La présente invention concerne un procédé de pré- par&tion de nouveaux agents progestatifs et plus particuliè- rement les 17a-alkyuyl-4,6-diène-38,178-dicle des séries de l'androstane et de l'oestrane en leufs esters de formule

dans laquelle R est un atome d'hydrogène ou un radical mé- thyle, et R' et R" peuvent être un atome d'hydrogène ou un groupe alkanoyle comprenant de 1 à 6 atomes de carbone.
Les groupes alkanoyles envisages pour R' et R" sont par exemple les groupes formyle, acétyle, propionyle, butyryle valéryle, caproyle et leurs isomères ramifiés, Les groupes alkynyle types comprennent .es groupes éthynyle, butynyle, propynyle et hexynyle.
Les composés de formule (I), obtenus par le pro- cédé de l'invention, sont utiles en raison de leurs proprié- tés pharmacologiques de valeur. @
Ce sont par exemple des agents ; progestatif s qui ne présentent pas les effets secondaire d'une activité ana- bolisante et androgénique, contrairement aux compositions connues jusqu'ici.

Conformément au procédé de la présente invention, un prépare les composés de formule (I) en mélangeant un composé 3-oxo correspondant de formule:

On conduit la réduction des composés de formule (II)avec un &gent qui réduit sélectivement un groupe @ sans réduire les liai sons insaturées carbone-carbone situées entre les atomes de carbone 4 et 5, et () et 7 -ou contenues dans le substituant 17a-alkyuyle.
Les agents réducteurs dé- rivés des métaux du groupe (III), de la classification pério- dique des éléments sont spécialement utiles et parmi eux les agents réducteurs dérivés des métaux du groupe III ayant un

poids moléculaire inférieur à 28, par exemple, les hydrures des métaux alcalins et de l'aluminium et les borohydrurea des métaux alcalins tels que l'hydrure de lithium-aluminium, l'hy- drure de lithium tri-(butoxy-tert) aluminium, et le borohy- drure de sodium, et leu alcoxydes d'aluminium tel que l'iso- propoxyde d'aluminium.
Dans le cas de la préparation des 17ss- alkanoate. c'est-à-dire des composées de formule (I) et (III) pour lesquels R" de la formule (II) est} un groupe alkanoyle on préfère utiliser comme agent réducteur l'hydrure de lithium tri-(butoxy-tertiaire) aluminium dans le but de prévenir le clivage du groupe ester.
On conduit l'acylation des composés de formule (II) avec un agent couramment utilisé pour l'estérification des groupes 3-hydroxy deu stéroides. Le agents préfères pour ladite acylation comprennent les halogénures des acides alka- noiques et les anhydrides des acides alkanoiques. On conduit de préférence le traitement avec un anhydride ou un halogénu- t re d'acide alkanoique en présence d'un accetaur d'acide, par exemple d'une base organique telle que les aminés tertio aires ( par exemple la pyridine) ou des bases minérales tel- les que les sels basiques (par exemple du carbonate de potas- sium). D'autres agents convenables pour la production des esters acétiques sont le cétène et l'acétate d'isopropényle.
Quand R" de la formule (II) est un atome d'hydrogène, l'uti- lisation de l'agent acylant à la température ambiante four- nit le 3-monoester, tandis que l'utilisation d'au moins 2 équivalents de l'agent acylant i des températures élevées fournit le 3,17-diester.
On peut alternativement préparer les composés de fornule (III) dans laquelle R" est un radical alkanoyle, par cliavge sélectif à la position 3 du 3,.17-diester, conve- nablement avec le borohydrure de sodium ou un ractif équi- valent .

On prépare les substances de départ de formule (II) à partir des composés 3-cétz-A4 correspondants. La dé- stydruation de ces composés #4, convenablement par traite- ment avec un caloranile, fournit les substances #4,6 corres- pondantes. On obtient les 17ss-alka oates correspondants en chauffant lesdites #4, '*;-cétones avec un agent acylant tel qu'un anhydride d'acide alkancique inférieur dans la pyri- dine.
Dans les exemples suivants les quantités de substan- ces sont exprimées en parties par poids, sauf indication
Contraire, les résultats obtenus pour le pouvoir rotatoire et les spectres infra-rouges se réfèrent à des déterminations faites'en chloroforme. Ces exemples sont fournis à titre d'illustration seulement et ne présentent aucun caractère limitatif quant au cadre et à l'esprit de l'invention.
EXEMPLE 1 A une solution d'une partie de la 17a-éthynyl-
17ss-hydroxyandrosta-4,6-diène-3-one dans 50 parties de tétra- hydrofurane, on ajoute 2 parties dhydrure de lithium tri-(bu- toxy tertiaire) aluminium et on agite le mélange résultant à température ambiante pendant 1 heure environ puis on l'aci- difie avec de l'acide acétique aqueux dilué. On extrait ce mélange aqueux, acidifié, avec du chloroforme et on sépare la couche de chloroforme, on la lave successivement à l'eau et au bicarbonate de codium aqueux, on la sèche sur sulfate de sodium anhydre et on la fait évaporer jusqu'à siccité.
On cristallise le résidu résultant à partir d'un mélange d'éther et de méthanol en obtenant le 17[alpha]-éthynylandrosta-
4,0-diène-3ss,17ss-diol qui fond à 228-230 C environ et montre des maxima dans le spectre ultra-violet à environ 232, 238, et 246 millimicrons avec des coefficients d'extinction molé- culaire d'environ 21.000, 247700 et 14.500 respectivement.

On agite le mélange et on le chauffe à 35 C pendant 45 minutes, puis on ajoute une quantité d'acétone suffisante pour décom- poser tout l'hydrure de lithium aluminium non entré en réac- ticn.
On ajoute de l'eau et on sépare 14 couche organique on la lava successivement avec de l'eau :et avec une solution saturée de chlorure de sodium. Apres séchage sur sulfate de soude anhydre et filtration, on élimine;le solvant sous vide.
On applique le résidu dans une solution à 10% d'acétate d'é- thyle en benzène à une colonne de chromatographie contenant 760 parties de gel de silice. On lave lecolonne au benzène puis avec des solutions à 5, 10 et 15 d'acétate d'éthyle en benzène.

The starting substances of formula (II) are prepared from the corresponding 3-ketz-A4 compounds. Dehydration of these # 4 compounds, suitably by treatment with caloranil, provides the corresponding # 4.6 compounds. The corresponding 17ss-alka oates are obtained by heating said # 4, '*; - ketones with an acylating agent such as lower alkancic acid anhydride in pyridine.
