Abstract
Description
プトレシン生成能を有するコリネバクテリウム属微生物であって、コリネバクテリウム・グルタミクムATCC13032ベースのプトレシン生産菌株KCCM11240P(特許文献6)、コリネバクテリウム・グルタミクムATCC13869ベースのプトレシン生産菌株DAB12−a△NCgl1469(argF欠損、NCgl1221欠損、大腸菌speC導入、argオペロンプロモーター置換、NCgl1469欠損; DAB12-bと命名、 特許文献6)からMFS(major facilitator superfamily)に属するパーミアーゼ(permease)であるNCgl2522を欠失させたとき、プトレシン生産量が減少したことを確認した。
参照例1で確認したところによると、NCgl2522がコードする膜タンパク質がプトレシンを細胞外に排出する機能があることを示した。そこで、本発明者は、NCgl2522のアミノ酸配列に基づいて、米国国立生物情報センター(NCBI、www.ncbi.nlm.noj.gov)のBlastPプログラムを通じて、相同性があるコリネバクテリウム属以外の遺伝子のうち、56%の相同性を有するアルカノバクテリウム・ヘモリティクムDSM 20595のARCH_0271塩基配列(配列番号5)及びアミノ酸配列(配列番号6)を確保した。
<2−1> ATCC13032ベースのプトレシン生産菌株における染色体内トランスポゾン遺伝子内へのARCH_0271、QWA _00075、SMD_2351導入
参照例1で作製したプトレシン排出能が減少したKCCM11240P△NCgl2522において、ARCH_0271、QWA_00075またはSMD_2351遺伝子を染色体内に挿入することによるプトレシン排出効果を確認するために、ARCH_0271、QWA_00075、またはSMD_2351を下記のような過程でトランスポゾン遺伝子内に導入した。
参照例1で作製したプトレシン排出能が弱化したDAB12−b△NCgl2522において、ARCH_0271遺伝子を染色体内に挿入することによるプトレシン排出効果を確認するために、先に製作したpDZTn−P(cj7)−ARCH_0271、pDZTn−P(cj7)−QWA_00075、pDZTn−P(cj7)−SMD_2351を実施例<2−1>と同様の方法でコリネバクテリウム・グルタミクムDAB12−b△NCgl2522に形質転換し、トランスポゾン内ARCH_0271、QWA_00075またはSMD_2351が導入されたことを確認した。
プトレシン生産菌株において、ARCH_0271導入時にプトレシン生産に及ぼす効果を確認するために、前記実施例<2−1>及び<2−2>で製作されたコリネバクテリウム・グルタミクム変異株を対象に、プトレシン生産能を比較した。
<3−1> L−リジン生産菌株であるコリネバクテリウム・グルタミクムKCCM11016Pにおける染色体内トランスポゾン遺伝子内へのARCH_0271導入
実施例<3−1>で製作されたARCH_0271が導入されたL−リジン生産菌株KCCM11016P Tn:P(cj7)−ARCH_0271にカダベリンを生成するための大腸菌由来のリジンデカルボキシラーゼ遺伝子をプラスミドの形で導入した。大腸菌由来のリジンデカルボキシラーゼldcCの塩基配列(配列番号32)及びアミノ酸配列(配列番号33)をNCBIのデータベースから確保した。
カダベリン生産菌株において、ARCH_0271導入時にカダベリン生産に及ぼす効果を確認するために、前記実施例<3−2>で製作されたコリネバクテリウム・グルタミクム変異株を対象に、カダベリン生産能を比較した。
<3−1> W3110にARCH_0271、QWA_00075、またはSMD_2351導入した菌株の製作
プトレシン生成能とカダベリン生合成経路を有する大腸菌野生型菌株W3110において、アルカノバクテリウム・ヘモリティクムDSM 20595のARCH_0271、アルカリゲネス・フェカリス由来のQWA_00075またはステノトロホモナス・マルトフィリア由来のSMD_2351を発現させる場合、プトレシンとカダベリンの生産を増加させることができるかどうかを確認するために、W3110のコリネバクテリウムと大腸菌シャトルベクターベースのpDZTn−P(cj7)−ARCH_0271、pDZTn−P(cj7)−QWA_00075、またはpDZTn−P(cj7)−SMD_2351をそれぞれ導入した。
前記で得られた菌株を対象に、プトレシンとカダベリン生産能を確認した。

Claims (8)
- 配列番号6で表されるアミノ酸配列又はこれと42%以上の配列相同性を有するアミノ酸配列を含むタンパク質の活性が導入または強化された、ジアミン生産能を有する微生物。
- 前記配列番号6で表されるアミノ酸配列と42%以上の配列相同性を有するアミノ酸配列は、配列番号23または26を含むグループから選択されるいずれか一つである、請求項1に記載の微生物。
- 前記微生物は、さらにジアミンアセチルトランスフェラーゼ活性が内在的活性に比べて弱化したものである、請求項1に記載の微生物。
- 前記ジアミンアセチルトランスフェラーゼは、配列番号12、13、及び14からなるグループから選択されたアミノ酸配列を含むものである、請求項3に記載の微生物。
- 前記ジアミンは、プトレシンまたはカダベリンである、請求項1に記載の微生物。
- 前記微生物は、コリネバクテリウム(Corynebacteirum)属微生物またはエシェリキア(Escherichia)属微生物である、請求項1に記載の微生物。
- (i)請求項1〜請求項6のいずれか一項に記載の微生物を培養して培養物を得るステップ、及び
(ii)前記培養された微生物または培養物からジアミンを回収するステップとを含む、ジアミンの生産方法。 - 前記ジアミンは、プトレシンまたはカダベリンである、請求項7に記載の生産方法。
