Procédé de préparation de diesters 16a,21 de l'oxyde 9p,11(3 de 16a,17a,21-triol-3,20-dicéto-04-prégnène La présente invention a pour objet un procédé de préparation de diesters 16a,21 de l'oxyde 9(3,11(3 de 16 ,17u.,21-triol-3,20-dicéto-A4-prégnène qui sont des intermédiaires précieux pour la préparation de 9a-halo-16a-hydroxy-hydrocortisones et leurs esters décrits dans le brevet suisse No 347523.
Ces diesters 16a,21 de l'oxyde 9[3,1l(3 de 16a, 17a,21-triol-3,20-dicéto-A4-prégnène répondent à la formule

dans laquelle R et R' sont des radicaux alcanoyle inférieurs, et sont obtenus, selon l'invention, par la mise en réaction d'un diester 16a,21 de 9a-bromo- 11p,16a,17a,21-tétrol-3,20-dicéto-A4-prégnène avec un acétate alcalin anhydre,
les diesters obtenus pou vant être saponifiés pour obtenir les triols corres- pondants.
<I>Exemple</I> On soumet au reflux pendant 181/2 heures une solution de 14,5 g de diacétate-16a,21 de 9a-bromo- 11 0,16a,17a,21-tétrol-3,20-dicéta-A4-prégnène et 3,2 g d'acétate de potassium anhydre dans 1 litre d'alcool absolu. On évapore le mélange réactionnel jusqu'à siccité et on dissout le résidu dans l'acétate d'éthyle et l'eau. On extrait la phase aqueuse à l'acétate d'éthyle et on lave les extraits réunis avec une solu tion saturée de bicarbonate de sodium et de sel.
On évapore l'extrait séché et on dissout le résidu dans 35 cm3 de pyridine, on traite par 10 cm3 d'anhydride acétique et on laisse reposer à la température am biante pendant une nuit. On ajoute du méthanol et du benzène et on évapore la solution jusqu'à siccité.
La cristallisation du résidu dans le méthanol donne 6,5 g de diacétate-16a,21 de 16a, 17a,21-triol-3,20- dicéto-9p,11(p-oxy-A4-prégnène brut à point de fusion de 171,5-191,50 avec ramollissement préalable, con venant pour servir à de nouvelles transformations.
On hydrolyse en milieu faiblement alcalin. le di- acétate-16a,21 de 16a,17 ,21-triol-3,20-dicéto-9a, 11 p-oxy-A4-prégnène ci-dessus, pour obtenir l'oxyde- %l1 <B>p</B> de 16a,17a,21-triol-3,20-dicéto-A4-prégnène correspondant.
Le produit de départ, c'est-à-dire le diacétate- 16a,21 de 9a-bromo-ll(3,16a,17a,21-tétrol-3,20-di- céto-A4-prégnène, peut être préparé comme suit On laisse reposer à la température ambiante pen dant 18 heures une solution à 0,70 g d'acétate-21 de 21-ol-3,20-dicéto-A4,9(11)16-prégnatriène et 0,50 g de tétroxyde d'osmium, dans 10 cm3 de benzène et 0,
5 cm3 de pyridine. On décompose l'osmiate en ajoutant 35 cm3 d'eau, 10 cm3 de benzène, 23 cm3 de méthanol, 3,58 g de sulfite de sodium et '3,58 g de bicarbonate de potassium. Après avoir agité le mélange pendant 5 heures, on ajoute environ 150 cm3 de chloroforme, et on continue d'agiter pendant 1/2 heure.
On filtre le mélange sur de la terre d'infusoi res, on lave le résidu au chloroforme chaud et on sépare la couche organique. On extrait la phase aqueuse à plusieurs reprises au chloroforme et on lave les extraits réunis avec une solution saline satu rée et avec de l'eau.
On évapore l'extrait séché, sous pression réduite, et on cristallise le résidu dans un mélange d'acétone-éther de pétrole pour obtenir 0,62 g de produit brut, à point de fusion de 172-1740 C, avec ramollissement préalable. Trois cristallisations dans le mélange d'acétone-éther<B>de</B> pétrole donnent 0,42 g d'acétate-21 de 16a,17a,21- -triol-3,20-dicéta-A4,q(")-prégnadiène à point de fu sion de 195-197,5, avec ramollissement préalable.
Une recristallisation supplémentaire ne change pas le point de fusion:

A une solution de 325 mg d'acétate-21 de 16 ,17 ,21-triol-3,20-dicéto-A4,9(ll)_prégnadiène dans 20 cms de pyridine, on ajoute 2 cmB d'anhydride acé- tique, et on laisse reposer le mélange à la tempéra ture ambiante pendant 3 jours.
On verse la solution dans l'eau, on refroidit et on filtre le produit, on le lave à l'eau pour obtenir 0,33 g de diacétate-16a,21 de 16a,17a,21-triol-3,20-dicéto-A4,9(lï)-prégnadiène pur, à point de fusion 193-1950, avec ramollissement préalable.
Deux recristallisations dans le mélange d'acétone-éther de pétrole donnent 0,28 g de diacé- tate pur, à point de fusion de 194-195o avec ramol lissement préalable

On extrait au chloroforme le solide mou résultant, et on lave l'extrait avec une solution saline saturée, on sèche, on filtre sur de la terre d'infusoires et on évapore sous pression réduite pour obtenir une huile.
La cristallisation dans un mélange d'acétone-éther de pétrole donne 0,45 g de bromhydrine brute, à point de fusion de 130-135,50 (décomposition), avec bru- nissement préalable.

<I> Example </I> A solution of 14.5 g of diacetate-16a, 21 of 9a-bromo-11 0.16a, 17a, 21-tetrol-3,20- is refluxed for 181/2 hours. diketa-A4-pregnene and 3.2 g of anhydrous potassium acetate in 1 liter of absolute alcohol. The reaction mixture is evaporated to dryness and the residue dissolved in ethyl acetate and water. The aqueous phase is extracted with ethyl acetate and the combined extracts washed with a saturated solution of sodium bicarbonate and salt.
The dried extract is evaporated and the residue is dissolved in 35 cm3 of pyridine, treated with 10 cm3 of acetic anhydride and left to stand at room temperature overnight. Methanol and benzene are added and the solution evaporated to dryness.
Crystallization of the residue from methanol gives 6.5 g of 16a, 17a, 21-triol-3,20-diketo-9p, 11-diacetate-16a, 21 (crude p-oxy-A4-pregnene with melting point of 171.5-191.50 with prior softening, suitable for further processing.
Hydrolysis is carried out in a weakly alkaline medium. the di-acetate-16a, 21 of 16a, 17, 21-triol-3,20-diketo-9a, 11 p-oxy-A4-pregnene above, to obtain the oxide-% l1 <B> p < / B> of the corresponding 16a, 17a, 21-triol-3,20-diketo-A4-pregnene.

The mixture is filtered through diatomaceous earth, the residue washed with hot chloroform and the organic layer separated. The aqueous phase is extracted several times with chloroform and the combined extracts washed with saturated saline solution and with water.
The dried extract is evaporated under reduced pressure and the residue is crystallized from an acetone-petroleum ether mixture to obtain 0.62 g of crude product, with a melting point of 172-1740 C, with prior softening.
