Procédé de préparation de la 6,17a-diméthyl-4,6-pregnadiène-3,20-dione La présente invention a pour objet un procédé de préparation de la 6,17a-diméthyl-4,6-prégnadiène-3,20- dione (VI), un agent progestinogène actif par voie orale et décrit par R. Deghenghi et R. Craudry dans J.Am. Chem. Soc., vol. 83, p. 4668 (1961).
La matière de départ du procédé selon l'invention est un 3p@hyâroxy-17a-méthyl-stéroïde de formule I.

(obtenue comme décrit par Plattner et colt. dans Helv. Chim. Acte,, vol. 32, p. 270 (1949) ) dans 52 ml d'acide formique. On refroi dit la solution et on ajoute 8,7 ml (Feau oxygénée à 30 0/0.
On maintient la solution à la température ordi- naire pendant 4 h, puis on ajoute de l'eau. On dissout le précipité dans 60 ml de méthanol chaud et on neu tralise la solution avec une solution méthanolique d'hydroxyde de potassium à 101%. Par dilution avec de l'eau, on obtient un solide cristallin, qui,
après une recristallisation dans du méthanol aqueux, fond à 278 280o C (II).
<I>B)</I> 3p-acétoxy 5a-hydroxy-17a-méthyl-pregnane- 6,20-dione.
On dissout la 3@,Sa:,613-tcihydioxy-,17-méUhylr-pre- gnan-20-one (Il), préparée ci-dessus dans<B>360</B> ml de méthanol et 360 ml d'éther et on .ajoute 220 ml d'eau à la solution.
Ensuite, on agite la solution et on ajoute 10 g de N-bromo-sucainimide. Lorsque ce dernier s'est dissous, on laisse reposer le mélange réactionnel dans une armoire frigorifique pendant 2 h.
On ajoute une faible quantité d'une solution saturée de bicarbonate de sodium pour éliminer la couleur orange de la solu tion. On chasse l'éther de la solution sous vide à la température ordinaire. Par dilution avec de l'eau, on obtient un solide cristallin: (III, R = H).
On dissout ce solide dans 20 ml de pyridine et 10 ml d'anhydride acétique et on chauffe 1e, solution .au bain-marie pendant 30 mn.
On dilue la solution refroi die avec de l'eau, ce qui précipite un solide, que l'on cristallise dans de l'éthanal aqueux chaud. On obtient ainsi le monoacétate (III, R = Ac).

On dissout 3,7 g de 3p, 5a-dihydroxy-17a-méthyl pregnane-6,20-dione (III, R = H) dans 90 ml de ben zène sec et 90 ml de tétrahydrofuranne sec.
On agite cette solution et on la refroidit dans un bain d'eau et de glace, tout en ajoutant 37 ml d'une solution éthé rée de bromure de méthylmagnésium 3 ZVI. Après une demi-heure ou davantage,
on ajoute 40 ml de tétrahy drofuranne et on agite la solution pendant 18 h à la température ordinaire.
On verse le mélange réactionnel dans 150 ml d'une solution de chlorure d'ammonium saturée et agitée, et on sépare la. -couche organique. On extrait la solution aqueuse avec de l'acétate d'éthyle et on lave les extraits réunis avec une solution de chlorure d'ammonium et avec de l'eau.
Après séchage de la solution organique sur du sulfate de magnésium, on chasse le solvant et on recristallise le résidu dans de l'acétone aqueuse, obtenant une substance fondant à 238-240o C (@. <I>D)</I> 6,17a-diméthyl-4,6-pregnadiène-3,20-dione On dissout 513 g de 3P,5a,6s-trihydroxy-6a,,17a- diméthyl-pregnan-20-one (IV)
dans 250 ml d'acétone purifiée et on refroidit la solution dans un bain d'eau et de glace.
A la solution agitée énergiquement, on ajoute un excès d'une solution d'acide chromique 8 N et on agite le mélange réactionnel pendant 4 mn après la fin de l'addition de l'acide chromique. Ensuite,
on verse le mélange réactionnel dans 400 ml d'eau et an extrait le mélange à l'éther. On lave les extraits réunis avec une solution de bicarbonate de sodium et avec de l'eau et on les sèche sur du sulfate de magnésium. L'élimination du solvant sous pression réduite laisse un semi-solide jaunâtre (V).
E) On dissout une partie de ce résidu (444 mg) dans 20 ml d'éthanol absolu et on ajoute quelques goutttes d'acide chlorhydrique concentré. On chauffe la solution au bain-marie pendant 3/4 d'heure, on la refroidit, on la dilue avec de l'eau et la traite de manière usuelle par extraction à l'éther.
On obtient une gomme jaunâ- tre dont le spectre ultra violet indique la présence d'environ 92 11/o d'une 44@6-cétone (az91 = 19,400), et d'environ 7 @/o d'une cétone a,(3-non saturée.
Par chro- matographie de cette matière sur de l'alumine, on obtient une substance dont le spectre I.R. est identique à celui de la 6,17a-diméthyl-4,6 prégnadiène-3,20-dione (VI).

The cooled solution is diluted with water to precipitate a solid, which is crystallized from hot aqueous ethanal. The monoacetate (III, R = Ac) is thus obtained.

3.7 g of 3p, 5a-dihydroxy-17a-methyl pregnane-6,20-dione (III, R = H) are dissolved in 90 ml of dry ben zene and 90 ml of dry tetrahydrofuran.
This solution is stirred and cooled in an ice water bath, while adding 37 ml of an ethereal solution of 3 ZVI methylmagnesium bromide. After half an hour or more,
40 ml of tetrahydrofuran are added and the solution is stirred for 18 h at room temperature.
The reaction mixture is poured into 150 ml of a saturated and stirred solution of ammonium chloride, and the mixture is separated. -organic layer. The aqueous solution was extracted with ethyl acetate and the combined extracts washed with ammonium chloride solution and with water.
After drying the organic solution over magnesium sulfate, the solvent is removed and the residue is recrystallized from aqueous acetone, obtaining a substance melting at 238-240o C (@. <I> D) </I> 6 , 17a-dimethyl-4,6-pregnadiene-3,20-dione 513 g of 3P, 5a, 6s-trihydroxy-6a ,, 17a-dimethyl-pregnan-20-one (IV) are dissolved
in 250 ml of purified acetone and the solution is cooled in an ice-water bath.
To the vigorously stirred solution, an excess of 8N chromic acid solution is added and the reaction mixture is stirred for 4 min after the addition of chromic acid is complete. Then,
the reaction mixture is poured into 400 ml of water and the mixture is extracted with ether. The combined extracts were washed with sodium bicarbonate solution and with water and dried over magnesium sulfate. Removal of the solvent under reduced pressure leaves a yellowish semi-solid (V).
E) Part of this residue (444 mg) is dissolved in 20 ml of absolute ethanol and a few drops of concentrated hydrochloric acid are added. The solution is heated in a water bath for 3/4 of an hour, cooled, diluted with water and treated in the usual way by extraction with ether.
A yellowish gum is obtained whose ultraviolet spectrum indicates the presence of about 92 11 / o of a 44 @ 6-ketone (az91 = 19.400), and of about 7% of an a ketone, (3-unsaturated.
Chromatography of this material on alumina gives a substance which has an I.R. spectrum identical to that of 6,17α-dimethyl-4,6 pregnadiene-3,20-dione (VI).
