Abstract
Description
更なる検討の結果、安全性がより高いとされるインドール化合物を発色基質とした場合においても、修飾酵素が染色効果を高めることが判明した。しかも、修飾酵素による染色は耐シャンプー性に優れていた。また、2種類以上のインドール化合物を併用すれば染色の色彩(色調)を調整可能であることも確認できた。
以下に示す発明は上記成果及び知見に基づく。
[1]EC 1.10.3.1で特定される酵素、EC 1.10.3.2で特定される酵素、EC 1.3.3.5で特定される酵素、EC 1.10.3.4で特定される酵素、EC 1.10.3.3で特定される酵素、及びEC 1.14.18.1で特定される酵素からなる群より選択される酵素に対して、アミン類の化学修飾により正電荷を付加して得られる修飾酵素を含む染色剤。

本発明の第1の局面は繊維又は繊維加工品の染色に用いられる染色剤に関する。本発明の染色剤は例えばケラチン、綿、ジアセテート、亜麻、リンネル、リオセル、ポリアクリル、ポリアミド、ポリエステル、ラミー、レーヨン、テンセル(登録商標)又はトリアセテートからなる繊維、糸、布、織物又は衣類などの染色に利用される。本発明の染色剤は、所定の修飾が施された酸化酵素(以下、「修飾酵素」と呼ぶ)を含む。本発明の染色剤は酸化染料に対して作用し、所望の染色効果を発揮する。従って、典型的には、本発明の染色剤を酸化染料と組み合わせて用いる。本明細書において表現「組み合わせて用いる」又は「組み合わせてなる」とは、酸化染料と修飾酵素が併用されることをいう。一態様として、本発明の染色剤は、酸化染料と修飾酵素が混合された組成物として提供される。当該態様の染色剤を一剤型染色剤と呼ぶ。一方、酸化染料を含む第1要素と、修飾酵素を含む第2要素とからなるキットの形態で本発明の染色剤を提供することもできる。当該態様の染色剤を二剤型染色剤と呼ぶ。二剤型染色剤の場合には各要素が別々の容器又は区画に収容された状態で提供され、使用に際して二つの要素が混合される。尚、一剤型染色剤は簡素な構成となることはもとより、より簡便な使用方法を実現できる点において二剤型染色剤に勝るといえる。

本発明では、天然の状態(ネイティブ)と比較して、保有する正電荷が増加している酸化酵素を用いる。換言すれば、正電荷が付加されるように修飾した酸化酵素が用いられる。酸化酵素には、カテコールオキシダーゼ(EC 1.10.3.1)、ラッカーゼ(EC 1.10.3.2)、ビリルビンオキシダーゼ(EC1.3.3.5)、o-アミノフェノールオキシダーゼ(EC 1.10.3.4)、アスコルビン酸オキシダーゼ(EC 1.10.3.3)、又はモノフェノールオキシダーゼ(EC 1.14.18.1)が用いられる。植物由来(例えば、アナカルジアセア(Anacardiacea)由来、マグニフェラ・インディカ(Magnifera indica)由来、シュイナス・モレ(Schinusmolle)由来、プレイオジニウム・ティモリエンス(Pleiogynium timoriense)由来、ポドカルパセア(Podocarpacea)由来)、動物由来、或いは細菌又は真菌由来(例えば、アスペルギルス(Aspergillus)由来、ボツリチス(Botrytis)由来、コリビア(Collybia)由来、ホメス(Fomes)由来、レンチヌス(Lentinus)由来、ミセリオフソラ(Myceliophthora)由来、ニューロスポラ(Neurospora)由来、プレウロトゥス(Pleurotus)由来、ポドスポラ(Podospora)由来、シタリジウム(Scytalidium)由来、トラメテス(Trametes)由来、リゾクトニア(Rhizoctonia)由来、フミコラ・ブレビス・バー・サーモイデア(H. thermoidea)由来、フミコラ・ブレビスポラ(H. brevispora)由来)の酵素を用いることができる。また、遺伝子組換え技術によって生産された酵素(組換え酵素)を用いることにしてもよい。
本発明の染色剤には、上記の各成分の他、追加の成分(任意成分)が含有され得る。任意成分として、アルカリ性化合物、チオ乳酸、亜硫酸ナトリウム、N-アセチル-L-システイン等の還元剤、界面活性剤、油性成分、シリコーン類、増粘剤、溶剤、水、キレート剤、アミノ酸、各種塩類、保湿剤、防腐剤、紫外線防止剤、アルコール、多価アルコール、香料等を例示することができる。
本発明の第2の局面は、本発明の染色剤を用いた染色方法に関する。本発明の染色方法では、酸素が存在する条件下(即ち酸素雰囲気下)、本発明の染色剤で繊維又は繊維加工品(以下、まとめて「被染色材料」と呼ぶ)を処理する。一剤型の染色剤の場合、例えば、染色剤を必要に応じて希釈した後、被染色材料に塗布する。或いは、染色剤を溶解した溶液に被染色材料を浸漬する。
本発明における修飾酵素はパーマネントウェーブ剤の成分としても有用である。一般にパーマネントウェーブの施術においては、還元剤(チオグリコール酸、チオグリコール酸塩類、システイン、ヒドロキシシステイン、ジヒドロキシシステイン、アセチル化システイン、亜硫酸ナトリウム等)を主成分とする第1剤によって毛髪中のジスルフィド(S-S)結合を切断し、次いで酸化剤を主成分とする第2剤により毛髪を処理し、切断したS-S結合を再形成させる。酸化剤には臭素酸ナトリウム、過ホウ酸塩、過酸化水素などが使用される。このような酸化剤に代えて、本発明の修飾酵素を用いることが可能である。本発明の修飾酵素を使用する場合、一剤型のパーマネントウェーブ剤を構成することも可能である。
1.方法
(1)アミノ基の修飾
以下の方法でアミノ基を付加する修飾をBOに施した。200 mgのBO(ミロセシウム・ベルカリア由来、天野エンザイム社製)に対して、モル比で1000当量のプトレシン、100当量のN-ヒドロキシサクシニミド、および200当量の水溶性カルボジイミド(EDC)を加えて、20mM HEPES(pH 6.5)で20mlとして4℃で1日反応させた。尚、プトレシンは希硫酸でpH調整した。
反応後のサンプルから未反応のプトレシンを除くため、20mM ホウ酸緩衝液(pH9.0)で平衡化させた10DGカラム(BIO-RAD)でサンプルを精製した。
試薬ビリルビン30 mgを0.05 mol/L リン酸塩緩衝液 (pH7.0)(0.05 mmol/L EDTA含有)1.0 mlに溶解して基質溶液とした。0.05mol/L リン酸塩緩衝液 (pH7.0)(0.05 mmol/L EDTA含有)に1%となるようにコール酸ナトリウムを溶解し、この3mlに基質溶液0.2 mlと酵素液0.1 mlを混合後、37℃で反応させ、460 nmの吸光度の減少より測定した。1 分間に1μmolのビリルビンを酸化させるのに要する酵素量を1単位と定義している。
未修飾BOとの電荷の比較のため、等電点電気泳動を行った。
以下の方法でアミノ基を定量した。測定サンプル(タンパク量:0.6-1.0 mg/mlの範囲で)中のタンパク量を揃えた後、調製サンプル(0.25ml)、4%NaHCO3(0.25ml)、0.1% TNBS(0.25ml)水溶液を混合した。40℃で2時間、反応させた。5% SDS(0.25ml)、1M HCl(0.125 ml)を加えて反応を停止させ、345nmの吸光度を測定した。測定サンプルの代わりに水を用いたものをブランクとし、ブランクの測定値を差し引いた値をΔODとした。未修飾BOにおけるアミノ基の結合量を8(リジンの分の7及びN末端の1)としてアミノ基の量を定量した。
上記の通り、調製したBOにはアミノ基が修飾されたことを確認できた。そこで、未修飾BOと修飾BOを用いて染毛試験を実施した。
(1)染毛基剤の調製
EMALEX HC-20(日本エマルジョン株式会社) 2%、乳酸1%、P-フェニレンジアミン(PPD)1%を蒸留水に溶解して、モノエタノールアミンを用いて所定のpHに調整した。次に、ヒドロキシエチルセルロース1.5%を加えて混合し、最終重量80%となるように蒸留水を加えた。続いて、酵素を加え、最終的に100%となるように調製した。
人白髪毛束(10cm, 1g, ビューラックス製)1本につき染毛基剤を2g塗布した。30℃で30分反応させた(途中15分で裏返した)。水で洗浄後、さらに1%SDSで洗浄して、水洗を色が抜けるまで行った。最後に一晩放置し、風乾させた。
分光測色計CM-700d (コニカミノルタ製)を使用し、10°視野、D65 Day Light、SCEモードにて測定した。CIEL*a*b表色系を用いて未処理の白髪との色差(ΔE*ab)で評価した。尚、L*は明度を表す。0~100の数値があり、数値が高いほど明るいことを示す。また、a*は色彩を示す。-60~+60の数値があり、-であるほど緑を示し、+であるほど赤を示す。b*は色彩を示す。-60~+60の数値があり、-であるほど青を示し、+であるほど黄色を示す。ΔE*abは[(ΔL)2+(Δa)2+(Δb)2]1/2で算出される値であり、ヘアカラーの評価において一般的に使用される単位である。基準値(染毛前の白髪の測定値)のL*値、a*値、b*値との差によりΔE*abを算出する。表3に色差の程度とΔE*abの関係を示す。尚、条件によって変動はするものの、一般的なヘアーマニュキュアの場合、ΔE*abが約40となり、過酸化水素を酸化剤に用いたものはΔE*abは50以上を示す。
(1)染毛試験1(pH8,9における染色効果の比較)
染毛試験1の結果を表4及び図2に示す。特にpH8の条件では修飾BOの方が染色効果が高い(色差で3程度の相違)。pH9.0の条件においても染色効果の差を認める。全体的に修飾BOの方が毛が赤く染まり、a*(赤み度)の値も高い(図2右)。肉眼による観察では、数値以上に修飾BOの方が良く染まっている印象であった。
染毛試験2の結果を表5及び図3に示す。特にpH9.0の条件で修飾BOの方が染色効果が高い。(図3左)。いずれのpHにおいても、修飾BOの方がa*が高くなる傾向を示した(図3右)。
染毛試験3の結果を表6及び図4示す。酵素濃度が低い条件下でも修飾BOは高い染色効果を示した(図4左)。また、本試験においても、修飾BOの方が赤み度が高いことを確認した。
発色基質としてPPD(パラフェニレンジアミン)以外に、PAP(パラアミノフェノール)、PTD(パラトルエンジアミン)についても染色効果を調べた(基質特異性の検討)。また、新たにブチルアミンを用いて修飾したものも調製し、その染色効果についても同様に比較した。結果を表7及び図5、6に示す。pH9.0の条件において、修飾BO(プトレシン)は高い染色効果を示すことを確認できる(図5左)。また、修飾BO(プトレシン)は赤み度が高くなる傾向を示した。基質特異性に関しては、非修飾BOと比べてプトレシンで修飾したBOの方が、PPD、PAPに対する染色効果が高い(図6)。また、ブチルアミンで修飾したものは特にPTDに対して染色効果が高くなる傾向を示した(図6)。
各種ポリアミンを用いてBOを修飾し、染色効果を比較した。
(1)アミノ基の修飾
100 mgのBOに対して、モル比で1000当量のポリアミン(プトレシン、3,3'-ジアミノジプロピルアミン、アグマチン硫酸塩)、100当量のN-ヒドロキシサクシニミド、および200当量の水溶性カルボジイミド(EDC)を加えて、20mM HEPES(pH 6.5)で10 mlとして4℃で1日反応させた。尚、プトレシン、3,3'-ジアミノジプロピルアミンは希硫酸でpH調整した。
反応後のサンプルから未反応のポリアミンを除くため、20mM ホウ酸緩衝液(pH9.0)で平衡化させた10DGカラム(BIO-RAD)でサンプルを精製した。
ビリルビンを基質に用いた波長460nmの吸光度変化による測定法を用いた。
未修飾BOとの電荷の比較のため、等電点電気泳動を行った。
TNBSを用いてアミノ基を定量した。
Bradford法によりタンパク量を定量した。尚、BSAを基準とした。
エマール20C(花王)10体積%溶液に、染毛後の毛束を浸漬し、30分間超音波処理を行った。その後、水でよく洗浄して乾燥後に、色差を測定した。耐シャンプー性は、洗浄前の毛髪の色差に対する、洗浄後の毛髪の色差(%)で表した。
調製した各修飾BOについて染毛試験を実施した。方法及び評価はB.の場合と同様である。但し、染色はすべてpH9.0の条件で行った。染毛試験の結果を表11と図8、9に示す。ポリアミンを修飾することで高い染色効果が得られることがわかる。特に、トリアミンを修飾したものは最も染毛効果が高かった(図8左)。最も低い酵素濃度で最大の色差を示したことから、酵素濃度を減らすことで更に染色効果が向上すると予想される。
耐シャンプー試験の結果を表12と図10、11に示す。修飾BOによる染色は耐シャンプー性に優れており、未修飾BOを用いた場合よりも色落ちし難い傾向を示した。修飾BOのこの特徴は染毛用途において極めて重要且つ有意義である。
パラフェニレンジアミンなどよりも安全性が高いとされるインドール化合物(4-アミノインドールや5-アミノインドール等)を発色基質とした場合の染色効果を調べた。
(1)アミノ基の修飾
100 mgのBOに対して、モル比で1000当量の3,3'-ジアミノジプロピルアミン、100当量のN-ヒドロキシサクシニミド、および200当量の水溶性カルボジイミド(EDC)を加えて、20mM HEPES(pH 6.5)で10mlとして4℃で1日反応させた。この反応液を精製したものをPMO(Polyamine Modified Oxidase)として使用した。なお3,3'-ジアミノジプロピルアミン添加後、希硫酸でpH調整した。
(1)染毛試験による染色効果の確認
試験結果を表13及び図12に示す。PMOを添加することで、未修飾BOや過酸化水素を用いた場合よりも有意に染毛効果が高い事が確認された。4AIや5AIを混合することでより、色彩を黒に近い色へ変えることが可能であった(図12)。

試験結果を表14及び図13に示す。未修飾BOよりもPMOの方が染毛効果に優れ、かつ色落ち(堅牢度が高い)も少ない。一方、4AI>4AI+5AI>5AIの順で堅牢度が高くなる傾向にあった。
酵素濃度と染色効果の関係、及び2種以上のインドール化合物を併用した場合の染色効果を調べた。
1.方法
修飾酵素としてPMOを使用した。その他の方法等はD.の場合と同様とした。
(1)染毛試験の結果
試験結果を表15、16及び図14~16に示す。いずれの条件においても、酵素濃度が50U/100g付近で色差はピークとなった(図14)。各種インドール化合物を併用したところ、色差に大きな違いは確認できなかったが(図15)、色彩の変化を認めた(表16、図16)。

試験結果を表17及び図17に示す。5AIを用いた場合には、酵素濃度を高めていくと堅牢度が高くなる(色落ちが少ない)傾向を示した(図17A)。

インドール化合物である5,6-ジヒドロキシインドールを発色基質とした場合の染色効果を調べた。5,6-ジヒドロキシインドールはメラニン前駆体であり、酸素存在下において重合し、メラニン色素が形成される。また、安全性が高いものの、その扱いにくさと染色効果の低さが問題とされている。
発色基質として5,6-ジヒドロキシインドールを0.3重量%用いたことを除いて、試験方法及び評価はD.(インドール類縁体を発色基質とした場合の染色効果1)に準じた。

- EC 1.10.3.1で特定される酵素、EC 1.10.3.2で特定される酵素、EC 1.3.3.5で特定される酵素、EC 1.10.3.4で特定される酵素、EC 1.10.3.3で特定される酵素、及びEC 1.14.18.1で特定される酵素からなる群より選択される酵素に対して、アミン類の化学修飾により正電荷を付加して得られる修飾酵素を含む染色剤。
- 酸化染料を組み合わせてなることを特徴とする、請求項1に記載の染色剤。
- 前記酸化染料と前記修飾酵素を含有した一剤型である、請求項2に記載の染色剤。

- 前記酸化染料が、フェニレンジアミン、アミノフェノール、クレゾール、トルエンジアミン、ナフトール、インドール、インドリン及びこれらの誘導体からなる群より選択される一以上の化合物である、請求項2~6のいずれか一項に記載の染色剤。
- 前記酸化染料が、パラフェニレンジアミン、パラアミノフェノール、パラトルエンジアミン、4-アミノインドール、5-アミノインドール、6-アミノインドール、4-ヒドロキシインドール及び5,6-ジヒドロキシインドールからなる群より選択される一以上の化合物である、請求項2~6のいずれか一項に記載の染色剤。
- 前記酵素がビリルビンオキシダーゼである、請求項1~8のいずれか一項に記載の染色剤。
- 請求項1~9のいずれか一項に記載の染色剤で繊維又は繊維加工品を処理するステップを含む染色方法。
- 前記繊維がケラチン繊維である、請求項10に記載の染色方法。
- 前記ケラチン繊維がヒト毛髪である、請求項11に記載の染色方法。
- pHが7.0~8.0の条件下で前記処理を行う、請求項12に記載の染色方法。
