Procédé de préparation d'un acylamidodiol aromatique, nitré dans le noyau. Dans le brevet principal, on a décrit un procédé de préparation du l'-1-p-tiitrophéiiyl- 2-dicliloroacétamidopropane-1,3-diol. Le pré sent brevet a pour objet un procédé de pré paration d'un autre aeylamidodiol aromati que, nitré dans le noyau.
Ce procédé est ca ractérisé en ce que l'on fait réagir le l'-1-p- nitrophényl-2-aminopropane-1,3-diol avec un dérivé < le l'acide fluoroacétique, contenant au moins un radical FCH2C0-, dans des condi tions telles que, par fixation d'un radical FCI12C0- sur le groupe amino, il se forme le 'F-1-p-nitrophényl-2-fluoroacétamidopro- pane-1,3-diol. Ce dernier est une nouvelle substance cristallisée,
dont la forme racémi- clue fond à<B>1.20-1210C.</B>
On peut citer comme dérivés appropriés pour effectuer ladite réaction de fluoroaeéty- lation, les halogénures, les alcoyl-esteis ou l'anhydride de l'acide fluoroacétique. Lors qu'on emploie un ester dudit acide, la réac tion est effectuée dans des conditions anhy- dres, de préférence en présence d'une petite quantité d'un catalyseur alcalin tel que le méthylate de sodium. Lorsqu'on emploie un halogénure ou l'anhydride de l'acide fluoro- aoétique, la réaction peut.
être effectuée soit. dans des conditions anhydres, soit en présence d'eau, et en présence ou non d'une substance alcaline. Si on utilise l'anhydride de l'acide :luoroacétique dans des conditions anhydre, il est préférable de ne pas employer un cata- ly seur alcalin.
De préférence, on. soumet à. ladite fluoro- acétylation la forme racémique (dl) ou l'iso mère optiduement actif lévogvre du #U-1-p- i!itrophényl-2-aminopropane-1,3-diol. Le pro duit obtenu par le procédé selon.le présent. firevet, surtout la forme racémique et. la, forme lévogvre, possède suie valeur thérapeutique comme antibiotique et se prête pour la prépa ration d'autres composés ayant une activité antibiotique.
Exemple <I>1:</I> On prépare une suspension de 1 @@ de dl-W- l-p-nitroplién -#-1=?-aminopropane-1.,3-diol dans <B>15</B> e1113 d'acétate d'éthyle et on la refroidit à 0<B>'</B> C. On ajoute ensuite, pendant l-0 minutes et tout en brassait fortement., 0,5 rma de chlorure de fluoroacétyle en plusieurs petites portions.
On laisse reposer le mélan-e réae- tionnel pendant une demi-heure et on le dilue ensuite avec 50 em3 d'acétate d'éthile. La so lution obtenue est lavée successivement avec de l'acide sulfurique 0,1 n, avec une solution â 2 /o de bicarbonate de soude et ensuite avec de l'eau.
Après avoir séché, on élimine Facé- t.ate d'éthyle par distillation et on purifie le résidu par cristallisation dans du diclilorure d'éthylène ou dans de l'acétate d'éth-le. Le dl - ! -1-p -nitrophényl-2-fhioroacétamiclopro- pane-1,3-diol ainsi obtenu est un corps cris- tallisé ayant un point de fusion de 120 à 1-21 C et répond à la formule

On peut obtenir des résultats légèremen t meilleurs dans le procédé ci-dessus décrit,
si on ajoute un équivalent de bicarbonate de soude au mélange réactionnel.
Exemple On suspend 1 g de (I)-V-1-p-nitrophényl- 2-aminopropane-1,3-diol dans 15 cms d'acétate d'éthyle et on refroidit le mélange à 0 C. On ajoute ensuite, pendant 10 minutes, 0,5 em- de chlorure de fluoroacétyle en petites por tions; pendant cette adjonction, le mélange doit, être brassé fortement. Après avoir laissé reposer ce mélange pendant une demi-heure, on dilue avec 50 em3 d'acétate d'éthyle.
La solution d'acétate d'éthyle est lavée avec de l'acide sulfurique 0,1 n, ensuite avec une solu tion à. 211/o de bicarbonate de soude et finale ment avec de l-'eau. On sèche la couche organi que et on évapore l'acétate d'éthyle dans le vide.
Le résidu est purifié par cristallisation dans du dichlorure d'éthylène ou dans clé l'acétate d'éthyle pour obtenir le (1)-!1'-1-p- nitr ophénpl-2-fluoroa céta niiclopropane-1., 3-zliol sous forme pure; point de fusion 1.1.4-145 C; (a) D dans l'alcool = -+ 9 4. On peut obtenir des résultats légèrement meilleurs si on em ploie dans le procédé décrit un équivalent de bicarbonate de soude (lui est ajouté a.ti mé lange réactionnel.
Process for preparing an aromatic acylamidodiol, nitrated in the nucleus. In the main patent, a process for the preparation of 1'-1-p-tiitrophéiiyl-2-dicliloroacétamidopropane-1,3-diol has been described. The present patent relates to a process for the preparation of another aromatic aeylamidodiol, nitrated in the nucleus.
This process is characterized in that the 1'-1-p-nitrophenyl-2-aminopropane-1,3-diol is reacted with a derivative <fluoroacetic acid, containing at least one FCH2C0- radical, under conditions such that, by attachment of an FCI12CO- radical to the amino group, the 'F-1-p-nitrophenyl-2-fluoroacetamidopropane-1,3-diol is formed. The latter is a new crystallized substance,
whose racemic form melts at <B> 1.20-1210C. </B>
Mention may be made, as derivatives suitable for carrying out said fluoro-ethylation reaction, of the halides, alkyl-esters or anhydride of fluoroacetic acid. When an ester of said acid is employed, the reaction is carried out under anhydrous conditions, preferably in the presence of a small amount of an alkaline catalyst such as sodium methoxide. When employing a halide or anhydride of fluoro-aoetic acid, the reaction can.
be carried out either. under anhydrous conditions, either in the presence of water, and in the presence or absence of an alkaline substance. If the anhydride of the luoroacetic acid is used under anhydrous conditions, it is preferable not to employ an alkaline catalyst.

if one equivalent of sodium bicarbonate is added to the reaction mixture.
Example 1 g of (I) -V-1-p-nitrophenyl-2-aminopropane-1,3-diol is suspended in 15 cms of ethyl acetate and the mixture is cooled to 0 C. The mixture is then added for 10 minutes, 0.5 em of fluoroacetyl chloride in small portions; during this addition, the mixture must be stirred vigorously. After allowing this mixture to stand for half an hour, it is diluted with 50 em3 of ethyl acetate.
The ethyl acetate solution is washed with 0.1 n sulfuric acid, then with a solution of. 211% of baking soda and finally with water. The organic layer is dried and the ethyl acetate evaporated in vacuo.
The residue is purified by crystallization from ethylene dichloride or from ethyl acetate to obtain (1) -! 1'-1-p-nitr ophenpl-2-fluoroa kéta niiclopropane-1., 3- zliol in pure form; melting point 1.1.4-145 C; (a) D in alcohol = - + 9 4.
