Le caractère amphotère de l'amino-acide A et la présence sur l'hydroxy-amine-acide B d'une fonction aminée rend cette réaction directe pratiquement impossible. Il faut, d'une part, libérer le carboxyle de l'amino-acide A en bloquant partiellement ou intégralement sa fonction amine et, d'autre part, bloquer la fonction aminée de l'hydroxy-amino-acide B.
On peut également protéger le carboxyle de B en vue de conférer une plus grande réactivité à l'hydroxyle et d'améliorer les- solubilités. Cette dernière protection du carboxyle est très avantageusement réalisée sous forme d'ester benzylique de B.
Or, la titulaire a maintenant trouvé qu'il est possible de préparer les 0-peptides par une méthode générale, facile à réaliser, ne faisant pas intervenir de réactifs coûteux et permettant, en outre, l'accès direct à des dérivés qui peuvent conduire d'emblée aux N,O- ou 0,0-peptides supérieurs.
Il est en effet possible de condenser sur ces dérivés un nouvel aminoacide ou peptide au niveau du groupement aminé provenant de A pour obtenir le N,O-peptide D, au niveau du groupement aminé issu de B pour préparer le N,O-peptide E ou encore au niveau du carboxyle provenant de B, afin d'aboutir aux N,O- ou 0,0-peptide F.

Les réactions que comporte le procédé selon l'invention ne faisant pas intervenir de racémisation, il est évidemment possible, au départ d'amino-acides ou peptides optiquement actifs, de réaliser la synthèse d'O-peptides optiquement actifs qui possèdent une activité physiologique propre ou conduisent à des dérivés d'importance thérapeutique extrême. Ainsi, au départ de 1'0-glycyl-L-sérine (formule IX du schéma II annexé), on peut préparer l'azasérine, substance récemment décrite, inhibitrice de tumeurs [Am. Soc., 76, 2878-2891 (1954)].
Lors de la mise en oeuvre du procédé selon l'invention, on emploie donc un halogénure d'acide ou un anhydride mixte d'un aminoacide ou d'un peptide dont la ou les fonctions aminées sont mono- benzylées, dibenzylées, tritylées ou acylées et un hydroxy-amino-acide ou hydroxy-peptide dont la ou les fonctions aminées sont monobenzylées, dibenzy- lées, tritylées ou acylées et dont le carboxyle est libre,
estérifié ou salifié. Ces matières premières ex- cellent par leur facilité de préparation et leur grande tendance à la cristallisation; elles sont parfaitement solubles en solvants organiques polaires ou non dans les conditions de leur utilisation.
Après isolement de 1'0-peptide N-substitué formé dans le procédé suivant l'invention, on peut débloquer les diverses fonctions aminées. On peut encore libérer sélectivement une seule ou plusieurs d'entre elles. Ce déblocage partiel se fait surtout avantageusement dans le cas des dérivés benzyl-tritylés, en mettant à profit l'instabilité du reste trityle en milieu acide. Les produits obtenus par déblocage partiel peuvent servir de composés de départ pour des condensations ultérieures avec d'autres aminoacides ou peptides suivant le schéma I.
L'O-peptide N-substitué, isolé après la condensation, peut, après transformation de son groupement carboxylique libre, estérifié ou salifié, en halogénure d'acide ou anhydride mixte, être amené à réagir sur la fonction hydroxylée ou aminée d'un aminoacide ou peptide convenablement substitué et donne ainsi naissance aux 0,N-peptides D et E ou 0,0-peptides F.
La condensation de l'halogénure d'acide ou de l'anhydride mixte d'un aminoacide ou peptide monobenzylé, dibenzylé ou tritylé sur l'hydroxyle libre d'un second aminoacide ou peptide mono- benzylé, dibenzylé, tritylé ou acylé, s'effectue convenablement dans un solvant organique anhydre tel que le chloroforme, le tétrahydrofuranne ou encore le diméthylformamide. Ainsi, dans le cas d'un anhydride mixte éthylcarbonique, il est recommandable d'opérer dans le solvant même qui a servi à la préparation de ce dernier.
La réaction s'effectue convenablement en présence d'un agent de condensation basique, tel que la triéthylamine, à une température comprise entre -10 et -I- 500 C, de préférence voisine de -I- 300 C.
Pour isoler le produit de la réaction, on peut reprendre le mélange réactionnel dans un solvant non miscible à l'eau, effectuer un lavage acide pour éliminer l'excédent d'agent de condensation basique, laver à l'eau pour enlever l'acide, sécher et évaporer à sec sous vide. On obtient ainsi avec un rendement quantitatif l'O-peptide N-substitué, suffisamment pur pour la suite des opérations.
Si l'on a affaire à un milieu réactionnel miscible à l'eau, on peut simplement-procéder à l'isolement de 1'0-peptide N-subs- titu6 par addition d'eau, la précipitation étant immé- diate.
Si le 0-peptide obtenu dans la condensation est N-tritylé, on peut détrityler le produit brut de la condensation par un hydracide, tel que l'acide chlorhydrique, en solution aqueuse et en présence d'un solvant tel que l'acétone ou l'acide acétique à une température de préférence voisine de 200 C. La détri- tylation peut aussi s'effectuer simplement au reflux de l'acide acétique dilué.
On peut isoler le 0-peptide détritylé, suivant les méthodes classiques, après séparation du triphénylcarbinol. La débenzylation peut

s'effectuer par hydrogénolyse en présence de charbon palladié. Il est à remarquer que cette dernière réaction, conduite à basse température, peut provoquer, comme on le sait, une monodébenzylation sélective, les dérivés N-dibenzylés conduisant aux composés N-monobenzylés correspondants.
Les dérivés obtenus selon le procédé de l'invention peuvent servir à la préparation de polypeptides supérieurs en chaîne droite ou cycliques, le déblocage sélectif d'un ou de plusieurs groupements actifs de la molécule visant à permettre la condensation d'un ou de plusieurs aminoacides ou peptides ou encore la cyclisation du polypeptide obtenu. La condensation sur l'hydroxyle de l'amino-acide B ou sur le peptide correspondant peut parfois conduire à de meilleurs rendements si ce dernier est utilisé à l'état d'ester ou de sel.
Les N-benzyl-amino-acides ou peptides utilisés sont obtenus selon le brevet suisse No 336838 ou les indications données dans l'article de L. Velluz et collaborateurs, paru dans Bull. Soc. Chim. France (1955) 201-204.
Les N-trityl-amino-acides ou peptides sont obtenus selon le brevet suisse N0338197. Ils trouvent ici leur première application dans la synthèse des 0-peptides.
Dans les exemples suivants, les chiffres romains se trouvant entre parenthèses se rapportent aux formules chimiques que l'on trouvera sur les schémas en annexe. Exemple 1 Préparation de la O-glycyl-DL-sérine (IX) par l'intermédiaire de la O-(N,N-dibenzyl-glycyl)- DL-N,N-dibenzyl-sérine (II) a) Action du chlorhydrate du chlorure de N,N-dibenzyl-glycyle sur la DL- N,N-dibenzyl-sérine On dissout 2,85 g de DL-N,N-dibenzyl-sérine préparée comme décrit dans l'article précité dans 30 cm3 de chloroforme contenant 2,
5 cm3 de tri- éthylamine. On refroidit à 01, C et ajoute progressivement 1,6 g de chlorhydrate de chlorure de N,N- dibenzyl-glycyle préparé comme décrit dans le brevet suisse No 331208, puis 2,5 cm3 de triéthylamine et, à nouveau, 1,6 g de chlorhydrate de chlorure de N,N-dibenzyl-glycyle. Après retour à la température ambiante, on agite 15 minutes, lave à l'eau, à l'acide chlorhydrique 5 N, à l'eau, sèche sur sulfate de magnésium et évapore à sec sous vide.
On obtient ainsi le chlorhydrate brut de la O-(N,N-dibenzyl- glycyl)-DL-N,N-dibenzyl-sérine avec un rendement quantitatif. Ce produit est amorphe. b) Dihydrogénolyse du chlorhydrate de la O-(N,N-dibenzyl-glycyl)-DL- N,N-dibenzyl-sérine On dissout 10 g de chlorhydrate de la O-(N,N- dibenzyl - glycyl) - DL - N,N - dibenzyl - sérine dans 200 ce d'alcool à 75 0/0,
ajoute 3 g de charbon palladié à 10 % de palladium et hydrogène pendant 20 minutes à la température ambiante (absorption de 2 molécules d'hydrogène), on filtre, évapore à sec sous vide, reprend le résidu dans un peu d'eau chaude et obtient par addition d'acétone le chlor- hydrate cristallisé de la O-(N-benzyl-glycyl)-DL-N- benzyl-sérine (III) avec un rendement presque quantitatif, F = 195 - 205o C (sur bloc).

Tétrahydrogénolyse du chlorhydrate de la O-(N,N-dibenzyl-glycyl)-N,N- dibenzyl-sérine On dissout 10 g de chlorhydrate de la O-(N,N- dibenzyl - glycyl) - DL - N,N - dibenzyl - sérine dans 200 cm3 d'alcool à 75 0/0, ajoute 3 g de charbon pal- ladié à 10 % de palladium, hydrogène pendant 20 minutes à la température ambiante (absorption de 2 molécules d'hydrogène)
puis pendant 45 minutes à la température de 50o C (absorption de 2 nouvelles molécules d'hydrogène). On filtre, concentre à petit volume sous vide et ajoute de l'alcool absolu jusqu'à trouble persistant. Le chlorhydrate de la O-glycyl- DL-sérine cristallise sous forme de petites aiguilles incolores qu'on obtient ainsi avec un rendement de 70 % par rapport à la DL-N,N-dibenzyl-sérine ini- tiale. Le produit est recristallisé par dissolution à froid dans 3 volumes d'eau et addition de 13 volumes d'alcool, F (tube capillaire) = 170 - 1750 C.
Il fournit une intense coloration violette avec la nin- hydrine et possède un spectre infrarouge identique à celui décrit (Am. Soc. 76, 2887 (1954)). Analyse: C5H1104N,,Cl = 198,6 Calculé C 30,2 % H 5,6 % O 32,2 % N 14,1 % Cl 17,8 0/0 Trouvé C 30,5 % H 5,9 % O 32,
1 % N 13,8 % Cl 17,3 0/0 Exemple 2 Action de l'anhydride mixte éthylcarbonique de la N,N-dibenzyl-glycine sur la DL-N,N-dibenzyl-sérine On dissout à chaud 2,55 g de N,N-dibenzyl- glycine dans 30 cm3 de chloroforme contenant 2 cm-3 de triéthylamine, refroidit à - 100 C, introduit 1,1 em3 de chloroformiate d'éthyle et abandonne 30 minutes à 00 C.
On ajoute alors une solution de 2,85 g de DL-N,N-dibenzyl-sérine et de 1,5 cm3 de triéthylamine dans 30 cm3 de chloroforme et laisse reposer une nuit à 30,, C. On lave

à l'eau, à l'acide chlorhydrique 5 N, à l'eau, sèche sur sulfate de magnésium et évapore à sec sous vide.
On obtient ainsi le chlorhydrate de la O-(N,N- dibenzyl-glycyl)-DL-N,N-dibenzyl-sérine identique au produit obtenu suivant l'exemple la et qui fournit par hydrogénolyse, comme décrit dans l'exemple 1 b et c, les chlorhydrates de la O-(N-benzyl-glycyl)- DL-N-benzyl-sérine, F = 195 - 2050 C (sur bloc), et de la O-glycyl-DLrsérine, F = 170 - 1750 C (en tube capillaire).
La preuve de la constitution du produit est aisément démontrée par le fait que la saponification alcaline du chlorhydrate de la O-(N,N-di- benzyl-glycyl)-DL-N,N-dibenzyl-sérine par traitement de ce produit à l'ébullition de la potasse méthanoli- que, dilution par l'eau et acidification, conduit quantitativement à un mélange de N,N-dibenzyl-glycine et de DL-N,N-dibenzyl-sérine que l'on peut aisément séparer, le second des constituants étant soluble dans l'eau chaude alors que le premier ne l'est pas.
Exemple 3 Préparation de la O-glycyl-DL-sérine (IX) par l'intermédiaire de la O-(N-trityl-glycyl)- DL-N-trityl-sérine (IV) a) Action de l'anhydride mixte éthyl- carbonique de la N - trityl - glycine sur la DL-N-trityl-sérine On dissout à chaud 3,17 g de N-trityl-glycine dans 30 cm2 de chloroforme contenant 2 cm0 de triéthylamine, refroidit à -100C, introduit 1 cm3 de chloroformiate d'éthyle et abandonne 45 minutes à 0 C.
On ajoute alors une solution de 3,5 g de DL-N-trityl-sérine et de 1,5 cm3 de triéthylamine dans 30 cm3 de chloroforme et laisse reposer une nuit à 301) C. On lave à l'eau, à l'acide chlorhydrique N, puis avec une solution saturée d'acétate de sodium et enfin à l'eau, sèche sur sulfate de magnésium et évapore à sec sous vide. On obtient ainsi la O-(N-trityl-glycyl)-DL-N-trityl-sérine amorphe brute avec un rendement quantitatif.
La preuve de la constitution du produit est démontrée par le fait qu'il est aisément saponifié à l'ébullition de la potasse méthanolique pour fournir un mélange de N-trityl- glycine et de DL-N-trityl-sérine. b) Détritylation de la O-(N-trityl-glycyl)- DL-N-trityl-sérine (IV) On dissout 4 g de O-(N-trityl-glycyl)-DL-N-tri- tyl-sérine (IV) brute dans 10 cm2 d'acétone, ajoute à la solution 3 cmJ d'acide chlorhydrique 5 N et abandonne une nuit à la température ambiante.
On introduit 3 ce d'eau, chasse l'acétone sous vide et essore le triphénylcarbinol qui a cristallisé. On ajuste la solution aqueuse résultante à pH 3 par la triéthylamine, concentre sous vide et précipite par adjonction d'alcool le chlorhydrate d'O-glycyl-DL- sérine qu'on obtient ainsi après essorage avec un rendement de 75 1% par rapport à la DL-N-trityl- sérine mise initialement en jeu.
Le produit est en tous points identique au chlorhydrate d'O-glycyl- DL-sérine obtenu selon l'exemple 1c ; F = 170 - 1750 C (en tube capillaire).
c) Détritylation de la O-(N-trityl-glycyl)- DL-N-trityl-sérine (IV) On dissout 2 g de O-(N-trityl-glycyl)-DL-N-tri- tyl-sérine brute dans 4 cors d'acide acétique, ajoute 1 cm3 d'acide chlorhydrique concentré et abandonne 30 minutes à la température ambiante. On dilue par 10 cm-' d'eau, essore le triphénylcarbinol et concentre la solution résultante sous vide à froid.
On ajuste à pH 3 par la triéthylamme et sépare, comme indiqué en b, 0,33 g (66 1% par rapport à la DL-N- trityl-sérine mise initialement en jeu) de chlorhydrate de O-glycyl-DL-sérine, F = 170 - 1750 C (en tube capillaire).
On peut également détrityler au reflux de l'acide acétique aqueux à 50 0/0 ; les rendements sont identiques. Exemple 4 Préparation de la O-(N,N-dibenzyl-glycyl)- DL-sérine (IV) par l'intermédiaire de la O-(N,N-dibenzyl- glycyl)-DL-N-trityl-sérine (V) a) Action de l'anhydride mixte éthyl- carbonique de la N,N - dibenzyl - glycine sur la DL-N-trityl-sérine On dissout à chaud 5,1 g de N,N-dibenzyl-gly- cine dans 60 ce de chloroforme contenant 4 cm3 de triéthylamine, refroidit à - 5o C, introduit 2,
1 cm3 de chloroformiate d'éthyle et abandonne 1 heure à 00 C. On ajoute alors une solution de 7 g de DL-N-trityl-sérine et de 3 crn3 de triéthylamine dans 60 cm3 de chloroforme, et laisse reposer une nuit à 300 C.
On lave à l'acide chlorhydrique N, à l'eau, puis avec une solution saturée d'acétate de sodium et enfin à l'eau, sèche sur sulfate de magnésium et évapore à sec sous vide. On obtient ainsi la O-(N,N-dibenzyl-glycyl)-DL-N-trityl-sérine amorphe brute avec un rendement quantitatif. \ b) Détritylation de la O-(N,N-dibenzyl- glycyl)-DL-N-trityl-sérine (V) On dissout 12g de O-(N,N-dibenzyl-glycyl)-DL- N-trityl-sérine dans 30 cm3 d'acétone, ajoute à la solution 6 cm3 d'acide chlorhydrique 5 N et 6 em3 d'eau, puis agite pendant 1 heure.
On dilue par 20 cm3 d'eau, chasse l'acétone sous vide et essore le triphényl-carbinol qui a cristallisé. Le filtrat aqueux est additionné d'une solution saturée d'acétate de sodium jusqu'à fin de précipitation. On es- sore, sèche à 60o C et obtient ainsi 5,5 g (80 % par rapport à la DL-N-trityl-sérine mise initialement en jeu) de O-(N,N-dibenzyl-glycyl)-DL-sérine, F = 170- 1720 (sur bloc).
Ce produit qui est nouveau se présente sous forme d'aiguilles incolores peu solubles dans l'eau, solubles dans l'alcool et l'acétone.

b) Détritylation de la O-(N-trityl-glycyl)- DL-N,N-dibenzyl-sérine (VII) On détrityle la O-(N-trityl-glycyl)-DL-N,N-di- benzyl sérine en suivant le mode opératoire indiqué à l'exemple 4b pour la détritylation de la O-(N,N- dibenzyl-glycyl)-DL-N-trityl-sérine et obtient, après purification par dissolution dans l'acide chlorhydrique N, neutralisation à pH 7, essorage et séchage, la O-glycyl-DLN,N-dibenzyl-sérine avec un rendement d'environ 40'0/() par rapport à la DL-N,

Velluz et coll., dans 30 cm3 de chloroforme contenant 2,5 cm3 de triéthyl-amine. On refroidit à 0o C et ajoute 1,6 g de chlorhydrate de chlorure de N,N- dibenzyl-glycyle puis 2,5 em3 de triéthyl-amine et, à nouveau, 1,6 g de chlorhydrate du chlorure précité. Après retour à la température ambiante, on traite comme indiqué à l'exemple la pour obtenir le chlorhydrate brut amorphe de la O-(N,N-dibenzyl-glycyl)- DL - N,N - dibenzyl - thréonine avec un rendement quantitatif.
On prépare la base libre correspondante par traitement d'une solution chloroformique du chlorhydrate par une solution d'acétate de sodium, séchage sur sulfate de magnésium et évaporation à sec sous vide.
Ce produit est amorphe. b) Hydrogénolyse de la O-(N,N-diberazyl- glycyl)-DL-N,N-dibenzyl-thréonine (X) On hydrogénolyse, à température ambiante, 10 g de O-(N,N-dibenzyl-glycyl)-DL-N,N-dibenzyl-thréo- nine dans 125 cm3 d'alcool à 80'% en présence de 3 g de charbon palladié à 10:
% de palladium et de 3,6 cm3 d'acide chlorhydrique 5 N et isole, selon le mode opératoire de l'exemple lb, le chlorhydrate de la O-(N-benzyl-glycyl)-DL-N-benzyl-thréonine (XI) avec un rendement de 80 %. Le produit qui est nou- veau se présente sous forme d'aiguilles incolores peu solubles dans l'eau, l'alcool, l'éther et l'acétone,
