保管及び輸送の簡便性、長期保管時の安定性等の観点から、製造中間体であっても結晶として得られることが好ましい。
本願発明は、4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンの結晶を提供することを目的とする。
その結晶化方法も鋭意検討したところ、4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンをアルコール類と混和することにより、結晶化が可能であることを見出した。
さらにまた、その結晶化方法についても、鋭意検討した結果、化合物1とアルコール類との混和物を極低温下に置くことで得ることができる少量の結晶を種結晶として用いることにより、より効率的に結晶化を行うことができることを見出した。
さらに、4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンの結晶は、当該結晶に対して難溶性を示す脂溶性溶媒中で懸濁撹拌することにより、容易に精製が可能であることを見出した。
すなわち、本発明の要旨は以下のとおりである。

油状物の化合物1と、アルコール類とを混和することで、当該化合物1の結晶を析出させることができる。アルコール類としては、メタノール、エタノール、1-プロパノール、2-プロパノール、またはこれらの混合物を挙げることができ、好ましいアルコール類としてメタノールまたはエタノールを挙げることができる。当該混和に用いるアルコール類としてメタノールまたはエタノールを用いることで、容易に結晶を析出させることができる。
結晶化および結晶化操作を容易にする観点から、化合物1とアルコール類との混和は、化合物1を可溶性溶媒に溶解させ、得られた化合物1の溶液とアルコール類とを混合することにより行なわれることが好ましい。当該混合は、可溶性溶媒に化合物1を溶解することにより得られる溶液をアルコール類に添加しても良いし、可溶性溶媒に化合物1を溶解することにより得られる溶液にアルコール類を添加しても良い。
また、回収率を向上させるために、化合物1とアルコール類との混和物に水を添加することが好ましい。具体的には、例えば、可溶性溶媒を溶媒とする化合物1の溶液とアルコール類との混和物を得るとともに、得られた当該混和物に水を添加するようにしてもよい。
例えば可溶性溶媒として、アセトニトリル等のニトリル系溶媒、アセトン、2-ブタノン等のケトン系溶媒、酢酸エチル、酢酸ブチル等のエステル系溶媒、テトラヒドロフラン等のエーテル系溶媒、トルエン等の炭化水素系溶媒、またはこれらの混液などが挙げられる。また、ジイソプロピルエーテルとアルコール類の混液なども当該可溶性溶媒として用いることができる。なお、可溶性溶媒に含まれるアルコール類としても、メタノール、エタノール、1-プロパノール、2-プロパノール、またはこれらの混合物が挙げられる。
結晶化および結晶化操作を容易にする観点から、可溶性溶媒として、好ましくは、ジイソプロピルエーテルとアルコール類の混液、アセトン、または酢酸エチルが挙げられ、特に好ましくは酢酸エチルである。
なお、可溶性溶媒とアルコール類の混合液に溶解した少量の化合物1を極低温(例えば-78℃)に冷却することにより、少量の結晶を得ることができる。得られた当該結晶を種結晶として化合物1の油状物又は溶液に添加することで結晶化を行うこともできる。
本実施形態の化合物1の結晶は、白色から淡黄色の結晶性の粉末である。ガラス製カバーグラスに挟み、熱板法融点測定器を用いて測定した本実施形態の化合物1の結晶の融点は46℃から49℃である。また、本実施形態の化合物1の結晶は、CuKα 放射を用いた回折角を2θとした粉末X線回折法により、以下のピークを含む粉末X線回折像が観察される。

具体的には、例えば、本実施形態の化合物1の結晶は、粉末X線回折により、図1と実質的に同一の粉末X線回折像が得られる。
なお、粉末X線回折は、例えば、後述する試験例2において用いた装置および操作条件により行うことができる。
なお、熱重量/示差熱分析(TG/DTA)は、例えば、後述する試験例3において用いた装置および操作条件により行うことができる。
本実施形態の化合物1の結晶は、例えば当該結晶に対して難溶性を示す脂溶性溶媒中で懸濁撹拌を行うことにより精製することができる。ここで「化合物1の結晶に対して難溶性を示す脂溶性溶媒」とは、当該結晶を溶媒の温度を化合物1の融点以下の温度としたときに化合物1の結晶を懸濁攪拌することができる脂溶性の溶媒を意味する。
化合物1の結晶に対して難溶性を示す脂溶性溶媒としては、例えば、ヘキサン、シクロヘキサン、ヘプタン等の脂肪族炭化水素系溶媒、ジイソプロピルエーテル、メチル-t-ブチルエーテル等のエーテル系溶媒、又はそれらの混合液が挙げられる。また、化合物1の結晶に対して難溶性を示す脂溶性溶媒は、化合物1の結晶が難溶性である性質を有する限り、上述の可溶性溶媒を含んでいてもよい。
精製効率の観点から、化合物1の結晶に対して難溶性を示す脂溶性溶媒として、溶媒の温度を20℃から40℃としたときに化合物1の結晶を懸濁攪拌することができる溶媒が好ましく、より好ましくは溶媒の温度を25℃から35℃としたときに化合物1の結晶を懸濁攪拌することができる溶媒である。より一層好ましくは、化合物1の結晶に対して難溶性を示す脂溶性溶媒として、ジイソプロピルエーテル又はヘキサンとジイソプロピルエーテルの混合液を用いることができる。ヘキサンとジイソプロピルエーテルの混合比は、体積比で、ジイソプロピルエーテル1部に対してヘキサン0~10部とすることができ、好ましくはジイソプロピルエーテル1部に対してヘキサン1部である。
撹拌温度は結晶が融解しない温度とすることでき、好ましくは20℃から40℃、より好ましくは25℃から35℃とすることができる。撹拌時間の長さは特に限定されないが、結晶の純度向上の観点から、例えば1時間から100時間とすることができ、好ましくは20時間から40時間である。
(実施例1)
3 L四径フラスコに、油状物の化合物1 (115 g)及び酢酸エチル230 mLを投入し、化合物1を溶解した。得られた化合物1の溶液にメタノール115 mLを加え溶解を確認した後、冷却した。-18.9°Cで晶析を確認し、15分間撹拌した。懸濁液にメタノール1035 mLを-19.6~-16.6°Cで滴下後、常水575 mLを-18.1~-2.7°Cで滴下した。懸濁液を-20.0°Cまで冷却し、9.5 cm桐山ロート(ろ紙 No. 3)で析出晶をろ過した。析出晶をメタノール518 mL及び常水57.5 mLの混液で洗浄後、30分間脱液し、湿潤結晶を118 g得た。送風乾燥機を使用し、35°C設定で19時間乾燥し、化合物1の結晶を白色粉末として110 g(回収率95.8%)得た。

50 mL三角フラスコに、化合物1(7.00 g)及び酢酸エチル7.0 mLを投入し、35℃水浴で加熱溶解した。得られた溶液をガラスろ過器で減圧ろ過し、ろ液を200 mL三径フラスコに受けた。50 mL三角フラスコ内を酢酸エチル7.0 mLで洗い込み後、ろ液にメタノール7.0 mLを加えた。
この溶液(化合物1の酢酸エチル(2倍量)及びメタノール(1倍量)溶液)を0.10 mLサンプリングし、これにメタノール0.20 mLを添加して白濁させ、-76℃冷媒浴で冷却し、化合物1を結晶化させた。得られた結晶を種結晶とし、以下の再結晶化に用いた。
上記の工程(化合物1溶液作成工程)で得られた化合物1の溶液を冷却撹拌し、-18.5℃で上記の工程(種結晶作成工程)で得られた種結晶を加え、晶析確認後、10分間撹拌した。懸濁液にメタノール63.0 mLを-19.8~-15.2℃でゆっくり滴下し、その後、精製水35.0 mLを-19.9~-14.3℃でゆっくり滴下した。懸濁液を-20.2~-16.2℃で10分間撹拌後、6.5cmφガラスろ過器で析出晶をろ過した。析出晶をメタノール31.5 mL及び精製水3.5 mLの混液で洗浄した。1時間脱液し、湿潤結晶6.81 gを得た。室温で3時間20分減圧乾燥し、化合物1の結晶を白色粉末として6.81 g(回収率97.2%)得た。
(実施例3)
200 mL三径フラスコに、実施例1で得られた化合物1の結晶 (7.00 g)、ヘキサン28.0 mL及びジイソプロピルエーテル28.0 mLを投入し、30.0~35.0℃で36時間撹拌した(9時間/日、4日間撹拌:夜間は室温で放置)。その際に懸濁精製の途中確認として、撹拌1~3日後の懸濁液から結晶の一部をサンプリングした。
(実施例4)
化合物1、表1に示す溶媒A及び溶媒Bからなる溶液を表1に晶析温度として示す温度に冷却し、冷却撹拌下で結晶を析出させた。次いで、その懸濁液に表1に示す溶媒C及び溶媒Dを添加し、懸濁液を表1にろ過温度として示す温度として結晶をろ過した。
例えば、Run2については以下のような手順で操作を行なった。
Run 2の操作:化合物1 (104 mg)、IPE (0.50 mL)、メタノール(0.10 mL)の混合物を加熱溶解し、-20℃に冷却してメタノール(0.20 mL)を添加した。晶析確認後(-20℃)、28℃に加熱し、結晶の大部分を溶解させた(一部の結晶は残っている)。これにメタノール(0.70 mL)を27℃で添加し、その後、水(0.50 mL)を27℃で添加した。0℃に冷却後、結晶をろ取し、乾燥して、化合物1 (90.4 mg)を収率86.9%で得た。
Run1、Run3~16についても同様に操作を行なったが、Run16のみはメタノールの添加とともに種結晶(重量未測定)の添加を行なった。
表1に結果を示す。また、表1において、IPEとはジイソプロピルエーテルを、IPAとは2-プロパノールを意味する。
実施例3における化合物1懸濁精製の効果(HPLC相対純度)
実施例3の懸濁精製法において、1日(9時間)撹拌ごとに、懸濁液から微量の結晶をサンプリングしてHPLC測定した。その結果を表3に示した。なお、保持時間が10.7分付近であるピークが化合物1のピークである。
サンプリングした結晶から調製した0.5 mg/mL溶液10 μLにつき、次の条件で液体クロマトグラフィー法により試験を行った。
検出器:紫外吸光光度計(測定波長:220 nm)
カラム:内径4.6 mm,長さ15 cmのステンレス管に5 μmの液体クロマトグラ
フィー用オクタデシルシリル化シリカゲルを充てんした逆相カラム(ジーエルサイエンス製,Inertsil ODS-3)を使用した。
カラム温度:30°C付近の一定温度
移動相A:薄めたリン酸(1→1000)
移動相B:液体クロマトグラフィー用アセトニトリル
移動相の送液:移動相A及び移動相Bの混液比を表2のように制御した。
ここで、薄めたリン酸(1 → 1000)とは、リン酸1mLを水に溶解し1000mLとした意味である。
化合物1結晶の粉末X線回折
実施例1で得られた化合物1の結晶を、ガラス製の平板試料ホルダーの充てん部に充てん成形し、次の操作条件で粉末X線回折測定法により測定した。図1に測定結果を示した。また、主なピークの2θ(deg)値を、表4に示した。

化合物1結晶の熱重量/示差熱分析(TG/DTA)
実施例1で得られた化合物1の結晶につき、熱分析法第1法(示差熱分析法:DTA)、第2法(熱質量測定法:TG)により試験を行った。測定結果を図2に示した。

測定温度範囲 : 30~200℃
雰囲気ガス : 乾燥窒素
雰囲気ガスの流量 : 毎分100mL
安定性試験
実施例1で得られた化合物1の結晶につき、加速条件下(40℃/75%RH)及び長期保存条件下(25℃/60%RH)で安定性試験を実施した。両条件とも試験開始から6ヶ月経過後で、結晶の外観に変化は無く、また分解物も生じなかった。
Claims (12)
- CuKα 放射を用いた回折角を2θとする粉末X線回折において下記の2θピークを含む粉末X線回折像が観察される、4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンの結晶。
2θ:9.7、12.9、16.4、16.8、17.6、19.5、21.7、22.6、22.9、23.3、24.5、24.8、26.0、26.4、27.2
- CuKα 放射を用いた回折角を2θとする粉末X線回折により、図1と実質的に同一の粉末X線回折像が得られる4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンの結晶。
- 熱板法により測定される融点が46℃から49℃である、請求項1又は2に記載の4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンの結晶。
- 熱重量/示差熱分析(TG/DTA)において、49℃までに重量の減少が確認できず、且つ50℃付近に単一の吸熱ピークを示す、請求項1から3の何れか一項に記載の4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンの結晶。
- 請求項1から4の何れか一項に記載された結晶を製造する方法であって、4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンをアルコール類と混和させて前記結晶を析出させる工程を含む製造方法。
- 4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンを当該化合物の可溶性溶媒に溶解させ、得られた前記4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンの溶液と前記アルコール類とを混合することにより、前記4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンと前記アルコール類とを混和させる請求項5に記載の結晶の製造方法。
- 前記可溶性溶媒が酢酸エチルである、請求項6に記載の製造方法。
- 前記アルコール類が、メタノール、エタノール、1-プロパノール、2-プロパノール、又はこれらの混合物である、請求項5から7の何れか一項に記載の製造方法。
- 前記4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンと前記アルコール類とを含む混和物に更に水を加える工程を含む、請求項5から8の何れか一項に記載の製造方法。
- -80℃から+10℃の範囲に前記4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンと前記アルコール類とを含む混和物の温度を調節しながら結晶を析出させる、請求項5から9の何れか一項に記載の製造方法。
- 請求項1から4の何れか一項に記載の結晶を、当該結晶に対して難溶性を示す脂溶性溶媒に懸濁して撹拌する工程を含む、4-(3-ベンジルオキシフェニルチオ)-2-クロロ-1-(3-ニトロプロピル)ベンゼンの精製方法。
- 前記脂溶性溶媒が、ヘキサンとジイソプロピルエーテルとの混合溶媒である、請求項11に記載の精製方法。
