Procédé de préparation de nouveaux dérivés de l'isothiazole On sait que parmi les très nombreux composés p-amino-benzènesulfamidés et leurs dérivés qui ont été préparés et contrôlés par différents chercheurs au point de vue de leur activité antibactérienne ou de leurs autres activités thérapeutiques, très peu ont été trouvés suffisamment intéressants pour être uti lisés en médecine humaine ou vétérinaire.
Malgré des recherches et des essais nombreux pendant ces der nières années, il était impossible de prévoir si un composé donné possédait une activité antibactérienne comparable à celle des composés hétérocycliques sul- famidés acceptés par le corps médical, tels que par exemple le (p-amino-benzènesulfamido)-2 thiazole, connu sous le nom générique de sulfathiazole.
On a maintenant trouvé que, d'une façon tout à fait inattendue, de nouveaux isothiazoles possé daient une activité antibactérienne aussi grande, si non plus grande, que le sulfathiazole, plus particu lièrement contre E. Coli.
La présente invention a donc pour objet un pro cédé de préparation des isothiazoles de formule

dans laquelle Ri représente un atome d'hydrogène ou un radical méthyle, un des radicaux R.-, et R3 représente un atome d'hydrogène et l'autre un radi cal p-amino-benzènesulfamido fixé au noyau isothia- zole par l'atome d'azote du radical sulfamido.
Le procédé selon l'invention est caractérisé en ce que l'on fait réagir un halogénure de p-R benzène- sulfonyle, R représentant un radical transformable en radical amino primaire, avec l'amino -4 (ou -5)- isothiazole correspondant puis que l'on transforme le radical R en un groupe amino primaire.
On peut transformer le produit obtenu en sels avec, par exem ple, des bases alcalines ou alcalino-terreuses, ou en dérivés N-acylès, par exemple les dérivés phtalylés ou succinylés. Le radical R est de préférence un ra dical acylamido, qui est transformé en radical amino primaire par hydrolyse du produit. La condensation peut être effectuée en présence d'un agent de con densation basique et d'un solvant organique ou aqueux. Le solvant et l'agent de condensation peu vent être la même substance, par exemple la pyridine.
Les amino-4 (ou -5)-isothiazoles de départ sont eux-mêmes nouveaux et peuvent être préparés par des méthodes qui ne font pas partie de la présente in- vention, mais sont néanmoins décrites dans les exem ples suivants <I>Exemple 1</I> A une solution refroidie sur glace de<B>12,5g</B> d'amino-4 (ou -5)-isothiazole dans 125 cm3 de pyri- dine on ajoute lentement, en dessous de 1011,
32 g de chlorure de p-acétamido-benzènesulfonyle. Après 16 heures, on verse la solution dans 4 litres d'eau et après encore 2 heures on filtre le produit solide, le lave à l'eau et le sèche : on en recueille 37,5 g. Par cristallisation dans l'éthanol on obtient le p-acéta- mido-benzènesulfamido-4 (ou -5)-isothiazole sous forme de petits cristaux incolores, fondant à 261 262o.
On fait bouillir à reflux, pendant deux heures, 2,1 g de p-acétamido-benzènesulfamido-4 (ou -5)- isothiazole dans 20 cm3 d'hydroxyde de sodium 2N. On ajoute du noir décolorant, on continue à faire bouillir pendant cinq minutes et on filtre la solution à chaud. Après refroidissement, on ajuste le pH du filtrat à 4-5 à l'aide d'acide chlorhydrique 2N et le mélange est réfrigéré pendant une nuit.
On filtre le p-amino-benzènesulfamido-4 (ou -5)-isothiazole, le lave à l'eau et après séchage, on en obtient 1,7 g ; par recristallisation dans l'éthanol, il se présente sous forme d'aiguilles incolores, fondant à 206o.
L'hydrolise peut aussi bien être effectuée avec de l'acide chlorhydrique 2N.
L'amino-4 (ou -5)-isothiazole de départ peut être obtenu comme suit On oxyde de l'amino-5-benz-1,2-isothiazole à l'aide de permanganate de potassium alcalin en acide isothiazole dicarboxylique-4,5 fondant à 145o (d) qui, après purification par l'intermédiaire du sel de sodium, est décarboxylé dans le mésitylène bouillant et donne l'acide isothiazole carboxylique-4 (ou -5) fondant à 1620.
L'ester méthylique de cet acide, fon dant à 55 , préparé en utilisant le diazométhane, est chauffé avec de l'hydrate d'hydrazine dans l'éthanol et on obtient la carboxy-hydrazide correspondante fondant à 176o (d).
Ce dernier corps est mis en suspension dans l'acide chlorhydrique; on recouvre d'éther et on traite au nitrite de sodium à une tem pérature comprise entre -50 et 0t. On chauffe l'iso- thiazole-4 (ou -5)-carboxyazide résultante (P. F. 320) avec de l'alcool benzylique et on obtient la benzyl isothiazole-4 (ou -5)-uréthane (P. F.
101o) qui par traitement à l'acide bromhydrique dans l'acide acé tique conduit au brombydrate d'amino-4 (ou -5)-iso- thiazole. La base libre, qui fond à 45a, est libérée par le carbonate de sodium, extraite à l'éther et cristal- lisée dans l'éther de pétrole.
<I>Exemple 2</I> On mélange à température inférieurre à 200 une solution de 20 g d'amino-5-méthyl-3-isothiazole dans 80 cm3 de pyridine et de 46,4 g de chlorure de p- acétamido-benzènesulfonyle dans 80 cm3 de pyridine. On laisse le mélange reposer à température ordinaire pendant 24 heures puis on le verse dans une solution de 250 cm3 d'acide sulfurique concentré dans 4 kg d'eau et de glace,
contenant suffisamment de glace pour maintenir la température finale inférieure à 10o. On filtre le solide qui précipite et le lave à l'eau. Par recristallisation dans l'acide acétique glacial on ob tient le p-acétamido-benzènesulfamido-5-méthyl-3- isothiazole qui fond à 271-2730 (d), sous forme de cristaux lenticulaires à peu près incolores.
On dissout 0,5 g de p-acétamido-benzènesulfa- mido-5 méthyl-3 isothiazole dans 10 cm3 d'une solu tion d'hydroxyde de sodium 2N et on fait bouillir la solution à reflux pendant 30 minutes. Après refroi dissement sur glace, on ajuste la solution à pH 6 par addition d'acide chlorhydrique 2N.
Le solide résultant est recueilli à 00 et par recristallisation dans l'eau on obtient le p-amino-benzènesulfamido- 5-méthyl-3-isothiazole, fondant à 191-1920, sous forme d'aiguilles jaune pâle. L'amino-5-méthyl-3-isothiazole de départ peut être obtenu de la façon suivante: on agite mécanique ment une solution de 136g d'hydroxyde de sodium dans 7 litres d'eau et on la refroidit à 12o par addi tion d'environ 500 g de glace.
On ajoute 460 g de persulfate de potassium en poudre et on agite la suspension pendant 10 minutes. On ajoute 197 g de (3-iminothiobutyramide par fractions, pendant 20 mi nutes, et assez de glace pour maintenir la température du mélange entre 12 et 180 (environ 2 kg). Après environ 30 minutes on obtient une solution rouge limpide. On agite cette solution pendant encore 6 heures et la laisse reposer une nuit. On filtre la petite quantité d'impuretés présentes et extrait la solution en continu avec 1 litre d'éther, en 24 heures.
On sèche l'extrait sur sulfate de sodium, le refroidit sur glace et le traite au gaz chlorhydrique sec. Le chlorhydrate d'amine qui précipite est recueilli im médiatement, lavé à l'éther sec et séché à l'exsicca- teur. On dissout 1 partie du chlorhydrate d'amine dans 10 parties d'eau et on filtre la solution. On ajoute 1 volume d'une solution d'hydroxyde de so dium à 500 g/litre et on extrait la solution en continu à l'éther pendant une nuit.
On sèche l'extrait sur sul fate de sodium et l'évapore ; on distille le résidu sous vide et on obtient l'amino-5-méthyl-3-isothiazole sous forme d'un solide jaune pâle, fondant à 52-53, bouillant à 90-92/0,1 mm.

Despite extensive research and testing over the past few years, it was impossible to predict whether a given compound possessed antibacterial activity comparable to that of the sulphamid heterocyclic compounds accepted by the medical profession, such as, for example, (p-amino -benzenesulfamido) -2 thiazole, generically known as sulfathiazole.
It has now been found that, quite unexpectedly, new isothiazoles possess as great, if not greater, antibacterial activity than sulfathiazole, more particularly against E. Coli.
A subject of the present invention is therefore a process for the preparation of isothiazoles of formula

in which R 1 represents a hydrogen atom or a methyl radical, one of the radicals R.-, and R3 represents a hydrogen atom and the other a radi cal p-amino-benzenesulfamido attached to the isothiazole ring by the nitrogen atom of the sulfamido radical.
The process according to the invention is characterized in that a pR benzene sulfonyl halide, R representing a radical convertible into a primary amino radical, is reacted with the corresponding amino -4 (or -5) - isothiazole then that the radical R is converted into a primary amino group.
The product obtained can be converted into salts with, for example, alkaline or alkaline-earth bases, or into N-acyl derivatives, for example phthalyl or succinyl derivatives. The R radical is preferably an acylamido radical, which is converted into the primary amino radical by hydrolysis of the product. The condensation can be carried out in the presence of a basic condensing agent and an organic or aqueous solvent. The hydrolysis can also be carried out with 2N hydrochloric acid.
The starting amino-4 (or -5) -isothiazole can be obtained as follows: Oxidizing 5-amino-benz-1,2-isothiazole using alkaline potassium permanganate to isothiazole-4 dicarboxylic acid , 5 mp 145 ° (d) which, after purification via the sodium salt, is decarboxylated in boiling mesitylene to give isothiazole-4 (or -5) carboxylic acid mp 1620.

<I> Example 2 </I> A solution of 20 g of amino-5-methyl-3-isothiazole in 80 cm3 of pyridine and 46.4 g of p-acetamido- chloride is mixed at a temperature below 200. benzenesulfonyl in 80 cm3 of pyridine. The mixture is left to stand at room temperature for 24 hours then it is poured into a solution of 250 cm3 of concentrated sulfuric acid in 4 kg of water and ice,
containing enough ice to keep the final temperature below 10o. The solid which precipitates is filtered off and washed with water. By recrystallization from glacial acetic acid, p-acetamido-benzenesulfamido-5-methyl-3-isothiazole is obtained, which melts at 271-2730 (d), in the form of almost colorless lenticular crystals.
0.5 g of p-acetamido-benzenesulfamido-5-methyl-3-isothiazole is dissolved in 10 cc of 2N sodium hydroxide solution and the solution is boiled under reflux for 30 minutes. After cooling on ice, the solution is adjusted to pH 6 by adding 2N hydrochloric acid.
The resulting solid is collected at 00 and recrystallization from water gives p-amino-benzenesulfamido-5-methyl-3-isothiazole, melting at 191-1920, in the form of pale yellow needles.
