Abstract
脂質組成物の製造方法は、極性溶媒及び非極性溶媒を有する抽出溶媒を用いて、含水原材料に含まれる脂質を抽出する抽出処理工程、並びに、抽出処理工程において得られた抽出液を、第1脂質分を含有する極性溶媒相と、第2脂質分を含有する非極性溶媒相とに分離する分離処理工程を含む。
Description
本開示は、脂質組成物及びその製造方法に関する。より詳細には、本開示は、エイコサペンタエン酸(EPA)、ドコサヘキサン酸(DHA)等の不飽和脂肪酸に富んだトリアシルグリセロール及びリン脂質を含有する脂質組成物、並びに、水産物資源又はそれらの加工時に発生する副生成物等からその脂質組成物を製造する方法に関する。
高度不飽和脂肪酸には、γ-リノレン酸やアラキドン酸等のω6系脂肪酸や、α-リノレン酸、EPA、DHA等のω3系脂肪酸があり、これらは体内で合成できにくい必須脂肪酸として知られている。特に、EPAやDHA等のω3系脂肪酸は、血中中性脂肪の低減や関節リュウマチ症状の緩和など様々な生理的機能を有する有用な物質として、食品、健康食品、医薬品等における利用が進められている。EPAとDHAの生理機能は互いに類似しているが同一ではなく、それぞれのω3系脂肪酸を摂取することに有益性が存在する。特に、EPAに関しては、DHAにはない優れた健康機能性、血小板凝固抑制作用や抗炎症作用などエイコサノイド産生に及ぼす生理活性が報告されている。
従来、ω3系不飽和脂肪酸は、青魚、例えばイワシから油脂を煮取ることで魚油として得られている。魚油は、リン脂質が少なく、主としてトリアシルグリセロールの形態でω3系脂肪酸を含んでいる。一方、ホタテガイの中腸腺や生殖巣、外套膜、鰓、またイカの眼球や口球、そして皮などには、トリアシルグリセロールに加えてリン脂質が含まれている。そして、トリアシルグリセロール及びリン脂質には、EPAやDHAを始めとする不飽和脂肪酸がエステル結合している。欧米では、リン脂質結合型のEPAやDHAを含む脂質製品として、クリルオイル(南極オキアミ由来)が流通している。
下記特許文献1~6には、リン脂質型のEPA又はDHAを含む脂質をホタテガイ又はスルメイカの加工時に発生する副生成物から製造する方法が記載されている。
不飽和脂肪酸の体内吸収性に関して、トリアシルグリセロールとリン脂質の比較研究により、リン脂質結合型のω3系脂肪酸の方がトリアシルグリセロール結合型のω3系脂肪酸よりも体内吸収性が良いことが示されている。このことから、リン脂質結合型のω3系脂肪酸を含む脂質組成物の需要の拡大が期待される。しかし、上記特許文献1~5に記載される従来の製造方法では、乾燥した原材料から脂質を抽出するので、原材料の乾燥に多大なエネルギーを必要とする。原材料がリン脂質を豊富に含むか否かに関わらず、一般に脂質を乾燥原料から抽出する場合には、このように投入エネルギーの大きさが問題になる。
又、上記特許文献6では、タンパク質を分解する酵素を作用させた後の水相から油分を分離・回収する。しかし、リン脂質は親水性部分を有するため、乳化が生じ易い。その結果、脂質分と水分の分離・回収が難しい。従って、この従来の脂質組成物の製造方法では、通常の3000×gを超える遠心操作が必要となる。
EPAやDHAを豊富に含有する脂質、特にEPAの割合が多い脂質を得るためには、ホタテガイの中腸腺や生殖巣、外套膜、鰓、またDHAの割合が多い脂質を得るためにはスルメイカの眼球や口球、皮を原材料として用いることが望ましい。しかし、これらの原材料の脂質には、リン脂質が豊富に含まれている。従って、上述したリン脂質の存在に起因する困難を伴う。このため、EPAやDHAを多く含有する脂質組成物を効率良く安価に製造できる技術の実用化を進める必要がある。
さらに、水産物資源及びそれらの加工時に発生する副生成物には、食物連鎖によってカドミウムなどの重金属やヒ素(もしくはヒ素含有化合物)、またダイオキシン類(ポリ塩化ジベンゾパラジオキシン、ポリ塩化ジベンゾフラン、及びダイオキシン様ポリ塩化ビフェニル)等の化合物が蓄積されている。水産物資源又はそれらの加工副生成物から抽出した脂質を栄養組成物として使用する場合には、これらの物質を取り除くことが望ましい。従来技術では、原材料から脂質を抽出する際、これらの物質が脂質に混入する。従って、栄養組成物とする脂質から不要な物質を分離及び除去することは容易ではなかった。そこで、カドミウム等の重金属やヒ素、ダイオキシン類等の物質の含有を抑えられた脂質組成物、特に、リン脂質、EPA、及び/又はDHAを豊富に含有しながらカドミウム等の重金属やヒ素、ダイオキシン類等の物質の含有を抑えた脂質組成物が望まれ、そのような脂質組成物の製造を可能にする方法が望まれる。
一側面において、本開示の課題は、上述の問題を解決し、原材料の乾燥に伴う多大なエネルギーを投入せずに、原材料から脂質の抽出及び分離・精製を簡便且つ容易に行うことが可能な、脂質組成物の製造方法を提供することである。
また、別の側面において、本開示の課題は、リン脂質を含み、且つ脂質を構成する脂肪酸の組成としてEPA及び/又はDHAを豊富に含み、且つカドミウム、ヒ素、ダイオキシン類等の不要な物質の含有を抑えた脂質を提供することである。
上記課題を解決するために、本発明者らは鋭意研究を重ねた結果、原材料を乾燥せずに含水状態のままで溶媒抽出により脂質を効率的に得る手法を見出した。
本開示の一態様によれば、極性溶媒と非極性溶媒を混合した抽出溶媒を用いて、含水原材料に含まれる脂質を抽出する抽出処理工程、及び、前記抽出処理工程において得られた抽出物を、第1脂質分を含有する極性溶媒相と、第2脂質分を含有する非極性溶媒相と、固形分相(不溶固形分相)とに分離する分離処理工程を含む、脂質組成物の製造方法が提供される。
本開示の上記態様による脂質組成物の製造方法は、更に、前記分離処理工程において得られた前記非極性溶媒相を吸着剤と接触させて、前記非極性溶媒相から不純物を除去する精製処理工程を含むことができる。
本開示の上記態様による脂質組成物の製造方法は、更に、前記分離処理工程において得られた前記極性溶媒相及び前記非極性溶媒相から、前記極性溶媒及び前記非極性溶媒をそれぞれ回収する溶媒回収工程を含むことができる。
前記極性溶媒は、低級アルコール、アセトン、アセトニトリル、THF、DMF、又はこれらの組合せを含んでいてもよい。
前記非極性溶媒は、炭素数5~8のアルカン、ジエチルエーテル、t-ブチルメチルエーテル、酢酸エチル、クロロホルム、塩化メチレン、ベンゼン、トルエン、又はこれらの組合せを含んでいてもよい。
前記含水原材料に含まれる全脂質の3質量%以上がリン脂質であってもよい。
前記抽出処理工程において、前記抽出溶媒は、予め前記極性溶媒及び前記非極性溶媒を用いて混合有機溶媒に調製した後に前記含水原材料に添加してもよいし、或いは、前記極性溶媒及び前記非極性溶媒を個別に前記含水原材料に順次添加して、前記添加によって前記混合有機溶媒に調製してもよい。
前記含水原材料は、ホタテガイ及びイカの加工時に発生する副生成物のうちの少なくとも1種を含んでもよい。
前記含水原材料は60~85質量%の含水率で水を含有してもよい。
前記抽出処理工程において、前記極性溶媒及び前記非極性溶媒は、原材料の含水重量1部に対してそれぞれ2~4部及び1~5部の重量比となる割合で使用されてもよい。
本開示の上記態様による脂質組成物の製造方法は、前記含水原材料を65℃以上の温度で加熱する加熱処理工程をさらに含むことができる。
本開示の上記態様による脂質組成物の製造方法は、前記含水原材料を破砕する工程をさらに含むことができる。本開示の実施態様による脂質組成物の製造方法のいくつかの側面は図1に要約されている。

本開示の上記態様による脂質組成物製造装置は、前記分離処理部において分離された極性溶媒相及び非極性溶媒相からそれぞれ前記極性溶媒及び前記非極性溶媒を回収するための溶媒回収部をさらに含むことができる。
本開示の別の態様によれば、脂質を構成する脂肪酸中のEPA対DHAの質量比(EPA/DHA)が1.2~7.2であり、全脂質中のリン脂質の割合が3質量%以上54質量%以下であり、カドミウム濃度が脂質1kg当たり0.4mg以下であり、ヒ素濃度が脂質1kg当たり3mg以下であり、且つ、ダイオキシン類濃度が脂質1g当たり2pg-TEQ以下である脂質組成物が提供される。この態様による脂質組成物において、脂質を構成する脂肪酸中のEPA対DHAの質量比(EPA/DHA)が3.4~7.2であり、全脂質中のリン脂質の割合が18質量%以上39質量%以下であってもよい。
本開示によれば、含水原材料を乾燥することなく含水状態のまま脂質を抽出できるだけでなく、水との共沸ために回収率が低い極性溶媒の添加量(使用量)が抑えられるため、全体として安価に脂質組成物を製造することができる。また、本開示によれば、原材料がリン脂質を豊富に含んでいるか否かに関わらず、乳化が起こらないために容易に脂質の抽出を行うことができる。従って、リン脂質と合わせてEPA及び/又はDHAを豊富に含む原材料からも効率よくEPA及び/又はDHAを含有する脂質組成物を製造することができる。
さらに、本開示によれば、混合有機溶媒を用いた脂質抽出液の二相分離を通じて、リン脂質に富んだ画分と中性脂肪に富んだ画分とに分けて脂質を取得することができる。このことにより、例えば、両画分を適正な比率で混合して所望の組成の脂質組成物を調製することが可能となり、季節の違いや産地の違いに起因する原材料の成分の変動に影響されない、成分が安定した脂質組成物を提供することができる。又、リン脂質及び中性脂肪を実質的に別個に処理することが可能になるので、各々に対して好適な酵素処理等を実施することができる。
さらに、本開示によれば、水産加工時に発生する副生成物を原材料として用いた場合においても、これら副生成物に含まれている脂質をカドミウム等の重金属やヒ素、ダイオキシン類などの物質と分離した状態で得ることができ、ごく簡単な精製工程で、それら不純物の濃度がきわめて低い、食品に係わる規制値を満たす安全な脂質を得ることができる。従って本開示は、ホタテガイやスルメイカ等の水産物の加工時に発生する副生成物の利用を促進させ、副生成物の処理費用に係わる問題の解消や海産資源の有効利用において有用である。
脂質には、トリアシルグリセロールのような単純脂質や、リン脂質、糖脂質等のような複合脂質がある。これらの脂質は分子の構成成分として脂肪酸を含有し、すなわち、分子内にエステル結合した状態で脂肪酸を含有し、加水分解によって脂肪酸を遊離する。脂肪酸は、飽和脂肪酸と不飽和脂肪酸とに分類できる。ラウリン酸、ミリスチン酸等は飽和脂肪酸である。一方、ヒトの体内で合成できにくい必須脂肪酸であるEPAやDHA等のω3系脂肪酸やアラキドン酸等のω6系脂肪酸は、複数の不飽和結合を有する高度不飽和脂肪酸(又は多価不飽和脂肪酸)である。
イワシやオキアミ等の海洋生物は、ω3系脂肪酸を含む水産資源として知られている。これら海産物を原材料とすることによって、トリアシルグリセロールやリン脂質等の脂質を含有する組成物を得ることができる。ω3系脂肪酸は、脂質の構成成分として組成物中に含まれることとなる。得られる脂質組成物は、サプリメントや食品添加物、また医薬品等に利用されている。脂質分子を構成する脂肪酸の体内吸収に関しては、親水基を有するリン脂質の方がトリアシルグリセロールの形態より有利である。従って、リン脂質の形態でω3系脂肪酸を豊富に含む脂質組成物は、ω3系脂肪酸の補給源として特に有用である。
リン脂質は、乾燥した原材料から抽出されてきた。このことは、リン脂質が親水性の構成部分(リン酸基)を有することに起因する。すなわち、水が共存する条件下での溶媒抽出においては、リン脂質が界面活性剤として作用し、脂質を水から分離することが難しくなるためである。この点に関し、本開示では、原材料を予め乾燥することなく、含水状態のままで有機溶媒を用いた脂質の抽出を行う。この際、極性溶媒と非極性溶媒の両方を好適な割合で組み合わせて混合有機溶媒を作製し、抽出溶媒として使用する。これにより、両親媒性を有するリン脂質をトリアシルグリセロール等のその他の脂質分と併せて良好に抽出することが可能となる。本開示による抽出処理は、リン脂質が乏しい含水原材料にも適用できる。
抽出物から回収される脂質には、リン脂質に加えて他の脂質も含まれ、具体的には、トリアシルグリセロールを筆頭とする中性脂肪や、ステロール類等の親油性の誘導脂質(脂質の加水分解によって生じる化合物)が含まれる。本開示では、抽出処理によって得られる抽出物(脂質を含有する混合有機溶媒)を、極性溶媒相と非極性溶媒相と固形分相とに分離する分離処理を行う。これにより、抽出物は三相に分画され、特に脂質は、極性溶媒相に含まれる第1脂質分と、非極性溶媒相に含まれる第2脂質分とに分離される。極性溶媒相の第1脂質分はリン脂質であり、中性脂肪は殆ど含まれない。非極性溶媒相の第2脂質分は、主に中性脂肪を含有するが、いくらかの量のリン脂質も含有する。
尚、抽出物には、リン脂質及び中性脂肪以外の不純物として、カドミウム、鉛、水銀、クロム、銅、亜鉛を始めとする重金属やマグネシウム(もしくはマグネシウム含有化合物)、ヒ素(もしくはヒ素含有化合物)、ダイオキシン類等の物質、またタンパク質系の有機物などが含まれる。そして、これら重金属やマグネシウム、ヒ素、ダイオキシン類等の物質、そしてタンパク質系の有機物は、極性溶媒相及び固形分相に選択的に分配させる。従って、これらの不純物は、後述するように脂質と分離することが可能である。特に、本開示に従って混合有機溶媒を用いた抽出及び相分離を行った場合、カドミウム、鉛、水銀等の重金属やマグネシウム、ヒ素、ダイオキシン類等の不純物は、極性溶媒相及び不溶固形分相に分離され、脂質が溶存する非極性溶媒相から排除される。従って、非極性溶媒相においては極めて低い不純物含有量の水準を容易に達成することができる。
以下に、本開示の脂質組成物の製造方法について詳細に説明する。
本開示において、脂質組成物を製造するための原材料は、脂質を含有する含水原材料であり、特に、リン脂質を含有する含水原材料が好ましく、脂質のアシルエステル部分を構成する成分としてはω3系脂肪酸を含有するものがより好ましい。原材料中のω3系脂肪酸は、リン脂質、中性脂肪、及びその他の脂質の何れの脂質を構成する成分であってもよいが、その少なくとも一部はリン脂質の構成成分であることが好ましい。すなわち、本開示の実施態様において、脂質組成物、好ましくはリン脂質を含有する脂質組成物、より好ましくはリン脂質構成ω3系脂肪酸を含有する脂質組成物の製造方法が提供される。好ましくは、原材料中の全脂質の3質量%以上、より好ましくは5質量%以上がリン脂質である。好ましくは、原材料中の全脂質の54質量%以下、より好ましくは39質量%以下がリン脂質である。原材料中の全脂質におけるリン脂質の質量割合は、例えば薄層クロマトグラフィ等、当業者に知られる方法によって測定することができる。
本開示において、用語「水産物資源」及び「水産資源」は同義に用いられ、これらの用語は、魚類及び非魚類水生生物を含み、海洋から採取されるものだけでなく淡水から採取されるものも含むものとする。用語「水産加工」は、これらの水生生物を食品製造等の目的で加工することを意味する。前述のようなリン脂質構成ω3系脂肪酸を含有し、本開示の一態様による方法の原材料として使用し得る水産資源として、例えば、ホタテ等の貝類やイカ等を含む軟体動物、オキアミ等の節足動物などの水生生物、またイクラ等の各種魚卵が挙げられる。ホタテガイの中腸腺や生殖巣、外套膜、鰓、またイカの眼球や、口球、皮等のような、水産加工に際して発生する副生成物も好適に利用できる。このような副生成物を用いると、副生成物の処理費用を削減することができ、かつ海産資源を有効に活用する上で好都合である。又、水産資源以外にも、大豆、卵黄、豚肝臓などもリン脂質の原材料となる。更に、近年、ボツリオコッカス属の微細藻類に、リノレン酸等の不飽和脂肪酸やそれらを構成成分とするリン脂質が含まれることが知られており、このようなものにも本開示の実施形態による方法において原材料として適用可能である。
このような原材料から、目的のω3系脂肪酸の含有量等を考慮して、適した原材料が選択される。例えば、ω3系脂肪酸成分としてEPA及びDHAを含有し、リン脂質に富んだ好ましい原材料として、ホタテガイの中腸腺、生殖巣、外套膜、鰓やイカの眼球、口球、皮、またオキアミやイクラ等が挙げられる。ホタテガイやスルメイカなどの水産加工時に発生する副生成物には、リン脂質又は中性脂肪の構成成分としてEPA及びDHAが含まれており、ω3系脂肪酸成分が豊富な極めて有用な原材料である。一例として、青森県において、ホタテの加工時に発生した中腸腺から抽出した脂質の約13質量%がリン脂質、約68質量%がトリアシルグリセロールであり(その他はコレステロール、ステロールエステル、糖脂質等)、それらの脂質を構成する脂肪酸の75~80%が不飽和脂肪酸であって、その中でもEPAが約34%と最も多かった(DHAは約7%)(図2及び図3)。上述のような原材料から必要に応じて複数種を組み合わせて用いてもよい。以下の記載において、EPA及びDHAを目的のω3系脂肪酸とした場合に関して本開示の各態様を説明するが、勿論、他のω3系脂肪酸やアラキドン酸等のω6系脂肪酸を目的とすることも可能である。
上述のような原材料は、乾燥せずに含水状態でそのまま使用することができる。概して、含水率が60~85質量%程度の含水原材料を好適に使用できるが、凍結乾燥等により含水率がこれより低下した原材料(例えば含水率が50%以上の原材料)にも本開示の方法は使用できる。含水率は、例えば105℃における温風乾燥又は恒温槽内静置により、脂質を抽出した後の試料を乾燥させ、恒量となった時点での乾燥重量を測定して、含水原材料の総重量からこの乾燥重量及び脂質重量を差し引くことによって算出することができる。本明細書において「含水」という場合の「水」は、原材料の生物学的組織に由来する水分及び外部から加えられた水分(海水、洗浄水等)の両方を含み得る。
原材料に含まれる酵素の生物学的反応が進行することを防止するために、抽出処理の前に、原材料を加熱して酵素を失活させる処理を施すことが好ましい。加熱処理は、含水原材料を例えば65℃以上、好ましくは70℃以上、より好ましくは80℃以上、さらに好ましくは90℃以上に加熱した水に浸漬することで行われる。加熱処理の時間は当業者が適宜決定することができ、通常は10分以内である。加熱処理の後の原材料は空冷、もしくは水冷した後に冷蔵保存又は冷凍保存してもよい。
又、抽出処理の前に、原材料を予め破砕(或いは粉砕)して用いると、抽出溶媒との接触面積の増大によって脂質の抽出効率が向上するので好ましい。含水状態での破砕を湿式破砕という。破砕方法は、切断刃を用いる物理的破砕が概して好適に利用できるが、公知の方法から適宜選択すればよい。破砕(或いは粉砕)により含水原材料をペースト様の状態に調製すると好ましい。他の物理的破砕方法には、超音波法、凍結融解法、浸透圧ショック法、ビーズや固体粉末等を用いる磨砕法、ホモジナイザーを用いる方法、小孔からの強制押出による剪断力を利用するフレンチプレス法などがある。
上述のような原材料に、極性溶媒及び非極性溶媒を添加して撹拌すること(抽出処理)により、混合有機溶媒に脂質が浸出する。抽出処理において、予め極性溶媒と非極性溶媒からなる混合溶媒を調製した後にその混合溶媒を原材料に添加してもよいし、或いは、極性溶媒及び非極性溶媒を個別に原材料に添加して、添加の結果として混合溶媒になるようにしてもよい。後者の場合、極性溶媒及び非極性溶媒の何れを先に原材料に添加してもよい。湿式破砕後の原材料に極性溶媒を添加して振とう、撹拌した後に非極性溶媒を添加すると、原材料に含まれる水と極性溶媒とが最初に混和して、溶媒と原材料とが接触し易い。
抽出処理は、雰囲気温度において好適に実施され、例えば10~40℃程度の抽出温度で行うことができる。抽出形態は、バッチ式及び連続式の何れでも良い。原材料と抽出溶媒とが良好に接触し得るように攪拌又は振とうにより十分に混合して、十分な抽出時間を設定するとよい。好ましくは、攪拌又は振とうを30分程度継続して原材料と抽出溶媒とを十分に混合する。バッチ式においては、溶媒抽出を複数回繰り返して抽出毎に得られる抽出物を合わせることによって脂質の回収率を高めることができる。向流抽出は、作業効率や経済性の点において好ましく、半向流多回抽出装置やミキサー-セトラー型抽出装置、塔型抽出装置、向流型遠心抽出装置等を用いると、連続向流抽出を好適に遂行できる。
抽出溶媒を構成する極性溶媒としては、例えば、メタノール、エタノール、1-プロパノール、2-プロパノール、1-ブタノール等の低級アルコール(炭素数1~5のアルコール)や、アセトン、アセトニトリル、THF(テトラヒドロフラン)、DMF(N,N-ジメチルホルムアミド)等が挙げられ、複数種を組み合わせて用いても良い。好ましくはエタノール又はメタノール、より好ましくはエタノールが用いられる。非極性溶媒としては、例えば、n-ヘキサンのような炭素数5~8の直鎖状、分枝状、又は環状のアルカン、ジエチルエーテル、t-ブチルメチルエーテル、酢酸エチル、クロロホルム、塩化メチレン、ベンゼン、トルエン等が挙げられ、複数種を組み合わせて用いても良い。好ましくはn-ペンタン、n-ヘキサン、n-ヘプタン又はt-ブチルメチルエーテル、より好ましくはn-ヘキサンが用いられる。極性溶媒が低級アルコールであって非極性溶媒が炭素数5~8の直鎖状アルカンであることが特に好ましく、極性溶媒がエタノールであって非極性溶媒がn-ヘキサンであることが最も好ましい。
極性溶媒の使用量は、含水原材料1重量部(含水重量)に対して1~5重量部程度、好ましくは2~4重量部程度となるように、非極性溶媒の使用量は、含水原材料1重量部に対して1~10重量部程度、好ましくは1~5重量部程度となるように各々調整するとよい。尚、脂質の過酸化を抑制するために、含水原材料に対して0.01~0.2質量%程度の抗酸化剤(例えば、γ‐トコフェロールやα‐トコフェロール、またはミックストコフェロール)を含水原材料にあらかじめ添加してもよい。
通常、水産加工原料の60~85質量%程度は水分である。溶媒抽出を開始すると、原材料に含まれる脂質(含水原材料の3~21質量%程度)が抽出溶媒へ移行すると共に、原材料に含まれる水分が極性溶媒に混和し、不溶物が固形分(15~31乾燥質量%程度)として残存する。
抽出処理によって得られる抽出液と不溶固形分との混合物から不溶固形分を除去することにより、脂質を含有する抽出液が得られる。固形分の除去は、沈降分離、濾過分離、遠心分離等の一般的な固液分離手法から適切な方法を選択することにより実施できる。固形分の除去は、後述する抽出液の分離処理の際に行ってもよい。抽出による脂質の回収率を上げるために、必要に応じて、回収した固形分に、上述と同様に極性溶媒及び非極性溶媒を再度添加して抽出処理を繰り返すことができる。概して、1回の抽出によって、原材料に含まれる全脂質の80質量%程度を、2回の抽出によって全脂質の98質量%程度以上を抽出できる。
抽出処理によって得られる抽出液は、極性溶媒と非極性溶媒との混合状態であり、これに分離処理を施して、極性溶媒相と非極性溶媒相とに分離する。分離処理としては、遠心分離等を用いて比重差による分離を促進する処理が適用される。抽出処理を複数回行う場合は、各回の抽出で固形分の除去後に得られる抽出液を1つに纏めて分離処理を行っても、或いは、各回の抽出で得られる抽出液毎に分離処理を行ってもよい。分離処理と共に前述の固形分の除去を行う場合は、不溶固形分を含んだままの抽出液を分離処理して、分離した極性溶媒相及び非極性溶媒相の各々を取り出す。この後、固形分に対して溶媒を添加すると抽出処理及び分離処理を繰り返すことができ、分離された極性溶媒相同士、非極性溶媒相同士を纏めて、各々、後続の処理が施される。遠心分離において使用される遠心力は、極性溶媒相と非極性溶媒相とを分けるために十分な遠心力であり、1,000~3,000×g程度、或いは1,500~2,500×g程度、例えば2,000×g程度の遠心力で行うとよい。遠心分離の時間は、極性溶媒相と非極性溶媒相とを分けるために十分な時間であり、例えば5分~10時間程度、好ましくは数分程度の遠心分離を行うとよい。遠心分離の温度は、例えば室温付近、具体的には10~30℃程度の温度で行い得る。これらのパラメータは抽出液の液量等によって適宜設定する。
相分離によって、抽出液に含まれる脂質は、極性溶媒相に含有される第1脂質分と、非極性溶媒相に含有される第2脂質分とに分けられる。概して、2回の抽出で得られる抽出液の分離処理によって、極性溶媒相には、原材料に含まれる全脂質の量の1~10質量%程度に相当する第1脂質分が含まれ、非極性溶媒相には、全脂質の量の90質量%以上に相当する第2脂質分が含まれることとなる。第1脂質分中のリン脂質の割合は、概して70質量%以上である。第2脂質分中のトリアシルグリセロールの割合は、概して35~75質量%程度、リン脂質の割合は概して10~30質量%程度となる。
分離処理によって分離された極性溶媒相及び非極性溶媒相に含まれる極性溶媒及び非極性溶媒は、それぞれ回収することができる。当業者に知られるあらゆる方法で溶媒を回収することができるが、蒸留によって溶媒回収を行うことが好適である。回収されたこれらの溶媒は再利用することができ、特に、上記で説明した抽出処理において再利用することができる。
水と混和する極性溶媒、特にエタノールは水と共沸するために、非極性溶媒、例えばn-ヘキサンと比較して、回収率が低い。しかしながら、本開示による抽出処理においては、極性溶媒は非極性溶媒と合わせて混合有機溶媒の一部として用いられるに過ぎない。従って、極性溶媒のみを抽出溶媒とする場合と比べて、極性溶媒の使用量を抑制できる。
溶媒回収工程は、極性溶媒相から極性溶媒を回収すること、非極性溶媒相から非極性溶媒を回収すること、又はそれらの両方を含み得る。後述するように、非極性溶媒相を吸着剤と接触させて第2脂質分の精製処理を行う場合には、精製処理の後に非極性溶媒の回収を行うことが好ましい。
溶媒の除去は、常法に従って、減圧留去、噴霧乾燥、凍結乾燥等の公知の乾燥技術を適宜利用して行うことができる。脂質の酸化又は変質を進行させる加熱及び酸素ガスとの接触は避けることが望ましく、非酸化性雰囲気中において80℃程度の温度で溶媒を留去するとよい。
極性溶媒相の第1脂質分には、水溶性又は親水性を有する物質が不純物として混在し、その具体例として、アルセノベタインやタンパク質系の水溶性有機物を挙げることができる。一方、非極性溶媒相の第2脂質分には、親油性の不純物が混在し、具体例として、マグネシウムや銅のポルフィリン錯体、もしくはその類似物を挙げることができる。非極性溶媒相の第2脂質分から不純物を除去するために、精製処理が行われ得る。これにより、製品の望ましくない着色や臭気を取り除いて、より高品質な脂質製品を供給できる。
非極性溶媒相に溶存するマグネシウムや銅のポルフィリン錯体等の不純物は、活性炭、活性白土、またはシリカゲル等の吸着剤を添加し、接触撹拌を行うことで除去できる。使用する吸着剤の添加量は、非極性溶媒相に溶存する脂質重量の1~10重量%程度が好適である。なお、脂質分の脱臭は、分子蒸留、水蒸気蒸留等を利用しても達成される。精製は、吸着剤を非極性溶媒相に添加して接触撹拌、もしくは非極性溶媒相を留去した後の脂質に対して分子蒸留、または水蒸気蒸留を行えばよく、これら処理の何れか1つ又は複数を実施してもよい。但し、水蒸気蒸留を行う場合には、脂質の成分の1つであるリン脂質を脱ガムで除去した後に実施する必要がある。
所望の組成の脂質組成物を得るために、脂質分、例えば第2脂質分の成分分析を行って、リン脂質、中性脂肪、及び、脂質を構成する目的のω3系脂肪酸成分(アシル残基として)の含有量を測定することが好ましい。これらの測定値に基づいて、別個の抽出操作より得た脂質分を組み合わせることができる。例えば、目的のω3系脂肪酸成分の含有量、及び、トリアシルグリセロールとリン脂質の含有量の各々の所定値を設定し、別個の抽出操作で得た第2脂質分を、この所定値になるような配合割合で組み合わせて、均質な脂質組成物に調製することができる。得られた脂質分の成分分析は、一般的な脂質の分析方法によって行えばよく、例えば、脂質の組成は薄層クロマトグラフィ/水素炎イオン化検出器(TLC/FID)を用いて、また脂肪酸組成についてはガスクロマトグラフィを利用して分析できる。脂質を構成する脂肪酸成分については、脂質分の鹸化及び不鹸化物のエーテル抽出を経て得られる遊離脂肪酸をメチルエステル化することにより、ガスクロマトグラフィによって分析できる。検出されるピーク面積、及び、予め作成した検量線に基づいて、各成分の含有量が決定される。脂質組成の分析は、順相TLC、順相HPLC等を利用して実施できる。
ホタテガイやスルメイカの加工時に発生する副生成物を原材料として脂質組成物を製造する場合、副生成物の産地や季節によって脂質の成分及び脂肪酸組成が変動する。又、副生成物に含まれる原材料の部位とその歩留りによっても、脂質の成分及び脂肪酸組成の割合にバラツキが生まれる。例えば、青森県において、ホタテの加工時に発生したホタテガイの中腸腺の脂質に含まれるリン脂質の割合は、3~16質量%程度の範囲で、EPA及びDHA及びの合計含有量は、38~43質量%程度の範囲で変動する。北海道紋別市、北海道宗谷郡猿払村、及び北海道二海郡八雲町において、ホタテの加工時に発生する中腸腺に加えて中腸腺以外の部位、すなわち生殖巣や外套膜、鰓をも含めた場合には、リン脂質の割合は7~54質量%程度の範囲で、EPA及びDHAの合計含有量は34~43質量%程度の範囲で変動する(図4及び図5)。従って、目的のω3系脂肪酸成分は、これらω3系脂肪酸の含有量及びリン脂質の含有量に係わる季節変動と、原材料の産地、そして原材料の月次発生量の変化を考慮に入れて設定する。原材料の成分に係わる季節変動と原材料の発生量に係わる季節変動に応じて適宜配合割合を調節することで、脂質成分の変動を抑制した、ω3系脂肪酸に富んだ脂質製品の提供が可能となる。
図1は、本開示の一態様による脂質組成物製造の概要を示す。この実施態様では、非極性溶媒相(第2脂質分)のみを使用して脂質組成物を製造し、製剤化している。極性溶媒であるエタノールと非極性溶媒であるn-ヘキサンは、それぞれ蒸留により回収して(溶媒回収工程)、抽出溶媒として再利用している。なお、エタノール及びn-ヘキサン以外の極性溶媒及び非極性溶媒を使用してもよい。廃水処理工程及び製剤化工程はそれぞれ任意である。加熱処理工程、湿式破砕処理工程、吸着(精製処理)工程、及び溶媒回収工程は、それぞれ省略可能であるが、省略しない方が好ましい。本図ではホタテガイの加工時に発生する副生成物を原材料としているが、この方法は脂質を含有するあらゆる種類の原材料に応用できる。
水産物資源及びそれらの加工時に発生する副生成物には、カドミウムや鉛、水銀、クロム、銅、亜鉛などの重金属や、マグネシウム(もしくはマグネシウム含有化合物)、ヒ素(もしくはヒ素含有化合物)、またダイオキシン類(ポリ塩化ジベンゾパラジオキシン、ポリ塩化ジベンゾフラン、及びダイオキシン様ポリ塩化ビフェニル)等の化合物が蓄積されている。本開示に従って得られた分離処理後の非極性溶媒相は、カドミウム、鉛、水銀、クロム、銅、亜鉛、マグネシウム、及びヒ素の含有量が著しく低いことが見出された。すなわち、分離処理を経ただけの非極性溶媒相において既に、例えばカドミウム濃度が脂質1kg当たり0.6mg以下、鉛濃度が脂質1kg当たり0.1mg以下、水銀濃度が脂質1kg当たり0.02mg以下、クロム濃度が脂質1kg当たり0.07mg以下、銅濃度が脂質1kg当たり1.6mg以下、亜鉛濃度が脂質1kg当たり4.2mg以下、マグネシウム濃度が脂質1kgあたり400mg以下、ヒ素濃度が脂質1kg当たり8mg以下、及びダイオキシン類濃度が脂質1kg当たり3.3pg‐TEQという低い水準が達成され得る。
第2脂質分を含むこの非極性溶媒相を、吸着剤と接触させて、非極性溶媒相から不純物を除去する精製処理により、上記よりさらに低いカドミウム濃度、鉛濃度、水銀濃度、クロム濃度、銅濃度、亜鉛濃度、マグネシウム濃度、ヒ素濃度、及びダイオキシン類濃度が達成される。具体的には、非極性溶媒相を吸着剤と接触させて30分間程度撹拌することによる精製処理後の第2脂質分のカドミウム濃度は、脂質1kg当たり0.4mg以下であり得、より好ましくは脂質1kg当たり0.3mg以下であり、さらに好ましくは脂質1kg当たり0.2mg以下である。又、精製処理後の第2脂質分のヒ素濃度は、脂質1kg当たり3mg以下であり得、より好ましくは脂質1kg当たり2mg以下であり、さらに好ましくは脂質1kg当たり1mg以下である。
この精製処理で使用される吸着剤は、活性炭、特に食品用途向けの活性炭が好適である。
従来の抽出方法では、これらの毒性成分の濃度が抽出液中で低減されていなかっただけではない。すなわち、従来の抽出方法では、カドミウム及びヒ素以外の不純物、例えばマグネシウムや鉄も多量に抽出液に含まれている。従って、吸着剤を適用しても、吸着剤上で不純物同士が競合しカドミウム及びヒ素の除去が妨げられていた。尚、本開示に従って得られた分離処理後の第2脂質分は、吸着剤の使用の有無にかかわらず、鉛、水銀、クロム、及びダイオキシン類の含有量も著しく低い安全なレベルに抑えられていた(それぞれ、脂質1kg当たりの鉛濃度は0.1mg未満、脂質1kg当たりの水銀濃度は0.01mg未満、脂質1kg当たりのクロム濃度は0.05mg未満、及び脂質1g当たりのダイオキシン類濃度は0.03pg-TEQ未満)。
例えばホタテガイの加工時に発生する副生成物のように、リン脂質を豊富に含み、かつ、EPAの含有量が高い原材料を使用して、本開示に従って抽出処理及び分離処理を行い、任意で非極性溶媒相に対して吸着剤による精製処理を行って、非極性溶媒相から第2脂質分を取得することにより、脂質を構成する脂肪酸におけるEPA対DHAの質量比(EPA/DHA)が1.2~7.2であり、全脂質中のリン脂質の割合が3質量%以上54質量%以下であり、カドミウム濃度が脂質1kg当たり0.4mg以下であり、ヒ素濃度が脂質1kg当たり3mg以下であり、且つ、ダイオキシン類濃度が脂質1gあたり2pg-TEQ以下である、脂質組成物を得ることができる。前記脂質組成物の脂質を構成する脂肪酸におけるEPA対DHAの質量比(EPA/DHA)は好ましくは1.9以上、より好ましくは3.4以上である。前記脂質組成物中の全脂質中のリン脂質の割合は好ましくは3質量%以上であり、より好ましくは10質量%以上であり、さらに好ましくは18質量%以上である。また、前記脂質組成物中の全脂質中のリン脂質の割合は好ましくは54質量%以下であり、より好ましくは45質量%以下であり、さらに好ましくは39質量%以下である。
本開示による脂質組成物製造装置又は脂質組成物製造システムは、上記において説明した脂質組成物の製造を実施することができる装置又はシステムである。すなわちこの装置又はシステムは、極性溶媒及び非極性溶媒を有する混合抽出溶媒を用いて含水原材料に含まれる脂質を抽出するための抽出処理部と、前記抽出処理部において得られた抽出物を、第1脂質分を含有する極性溶媒相と第2脂質分を含有する非極性溶媒相と固形分相とに分離するための分離処理部とを含む。
この装置又はシステムは、前記分離処理部において分離された非極性溶媒相を吸着剤と接触させて、前記非極性溶媒相に残存する不純物を除去するための精製処理部をさらに含むこともできる。この装置又はシステムは又、前記分離処理部において分離された極性溶媒相及び非極性溶媒相からそれぞれ前記極性溶媒及び前記非極性溶媒を回収するための溶媒回収部をさらに含むこともできる。この装置又はシステムは又、前記含水原材料を65℃以上の温度で加熱するための加熱処理部をさらに含むこともできる。この装置又はシステムは又、前記含水原材料を破砕するための湿式破砕部をさらに含むこともできる。
<ホタテ加工時に発生する副生成物の内容分析>
北海道二海郡八雲町において、ホタテの加工時に発生したホタテガイの中腸腺、生殖巣、外套膜、そして鰓を入手し、これに熱湯処理を施した試料1を用いて、以下の測定を行った。

合計量を100gに調整したホタテガイの中腸腺、生殖巣、外套膜、鰓を細かく破砕し、クロロホルム-メタノール混合溶媒(容量比=2:1)700mLを加えて溶媒抽出を行った。抽出液から固形分を除去した後に溶媒を留去することにより、総脂質として油状物を得た。この油状物の質量を測定したところ、7.6g(含水原料の7.6質量%)であった。又、抽出後の残渣を温度を105℃に設定した恒温槽内において恒量となるまで乾燥したところ、21.8gの固形分が回収された。
合計量を100gに調整したホタテガイの中腸腺、生殖巣、外套膜、鰓を細かく破砕し、クロロホルム-メタノール混合溶媒(容量比=2:1)700mLを加えて溶媒抽出を行った。抽出液から固形分を除去した後に溶媒を留去することにより、総脂質として油状物を得た。この油状物の質量を測定したところ、7.6g(含水原料の7.6質量%)であった。又、抽出後の残渣を温度を105℃に設定した恒温槽内において恒量となるまで乾燥したところ、21.8gの固形分が回収された。
北海道紋別市において、ホタテの加工時に発生した中腸腺、生殖巣、外套膜、そして鰓を入手し、これに熱湯処理を施した試料2を用いて、上記と同様の内容分析を行ったところ、総脂質の量は、含水質量の5.6質量%であった。また、北海道宗谷郡猿払村において、ホタテの加工時に発生した中腸腺、生殖巣、外套膜、そして鰓を入手し、これに熱湯処理を施した試料3についても上記と同様の内容分析を行ったところ、総脂質量は、含水質量の6.6質量%であった。同様に、青森県において、ホタテの加工時に発生したホタテガイの中腸腺を入手し、これに熱湯処理を施した試料4についても上記と同様の内容分析を行ったところ、総脂質量は含水質量の12.9質量%であった。
<試料1>
(抽出処理及び分離処理)
フードプロセッサーを用いて、ホタテガイの中腸腺、生殖巣、外套膜、鰓219gをペースト状に破砕し、これに95%エタノール100mL及びn-ヘキサン250mLを加えて30分間振とうした(1回目の抽出)。これを3000rpm(900×g)で10分間遠心分離して、抽出液をn-ヘキサン相、エタノール‐水相及び固形分に分離し、n‐ヘキサン相及びエタノール‐水相の各々を個別に容器に取り出した(1回目の分離処理)。残った固形残渣に再度95%エタノール100mL及びn-ヘキサン250mLを加えて、上述と同様の条件で2回目の抽出処理及び2回目の分離処理を行った。2つのエタノール‐水相を纏めて、これに含まれる第1脂質分の量を調べたところ、0.75gであり、2つのn‐ヘキサン相を各々纏めて、これに含まれる第2脂質分の量を調べたところ、16.15gであった。従って、第1脂質分は総脂質量の約4.4%、第2脂質分は約94.4%であり、抽出処理における脂質の回収率は98.8%となった。
(抽出処理及び分離処理)
フードプロセッサーを用いて、ホタテガイの中腸腺、生殖巣、外套膜、鰓219gをペースト状に破砕し、これに95%エタノール100mL及びn-ヘキサン250mLを加えて30分間振とうした(1回目の抽出)。これを3000rpm(900×g)で10分間遠心分離して、抽出液をn-ヘキサン相、エタノール‐水相及び固形分に分離し、n‐ヘキサン相及びエタノール‐水相の各々を個別に容器に取り出した(1回目の分離処理)。残った固形残渣に再度95%エタノール100mL及びn-ヘキサン250mLを加えて、上述と同様の条件で2回目の抽出処理及び2回目の分離処理を行った。2つのエタノール‐水相を纏めて、これに含まれる第1脂質分の量を調べたところ、0.75gであり、2つのn‐ヘキサン相を各々纏めて、これに含まれる第2脂質分の量を調べたところ、16.15gであった。従って、第1脂質分は総脂質量の約4.4%、第2脂質分は約94.4%であり、抽出処理における脂質の回収率は98.8%となった。
(成分分析)
上記第1脂質分及び第2脂質分の各々について、リン脂質及び中性脂肪の含有量を、薄層クロマトグラフィを利用して測定した。その結果、第1脂質分中のリン脂質含有量は56質量%、トリアシルグリセロール含有量は1質量%以下であり、第2脂質分中のリン脂質含有量は9質量%、トリアシルグリセロール含有量は43質量%であった。又、ガスクロマトグラフィによって、脂質に含まれる脂肪酸中のEPA及びDHAの割合を調べたところ、第1脂質分のEPAの割合は26質量%、DHAの割合は11質量%であった。第2脂質分のEPAの割合は32質量%、DHAの割合は9質量%であった。
上記第1脂質分及び第2脂質分の各々について、リン脂質及び中性脂肪の含有量を、薄層クロマトグラフィを利用して測定した。その結果、第1脂質分中のリン脂質含有量は56質量%、トリアシルグリセロール含有量は1質量%以下であり、第2脂質分中のリン脂質含有量は9質量%、トリアシルグリセロール含有量は43質量%であった。又、ガスクロマトグラフィによって、脂質に含まれる脂肪酸中のEPA及びDHAの割合を調べたところ、第1脂質分のEPAの割合は26質量%、DHAの割合は11質量%であった。第2脂質分のEPAの割合は32質量%、DHAの割合は9質量%であった。
<試料2>
ホタテガイの中腸腺、生殖巣、外套膜、及び鰓50.3gを用いて、試料1と同様に破砕、抽出処理及び分離処理を行って、エタノール‐水相及びn‐ヘキサン相を得た。各相に含まれる第1脂質分及び第2脂質分の量を調べたところ、第1脂質分は0.75gであり、第2脂質分は0.96gであった。従って、第1脂質分は総脂質量の約27%、第2脂質分は約34%であり、抽出における回収率は61%となった。又、第1脂質分及び第2脂質分の各々について、リン脂質及びトリアシルグリセロールの含有量、並びにEPA及びDHAの割合を試料1と同じ方法で調べたところ、第1脂質分のリン脂質含有量は47質量%、トリアシルグリセロール含有量は1質量%以下、EPAの割合は16質量%、DHAの割合は25質量%であった。第2脂質分のリン脂質含有量は31質量%、トリアシルグリセロール含有量は33質量%、EPAの割合は24質量%、DHAの割合は15質量%であった。
ホタテガイの中腸腺、生殖巣、外套膜、及び鰓50.3gを用いて、試料1と同様に破砕、抽出処理及び分離処理を行って、エタノール‐水相及びn‐ヘキサン相を得た。各相に含まれる第1脂質分及び第2脂質分の量を調べたところ、第1脂質分は0.75gであり、第2脂質分は0.96gであった。従って、第1脂質分は総脂質量の約27%、第2脂質分は約34%であり、抽出における回収率は61%となった。又、第1脂質分及び第2脂質分の各々について、リン脂質及びトリアシルグリセロールの含有量、並びにEPA及びDHAの割合を試料1と同じ方法で調べたところ、第1脂質分のリン脂質含有量は47質量%、トリアシルグリセロール含有量は1質量%以下、EPAの割合は16質量%、DHAの割合は25質量%であった。第2脂質分のリン脂質含有量は31質量%、トリアシルグリセロール含有量は33質量%、EPAの割合は24質量%、DHAの割合は15質量%であった。
<試料3>
ホタテガイの中腸腺、生殖巣、外套膜、鰓50.1gを用いて、試料1と同様に破砕、抽出処理及び分離処理を行って、エタノール‐水相及びn-ヘキサン相を得た。各相に含まれる第1脂質分及び第2脂質分の量を調べたところ、第1脂質分は0.07gであり、第2脂質分は3.2gであった。従って、第1脂質分は総脂質量の約2%、第2脂質分は約97%であり、抽出における回収率は99%となった。又、第1脂質分及び第2脂質分の各々について、リン脂質及びトリアシルグリセロールの含有量並びにEPA及びDHAの割合を試料1と同じ方法で調べたところ、第1脂質分のリン脂質含有量は47質量%、トリアシルグリセロール含有量は1質量%以下、EPAの割合は23質量%、DHAの割合は16質量%であった。第2脂質分のリン脂質含有量は29質量%、トリアシルグリセロール含有量は49質量%、EPAの割合は36質量%、DHAの割合は8質量%であった。

<試料4>
ホタテガイの中腸腺50.3gを用いて、試料1と同様に破砕、抽出処理及び分離処理を行って、エタノール‐水相及びn‐ヘキサン相を得た。各相に含まれる第1脂質分及び第2脂質分の量を調べたところ、第1脂質分は0.3gであり、第2脂質分は6.1gであった。従って、第1脂質分は総脂質量の約5%、第2脂質分は約95%であり、抽出における回収率は約100%となった。又、第1脂質分及び第2脂質分の各々について、リン脂質及びトリアシルグリセロールの含有量並びにEPA及びDHAの割合を試料1と同じ方法で調べたところ、第1脂質分のリン脂質含有量は58質量%、トリアシルグリセロール含有量は1質量%以下、EPAの割合は30質量%、DHAの割合は6質量%であった。第2脂質分のリン脂質含有量は20質量%、トリアシルグリセロール含有量は50質量%、EPAの割合は35質量%、DHAの割合は7質量%であった。
ホタテガイの中腸腺50.3gを用いて、試料1と同様に破砕、抽出処理及び分離処理を行って、エタノール‐水相及びn‐ヘキサン相を得た。各相に含まれる第1脂質分及び第2脂質分の量を調べたところ、第1脂質分は0.3gであり、第2脂質分は6.1gであった。従って、第1脂質分は総脂質量の約5%、第2脂質分は約95%であり、抽出における回収率は約100%となった。又、第1脂質分及び第2脂質分の各々について、リン脂質及びトリアシルグリセロールの含有量並びにEPA及びDHAの割合を試料1と同じ方法で調べたところ、第1脂質分のリン脂質含有量は58質量%、トリアシルグリセロール含有量は1質量%以下、EPAの割合は30質量%、DHAの割合は6質量%であった。第2脂質分のリン脂質含有量は20質量%、トリアシルグリセロール含有量は50質量%、EPAの割合は35質量%、DHAの割合は7質量%であった。
<非極性溶媒相(第2脂質分)中の不純物の分析>
ホタテガイの加工時に発生する副生成物を原材料として、本開示に従った加熱処理、湿式破砕処理、抽出処理及び分離処理を実施した後に、n-ヘキサン(非極性溶媒)相におけるカドミウム、鉛、ヒ素、及びクロムの濃度を測定した。元素分析は、溶媒を除去した後にICP-AESを用いて行った。又、n-ヘキサン相に溶存する脂質重量に対して5重量%の粉末活性炭吸着剤を添加して30分間接触撹拌する精製処理を行った後、再びこれらの不純物の濃度を測定した。結果を図6に示す。これらの不純物の濃度は、脂質1kg当たりのmg量として表している。グラフ中の横線は、参考のための基準値を示す。カドミウム濃度の基準値は、FAO/WHO Codex Alimentariusの精米に対する国際基準値(0.4mg/kg)に基づく(尚、海産二枚貝の国際基準値はより緩く、2mg/kgである)。鉛濃度の基準値はFood Chemical Codex、ヒ素濃度の基準値はFood Chemical Codex及びEuropean Union E322にそれぞれ基づく。尚、クロムは、鉄、銅、亜鉛と共に健康維持に欠かすことのできない元素であるが、フランスでは1日あたりのクロム摂取量を30μg以下に抑えることが推奨されている。そこで、EPA及びDHAの1日あたりの摂取量(1g)と上記において見出された第2脂質分中のEPA及びDHAの濃度(約40%)から、クロム濃度の上限値(12mg/kg)を算出した。この図から、第2脂質分は毒性不純物の濃度がきわめて低く抑えられており、そのままでも十分に安全性が確保されているが、精製処理によりさらに厳しい基準をも満たすことができることがわかる。
ホタテガイの加工時に発生する副生成物を原材料として、本開示に従った加熱処理、湿式破砕処理、抽出処理及び分離処理を実施した後に、n-ヘキサン(非極性溶媒)相におけるカドミウム、鉛、ヒ素、及びクロムの濃度を測定した。元素分析は、溶媒を除去した後にICP-AESを用いて行った。又、n-ヘキサン相に溶存する脂質重量に対して5重量%の粉末活性炭吸着剤を添加して30分間接触撹拌する精製処理を行った後、再びこれらの不純物の濃度を測定した。結果を図6に示す。これらの不純物の濃度は、脂質1kg当たりのmg量として表している。グラフ中の横線は、参考のための基準値を示す。カドミウム濃度の基準値は、FAO/WHO Codex Alimentariusの精米に対する国際基準値(0.4mg/kg)に基づく(尚、海産二枚貝の国際基準値はより緩く、2mg/kgである)。鉛濃度の基準値はFood Chemical Codex、ヒ素濃度の基準値はFood Chemical Codex及びEuropean Union E322にそれぞれ基づく。尚、クロムは、鉄、銅、亜鉛と共に健康維持に欠かすことのできない元素であるが、フランスでは1日あたりのクロム摂取量を30μg以下に抑えることが推奨されている。そこで、EPA及びDHAの1日あたりの摂取量(1g)と上記において見出された第2脂質分中のEPA及びDHAの濃度(約40%)から、クロム濃度の上限値(12mg/kg)を算出した。この図から、第2脂質分は毒性不純物の濃度がきわめて低く抑えられており、そのままでも十分に安全性が確保されているが、精製処理によりさらに厳しい基準をも満たすことができることがわかる。
本開示の実施形態は、水産資源等の原材料を乾燥させずに含水状態のまま使用でき、リン脂質を含む原材料からの脂質の抽出及び分離・回収・精製を効率的且つ簡便に行え、リン脂質及びω3系脂肪酸、特にEPAを豊富に含み且つ不純物が国際的な基準値以下である脂質製品を製造することができる。従って、サプリメント、食品添加物、あるいは医薬品原材料等として製品を提供することが可能となる。本開示の実施形態は、産地及び季節による原材料の成分変動に対応して、成分が一定の範囲に収まる脂質製品を提供するための管理が容易で、水産資源の加工時に発生する副生成物の処理費用の削減や未利用資源の有効利用にも寄与し、リサイクルの促進及び環境保護に貢献することが可能である。
Claims (17)
- 極性溶媒及び非極性溶媒を有する抽出溶媒を用いて、含水原材料に含まれる脂質を抽出する抽出処理工程、並びに
前記抽出処理工程において得られた抽出液を、第1脂質分を含有する極性溶媒相と、第2脂質分を含有する非極性溶媒相とに分離する分離処理工程
を含む、脂質組成物の製造方法。 - 前記分離処理工程において得られた前記非極性溶媒相を吸着剤と接触させて前記非極性溶媒相から不純物を除去する精製処理工程をさらに含む、請求項1に記載の脂質組成物の製造方法。
- 前記分離処理工程において得られた前記極性溶媒相及び前記非極性溶媒相から、前記極性溶媒及び前記非極性溶媒をそれぞれ回収する溶媒回収工程をさらに含む、請求項1又は2に記載の脂質組成物の製造方法。
- 前記極性溶媒は、低級アルコール、アセトン、アセトニトリル、THF、DMF、又はこれらの組合せを含む、請求項1~3の何れか一項に記載の脂質組成物の製造方法。
- 前記非極性溶媒は、炭素数5~8のアルカン、ジエチルエーテル、t-ブチルメチルエーテル、酢酸エチル、クロロホルム、塩化メチレン、ベンゼン、トルエン、又はこれらの組合せを含む、請求項1~4の何れか一項に記載の脂質組成物の製造方法。
- 前記含水原材料に含まれる全脂質の3質量%以上がリン脂質である、請求項1~5の何れか一項に記載の脂質組成物の製造方法。
- 前記抽出処理工程において、前記抽出溶媒は、予め前記極性溶媒及び前記非極性溶媒を用いて混合有機溶媒に調製した後に前記含水原材料に添加するか、或いは、前記極性溶媒及び前記非極性溶媒を個別に前記含水原材料に順次添加して、前記添加によって前記混合有機溶媒に調製する、請求項1~6の何れか一項に記載の脂質組成物の製造方法。
- 前記含水原材料は、ホタテガイの中腸腺、生殖巣、外套膜、鰓及びイカの眼球、口球、皮のうちの少なくとも1種の水産加工副生成物を含む、請求項1~7の何れか一項に記載の脂質組成物の製造方法。
- 前記含水原材料は60~85重量%の含水率で水を含有する、請求項1~8の何れか一項に記載の脂質組成物の製造方法。
- 前記抽出処理工程において、前記極性溶媒及び前記非極性溶媒は、原材料の含水重量1部に対してそれぞれ2~4部及び1~5部の重量比となる割合で使用される、請求項1~9の何れか一項に記載の脂質組成物の製造方法。
- 前記含水原材料を65℃以上の温度で加熱する加熱処理工程をさらに含む、請求項1~10の何れか一項に記載の脂質組成物の製造方法。
- 前記含水原材料を破砕する湿式破砕工程をさらに含む、請求項1~11の何れか一項に記載の脂質組成物の製造方法。
- 極性溶媒及び非極性溶媒を有する抽出溶媒を用いて含水原材料に含まれる脂質を抽出するための抽出処理部と、
前記抽出処理部において得られた抽出物を、第1脂質分を含有する極性溶媒相と第2脂質分を含有する非極性溶媒相と固形分相とに分離するための分離処理部と
を含む、脂質組成物製造装置。 - 前記分離処理部において分離された非極性溶媒相を吸着剤と接触させて前記非極性溶媒相から不純物を除去するための精製処理部をさらに含む、請求項13に記載の脂質組成物製造装置。
- 前記分離処理部において分離された極性溶媒相及び非極性溶媒相からそれぞれ前記極性溶媒及び前記非極性溶媒を回収するための溶媒回収部をさらに含む、請求項13又は14に記載の脂質組成物製造装置。
- 脂質を構成する脂肪酸中のEPA対DHAの質量比(EPA/DHA)が1.2~7.2であり、全脂質中のリン脂質の割合が3質量%以上54質量%以下であり、カドミウム濃度が脂質1kg当たり0.4mg以下であり、且つ、ヒ素濃度が脂質1kg当たり3mg以下であり、且つ、ダイオキシン類濃度が脂質1g当たり2pg-TEQ以下である、脂質組成物。
- 脂質を構成する脂肪酸中のEPA対DHAの質量比(EPA/DHA)が3.4~7.2であり、全脂質中のリン脂質の割合が18質量%以上39質量%以下である、請求項16に記載の脂質組成物。
