Definitions
- the present invention relates to a microorganism having a diamine production capacity and a method for producing a diamine using the same.
- Biogenic amines are nitrogen compounds produced mainly by decarboxylation of amino acids, amination of aldehydes and ketones, and amino group transfer reactions. Such biogenic amines are low molecular weight and are synthesized during the metabolism of microorganisms, plants and animals, and thus are known as components commonly found in these cells.
- polyamines such as spermidine, spermine, putrescine or putrescine or 1,4-butanediamine and cadaverine are representative examples.
- putrescine is an important raw material for synthesizing polyamine nylon-4,6 by reacting with adipic acid, mainly acrylonitrile and succinonitrile from propylene.
- the fermentation is mainly produced by chemical synthesis.
- cadaverine is a odorless diamine compound produced by hydrolysis of proteins during rot of animal tissues.
- Cadaverine has a chemical formula of NH 2 (CH 2 ) 5 NH 2 and is similar to putrescine.
- the cadaverine may be used as a component of a polymer such as polyamide or polyurethane, a chelating agent or other additives.
- polyamide-5,4 which has a global market of 3.5 million tons per year, is known to be produced by the condensation of cadaverine or succinic acid, and has attracted great attention as an industrial base compound.
- This cadaverine is a diamine found in several microorganisms (Tabor and Tabor, Microbiol Rev., 49: 81-99,1985), and L-lysine decarboxylase from L-lysine in the Gram-negative bacterium Escherichia coli. Direct biosynthesis using Cardaberine concentrations in E. coli are regulated by biosynthesis, degradation, absorption and release of cardaberine (Soksawatmaekhin et al., Mol Microbiol., 51: 1401-1412,2004).
- the inventors of the present invention sought to increase the production capacity of diamines in microorganisms having diamine production ability by identifying proteins having the ability to release diamines such as putrescine or cadaverine, resulting in alkanobacterium hamorityum Derived proteins or proteins having high amino acid sequence homology with diamine have a diamine-releasing ability, which is introduced into a microorganism having a diamine-producing ability to enhance its activity, thereby reducing the diamine-like ability of diamines such as putrescine and cadaverine. It was confirmed that it can increase significantly, and completed the present invention.
- One object of the present invention is to provide a microorganism having a diamine production capacity.
