Definitions
- the present invention relates to a method for liquid phase synthesis of nucleic acids. More particularly, the present invention provides that the base portion of the nucleoside is (i) an aromatic hydrocarbon ring carbonyl group having at least one hydrophobic group or (ii) an aromatic hydrocarbon ring having at least one hydrophobic group.
- the present invention relates to a novel nucleoside monomer compound substituted with a thiocarbonyl group, and a method for producing a nucleic acid oligomer that can avoid column chromatographic purification for each reaction and can be efficiently synthesized in large quantities using the nucleoside monomer compound as a synthesis unit.
- the present invention also relates to a method for producing the nucleic acid oligomer in a liquid phase. [Background of the invention]
- nucleic acid oligomers such as DNA probes, siRNA, antisense DNA, and antisense RNA have been actively used. Yes.
- a solid phase synthesis method using a phosphoramidite method is mainly known.
- Patent Document 1 discloses a nucleoside compound into which polyethylene glycol (PEG), which is a hydrophilic polymer having a molecular weight distribution in the base part of a nucleoside, is introduced, as a terminal starting material or a building block. It has been proposed to synthesize nucleic acid oligomers by facilitating the separation and purification of intermediate products.
- PEG polyethylene glycol
- Non-Patent Document 1 synthesizes nucleotide dimers or trimers in which adenine, which is the base part of a nucleoside, is substituted with an unsubstituted benzoyl group by repeating reaction in a liquid phase and column chromatography purification, and these are synthesized. There has been proposed a method for producing a nucleic acid oligomer by performing column chromatographic purification after binding by the loamidite method.
- Non-Patent Document 2 uses a phosphotriester method in a liquid phase using a nucleoside in which thymine, which is a base moiety of a nucleoside, is substituted with a p-methoxybenzoyl group (p-toluyl group).
- p-toluyl group p-methoxybenzoyl group
- Patent Document 2 a compound having a property of changing from a liquid phase state to a solid phase state due to a change in solution composition or solution temperature after an organic synthesis reaction is used as an organic synthesis reagent. It has been proposed to perform the separation step easily.
- an organic synthesis reagent a compound having a hydrophobic holding group on an aromatic ring, for example, benzylamine substituted with two docosyloxy groups, which is an alkoxy group having 22 carbon atoms, is described. The use for peptide synthesis has been proposed.
- the production method mainly used in the production of nucleic acid oligomers is a solid phase synthesis method.
- the solid-phase synthesis method can easily achieve the production of nucleic acid oligomers, but due to the reaction on the resin, the reaction field is limited, requires a dedicated synthesizer, and it is difficult to increase the scale. is there.
- synthesis in a liquid phase that can use a general-purpose reactor is effective, but it is necessary for synthesis of nucleic acid oligomers such as siRNA and antisense nucleic acids.
- the problem of the present invention is that it is easy to remove unreacted reagents and activators for each reaction, and can avoid complicated steps such as column chromatography purification, both in the 3 ′ direction and in the 5 ′ direction. It is an object of the present invention to provide a novel nucleoside monomer compound capable of base extension and enabling the synthesis of a nucleic acid oligomer utilizing a building block, a method for producing a nucleic acid oligomer using the same, and the like.
- the base portion of the nucleoside is (i) an aromatic hydrocarbon ring carbonyl group having at least one hydrophobic group or (ii) at least one.
- the use of the novel nucleoside monomer compound makes it possible to extend the base in both the 3 'direction and the 5' direction. Furthermore, the inventors have found that a large amount of nucleic acid oligomers can be efficiently synthesized by synthesizing nucleic acid oligomers in a liquid phase, and as a result of further research based on such knowledge, the present invention has been completed.
