Procédé de préparation d'un acylamidodiol aromatique, nitré dans le noyau. Dans le brevet principal, on a décrit un procédé de préparation du l'-1-p-nitrophényl- 2-clieliloroacétamidopropane-7,3-diol. Le pré sent brevet a pour objet un procédé de pré paration d'un autre acylamidodiol aromati que, nitré dans le noyau.
Ce procédé est ea- ractérisé en ce que l'on fait réagir le ïf-1-p- niti-opliényl-2-aniinopropane-1,3-diol avec un dérivé de l'acide ehloroacétiqtte contenant au moins un radical Cl--CI-12-C0, dans des conditions telles que, par fixation d'un radical Cl-C132-CO sur le groupe amino, il se forme le '1'-1-p-nit.rophényl-\?-chloroacétainido- propane-1,3-diol. Ce dernier est. une nouvelle substance cristallisée.
La forme racémique (dl) fond à 99-100 C, tandis que les isomères optiquement actifs (1 et d) présentent un point de fusion de 83-86 C.
On peut citer comme dérivés appropriés pour effectuer ladite réaction de chloroacéty- la-tion, les halogénures, les alcoyl-esters ou L'anhydride de l'acide chloroacétique. On em ploie de préférence un alcoyl-ester inférieur de l'acide chloroacétique. Lorsqu'on emploie un ester dudit acide, la réaction est effectuée dans des conditions anhydres et de préférence en présence d'une petite quantité d'un cataly seur alcalin tel. que le méthylate de sodium.
Lorsqu'on emploie un halogénure ou l'anhy dride (le l'acide ehloroacétique, la réaction peut être effectuée soit dans des eonditions anhydres, soit, en présence d'eau et en pré- @ence ou non d'une substance alealine. Toute- fois, si on emploie l'anhydride chloroacétique, il est. préférable de ne pas effectuer la réac tion dans des conditions anhydres dans le cas ou on utilise un catalyseur alcalin.
De préférence, on soumet. à ladite ehloro- acétylation la forme racémique (dl) ou l'iso mère optiquement actif lévogy re du !Y-1-p- nitrophényl-2-aniinopropane-1,3-diol. Le pro duit obtenu par le procédé du présent brevet, surtout la forme racémique et la forme lévo- gy re,
a tune valeur thérapeutique comme anti biotique et se prête pour la préparation d'au tres composés ayant une activité antibiotique. Exemple <I>1:</I> Un mélange composé de 1 g de dl-#u-1-p- nitrophényl-2-aminopropane-1,3-diol, de 50 cni3 d'acétate d'éthyle et de 50 cm3 d'une solution <B>0,5</B> n de hOII est traité à 0 C, en brassant, avec 1,:5 g de chlorure de chloroacéty le.
On sépare la phase organique du mélange et on extrait la phase aqueuse avec deux portions égales d'acétate d'éthyle. Les extraits d'acé tate d'éthyle sont réunis, lavés avec de l'acide sulfurique 0,1 n, puis avec une solution de bicarbonate de soude à 51/o et finalement avec de l'eau. On sèche les extraits au moyen de sulfate de magnésium anhydre et on élimine l'acétate d'éthyle par distillation.
Le résidu est cristallisé plusieurs fois dans du diehlorure d'éthylène pour obtenir le dl-#u-1-p-nitro- phényl-?-ehloroacétamidopropane-1,3-diol re cherché sous forme d'ur-ie substance cristallisée du point de fusion 99-100 C.

In the main patent, a process for the preparation of 1'-1-p-nitrophenyl-2-clieliloroacetamidopropane-7,3-diol has been described. The present patent relates to a process for the preparation of another aromatic acylamidodiol, nitrated in the nucleus.
This process is characterized by reacting ïf-1-p-niti-opliényl-2-aniinopropane-1,3-diol with a derivative of chloroacétiqtte acid containing at least one radical Cl-- CI-12-C0, under conditions such that, by attachment of a C1-C132-CO radical to the amino group, the '1'-1-p-nit.rophenyl - \? - chloroacetainidopropane is formed -1,3-diol. This last one is. a new crystallized substance.
The racemic form (dl) melts at 99-100 C, while the optically active isomers (1 and d) have a melting point of 83-86 C.
Mention may be made, as derivatives suitable for carrying out said chloroacetylation reaction, of the halides, alkyl esters or anhydride of chloroacetic acid. Preferably, a lower alkyl ester of chloroacetic acid is employed. When an ester of said acid is employed, the reaction is carried out under anhydrous conditions and preferably in the presence of a small amount of such an alkaline catalyst. than sodium methoxide.
When a halide or anhydride (chloroacetic acid is employed, the reaction can be carried out either under anhydrous conditions or in the presence of water and in the presence or absence of an aleal substance. However, if chloroacetic anhydride is employed, it is preferable not to carry out the reaction under anhydrous conditions in the case where an alkaline catalyst is used.

The organic phase is separated from the mixture and the aqueous phase is extracted with two equal portions of ethyl acetate. The ethyl acetate extracts are combined, washed with 0.1 n sulfuric acid, then with 51% sodium bicarbonate solution and finally with water. The extracts were dried with anhydrous magnesium sulfate and the ethyl acetate was removed by distillation.
