distille à sec. On répète cette opération une deuxième fois sur les liqueurs aqueuses, saturées de sel.
Les résidus de dis tillation réunis pèsent 8,6 g et sont constitués par le composé V, directement utilisable pour la suite de la synthèse. Ce produit n'est pas décrit dans la litté rature.
<I>d) Préparation de la</I> lactone <I>du</I> 165-carboxy-I8p- hydroxy-11,17a-diméthoxy-5-méthyl-3 -oxo-2 <I>-3</I> - séco-20a-yohimbane <I>(VI) et séparation de ses</I> <I>deux isomères en</I> C;;.
On mélange 8;5 g de composé V, obtenu sous forme d'une résine, 70 cm3 d'acide acétique, 80 crie d'anhydride acétique et 4 g d'acétate de lithium et chauffe à 85o C pendant deux heures et demie. Après addition d'eau, on abandonne la solution formée pen dant d'eux heures à la température ambiante, extrait ensuite au chlorure de méthylène et lave les extraits à l'eau, puis à l'eau saturée de bicarbonate de soude et à l'eau pure.
On filtre, distille à sec sous vide et on obtient 7,5 g de résine solvatée, constituée par le composé VI. On reprend ce résidu par 15 cm3 d7acé- tate d'éthyle, amorce, gratte et essore un premier jet de 1,08 g et un deuxième jet de 0,2 g. F. = 230o C, [a]20 _<B>-480</B> 5 (c = 0,5 %, diméthylformamide, D d'un composé qu'on désignera par Vla.
Ce composé est soluble en diméthylformamide, peu soluble en chloroforme et pyridine et insoluble dans l'éther.

On distille, d'autre part, la liqueur-mère à sec et la résine résiduelle de 4,8 g est constituée par l'iso mère VIb, contaminé par l'isomère VIa. On l'utilise sans autre purification pour la suite de la synthèse (voir exemple 2a).
Les deux isomères sont des produits nouveaux. <I>e) Préparation de la lactose du</I> 16(i-carboxy-18a- carboxy-1 Sa-hydroxy-11,17a-diméthoxy-5a-mé- thyl-1\3-20a-yohimbène (Vlla) <I>(base quaternaire).</I> On porte au reflux pendant deux heures 780 mg de lactame, VIa, avec 4 cm3 d'oxychlorure de phos phore et distille ensuite à sec sous vide. La résine rouge obtenue est constituée par le composé VIIa qui est nouveau.
<I>f) Préparation de la lactose du</I> 16(3-carboxy-18p- hydroxy-11,17a-diméthoxy-5a-méthyl-2 0a-yohim - bane (VI1Ia).
On dissout le composé VIIa, obtenu à partir de 780 mg de lactose VIa, dans 10<I>ce</I> de méthanol à 0 C et ajoute par petites fractions du borohydrure de potassium jusqu'à pH 10-11. Il se forme des mous ses. Après addition d'eau, on gratte, essore, sèche et recueille 680 mg (soit 80 %) d'un solvat à point de fusion se situant vers 1350 C - 138 C et 2500 C, [a]D = - 2160 (c = 0,25 %, chloroforme). Ce nou veau produit, VIIIa, est soluble en chloroforme, peu soluble en éthanol ou méthanol et insoluble dans l'éther.

2.42 mg de composé VIIIa dans 3 cm3 d'acide formique sont portés au reflux pendant 15 minutes, on glace, ajoute de l'eau et du bicarbonate jusqu'à pH 8, essore, lave et sèche. Rendement: 225 mg (soit 90 %) de composé IXa brut. On le redissout en acétone, filtre, concentre, glace et essore pour recueil lir 140 mg de cristaux fondant à 2800 C, [a]20 = -I- 19o 1 (c = 0,25 %, chloroforme), insolubles dans l'eau et l'éther, peu solubles en acétone et chlo roforme.

<I>h) Préparation du 180-hydroxy-11,17a-diméthoxy-</I> 16(3-méthoxycarbonyl-5a-méthyl-35,20a-yohim- bane (Xa).
On ajoute à 405 mg de lactose IXa, obtenus selon g), dans 12 ce de méthanol, 4 em8 de métha nol contenant 1 mg de sodium par cm3 et chauffe pendant 2 heures au reflux. On refroidit en glaçant, ajoute 25 cm-' d'eau et extrait au chlorure de méthy lène. Les extraits lavés à l'eau, sont distillés à sec et fournissent 480 mg d'une résine constituée par le composé Xa. Ce nouveau produit est directement utilisable pour l'estérification finale.
<I>i) Préparation de la</I> 5a-méthyl-réserpine lévogyre (pyridine) (XIa).
On mélange 480 mg de composé Xa avec 1,2 g de chlorure de 3,4,5-triméthoxy-benzoyle et 5 cm3 de pyridine, et chauffe en vase clos sous atmosphère d'azote pendant 16 heures à 80o C. Après refroidis sement, on ajoute quelques emB d'eau, chauffe à 50 C, puis abandonne la solution obtenue pendant une heure et demie. On extrait ensuite plusieurs fois au chlorure de méthylène, lave les extraits à l'acide chlorhydrique, au bicarbonate de soude et à l'eau, puis distille à sec et recueille 1,1 g de résine brune.
La résine est reprise par le méthanol, on additionne 0,6 cm3 d'acide nitrique 2N, gratte, glace et essore le nitrate de la 5a-méthyl réserpine, XIa. On le met en suspension dans quelques cm3 dé méthanol et le redissout par addition d'ammoniaque concentrée. On verse dans l'eau, essore, lave et sèche pour obtenir 485 mg (soit 80 % de rendement pour les deux der niers stades) de la 5a-méthyl@réserpine, XIa, F.
_ 150- C environ, [a] D = - 10'80 10 (c = 0,5 %, chloroforme) qui est un composé nouveau, solvaté avec une molécule d'eau, soluble en alcool, acétone, benzène et chloroforme, insoluble dans l'éther.

On introduit 4,8 g de composé VIb, obtenu sous forme d'une résine selon l'exemple 1d, dans 9 cm'- d'oxychlorure de phosphore, porte au reflux pendant deux heures, distille à sec sous vide et reprend le résidu par 30 cm3 de méthanol. Le composé VIIb ainsi obtenu est utilisé tel quel pour le stade suivant de la synthèse.
Il n'est pas décrit dans la littérature. <I>b) Préparation de la</I> lactone <I>du 16a-carboxy-185-</I> hydroxy-11,17a-diméthoxy-5b-méthyl-20a-yohim- bane lévogyre (pyridine) (VIIIb).
A la solution méthanolique du composé VIIb, on ajoute par petites fractions du borohydrure de po tassium jusqu'à pH 10-11, puis de l'eau. On extrait au chlorure de méthylène, distille les extraits à sec et cristallise le composé VIIIb, dans l'acétate d'éthyle. On essore 630 mg de cristaux, F. = 2400 C, qu'on recristallise en acétate d'éthyle. On obtient 500- mg de cristaux fondant à 2450 C, [a]20 = -900 10 (c = 0,75 %, chloroforme) solubles en chloroforme, insolubles en éther.

Rende ment: 180 mg (soit 90 %) de composé IXb, F. = 320- C, [a] D = -I- 38,51, (c = 0,25 %, chloroforme) qu'on purifie pour l'analyse (sans que les constantes changent) par dissolution dans l'acétone et filtration. Le nouveau produit est hygroscopique, insoluble dans l'eau et l'éther.

On prépare le mélange de 2.75 mg de composé IXb, 10 cm3 de méthanol, 4 cm3 de tétrahydrofuran et 2,2 cm3 de méthanol contenant 1 mg de sodium par cm3 et chauffe au reflux deux heures et demie. Après refroidissement, on ajoute de l'eau, extrait au chlorure de méthylène, lave les extraits jusqu'à pH 7 et distille à sec. Le résidu, constitué par le composé Xb, pèse 280 mg. II est utilisable sans autre purifi cation pour l'estérification finale.
Pour l'analyse, on cristallise le nouveau composé en acétate d'éthyle, F. = 230o C, [a] D = -<B>1340</B> (c = 0,25 %, chloro forme) et vérifie sa structure par le spectre I.R. qui est concordant.
<I>e) Préparation de la</I> 56-méthyl-réserpine lévogyre <I>(chloroforme)</I> (XIb).
On ajoute à 280 mg de composé Xb brut, 4,5 cm3 de pyridine et 1 g de chlorure de 3,4,5-triméthoxy- benzoyle et chauffe sous atmosphère d'azote en vase clos 16 heures à 800 C. Après refroidissement, on ajoute quelques cm3 d'eau, abandonne pendant une heure à la température ambiante et sous agitation intermittente et extrait au chlorure de méthylène. Les extraits sont lavés successivement par l'acide chlor hydrique, l'eau, le bicarbonate de soude et l'eau et distillés à sec.
Ils fournissent 860 mg d'un résidu qu'on reprend par 4 cm3 de méthanol et précipite par addition d'acide nitrique concentré. On glace, essore, lave au méthanol. Le nitrate de la 5b-méthyl-réserpine fond au-dessus de 2500 C. On le met en suspension dans le méthanol et ajoute de l'ammoniaque. II y a dissolution puis cristallisation de la base libre. On glace, essore, sèche et recueille 190 mg de composé XIb, F. = 250o C, [a1= -136o (c = 0,2.5 20 % , chloroforme).
Le nouveau produit, non décrit dans la littérature, se présente sous forme de cristaux incolo res, solubles en chloroforme, peu solubles en acétone et alcool, insolubles dans l'eau et l'éther.

-20a-yohimbene-3-oic thus obtained by an alkali borohydride to obtain a C5 isomer mixture of 18 [P-hy droxy-11,17a-dimethoxy-16p-methoxycarbonyl-5-methyl-3-oxo -2-3-seco-20a-yohimbanes, saponifies the obtained isomeric mixture into a C5 isomeric mixture of 16a-carboxy-18µ-hydroxy-11,17a-dimethoxy-5-methyl-3-oxo-2- 3-seco-2.Oa-yohimbanes,
transforms this mixture of yohimbanes into a mixture of corresponding lactones, separates the two C5 stereoisomers, and continues to subject each isomer separately to the following operations, i.e. cyclizes by means of phosphorus oxy-chloride to lactones of the 16 [i-carbo xy-180-hydroxy-11,7a-dimethoxy-5-methyl-03-20a- yohimbene corresponding,
reduces these lactones with an alkali borohydride, converts the lactones of 16p-carboxy-18 (3-hydroxy-11,17a-dimethoxy-5 -methyl-20a-yohimbanes formed into their 3p isomers by the action of formic acid, subject to the methanolysis these latter compounds and esterified with 3.4 chloride,
5-Trimethoxy-benzoyl 18 (3-hydroxy-11,17a-dimethoxy-16p-methoxycarbonyl-5-methyl-3p, 20a-yohimbans obtained in 5-methyl-r-serpmes.
Racemic 1- (6'methoxy-3'-indolyl) -2-propylamine, one of the starting compounds used in the present process, can be obtained according to the process described in Swiss patent NI, 369761.
The other starting compound, the methyl ester of 1 (3-carboxy-methyl-2p-methoxycarbonyl-3a-methoxy-40-acetoxy-6a-formyl-cyclohexane, racemic, [6120 = -I- 42.50 (c = 1%, pyridine), can be obtained according to the process described in Swiss patent No. 372290. It is advisable to carry out the condensation of these two compounds in a neutral solvent such as methylene chloride.
The transformation of the condensation product into compound IV, by reduction of the double bond at 4 (21) and subsequent cyclization accompanied by partial saponification, is preferably carried out in alcoholic solution using potassium borohydride.
The separation of the isomeric lactones of 16p-carboxy-18a-hydroxy-11,17a-dimethoxy-5-methyl-3-oxo-2-3-seco-20a-yohimbane can be accomplished due to the insolubility in the Isomer ethyl acetate, mp 2300 C, [] D = - 480 (c = 0.5%, dimethylformamide). The reduction of isomeric lactones of 165-carboxy-18a-hydroxy-11,
17α-dimetho-xy-5-methyl-A3-20a-yohimbene (quaternary base) VII is also preferably carried out in alcoholic solution using potassium borohydride.
The two stereoisomers of levorotatory 5-methyl-reserpine belong to the physiologically active compounds of the reserpine family and possess valuable pharmacodynamic properties. In the following examples, the melting points are instantaneous melting points, determined on a Maquenne block. Temperatures are expressed in degrees centigrade.

After decanting of the organic phase, the aqueous phase is extracted twice with methylene chloride, the combined organic liquors are washed with lithine and with water and dried over potassium hydroxide. It is filtered, distilled to dryness and 6 g of a strongly solvated residue, consisting of the free amine, is collected.
<I> B) Esterification of (acid of compound II </I> 7.5 g of 1p-carboxymethyl-2S-methoxy-carbonyl- 3a-methoxy-4S-acetoxy-6p-formyl-cyclohexane-dex-trorotatory (pyridine ), II, dissolved in methylene chloride, are methylated with diazomethane at 0,) C.
The solution is then concentrated under vacuum to 50 cm3. <I> C) Condensation </I> To the solution of compound II, obtained according to B) is added tryptamine I, obtained according to A), and the reaction mixture is given for half an hour in the dark at 00 C.
The temperature is then allowed to slowly rise to 18 ° C. and after half an hour is cooled again to 50 ° C. The compound III thus formed is used without further purification for the reduction which is carried out at the following stage of the synthesis. This product is new.
<I> b) Preparation of </I> 18S-hydroxy-11,17a-dimethoxy-16p-methoxycarbonyl-5-methyl-3-oxo-2 <I> -3 </I> -séco- 20a-yohimbane < I> (IV) </I> At the base of Sehiff, <B> 111, </B> obtained from 10 g of picrate of amine I and contained in <B> 100 </B> cm3 of Methylene chloride, 70 cm 3 of methanol are added, then, in small fractions, 2., 5 g of potassium borohydride.
The reaction mixture is left in an ice bath for one hour, left at room temperature for the next hour and then heated under reflux for 65 minutes. Ice, acetic acid is added to pH 6, then hydrochloric acid to pH 1 and extracted with methylene chloride. The extracts are washed with water and distilled to dryness. The residue, consisting of compound IV strongly solvated, weighs 13 g. This compound is not described in the literature.
<I> c) Preparation of </I> 16p-carboxy-18p-hydroxy-11,17a-dimethoxy-5-methyl-3-oxo-2-3 -séco-20a-yohimbane <I> (V) . </I>
13 g of compound IV, obtained in the form of a solvated resin, are introduced into a solution containing 8 cm3 of 10 N sodium hydroxide, 35 cc of methanol and 18 cm3 of water, and refluxed for one hour. After cooling and adding water, extracted twice with methylene chloride, added, with stirring with aqueous liquor, hydrochloric acid to pH 7, then ethyl acetate and more hydrochloric acid to pH 1. Extracted with ethyl acetate, the extracts washed with salted water, filtered and distilled to dryness. This operation is repeated a second time on the aqueous liquors, saturated with salt.
The combined distillation residues weigh 8.6 g and consist of compound V, which can be used directly for the remainder of the synthesis. This product is not described in the literature.
<I> d) Preparation of </I> lactone <I> of </I> 165-carboxy-I8p-hydroxy-11,17a-dimethoxy-5-methyl-3 -oxo-2 <I> -3 < / I> - seco-20a-yohimbane <I> (VI) and separation of its </I> <I> two isomers in </I> C ;;.
8.5 g of compound V, obtained in the form of a resin, 70 cm3 of acetic acid, 80% of acetic anhydride and 4 g of lithium acetate are mixed and heated at 85 ° C. for two and a half hours. After adding water, the solution formed is left for several hours at room temperature, then extracted with methylene chloride and the extracts washed with water, then with water saturated with sodium bicarbonate and with water. It is filtered and distilled to dryness under vacuum and 7.5 g of solvated resin, consisting of compound VI, are obtained. To 280 mg of crude compound Xb, 4.5 cm3 of pyridine and 1 g of 3,4,5-trimethoxy-benzoyl chloride are added and heated under a nitrogen atmosphere in a closed cup for 16 hours at 800 C. After cooling, a few cm 3 of water are added, left for one hour at room temperature and with intermittent stirring and extracted with methylene chloride. The extracts are washed successively with hydrochloric acid, water, sodium bicarbonate and water and distilled to dryness.
