본 발명은 생약재를 이용한 건강보조식품 조성물 및 이를 이용한 건강보조식품에 관한 것이다. 본 발명에 따른 생약재를 이용한 건강보조식품 조성물은 생약재로 우슬, 지모, 백출, 당삼, 단풍마, 목단피, 황백, 숙지황, 현삼, 택사, 황기, 사상자, 선모, 패모, 구기자, 상백피, 황정, 오미자 및 반지련을 포함한다. 상기한 구성에 의해 본 발명에 따른 생약재를 이용한 건강보조식품은 천연생약재의 유용성분을 용이하게 섭취할 수 있고 건강한 삶을 영위할 수 있도록 섭취자에게 영양분을 제공할 수 있다.
Description
본 발명은 생약재를 이용한 건강보조식품 조성물 및 이를 이용한 건강보조식품에 관한 것으로서, 더욱 상세하게는 천연생약재 성분을 유효성분으로 포함하여 건강보조식품 조성물을 구성함으로써 천연생약재의 유용성분을 용이하게 섭취할 수 있고 건강한 삶을 영위할 수 있도록 영양분을 제공할 수 있는 생약재를 이용한 건강보조식품 조성물 및 이를 이용한 건강보조식품에 관한 것이다.
경제성장에 따른 국민소득 향상으로 생활수준이 크게 향상되고, 건강에 대한 관심이 높아지면서 영양의 중요성이 재인식되었다. 일상의 음식만으로는 인체의 균형 및 영양유지가 불충분하다는 인식이 확산됨에 따라 건강 식품의 소비가 점차 증가하고 있다.
건강 기능 식품이 각광 받는 이유는 고령화 사회로의 진입, 식습관에 기인하는 고혈압, 당뇨병, 심혈관계 질환 등의 만성질환의 증가와 인스턴트 음식의 발달로 미량 영양소의 섭취가 줄어들고 있기 때문이다. 식품의 유효성분에 의한 건강 증진 및 질병 예방의 효과 등이 증명되면서 식품의 기능성, 즉, 식품의 생체조절기능이 주목받고 있고, 이를 이용한 많은 제품들이 만들어지고 있다.
건강 기능 식품은 유용한 기능성을 가진 원료나 성분을 사용하여 제조한 식품을 말하며 여기서 기능성이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용등과 같은 보건용도에 유용한 효과를 얻는 것을 말한다.
한편, 종래 생약재(한약재)를 복용하는 방법으로는 재래식 약탕기로 한약을 달여내어 복용하는 방법을 들 수 있는데, 한약재에 찬물을 붓고 장시간, 보통 2시간 끓여서 추출하는 방식으로 유효성분을 추출하게 된다. 이러한 방법은 장시간 달이고 적당히 식혀 먹도록 되어 있어, 약의 투여가 급한 응급 환자에 있어서는 그 대응력이 양약에 비해 현저히 떨어지는 단점을 가진다.
이러한 복용 상의 불편함으로 인하여, 액상 형태로 미리 제조된 갈근탕 또는 쌍화차와 같은 한방차들이 시중에 나와 있지만, 금방 달여진 한약액이나 한방차와 비교할 때, 약효, 맛 또는 향 등이 그대로 보존되기 어려운 문제점이 있으며, 또한, 이러한 한방차도 끓는 물에 다시 데워야 하는 불편함이 있다.
상기와 같은 복용방법의 문제점으로 인하여 다양한 형태로 복용할 수 있는 한약재가 개발되고 있는데, 예를 들어, 대한민국 특허공개 제10-2005-0053572호는, 인삼, 대추, 밤 및 마늘을 건조한 후 미세하게 분쇄하여 일정한 비율로 혼합한 분말가루를 티백에 밀봉하여 이용하는 '건강식 한약제 티백'을 개시하고 있으며, 대한민국 등록실용신안공보 제20-399746호에는, 오가피 건조분쇄물을 티백에 밀봉하여 이용하는 '한방 오가피 티백'이 개시되어 있다.
그러나 상기 티백의 내용물로 사용되는 한약재는 건조, 분쇄 등의 단순처리를 통하여 티백에 담아 사용할 수 있는 것으로, 황기, 당귀, 작약, 황정, 천궁, 백출, 하수오 등의 약재를 상기와 같이 단순한 건조분쇄의 방법으로 처리하여 티백에 담아 사용하게 되면 각 약재의 유효성분이 온전히 용출되기 어려운 문제점이 있었다.
본 발명이 속하는 기술분야의 선행기술문헌에는 한국 등록특허공보 제10-1966021호, 한국 등록특허공보 제10-1812725호, 한국 공개특허공보 제10-2016-0074300호 등이 있다.
본 발명은 천연생약재 성분을 유효성분으로 포함하여 건강보조식품 조성물을 구성함으로써 천연생약재의 유용성분을 용이하게 섭취할 수 있고 건강한 삶을 영위할 수 있도록 영양분을 제공할 수 있는 생약재를 이용한 건강보조식품 조성물 및 이를 이용한 건강보조식품을 제공하는데 있다.
또한, 본 발명은 남성 건강에 유익한 천연생약재 성분을 포함함으로써 남성호르몬인 테스토스테론의 분비가 증강되어 남성 건강에 효과적인 생약재를 이용한 건강보조식품 조성물 및 이를 이용한 건강보조식품을 제공하는데 있다.

상기 발효는 상기 생약재 분말과 홍국균을 혼합한 후 60 내지 70℃의 온도에서 20 내지 50시간 동안 발효하여 진행될 수 있다.

또한, 본 발명에 따른 생약재를 이용한 건강보조식품 조성물은 우슬 3 내지 7 중량부, 지모 3 내지 7 중량부, 백출 3 내지 7 중량부, 당삼 3 내지 7 중량부, 단풍마 3 내지 7 중량부, 목단피 3 내지 7 중량부, 황백 3 내지 7 중량부, 숙지황 3 내지 7 중량부, 현삼 3 내지 7 중량부, 택사 3 내지 7 중량부, 황기 3 내지 7 중량부, 사상자 3 내지 7 중량부, 선모 3 내지 7 중량부, 패모 3 내지 7 중량부, 구기자 3 내지 7 중량부, 상백피 3 내지 7 중량부, 황정 3 내지 7 중량부, 오미자 3 내지 7 중량부 및 반지련 3 내지 7 중량부의 중량 비율로 포함될 수 있다.
상기 우슬은 한국의 중부 이남의 산과 들에 자라는 여러해살이 식물이며, 중요성분은 사포닌, 칼륨, 점액질이 있어, 간과 신장의 병은 치료하는 약재로 쓰이고 있으며, 엑디스테로이드 성분이 있어서 염증을 잡아주고 뼈를 튼튼하게 하는 작용을 하며, 트리페리핀성분과 사포닌 성분이 면역력을 높이므로 약제로 사용되고 있다. 본 발명에서 상기 우슬은 술에 20 내지 30시간 동안 침지시킨 후 건조하고 볶아 사용할 수도 있다.
상기 지모(Anemarrhena asphodeloides)는 나리(Liliaceae)과에 속하는 식물로서, 중국이 원산지이며 한국에서는 중부지방에서 재배되는 다년초이다. 보통 근경을 건조하여 약재로 사용되어 왔으며 소염, 해열, 지사, 이뇨 작용과 요통 완화 및 진정 효과가 있는 것으로 알려져 있다. 지모의 유효성분으로서 아스포닌(Asphonin) 6% 이외에도, 살사사포닌(Sarsasaponin), 말코게닌(Markogenin; 2-hydroxy sarsasapogenin) 등의 스테로이드 사포게닌, 플라보노이드(Flavonoids) 및 탄닌 등이 함유되어 있는 것으로 알려져 있다. 본 발명에서 상기 지모는 술에 20 내지 30시간 동안 침지시킨 후 건조하고 볶아 사용할 수도 있다.
상기 백출(白朮)은 삽주(Atractylodes japonica Koidzumi) 또는 백출(Atractylodes macrocephala Koidzumi, 국화과 Compositae)의 뿌리줄기로서 그대로 또는 주피를 제거한 것으로, 지라와 위장의 기능이 약해 소식, 권태감이 생기고 얼굴빛이 황색이며 대변을 묽게 보거나 설사에 좋으며 수분이 정체되어 전신이 붓고 소화가 안될때 수분 배설을 돕는다. 위장에 수(水)와 습(濕)이 쌓여 가슴뜀, 기침, 맑은 가래 등에도 쓰며 지라의 기운 허약으로 피부에 땀이 저절로 날때, 임신 구토에도 사용한다. 위장 장애가 있는 감기, 사지동통 에도 쓴다. 또한 식욕증진에도 탁월한 효능이 있다고 알려져 있다.
약리적으로 생쥐의 체중 증가와 지구력 증가, 탐식 능력을 증가, 세포 면역 기능 촉진이 보고되었다. 또한 흥분 작용을 조절, 항궤양 및 간 기능 보호 작용, 면역 기능 항진작용, 혈관 확장 작용, 이뇨 작용 및 혈당 강하 작용, 항암작용 등이 보고되었다. 본 발명에서 상기 백출은 쌀뜨물에 20 내지 30시간 동안 침지시킨 후 건조하고 볶아 사용할 수도 있다.
상기 당삼은 초롱꽃과(길경과: Campanulaceae)에 속하는 다년생 초본인 만삼(黨蔘, Pilose bellflower 학명: Codonopsis pilosula)의 뿌리이다. 맛이 달고, 성질은 평하며, 폐경에 작용하여 원기를 도우며 피로와 영양 실로조 인한 열이나 땀을 제거하고 기를 보충하고 몸의 진액과 혈을 보양하는 효능이 있으므로, 능히 지라와 위장, 폐의 기운이 약하거나 병후로 신체가 약한 증후에 상용하는 약물이다. 당삼은 작용이 인삼과 비슷하여 일반적으로 보기와 건비의 방제에 인삼의 대체약물로 많이 사용한다.
상기 당삼의 화학성분으로는 saponin 및 미량의 알카로이드, 단백질, 전분,비타민 A1, A2 등을 함유한다. 만삼(黨蔘, Pilose bellflower 학명: Codonopsis pilosula)은 민간에서 옛날부터 강장, 건위, 쇠약, 빈혈, 백혈병에 유효하다고 하였으며, 약리작용에 관해서는 적혈구 증가, 백혈구 감소, 임파구 감소에 효과가 있으며, 또한 혈관 확장으로 인한 혈압 강하 작용이 있고, 그의 수침액은 혈중 당량을 증가시켜 초기 당뇨병에 유효하다.
상기 단풍마(Dioscorea quinquelobata Thunb)는 여러해살이 덩굴풀이며, 굵고 단단한 뿌리줄기가 있고, 질긴 줄기는 길게 뻗어나가며 다른 물체로 기어오르는 특징이 있다. 잎은 서로 어긋나게 자리하는데 전체적으로 심장꼴인 잎 몸은 5~9 갈래로 중간 정도까지 갈라지고, 갈라진 조각의 끝은 뾰족하며 가장자리는 밋밋하다.
상기 단풍마의 잎의 길이는 6~12cm이고, 암꽃과 수꽃이 각기 다른 포기에 핀다. 꽃은 모두 잎겨드랑이로부터 자라나는 긴 꽃대에 이삭 모양으로 달려 아래로 처진다. 수꽃 이삭은 여러 갈래로 갈라지는데 암꽃 이삭은 갈라지지 않는다. 대롱 모양인 꽃은 여섯 갈래로 끝이 갈라진다. 꽃의 지름은 4mm 안팎이고 빛깔은 초록빛이다.
상기 단풍마의 성미는 쓰고 성질은 약간 차며 간경 및 폐경으로 귀경한다. 한방학적으로 단풍마는 풍습을 없애고 혈을 잘 돌게하며 경락을 통하게 하며 담을 삭이고 기침을 멈추는 것으로 알려져 있다. 본 발명에서 상기 단풍마는 쌀뜨물에 20 내지 30시간 동안 침지시킨 후 건조하고 볶아 사용할 수도 있다.
상기 목단피는 작약과(Paeoniaceae)에 속하는 목단(Paeonia suffruticosa Andrews)의 뿌리 껍질로 한의학적으로 청열양혈, 활혈산어, 퇴허혈 작용을 가지고 있어 널리 이용되어 왔다. 목단피 추출물은 생체 외에서 케모카인(chemokine)의 일종인 IL-8 및 MCP-1의 유리를 억제하며, 또한 염증 매개인자인 iNOS 및 COX-2 발현은 억제하는 활성이 보고되었다. 본 발명에서 상기 목단피는 까맣게 볶아 사용할 수도 있다.
상기 황백(, Phellodendri Cortex)은 운향과(Rutaceae)에 속하는 황백 나무(Phellodendron amurense Rupr)의 껍질을 말린 것으로, 나무줄기에서 껍질을 벗겨내어 조피(거친껍질)를 제거하거나 썰어서 햇볕에 말려 약제로 사용하며 혈압강하작용, 중추신경 억제작용(KulkarniSK & Dhir A, Phytother Res 24, pp317-324, 2009), 아세틸콜린 증강작용 및 항염작용(Uchiyama T et al,Yakugaku Zasshi 109, pp672-676, 1989), 항산화와 노화예방 효과(Kong LD et al,Zhongguo Zhong Yao Za Zhi 26, pp245-248, 2001), 및 피부면역력을 강화시키는 효능이 있다고 알려져 있다. 본 발명에서 상기 황백은 술에 20 내지 30시간 동안 침지시킨 후 건조하고 볶아 사용할 수도 있다.
상기 숙지황은 지황(Rehmannia glutinosa Liboschtz var purpurea Makino)의 생뿌리를 물에 씻어 술로 여러 번 쪄서 검은색의 윤기가 돌게하여 말린 것이다. 지황은 현삼과 식물이며, 높은 15~25cm 되는 여러해살이풀이다. 긴 타원형 또는 긴 달걀 모양이다. 이른 여름에 줄기 윗부분에 가지색을 띤 분홍색 꽃이 핀다. 뿌리에는 이리도이드 배당체인 카탈폴 및 만니톨, 레흐마닌이 있다. 뿌리를 그대로 말리거나 쪄서 말릴 때 검은색을 띠는 것은 카탈폴에 의한 것이다.
지황(, Rehmannia glutinosa Libosch var purpurea Makino 또는 동속식물의 뿌리)은 현삼과(Scrophulariaceae)에 속하는 다년생 초본으로 지황의 근엽과 잔뿌리를 제거하고 흙을 깨끗이 씻은 것을 생지황 또는 선지황이라 한다. 생지황은 보혈, 강장, 진정, 빈혈, 토혈, 하혈, 자궁출혈, 결핵성 쇠약 등에 그 효능이 널리 인정되고 있다. 보통은 생지황을 말린 건지황을 황주 또는 백주등의 술에 넣고 주침(酒侵, 약재에 술이 배어들도록 술에 일정한 시간 동안 담가 두는 것)하여 증숙(蒸熟, steaming)한 후 건조하는 것을 반복한 것을 숙지황(Rehmanniae Radix Preparata)이라고 일반적으로 분류하고 각각 약리를 달리하여 사용하고 있다.
상기 숙지황은 보혈, 자음의 효능이 있으며, 생리불순, 허약체질, 어린이의 발육부진, 치매, 조루증, 발기부전 등에 사용되고 있다. 또한, 최근 혈액 순환을 개선시켜 여러 만성 질환의 말초미세 순환을 개선시키는 효과, 혈중 과산화물 제거효소(superoxide dismutase), 글루타티온 산화효소(glutathione peroxidase)의 증가와 지질과산화의 억제, 면역증가 효과, 항암작용, 섬유소용해계(fibrinolytic system)의 활성증가, 폴리브렌에 의해 유도되는 적혈구 집괴(polybrene-induced erythrocyte aggregation)의 감소, 적혈구변형능(erythrocyte deformability)과 ATP 함량(content)의 증가 등이 보고되고 있다.
상기 현삼(Scrophularia Buergeriana)은 현삼과에 속하는 다년생 초본식물이다. 현삼은 산지에서 자라고, 해열제로 인후염, 종기, 림프선염에 사용하며, 울릉도에서 자라는 섬현삼(S takesimensis), 산지에서 자라는 큰개현삼(S kakudensis) 및 토현삼(S koraiensis) 등이 있다.
상기 현삼(玄蔘, Scrophularia Buergeriana)은 한방에서 알려져 있는 효능으로는 녹농간균에 대하여 억제 작용이 있고 혈관을 확장하고 혈압을 하강시키며, 혈당을 낮추는 것으로 알려져 있다. 소량을 쓰면 가벼운 강심작용을 다량을 쓰면 심장 중독을 일으키는 한약재이다. 본 발명에서 상기 현삼은 술에 20 내지 30시간 동안 침지시킨 후 건조하고 볶아 사용할 수도 있다.
상기 택사(澤瀉, Alismatis Rhizoma)는 택사과(Alismataceae)에 속하는 다년생 소택(연못) 식물로서, 국내에는 택사(A canaliculatum)와 질경이택사(A orientale (Sam) Juz) 2종류가 분포한다. 그중에서도 질경이택사를 주로 한약재로 이용하는데, 질경이택사의 덩이줄기의 잔뿌리 및 주피를 제거하여 한약재인 택사(Alismatis Rhizoma)로서 사용한다. 택사는 요독증 개선 작용이 있으며, 고혈압, 알레르기, 콜레스테롤 혈증, 지방간, 고지혈증의 치료에 사용된다. 본 발명에서 상기 택사는 500g 당 식염수 6.25g를 혼합한 후 볶아 사용할 수도 있다.
상기 황기(Astragalus membranaceus)는 쌍떡잎식물 장미목 콩과의 여러해살이풀로서 산지의 바위틈에 자란다. 한국, 일본, 만주, 중국 북동부, 시베리아 동부 등지에 주로 분포한다. 황기는 흔히 약초로서 재배하며 한방에서는 가을에 채취하여 노두와 잔뿌리를 제거하고 햇빛에 말린 것을 한약재의 황기라 하며, 강장, 지한, 이뇨, 소종 등의 효능이 있어 신체허약, 피로권태, 기혈허탈, 탈항, 자궁탈, 내장하수, 식은땀, 말초신경 등에 처방한다.
상기와 같은 약효를 지니는 황기는 한약재의 일부로 사용되나, 가장 흔히 사용되는 형태로는 여름 복날 삼계탕을 섭취시 닭과 함께 황기를 넣고 끓이는데 사용되고 있다. 황기는 주로 식품보다는 한방에서 주로 사용되어지는데 지산, 이뇨, 강장, 혈압강하 등의 목적으로 사용되며 약리실험에서도 이뇨작용, 강장작용, 혈압강하작용, 혈당강하작용, 면역증강작용, 항종양작용, 항바이러스 작용 등 다양한 생리적 기능성이 확인되고 있으며 황기에 존재하는 생리활성 물질로는 isoflavone 배당체 중의 하나인 formononetin과 triterpenoide glycoside 및 β-sitosterol, stimast-4-en-6β-ol-3-one 등의 물질과 saponin류로써 astragaloside류 및 당이 주성분으로 알려져 있다. 본 발명에서 상기 황기는 볶아 사용할 수도 있다.
상기 사상자(Cnidi Fructus)는 산형과에 속하는 식물인 사상자(Torilis japonica Decandolle) 또는 벌사상자(Cnidium monnieri (L) Cussion)의 건조한 열매를 의미하고, 천연, 잡종 또는 변종의 사상자를 모두 포함한다. 사상자는 미나리과의 두해살이풀로서 어린순은 나물로 먹고, 열매는 수렴제, 소염제, 살충제 등으로 쓴다. 또한, 사상자는 피부질환에 대해서도 치료효과가 있으므로 피부습진, 알레르기성 피부염 및 진물 등의 외용약으로도 이용되어 왔다. 본 발명에서 상기 사상자는 볶아 사용할 수도 있다.
상기 선모(仙茅)는 수선화과(Amarylidaceae)에 속하는 선모(Curculigo orchioides Gaertner)의 뿌리줄기로, 신장을 따뜻하게 하고 양기를 보충해주며 근육과 뼈를 강하고 튼튼하게 하는 등의 효능이 있으며, 그 성분으로 쿠르쿠리고시드, 리코린, 지방족 히드록시케톤류, 뮤코라지 등이 함유되어 있으며, 선모의 메탄올 추출물이 대식세포의 탐식능을 향상시키며, 글루코시드 분획에서 효과가 있음이 알려져 있다. 본 발명에서 상기 선모는 대나무칼이나 구리칼로 잘라 쌀뜨물에 담궜다가 빨간물이 나오면 버리고 술에 20 내지 30시간 동안 침지시킨 후 건조하고 볶아 사용할 수도 있다.
상기 패모는 백합과(Liliaceae)에 속하며, 라틴명은 Fritillariae Bulbus이다. 상기 패모는 심아를 제거한 대패와 심아를 제거하지 않은 주패를 포함한다. 상기 대패는 비늘줄기로 비늘줄기 바깥층의 한 개로 된 비늘잎은 거의 초생달 모양을 이루고 높이 1~2cm, 너비 20~35mm, 바깥면은 유백색~연한 노란색이며 안쪽 면은 흰색 또는 연한 갈색이고 하얀 가루가 덮여 있다.
상기 패모(Fritillaria thunbergii)의 뿌리는 종양 및 유방암과 같은 항암효과 및 고혈압 강하 작용을 가지고, 기침, 가래, 폐렴, 편도염, 인후동통, 기관지염, 구갈에 효과가 있으며, 베인 상처, 종기, 벌레 물린데에 효과가 있는 것으로 알려져 있다.
상기 구기자(Lycium chinense)는 가지 과에 속하는 구기자나무의 열매이다. 구기자는 원형 또는 타원형의 장과이고 건조시켰을 때에 겉이 쭈글쭈글하고 속에는 많은 씨가 들어있다. 구기자의 성분은 베타인(betaine), 제아잔틴(zeazantin), 카로틴(carotene), 티아민(thiamine), 비타민 A, B1, B2, C, 피잘리엔 및 β-시스토스테롤(β-sistosterol)과 각종 무기성분 등을 다량 함유하고 있고, 저비중 콜레스테롤의 장내흡수를 억제 강하하여 간장보호에 효과적이며, 고혈압, 협심증, 동맥경화 및 당뇨병 등의 성인병에 탁월한 예방 및 치료 효과를 지니고 있는 동시에, 시력보호 효과도 뛰어난 것으로 알려져 있다. 본 발명에서 상기 구기자는 막걸리에 20 내지 30시간 동안 침지시킨 후 9증 9포로 쪄서 사용할 수도 있다.
상기 상백피는 뽕나무과에 속한 낙엽교목인 뽕나무속 식물의 근피로써 예로부터, 다양한 효과가 있다고 알려져 있다. 상백피 추출물이 인간 기관지 상피세포에서 기도점막의 염증을 유발 시키는 IL-8과 GM-CSF mRNA를 유의하게 감소시킴도 보고되어 있다. 또한, LPS로 염증이 유발된 마크로파지 2647 세포에서 상백피 추출물이 IL-1β 및 TNF-α의 분비를 감소시킴도 보고되어 있는 등 많은 연구에서 상백피 추출물의 항염증 및 항알레르기 작용이 연구되었다.
상기 황정은 백합과에 속한 다년생 초본인 층층갈고리둥굴레의 뿌리줄기를 건조한 것으로, 뿌리에는 글로코키닌, 사포닌 성분 등이 포함되어 있어, 힘줄과 뼈를 든든하게 하고, 비위를 보하며, 식용부진, 기력 회복, 당뇨, 노화방지 및 피부미용 등에 좋다. 본 발명에서 상기 황정은 술에 20 내지 30시간 동안 침지시킨 후 9증 9포로 쪄서 사용할 수도 있다.
상기 오미자는 목련과(Magnoliaceae)에 속하며, 라틴명은 Schizandrae Fructus이다. 상기 오미자는 고르지 않은 구형-편구형을 이루고 지름 약 6mm로 어두운 적색-흑갈색을 띄며 바깥면에는 주름이 있고 때때로 흰 가루가 묻어 있다. 과육을 벗기면 길이 2-5mm의 콩팥모양의 씨가 1-2개 들어 있고 그 씨의 바깥면은 광택이 있는 황갈색-어두운 적갈색이며 등쪽에 명확한 봉선이 있다. 외종피는 벗겨지기 쉬우나 내종피는 배유에 밀착되어 있다. 상기 오미자는 땀을 멎게하는 효능, 갈증을 그치게 하는 효능, 폐가 허해서 숨이 차거나 기침을 개선시키는 효능 등이 보고 되어진다. 본 발명에서 상기 오미자는 볶아 사용할 수도 있다.
상기 반지련(Portulaca grandiflora Hooker)은 남아메리카가 원산이며 일반적으로 채송화라고도 지칭된다. 성분으로 전초에 알칼로이드(Alkaloid), 라보노이드(Flavonoid)류 등이 함유되어 있으며 주성분으로는 포츌랄(Portulal), 베타닌(Betanin), 베타시아닌(Betacyanin), 베타니딘(Betanidin) 등이 알려져 있다.
상기 반지련은 청열 작용으로 인후염이나 뜨거운 물에 데었을 때, 타박상과 피부가 헐어 생긴 발진 등 주로 외용약으로 사용된 바 있다(중약대사전, 도서출판 정담, 1997) 반지련의 선행연구로는 항돌연변이 활성(antimutagenic effect)(Zhongguo Zhong Yao Za Zhi, 15:617-22, 1990) 등이 보고된 바 있고, 기타 항산화 항염 활성(특허등록 제10-793019호), 신경 염증 또는 퇴행성 뇌신경 질환 예방 또는 치료 활성(특허공개 제10-2016-0025415호), 모발 성장 촉진 또는 탈모방지 활성(특허공개 제10-2012-0117128호)등이 보고된 바 있다.
한편, 본 발명에 따른 생약재를 이용한 건강보조식품 조성물은 상기 우슬, 지모, 백출, 당삼, 단풍마, 목단피, 황백, 숙지황, 현삼, 택사, 황기, 사상자, 선모, 패모, 구기자, 상백피, 황정, 오미자 및 반지련으로 이루어진 조성물 이외에, 숙잠 분말, 함초 분말, 초록입홍합 오일 및 초유분말을 더 포함할 수 있다.
구체적으로, 본 발명에 따른 생약재를 이용한 건강보조식품 조성물에서 상기 숙잠 분말 1 내지 3 중량부, 함초 분말 2 내지 4 중량부, 초록입홍합 오일 1 내지 3 중량부 및 초유분말 0.5 내지 1.5 중량부가 더 포함될 수 있다.
상기 숙잠 분말은 하기의 방법으로 제조된 숙잠 분말이 이용될 수 있다.
상기 숙잠 분말을 제조하기 위하여, 먼저, 숙잠을 준비할 수 있다.
상기 숙잠은 누에 몸속에 견사단백질이 가득 차 있는 누에로서 고치를 완전히 지어 번데기로 탈피하기 전까지의 견사단백질이 누에 몸속에 들어있는 누에를 말한다.
즉, 통상적으로 4면잠 누에의 경우 5령3일 누에란 누에가 마지막 잠을 자고 5령이 되어 3일째 되는 것으로, 약 3일이 지나면 체내에서 단백질 성분의 견사단백질(견사선)이 급속히 비대해지기 시작하고, 내장이 점차 퇴화되며 5령7일~8일 정도가 되면 견사단백질만으로 가득차게 되는데, 이때를 숙잠이라고 한다.
일반적으로 누에는 알에서 깨어나 4번의 탈피를 거쳐 누에고치를 짓고 누에번데기로 변태한 후 나방이 되어 알을 낳고 약 2주 후면 죽게 되는 일생의 과정을 가진 완전변태 곤충의 하나이다.
그리고 누에분말은 식용 및 약용으로 이용되고 있는 것으로, 누에 성분 및 약리효과에 대한 관심이 높아지고 있어 이와 관련된 여러 연구가 이루어지고 있다.

상기 뽕잎은 글루코사이드(Glucoside), 비타민 C, 비타민 B1, 비타민 B2 등 50여 종의 성분이 포함되어 있고 당뇨병 치료와 혈압 및 혈당 강하, 항암, 노화 방지, 동맥경화 등 성인병 예방에 효과가 있는 것으로 알려지고 있는데, 상기 뽕잎은 숙잠과 함께 가열되고, 상기 뽕잎의 향이 숙잠에 침투됨으로써, 상기 숙잠의 잡냄새를 제거할 수 있다.
상기 단계에서 상기 뽕잎은 상기 숙잠 전체 100 중량부에 대해 1 내지 5 중량부의 중량 비율로 혼합하고, 상기 가열은 120 내지 130℃의 온도의 수증기로 1 내지 3시간 동안 수행될 수 있다.
그 다음으로, 상기 가열하여 쪄진 숙잠을 동결 건조할 수 있다.
일반적으로 동결 건조 방법으로는 상압 동결 건조법과 감압 동결 건조법이 사용될 수 있는데, 본 발명에서는 상기 숙잠의 손상을 줄이기 위하여 0.5 내지 3mmHg의 압력 및 -65 내지 -55℃의 온도로 30 내지 50시간 동안 동결 건조하여 수행될 수 있다.
상기 단계에서 상기 동결 건조가 상기한 범위를 벗어나서 수행되는 경우에는 숙잠 내에 함유되어 있는 수분이 충분히 승화되지 않거나 과도하게 수분의 증발이 일어나 숙잠의 손상이 발생할 수 있고, 생물/생화학적인 활성의 유지가 어려울 수 있다.
이어서, 상기 동결 건조된 숙잠을 분쇄하여 숙잠 분말을 제조할 수 있다.
상기 숙잠의 분쇄는 분쇄기를 이용하여 당해 기술분야에서 공지된 다양한 방법으로 수행될 수 있다.
상기 함초 분말은 하기의 방법으로 제조된 함초 분말이 사용될 수 있다.
상기 함초 분말을 제조하기 위하여, 먼저, 함초를 준비한 후 세척할 수 있다.
상기 함초(Salicornia spp.)는 명아주 과의 한해살이 풀로 국내 자생하는 대표적인 식용 및 약용 염생식물이다. 상기 함초는 오랫동안 민간약으로 시력저하, 소화불량, 위장병, 간염, 신장병 등에 사용되어 왔고, 항산화, 항당뇨, 항암, 항고혈압, 항고지혈증, 피부미백 효과가 있음이 과학적으로 입증되어오고 있다.
다음으로, 상기 세척된 함초를 혼합용액에 침지시킬 수 있다.
상기 단계에서는 상기 세척된 함초를 혼합용액에 침지시킴으로써 함초가 변색되거나 쉽게 변질되는 것을 방지할 수 있는데, 상기 혼합용액은 포도즙, 레몬 과즙 및 죽염수로 이루어질 수 있다.
또한, 상기 단계에서 상기 포도즙은 혈전 방지 및 고혈압, 동맥경화 및 심장질환 등 성인병 예방에 좋고 기억력 향상에 도움을 주며, 상기 레몬 과즙은 비타민 C의 함량이 많고 산미가 강하며 피부건강, 피로회복, 감기예방, 두통 등에 효능이 좋으며, 상기 죽염수는 상기 함초를 살균하여 쉽게 부패하는 것을 방지할 수 있다.
또한, 상기 레몬 과즙과 죽염수는 함초가 시간이 경과함에 따라 변색되는 것을 방지할 수 있는데, 상기 혼합용액은 포도즙 1 중량부, 레몬 과즙 1 중량부 및 죽염수 8 중량부의 중량 비율로 혼합되어 제조되고, 상기 죽염수는 농도가 1 내지 3 중량%일 수 있다.
또한, 상기 단계에서는 상기 세척된 함초를 혼합용액 침지시킨 후 20 내지 25℃의 온도에서 50 내지 100분 동안 유지시켜 진행될 수 있다.
그 다음으로, 상기 혼합용액에 침지된 함초를 꺼낸 후 상기 함초를 수증기로 증숙할 수 있다.
상기 단계에서는 상기 포도즙, 레몬 과즙 및 죽염수의 성분이 침투된 함초를 계피가루 및 설탕의 혼합물과 혼합한 후, 증숙기에서 찌는 과정으로 이루어질 수 있는데, 상기 단계에서 상기 계피가루 및 설탕은 계피가루 20 내지 30 중량부 및 설탕 10 내지 20 중량부의 중량 비율로 혼합되고, 증숙기에서 120 내지 140℃의 수증기로 20 내지 4O분 동안 가열하여 찜으로써 수행될 수 있다.
상기 단계에서 상기 계피가루는 면역력을 높이고 혈액 순환을 촉진시켜 혈관질환 예방에도 도움을 주며 식중독 예방 및 살균, 살충 효과가 있으며, 상기 설탕은 침지된 함초의 표면 윤기 및 당도를 증진시키고 방부 효과가 있어 저장성을 향상시킬 수 있다.
이어서, 상기 증숙된 함초를 분쇄한 후 건조할 수 있다.
상기 단계에서는 상기 증숙된 함초를 입자가 큰 상태로 분쇄한 후 건조함으로써 상기 함초의 건조 효율을 증진시킬 수 있는데, 예를 들어, 상기 함초를 0.5 내지 2mm의 크기로 분쇄한 후, 상기 분쇄된 함초를 25 내지 35℃에서 20 내지 30 시간 동안 건조함으로써 진행될 수 있다.
다음으로, 상기 건조된 함초를 분쇄하여 분말화함으로써 함초 분말을 제조할 수 있다.
상기 단계에서는 상기 건조된 함초를 공지의 분쇄기를 이용하여 분쇄하여 분말화할 수 있다.
상기 초록입홍합 오일은 초록입홍합(Perna Canaliculus)을 사용하여 제조되는데, 상기 초록입홍합(Perna Canaliculus)은 뉴질랜드 해안에 서식하는 홍합의 일종으로 녹색홍합(Green-lipped Mussel)으로 불리며 학명은 Perna Canaliculus 이다.
상기 초록입홍합은 강렬한 자외선에 대항할 수 있도록 진화된 플랑크톤을 주 먹이로 하므로, 다른 지역에서 자생하는 홍합에서는 볼 수 없는 독특한 성분 즉, 항산화와 항염에 관련된 오메가 불포화 지방산을 함유하고 있다고 보고 되었다.
또한, 상기 초록입홍합에서 발견된 불포화 지방산은 오메가-3 지방산을 함유하는 물고기 기름, 아마인유, 달맞이 꽃 오일 등보다도 200배 이상의 강한 염증 억제효과가 있는 것으로 임상실험을 통해 알려졌다. 오메가-3계열의 지방산은 오메가-6계열의 리놀레산과 경쟁적으로 작용함으로써 오메가-6 계열 지방산으로부터 만들어지는 염증 매개 물질의 생성을 방해하여 동맥 경화증과 류머티스 관절염 등의 염증성질환 발생을 억제하는 것으로 규명된 바 있다.
상기 초록입홍합 오일은 하기의 방법으로 제조된 초록입홍합 오일이 사용될 수 있다.
먼저, 초록입홍합의 껍질을 제거하고, 상기 껍질이 제거된 초록입홍합을 세척할 수 있다.
상기 단계에서는 상기 껍질이 제거된 초록입홍합을 10 내지 20℃ 온도 및 1 내지 3(w/w)% 농도 범위를 가지는 탄산수소나트륨 용액으로 세척할 수 있는데, 상기 탄산수소나트륨(NaHCO3)은 식품첨가물로도 이용되는 것으로, 독성이 없으며 침투, 확산, 팽창 등의 기능을 가질 수 있다.
다음으로, 상기 세척된 초록입홍합을 건조할 수 있다.
상기 세척된 초록입홍합의 건조는 제1 건조 및 제2 건조로 이루어질 수 있는데, 상기 제1 건조는 상기 세척된 초록입홍합을 70 내지 80℃의 온도에서 3 내지 5시간 동안 건조시켜 상기 세척된 초록입홍합에 잔류하는 수분을 1차로 제거할 수 있다.
또한, 상기 제2 건조는 상기 제1 건조된 초록입홍합을 열풍으로 건조할 수 있는데, 상기 제2 건조는 상기 제1 건조된 초록입홍합을 110 내지 130℃ 온도의 열풍을 1 내지 3시간 동안 가하여 줌으로써 수행될 수 있다.
그 다음으로, 상기 제2 건조된 초록입홍합을 분쇄하여 분말화한 후 숙성하여 초록입홍합 숙성분말을 제조할 수 있다.
상기 단계에서 상기 제2 건조된 초록입홍합의 분쇄는 상기 제2 건조된 초록입홍합을 5 내지 10mm의 입경으로 분쇄하여 분말화하고, 상기 분말화된 초록입홍합의 숙성은 상기 분말화된 초록입홍합을 5 내지 10℃의 온도에서 3 내지 7시간 동안 보관함으로써 수행될 수 있다.
이어서, 상기 초록입홍합 숙성분말에 효소와 산을 혼합한 후 보관하여 효소 분해할 수 있다.
즉, 상기 단계에서는 상기 초록입홍합 숙성분말에 효소와 산을 혼합한 후 상기 효소 및 산이 혼합된 초록입홍합 숙성분말을 36 내지 38℃의 온도에서 20 내지 30시간 동안 보관함으로써 효소 분해할 수 있다.
상기 단계에서 상기 효소로는 시중에 판매되고 있는 공지된 프로테아제(Protease), 키티나제(chitinase), 펙티나제(Pectinase) 및 자일라나제(Xylanase)가 이용되고, 상기 산으로는 구연산 및 아스코르빈산이 이용될 수 있는데, 구체적으로는 상기 초록입홍합 숙성분말 전체 100 중량부에 대해 상기 프로테아제(Protease) 0.01 내지 0.03 중량부, 키티나제(chitinase) 0.03 내지 0.05 중량부, 펙티나제(Pectinase) 0.005 내지 0.009 중량부, 자일라나제(Xylanase) 0.008 내지 0.012 중량부, 구연산 0.01 내지 0.1 중량부 및 아스코르빈산 0.01 내지 0.1 중량부의 중량 비율로 혼합할 수 있다.
다음으로, 상기 효소 분해된 초록입홍합 숙성분말에 유산균 배양액을 혼합한 후 발효하여 초록입홍합 발효물을 제조할 수 있다.
상기 단계에서는 효소 분해된 초록입홍합 숙성분말 100 중량부에 대해 유산균 배양액 1 내지 3 중량부의 중량 비율로 혼합한 후, 40 내지 42℃의 온도에서 20 내지 30시간 동안 발효함으로써 진행될 수 있다.
또한, 상기 단계에서 상기 유산균 배양액은, 1.2N HCl을 이용하여 배지의 pH를 2.8~3.2로 조정한 다음 김치로부터 유산 균주를 분리하고, 상기 분리된 유산 균주를 MRS broth(Oxoid, England)를 이용하여 37~39℃에서 20 내지 25시간 동안 배양한 후 1×1.08 ~ 5×1.08 CFU/mL이 되도록 희석하며, 이후 10,000 내지 15,000rpm에서 10~15분간 원심분리하여 상청액(supernatant)만을 분리하고, 상기 상청액(Supernatant)을 0.45㎛ 시린지 필터(syringe filter)로 여과 후, 상기 여과된 상청액 100 중량부에 대해 멸균한 증류수 1,000 내지 2,000 중량부의 중량 비율로 혼합하여 희석함으로써 제조되고, 상기 유산균은 발효 식품인 김치로부터 분리된 유산 균주로, 구체적으로, 상기 유산 균주로는 락토바실러스 쿠르바투스(Lactobacillus curvatus), 바이셀라 비리데센스(Weissella
viridescens), 락토바실러스 플란타럼(Lactobacillus
plantarum) 및 류코노스톡 락티스(Leuconostoc lactis)로 이루어진 군에서 선택된 어느 하나 이상의 공지된 유산 균주가 사용될 수 있다.
그 다음으로, 상기 초록입홍합 발효물을 이용하여 오일을 분리함으로써 초록입홍합 오일을 제조할 수 있다.
상기 단계에서 상기 초록입홍합 발효물을 이용하여 오일을 분리하는 등의 구성은 당해 기술 분야에서 통상의 지식을 가진 자에게는 공지의 기술인 바, 설명의 편의 및 본 발명의 기술적 사상의 명확성을 위하여 이에 대한 구체적인 설명은 생략하기로 한다.
상기 초유분말은 초유를 이용하여 제조되는데, 상기 초유는 포유동물이 분만 후 1주일 이내에 분비하는 유즙을 말하며, 최근에는 분만 후 48~72시간 이내에 분비하는 유즙을 초유로 정의하고 있다.
일반적으로 48시간 이내에 분비되는 초유 속에는 기능성 물질들이 다량 함유되어 있다. 화학적으로 젖소 초유는 매우 복잡한 액상의 물질로서 단백질, 탄수화물, 지방, 비타민, 미네랄 등이 풍부한데, 갓 태어난 송아지에게 영양소를 공급하는 풍부한 에너지원이며 병원성 미생물에 대한 방어, 미성숙한 장관의 발달, 각종 장기와 조직의 성장, 면역체계의 발달 등에 관여하는 면역조절인자, 생리활성인자, 항균인자, 성장호르몬, 성장인자, 세포분열 활성인자 등을 다량 함유하고 있다.
이러한 면역적, 생리활성적 특성을 지니고 있는 물질과 인자들은 주로 초유 유장(whey)에 함유되어 있으며, a-lactalbumin, b-lactoglobulin이 70~80%를 차지하고 이외의 단백질 성분들은 serum albumin, lactoferrin, immunoglobulin, growth factor, lysozyme, 미량의 bioactive factor 등이며 각각의 특성을 규명하기 위하여 많은 연구들이 진행되고 있다(Korean J. Food Sci. Technol. Vol.34, No.4, pp.694~699 (2002)).
연구에 따르면, 죽상동맥경화증 및 심혈관 질환은 변경된 면역과 관련이 있으며, 심장 및 혈관 조직이 손상되면 항체가 생성되어 면역계가 더욱 손상되며, 여러 연구에서 클라미디아 감염성 폐렴 등은 죽상 경화성 병변의 초기 단계 활성화 및 발달 원인으로 중점을 두고 있다. 소 초유의 면역인자는 이 박테리아와 싸우는데 도움이 되며, 특히 초유에 함유된 프롤린-리치-폴리펩타이드(proline-rich-polypeptide)는 심근합병증으로 진단된 환자에 대해 심근 보호 효과를 갖는 것으로 예상되며, 면역 반응을 조절하여 손상된 혈관의 콜레스테롤 침착을 억제하고, 성장인자는 심장 세포의 복구 및 재생의 촉진, 측부 순환(collateral circulation)을 위한 새로운 관상동맥 혈관 재생을 촉진한다.
또한, 혈압조절 기능과 관련하여, 초유에 존재하는 펩타이드는 레닌-안지오텐신 계에 통합적으로 관여하는 효소인 안지오텐신 I-전환효소(ACE)를 억제하여 혈액의 부피를 낮춤으로서 혈압을 낮추고 심장의 산소소모량을 감소시켜 항 고혈압 및 심혈관질환 개선 효능이 있음을 연구결과로 보고된 바 있다.
최근 연구에서 심근경색 쥐(rat) 모델 대상 연구에서 28일간 초유 섭취 후 지질과 산화(항산화)의 억제 및 심장 손상 지표물질(CKMB, LDH)의 감소를 통해 심장 조직 손상 보호 효능이 관찰되었으며, 심부전 치료제(ACE 억제제, 혈압조절제 enalapril)와 복합 섭취 시 그 효능이 더욱 증진됨을 확인되었다(Journal of pharmacology and toxicology 9(1):37-45, 2014).
또한, 상기 초유에는 Insulin-like Growth Factor-1(IGF-1)이 함유되어 있는데, IGF-1은 전체적인 성장을 촉진하며, 세포 성장과 증폭을 조절할 수 있는 능력을 가지고 있는 것으로 알려져 있다.
본 발명에서 상기 초유분말은 하기의 방법으로 제조된 초유분말이 사용될 수도 있다.
상기 초유분말을 제조하기 위하여, 먼저, 소 초유를 준비하고 상기 준비된 소 초유에 효소를 혼합한 후 보관하여 효소 분해할 수 있다.
즉, 상기 단계에서는 상기 준비된 소 초유에 효소를 혼합한 후 상기 효소가 혼합된 소 초유를 36 내지 38℃의 온도에서 20 내지 30시간 동안 보관함으로써 효소 분해할 수 있다.
상기 단계에서 상기 효소로는 시중에 판매되고 있는 공지된 프로테아제(Protease) 및 자일라나제(Xylanase)가 이용될 수 있는데, 예를 들어, 상기 프로테아제(Protease)로는 키모트립신(chymotrypsin)이 사용될 수 있고, 상기 키모트립신은 단백질 분해를 일으키는 소화 효소이다.
즉, 상기 키모트립신은 단백질 분해를 일으키는 소화 효소로 펩티드 결합을 끊는데, 상기 키모트립신은 포유동물과 다른 생물체의 소화계에서 단백질 분해효소로 작용한다. 상기 키모트립신의 기질로는 트립토판, 티로신, 페닐알라닌, 류신, 메티오닌이 있으며 이들의 카복시 말단을 잘라낸다. 다른 단백질 분해효소와 같이 펩티드 결합을 끊는 반응을 일으킨다.
또한, 상기 자일라나제(Xylanase)는 자일란(xylan)을 자일로올리고당(xylo-oligosaccharide)나 자일로스(Xylose)로 가수분해하는 효소로서 바이오에너지, 식품, 섬유, 제지 및 펄프 산업에 사용되고 있는 효소이다. 지구상에서 가장 풍부한 생물자원인 식물성 섬유소를 분해하고 당화하여 바이오에탄올 생산하는 분야에서 연구가 활발히 진행되고 있으며 고급 용지의 생산 및 폐지재생, 과일음료 및 맥주의 혼탁물 제거, 커피와 식물성 기름 및 전분의 추출 그리고 단위동물의 사료이용 효율 증대와 같은 용도로 이미 산업적으로 널리 사용되고 있는 효소이다.
구체적으로는 상기 단계에서 상기 효소는 상기 준비된 소 초유 전체 100 중량부에 대해, 상기 키모트립신과 같은 프로테아제(Protease) 0.01 내지 0.03 중량부 및 자일라나제(Xylanase) 0.008 내지 0.012 중량부의 중량 비율로 혼합될 수 있다.
다음으로, 상기 효소 분해된 소 초유에 배양액 및 유산균으로 이루어진 발효액을 혼합할 수 있다.
상기 단계에서 상기 유산균으로는 젖산균(Lactobacillales), 스트렙토코쿠스 테르모필루스(Streptococcus
thermophiles) 및 비피도박테리움 롱굼(Bifidobacterium longum)으로 이루어진 군에서 선택된 어느 하나의 공지된 유산균이 이용될 수 있고, 상기 배양액은 펩톤, 맥아추출물(Malt extract), 시트르산나트륨(Sodium citrate), 제2인산칼륨(Potassium phosphate dibasic), 글루코스(Glucose) 및 증류수로 이루어지고, 상기 배양액은 펩톤 15 내지 25 중량부, 맥아추출물 5 내지 15 중량부, 시트르산나트륨 0.5 내지 1.5 중량부, 제2인산칼륨 0.1 내지 0.3 중량부, 글루코스 1 내지 3 중량부 및 증류수 100 내지 200 중량부의 중량 비율로 혼합되어 이루어질 수 있다.
상기 배양액에서 상기 펩톤은 단백질을 공급해주는 질소원으로서 미생물의 성장에 필요한 단백질을 공급해 주며, 상기 맥아추출물은 각종 미네랄, 비타민 B군, 아미노산이 포함되어 있어 미생물 성장에 도움을 줄 수 있으며, 상기 시트르산나트륨은 배양액의 pH를 조절하여 미생물의 성장에 최적화된 상태를 유지해 주는 무기질로, 영양을 공급해 줄 수 있고, 상기 글루코스는 유산균이 당을 대사하여 유기산을 만들어내는 중요한 성분으로 기능할 수 있다.
그 다음으로, 상기 발효액이 혼합된 소 초유를 발효시켜 소 초유 발효물을 제조할 수 있다.
상기 단계에서 상기 발효는 상기 발효액이 혼합된 소 초유를 38 내지 42℃의 온도에서 1 내지 3일 동안 보관하여 발효시킬 수 있다.
이어서, 상기 소 초유 발효물을 여과하여 불순물을 제거하고 동결 건조한 후 분쇄하여 초유분말을 제조할 수 있다.
상기 단계에서는 공지의 여과기를 이용하여 소 초유 발효물을 여과함으로써 상기 소 초유 발효물에 잔류하는 불순물을 제거할 수 있고, 상기 여과된 소 초유 발효물을 -40℃ 내지 -30℃ 온도의 범위에서 1 내지 3시간 동안 동결 건조한 후 분쇄하여 초유분말을 제조할 수 있다.
이하, 본 발명에 따른 생약재를 이용한 건강보조식품에 대하여 바람직한 실시예를 들어 더욱 상세하게 설명한다.
본 발명에 따른 생약재를 이용한 건강보조식품은 우슬, 지모, 백출, 당삼, 단풍마, 목단피, 황백, 숙지황, 현삼, 택사, 황기, 사상자, 선모, 패모, 구기자, 상백피, 황정, 오미자 및 반지련으로 이루어진 조성물을 분쇄하여 생약재 분말을 제조하고, 상기 생약재 분말을 홍국균으로 발효한 후 분말, 과립, 정제, 캡슐, 음료 또는 추출액 등의 형태로 성형함으로써 제조될 수 있다.
상기 홍국균(Monascus sp.)은 붉은 색소를 함유하고 있는 누룩 곰팡이의 일종으로, 혈압, 혈당강하, 항균, 항암 및 콜레스테롤 수치 저하 등에 탁월한 효과를 나타내는 균류이다. 또한, 상기 홍국균은 한방에서는 어혈해소제로 사용되어지고 있으며 혈압을 떨어뜨리는데 효과가 높으며 콜레스테롤을 분해하는 성분인 모나콜린- K라는 물질을 분비하는 것으로 알려져 있다.
본 발명에서는 시중에 판매되고 있는 공지된 홍국균을 사용하여 상기 생약재분말을 발효할 수 있는데, 예를 들어, 상기 생약재 분말과 홍국균을 혼합한 후 60 내지 70℃의 온도에서 20 내지 50시간 동안 발효함으로써 진행될 수 있다.
이하, 본 발명에 따른 생약재를 이용한 건강보조식품에 대한 실시예를 들어 더욱 구체적으로 설명하기로 한다.
< 실시예 1 >
먼저, 우슬 5 중량부, 지모 5 중량부, 백출 5 중량부, 당삼 5 중량부, 단풍마 5 중량부, 목단피 5 중량부, 황백 5 중량부, 숙지황 5 중량부, 현삼 5 중량부, 택사 5 중량부, 황기 5 중량부, 사상자 5 중량부, 선모 5 중량부, 패모 5 중량부, 구기자 5 중량부, 상백피 5 중량부, 황정 5 중량부, 오미자 5 중량부 및 반지련 5 중량부로 혼합된 생약재를 분쇄하여 생약재 분말을 제조하였고, 상기 생약재 분말과 홍국균을 혼합한 후 65℃의 온도에서 35시간 동안 발효하였다.
이어서, 상기 발효된 생약재 분말을 추출하여 생약재 유용성분이 추출된 추출액을 제조하였고, 상기 추출액에서 고형분을 분리하여 제거함으로써, 생약재의 유용성분이 함유된 액상 추출액을 제조하였다.
< 실시예 2 >
실시예 1과 동일한 조성물을 이용하여 액상 추출액을 제조하였는데, 실시예 2에서는 생약재로 숙잠 분말 2 중량부, 함초 분말 3 중량부, 초록입홍합 오일 2 중량부 및 초유분말 1 중량부가 더 포함되도록 하였다.
실험예 1. 독성 실험
실시예 1, 2의 액상 추출액을 단기간에 과량을 섭취하였을 때 급성적(24시간 이내)으로 동물체내에 미치는 독성을 조사하고, 치사율을 결정하기 위하여 본 실험을 수행하였다. 일반적인 마우스인 ICR 마우스 계통 30마리를 대조군과 실험군에 각각 10마리씩 배정하였다. 대조군에는 PEG-400/tween-80/EtOH(8/1/1, v/v/v)만을 투여하고, 실험군은 본 발명의 실시예 1, 2의 액상 추출액을 상기 PEG-400/tween-80/EtOH(8/1/1, v/v/v)에 녹여 각각 경구 투여하였다. 투여 24시간 후에 각각의 치사율을 조사한 결과, 대조군과 2g/㎏/day 농도의 본 발명의 실시예 1, 2의 액상 추출액을 투여한 실험군에서 마우스가 모두 생존하는 것으로 확인되었다.
실험예 2. 세포 독성 평가 시험(MTT assay)
한국세포주은행에서 TM3(BALB/cnu
/+ mouse)를 분양받아 실험에 사용하였다. 배지는 DEME에 10% FBS와 1% penicillin/streptomycin을 첨가한 것을 이용하였고, 배지에서 37℃, 5% CO2 조건에서 24시간 배양하며 실험에 사용하였다.
TM3 세포를 96-well plate에 2x104 cell/well이 되도록 넣고 인큐베이터에서 37℃, 5% CO2 조건으로 12시간 배양하였다. 12시간 후 배지를 모두 버린 후 실시예 1을 원액, 10배, 30배 희석하여 처리하고, 37℃에서 24시간 배양한 후 배지를 모두 버리고 MTT(Thiazoly blue tetrazolium bromide, 1 mg/ml) 시약이 포함된 배지에 4시간 동안 배양하였다. 배지를 모두 버리고 DMSO를 200ul씩 넣은 후 세포 내 formazan crystal을 용해시키고 흡광도 570nm에서 흡광도를 측정하여 그 결과를 도 1에 나타내었다. 대조구는 실시예 1 대신에 증류수로 처리한 증류수 처리구를 의미한다.
도 1은 실시예 1의 액상 추출액에 대한 MTT assay를 통한 세포독성 결과를 나타낸 그래프이다.
도 1을 참조하면, 실시예 1의 원액, 10배, 30배 희석하여 처리한 결과 모든 농도에서 세포독성은 없는 것을 확인할 수 있다.
실험예 3. 남성호르몬 합성 관련 효능 평가 실험 진행(in vitro test)
실시예 1의 처리에 따른 TM3 cell에 대한 testosterone 변화 정도를 실험하였다. Standard는 kit로 제공된 50,000pg/㎖ testosterone을 phosphate buffer saline(PBS)을 이용하여 0, 1.95, 7.81, 31.25, 125, 500, 2000pg/㎖의 standard를 만들었다. Assay buffer는 고순도의 증류수를 이용하여 1:10까지 희석하고, wash buffer는 고순도의 증류수를 이용하여 1:20까지 희석하였다. 측정방법은 6-well plate에 1×106 cells/well의 세포를 넣고 37℃, 5% CO2의 조건으로 12시간 배양하여 세포를 well plate의 바닥에 붙인 다음, 배지를 모두 버린 후 원액, 10배 및 30배로 희석한 실시예 1 액상 추출액을 동일한 용량으로 넣고 37℃, 5% CO2의 조건으로 24시간 배양한 후 배지를 회수하여 testosterone을 측정하였다. 즉 회수된 배지 100㎕를 goat anti-mouse IgG microtiter plate(assay designs, USA)에 옮긴 후 testosterone EIA antibody를 첨가 후 1시간 동안 배양하였다. 배양이 끝난 후 wash buffer로 세척하고, 세척 후 stop solution을 첨가한 후 반응을 중지하고 405nm에서 microplate reader를 이용하여 흡광도를 측정하였다. 실험결과를 도 2에 나타내었다. 대조구는 실시예 1 대신에 증류수로 처리한 증류수 처리구를 의미한다.
도 2는 실시예 1의 액상 추출액의 테스토스테론(testosterone) 호르몬 활성 평가결과를 나타낸 그래프이다.
도 2를 참조하면, 대조구에 비해 실시예 1을 원액 처리시 testosterone이 2.3배 정도 증가하였으며, 10배와 30배로 희석한 경우에도 testosterone 호르몬 수치가 증가하는 것을 확인할 수 있다.
실험예 4. 관능 평가
실시예 1의 액상 추출액을 40인의 성인남녀로 구성되는 평가단에서 30㎖를 물과 함께 복용하게 하고, 복용의 편의성을 아래 각 항목에 따라 5점 척도법에 의거하여 관능 평가하여 그 결과를 아래 표 1에 나타내었다(1: 아주 나쁨, 2: 나쁨, 3: 보통, 4: 좋음, 5: 매우 좋음)

상기 [표 1]을 참조하면, 실시예 1의 액상 추출액은 맛, 향, 목넘김, 기호도 등의 관능성이 우수함을 확인할 수 있다.
이상, 본 발명의 바람직한 일 실시예를 설명하였지만, 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자라면 본 발명이 그 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 일 실시예는 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
Claims (6)
- 생약재로 우슬, 지모, 백출, 당삼, 단풍마, 목단피, 황백, 숙지황, 현삼, 택사, 황기, 사상자, 선모, 패모, 구기자, 상백피, 황정, 오미자 및 반지련을 포함하는 것을 특징으로 하는 생약재를 이용한 건강보조식품 조성물.
- 제 1항에 있어서,상기 우슬 3 내지 7 중량부, 지모 3 내지 7 중량부, 백출 3 내지 7 중량부, 당삼 3 내지 7 중량부, 단풍마 3 내지 7 중량부, 목단피 3 내지 7 중량부, 황백 3 내지 7 중량부, 숙지황 3 내지 7 중량부, 현삼 3 내지 7 중량부, 택사 3 내지 7 중량부, 황기 3 내지 7 중량부, 사상자 3 내지 7 중량부, 선모 3 내지 7 중량부, 패모 3 내지 7 중량부, 구기자 3 내지 7 중량부, 상백피 3 내지 7 중량부, 황정 3 내지 7 중량부, 오미자 3 내지 7 중량부 및 반지련 3 내지 7 중량부의 중량 비율로 포함된 것을 특징으로 하는 생약재를 이용한 건강보조식품 조성물.
- 우슬, 지모, 백출, 당삼, 단풍마, 목단피, 황백, 숙지황, 현삼, 택사, 황기, 사상자, 선모, 패모, 구기자, 상백피, 황정, 오미자 및 반지련으로 이루어진 조성물을 분쇄하여 생약재 분말을 제조하고, 상기 생약재 분말을 홍국균으로 발효하여 제조된 것을 특징으로 하는 생약재를 이용한 건강보조식품.
- 제 3항에 있어서,상기 우슬 3 내지 7 중량부, 지모 3 내지 7 중량부, 백출 3 내지 7 중량부, 당삼 3 내지 7 중량부, 단풍마 3 내지 7 중량부, 목단피 3 내지 7 중량부, 황백 3 내지 7 중량부, 숙지황 3 내지 7 중량부, 현삼 3 내지 7 중량부, 택사 3 내지 7 중량부, 황기 3 내지 7 중량부, 사상자 3 내지 7 중량부, 선모 3 내지 7 중량부, 패모 3 내지 7 중량부, 구기자 3 내지 7 중량부, 상백피 3 내지 7 중량부, 황정 3 내지 7 중량부, 오미자 3 내지 7 중량부 및 반지련 3 내지 7 중량부의 중량 비율로 포함된 것을 특징으로 하는 생약재를 이용한 건강보조식품.
- 제 4항에 있어서,상기 발효는 상기 생약재 분말과 홍국균을 혼합한 후 60 내지 70℃의 온도에서 20 내지 50시간 동안 발효하여 진행된 것을 특징으로 하는 생약재를 이용한 건강보조식품.
- 제 5항에 있어서,상기 생약재 분말을 홍국균으로 발효한 후 분말, 과립, 정제, 캡슐, 음료 또는 추출액의 형태로 제조된 것을 특징으로 하는 생약재를 이용한 건강보조식품.
