<B>Procédé de</B> préparation <B>de nouveaux dérivés de la</B> phénothiazine La présente invention a pour objet un pro cédé de préparation de nouveaux dérivés de la phénothiazine substitués à l'azote par le groupement<I>-</I> (CH2)n <I>- CO - Z,</I> où<I>n</I> est un nombre entier inférieur à 3 et Z représente un reste aminé secondaire ou tertiaire.
Ces dérivés peuvent être représentés par la formule générale

dans laquelle les noyaux benzéniques de la phénothiazine peuvent être substitués par des atomes d'halogène ou des restes alcoyle ou alcoxy inférieurs.
Le procédé selon l'invention est caracté risé en ce qu'on fait agir une amine primaire ou secondaire sur une phénothiazine substituée à l'azote par un reste -(CH2),, -COX, dans lequel X est un atome ou groupe d'atomes qui s'élimine en se combinant avec un hydrogène lié à l'azote de l'amine. X peut être, en parti culier, un reste OR, R étant un alcoyle infé rieur. X peut être aussi un halogène, en parti culier le chlore. Dans ces deux cas, la réac tion est effectuée de préférence en présence d'un solvant.
Lorsque l'on utilise une phénothiazine substituée à l'azote par un reste -(CH2)n -COOR, la réaction peut dans certains cas être réalisée à température ordinaire ; il est cependant préférable d'opérer à température élevée (jusqu'à 1500, éventuellement dans un autoclave. Quand on emploie une phénothia- zine substituée à l'azote par un reste tel que -(CH2)n -COCI, la réaction se fait de pré férence dans un solvant à température ordi naire ou à une température inférieure à 100 C.
Les produits obtenus par le procédé selon l'invention sont des intermédiaires importants pour la préparation de produits pharmaceu tiques.
Les exemples suivants montrent comment le procédé selon l'invention peut être mis en aeuvre. Les points de fusion indiqués ont été déterminés au banc Kofler. <I>Exemple 1</I> On chauffe à 1000 pendant 16 heures 3 g de phénothiazinyl -10'-acétate d'éthyle avec 20 cm3 d'une solution éthylique à 18 % de
monométhylamine. Après refroidissement, on essore le produit formé, lave à l'alcool et sèche. On obtient 2,4 g de phénothiazinyl - 10' - acétyl - méthylamine qui, après recristallisation dans l'alcool absolu, fond à 228o. Le phénothiazinyl - 10' - acétate d'éthyle utilisé comme matière première est préparé à partir de phénothiazine et de bromo- acétate d'éthyle selon Cauquil et Cassadevall Cr. 225, 528 (1947).
<I>Exemple 2</I> On chauffe en tube scellé à 100o pendant 17 heures 2 g de l'ester méthylique de l'acide (phénothiazinyl - 10') - 3 - propionique avec 15 cm3 d'une solution alcoolique à 18 % de monométhylamine.
On obtient 1,7 g de (phénothiazinyl - 10') - 3 - propionyl-méthylamine qui, après recristallisation dans l'alcool méthylique, fond à 96o.
De même, en chauffant en tube scellé à 150 pendant 16 heures 1 g du même ester avec 1,5 g de pipéridine et 5 cm-3 de toluène, on obtient 0,8 g de (phénothiazinyl - 10') - 3 - propionyl - N pipéridine qui, après recristallisation dans 3 cm3 d'alcool méthyli que, fond à 120 .
On peut également chauffer en tube scellé à 100 pendant 20 heures 2 g de l'ester avec 14 cm3 d'une solution alcoolique à 29 % de monoéthylamine ; on obtient 2,1 g de (phéno- thiazinyl - 10') - 3 - propionyl - éthyla- mine qui, après recristallisation dans l'alcool éthylique à 500, fond à 1200.
L'ester méthylique de l'acide (phénothiazi- nyl - 10') - 3 - propionique est préparé de la manière suivante : on ajoute 20 g d'acide (phénothiazinyl - 10') - 3 - propionique préparé selon Smith, J. Org. Chem ; 15, 1125 (1950) dans 100 cm2 de méthanol, contenant 36,5 g de gaz chlorhydrique.
On laisse réagir à 50 pendant une heure, puis on distille sous vide, traite le résidu par 40 cm3 de solution aqueuse à 30 % de carbonate de sodium et on extrait avec 100 cm-' d'acétate d'éthyle. On ob tient 18 g de l'ester méthylique de l'acide (phénothiazinyl - 10') - 3 - propionique qui, après recristallisation dans le méthanol, fond à 680.
<I>Exemple 3</I> On dissout à chaud 0,64 g d'ester méthyli que de l'acide (chloro - 3' - phénothiazinyl - 10') - propionique dans 2 cm3 d'alcool méthylique. On refroidit à 200 et on ajoute 2 cm3 d'une solution alcoolique à 18 0i0 de monométhylamine. On laisse réagir pendant 4 heures, puis on chasse les solvants sous vide et on cristallise dans 4 cm3 d'alcool méthyli que.
On obtient ainsi la (chloro - 3' - phé- nothiazinyl - 10') - 3 - propionyl - mé- thylamine qui fond à 850.
Pour obtenir l'ester méthylique de l'acide (chloro - 3' - phénothiazinyl - 10') - 3 - propionique, on prépare tout d'abord le nitrile (P. F. 178-179)à partir de la chloro - 3 - phénothiazine et du nitrile acrylique par analogie avec la méthode décrite par Smith, J. Org. Chem. 15, 1129 (1950). On saponifie ce nitrile par la soude alcoolique et on obtient l'acide (chloro - 3' - phénothia- zinyl - 10') - 3 - propionique (P.
F. 156o). En opérant ensuite comme dans l'exemple 2, on prépare l'ester méthylique qui, après recris- tallisation dans le méthanol, fond à 740. <I>Exemple 4</I> Dans une suspension de 5 g d'acide phéno- thiazinyl-propionique [préparé selon Smith - J. Org. Chem. 15, 1125-1130 (1950)] dans 20 cm3 d'oxychlorure de phosphore, on coule, à une température inférieure à 10 , 3,
85 g de pentachlorure de phosphore dissous dans 38 cm2 d'oxychlorure de phosphore. On chauffe pendant une\ demi-heure à 500, puis chasse l'oxychlorure de phosphore sous vide à froid.
Le chlorure d'acide brut ainsi obtenu est repris par 75 cm3 de benzène et on coule peu à peu, en agitant, cette solution dans une solu tion de 3,4 g de pipéridine dans 25 cm3 de benzène anhydre, en maintenant la tempéra ture inférieure à 10,). On lave la solution benzénique successivement avec 50 cm3 d'eau, 40 cm3 d'acide chlorhydrique normal, 40 cm3 de solution saturée de bicarbonate de sodium, puis 25 cm3 d'eau. On la sèche sur sulfate de sodium, puis chasse le benzène sous vide.
On obtient 6 g de N - [(phénothiazinyl - 1M - 3 - propionyl] - pipéridine qui cristal lise par amorçage. Après recristallisation dans l'acétate d'éthyle, elle fond à 128-129.

The process according to the invention is characterized in that a primary or secondary amine is made to act on a phenothiazine substituted for nitrogen by a residue - (CH2) ,, -COX, in which X is an atom or group d atoms which get rid of by combining with a hydrogen bonded to the nitrogen of the amine. X can be, in particular, an OR residue, R being a lower alkyl. X can also be a halogen, in particular chlorine. In these two cases, the reaction is preferably carried out in the presence of a solvent.
When using a phenothiazine substituted on nitrogen with a residue - (CH2) n -COOR, the reaction can in some cases be carried out at room temperature; however, it is preferable to operate at high temperature (up to 1500, optionally in an autoclave. When using a phenothiazine substituted for nitrogen by a residue such as - (CH2) n -COCI, the reaction takes place preferably in a solvent at ordinary temperature or at a temperature below 100 C.
The products obtained by the process according to the invention are important intermediates for the preparation of pharmaceutical products.
The following examples show how the process according to the invention can be implemented. The indicated melting points were determined on the Kofler bench. <I> Example 1 </I> 3 g of phenothiazinyl -10′-ethyl acetate are heated at 1000 for 16 hours with 20 cm3 of an 18% ethyl solution of
monomethylamine. After cooling, the product formed is filtered off, washed with alcohol and dried. 2.4 g of phenothiazinyl - 10 '- acetyl - methylamine are obtained which, after recrystallization from absolute alcohol, melts at 228 °. Ethyl phenothiazinyl - 10 '- acetate used as raw material is prepared from phenothiazine and ethyl bromoacetate according to Cauquil and Cassadevall Cr. 225, 528 (1947).
<I> Example 2 </I> 2 g of the methyl ester of (phenothiazinyl - 10 ') - 3 - propionic acid are heated in a sealed tube at 100o for 17 hours with 15 cm3 of an alcoholic solution at 18 % monomethylamine.
1.7 g of (phenothiazinyl - 10 ') - 3 - propionyl-methylamine are obtained which, after recrystallization from methyl alcohol, melts at 96 °.
Likewise, by heating 1 g of the same ester in a sealed tube at 150 for 16 hours with 1.5 g of piperidine and 5 cm-3 of toluene, 0.8 g of (phenothiazinyl - 10 ') - 3 - propionyl is obtained. - N piperidine which, after recrystallization from 3 cm3 of methyl alcohol, melts at 120.
It is also possible to heat in a sealed tube at 100 for 20 hours 2 g of the ester with 14 cm3 of a 29% alcoholic solution of monoethylamine; 2.1 g of (phenothiazinyl - 10 ') - 3 - propionyl - ethylamine are obtained which, after recrystallization from ethyl alcohol at 500, melts at 1200.
The methyl ester of (phenothiazinyl - 10 ') - 3 - propionic acid is prepared as follows: 20 g of (phenothiazinyl - 10') - 3 - propionic acid prepared according to Smith, J. Org. Chem; 15, 1125 (1950) in 100 cm2 of methanol, containing 36.5 g of hydrochloric gas.
Allowed to react at 50 for one hour, then distilled in vacuo, the residue treated with 40 cm 3 of 30% aqueous sodium carbonate solution and extracted with 100 cm 3 of ethyl acetate. 18 g of the methyl ester of (phenothiazinyl - 10 ') - 3 - propionic acid are obtained which, after recrystallization from methanol, melts at 680.
<I> Example 3 </I> 0.64 g of methyl ester of (chloro - 3 '- phenothiazinyl - 10') - propionic acid is dissolved hot in 2 cm3 of methyl alcohol. The mixture is cooled to 200 and 2 cm3 of an 18010 alcoholic solution of monomethylamine are added. Allowed to react for 4 hours, then the solvents are removed in vacuo and crystallized from 4 cm3 of methyl alcohol.
There is thus obtained (chloro - 3 '- phenothiazinyl - 10') - 3 - propionyl - methylamine which melts at 850.
To obtain the methyl ester of (chloro - 3 '- phenothiazinyl - 10') - 3 - propionic acid, the nitrile (PF 178-179) is first prepared from chloro - 3 - phenothiazine and acrylic nitrile by analogy with the method described by Smith, J. Org. Chem. 15, 1129 (1950). This nitrile is saponified with alcoholic soda and the acid (chloro - 3 '- phenothiazinyl - 10') - 3 - propionic (P.
F. 156o). Then operating as in Example 2, the methyl ester is prepared which, after recrystallization from methanol, melts at 740. <I> Example 4 </I> In a suspension of 5 g of phenolic acid thiazinyl-propionic [prepared according to Smith - J. Org. Chem. 15, 1125-1130 (1950)] in 20 cm3 of phosphorus oxychloride, the mixture is poured at a temperature below 10, 3,
85 g of phosphorus pentachloride dissolved in 38 cm2 of phosphorus oxychloride. The mixture is heated for half an hour at 500, then the phosphorus oxychloride is removed in a cold vacuum.
The crude acid chloride thus obtained is taken up in 75 cm3 of benzene and this solution is gradually poured, with stirring, into a solution of 3.4 g of piperidine in 25 cm3 of anhydrous benzene, while maintaining the temperature. less than 10,). The benzene solution is washed successively with 50 cm3 of water, 40 cm3 of normal hydrochloric acid, 40 cm3 of saturated sodium bicarbonate solution, then 25 cm3 of water. It is dried over sodium sulfate, then the benzene is removed under vacuum.
6 g of N - [(phenothiazinyl - 1M - 3 - propionyl] - piperidine are obtained, which crystallizes on initiation. After recrystallization from ethyl acetate, it melts at 128-129.
