Procédé pour la préparation de nouveaux prégnènes L'usage largement répandu de la prednisone et de la prednisolone est bien ,connu dans le domaine des stéroïdes. Bien que ces composés soient des 1,4- prégnadiènes et thérapeutiquement actifs, leur usage continu, en particulier sur de longues périodes de temps, peut provoquer des réactions secondaires in désirables telles que rétention de sodium et d'eau, l'ulcère peptique, l'interférence avec l'équilibre psy chique.

dans laquelle R est un radical alcanoyle inférieur et X est le radical divalent - CHOH - .
Les présents composés sont des solides cristallins à point de fusion relativement élevé, habituellement insolubles dans l'eau et légèrement solubles dans les solvants organiques usuels.
Le procédé selon l'invention est caractérisé par le fait qu'on met en réaction un 3,20-bis-alcoylène-di- oxy-5a,6a-époxy-prégnane-11(3,17a,21-triol avec un Grignard méthylique dans un solvant, afin de pro duire le 5a-hydroxy-6(3-méthyl-prégnane que d'on sou met ensuite à une oxydation pour obtenir le I1-céto- prégnane correspondant,
qu'on hydrolyse le composé ainsi obtenu dans un milieu acide pour éliminer le groupe éthylènedioxy en position 3 tout en produisant le 3-céto-prégnane correspondant, qu'on déshydrate le produit obtenu à l'aide d'un hydroxyde d'un métal alcalin pour obtenir le 20-éthylène-dioxy-17a,21-di- hydroxy-6a-méthyl-4-prégnène-3,11-dione,
qu'on met en réaction le composé formé avec un anhydride al- canoyle inférieur .pour former un groupe alcanoyloxy inférieur en position 21, qu'on fait ensuite agir un glycol et un agent acide sur le produit .obtenu pour former un groupe 3-alcoylènedioxy, qu'on met en ré action ce produit avec un chlorure de thionyle pour former une double liaison en position 16,17, qu'on soumet le produit obtenu à une réduction pour ob tenir un 3,
20 - bis - alcoylènedioxy - 6 - m6thyl-5,16-pré- gnadiène-lla,21-diol, que l'on traite à l'aide d'acide osmique et décompose le produit pour obtenir un 3,20 -bis - alcoylènedioxy-6-méthyl-5-prégnène-11 S, 16a,17a-21-,tétrol, qui fournit, après hydrolyse acide, le 11,16a,17a,21-tétrahydroxy-6a-méthyl-4-prégnène- 3,20-dione,
après quoi on estérifie ce produit à .l'aide d'un anhydride alcanoyle inférieur et fait réagir le produit obtenu avec SeO2 afin de produire une 16a, 21- di.- alcanoyloxy - (inférieur) -11(3,17a-dihydroxy-6a- m6thyl-1,4-prégnadiène-3,20-dione.
Le produit ainsi obtenu peut être hydrolysé afin d'obtenir la 11a,16a,17a,21-tétrahydroxy-6a-méthyl- 1,4-prégnadiène-3,20-dione.
Dans la mise en couvre de ce procédé, on peut poursuivre la réaction sur une période d'environ 5 à 48 heures et à une température d'environ 30 à 1750. On peut conduire le procédé partiellement ou en to- talité en atmosphère inerte.
De plus, selon ce procédé, quand un quelconque autre substituant sur le 5a,6u- époxystéroïde qui peut réagir avec ,les réactifs de Gri gnard est ;présent, on peut :le régénérer pendant l'iso lement de 6P-R-substitué-5a-hydroxystéroïde.
On prépare ,les composés par un .procédé à pha ses multiples en partant, par exemple, du 3,20-bis- éthylène- dioxy-5a,6a-époxy-prégnane-1 l [3,17a,21-triol décrit dans le J.A.C.S. <I>78,</I> page 984 (mars 1956). On fait réagir ce composé, par exemple, avec du bromure de méthyl-magnésium dans un solvant pour obtenir le 5a-hydroxyl-6(3-méthylprégnane correspondant (II).
On traite le 3-céto-prégnane correspondant (III, R = éthylène, R' = alkanoyle inférieur) par exemple par un hydroxyde alcalin pour obtenir la 20-éthylène- dioxy-17a- 21- dihydroxy -6a-méthyl-4-prégnène-3,11- dione (IV, R = éthylène, R' = alkanoyle inférieur).
Le schéma annexé illustre leprocédé général de la présente invention.
Dans la préparation des composés, on conduit la phase finale de préférence en dissolvant le 4-prégnène dans un alcool alkylique inférieur. L'addition d'une petite quantité d'un acide accroît habituellement la vitesse de réaction. On conduit de préférence la ré action au point d'ébullition du solvant, en conditions de reflux. La .température peut donc varier d'environ 40 à<B>1000</B> C. La réaction est pratiquement complète en 10 à 48 heures.
Après achèvement de la réaction, on peut purifier les produits désirés par séparation chromatographique ou autres méthodes connues dans la technique des stéroïdes.
Les composés obtenus selon la .présente invention peuvent être utilisés en combinaison avec des char ges, .des excipients, etc., dans la préparation de com primés, de poudre, de pilules et analogues. On -peut également les utiliser par voie parentérale en .solution ou en suspension.
<I>Exemple</I> (IA @ IIA) A 100 cc de tétrahydrofurane sous un courant d'azote, on ajoute 50 cc de bromure de méthylma- gnésium en solution 3M dans l'éther éthylique avec agitation vigoureuse.
A ce mélange, on ajoute une solution de 5,8g de 3,20-bis-éthylènedioxy-5a,6a-ép- oxy-prégnane-11,17a,21-triol dans 30 cc de tétrahydro- furane. On poursuit l'agitation et d'exclusion d'oxy gène, tandis que la réaction est soumise au reflux pendant 24 heures. Après refroidissement à tempéra ture ambiante, on ajoute 32 cc d'une solution saturée de chlorure d'ammonium jusqu'à formation d'un pré cipité blanc gélatineux.
On décante de liquide sur nageant, on lave :plusieurs .fois le .résidu solide au tétrahydrofurane chaud et on évapore .les décantats réunis, à sec, sous pression réduite.
On dissout l'huile jaune ainsi obtenue dans l'acé tate d'éthyle, on la -lave 3 fois avec une solution saline saturée et on la traite sur du sulfate de magnésium anhydre et du ,charbon de bois activé. La filtration sur terre à diatomées suivie par une évaporation à sec sous pression réduite donne un verre blanc auquel on ajoute 50cc d'acétone pour induire la cristallisa-. tion puis 200 cc d'éther de pétrole pour achever la précipitation.
Par refroidissement et filtration, on ob tient 4,55 g (76 %) de 3,20-bis-éthylènedioxy-6(3-mé- thyl-prégnane-5a,11p,17u.,21-tétrol, P.F. 170-172 C. Dans une autre expérience, en partant de <B>17,0</B> g de 3,20-bis-éthylènedioxy-5a,6a-époxy-pré- gn.ane-11(1,17a,21-triol, le processus ci-dessus donne le 3,20-bis-éthylènedioxy-6p-méthyl=prégnane-5a,11(3, 17a,21-tétrol, P.F. 2101, C.
Cinq cristallisations de cette matière dans le mélange acétone-éther de pétrole don nent le 3,20-bis-éthylènedioxy-6(3-méthyl-prégnane-5u., 11(3,17a,21-tétrol pur, P.F. 229-231(1 C ;<B>[a] 25</B> = 21,5e (C<B>1,995,</B> pyridine).

L'addition d'éther donne après filtration 200 mg d'un solide brun à bas point de fusion.
On cristallise dans le méthanol après évaporation à sec de la liqueur mère, ce qui donne 500 mg d'un .solide blanc, la 3,20-bis - éthylènedioxy- 5a,17a,21- trihydroxy-6(3-mé- thyl-prégnane-11-one, laquelle, par cristallisation ré pétée dans divers solvants, ne donne pas d'échantil lons analytiquement :purs.
(IIBD <U>-</U> IIIBD) A 230 mg de 21-acétoxy-3,20-bis-éthylènedioxy- 5a,17a-dihydroxy-6(3-méthyl-prégnane-11-one dans 3 cc d'acide acétique glacial chauffé sur un cône de vapeur, on ajoute 1 cc d'eau.

(VIIA D VIIIA) On ajoute goutte à goutte une solution de<B>1,64g</B> d'acide osmique dans 100 cc de benzène à 2,78 g de 3,20-bis-éthyèlnedioxy-6-méthyl-5,16-prégnadiène- l Ip-21-diol dissous dans 150 cc de benzène et 2 cc de pyridine. On agite le mélange pendant une heure, puis on le laisse reposer à la température ambiante pendant 16 heures.
On décompose l'ester osmiate par addition de 75 cc de méthanol, 10g de sulfite de sodium et 10g de bicarbonate de potassium dans 120 cc d'eau suivie d'une vigoureuse agitation pendant 3 heures. Le précipité minéral est filtré, lavé trois fois avec une solution saturée de chlorure de sodium, traitée au sulfate de magnésium anhydre et au char bon de bois activé, filtré et évaporé sous pression réduite pour donner 2,9 g d'huile, la cristallisation dans le mélange acétone-éther donne 1,35 g de pou dre blanche, point de fusion 277-279 C.
On cristal lise une .petite portion deux fois dans l'acétone et deux fois dans le méthanol pour obtenir le 3,20-bis éthylènedioxy-6-méthyl-5-prégnène-l1a-16a,17a,21-té- trol pur, .point de fusion: 280-282 C.
(VIIA @ VIIIaA) A une solution de 1,1 g de 3,20-bis-éthylènedioxy- 6-méthyl-5,16-prégnadiène-11(3,21-dioI dans 50cc d'éthanol, on ajoute 5 cc d'acide sulfurique à 8<B>%</B>. Après reflux pendant une heure, on concentre le mé lange réactionnel à environ 1/3 de son volume, on l'extrait à l'acétate d'éthyle, on le lave une fois au bicarbonate de sodium saturé et deux fois avec une solution saline saturée.
Le traitement de l'extrait par du sulfate de magnésium anhydre et du charbon de bois activé suivi d'une filtration et d'une évaporation sous pression réduite donne 700m- d'huile.
On soumet cette huile à une séparation chromato- graphique sur 250 g de terre à diatomées en utilisant le système acétate d'éthyle (2 parties), éther de pé trole (point d'ébullition 90-100, 4 parties), méthanol (3 parties) et eau (2 parties). Le volume en retour de 2,5 à 3 est évaporé sous pression réduite et cristal lisé dans le mélange acétone-benzène pour donner 135 mg de 11(3,21-dihydroxy-6a-.méthyl-4,16-prégna- diène-3,20-dione, point de fusion<B>:</B> 115-1200 C.

A 295 mg de 3,20-bis-éthylènedioxy-6-méthyl-5- prégnène-115,16u,17a,21-tétroi, en suspension dans 20 cc de méthanol, on ajoute 2 cc d'acide sulfurique à 8 % et on soumet la solution au reflux pendant 1 heure. On concentre le mélange réactionnel à envi ron 1/3 de son volume, on l'extrait à l'acétate d'éthyle, on le lave une fois avec une solution saturée de bi carbonate de sodium et deux fois avec une solution saline saturée.
On traite l'extrait au sulfate de sodium anhydre, on le filtre et on l'évapore sous pression ré duite pour obtenir, après cristallisation dans l'éther, 110mg de 11(3,16a,17a,21-tétrahydroxy-6a-méthyl-4- prégnène-3,20-dione ; point -de fusion 200-210 C.
(VIIIaA --a IXAC) A une solution agitée de 120mg de 11(3,21-dihydr- oxy-6a.-méthyl-4,16-prégnadiène-3,20-dione dans 10 cc de dichlorométhane, 10 cc de benzène et 0,1 cc de py- ridine, on ajoute 90 mg d'acide osmique.
On agite la solution à température ambiante pendant 1 h 1/.c, puis on ajoute 5 cc de méthanol, puis 650 mg de bicarbo nate de -potassium et autant de sulfite de sodium, dans 7 cc d'eau. On agite vigoureusement le mélange pen dant 3 heures, on ajoute 50 cc de chloroforme, on filtre la matière minérale, on lave le gâteau avec 200 cc .de chloroforme et on lave trois fois le filtrat à l'eau et finalement avec une solution saline saturée. Le traitement au sulfate de magnésium anhydre et au charbon de bois activé, la filtration et l'évaporation sous pression réduite donnent un solide blanc.
La cris tallisation dans le mélange acétone-éther donne 20 mg de 11(3,16a,17a,21-tétrahydroxy-6a-méthyl-4-,prégnène- 3,20-dione, point .de fusion 216-217 C.
(IXAC @ IXBC) A 200 mg de 11p,16a,17a,21-tétrahydroxy-6a-mé- thyl-4-prégnène-3,20-dione impure dans 3 cc de pyri- dine, on ajoute 1,5 cc d'anhydride acétique et on laisse le mélange reposer à la température ambiante pendant 18 heures. On dilue le mélange réactionnel avec de l'acétate .d'éthyle, on le lave une fois à l'acide chlorhydrique dilué froid, une fois au bicarbonate de sodium saturé et à l'eau jusqu'à neutralité.
Le traite ment au sulfate de magnésium anhydre et au charbon de bois activé, suivi de la filtration et de l'évaporation sous pression réduite donne 210 mg d'huile. La sé paration chromatographique sur 220 g de terre à dia tomées en utilisant un mélange de 2 parties d'acétate d'éthyle, 4,5 parties d'éther de pétrole (point d'ébul lition 90-1000, 3 parties de méthanol et 2 parties d'eau, donne, après évaporation du quatrième volume en retour, un solide à bas .point de fusion. Une cris tallisation dans le mélange acétone-éther de pétrole donne 33 mg. Point de fusion 174-176 C.

After completion of the reaction, the desired products can be purified by chromatographic separation or other methods known in the steroid art.
