Procédé de préparation d'un amino#alcool tertiaire. La présente invention a pour objet un procédé de préparation de 3-(N-pipéridyl)-1- eyclohexyl-1-phényl-propanol-(1), qui est un composé nouveau. Ce procédé consiste à faire réagir, dans un solvant anhydre, l'oméga-(N- pipéridyl)-propiophénone et une quantité su périeure à un équivalent moléculaire d'un halogénure de cyclohexyl-magnésium, à hydro lyser ce mélange réactionnel et à isoler l'alcool tertiaire ainsi obtenu.
Le composé ainsi obtenu, correspondant à. la formule:

forme des aiguilles blanches et brillantes, dont le point de fusion corrigé est de 114,3 à 115 0 C, insolubles dans l'eau Iroide et l'alcool et peu solubles dans l'alcool chaud et les acides minéraux dilués.
Analyse: calculé pour C2oII3iN0: C 79,68; H 10,37; N 4,65 trouvé: C 79,7; H 10,3; N 4,47.
Il forme aisément des sels cristallisés tels que le chlorhydrate, le nitrate, le citrate, etc. C'est ainsi que le chlorhydrate, par exemple, se forme aisément et est soluble dans l'eau. Certains sels sont même tellement solubles dans l'eau qu'ils sont extrêmement hygrosco- piques et qu'ils doivent être manipulés avec précaution sous leur forme cristallisée. En so lution aqueuse, ils présentent la propriété pré cieuse de rester stables pendant longtemps. Les sels sont par suite très utiles pour l'appli cation pharmaceutique.
Il est remarquable que le propanol pré paré suivant le procédé de l'invention pré sente des propriétés antispasmodiques, si l'on considère qu'il lui manque le groupe ester jusqu'alors considéré comme nécessaire. Ce point est tout particulièrement surprenant, du fait que les alcools secondaires de structure analogue, par exemple le 3-(N-pipéridyl)- phényl-propanol-(1), ne manifestent aucune activité thérapeutique appréciable en tant que spasmolytiques.
Le produit semble actif à la fois contre le spasme direct et le spasme neurotropique; il est particulièrement efficace contre ce dernier. En plus de son activité qui est remarquable ment élevée, puisqu'il est plus actif que l'atro pine et d'autres substances antispasmolytiques; il se caractérise par une toxicité remarquable ment faible. Si l'on envisage le rapport de ces deux facteurs, activité et toxicité, comme un indice thérapeutique, on pourra dire que ce composé est beaucoup plus favorable que des substances connues.
Comme le premier stade de la réaction doit être effectué dans des conditions qui ne con duisent pas à l'hydrolyse, on a constaté qu'un éther dialcoylique, par exemple les éthers di- éthylique, dibutylique, etc., ou un mélange de benzène et de pyridine, constituent des sol vants entièrement satisfaisants, pourvu qu'ils soient anhydres.
La proportion de réactif de Grignard utili sée exerce aussi une influence sur le rende ment. Il semble que la cause en soit la for mation d'un complexe qui se dissocie lors de l'hydrolyse. On pense que c'est pour cette rai son qu'une partie du réactif n'est plus dispo nible pour la suite de la réaction. Quoi qu'il en soit, en augmentant la proportion du ré actif de Grignard notablement au-delà de la quantité équimoléculaire, on obtient en géné ral un rendement nettement amélioré. En re vanche, au-delà de deux équivalents molécu laires, toute nouvelle augmentation de la pro portion utilisée diminue sensiblement le rende ment final.
La proportion de deux molécules environ de réactif de Grignard par molécule de cétone semble donc être la plus favorable.
Dans l'exemple qui suit, toutes les propor tions sont données en poids, sauf indication contraire.
A une solution, dans l'éther éthylique ab solu, de bromure de cvclohexyl-magnésium (préparée à partir de 261 parties de bromure de cyclohexyle, 38,8 parties de tournures de magnésium et 700 parties en volume d'éther éthylique absolu), on ajoute, sous agitation, une solution sèche de 174 parties d'oméga-(N- pipéridyl) -propiophénone dans 600 parties en volume d'éther, à une vitesse assurant le main tien d'une douce ébullition à reflux, sans re froidissement ni chauffage extérieurs. On agite le mélange réactionnel pendant environ 5 heures, puis on le laisse reposer à la tempé rature ambiante jusqu'à ce que la réaction pa raisse achevée.
Quand il est refroidi, le mé lange réactionnel est alors hydrolysé par addi tion goutte à goutte de 500 parties en volume d'acide chlorhydrique 2,5 N, et enfin on le rend fortement acide vis-à-vis du rouge Congo par addition d'acide chlorhydrique concentré. On recueille le solide blanc résultant sur un filtre, on le sèche à l'air, on le redissout dans 2500 parties d'eau à 95 , on traite cette solu tion avec du charbon décolorant et on la cla rifie par filtration. Le filtrat refroidi est rendu basique au moyen d'ammoniaque et l'on recueille le 3-(N-pipéridvl)-1-cyclohexvl-1- phényl-propanol- (1).
Ainsi qu'on l'a signalé plus haut, le com posé préparé suivant la présente invention est utile en tant qu'antispasmodique. On pourra se faire une idée de son efficacité au moyen de l'essai courant. qui consiste à déterminer sa capacité à. assurer le relâchement de bandes isolées de l'intestin du lapin plongées dans une solution d'essai à température constante.
Dans cet essai, l'état. spasmodique était provo qué par de l'iodure de furfuryltriméthvl- ammonium à raison de 0,1 mg/100 ml de solu tion de Tyrode. On admet que la relaxation du fragment intestinal est égale à 100% lorsque l'état spasmodique a été surmonté au point de rendre au fragment intestinal son activité normale. Si l'intestin ne présente essentiellement aucune contraction après l'ad ministration du médicament antispasmodique, cet état est noté comme correspondant à une relaxation complète.

A process for the preparation of a tertiary amino alcohol. The present invention relates to a process for the preparation of 3- (N-piperidyl) -1-eyclohexyl-1-phenyl-propanol- (1), which is a new compound. This process consists of reacting, in an anhydrous solvent, omega- (N-piperidyl) -propiophenone and an amount greater than a molecular equivalent of a cyclohexyl-magnesium halide, hydrolyzing this reaction mixture and isolating. the tertiary alcohol thus obtained.

forms white and shiny needles, the corrected melting point of which is 114.3 to 115 ° C., insoluble in cold water and alcohol and poorly soluble in hot alcohol and dilute mineral acids.

It readily forms crystalline salts such as hydrochloride, nitrate, citrate, etc. Thus, the hydrochloride, for example, is easily formed and is soluble in water. Some salts are even so soluble in water that they are extremely hygroscopic and must be handled with care in their crystallized form. In aqueous solution, they have the valuable property of remaining stable for a long time. The salts are therefore very useful for pharmaceutical application.
It is remarkable that the propanol prepared according to the process of the invention exhibits antispasmodic properties, if it is considered that it lacks the ester group hitherto considered necessary. This point is particularly surprising, since secondary alcohols of similar structure, for example 3- (N-piperidyl) - phenyl-propanol- (1), do not show any appreciable therapeutic activity as spasmolytics.
The product appears to be active against both direct spasm and neurotropic spasm; it is particularly effective against the latter. In addition to its activity which is remarkably high, since it is more active than atropine and other antispasmolytic substances; it is characterized by remarkably low toxicity. If we consider the ratio of these two factors, activity and toxicity, as a therapeutic index, we can say that this compound is much more favorable than known substances.
Since the first stage of the reaction should be carried out under conditions which do not lead to hydrolysis, it has been found that a dialkyl ether, for example diethyl, dibutyl ethers, etc., or a mixture of benzene and pyridine, are entirely satisfactory solvents, provided they are anhydrous.
The proportion of Grignard reagent used also has an influence on the yield. It seems that the cause is the formation of a complex which dissociates during hydrolysis. It is believed that it is for this reason that part of the reagent is no longer available for the rest of the reaction. In any event, by increasing the proportion of the Grignard reagent notably above the equimolecular amount, a markedly improved yield is generally obtained. On the other hand, beyond two molecular equivalents, any further increase in the proportion used appreciably decreases the final yield.
The proportion of approximately two molecules of Grignard reagent per ketone molecule therefore seems to be the most favorable.
In the example which follows, all the proportions are given by weight, unless otherwise indicated.
To a solution, in dissolved ethyl ether, of cyclohexyl-magnesium bromide (prepared from 261 parts of cyclohexyl bromide, 38.8 parts of magnesium turnings and 700 parts by volume of absolute ethyl ether), a dry solution of 174 parts of omega-(N-piperidyl) -propiophenone in 600 parts by volume of ether is added, with stirring, at a speed ensuring the maintenance of a gentle boiling at reflux, without cooling nor outdoor heating. The reaction mixture is stirred for about 5 hours, then allowed to stand at room temperature until the reaction is complete.
When it is cooled, the reaction mixture is then hydrolyzed by the dropwise addition of 500 parts by volume of 2.5N hydrochloric acid, and finally it is made strongly acid vis-à-vis Congo red by the addition of concentrated hydrochloric acid. The resulting white solid is collected on a filter, dried in air, redissolved in 2,500 parts of 95% water, treated with decolourizing carbon and clarified by filtration. The cooled filtrate is made basic with ammonia and 3- (N-piperidvl) -1-cyclohexvl-1-phenyl-propanol- (1) is collected.
As noted above, the compound prepared according to the present invention is useful as an antispasmodic. An idea of its effectiveness can be obtained from the current test. which is to determine its ability to. ensure the release of isolated strips of rabbit intestine immersed in a test solution at constant temperature.
In this essay, the state. spasmodic was caused by furfuryltrimethyl ammonium iodide at 0.1 mg / 100 ml of Tyrode's solution. It is assumed that the relaxation of the intestinal fragment is equal to 100% when the spasmodic state has been overcome to the point of returning the intestinal fragment to its normal activity. If the intestine shows essentially no contraction after administration of the antispasmodic drug, this condition is noted as corresponding to complete relaxation.
