Verfahren zur Herstellung von neuen 5, 6, 7, 8-Tetrahydro-pyrido [4, 3-d] pyrimidinen
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 5, 6, 7, 8-Tetrahydro-pyrido [4, 3-d] pyrimidinen der Formel I :

und von ihren Salzen mit physiologisch verträglichen anorganischen oder organischen Säuren.
In dieser Formel bedeuten R ein Wasserstoffatom, einen Alkyl-, Aryl- oder Aralkylrest, R einen geradkettigen oder verzweigten, gesättigten oder ungesättigten Alkylrest, Rz ein Wasserstoffatom, einen Alkyl- oder Aralkylrest, eine mono- oder disubstituierte Aminogruppe, deren Stickstoffatom auch Glied eines gesättigten heterocyclischen Ringes, der gegebenenfalls ein weiteres Heteroatom enthält, sein kann, oder eine freie oder durch einen Alkyl- oder Aralkylrest substituierte Mercaptogruppe.
Die neuen Verbindungen lassen sich erfindungsgemäss durch Umsetzung eines Piperidoncarbonsäureesters der Formel II :

in der R eine der oben angeführten Bedeutungen besitzt oder mit einem Säureadditionssalz dieses Amidins mit einer anorganischen oder organischen Säure erhalten.
Diese Umsetzung wird vorzugsweise in Gegenwart eines alkalischen Kondensationsmittels und zweckmässig in Gegenwart eines Lösungsmittels bei Zimmertemperatur oder mässig erhöhter Temperatur durchgeführt. Als alkalisches Kondensationsmittel kann beispielsweise ein Alkalihydroxyd, ein Alkalicarbonat oder ein Alkalialkoholat verwendet werden, als Lösungsmittel Wasser oder ein organisches Lösungsmittel, beispielsweise Methylalkohol, Äthylalkohol, Benzol, Tetrahydrofuran usw.
Falls eine Verbindung der Formel I erhalten wird, in der R eine freie oder substituierte Mercaptogruppe bedeutet, so kann diese gewünschtenfalls nachträglich mit Ammoniak, einem primären oder sekundären Amin in einen der oben bei der Definition von R angeführten basischen Reste überführt werden. Diese Umsetzung wird vorteilhaft unter Anwendung eines Überschusses des betreffenden Amins, gegebenenfalls in Gegenwart eines inerten-organischen Lösungsmittels, wie Alkohol, Benzol, Tetrahydrofuran usw., durchgeführt ; als Lösungsmittel kann auch ein grösserer Überschuss des eingesetzten Amins verwendet werden.
Die Umsetzung wird bei Temperaturen zwischen 20 und 180 C, vorzugsweise bei der Siedetemperatur des verwendeten Lösungsmittels bzw. des eingesetzten Amins durchgeführt ; falls niedrigsiedende Amine oder Lösungsmittel verwendet werden, empfiehlt es sich, im geschlossenen Gefäss zu arbeiten.
Verbindungen der Formel I, in denen R2 einen der angegebenen basischen Reste bedeutet, lassen sich also nach einem einstufigen oder nach einem zweistufigen Verfahren herstellen. Nach der üblichen Aufarbeitung können die erhaltenen freien Basen gewünschtenfalls in ihre Salze mit einer physiologisch verträglichen anorganischen oder organischen Säure, beispielsweise Salzsäure, Schwefelsäure, Phosphorsäure, Bernsteinsäure, Weinsäure, Zitronensäure, Maleinsäure usw. überführt werden. Sind mehrere basische Stickstoffatome vorhanden, lassen sich auch Salze mit mehreren Äquivalenten der betreffenden Säure herstellen.
Die als Ausgangsstoffe verwendeten Piperidoncarbonsäureester der Formel II waren teils bekannt, teils lassen sie sich auf bekannte Weise aus Imino-dipropionsäureestern der Formel IV :

Eine besonders vorteilhafte Variante des erfindungsgemässen Verfahrens besteht darin, dass die Ausgangsstoffe der Formel II nicht isoliert werden, sondern dass das bei der Dieckmann-Kondensation anfallende Reaktionsgemisch direkt mit einem Amidin der Formel III umgesetzt wird, zweckmässig in Gegenwart von Wasser oder in Gegenwart eines organischen Lösungsmittels.
Die neuen Verbindungen weisen wertvolle therapeutische Eigenschaften auf und sind in dieser Hinsicht den bisher bekannten Pyrido [4, 3-d] pyrimidinen wesentlich und in überraschender Weise überlegen.
Sie sind insbesondere antipyretisch, antiphlogistisch, diuretisch, bakteriostatisch, sedativ und coronardilatorisch wirksam, wobei natürlich bei den einzelnen Verbindungen die eine oder die andere Wirkungskomponente überwiegt.
Die folgenden Beispiele sollen die Erfindung erläutern, ohne sie zu beschränken.
Beispiel l : 22, 2gl, 5-Dimethylpiperidon- (4)-3-carbonsäure-methylester. HC1, 19, lgPhonyl- acetamidin. HCl und 20, 8 g Kaliumcarbonat werden in je 50 ml Wasser gelöst und vermischt. Nach kurzem Erwärmen auf 60 C beginnt die Kristallisation, man lässt noch einige Stunden stehen und saugt dann ab. Nach Umkristallisation aus Methanol erhält man 20 g 2-Benzyl-4-hydroxy-6, 8-dimethyl- - 5, 6, 7, 8-tetrahydro-pyrido[4, 3-d] pyrimidin vom F. = 193-195 C.
Beispiel 2 : Zur Lösung von 11, 5 g Natriummetall in 300 ml absolutem Methanol werden 115 g Methylamino-dipropionsäure-diäthylester gegeben. Das Lösungsmittel wird im Vakuum abgezogen und der Rückstand noch 1 h im Hochvakuum auf 1300C erhitzt. Das entstandene Natriumenolat des N-Methyl- - piperidon- (4)-3-carbonsäureäthylesterswird in 100 ml Wasser gelost und die Lösung von 74 g S-Äthylisothioharnstoff. HBr in 200 ml Wasser zugegeben. Das Reaktionsgemisch wird 3-4 h kräftig gerührt und dann der entstandene Niederschlag abgenutscht. Nach Auswaschen mit kaltem Wasser wird aus Essigester um-

erhältisothioharnstoffsulfat und 41 g Kaliumcarbonat werden in je 40 ml Wasser unter gelindem Erwärmen gelöst und dann zusammengegeben.
Nach 5stündigem Rühren wird das entstandene Kristallisat gesammelt und aus Äthanol umkristallisiert. Man erhält 23 g 2-Methylmercapto-4-hydroxy-6, 8-dimethyl-5, 6, 7,8-tetrahydro-pyrido [4, -d] pyrimidin vom F. = 212 C.
Beispiel 4 : 11, 2 g 2-Methylmercapto-4-hydroxy-6, 8-dimethyl-5, 6, 7, 8-tetrahydro-pyrido- [4, 3-d] pyrimidin und 26 g Dibutylamin werden in einem V 4A-Autoklaven 32 h auf 170 C erhitzt, Nach Abziehen des überschüssigen Dibutylamins wird der Rückstand mit etwas Äthanol angerieben und der er- haltene Kristallbrei abgenutscht. Nach Umkristallisation aus Äthanol erhält man 9 g 2-Dibutylamino-4- - hydroxy-6, 8-dimethyl-5, 6, 7, 8-tetrahydro-pyridor4, 3-djpyrimidin vom F. = 134-135 C.
Beispiel 5 : Die Mischung aus 10 g 2-Methylmercapto-4-hydroxy-6-butyl-5, 6, 7, 8-tetrahydro-

pyrido[4, 3-d]pyrimidinschüssigen Amins wird der Rückstand mit Äther verrieben, abgenutscht und aus Äthanol umkristallisiert.
Man erhält 8, 5 g 2-Hexylamino-4-hydroxy-6-butyl-5,6,7,8-tetrahydro-pyrido[4,3-d]pyrimidin vom F. = 129 C.
Beispiel 6 : Zu einer Lösung von 1, 4 g Natriummetall in 150 ml absolutem Äthanol werden 4, 1 g N, N-Dimethylguanidin-sulfat und 6, 7 g 1-Methyl-3-carbäthoxy-piperidon-(4)-hydrochlorid gegeben und das Gemisch 5 h unter Rühren zum Rückfluss erhitzt, vom ausgefallenen anorganischen Material abfiltriert und das Filtrat unter vermindertem Druck eingeengt. Nach Umkristallisieren des festen Rückstandes aus Äthanol erhält man 3, 4 g 2-Dimethylamino-4-hydroxy-6-methyl-5,6,7,8-tetrahydro-pyrido[4,3-d] pyrimidin vom F. = 204-205 C..
