[1]カチオン性グラフト重合体を用いた、標的物を分離濃縮する方法であって;
(1)塩基性条件下でカチオン性グラフト重合体と標的物を含む試料と接触させ、標的物をカチオン性グラフト重合体に結合させるステップ;
(2)カチオン性グラフト重合体と標的生体分子との結合体を分離するステップ;及び
(3)前記結合体から標的物を溶出するステップ;を
含む方法
<ここで前記カチオン性グラフト重合体は、少なくとも1のアミノ基を有する高分子化合物(a)と、少なくとも1のエポキシ基を有する化合物(b)とを反応して得られるポリアミン誘導体と、エチレン性不飽和単量体(c)とを重合して得られる、ポリアミングラフト重合体である>;
[2]ステップ(1)において、さらに二価又は三価の金属イオンを添加することを含む、上記[1]に記載の方法;
[3]ステップ(2)において、結合体を洗浄する工程をさらに含む、上記[1]又は[2]に記載の方法;
[4]前記カチオン性グラフト重合体が固相面に固定されている、上記[1]~[3]のいずれか一項に記載の方法。

「少なくとも1のアミノ基を有する高分子化合物(a)」における「アミノ基」の数には、特に制限はないが、反応性、使いやすさ等の観点から、1高分子あたり5~15000個であることが好ましく、8~3000個であることが特に好ましい。また、高分子化合物の分子量あたりの個数に換算すると、分子量10000あたり、5個から230個であることが好ましく、10個から130個であることが特に好ましい。
また、本願第1発明において用いられる少なくとも1のアミノ基を有する高分子化合物(a)が有する繰り返し単位の数にも特に制限は無いが、反応性、粘度、ハンドリング、歩留などの観点から、10~150000個であることが好ましく、10~3000個であることが特に好ましい。
一般式(a1)中、R9及びR10はその少なくとも一方が水素原子であり、他方が水素又は炭素数1から8の炭化水素基であり、水素原子又は炭素数1から3のアルキル基であることが好ましい。
また、一般式(a1)において、R9及びR10のいずれか一方もアリル基であること、すなわち2のアリル基を有するジアリルアミン系単量体であることが、重合性等の観点から特に好ましい。
高い重合性を得る観点からは、アリルアミン系単量体(a’)のうち少なくとも一部は、2のアリル基を有するジアリルアミン系単量体であることが好ましく、該アリルアミン系単量体(a’)の30モル%以上がジアリルアミン系単量体であることがより好ましく、50モル%以上がジアリルアミン系単量体であることが特に好ましい。
前記少なくとも1のエポキシ基を有する化合物(b)が、一般式(6)で表される化合物である、ポリアミングラフト重合体(ただし、一般式(6)中のRは、置換又は非置換の1価の炭化水素基である。)であってよい。
前記少なくとも1のエポキシ基を有する化合物(b)が、一般式(7)で表される化合物であって、(1)式中R1、R2、R3、及びR4が、水素原子である、エチレンオキサイド、(2)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または鎖中に水酸基を含む炭素数1~8の直鎖もしくは分岐している飽和炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(3)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または置換若しくは非置換の1価の炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)エポキシ化合物、(4)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または鎖中にエーテル結合を含む炭素数1~8の直鎖もしくは分岐している飽和炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(5)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子またはハロゲンである(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(6)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または不飽和炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(7)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または脂環式または不飽和結合を有する環式炭化水素基を含む炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(8)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または芳香環もしくは複素環を含む炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(9)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子またはエポキシ環を含む炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、多官能エポキシ化合物、(10)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または鎖中にアルコキシシリルを含む炭素数3~12の直鎖もしくは分岐している飽和炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(11)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または鎖中にフッ素を有する炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(12)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または鎖中にカルボキシル基を有する炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、(13)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または鎖中にエステル結合もしくはアミド結合を含む炭素数1~12の直鎖もしくは分岐している飽和炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、並びに(14)式中R1、R2、R3、及びR4が、それぞれ独立に水素原子または鎖中にスルホン酸エステル基を有する炭化水素基である(但し、R1、R2、R3、及びR4の全てが水素原子ではない)、エポキシ化合物、からなる群より選ばれる、ポリアミングラフト重合体であってよい。
前記エチレン系不飽和単量体(c)が、ビニル系単量体、スチレン系単量体、メタクリル酸エステル系単量体、アクリル酸エステル系単量体、アクリルアミド系単量体、アリル系単量体、ジアリル系単量体、及び不飽和カルボン酸からなる群より選ばれる、ポリアミングラフト重合体であってよい。
エチレン系不飽和単量体(c)の分子量には特に制限はないが、グラフト効率などの観点から、分子量28から1100であることが好ましく、28から500であることが特に好ましい。
エチレン系不飽和単量体(c)の炭素原子数にも特に制限はないが、2個から50個であることが好ましく、2個から30個であることが特に好ましい。
これらの単量体の化学構造、CAS番号等は、当業者において自明であるので、その記載を省略する。
20質量%のポリアリルアミン(重量平均分子量3000)を氷水で冷却及び撹拌しながらプロピレンオキシド(アミンに対し2当量)を滴下した。20℃にて24時間反応させた後、溶液を濃縮し、プロピレンオキシド変性ポリアリルアミンを水溶液として得た。
上記に従い調製した42質量%のプロピレンオキシド変性ポリアリルアミンに、14質量%となるように水を加え、20℃で撹拌した。反応溶液はpH10であった。次に65質量%のジメチルアクリルアミド(アミンに対し3当量)と28.5質量%のAPS水溶液12.01g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させ、プロピレンオキシド変性ポリアリルアミンとジメチルアクリルアミドのグラフト重合体を水溶液として得た(重量平均分子量120000)。
(1)と同様に調製した50質量%のプロピレンオキシド変性ポリアリルアミンに、30質量%となるように水を加え、20℃で撹拌した。反応溶液はpH10であった。次に65質量%ジアリルジメチルアンモニウムクロライド(アミンに対し3当量)を加えた。さらに28.5質量%のAPS水溶液96.08g(モノマーに対して20モル%)を分割して加え72時間重合させ、プロピレンオキシド変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た(重量平均分子量24000)。
20質量%のポリアリルアミンを氷水で冷却及び撹拌しながらプロピレンオキシド(アミンに対し0.1当量)を滴下した。20℃にて24時間反応させた後、溶液を濃縮し、プロピレンオキシド変性ポリアリルアミンを水溶液として得た。
プロピレンオキシド変性ポリアリルアミンに20質量%となるように水を加え、次にスチレン (アミンに対し0.3当量)を加え、20℃で撹拌した。反応溶液はpH12で あった。その後、28.5質量%のAPS水溶液12.01g(モノマーに対して20モル%)を滴下し、24時間重合させた。その後70℃で24時間加温し、プロピレンオキシド変性ポリアリルアミンとスチレンの以下に推定される構造のグラフト重合体を得た(ただし、下記一般式中、qは0から3,000の整数であり、rは0から3,000の整数であり、sは0から3,000の整数であり、tは0から10,000の整数であり、uは0から10,000の整数であり、wは0から10,000の整数である)。平均粒径は、120nmであった。
(1)と同様に調製した42質量%のプロピレンオキシド変性ポリアリルアミンに、14質量%となるように水を加え、20℃で撹拌した。次に35質量%の塩酸を加えpHを7に調整し、ジメチルアクリルアミド(アミンに対し3当量)と28.5質量%のAPS水溶液12.01g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させ、プロピレンオキシド変性ポリアリルアミンとジメチルアクリルアミドのグラフト重合体を水溶液として得た(重量平均分子量210000)。
20質量%のポリアリルアミンに13質量%となるように水を加え、氷水で冷却及び撹拌しながらエポキシオクタン(アミンに対し0.1当量)を滴下した。滴下終了後40℃にて24時間反応させた後、溶液を濃縮し、エポキシオクタン変性ポリアリルアミンを水溶液として得た。
上記に従い調製した30質量%のエポキシオクタン変性ポリアリルアミンに、19質量%となるように水を加え、20℃で撹拌した。反応溶液はpH12であった。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させた。その後、28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)を分割してさらに追加し、エポキシオクタン変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た。
20質量%のポリアリルアミンに13質量%となるように水を加え、氷水で冷却及び撹拌しながらエチレングリコールジグリシジルエーテル(アミンに対し0.05当量)を滴下した。滴下終了後40℃にて24時間反応させた後、溶液を濃縮し、エチレングリコールジグリシジルエーテル変性ポリアリルアミンを水溶液として得た。
上記に従い調製した30質量%のエチレングリコールジグリシジルエーテル変性ポリアリルアミンに、19質量%となるように水を加え、20℃で撹拌した。反応溶液はpH12であった。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させ、エチレングリコールジグリシジルエーテル変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を黄色ゲルとして得た。
20質量%のポリアリルアミンに13質量%となるように水を加え、氷水で冷却及び撹拌しながらスチレンオキシド(アミンに対し0.1当量)を滴下した。滴下終了後40℃にて24時間反応させた後、溶液を濃縮し、スチレンオキシド変性ポリアリルアミンを水溶液として得た。

上記に従い調製した30質量%のグリシジルブチレート変性ポリアリルアミンに、19質量%となるように水を加え、20℃で撹拌した。反応溶液はpH12であった。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させた。その後、28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)を分割してさらに追加し、グリシジルブチレート変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た。
(3)と同様に調製したプロピレンオキシド変性ポリアリルアミンに20質量%となるように水を加え、次にスチレン (アミンに対し0.3当量)を加え、20℃で撹拌した。反応溶液はpH12であった。その後、28.5質量%のSPS水溶液12.53g(モノマーに対して20モル%)を滴下し、24時間重合させた。その後70℃で24時間加温し、プロピレンオキシド変性ポリアリルアミンとスチレンのグラフト重合体を得た。平均粒径は、135nmであった。
(3)と同様に調製したプロピレンオキシド変性ポリアリルアミンに20質量%となるように水を加え、次にスチレン (アミンに対し0.3当量)を加え、80℃で撹拌した。反応溶液はpH12であった。その後、28.5質量%のAPS水溶液12.01g(モノマーに対して20モル%)を滴下し、24時間重合させ、プロピレンオキシド変性ポリアリルアミンとスチレンのグラフト重合体を得た。平均粒径は、144nmであった。
ジアリルアミンに79質量%となるように水を加え、氷水で冷却及び撹拌しながらグリシドール(アミンに対し1当量)を滴下した。滴下終了後45℃にて24時間反応させた後、溶液を濃縮し、グリシドール変性ジアリルアミンを水溶液として得た。
78質量%のグリシドール変性ジアリルアミンに35質量%の塩酸(アミンに対し1当量)を加えた。その後、50質量%となるように水を加え、60℃に加温し、2,2’-アゾビス(2-メチルプロピオンアミジン)2塩酸塩(モノマーに対し6モル%)を分割して加え、24時間重合させた。得られた溶液を電気透析によって精製し、グリシドール変性ポリジアリルアミンを水溶液として得た。
上記に従い調製した、43質量%のグリシドール変性ポリジアリルアミンに30質量%となるように水を加え、20℃で撹拌した。反応溶液はpH11であった。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液36.04g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させた。その後、28.5質量%のAPS水溶液36.04g(モノマーに対して10モル%)を分割してさらに追加し、50℃で24時間重合させ、グリシドール変性ポリジアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た(重量平均分子量84000)。
47質量%のポリエチレンイミン(重量平均分子量2000)を撹拌しながらプロピレンオキシド(アミンに対し0.1当量)を滴下した。20℃にて24時間反応させ、プロピレンオキシド変性ポリエチレンイミン水溶液を得た。
上記に従い調製した50質量%のプロピレンオキシド変性ポリエチレンイミンに、14質量%となるように水を加え、20℃で撹拌した。反応溶液はpH12であった。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液31.23g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させ、プロピレンオキシド変性ポリエチレンイミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た。
15質量%のポリビニルアミン(重量平均分子量150000)を撹拌しながらプロピレンオキシド(アミンに対し0.1当量)を滴下した。20℃にて24時間反応させた後、溶液を濃縮し、23質量%のプロピレンオキシド変性ポリビニルアミン水溶液を得た。
上記に従い調製した23質量%のプロピレンオキシド変性ポリビニルアミンに、15質量%となるように水を加え、20℃で撹拌した。反応溶液はpH12であった。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液31.23g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させ、プロピレンオキシド変性ポリビニルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た。
ジアリルアミンに78質量%となるように水を加え、氷水で冷却及び撹拌しながらプロピレンオキシド(アミンに対し0.1当量)を滴下した。20℃にて24時間反応させ、プロピレンオキシド変性ジアリルアミンを水溶液として得た。
79質量%のプロピレンオキシド変性ジアリルアミンに35質量%の塩酸(アミンに対し1当量)を加え、59質量%のプロピレンオキシド変性ジアリルアミン塩酸塩を水溶液として得た。得られた59質量%のプロピレンオキシド変性ジアリルアミン塩酸塩41.33gに7質量%となるように水を加え、60℃に加温した。その後、59質量%のプロピレンオキシド変性ジアリルアミン塩酸塩123.99gと2,2’-アゾビス(2-メチルプロピオンアミジン)2塩酸塩(単量体に対し5.6モル%)を分割して加え、24時間重合させた。得られた溶液を電気透析によって精製し、プロピレンオキシド変性ポリジアリルアミンを水溶液として得た。
上記に従い調製した30質量%のプロピレンオキシド変性ポリジアリルアミンに、24質量%となるように水を加え、20℃で撹拌した。反応溶液はpH12であった。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液14.42g(単量体に対して10モル%)をそれぞれ分割して加え24時間重合させた。その後、28.5質量%のAPS水溶液14.42g(単量体に対して10モル%)を分割してさらに追加し、60℃で24時間重合させ、プロピレンオキシド変性ポリジアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た(重量平均分子量19000)。
15質量%のポリアリルアミン(重量平均分子量1600)を氷水で冷却及び撹拌しながらプロピレンオキシド(アミンに対し0.1当量)を滴下した。20℃にて24時間反応させた後、溶液を濃縮し、プロピレンオキシド変性ポリアリルアミンを水溶液として得た。
上記に従い調整した30質量%のプロピレンオキシド変性ポリアリルアミンに、18質量%となるように水を加え、20℃で撹拌した。反応溶液はpH10であった。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液14.42g(単量体に対して10モル%)をそれぞれ分割して加え24時間重合させた。その後、28.5質量%のAPS水溶液14.42g(単量体に対して10モル%)を分割してさらに追加し、60℃で24時間重合させ、プロピレンキシド変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た(重量平均分子量16000)。
(3)と同様にして調整したプロピレンオキシド変性ポリアリルアミンに20質量%となるように水を加えた。次に2-ビニルピリジン (アミンに対し0.3当量)を加え、20℃で撹拌した。反応溶液はpH12であった。その後、28.5質量%のAPS水溶液12.01g(単量体に対して20モル%)を滴下し、24時間重合させ、プロピレンオキシド変性ポリアリルアミンと2-ビニルピリジンのグラフト重合体を得た。平均粒径は、225nmであった。
出発原料等から、得られたグラフト重合体は下式の構造を有するものと推定された(ただし、下記一般式中、qは0から3,000の整数であり、rは0から3,000の整数であり、sは0から3,000の整数であり、tは0から10,000の整数であり、uは0から10,000の整数であり、wは0から10,000の整数である)。
必要に応じてこれらを精製分離処理操作した後のものであってもかまわない。たとえば、血液から得られる血漿(blood plasma)ないし血清(blood serum)がこれにあたる。
また、金属イオン塩を加えたものであってもかまわない。
細胞とは生体内に存在するものだけでなく、培養細胞であってもかまわない。また正常な細胞だけでなく、がん細胞(たとえば血中循環がん細胞(CTC))であってもよい。
真菌とは一般にキノコ・カビ・酵母と呼ばれる生物の総称であり、白癬菌、カンジダ、ならびにアスペルギルスなどがこれにあたる。
細菌とは細胞膜を持つ原核生物である。ブドウ球菌、大腸菌、サルモネラ菌、緑膿菌、コレラ菌、赤痢菌、炭疽菌、結核菌、ボツリヌス菌、破傷風菌、ならびにレンサ球菌などがこれにあたる。
ウィルスとは、他の生物の細胞を利用して、自己を複製させることのできる微小な構造体で、タンパク質の殻とその内部に入っている核酸からなるものである。ノロウイルス、ロタウイルス、インフルエンザウイルス、アデノウイルス、コロナウイルス、麻疹ウイルス、風疹ウイルス、肝炎ウイルス、ヘルペスウイルス、ならびにHIVなどがこれにあたる。
細胞、真菌、細菌及びウィルスの分解物とは、これらを構成する因子であり、各オルガネラ(核、ゴルジ体、ミトコンドリアなど)、細胞、真菌、細菌の膜画分などのリン酸脂質を含むもの(反転小胞を含む)、細胞、真菌、細菌及びウィルスを構成する因子の複合体などであってもかまわない。また正常な細胞などには存在しないが、アポトーシスによって生じる微小な膜小胞(たとえばEndotherialMicroparticle:EMP)やベクシルであってもよい。
ペプチドとは様々なアミノ酸がつながってできた分子の系統群を指す、生体内で機能している完全なたんぱく質を含み、様々ないわゆる翻訳後修飾(たとえばグリコシル化:糖鎖修飾)を含んでもよい。
核酸とはデオキシリボヌクレオチド(DNA)、リボヌクレオチド(RNA)及びこれらの混合物及び結合物であってもよい。またその構成塩基は天然に存在するヌクレオチド、例えばグアニン(G),アデニン(A),チミン(T),シトシン(C)、ウラシル(U)であるが、それ以外の天然及び人工の修飾塩基を含んでいてもよい。ここで「修飾塩基」とは、上記5つのヌクレオチドが化学的修飾を受けた塩基を意味する。これに限定されないが、メチルシチジン、シュードウリジン、4-チオウリジン、ジヒドロウリジン、キューオシン、ヒポキサンチン(イノシン(I))等がこれにあたる。RNAを鋳型とした逆転写反応により作製されるcDNAも含まれる。
酸性条件下とは、pH7以下、好ましくはpH6以下、より好ましくはpH5以下を指す。酸性溶液を用いて、酸性条件にするのが好ましい。酸性溶液としてはグリシンbuffer、ギ酸溶液、酢酸溶液などがあげられる。
キレート剤としてはエチレンジアミン四酢酸(EDTA)、グリコールエーテルジアミン四酢酸(EGTA)などがこれにあたる。キレート剤のプロトンが乖離しているpHであれば限定されないが、pHは6.0からpH8.0が好ましい。
また、カラムなどに「カチオン性グラフト重合体」を充填した状態であってもよい。
大腸菌の分離、および回収率の確認
1.方法
カチオン性グラフト重合体を用いて回収した大腸菌の回収率を血球分析装置(シスメックス)を用いて求めた。
試料、および試薬は以下の通りである。
1)試料
大腸菌はバイオラッド社製のGFP発現Kitを用いて作製した。
作製した大腸菌をノーマルヒト血清中に必要量(1~2×104cell/μL) 添加したものを試料として用いた。
2)試薬
I : 反応液:1M グリシン-NaOH pH11.0、3M 塩化マグネシウム
II: 分散液:500mM グリシン-HCl pH5.0
3)カチオン性グラフト重合体
20質量%のポリアリルアミン(重量平均分子量3000)を氷水で冷却及び撹拌しながらプロピレンオキシド(アミンに対し0.1当量)を滴下した。20℃にて24時間反応させた後、溶液を濃縮し、プロピレンオキシド変性ポリアリルアミンを水溶液として得た。
プロピレンオキシド変性ポリアリルアミンに20質量%となるように水を加え、次にスチレン (アミンに対し0.3当量)を加え、20℃で撹拌した。その後、濃度28.5質量%の過硫酸アンモニウム(APS)水溶液12.01g(モノマーに対して20モル%)を滴下し、24時間重合させた。その後70℃で24時間加温し、プロピレンオキシド変性ポリアリルアミンとスチレンのカチオン性グラフト重合体を得た(平均粒径120nm)(上記(3)プロピレンオキシド変性ポリアリルアミンとスチレンとのグラフト重合体)。
4)血球分析装置
大腸菌は血球分析装置で血小板(PLT)に検出される。
測定は血球分析装置のプロトコルに従い行った。
試料500μLと20%カチオン性グラフト重合体50μL、および反応液100μLを混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清を回収し、得られた沈殿凝集塊を分散液500μLで完全に分散し分散溶液を得た。上清、および分散溶液は血球分析装置を用いて大腸菌の回収率を求めた。また、対照としてカチオン性グラフト重合体と反応させる前の大腸菌を用いた。
血球分析装置の測定結果を表1に示す。
分離した大腸菌の生存確認
1.方法
カチオン性グラフト重合体を用いて血液中の大腸菌を分離し、分離した大腸菌の生存を培養にて確認した。
試料、および試薬は以下の通りである。
1)試料
実施例1と同様にバイオラッド社製のGFP発現Kitを用いて作製した大腸菌をノーマルヒト血清中に必要量添加し試料として用いた。
2)試薬
I : 反応液:1M グリシン-NaOH pH11.0、3M 塩化マグネシウム
II: 分散液:500mM グリシン-HCl pH5.0
3)カチオン性グラフト重合体
実施例1と同様に調製した。
試料500μLと20%カチオン性グラフト重合体50μL、および反応液100μLを混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清を除去し、得られた沈殿凝集塊を分散液500μLで完全に分散し分散溶液を得た(図2)。上清、および分散溶液の5000分の1量をLB細菌培養寒天培地を用いて37℃で18時間培養し、生存を確認した(図3)。
また、比較例として実施例と同様の方法で凝集沈殿塊を得た後、1.5M NaCl溶液500μL(比較例1)と1.5%TritonX-100 500μL(比較例2)をそれぞれ添加し、凝集塊を完全に分散させた。それぞれの上清、および分散溶液を実施例と同様の方法で培養し生存を確認した。
培養結果を表2に示す。
質量分析器での測定
1.方法
カチオン性グラフト重合体を用いて回収した大腸菌をMALDI TOF (マトリックス支援レーザー脱離イオン化飛行時間型)質量分析器MALDI BioTyper(登録商標)(ブルカー・ダルトニクス)を用いて細菌同定を行った。
試料、および試薬は以下の通りである。
1)試料
大腸菌はバイオラッド社製のGFP発現Kitを用いて作製した。
作製した大腸菌を生理食塩水中に必要量添加したものを試料として用いた。
2)試薬
I :反応液:1M グリシン-NaOH pH11.0、3M 塩化マグネシウム
II :分散液:70% ギ酸
III:凝集液:100% アセトニトリル
3)カチオン性グラフト重合体
実施例1と同様に調製した。
4)質量分析器
マトリクスはブルカー・ダルトニクス社のmatrix HCCA portionedを用いた。
測定はMALDI BioTyper(登録商標)のプロトコルに従い行った。細菌同定の判定はBioTyper(登録商標)に指定するスコアにより評価した。
試料500μLと20%カチオン性グラフト重合体50μL、および反応液100μLを混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た(図1)。上清を除去し、得られた沈殿凝集塊を分散・抽出液50μLで完全に分散させ、菌体中のタンパク質を抽出した。その後、凝集液200μLを添加し、卓上小型遠心機にて30秒間遠心し、カチオン性グラフト重合体を沈殿させた。上清を回収し、質量分析器の測定用試料とした。
質量分析器の測定結果を表3に示す。
血清中の核酸の分離、および回収率の確認
1.方法
カチオン性グラフト重合体を用いて血清中に添加した核酸を分離し、得られた核酸はアガロース電気泳動を行い、SYBR Safe(invitrogen)を用いて染色し、LAS4000(GE)により検出した。回収率はスポットの蛍光強度から求めた。
試料、および試薬は以下の通りである。
1)試料
核酸はプラスミドとゲノムを対象とした。プラスミドは大腸菌から抽出し(実施例4)、ゲノムはHela細胞から抽出した(実施例5)。抽出したプラスミド溶液はLB細菌培養寒天培地を用い培養し、コロニーが形成されなかったことから大腸菌の混入がないことを確認した。また、抽出したゲノム溶液は蛍光顕微鏡にて、細胞が混入していないことを確認した。
抽出したDNAをノーマルヒト血清中に必要量添加し試料とした。また、対照としてDNA未添加のノーマルヒト血清を用いた。
2)試薬
I : 反応液:1M グリシン-NaOH pH11.0、3M 塩化マグネシウム
II: 分散液:500mM EDTA-2Na pH8.0
3)カチオン性グラフト重合体
実施例1と同様に調製した。
4)電気泳動
1.0%アガロースゲルを用いた。
電気泳動用のサンプル調整は6×Loading Dye(TOYOBO)を用いた。
試料500μLと20%カチオン性グラフト重合体50μL、および反応液100μLを混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清、および分散溶液にnacalai tesqueの核酸抽出用試薬フェノール・クロロホルム・イソアミルアルコール(25:24:1)500μL添加し、フェノール相、および水相に分離した。水相中のDNAを300μL回収し、エタノールを600μLおよび酢酸ナトリウム水溶液30μLを混合し、エタノール沈殿によりDNAを沈殿させた。沈殿物は蒸留水100μLで懸濁し、Loading Dyeと混合し電気泳動用の試料とした。1%アガロースゲルを用いて電気泳動を行ない、SYBR Safeにより染色した後、LAS4000にて蛍光検出した(図4)。
DNA回収量を表4に示す。
血清中の細胞の分離、回収率、および立体構造・表面構造の確認
1.方法
カチオン性グラフト重合体を用いて血清中に添加した細胞を分離した。細胞の回収率は血球分析装置(シスメックス)を用いて確認した。さらに細胞表面の抗原抗体反応を蛍光顕微鏡(Life technologies)を用いて確認した。
試料、および試薬は以下の通りである。
1)試料
細胞はHela細胞を用い、ノーマルヒト血清中に必要量(1~2×106cell/mL)添加したものを試料として用いた。
2)試薬
I : 反応液:1M グリシン-NaOH pH11.0、3M 塩化マグネシウム
II: 分散液:500mM EDTA-2Na pH8.0
3)カチオン性グラフト重合体
実施例1と同様に調製した。
4)血球分析装置
細胞はWBCとして測定される。
測定は実施例1同様に血球分析装置のプロトコルに従い行った。
5)抗体
一次抗体は抗ヒトCD146(MCAM)、モノクローナル抗体(N1238)(MONOSAM)を50倍希釈し用いた。
二次抗体はAnti-Mouse IgG Alexa Fluor 594(ライフテクノロジー)を500倍希釈し用いた。
一次抗体、および二次抗体を遮光状態で1時間転倒混和により反応させ、蛍光標識抗体を調製した。
試料500μLと20%カチオン性グラフト重合50μL、および反応液100μLを混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清を回収し、得られた沈殿凝集塊を分散液500μLで完全に分散した(図5)。回収した上清、および得られた分散溶液は血球分析装置を用いて回収率を測定した。対照としてカチオン性グラフト重合体と反応させていない細胞を用いた。
次いで、分散溶液を蛍光標識抗体と混合し、遮光状態で1時間転倒混和により反応させ、蛍光顕微鏡にて抗原抗体反応を確認した(図6)。
血球分析装置でのWBC測定値を表5に示す。
血球分析装置は核を持つ物質をWBCとして測定することから、本発明方法を用いることで、細胞は立体構造を維持した状態で回収可能といえる。
また、Hela細胞とMCAM抗体の抗原抗体反応を蛍光顕微鏡により確認した。この結果から、細胞の表面構造を維持した状態で回収可能といえる。
血清中のベシクルの分離、回収率、および立体構造・表面構造の確認
1.方法
カチオン性グラフト重合体を用いて血清中に添加したベシクルを分離した。ベシクルの回収率は血球分析装置を用いて確認した。また、分離したベシクルの立体構造、および表面構造を維持しているかは蛍光顕微鏡(Life technologies)を用いて確認した。
試料、および試薬は以下の通りである。
1)試料
ベシクルはHela細胞をソニケータで破砕し作製した。
ベシクルを蛍光顕微鏡で観察するために、細胞破砕はDyLight 488 Antibody Labeling kit(Thermo)付属の蛍光色素(DyLight,Ex/Em:493/518)水溶液中で行ないベシクル内部に蛍光色素を取り込ませた。
作製したベシクルをノーマルヒト血清中に必要量添加し試料として用いた。
2)試薬
I : 反応液:1M グリシン-NaOH pH11.0、3M 塩化マグネシウム
II: 分散液:500mM EDTA-2Na pH8.0
3)カチオン性グラフト重合体
実施例1と同様に調製した。
4)血球分析装置
ベシクルはWBCとして測定される。
測定は実施例1と同様に血球分析装置のプロトコルに従い行った。
5)抗体
一次抗体は抗ヒトCD146(MCAM)、モノクローナル抗体(N1238)(MONOSAM)を50倍希釈し用いた。
二次抗体はAnti-Mouse IgG Alexa Fluor 594(ライフテクノロジー)を500倍希釈し用いた。
一次抗体、および二次抗体を遮光状態で1時間転倒混和により反応させ、蛍光標識抗体を調製した。
試料500μLと10%カチオン性グラフト重合体100μL、および反応液50μLを混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清を回収し、得られた沈殿凝集塊を分散液500μLで完全に分散し、分散溶液を得た(図7)。回収した上清、および得られた分散溶液は血球分析装置を用いて回収率を測定した。対照としてカチオン性グラフト重合体と反応させていないベシクルを用いた。
次いで、分散溶液を蛍光標識抗体と混合し、遮光状態で1時間転倒混和により反応させ、蛍光顕微鏡にて抗原抗体反応を確認した(図8)。
血球分析装置でのベシクル測定値を表6に示す。
また、分散溶液中のベシクルが蛍光色素を取り込んだままの状態であり、そのベシクルの淵に蛍光標識抗体が取り囲んでいる様子を蛍光顕微鏡で確認した。この結果から、ベシクルの立体構造、および表面構造を維持した状態で回収可能といえる。
ウィルス粒子の分離、回収確認
1.方法
タンパク質の殻とその内部に入っている核酸からなるウィルスのモデルとして、バクテリオファージを用いる。一定力価(titer)のファージ溶液を調整し、本発明の方法でバクテリオファージを回収し、回収後力価を大腸菌を溶菌させたプラークをカウントすることにより計算する。

バクテリオファージ試料500μLと10%カチオン性グラフト重合体100μL、および反応液50μLを混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清を回収し、得られた沈殿凝集塊を分散液500μLで完全に分散し、分散溶液を得た。得られた溶液を段階希釈し、希釈した溶液を宿主大腸菌培養液と混合し、軟寒天に加えて、寒天プレート上で一晩培養し、そのプラーク数から回収できたファージの力価を計算する。
界面活性剤入り溶液にて回収した細菌の生存確認
1.方法
採血管の分離剤上に沈殿した大腸菌を非イオン性界面活性剤入り溶液を用いて、生存した状態で大腸菌を回収させる。
試料、および試薬は以下の通りである。
1)試料
大腸菌はバイオラッド社製のGFP発現Kitを用いて作製した。
生 理食塩水中に作製した大腸菌2×108cell/mLを添加したものを試料として用いた。
2)試薬
I :1% 界面活性剤溶液
界面活性剤は実験1~6として、スクロースラウレート(同仁化学)、サポニン(シグマ・アルドリッチ)、BPSH(NIKKOL)、ノイゲンTDS-70(第一工業製薬)、TritonX-705(シグマ・アルドリッチ)、またはCHAPS(同仁化学)を用いた。
II :反応液:1M グリシン pH11.0、3M 塩化マグネシウム
III:分散液:500mM グリシン pH5.0
3)カチオン性グラフト重合体
実施例1と同様に調製した。
生理食塩水中に大腸菌を分散させた菌液3mLを採血管に加え、遠心3000rpm、5分間遠心を行った後、上清を除去する。つぎに1%界面活性剤溶液0.5mLを採血管に添加し、分離剤表面に沈殿した菌体を分散させる。
菌体分散液は20℃で30分間、および3時間静置後、10%カチオン性グラフト重合体100μL、および反応液100μLを混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清を除去し、得られた沈殿凝集塊を分散液500μLで完全に分散し分散溶液を得た。分散溶液の100分の1量をLB細菌培養寒天培地を用いて37℃で18時間培養し、コロニー数にて生菌数を確認した。
対照として、界面活性剤溶液の代わりに生理食塩水を用いた(対照1)。
界面活性剤入り溶液を用いて回収した大腸菌の質量分析器での測定
1.方法
採血管の分離剤上に沈殿した大腸菌を界面活性剤入り溶液用いて回収し、カチオン性ポリマーと反応後、70%アセトニトリルにより反応物を洗浄することにより血液成分の影響を抑え、かつ質量分析器での測定が可能な試料を調製した。
試料、および試薬は以下の通りである。
1)試料
大腸菌はバイオラッド社製のGFP発現Kitを用いて作製した。
全血と作製した大腸菌を細菌培養用の培養ボトル(BD社)に2×108cell/mL添加したものを試料として用いた。
2)試薬
I :回収液:0.1% 界面活性剤
II :反応液1:1M グリシン pH11.0
III:反応液2:3M 塩化マグネシウム
IV :洗浄液:70% アセトニトリル
V :抽出液:70% ギ酸
VI :分離液:100% アセトニトリル
3)カチオン性グラフト重合体
実施例1と同様に調製した。
4)電気泳動
E-R155e-パジェル 15%ゲル(ATTO)を用いた。
ゲル染色は2D-銀染色試薬II(コスモ・バイオ)を用いた。
5)質量分析
Auto FlexII(Bruker)のflexControl 2.0を用い取扱い説明書に従って測定をおこなった。
マススペクトル解析は、flexAnalysisを用いた。
採血し、得られた血液10mLを培養ボトルに加え、そこから3mLを採血管に移す。採血管は3000rpm、5分間の遠心を行い、血球成分と菌体を分離する。採血管中の分離剤表面に吸着した大腸菌は回収液500μLを添加し、1.5mLチューブに回収した。ここで回収液に含まれる界面活性剤として実験7~12ではスクロースラウレート、ノイゲンXL60、サポニン、BPSH、TritonX-705、CHAPSを用いた。
また対照2として、回収液に加える界面活性剤の代わりに蒸留水を用いて回収を検討した。回収溶液の入ったチューブに10%カチオン性グラフト重合体100μL、反応液1、および反応液2をそれぞれ100μL混合し、1分間反応させた。その後、卓上小型遠心機にて30秒間遠心し、凝集沈殿塊を得た。上清を除去し、得られた凝集沈殿塊を洗浄液500μLで完全に分散し、卓上遠心機にて30秒間遠心し、凝集沈殿塊を得た。上清は1.5チューブに回収し、試料1とした。凝集沈殿塊は抽出液50μLに完全に分散後、凝集沈殿液150μLを加え、卓上遠心機にて30秒間遠心した。その上清を新しい1.5チューブに移し、試料2とした。
これらはAuto FlexIIにてマススペクトルを測定した。
また、夾雑物の影響を確認するため電気泳動による確認を実施した。測定に用いた試料は対照2、および実験7~12のそれぞれから得られる試料1および2を15000rpm、30分間遠心し、その上清を回収し、さらにその上清は濃縮するために凍結乾燥を一晩行った。乾燥させた試料をSDS sample Buffer 50μLで溶解し、電気泳動用の試料とした。E-R155e-パジェル 15%ゲルを用いて、電気泳動を行った。
電気泳動結果を図9に示す。
試料1は分子量10000、および30000付近にメジャーバンドが確認された。この結果から、洗浄液により、カチオン性ポリマーに吸着した血液成分を除去可能であることが分かった。また、試料2は対照2に対して、30000以下のバンド数が多い。この結果から、洗浄液により血液成分が除去されたことで、菌体由来のタンパク質バンドが検出された。
用いた界面活性剤により質量分析器でのイオン化を阻害しないかどうかを判定するために、得られたマススペクトルからデータ処理用ソフトウェアflexAnalysisのFind Mass List機能によりマスピーク数をカウントした。その結果を表9に示す。カウント条件は、表10に示す。
また、血液成分の影響回避効果を確認するために、菌体由来のマスピークに対する血液成分由来のマスピークのピーク強度の比率を求めた。菌体由来のマスピークはコロニーをサンプルとした場合に検出されたマスピークから6250、9740m/zを選んだ。血液成分由来のマスピークはヘモグロビンのマスピークである15130m/zを選んだ。その結果を表11に示す。
表11より、実験7、8は菌体由来と血液由来のマスピーク比率が1.0以上であり、菌体由来ピークが血液由来ピークよりも高い強度で検出されている。それに対して、実験9~12は菌体由来と血液由来のマスピーク比率が1.0以下であり血液由来ピークが菌体由来ピークよりも高い強度で検出されており、血液成分の影響回避効果が低かった。
この結果から、実験7、8の方法は、血液成分の影響を抑え、かつ質量分析器での測定が可能である。
ウィルス粒子の分離、回収確認(参考例1の追試)
1.方法
1)試料
NBRC(独立行政法人製品評価技術基盤機構 バイオテクノロジーセンター)より購入したEscherichia coli phage T7 (コード:20007)を用い、NBRC指定プロトコルおよび一般的な実験手順、例えば「無敵のバイオテクニカルシリーズ・遺伝子工学実験ノート(上)・2.バクテリオファージの項(羊土社)」に従ってバクテリオファージを調製した。
復水液(10% polypepton、2% Yeast extract、1% MgSO4)200μLで乾燥菌体を懸濁させた後、大腸菌105cell程度を含んだ0.6%寒天培地(10% polypepton、5% Yeast extract、2.5% NaCl、1% 寒天)上に重層した。この培地プレートを37℃、一晩培養し、コロニーのできていない部分の寒天を削り取り(数cm2 程度)、1.5mLチューブに移した。10mM Tris-HCl (pH7.5) 1mLを入れ、90分間20℃にて振盪した後、3000rpm、15分間遠心し上清を回収し、バクテリオファージサンプルとした(冷蔵保存)。
2)試薬
I :反応液1:1M グリシン-NaOH pH11.0
II :反応液2:3M 塩化マグネシウム溶液
III:分散液:1M グリシン-HCl pH5.0
3)カチオン性グラフト重合体
実施例1と同様に調製した。
試料500μLと20%カチオン性グラフト重合体100μL、反応液1:100μL、および反応液2:100μLを混合した。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清を除去し、得られた沈殿凝集塊を分散液100μLで完全に分散し分散溶液を得た。分散溶液を大腸菌105cell程度を含む寒天培地と混合し、37℃、18時間培養し、大腸菌の溶菌を確認した(図11)。
また、ポジティブコントロールとしてカチオン性グラフト重合体と反応させる前のバクテリオファージ試料を大腸菌に感染させた。
グラフト重合体により、バクテリオファージが分離できることがわかった。
分離した大腸菌の生存確認
1.方法
1)試料
実施例1と同様にバイオラッド社製のGFP発現Kitを用いて作製した大腸菌をノーマルヒト血清中に必要量添加し試料として用いた。大腸菌108cell程度。
2)試薬
I: 反応液1:1M グリシン-NaOH pH11.0
II: 反応液2:3M 塩化マグネシウム溶液
III:分散液:1M グリシン-HCl pH5.0
3)カチオン性グラフト重合体
上記(5)エポキシオクタン変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドとのグラフト重合体(PAAEpo-g-DADMAC)、および上記(8)グリシジルブチレート変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドとのグラフト重合体(PAAGB-g-DADMAC)を用いた。
具体的には、20質量%のポリアリルアミンに13質量%となるように水を加え、氷水で冷却及び撹拌しながらエポキシオクタン(アミンに対し0.1当量)を滴下した。滴下終了後40℃にて24時間反応させた後、溶液を濃縮し、エポキシオクタン変性ポリアリルアミンを水溶液として得た。調製した30質量%のエポキシオクタン変性ポリアリルアミンに、19質量%となるように水を加え、20℃で撹拌した。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させた。その後、28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)を分割してさらに追加し、エポキシオクタン変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た。
あるいは、20質量%のポリアリルアミンに13質量%となるように水を加え、氷水で冷却及び撹拌しながらグリシジルブチレート(アミンに対し0.1当量)を滴下した。滴下終了後40℃にて24時間反応させた後、溶液を濃縮し、グリシジルブチレート変性ポリアリルアミンを水溶液として得た。調製した30質量%のグリシジルブチレート変性ポリアリルアミンに、19質量%となるように水を加え、20℃で撹拌した。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させた。その後、28.5質量%のAPS水溶液14.42g(モノマーに対して10モル%)を分割してさらに追加し、グリシジルブチレート変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た。
上記グラフト重合体を終濃度8%、公知方法にて製造されるラテックスビーズ、例えばポリスチレンラテックスLE(平均粒径:120nm、ニットーボーメディカル社製)を終濃度5%となるように両者を混合させ、グラフト重合体溶液とした。
さらに実施例1で用いた上記プロピレンオキシド変性ポリアリルアミンとスチレンとのグラフト重合体(PAA-g-PSt)もポジティブコントロールとして用いた。
試料500μLと上記グラフト重合体溶液100μL、反応液1:100μL、および反応液2:200μLを混合した。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清は別の遠心チューブに移し、さらに15000rpm、5分間遠心し、得られた沈殿物を分散液500μLに分散させた。
上清を除去することにより得られた沈殿凝集塊は、分散液500μLで完全に分散し分散溶液を得た。
上清由来、および沈殿凝集塊由来の分散溶液100μLを別々のLB液体培地5mLに加え、37℃、18時間培養し、大腸菌の生存を確認するために培養前後の培養液試料の吸光度を分光光度計により測定し、その変化量を大腸菌量とした。
また、上記の上清由来、および沈殿凝集塊由来の分散溶液50μLをLB細菌培養寒天培地上に塗布し、37℃、18時間培養し、コロニーの形成によって大腸菌の生存を確認した(図12)。
また、ネガティブコントロールとして、ポリスチレンラテックスLEを含み上記グラフト重合体を含まない場合についても検討した。
分離した大腸菌の生存確認
1.方法
1)試料
実施例1と同様にバイオラッド社製のGFP発現Kitを用いて作製した大腸菌をノーマルヒト血清中に必要量添加し試料として用いた。大腸菌108cell程度
2)試薬
I :反応液1:1M グリシン-NaOH pH11.0
II :反応液2:3M 塩化マグネシウム溶液
III:分散液:1M グリシン-HCl pH5.0
3)カチオン性グラフト重合体
上記(5)エポキシオクタン変性ポリアリルアミンとジアリルジメチルアンモニウムクロライドとのグラフト重合体(PAAEpo-g-DADMAC)を用いた。
試料500μLと上記グラフト重合体溶液100μL、反応液1:100μL、および反応液2:200μLを混合した。試料と上記グラフト重合体を混合させた後、公知方法にて製造されるラテックスビーズ、例えばポリスチレンラテックスLE(平均粒径:120nm、ニットーボーメディカル社製)20μLを添加しよく混合させてから、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清は別の遠心チューブに移し、さらに15000rpm、5分間遠心し、得られた沈殿物を分散液500μLに分散させた。
上清を除去することにより得られた沈殿凝集塊は、分散液500μLで完全に分散し分散溶液を得た。
上記の上清由来、および沈殿凝集塊由来の分散溶液50μLをLB細菌培養寒天培地上に塗布し、37℃、18時間培養し、コロニーの形成によって大腸菌の生存を確認した。
グラフト重合体ははじめにラテックスビーズと混合しておいても、菌体と反応させた後にラテックスビーズを添加しても同じように菌体捕獲した(図13)。
分離した大腸菌の生存確認
1.方法
1)試料
実施例1と同様にバイオラッド社製のGFP発現Kitを用いて作製した大腸菌をノーマルヒト血清中に必要量添加し試料として用いた。大腸菌108cell程度
2)試薬
I : 反応液1:1M グリシン-NaOH pH11.0
II : 反応液2:3M 塩化マグネシウム溶液
III: 分散液:1M グリシン-HCl pH5.0
3)カチオン性グラフト重合体
上記(11)グリシドール変性ポリジアリルアミンとシアリルジメチルアンモニウムクロライドとのグラフト重合体を用いた。
具体的には ジアリルアミンに79質量%となるように水を加え、氷水で冷却及び撹拌しながらグリシドール(アミンに対し1当量)を滴下した。滴下終了後45℃にて24時間反応させた後、溶液を濃縮し、グリシドール変性ジアリルアミンを水溶液として得た。
78質量%のグリシドール変性ジアリルアミンに35質量%の塩酸(アミンに対し1当量)を加えた。その後、50質量%となるように水を加え、60℃に加温し、2,2’-アゾビス(2-メチルプロピオンアミジン)2塩酸塩(モノマーに対し6モル%)を分割して加え、24時間重合させた。得られた溶液を電気透析によって精製し、グリシドール変性ポリジアリルアミンを水溶液として得た。調製した43質量%のグリシドール変性ポリジアリルアミンに30質量%となるように水を加え、20℃で撹拌した。次に65質量%のジアリルジメチルアンモニウムクロライド(アミンに対し3当量)と28.5質量%のAPS水溶液36.04g(モノマーに対して10モル%)をそれぞれ分割して加え24時間重合させた。その後、28.5質量%のAPS水溶液36.04g(モノマーに対して10モル%)を分割してさらに追加し、50℃で24時間重合させ、グリシドール変性ポリジアリルアミンとジアリルジメチルアンモニウムクロライドのグラフト重合体を水溶液として得た。
上記グラフト重合体を終濃度8%、公知方法にて製造されるラテックスビーズ、例えばポリスチレンラテックスLE(平均粒径:120nm、ニットーボーメディカル社製)を終濃度5%となるように両者を混合させ、グラフト重合体溶液とした。
さらに実施例1で用いた上記(3)プロピレンオキシド変性ポリアリルアミンとスチレンとのグラフト重合体(PAA-g-PSt)もポジティブコントロールとして用いた。
試料500μLと上記グラフト重合体溶液100μL、反応液1:100μL、および反応液2:200μLを混合した。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清は別の遠心チューブに移し、さらに15000rpm、5分間遠心し、得られた沈殿物を分散液500μLに分散させた。
上清を除去することにより得られた沈殿凝集塊は、分散液500μLで完全に分散し分散溶液を得た。
上清由来、および沈殿凝集塊由来の分散溶液100μLを別々のLB液体培地5mLに加え、37℃、18時間培養し、大腸菌の生存を確認するために培養前後の培養液試料の吸光度を分光光度計により測定し、その変化量を大腸菌量とした。
また、上記の上清由来、および沈殿凝集塊由来の分散溶液50μLをLB細菌培養寒天培地上に塗布し、37℃、18時間培養し、コロニーの形成によって大腸菌の生存を確認した(図14)。
また、対照として、ポリスチレンラテックスLEを含み上記グラフト重合体を含まない場合についても検討した。
質量分析器での測定
1.方法
1)試料
実施例1と同様にバイオラッド社製のGFP発現Kitを用いて作製した大腸菌をノーマルヒト血清中に必要量添加し試料として用いた。大腸菌108cell程度
2)試薬
I : 反応液1:1M グリシン-NaOH pH11.0
II : 反応液2:3M 塩化マグネシウム溶液
III: 分散液:70%ギ酸
IV : 凝集液:100%アセトニトリル
3)カチオン性グラフト重合体
実施例12で用いた上記(11)グリシドール変性ポリジアリルアミンとシアリルジメチルアンモニウムクロライドとのグラフト重合体を用いた。
上記グラフト重合体を終濃度8%、公知方法にて製造されるラテックスビーズ、例えばポリスチレンラテックスLE(平均粒径:120nm、ニットーボーメディカル社製)を終濃度5%となるように両者を混合させ、グラフト重合体溶液とした。
さらに実施例1で用いた上記(3)プロピレンオキシド変性ポリアリルアミンとスチレンとのグラフト重合体(PAA-g-PSt)もポジティブコントロールとして用いた。
試料500μLと上記グラフト重合体溶液50μL、反応液1:100μL、および反応液2:200μLを混合した。その後、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。
上清を除去し、得られた沈殿凝集塊を分散液30μLで完全に分散させた。続いて、凝集液100μLを混合した後、卓上小型遠心機にて60秒間遠心した。得られた上清を質量分析器の測定試料とした。質量分析器の取り扱いについては、実施例3と同様に行った。
また、対照として、ポリスチレンラテックスLEを含み上記グラフト重合体を含まない場合についても検討した。
分離した大腸菌の生存確認
1.方法
1)試料
実施例1と同様にバイオラッド社製のGFP発現Kitを用いて作製した大腸菌をノーマルヒト血清中に必要量添加し試料として用いた。大腸菌108cell程度
2)試薬
I : 反応液1:1M グリシン-NaOH pH11.0
II : 反応液2:3M 塩化マグネシウム溶液
III: 分散液:1M グリシン-HCl pH5.0
3)カチオン性グラフト重合体
実施例12で用いた上記(11)グリシドール変性ポリジアリルアミンとシアリルジメチルアンモニウムクロライドとのグラフト重合体を用いた。
試料500μLと上記グラフト重合体溶液100μL、反応液1:100μL、および反応液2:200μLを混合した。試料と上記グラフト重合体を混合させた後、公知方法にて製造されるラテックスビーズ、例えばポリスチレンラテックスLE(平均粒径:120nm、ニットーボーメディカル社製)20μLを添加しよく混合させてから、卓上小型遠心機にて30秒間遠心し、沈殿凝集塊を得た。上清は別の遠心チューブに移し、さらに15000rpm、5分間遠心し、得られた沈殿物を分散液500μLに分散させた。
上清を除去することにより得られた沈殿凝集塊は、分散液500μLで完全に分散し分散溶液を得た。
上記の上清由来、および沈殿凝集塊由来の分散溶液50μLをLB細菌培養寒天培地上に塗布し、37℃、18時間培養し、コロニーの形成によって大腸菌の生存を確認した。
グラフト重合体ははじめにラテックスビーズと混合しておいても、菌体と反応させた後にラテックスビーズを添加しても同じように菌体捕獲した(図15)。
Claims (11)
- カチオン性グラフト重合体を用いた、標的物を分離濃縮する方法であって;
(1)塩基性条件下でカチオン性グラフト重合体と標的物を含む試料と接触させ、標的物をカチオン性グラフト重合体に結合させるステップ;
(2)カチオン性グラフト重合体と標的生体分子との結合体を分離するステップ;及び
(3)前記結合体から標的物を溶出するステップ;を
含む方法
[ここで前記カチオン性グラフト重合体は、少なくとも1のアミノ基を有する高分子化合物(a)と、少なくとも1のエポキシ基を有する化合物(b)とを反応して得られるポリアミン誘導体と、エチレン性不飽和単量体(c)とを重合して得られる、ポリアミングラフト重合体である]。 - ステップ(1)において、さらに二価又は三価の金属イオンを添加することを含む、請求項1に記載の方法。
- ステップ(2)において、結合体を洗浄する工程をさらに含む、請求項1又は2に記載の方法。
- 前記カチオン性グラフト重合体が固相面に固定されている、請求項1~3のいずれか一項に記載の方法。
- ステップ(3)において、標的物と結合していないカチオン性グラフト重合体を凝集させるステップを含む、請求項1~3のいずれか一項に記載の方法。
- 請求項1~5のいずれか一項に記載の方法のために用いられるキットであって、カチオン性グラフト重合体を含むキット。
- さらに、二価又は三価の金属イオン塩を含む請求項6に記載のキット。
- さらに、塩基性溶液と二価又は三価の金属イオン塩を含む反応液を含む請求項6に記載のキット。
- さらに、洗浄液を含む請求項6~8のいずれか一項に記載のキット。
- さらに、酸性溶液又はキレート剤を含む分散液を含む請求項6~9のいずれか一項に記載のキット。
- さらに、凝集液を含む請求項6~10のいずれか一項に記載のキット。
