Procédé de préparation d'un acylamidodiol aromatique. Dans le brevet principal, on a décrit un procédé de préparation du !U-1-phényl-2-acét- amidopropane-1;3-diol.
Le présent brevet a pour objet un procédé de préparation d'un autre acylamidodiol aro matique.
Ce procédé est caractérisé en ce que l'on fait réagir le rég.-l-phényl-2-aminopropane- 1,3-diol avec un dérivé de l'acide dichloroacé- tique contenant au moins un radical C12CH-CO-, dans des conditions telles que, par fixation d'un radical C12CH-CO- sur le groupe amino, il se forme le rég.-l-phényl-2-dichloro- acétamidopropane-1,3-diol.
Le rég.-l-phényl-2-dichloroacétamidopro- pane-1,3-diol obtenu est une nouvelle subs tance, laquelle est cristallisée et présente un point de fusion de 158-159 C (forme racé mique). On peut citer comme dérivés appro priés pour effectuer ladite réaction de di- chloroacétylation, les halogénures, les alcoyl- esters ou l'anhydride de l'acide dichloroacé- tique. Lorsqu'on emploie un ester dudit acide, la réaction est effectuée dans des conditions anhydres.
Lorsqu'on emploie un halogénure ou l'anhydride de l'acide dichloroacétique, la réaction peut être effectuée en milieu anhydre ou en présence d'eau et en présence ou non d'une substance alcaline. Si on emploie l'an hydride dichloroacétique, il est toutefois pré- férable de ne pas effectuer la réaction en milieu -anhydre lorsqu'on. utilise un cataeur alcalin.
De préférence, on soumet à la dichloroacé- tylation la forme racémique (dl) du rég.-l- phényI-2-aminopropane-1,3-diol. Le produit obtenu par le procédé du présent brevet est un produit intermédiaire pour la préparation de composés ayant une activité antibiotique. <I>Exemple:</I> On traite 1,7 g de dl-rég.-l-phényl-2-amino- propane-1,3-diol avec 1,6 g de dichloroacétate de méthyle et on chauffe le mélange à 100 C pendant une heure et quart.
Le résidu est lavé avec deux portions d'éther de pétrole à 20 cms chacune, et le produit insoluble est recueilli. Par recristallisation dans de l'acétate d'éthyle, on obtient le dl-rég.-l-phényl-2-dichloroacét- amidopropane-1,3-diol recherché sous forme pure; point de fusion: 158-159 C. Process for preparing an aromatic acylamidodiol. In the main patent, a process for the preparation of! U-1-phenyl-2-acet-amidopropane-1; 3-diol has been described.
The present patent relates to a process for preparing another aromatic acylamidodiol.
This process is characterized in that the reg.-1-phenyl-2-aminopropane-1,3-diol is reacted with a dichloroacetic acid derivative containing at least one C12CH-CO- radical, in conditions such that, upon attachment of a C12CH-CO- radical to the amino group, the reg.-1-phenyl-2-dichloroacetamidopropane-1,3-diol is formed.
The reg.-1-phenyl-2-dichloroacetamidopropane-1,3-diol obtained is a new substance, which is crystallized and has a melting point of 158-159 C (racemic form). Mention may be made, as suitable derivatives for carrying out said dichloroacetylation reaction, of the halides, alkyl esters or anhydride of dichloroacetic acid. When an ester of said acid is employed, the reaction is carried out under anhydrous conditions.
When a halide or anhydride of dichloroacetic acid is employed, the reaction can be carried out in an anhydrous medium or in the presence of water and in the presence or absence of an alkaline substance. If dichloroacetic anhydride is used, however, it is preferable not to carry out the reaction in anhydrous medium when. uses an alkaline catalyst.
Preferably, the racemic form (dl) of reg.-1-phenyl-2-aminopropane-1,3-diol is subjected to dichloroacetylation. The product obtained by the process of the present patent is an intermediate product for the preparation of compounds having antibiotic activity. <I> Example: </I> 1.7 g of dl-reg.-1-phenyl-2-amino-propan-1,3-diol are treated with 1.6 g of methyl dichloroacetate and the mixture is heated at 100 C for an hour and a quarter.
The residue is washed with two portions of petroleum ether at 20 cms each, and the insoluble product is collected. By recrystallization from ethyl acetate, the desired dl-reg.-1-phenyl-2-dichloroacet-amidopropane-1,3-diol is obtained in pure form; melting point: 158-159 C. This product meets the formula
