Abstract
Description
Claims (14)
- 標的タンパク質を含む調製物の凝集体含有量を低下させる方法であって、
前記調製物をアリールアニオンと接触させて、混合物を形成するステップ、及び
前記混合物を少なくとも1種の電気陽性固体と接触させて、過剰のアリールアニオンを除去するステップ
を含む方法。 - 前記混合物を、同時に又は順次、少なくとも1種の追加の電気陽性固体と接触させるステップをさらに含む、請求項1に記載の方法。
- 前記混合物を過飽和アラントインと接触させるステップをさらに含む、請求項1に記載の方法。
- アラントインが、(a)約0.6から約50%、(b)約1から約10%、及び(c)約1から約2%からなる群から選択される濃度範囲で存在する、請求項3に記載の方法。
- 前記アリールアニオンがメチルブルーである、請求項1に記載の方法。
- 前記アリールアニオンが、(a)約0.1%から約0.5%、(b)約0.02から約0.4%、及び(c)約0.25から約0.3%からなる群から選択される濃度範囲で存在する、請求項1から5のいずれか一項に記載の方法。
- 操作導電率が、(a)約0.1から約50mS/cm、(b)約1から約30mS/cm、及び(c)約5から約15mS/cmからなる群から選択される範囲である、請求項1から6のいずれか一項に記載の方法。
- 操作pHが、(a)約3から約8、(b)約4から約7、(c)約5から約6、(d)約4.5から5.5、(e)約5.1から約5.3からなる群から選択される範囲である、請求項1から7のいずれか一項に記載の方法。
- 前記混合物が、抗ウイルス剤リン酸トリ(n−ブチル)をさらに含む、請求項1から8のいずれか一項に記載の方法。
- 前記少なくとも1種の電気陽性固体の表面が、静電相互作用、疎水性相互作用、水素結合及び金属親和性からなる群から選択される化学相互作用を促進する、請求項1から9のいずれか一項に記載の方法。
- 前記少なくとも1種の電気陽性固体が粒子である、請求項1から10のいずれか一項に記載の方法。
- 前記標的タンパク質が、組換えタンパク質、抗体、成長ホルモン及び凝固因子からなる群から選択される一種を含む、請求項1から11のいずれか一項に記載の方法。
- 前記調製物が、細胞含有細胞培養収集物、実質的に無細胞の細胞培養収集物、及び部分精製タンパク質からなる群から選択される一種である、請求項1から12のいずれか一項に記載の方法。
- 請求項1から13のいずれか一項に記載の方法の簡便な実施のために提供されるキット。
