Procédé de préparation de sels de l'a-L-glutamyl-chollne
L'invention a pour objet un procédé de préparation de l' -L-glutamyl-choline et de ses sels. On a décrit dans le brevet américain No 2776923 un procédé d'obtention de la L-glutamyl-choline et de son chlorhydrate par réaction de l'anhydride N-carbohenzoxy-
L-glutamique avec la choline ou le chlorhydrate de choline. On obtient ainsi un mélange des isomères a et y de la L-glutamyl-choline ou de son chlorhydrate
N-carbobenzoxylés. L'hydrogénolyse en présence de platine ou de palladium fournit le mélange des isomères a et y de la L-glutamyl-choline ou de son chlorhydrate.
On ne connaissait pas de procédé permettant l'obtention préférentielle de l'a-L-glutamyl-choline et de ses sels et la présente invention se propose de remédier à cette lacune.
Suivant le procédé de la présente invention, on fait réagir la monochlorhydrine du glycol sur l'acide
L-glutamique en présence d'acide chlorhydrique gazeux, traite le chlorhydrate du L-glutamate de di-(ss'- chloroéthyle) par une base appropriée, la soude par exemple, obtient le L-glutamate de di-(ss'-chloro- éthyle), fait réagir sur ce dernier le chlorure de trityle en présence d'un agent fixant les acides par exemple la triéthylamine, reprend le N-trityl-L-glutamate de di-(ss'-chloroéthyle) obtenu dans le méthanol en présence de méthylate de sodium pour obtenir le N-trityl-L-glutamate d' -(ss'-chloroéthyle) et y-méthyle, saponifie ce dernier, par exemple par la soude en solution dioxanique, pour obtenir le N-trityl-L-glutamate d'a-(ss'-chloroéthyle),
qu'on traite ce dernier en vase clos par l'iodure de sodium et la triméthylamine en acétone, puis détrityle l'iodure de N-trityl-L-glutamate d'a-choline résultant, par exemple par l'acide acétique aqueux à 50% pour obtenir l'iodure d'a-L-glutamyl- choline. On peut transformer celui-ci en nitrate par l'action du nitrate d'argent en solution aqueuse ou en tout autre sel par réaction avec un réactif approprié.
Comme indiqué dans le brevet américain
No 2776923 précité, la glutamyl-choline et ses sels sont des produits montrant une activité pharmacodynamique intéressante; on en a préconisé l'emploi dans le traitement de la poliomyélite, de l'hypertension et comme agent promoteur de croissance.
D'après Yoshito Nishizawa, Archives françaises de Pédiatrie, 15, 195-216 (1958), l'ct-L-glutamyl-cho- line présente une activité thérapeutique double de celle de la y-L-giutamyl-choline.
Les exemples suivants illustrent l'invention. Les formules développées des différents composés, intervenant dans le procédé suivant l'invention, figurent sur le schéma annexé.
Exemple a) Chlorhydrate du L-glutamate de di-(B'-chloro-
éthyle) (II).
On introduit 20 g d'acide L-glutamique (I) dans 200 cm3 de monochlorhydrine du glycol, chauffe au bain d'huile à 1050 C + 5 et fait passer dans le mélange de l'acide chlorhydrique gazeux en agitant jusqu'à saturation et dissolution complète, ce qui demande environ 30 minutes.
On abandonne encore 3 heures à la même tempé rature, distille au bain-marie sous pression réduite, environ 150 cm3 de monochlorhydrine et ajoute 250 cm3 d'éther. On glace, essore, lave à l'éther par empâtage et sèche à 500 C pour obtenir 39 g (93%) de chlorhydrate du L-glutamate de di-(0'-chloroéthyle) (11), F. = 115-1170 C, [a]2u0 DO = + 16 + 0, 5 (c = 2%, eau). Ce produit qui n'a pas encore été décrit se présente sous forme de feuillets incolores, solubles dans d'eau glacée et abandonne au réchauffement pendant une nuit à la température ambiante.
On évapore sous vide la majeure partie des solvants, ajoute 50 cm3 d'eau et termine la distillation au dioxane. On ajoute 11 cm3 d'acide chlorhydrique
N en présence de 40 cm3 de chloroforme, agite puis décante, extrait par deux fois 5 cm3 de chloroforme, réunit les solutions chloroformiques, les lave avec 10 cm3 d'eau, sèche sur sulfate de magnésium et évapore à sec sous vide. On obtient ainsi, avec un rendement quantitatif, le N-trityl-L-glutamate d'a-(f\'- chloroéthyle) (IV: R = H).
Cette substance qui se présente sous forme d'une huile n'a pas encore été décrite; elle peut être directement utilisée pour la réaction décrite sous f).
Le N-trityl-L-glutamate d'a-('-chloroéthyle) peut être caractérisé sous forme de son sel de diéthylamine, comme suit:
L'huile brute obtenue selon e) est reprise dans 10cm3 d'éther; on ajoute 1,1 cm3 de diéthylamine puis, en deux fois, 20 cm3 de cyclohexane, glace pendant deux heures, essore, lave au cyclohexane et essore 4,7 g de sel de diéthylamine du N-trityl-L-gluta- mate d'α-(ss'-chloroéthyle), F. vers 100 C, [α]D20 =
+ 460 + 1 (c = 2%, chloroforme). Ce produit qui est nouveau se présente sous forme de petites aiguilles incolores, solubles dans l'eau, l'alcool, l'acétone, le benzène et le chloroforme, peu solubles dans l'éther et le cyclohexane.

On introduit 16,5 g du sel de diéthylamine du
N-trityl-L-glutamate d'α-(ss'-chloroéthyle) dans 100 cm3 de chloroforme, ajoute 20 cmi d'eau puis, par fraction et en agitant, 30 cm3 d'acide chlorhydrique
N. On décante, extrait par 20 cm3 de chloroforme, réunit les extraits chloroformiques, les lave avec 30 cmS d'eau, sèche sur sulfate de magnésium et évapore à sec sous vide.
On introduit 6 g d'iodure de sodium fondu dans un tube à sceller, puis le N-trityl-L-glutamate d'a-(p'- chloroéthyle) obtenu au paragraphe précédent, en solution dans 20 cm3 d'acétone sèche. On ajoute 12 cm3 de triméthylamine anhydre, scelle le tube et chauffe pendant une nuit à 750 C. On glace, ouvre le tube, évapore à sec sous vide, reprend par 60 cm3 de chloroforme et filtre l'insoluble. La solution filtrée est
additionnée de 20 cm3 d'eau contenant 1 cm3 d'acide
acétique. On acidifie à pH 2 par une solution aqueuse
diluée d'acide iodhydrique, décante, extrait au chloroforme, lave avec 10 cm3 d'eau, sèche la solution chloroformique sur sulfate de magnésium et évapore à sec sous vide. On ajoute 120 cm3 d'éther anhydre,
agite, filtre et sèche sous vide.
On obtient ainsi 10,7 g
d'un produit qu'on pulvérise et traite à nouveau par l'éther comme précédemment pour obtenir 9,8 g
(54%) d'iodure du N-trityl-L-glutamate d'a-choline. l'eau et l'alcool, peu solubles dans l'acétone et le chloroforme, insolubles dans l'éther.

On introduit 31 g de chlorhydrate de glutamate de di-(ss'-chloroéthyle) obtenu selon a) dans 50 cm3 d'eau glacée, ajoute 250 cm3 de chloroforme, 2 gouttes de phénolphtaléine, puis, par fractions, 12,5 cm3 de lessive de soude.
On décante, extrait la solution aqueuse par 25 cm3 de chloroforme, réunit les solutions chloroformiques, lave avec 15 cm3 d'eau et sèche sur sulfate de magnésium. c) N-trityl-L-glutamate de di-(ss'-chloroéthyle)
(1V: R =-CH2-CHS-C1).
On ajoute à la solution chloroformique ci-dessus 30 g de chlorure de trityle et, par portions, 15 cm3 de triéthylamine puis abandonne une nuit à 300 C.
On lave avec 50 cm9 d'eau, un mélange de 50 cm3 d'eau et 45 cm3 d'acide chlorhydrique N et enfin avec 40 cm3 d'eau, sèche sur sulfate de magnésium et évapore à sec sous vide. d) N-trityl-L-glutamate d'a-(ss'-chloroéthyle) et y-mé
thyle (IV : R = - CH3).
On dissout le résidu obtenu ci-dessus dans 50 cm3 de méthanol, évapore à sec sous vide, reprend dans 200 cm3 de méthanol à 350 C, ajoute 2,5 cm3 d'une solution normale de méthylate de sodium dans le méthanol et abandonne pendant 20 minutes à 350 C.
On ajoute encore 1 cm3 de la solution de méthylate de sodium puis, après 40 minutes, de nouveau 1 cm3.
On abandonne 1 heure à 350 C, une heure à la température ambiante, glace, essore, rince au méthanol glacé et sèche à 400 C pour obtenir 30 g de produit qu'on reprend dans 50 cm3 de tétrachlorure de car bone. On glace, filtre l'insoluble, concentre à demivolume, ajoute 50 cm3 de méthanol et distille ce qui reste de tétrachlorure de carbone. On glace, essore, rince au méthanol glacé et sèche à 400 C pour obtenir 29,2 g (63%) de N-trityl-L-glutamate d'a-(ss'-chloro-
éthyle) et y-méthyle, F. = 83-860 C, [l2D0 = + 35,50
+ 0,5 (c = 2%, chloroforme).

On dissout 4,7 g de N-trityl-L-glutamate d'α-(ss'-
chloroéthyle) et y-méthyle (IV R = - CH0), obtenu
selon d), dans 40 cm3 de dioxane, ajoute 10 cm3 de
méthanol et 10,5 cm3 de soude N. On place sur bain g) Iodure d'a-L-glutamyl-choline (V: X = I)
On ajoute à 3 g d'iodure brut du N-trityl-L-glutamate d'a-choline obtenu selon f), 7,2 cm3 d'acide acétique aqueux à 50%, agite et chauffe à 350 C pendant quinze minutes.
On ajoute 18 cm3 d'acétone puis, par portions, 25 cm3 d'acétone. On essore, lave à l'acétone et sèche sous vide pour obtenir 1,35 g (77%) d'iodure d'α-L-glutamyl-choline, F. = 170180 C, [α]D20 = + 13,5 # 1 (c = 2 %, eau).
Ce produit, qui n'a pas encore été décrit, se présente sous forme de petites aiguilles incolores solubles dans l'eau, insolubles dans l'alcool, I'éther, l'acétone et le benzène.

(V:X = NO)
On introduit 1,08 g d'iodure d'a-L-glutamyl-choline obtenu selon g) dans une solution de 0, 510 g de nitrate d'argent dans 4 cm3 d'eau. On filtre, concentre à demi-volume, ajoute 4 cm3 d'alcool et 40 cm3 d'acétone, glace et essore. On recristallise dans 2 cm3 d'eau légèrement alcoolisée, par addition d'acétone pour obtenir 650 mg (75%) de nitrate d'α-L-glutamyl-cho- line, F.
(en tube capillaire) = environ 90 C, [α]D20 = -t 16,5 + 1 (c = 2%, eau).
Ce produit, qui est nouveau, se présente sous forme de petits feuillets incolores, solubles dans l'eau, peu solubles en alcool, insolubles en éther, acétone et benzène.
Process for the preparation of α-L-glutamyl-chollne salts
The subject of the invention is a process for preparing ′ -L-glutamyl-choline and its salts. A process for obtaining L-glutamyl-choline and its hydrochloride by reaction of N-carbohenzoxy anhydride has been described in US Patent No. 2776923.
L-glutamic with choline or choline hydrochloride. A mixture of the α and γ isomers of L-glutamyl-choline or its hydrochloride is thus obtained.
N-carbobenzoxylates. Hydrogenolysis in the presence of platinum or palladium provides the mixture of the α and γ isomers of L-glutamylcholine or its hydrochloride.
There was no known process allowing the preferential production of α-L-glutamyl-choline and its salts and the present invention proposes to remedy this shortcoming.
According to the process of the present invention, the monochlorohydrin of the glycol is reacted with the acid
L-glutamic in the presence of gaseous hydrochloric acid, treats the hydrochloride of di- (ss'-chloroethyl) L-glutamate with a suitable base, for example soda, obtains di- (ss'-chloro L-glutamate - ethyl), reacts with the latter trityl chloride in the presence of an acid-binding agent for example triethylamine, takes up the N-trityl-L-glutamate of di- (ss'-chloroethyl) obtained in methanol by presence of sodium methoxide to obtain - (ss'-chloroethyl) and y-methyl N-trityl-L-glutamate, saponifies the latter, for example with sodium hydroxide in dioxane solution, to obtain N-trityl-L -a- (ss'-chloroethyl) glutamate,
that the latter is treated in a closed vessel with sodium iodide and trimethylamine to acetone, then detritylated the resulting N-trityl-L-glutamate α-choline iodide, for example with aqueous acetic acid to 50% to obtain α-L-glutamylcholine iodide. This can be converted into nitrate by the action of silver nitrate in aqueous solution or into any other salt by reaction with an appropriate reagent.
As shown in the US patent
No. 2776923 mentioned above, glutamylcholine and its salts are products showing interesting pharmacodynamic activity; it has been advocated for use in the treatment of poliomyelitis, hypertension and as a growth promoting agent.
According to Yoshito Nishizawa, Archives Françaises de Pédiatrie, 15, 195-216 (1958), α-L-glutamyl-choline has a therapeutic activity double that of γ-L-giutamyl-choline.
The following examples illustrate the invention. The structural formulas of the various compounds involved in the process according to the invention appear in the attached diagram.
Example a) Di- (B'-chloro- L-glutamate hydrochloride
ethyl) (II).
20 g of L-glutamic acid (I) are introduced into 200 cm3 of glycol monochlorohydrin, heated in an oil bath at 1050 C + 5 and passed through the mixture of gaseous hydrochloric acid while stirring until saturation. and complete dissolution, which takes about 30 minutes.
Is left for another 3 hours at the same temperature, distilled in a water bath under reduced pressure, about 150 cm3 of monochlorohydrin and added 250 cm3 of ether. This product, which has not yet been described, is in the form of colorless sheets, soluble in ice-water and leaves on warming up overnight at room temperature.
Most of the solvents are evaporated off under vacuum, 50 cm3 of water are added and the dioxane distillation is completed. Add 11 cm3 of hydrochloric acid
N in the presence of 40 cm3 of chloroform, stirred then decanted, extracted with twice 5 cm3 of chloroform, combined the chloroform solutions, washed with 10 cm3 of water, dried over magnesium sulphate and evaporated to dryness under vacuum. In this way, in quantitative yield, α- (f \ '-chloroethyl) N-trityl-L-glutamate (IV: R = H) is obtained.
This substance, which is in the form of an oil, has not yet been described; it can be used directly for the reaction described under f).
Α - ('- chloroethyl) N-trityl-L-glutamate can be characterized as its diethylamine salt, as follows:
The crude oil obtained according to e) is taken up in 10cm3 of ether; 1.1 cm3 of diethylamine are added then, in two steps, 20 cm3 of cyclohexane, ice for two hours, drained, washed with cyclohexane and 4.7 g of diethylamine salt of N-trityl-L-glutamate d 'α-( ss'-chloroethyl), F. around 100 C, [α] D20 =
+ 460 + 1 (c = 2%, chloroform). This new product is in the form of small, colorless needles, soluble in water, alcohol, acetone, benzene and chloroform, poorly soluble in ether and cyclohexane.

16.5 g of the diethylamine salt of the
Α-( ss'-chloroethyl) N-trityl-L-glutamate in 100 cm3 of chloroform, add 20 cm3 of water then, by fraction and stirring, 30 cm3 of hydrochloric acid
N. Decanted, extracted with 20 cm3 of chloroform, combined the chloroform extracts, washed with 30 cmS of water, dried over magnesium sulfate and evaporated to dryness under vacuum.
6 g of molten sodium iodide are introduced into a tube to be sealed, then the α- (p'-chloroethyl) N-trityl-L-glutamate obtained in the preceding paragraph, dissolved in 20 cm3 of dry acetone. 12 cm3 of anhydrous trimethylamine are added, the tube is sealed and heated overnight at 750 C. Ice, the tube is opened, evaporated to dryness under vacuum, taken up in 60 cm3 of chloroform and the insoluble material filtered. The filtered solution is
added to 20 cm3 of water containing 1 cm3 of acid
acetic. Acidified to pH 2 with an aqueous solution
diluted with hydriodic acid, decanted, extracted with chloroform, washed with 10 cm3 of water, dried the chloroform solution over magnesium sulfate and evaporated to dryness under vacuum. 120 cm3 of anhydrous ether are added,
stir, filter and dry under vacuum.
This gives 10.7 g
of a product which is pulverized and treated again with ether as above to obtain 9.8 g
(54%) α-choline N-trityl-L-glutamate iodide. water and alcohol, sparingly soluble in acetone and chloroform, insoluble in ether.

31 g of di- (ss'-chloroethyl) glutamate hydrochloride obtained according to a) are introduced into 50 cm3 of ice-cold water, added 250 cm3 of chloroform, 2 drops of phenolphthalein, then, in fractions, 12.5 cm3 of sodium hydroxide solution.
Decanted, the aqueous solution extracted with 25 cm3 of chloroform, combined the chloroform solutions, washed with 15 cm3 of water and dried over magnesium sulfate. c) Di- (ss'-chloroethyl) N-trityl-L-glutamate
(1V: R = -CH2-CHS-C1).
30 g of trityl chloride and, in portions, 15 cm3 of triethylamine are added to the chloroform solution above, then left overnight at 300 C.
Washed with 50 cm3 of water, a mixture of 50 cm3 of water and 45 cm3 of N hydrochloric acid and finally with 40 cm3 of water, dried over magnesium sulfate and evaporated to dryness under vacuum. d) a- (ss'-chloroethyl) N-trityl-L-glutamate and y-me
thyl (IV: R = - CH3).
The residue obtained above is dissolved in 50 cm3 of methanol, evaporated to dryness in vacuo, taken up in 200 cm3 of methanol at 350 ° C., 2.5 cm3 of a normal solution of sodium methoxide in methanol are added and left for 20 minutes at 350 C.
A further 1 cm3 of the sodium methoxide solution is added and then, after 40 minutes, again 1 cm3.
Is left for 1 hour at 350 C, one hour at room temperature, ice, filtered, rinsed with ice-cold methanol and dried at 400 C to obtain 30 g of product which is taken up in 50 cm3 of carbon tetrachloride. Ice, filter the insoluble, concentrate to half volume, add 50 cm3 of methanol and distill off what remains of carbon tetrachloride. Ice, filtered off, rinsed with ice-cold methanol and dried at 400 ° C. to obtain 29.2 g (63%) of α- (ss'-chloro- N-trityl-L-glutamate.

To 3 g of crude iodide of α-choline N-trityl-L-glutamate obtained according to f), 7.2 cm3 of 50% aqueous acetic acid are added, stirred and heated at 350 ° C. for fifteen minutes.
18 cm3 of acetone are added then, in portions, 25 cm3 of acetone. It is filtered off, washed with acetone and dried under vacuum to obtain 1.35 g (77%) of α -L-glutamyl-choline iodide, F. = 170 180 C, [α] D20 = + 13 , 5 # 1 (c = 2%, water).
This product, which has not yet been described, is in the form of small, colorless needles soluble in water, insoluble in alcohol, ether, acetone and benzene.

1.08 g of α-L-glutamyl-choline iodide obtained according to g) is introduced into a solution of 0.510 g of silver nitrate in 4 cm3 of water. Filter, concentrate to half volume, add 4 cm3 of alcohol and 40 cm3 of acetone, ice and wring. Recrystallized from 2 cm3 of slightly alcoholic water by adding acetone to obtain 650 mg (75%) of α -L-glutamyl-choline nitrate, F.
(in capillary tube) = approximately 90 C, [α] D20 = -t 16.5 + 1 (c = 2%, water).
This product, which is new, is in the form of small, colorless sheets, soluble in water, poorly soluble in alcohol, insoluble in ether, acetone and benzene.
