The object of the present invention is a process for preparing aldehyde acids, of general formula III:
EMI1.1
in which a and b represent: two carbon atoms, originally neighboring and coming from a particular opening of the nuclei A, B, C or D of a sterol- of the normal or allo series whose D ring can belong to the homo series as well as to the normal series (pentagonal D nucleus). These acids are of interest as intermediates in the synthesis of physiologically active steroid derivatives and also form part of the invention as new industrial products and not as pharmaceuticals or remedies.
'In its French patent application entitled "Process for the preparation of new derivatives of ketosteroids" of June 15, 1956 and in the two certificates of addition to said application of July 19, 1956 and January 22, 1957, the applicant Company has describes the preparation of germinated bis-hydroperoxides, of f, general formula:
EMI1.2
as well as their esters, said bis-hydroperoxides obtainable by the action of anhydrous hydrogen peroxide in an anhydrous solvent such as tertiary butyl alcohol, ether and ethyl acetate on a ketosteroid.
It has now been found that if one subjects a bis-hydroperoxide, .I, resulting from the transformation of an unsaturated [alpha] ss steroid ketone to the action of acylating agents and in particular of anhydrides of. acids in the presence of pyridinic bases such as pyridine or methylethylpyridine, instead of the expected diester, the enolic lactone, II, is obtained by insertion of an oxygen between the starting carbonyl and the double bond. By saponification, these lac- -tones open to the sought aldehyde III acid.
The structure of lactone II and of aldehydic acid III is confirmed by the oxidation of III to the alkaline potassium permangana- te which furnishes the diacid, IV, and on the other hand, by obtaining a ketonic acid, V, which results from the parmanganic oxidative cleavage of the double bond of II with removal of the 0 <carbon from the original ring.
In order to obtain the aldehyde acids, III, according to the invention, an acid anhydride such as benzoic, acetic, propionic or succinic anhydride is reacted at low temperature, preferably in the vicinity of 0, in the presence of. a pyridine base and isolates the desired lactone II directly by pouring the reaction mixture into water. After possible recrystallization, the lactone II is saponified to the desired aldehyde III acid by the action of sodium hydroxide or hydroalcoholic or hydromthanolic potassium hydroxide. This saponification can also be carried out by alkali metal alcoholates. When the saponification is complete, the acid is acidified and the desired acid III is separated, which is easy to purify by crystallization.
The following examples illustrate the invention without, however, limiting it. It is in particular possible to use as starting products the bis-hydroperoxides of other steroid ketones than those mentioned, to use other acid anhydrides than those indicated, to vary the nature of the pyridine base or the temperature without moving away for this. within the scope of the invention. The points of fusion are instantaneous points of fusion determined on a Maquenne block.
