このように、in vivoでの酸化ストレスの研究は、酸化ストレスと関係する神経変性疾患、心血管疾患、糖尿病、関節リウマチなどの疾患の病理、発症及び細胞生物学に有用な情報を与えるはずである。

(1)Nrf2タンパク質において少なくともNeh2ドメイン配列を含み、かつ、Neh1ドメイン配列又はNeh1−Neh3ドメイン配列を実質的に欠失させるか又は機能欠損させてなる部分タンパク質をコードする核酸配列と、該核酸配列の上流に配置されたストレス誘導性プロモーター配列と、該核酸配列の下流に配置された、検出可能シグナルを発することが可能なタンパク質(例えば、発光若しくは蛍光タンパク質、又は標識可能なタグ配列が結合されたタンパク質)をコードする核酸配列とを含む、酸化ストレスインジケーター発現用核酸構築物。
(2)部分タンパク質が、Nrf2タンパク質からNeh1ドメイン配列又はNeh1−Neh3ドメイン配列を実質的に欠失させてなるアミノ酸配列を含む、上記(1)に記載の核酸構築物。
(3)Nrf2タンパク質が哺乳動物由来Nrf2である、上記(1)又は(2)に記載の核酸構築物。
(4)哺乳動物由来Nrf2がヒトNrf2又はマウスNrf2である、上記(3)に記載の核酸構築物。

本発明のインジケーターは、細胞又は動物においてNrf2−Keap1経路に関連した酸化ストレスを特異的に検出可能にし、また、酸化ストレス時におけるストレス応答検出のS/N比を向上させるという利点を有する。
本明細書は本願の優先権の基礎である日本国特許出願2011−010833号の明細書および/または図面に記載される内容を包含する。
図2は、ヒトNrf2及びマウスNrf2のアミノ酸配列アラインメントを示す。
図3は、p(3xARE)TKbasal−hNrf2(1−433)−GL4−Flagからなる核酸構築物(OKD48)、ならびに、通常条件と酸化ストレス条件での該核酸構築物の転写の抑制と誘導を模式的に示す。OKD48構築物としてp(3xARE)TKbasal−hNrf2(1−433)GL4−Flagを作製した。ヒトNrf2では、Neh2ドメインはKeap1結合及びユビキチン化領域として機能し、Neh1ドメインはDNA結合領域として機能する。OKD48構築物は、3×AREプロモーター、ヒトNrf2(アミノ酸1−433)コード配列及びFlagタグ結合Luciferase(GL4)コード配列を有する。通常条件下では、OKD48構築物の転写は誘導されなかった。また、漏出したOKD48タンパク質は、Keap1系によって分解された。酸化ストレス下では、OKD48は、3×AREプロモーターによって転写誘導され、得られたOKD48タンパク質はKeap1系によって安定化された。このように、酸化ストレスを受けた細胞でのみ発光を検出した。
図4は、p(3xARE)TKbasal−hNrf2(1−433)−GL4−Flagからなる核酸構築物(OKD48)を含むベクターの構造を示す。

1.酸化ストレスインジケーター発現用核酸構築物
本発明の酸化ストレスインジケーター発現用核酸構築物は、Nrf2タンパク質において少なくともNeh2ドメイン配列を含み、かつ、Neh1ドメイン配列又はNeh1−Neh3ドメイン配列を実質的に欠失させるか又は機能欠損させてなる部分タンパク質をコードする核酸配列と、該核酸配列の上流に配置されたストレス誘導性プロモーター配列と、該核酸配列の下流に配置された、発光若しくは蛍光タンパク質、又は(各種試薬で)標識可能なタグ配列が結合されたタンパク質、をコードする核酸配列とを含むことを特徴とする。
本明細書中の「核酸」という用語は、遺伝子、DNA、cDNA、RNA、mRNAそれらの化学修飾体などを含む意味で使用される。
本明細書中の「酸化ストレスインジケーター」という用語は、細胞又は生体内で生じた酸化ストレスのインジケーター(指示物質又は指示薬)であり、本発明では、Nrf2タンパク質において少なくともNeh2ドメインを機能的に保持させ、一方Neh1ドメイン又はNeh1−Neh3ドメインを機能欠損させたものを指す。
Neh2ドメインに関連して使用される「機能的に保持され(る)」という用語は、Keap1と結合してユビキチン化される機能を保持された上で、配列番号1(ヒトNrf2タンパク質)又は配列番号2(マウスNrf2タンパク質)のアミノ酸配列中のアミノ酸1位~93位の配列と80%以上、好ましくは90%以上、より好ましくは95%以上の同一性をもつアミノ酸配列を有するNeh2ドメインをいう。Neh2ドメインの機能は、酸化ストレスのない通常時に酸化ストレスインジケーターがNrf2による分解を受ける上で必須となる。
さらにまた、「Neh1ドメイン又はNeh1−Neh3ドメインを実質的に欠失させるか又は機能欠損させる」という用語は、Neh1ドメインの機能である「DNA結合能」が失われていればよい状態を指し、したがって必ずしもNeh1ドメインの全体が欠失される必要がなく、すなわちNeh1ドメインの一部、例えばその1~50個、好ましくは1~20個のアミノ酸が欠失されずに残存してもよいことを意味する。また、Neh3は非存在であることが好ましい。いずれにしても、Neh1ドメインの存在により、酸化ストレスがない場合であっても酸化ストレス応答を示すことがあるため、Neh1ドメインを非機能的にすることが必要である。
「酸化ストレス」という用語は、生体内で活性酸素種の生成と消去システムのバランスが乱れ、活性酸素種が過剰になる状態を指し、本発明では特にKeap1−Nrf2経路に関連する。酸化ストレスにより誘導されるヘムオキシナーゼ1(HO−1)が紫外線(UV−A)照射により誘導され、この誘導にNrf2タンパク質が関与することが知られている(Allanson M and Reeve VE,J.Invest.Dermatol.2004;122:1030−1036;Zhong JL et al.Photochem.Photobiol.Sci.2010;9:18−24)。従って、例えば、本発明によれば紫外線照射によって引き起こされる生理条件に近い低レベルの酸化ストレスであっても検出することができる。紫外線照射の強度は、例えば1~100mW/cm2、好ましくは2~50mW/cm2、より好ましくは5~30mW/cm2である。酸化ストレスはアテローム性動脈硬化症などの心血管疾患、アルツハイマー病、パーキンソン病、ハンチントン病、筋萎縮性側索硬化症などの神経変性疾患、肝疾患を始めとした各臓器の変性疾患、糖尿病、関節リウマチなどの多くの疾患と関連する。また、急性炎症や慢性炎症などの炎症反応との関連も報告されている。本発明では、上記酸化ストレスインジケーターを用いて酸化ストレスをin vitro又はin vivoで測定することを可能にする。
「Nrf2タンパク質」という用語は、動物細胞内において酸化ストレスに応答して、これに対する防御機能を担う抗酸化タンパク質の発現を誘導する塩基性ロイシンジッパー(bZip)型転写因子として働くタンパク質を指す。細胞に酸化ストレスがない状態では、Nrf2はKeap1と結合した形で細胞質に存在している。このときKeap1はさらにユビキチンリガーゼ複合体と結合して、Nrf2のユビキチン化を促進している。ユビキチン化されたNrf2は蛋白質分解酵素複合体であるプロテアソームによって分解を受けるため、その機能が抑えられている。一方、細胞が酸化ストレスにさらされると、活性酸素がKeap1と反応し、その構造に変化を与える。この構造変化によりKeap1がNrf2から解離することでNrf2のユビキチン化が抑えられる。これにより分解が抑制されたNrf2は、安定化されて細胞内での量を増加させ、核内に移行して転写因子としての機能を発揮するようになる。このときNrf2は抗酸化剤応答配列(antioxidant responsive element;ARE)に結合し酸化ストレス防御遺伝子を発現する。Nrf2−Keap1経路を介した酸化ストレス防御機構については、伊東健、生化学(Biochemistry)78(2)pp.79−92(2006)(日本)に記載されている。
Nrf2は、図1Aに示すように、N末端側から順番に、Neh2ドメイン、Neh4ドメイン、Neh5ドメイン、Neh6ドメイン、Neh1ドメイン及びNeh3ドメインからなる6つのNeh(Nrf2−ECH homology)ドメインによって構成されており、種間で高度に保存されている(伊東健 2006,上掲)。ECHは、ニワトリNrf2の別称である。ここで、Neh2は、Nrf2活性の抑制性の制御ドメインであり、Keap1と結合する部位及びユビキチン化部位を有している。Neh4及びNeh5はともに、転写活性化ドメイン(TAD;Trans−Activation Domain)であり、補助因子である転写コアクチベーター(CBP)と結合する。Neh6は核において恒常的に分解されるドメイン(Degron)である。Neh1は、塩基性−CNC特異的領域(Basic)とロイシンジッパー領域(L−Zip)からなり、BasicはDNAと特異的に結合する、及び、核への移行に関与する領域であり、L−Zipは小Maf群因子とのヘテロ二量体形成に重要な領域である。Neh3は転写活性化に関わる領域(TAD)である(Nioi P et al.Mol Cell Biol 2005;25:10895−10906)。
本明細書で使用する「Neh2−Neh6ドメイン」という用語は、Nrf2タンパク質のNeh2ドメインのN末端アミノ酸残基からNeh1ドメインのN末端アミノ酸の直前のアミノ酸残基までを指す。また、「Neh1−Neh3ドメイン」とは、Neh1ドメインのN末端アミノ酸残基からNrf2タンパク質のC末端アミノ酸残基までを指す。
本明細書で使用される「Nrf2」という用語は、脊椎動物、無脊椎動物、温血動物、哺乳動物、鳥類などの動物に由来のNrf2、その相同体(ホモログ、もしくはオーソログ)、及びその類似体(アナログ)(例えば変異体(Nrf2の生物活性を保持するが、部分的にアミノ酸残基の欠失、置換、挿入もしくは付加を含む)又は化学修飾体)を含む。動物由来のNrf2のアミノ酸配列又は塩基配列は、NCBI(GenBank)、EMBL、DDBJなどの配列データベースにアクセスすることによって入手可能である。
例えば、図2に、哺乳動物由来のNrf2のなかで、特にヒトNrf2及びマウスNrf2のアミノ酸配列(それぞれ配列番号1、配列番号2)のアラインメントが示されている。ヒトとマウスのNrf2間の配列同一性は約80%である。また、図1Bには、ヒトとマウスのNrf2タンパク質のNeh2、Neh4、Neh5、Neh6、Neh1、Neh3の各ドメインの位置と%同一性が示されている。配列同一性は、BLAST(Basic Local Alignment Search Tool)、FASTAなどの公知のアルゴリズムを利用して決定できるし(Altschul SF et al,J Mol Biol 215(3):403−10,1990)、また%同一性は、2つのアミノ酸配列を、一致度が最大となるようにギャップを導入してか又はギャップを導入しないでアラインメントさせたとき、総アミノ酸残基数(ギャップの数も含む)に対する同一アミノ酸残基数のパーセンテージを意味する。この定義は、DNAの塩基配列の%同一性に関しても同様に適用する。この場合、上記アミノ酸配列に代えて塩基配列、上記アミノ酸に代えてヌクレオチド(又は塩基)がそれぞれ対象となる。
このように、本発明では、Nrf2として、哺乳動物由来Nrf2が好ましく、例えばヒト由来Nrf2及びマウス由来Nrf2がより好ましいが、保存性の高いドメインであるNeh2以外のNrf2領域に全成熟アミノ酸残基数の約20%以下、好ましくは約10%以下、さらに好ましくは約5%以下、例えば約4%以下、約3%以下、約2%以下又は約1%以下のアミノ酸変異(欠失、置換、挿入又は付加)があってもよい。すなわち、ヒトNrf2は、配列番号1に示されるアミノ酸配列、又は該アミノ酸配列と80%以上、好ましくは90%以上、より好ましくは95%以上、例えば96%、97%、98%又は99%以上の同一性を有するアミノ酸配列、を含むタンパク質であり、一方、マウスNrf2は、配列番号2に示されるアミノ酸配列、又は該アミノ酸配列と80%以上、好ましくは90%以上、より好ましくは95%以上、例えば96%、97%、98%又は99%以上の同一性を有するアミノ酸配列を含むタンパク質である。
本発明の核酸構築物は、少なくとも、上記インジケーターをコードする核酸配列とストレス誘導性プロモーター配列とを含む。
「ストレス誘導性プロモーター」という用語は、ストレス防御遺伝子の遺伝子発現を制御するプロモーターを指す。そのようなプロモーターは、以下のものに限定されないが、例えばキノンレダクターゼ(QR)プロモーター、Nrf2の標的遺伝子プロモーター、プロモーター活性化部位である抗酸化剤応答配列(ARE)又はMaf認識配列(MARE;Maf−recognition element)などである。或いは、これらのプロモーター又はプロモーター活性化部位と、別のプロモーター又はエンハンサーとの組み合わせであってもよい。ここで、別のプロモーター又はエンハンサーは、ウイルス由来プロモーター、哺乳動物由来プロモーター、組織特異的プロモーターなどを含み、例えばHSV−TK basal promoter(Exp Cell Res.2009 315(15):2496−504)、HSV−TK promoter(Cell 1981;25:385−398,Arch.Biol.Sci.Belgrade 2006;58(4):197−203,phRL−TK(Promega))、CMV(サイトメガロウイルス)プロモーター/エンハンサー(J.Vitol.1995;69:2194−2207,pKM2L−pvCMV(RDB No.5551;RIKEN,pRc/CMV(Invitrogen),pCMVTNT(Promega))やSV−40ウイルスプロモーター/エンハンサー(Cell 1981;27:299−308,J.Virol.1991;65(12):6900−6912,pKM2L−pvSV40(RDB No.5550;RIKEN),pCAT(登録商標)3−Enhancer(Promega),pCAT(登録商標)3−Promoter(Promega),pGL3−control(Promega))、伸長因子 1(EF−1)プロモーター(Nucleic Acids Res.1999;27(24);4775−4782)などである。また、エンハンサーの例は、Nrf2の標的遺伝子由来エンハンサー、例えばヘムオキシゲナーゼ−1(HO−1)エンハンサー(薬学雑誌2007;127(4):757−764(日本),Am.J.Physiol.Renal Physiol.2003;285:F515−F523)、NAD(P)Hデヒドロゲナーゼ−1(NQO1)エンハンサー(Proc Natl Acad Sci U S A.1996;93:14960−5)などである。上記組み合わせは、例えばAREとHSV−TK基本プロモーター(basal promoter)の組み合わせ、AREとHO−1エンハンサーの組み合わせなどであり、好ましくは、AREとHSV−TK basal promoterの組み合わせである。
上記のプロモーター/エンハンサーの配列の具体例を以下に示す。

AREは、酸化ストレスの原因物質である親電子性物質(例えばマレイン酸ジエチル(DEM)、亜ヒ酸ナトリウム(ASN)、t−ブチルヒドロキノン(t−BHQ)など)に応答して生体防御遺伝子の発現誘導を仲介する遺伝子発現制御配列(又は、cis−acting enhancer region)である。本発明の実施形態によれば、AREは複数回リピート、好ましくは3~5回のリピートであり、より好ましくは3回リピートである。複数回リピートでは、ARE配列とARE配列との間に、例えば1~20個、例えば1~10個、の任意の塩基からなる配列がスペーサーとして存在することが好ましい。AREの塩基配列は、TGA(G/C)NNNGC(ここでNはG,C,A又はTである。)(Trends Mol Med.2004;10(11):549−57)であり、例えばTGACATTGC(配列番号8)及び/又はTGACAAAGC(配列番号9)である。後述の実施例で使用されたAREの3回リピート配列(3×ARE)は、次のとおりである(配列番号10)。

本発明の核酸構築物は、ターミネーター、ポリA配列、リボソーム結合配列などの調節配列、必要に応じて選択マーカー配列、をさらに含むことができる。さらにまた、該核酸構築物は、上流及び下流にNrf2遺伝子の5’−非翻訳領域配列及び3’−非翻訳領域配列を含むことが可能であり、これは細胞ゲノムへの組み込みを意図したものである。3’−非翻訳領域配列又は5’−非翻訳領域配列のサイズは、非限定的に、例えば1kb以上、好ましくは1~7kbである。
本発明の酸化ストレスインジケーターは、Nrf2タンパク質において、少なくともNeh2ドメインを含み、かつ、Neh1ドメイン又はNeh1−Neh3ドメイン、好ましくはNeh1−Neh3ドメイン、が発光又は蛍光タンパク質、又は標識可能なタグ配列が結合されたタンパク質、によって置換された改変Nrf2タンパク質である。本発明の核酸構築物では、改変Nrf2タンパク質の発現によって発光又は蛍光の観察が可能となる。
選択マーカー配列は、本発明の核酸構築物が組み込まれた細胞を選択するために有用であり、例えば薬剤耐性遺伝子(例えばカナマイシン耐性遺伝子、アンピシリン耐性遺伝子、ハイグロマイシン耐性遺伝子など)などを含むことができる。また、細胞ゲノムへの組込み後に選択マーカー配列を除去する場合、核酸構築物において、選択マーカー配列の両端にloxP配列、FRT配列などのリコンビナーゼ認識配列を配置し、Cre−loxP系、FLP−FRT系などのリコンビナーゼ−リコンビナーゼ認識配列系を利用して除去を行うことができる。必要であればHSV−TK遺伝子、或いは、ジフテリア毒素(DT)遺伝子もしくはその断片などのネガティブ選択マーカー配列を核酸構築物に挿入することができる。
本発明において酸化ストレスの検出には、検出可能シグナルを発することが可能な任意のタンパク質を使用することができる。また、例えば、細胞死誘導分子などを利用することで酸化ストレスを受けた細胞や組織を特異的に死滅させることもできる。好ましくは発光若しくは蛍光タンパク質、又は標識可能なタグ配列が結合されたタンパク質が使用される。本明細書中ではこれらについて本発明を説明するが、本発明はこれらに限定されるものではない。
本発明で使用される発光又は蛍光タンパク質は、特に制限されずに任意のものを使用できる。蛍光タンパク質としては、オワンクラゲ由来蛍光タンパク質及びその誘導体、例えばGFP,YFP,EBFP,ECFP,EGFP,EYFP,Venusなど、またサンゴ由来蛍光タンパク質及びその誘導体、例えばDsRed,HcRed,mCherryなどを使用することができる。発光タンパク質は、代表的な例として、ホタル、コメツキムシ等の甲虫類、ウミホタル、バクテリアなどの生物がもつ生物発光反応を触媒する酵素であるルシフェラーゼを含む。蛍光タンパク質に関しては、例えばShimomura O et al(1962)J Cell Comp Physiol 59:223−39;Phillips G(2001)FEMS Microbiol Lett 204(1):9−18;Shaner N et al(2005)Nat Methods 2:905−909;Heim R et al(1995)Nature 373:663−664などの文献を挙げることができる。また、ルシフェラーゼに関しては、例えばde Wet JR et al(1985)Proc Natl Acad Sci USA 82:7870−7873、特開2008−289475号公報、特開2005−245457号公報などの文献を挙げることができる。このような蛍光又は発光タンパク質は、Clontech、Roche Diagnostics、チッソ(日本)などからも市販されている。
標識可能なタグ配列が結合されたタンパク質は、タグタンパク質とも称され、例えばテトラシステインタグ(標識試薬:FlAsHラベリング試薬又はReAsHラベリング試薬;Invitrogen)、HaloTag(登録商標)(標識試薬:多数あり;Promega)、SNAP−tag、CLIP−tag、ACP−tag及びMCP−tag(以上、標識:多数あり;New Englnad Biolabs)、Fluorogen activating proteins(FAPs)(Nat.Biotechnol.2008;26(2):235−240)などを含むが、これらに限定されない。これらのタグタンパク質は、タンパク質を蛍光タグ配列と結合してラベルしたものである。
本発明の核酸構築物は、次のようにして作製することができるが、以下は例示であって当該方法に制限されるものではない。
ヒト、マウスなどの動物種由来のNrf2遺伝子をcDNAクローニングし、PCR増幅し、ベクターに組み込んだのち、Nrf2のNeh1コード領域又はNeh1−Neh3コード領域の実質的に両末端を制限酵素で切断・除去し、予め作製しておいた発光若しくは蛍光タンパク質又はタグタンパク質、或いはレポーター配列(例えばFlagタグなど)を連結した発光若しくは蛍光タンパク質又はタグタンパク質、をコードする核酸を該切断サイトに挿入する。Neh1コード領域及びNeh3コード領域はそれぞれ、例えば、ヒトNrf2の場合、配列番号1のアミノ酸配列の434位~561位、562位~605位の配列をコードする領域に相当し、一方、マウスNrf2の場合、Neh1コード領域及びNeh3コード領域はそれぞれ、配列番号2のアミノ酸配列の426位~553位、554位~597位の配列をコードする領域に相当する(図1B参照)。Neh1コード領域又はNeh1−Neh3コード領域の置換において、該領域の機能が不全若しくは機能欠損になるのであれば、該領域の全部でなく一部分を発光若しくは蛍光タンパク質又はタグタンパク質をコードする核酸で置換してもよい。このとき、Nrf2タンパク質からNeh1又はNeh1−Neh3を欠失した残りのタンパク質部分をコードする核酸は、少なくともNeh2を含むアミノ酸配列をコードする核酸からなる。このようにして得られた改変Nrf2タンパク質をコードする核酸をPCRによって増幅し、増幅産物を回収する。Neh3コード領域は必ずしも必須ではないため、PCR増幅の際に、該領域を増幅しないようにプライマーを設計することによってNeh3コード領域を含有しない増幅産物を作製することができる。後述の実施例では、hNrf2(1−433)−Luc、mouse Nrf2(1−426)−Luc(ここで、LucはルシフェラーゼをコードするDNAを表す。)をコードする核酸が増幅産物として得られる。これらの核酸構築物は、未改変Nrf2−Luc核酸構築物と比べてより高いS/N比を付与することができる。
後述の実施例で実際に使用した配列(3xARE−TKbasal−OKD48−LUC)を次に示す(配列番号12)。

PCR増幅のためのプライマーは、目的の増幅産物の塩基配列に基づいて設計可能であるし、またPCR条件は慣用の条件を使用可能である。
一般にPCR反応は、二本鎖DNAを変性して一本鎖DNAに分離するステップ、一本鎖DNAにプライマーをアニーリングするステップ、及び一本鎖DNAを鋳型にしてプライマーを伸長するステップを1サイクルとし、これを約20~45サイクル反復することを含む。変性ステップは、例えば94~98℃、約10秒~約5分の処理からなり、アニーリングステップは、例えば約50~68℃、約10~60秒の処理からなり、伸長ステップは、例えば72℃、約20秒~約10分の処理からなる。全サイクルの開始前に94~98℃、約30秒~約5分の処理を行ってもよく、また、全サイクルの完了後に72℃、約1~10分の処理を行ってもよい。プライマーは、正方向プライマー及び逆方向プライマーからなり、鋳型DNAの塩基配列に基づいて設計される、また、プライマーのサイズは、一般に、15~30塩基、好ましくは20~25塩基である。PCR反応液は、鋳型DNA、耐熱性ポリメラーゼ、Mg2+、dNTP(N=A,T,C,G)などを含むPCRバッファーからなる。サーマルサイクラーなどのPCR装置を使用すると便利にPCR反応を実施することができる。PCRの手法の具体例については、FM Ausubelら、Short Protocols in Molecular Biology(2002),John Wiley & Sons、RA Sikiら,Science 1985,230:1350−1354、HA Erlichら,Science 1991,252:1643−1651などを参照することができる。
一方、核酸構築物のもうひとつの要素であるストレス誘導性プロモーターについては、それを合成するか、又は、それを含むライブラリー、例えば動物組織由来ゲノムライブラリーから調製し、ユニーク制限サイト又はマルチクローニングサイトを含むベクターに組み込む。このベクター中、ストレス誘導性プロモーターの3’側の制限サイト又はクローニングサイトに、改変Nrf2タンパク質をコードする核酸を挿入する。必要に応じて、作製された核酸構築物をベクターから切り出し、電気泳動などの精製法を用いて回収する。ここで、ストレス誘導性プロモーターは、上に例示したようなものであり、好ましくは酸化ストレス誘導性プロモーターであり、本発明ではARE、AREとプロモーター(例えばHSV−TK basal promoterなど)の組み合わせが好ましい。AREは、前述のとおり、AREの複数回リピートであることが好ましく、例えばAREの3回リピート(3×ARE)がより好ましい。
このようにして、図3に示すような本発明の核酸構築物を作製することができる。もしこの核酸構築物をゲノムに組み込むことが意図される場合には、核酸構築物の5’側及び3’側にそれぞれNrf2遺伝子の約1~7kbの5’−非翻訳領域配列、約1~7kbの3’−非翻訳領域配列を連結するとよい。このような核酸構築物を含むベクターは、細胞ゲノム上のNrf2遺伝子(又はNrf2遺伝子座)と相同組換えを起こし、これによって核酸構築物が該ゲノムに組み込まれる。この手法は、特に該核酸構築物を含むトランスジェニック非ヒト動物、例えばトランスジェニックマウスを作製するために利用しうる。
2.ベクター
本発明はさらに、上記の核酸構築物を含むベクターも提供する。
ベクターは、プラスミド(例えばpBruescript系、pUC系など)、ファージ、コスミド、ウイルス、BAC、PACなどのいずれでもよく、これらに制限されない。プラスミドは、動物細胞用プラスミド、例えば哺乳動物用プラスミドであり、便利には市販のプラスミドを使用できる。また、ウイルスは、アデノウイルス、アデノ随伴ウイルス、レトロウイルス、センダイウイルス、レンチウイルスなどのウイルスベクターである。BAC及びPACは人工染色体である。
本発明のベクター構築物の例を図4に示す。
3.細胞
本発明はさらに、上記の核酸構築物又は上記のベクターを含む細胞を提供する。
細胞は、一般に動物細胞であり、好ましくは温血動物細胞、より好ましくは哺乳動物細胞、さらに好ましくはヒト細胞又はマウス細胞である。細胞はまた、培養細胞、初代細胞、継代細胞、株化細胞、形質転換細胞、トランスフェクション細胞、体細胞、生殖細胞、胚性幹(ES)細胞、組織幹細胞、遺伝子操作等により分化多能性が付与された細胞(例えば人工多能性(iPS)細胞を含む)および当該細胞より分化した細胞などを非限定的に包含する。哺乳動物細胞の例は、HEK293細胞、CHO細胞、BHK細胞、COS細胞、HeLa細胞などを含む。
上記細胞は、本発明の核酸構築物又はベクターによって形質転換又はトランスフェクションされる。形質転換又はトランスフェクションの手法は、通常、動物細胞の形質転換法であり、例えばウイルス感染法、エレクトロポレーション、マイクロインジェクション、リポソーム法、リン酸カルシウム法などの公知の手法を含む。
ウイルス感染法は、レトロウイルス、レンチウイルス、アデノウイルス、アデノ随伴ウイルス、センダイウイルスなどの公知のウイルスベクターを利用する手法である。この手法は、ウイルスが動物細胞に感染しやすい特性を利用している。
リポソーム法は、カチオン性リポソーム、例えばコレステロール系カチオン性リポソーム、などのリポソームを利用する手法であり、リポソームによる形質転換法はリポフェクションとも呼ばれる。この手法は、細胞の表面がアニオン性の電気的性質を有することを利用している。また、リポソームの表面に、細胞膜透過性ペプチド(例えばHIV−1 Tatペプチド、ペネトラチン(penetratin)、オリゴアルギニンペプチドなど)を結合したリポソームを利用することもできる。
4.非ヒト動物
本発明はさらに、上記の核酸構築物又は上記のベクターを含む非ヒト動物、すなわちトランスジェニック動物(ヒトを除く)又はその子孫動物を提供する。
トランスジェニック動物(ヒトを除く)の作製には、例えば核移植法や、ES細胞、iPS細胞などの分化多能性細胞を利用することができる。
本明細書において「動物」は、ヒトを除く動物、好ましくは鳥類、哺乳類、例えばサル、チンパンジーなどの霊長目、マウス、ラット、ハムスターなどのげっ歯目、家畜動物として有用なウシ、ヤギ、ヒツジ、ブタ、などの偶蹄目、イヌ、ネコなどの食肉目、ニワトリなどの鳥類などを包含するがこれらに限定されない。
核移植法は、例えば線維芽細胞などの体細胞のゲノムに本発明の核酸構築物又はベクターを導入したのち、該細胞から取出した核を、除核した受精卵又は未受精卵にマイクロインジェクトし、(除核した未受精卵の場合、電気刺激を与えたのち)これを仮親の子宮又は卵管に移植し、発生、出産させてキメラ動物を得ることを含む。
ES細胞を利用する方法は、動物(ヒトを除く)の卵子の胚盤胞から取出した内部細胞塊に本発明の核酸構築物又はベクターをマイクロインジェクション、ミクロセル融合法、エレクトロポレーションなどの手法によって導入したのち、ES細胞を別の胚の胚盤胞に注入し移植胚を得、この胚を仮親の子宮に移植し、出産させてキメラ動物を得ることを含む(例えば、Evans MJ and Kaufman MH 1981,Nature 292:154−156、押村光雄ら編、クロマチン・染色体実験プロトコール(2004年)羊土社)。
iPS細胞を利用する方法として一例を挙げると、動物由来の体細胞に、改変Nrf2タンパク質をコードする核酸が発現可能な状態で上記の核酸構築物又はベクターを導入して形質転換細胞を作製したのち、文献記載の手法(例えばTakahashi K and Yamanaka S 2006,Cell 126:663−676)によって、この細胞に転写因子(例えばOct3/4、Sox2、Klf4、c−Mycなど)又はそれをコードする核酸(ベクターを含む)を導入し、培養することによってES細胞様の分化多能性細胞を得て、このiPS細胞を胚盤胞に注入し移植胚を得、さらにこの胚を仮親の子宮に移植し、出産させてキメラ動物を得ることができる。キメラ動物の選抜は、動物組織からゲノムDNAを取り出し、サザンハイブリダイゼーション、in situハイブリダイゼーション、PCRなどの手法で導入核酸の存在を調べるか、或いは、マウス等の動物の毛色の変化で判定することもできる。
上記のように作製されたキメラ動物はさらに、同種の野生型と交配し、さらに、得られたヘテロ接合性の動物個体同士の交配を繰り返すことによってホモ接合性の子孫動物(ヒトを除く)を得ることができる。
また、上記のヘテロ接合性又はホモ接合性の子孫動物を、任意の同種の動物と交配させて酸化ストレスインジケーターと共に更なる特徴を付与させることもできる。そのような任意の動物としては、小胞体ストレスインジケーター発現用核酸構築物を含む同種の動物や低酸素ストレスインジケーター発現用核酸構築物を含む同種の動物を例示することができるが、これらに限定される訳ではない。また、交配を続けた子孫動物においても本発明の酸化ストレスインジケーターの機能を示す限り、本発明にかかる子孫動物と理解される。
例えば、小胞体ストレスインジケーター発現用核酸構築物を含む同種の動物と交配させる場合には、酸化ストレスインジケーター発現用核酸構築物と小胞体ストレスインジケーター発現用核酸構築物とを含む非ヒト動物を作製することができ、これと同様にして低酸素ストレスインジケーター発現用核酸構築物を含む同種の動物と交配させる場合には、酸化ストレスインジケーター発現用核酸構築物と低酸素ストレスインジケーター発現用核酸構築物とを含む非ヒト動物を作製することができる。
小胞体ストレスは、IRE1a−XBP1経路、ATF6経路、PERK−ATF4経路などと関連し、アルツハイマー病、パーキンソン病、ポリグルタミン病、筋萎縮性側索硬化症などの神経変性疾患、糖尿病、高脂血症、肥満などの代謝性疾患、癌などの疾患と関連している。小胞体ストレスは、小胞体膜タンパク質ATF6、IRE1、PERKなどによって感知され、例えばIRE1は、XBP1のスプライシングを誘導するため、この反応を利用して、小胞体ストレスをインビボで検査できるシステム(ERAIシステム)が開発されているし、さらにまた、ATF6は切断されて活性型ATF6タンパク質に変換され、一方、PERKはeIF−2αをリン酸化して機能を低下させATF4の合成を促進するため、これらの誘導物が小胞体ストレスの分析のために利用されている(特開2005−204516号公報;Yoshida H.,FEBS J.2007 Feb;274(3):630−58.ER stress and diseases.)。
低酸素ストレスは、細胞が正常に生育する酸素濃度に比べて低い酸素濃度の条件下に晒された際に引き起こされるストレス応答を指し、HIF−1αが関与する経路などと関連することが知られている。
小胞体ストレスインジケーター発現用核酸構築物を含む非ヒト動物は、プロモーター(及び、場合により、エンハンサーと組み合わせてもよい。)の下流にXBP1遺伝子(イントロンを含む)、その3’側に発光若しくは蛍光タンパク質又はタグタンパク質コード配列を連結して含む小胞体ストレスインジケーター発現用核酸構築物又は該構築物を含むベクターを有する非ヒト動物である。プロモーター等の例は、限定されないが、CMV(サイトメガロウイルス)やSV−40ウイルスなどのウイルス由来のプロモーター及び/又はエンハンサー、β−アクチンプロモーター、伸張因子1(EF−1)プロモーター、それらの組み合わせなどである。XBP1遺伝子には、例えばその5’端に、Flagなどのタグをコードする配列を連結してもよい。
小胞体ストレスインジケーター発現用核酸構築物の例は、CMVエンハンサー及びβ−アクチンプロモーターを含む配列の下流に、タグコード配列(タグの例:Flag)、XBP1遺伝子、発光若しくは蛍光タンパク質又はタグタンパク質コード配列を順番に含む構築物である。
XBP1(X−Box Binding Protein 1)の塩基配列及びアミノ酸配列は、GenBank(米国NCBI)などから入手可能であり、例えば登録番号の例は、NM_013842(マウス)、NM_00107953、(ヒト)、NM_005080(ヒト)などである。
本発明の非ヒト動物はまた、上記のような酸化ストレス関連疾患を有する非ヒト動物、例えば該疾患のモデル動物(例えばマウス)と交配させて、酸化ストレスインジケーター発現用核酸構築物、及び場合により小胞体ストレスインジケーター発現用核酸構築物、低酸素ストレスインジケーター発現用核酸構築物などの他のストレスインジケーター発現用核酸構築物、を含む上記疾患を有する非ヒト動物を作製することができる。
例えば酸化ストレス関連疾患を有する非ヒト動物として、ヒト型Aβを産生し得るキメラAPP遺伝子を含むADモデル非ヒト動物と交配させる場合には、酸化ストレスインジケーター発現用核酸構築物を含むADモデル非ヒト動物を作製することができる。他の疾患モデル非ヒト動物においても同様である。
また、酸化ストレスインジケーター発現用核酸構築物と小胞体ストレスインジケーター発現用核酸構築物を含む非ヒト動物と、ヒト型Aβを産生し得るキメラAPP遺伝子を含むADモデル非ヒト動物と交配させる場合には、酸化ストレスインジケーター発現用核酸構築物と小胞体ストレスインジケーター発現用核酸構築物とを含むADモデル非ヒト動物を作製することができる。酸化ストレスインジケーター発現用核酸構築物以外の、他のストレスインジケーター発現用核酸構築物は上述した作製方法のように任意に選択すればよく、交配させる他の疾患モデル非ヒト動物においても同様である。
以上のように、酸化ストレスインジケーター発現用核酸構築物を保持させる限りにおいて、酸化ストレスのインジケーターの特徴を有して、他の更なる特徴を含む同種の動物を任意に作製し得る。
酸化ストレス関連疾患を有する非ヒト動物は、ヒト型Aβを産生し得るキメラAPP遺伝子を含むADモデル非ヒト動物(特開2008−000027号公報)、ヒト・プレセニリン2遺伝子の変異体を含むADモデル非ヒト動物(特開平11−146743号公報)、α−シヌクレイン遺伝子を含むパーキンソン病モデル非ヒト動物(再表2005/041649号公報)、Parkin遺伝子の変異体を含むパーキンソン病モデル非ヒト動物(特開2003−018992号公報)、糖尿病モデル非ヒト動物(Diabetes 1997 46:887−894)、肥満症モデル非ヒト動物(Nature 372 425−432,1994)、慢性炎症モデル非ヒト動物(Cell Metab 2009 10 178−88)などを包含するが、これらに限定されない。
5.酸化ストレス測定法
本発明はさらに、本発明の上記細胞又は上記非ヒト動物において、酸化ストレスを提供した際に増大する検出可能シグナル(例えば、発光又は蛍光シグナル)の強度を測定することを含む、酸化ストレスのレベルを測定するための方法を提供する。
本発明の測定方法において、酸化ストレスを測定する際の対照として、例えば、酸化ストレスインジケーター発現用核酸構築物を含まない細胞または非ヒト動物、酸化ストレスを与えていない細胞または非ヒト動物、酸化ストレスの陽性または陰性対照を与えた細胞または非ヒト動物、などを使用することができる。
細胞を使用する場合には、例えば、培養細胞の培地に酸化ストレス物質を添加し、細胞に酸化ストレスを誘導し、細胞内の発光又は蛍光強度の増大を蛍光顕微鏡及び画像化装置、或いは細胞ライセート(lysate)を使用したルシフェラーゼアッセイや蛍光測定法などを用いて測定することができる。また、いかにして酸化ストレス物質が細胞に対し影響を及ぼすかをリアルタイムで観察することができる。
培地としては、通常の動物細胞培地、例えばDMEM、BME、ハムF12、RPMI1640、Fisher培地、ES培地、霊長類ES培地などを基本培地とし、これに、抗生物質(例えばペニシリン、ストレプトマイシンなど)、血清(例えばウシ胎児血清など)、タンパク質因子(例えば塩基性線維芽細胞増殖因子(bFGF)、トランスフェリン、インスリン、白血病抑制因子(LIF)など)、非必須アミノ酸、メルカプトエタノールなどを適宜添加することができる。
培養は、固体培養、液体培養等のいずれでもよく、例えば35~40℃の範囲の温度で、CO2ガス含有空気の雰囲気下で、必要であれば線維芽細胞などのフィーダー細胞を使用して行うことができる。
非ヒト動物を使用する場合には、動物を個体レベルで解析することができる、例えばXengen社のIVIS Imaging Systemを使用することによって、酸化ストレスの存在下で、生きた動物個体で発光又は蛍光シグナルを定量的に測定することができる。この方法であれば、酸化ストレスを受けている細胞を組織的に調べることが可能である。非ヒト動物として、酸化ストレス関連の疾患を有する動物を使用する場合には、酸化ストレスと疾患との関係を視覚的に調べることができるし、一方で、抗酸化ストレス薬、すなわち該疾患の治療薬の開発のために上記動物を使用することができる。また、このとき発光シグナルを得るためには、例えば、核酸構築物にルシフェラーゼ遺伝子を有する非ヒト動物であれば、ルシフェラーゼ基質を該動物に注射するだけで、酸化ストレスに応じて発光を検出することができる。ルシフェラーゼ基質は、例えばルシフェリンであり、ルシフェリンはルシフェラーゼによって酸化されて発光物質に変換される。
非ヒト動物を使用して本発明の酸化ストレス測定方法を実施する場合、非ヒト動物を反復して試験に供することができる。すなわち、非ヒト動物を酸化ストレスに曝露し、一定の期間の間に当該動物を酸化ストレスの影響から回復させ、再度非ヒト動物を酸化ストレスに曝露するとき、この非ヒト動物を使用して酸化ストレスを測定することができる。これにより、経時的な評価が必要な事例や、長期にわたり進行する疾患等への応用が可能となる。上記期間は、例えば1日以上、好ましくは2日以上、より好ましくは3日以上、かつ、例えば10日以下、好ましくは7日以下、より好ましくは5日以下、最も好ましくは約4日である。
6.スクリーニング法
本発明はさらに、本発明の上記細胞又は上記非ヒト動物に、候補薬剤の存在下で、ある特定の酸化ストレスを提供し、薬剤を含まない対照と比べて検出可能シグナル(例えば、発光又は蛍光シグナルなど)の強度が減少するときに該候補薬剤が酸化ストレス抑制能を有すると判定することを含む、酸化ストレス抑制剤をスクリーニングする方法を提供する。
候補薬剤は、天然物、非天然物(又は合成物)、有機物質、無機物質、低分子化合物、タンパク質、糖タンパク質、リポプロテイン、ペプチド、糖類、脂質、核酸、ヌクレオチド、オリゴヌクレオチド、ポリヌクレオチド、ヌクレオシドなど、或いは公知の治療用薬剤又は生物活性物質などから選択可能である。特に公知の治療用薬剤又は生物活性物質であれば、酸化ストレス抑制効果を有することが知られていなかった物質の新しい効果としてスクリーニングされる可能性があり、酸化ストレス抑制効果をもつ物質はさらに、アテローム性動脈硬化症などの心血管疾患、アルツハイマー病、パーキンソン病、ハンチントン病などの神経変性疾患、肝疾患を始めとした各臓器の変性疾患、糖尿病、関節リウマチなどの酸化ストレス関連疾患の治療薬となりうる可能性を有している。
また、例えば本発明の非ヒト動物が、酸化ストレスインジケーター発現用核酸構築物及び小胞体ストレスインジケーター発現用核酸構築物を含む動物である場合には、酸化ストレス及び/又は小胞体ストレスを抑制する薬剤のスクリーニングのために、該動物を使用することができる。小胞体ストレスは、前述したように、神経変性疾患、糖尿病、高脂血症、肥満などの代謝性疾患、慢性炎症、癌などの疾患と関連があり、この動物を用いれば、それぞれのストレスに対するストレス抑制剤を見出すことが可能になる。このように、酸化ストレスインジケーターと共に更なる機能を獲得させた動物については、適宜その機能を利用して、酸化ストレスの状態と合わせて他の生体内の反応等を解析することが可能である。
スクリーニング法は、「5.酸化ストレス測定法」で記載した手法又は実験系により実施することができる。
すなわち、細胞であれば、培養細胞にストレスをかける前に、かけると同時に、またはかけた後に、培地に候補薬剤を添加して細胞を培養し、細胞内の、例えば発光又は蛍光シグナルなどの検出可能シグナルを測定して、対照(候補薬剤なし)と比較して該シグナルを減少させうる物質を選択する。
非ヒト動物であれば、酸化ストレスをかける前に、かけると同時に、またはかけた後に、該動物に、経口投与、静脈内投与、直腸内投与、皮下投与、筋肉内投与、経粘膜投与などの投与経路によって候補薬剤を投与するか、或いは食餌と一緒に摂取させ、蛍光又は発光シグナルなどの検出可能シグナルを測定し、該シグナルを減少させる薬剤を選択する。
本発明の非ヒト動物が、酸化ストレス関連疾患をもつ動物との交配によって作製された動物であれば、疾患の治療効果も同時に判定することが可能になるため、このような動物を使用することで該疾患の治療剤を容易に見出すことが可能になると考えられる。
上記の方法で選抜された薬剤は、賦形剤、溶剤又は担体と組み合わせて医薬品の形態に製剤化されうる。医薬品は、固体製剤(例えば錠剤、顆粒剤、粉末剤、カプセル剤など)、液体製剤(例えば溶液剤、懸濁剤など)、エーロゾル製剤、遅延放出製剤(例えば腸溶剤、多層製剤、コーティング製剤など)、などの多様な製剤に処方されうる。製剤化においては、種々の添加剤、例えば、崩壊剤、結合剤、安定化剤、滑沢剤、乳化剤、香味剤、着色料などを製剤に適宜添加することができる。
実施例1
<方法>
[プラスミドの構築]
p(3xARE)TKbasalを作製するために、PCRによって増幅されたARE断片(マウスGSTYaプロモーター由来;ACTAGTACTAGTGGAAATGACATTGCTAATGGTGACAAAGCAACTTTTCTAGA(配列番号14);結合された制限部位を太字で示した。)を、SpeI−XbaIで消化し、自己連結して、3回リピート断片を形成し、さらにXbaI−SpeI部位を有するpTKbasalに挿入した。XbaI−SpeI部位はTK basal promoterの5’部位に位置する。ヒトNrf2(図1の1−93,1−433,全長(full−length))をコードするcDNAをPCRで増幅し、KpnI−XhoI部位を有するp(3xARE)TKbasal(又は図6のpTKX)に挿入した。ルシフェラーゼ(GL4)をコードするcDNAを、その3’末端において1×Flagタグを用いてPCR増幅し、XhoI−NheI部位を有するp(3xARE)TKbasal−hNrf2(1−433)に挿入した。得られたp(3xARE)TKbasal−hNrf2(1−433)−GL4−FlagをOKD48−Lucプラスミドとして使用した。同様の手法でGFPバージョン、すなわちp(3xARE)TKbasal−hNrf2(1−433)−Venus−Flagを作製し、OKD48−Venusプラスミドとして使用した。
ヒトNrf2の過剰発現ベクター(pCAX−hNrf2)を、KpnI−XhoIを有するpCAXにPCR増幅ヒトNrf2断片を挿入することによって構築した。また、ヒトKeap1の過剰発現ベクター(pCAX−hKeap1)を、HindIII−XhoI部位を有するpCAXにPCR増幅ヒトKeap1断片を挿入することによって構築した。同様の手法で、末端切断型バージョンのpCAX−hKeap1(1−314)を構築した。
[細胞培養、トランスフェクション及び処理]
HeLa細胞及びHEK293T細胞を、100U/mlペニシリン、100μg/mlストレプトマイシン及び10%ウシ胎児血清を補充したDMEM培地中で、5%CO2含有の雰囲気下、37℃で培養した。リン酸カルシウム−DNA沈殿法を用いて細胞内にプラスミドDNAを挿入した。薬剤に対する細胞応答をテストするために、細胞を、種々の時間にわたって、10μM ASN(sodium arsenite),100μM DEM(diethylmaleate),200μM H2O2,2.5μg/ml tunicamycin,1μM thapsigargin,1mM DTT,100μM etoposide又は100μg/ml TTFA(thenoyltrifluoroacetone)で処理した。
[ルシフェラーゼアッセイ]
OKD48−Lucレポーターを使用する二連のルシフェラーゼアッセイでは、phRL−TK(Promega)を内部対照として使用した。HeLa細胞を24ウエルプレートに撒き、その後、プラスミドDNAでトランスフェクションした。トランスフェクション後24時間目に、ルシフェラーゼアッセイのために細胞を溶解した。各レポーター活性の測定を、ルシフェラーゼアッセイ系(Promega)とルミノメーター(Berthold)を用いて行った。その結果は、3回の実験の平均±SEMとして示されている。それぞれの値を、誘導倍率として示し、この倍率は、未処理(NT)の倍率(図5A)、Nrf2過剰発現のない未処理(NT)の倍率(図5D)、又はKeap1過剰発現のない未処理(NT)の倍率(図5E)に対してそれぞれ正規化された。このとき各NTの倍率を1.0とした。
[蛍光画像化及び蛍光強度測定]
細胞の画像をとるために、HEK293T細胞を6ウエルプレートに撒き、その後、プラスミドDNAでトランスフェクションした。トランスフェクション後24時間目に、FSX100(Olympus)によって蛍光画像を得た。細胞溶解液の蛍光を測定するために、細胞を、digitoninバッファー(50mM Tris−HCl(pH7.5),1mM EDTA,10mM EGTA及び10μM digitonin)中で、37℃、30分間、溶解した。16,500gの遠心分離によって溶解液を透明にした。蛍光光度計(ARVO MX−2,PerkinElmer)を用いて上清の蛍光を測定した(発光波長535nm;励起波長485nm)。pCAG−GL3を内部対照として使用した。各サンプルの蛍光強度は、同時トランスフェクションされたルシフェラーゼ活性に対して正規化された。値は、3回の実験の平均±SEMとして示された。
[ウエスタンブロット分析]
細胞を、SDSサンプルバッファー(50mM Tris−HCl(pH6.8),2% SDS,50mM DTT,10%グリセロール及び1μg/mlブロモフェノールブルー)中で溶解した。溶解液を、98℃で、10分間加熱し、SDS−PAGEを用いて溶解液中のタンパク質を分離した。電気泳動後、タンパク質をフッ化ポリビニリデン多孔質メンブレンに電気的に転写し、標準的手法で、Luciferaseに対するモノクローナル抗体(Promega),GFPに対するモノクローナル抗体(Nacalai tesque),GAPDHに対するモノクローナル抗体(Cell Signaling Technology),又はNrf2に対するポリクローナル抗体(Santa Cruz)を用いて免疫学的に検出した。
[トランスジェニックマウス]
p(3xARE)TKbasal−hNrf2(1−433)−GL4−Flagの4.5−kb SpeI−SfiI断片を、トランスジーンとして、C57BL/6マウス受精卵のなかにマイクロインジェクションし、トランスジェニック子孫を、5’−ATC ACC AGA ACA CTC AGT GG−3’(配列番号15)及び5’−ACT CGG CGT AGG TAA TGT CC−3’(配列番号16)のプライマーを用いるPCRによって選抜した。得られたマウスをin vivoでの画像化アッセイのために使用した。D−ルシフェリン(4.5mg)を腹腔内注射したのち、マウスを、標準的プロトコールに従って、in vivo画像化システムIVIS(Xenogen)を用いて分析した。動物を含む実験プロトコールは、RIKEN(日本)の動物研究委員会によって承認された。
<結果>
[新規の酸化ストレスインジケーター(OKD48)の設計と構築]
今回、本発明者らは、新しいタイプの酸化ストレスインジケーターであるOKD48の作製に、Keap1−Nrf2経路を利用した。ストレス依存的な安定化に関連するNrf2の部分断片をルシフェラーゼ(Luc)遺伝子と融合させ、ストレス誘導性のプロモーターから発現させた。図3に示すように、通常時、内在性のNrf2は核内に存在しないので、レポーターの転写レベルの発現は誘導されず、また、漏出してくるレポータータンパク質もKeap1による分解抑制を受けシグナルは検出されない。しかし、酸化ストレス時には、内在性Nrf2量の増加と核内への移行に伴いレポーター遺伝子の転写が誘導され、さらに、Keap1による分解抑制も解除されるので、レポータータンパク質の発現が増強され、シグナルが検出される。
図6にその一例を示すように、用いるNrf2の部分断片と、プロモーターの組み合わせについて、いくつかのバリエーションを作製し、最も良い応答性を示すものを探索した。Nrf2断片に関しては、アミノ酸(a.a.)1~93、1~433、または全長(full length)について調べた。プロモーターについては、陰性対照として用いたHSV−TKプロモーターの他、HO−1エンハンサー、3×AREプロモーターを検討した。その結果、Nrf2のa.a.1~433領域と3×AREプロモーターの組み合わせが、酸化ストレス誘導剤であるASNやDEMに対して最も良い応答性を示した。
[In vitroでのOKD48の特性決定]
このように構築したOKD48(ARE−Nrf2 jointed stress associated indicator)−Lucが、実際の哺乳動物細胞で酸化ストレスインジケーターとして機能するかテストするために、融合遺伝子の発現ベクターを培養細胞に導入し、アッセイを行った(図5)。HeLa細胞を用いて、各種薬剤に対する応答性をルシフェラーゼアッセイで評価したところ、OKD48−Lucは、ASN(亜ヒ酸ナトリウム)やDEM(マレイン酸ジエチル)など酸化ストレス誘導剤に対して特異的に応答し、Tun(tunicamycin)やTg(thapsigargin)などの小胞体ストレス誘導剤、還元剤であるDTT(dithiothreitol)、トポイソメラーゼIIの阻害剤でアポトーシスを誘導するEtp(etoposide)、chemical hypoxia誘導剤のTTFA(thenoyltrifluoroacetone)では、ほとんど活性化されなかった(図5A)。このレポーターアッセイの結果と一致して、Luc抗体を用いたウエスタンブロットによっても、OKD48−LucのASNやDEMに特異的なタンパク質レベルでの上昇が確認できた(図5B)。図5Cに示すように、これらの薬剤の中で内在性Nrf2を顕著に誘導するのは、ASNとDEMのみであった。これらの結果は、作製したレポーターの特異性が内在性Nrf2と一致することを示しており、OKD48−Lucの酸化ストレスインジケーターとしての有用性を意味する。
次に本発明者らは、OKD48−Lucに対する関連因子の過剰発現の影響を調べた。図5Dに示すように、Nrf2の過剰発現により、通常条件(normal condition)においてもOKD48−Lucの活性は100倍以上上昇し、さらにその活性はASN処理によって若干上昇した。一方、Keap1の過剰発現によって、ストレスの有無にかかわらずOKD48−Lucの活性は低下し、Nrf2との結合領域を欠くKeap1(1−314)の過剰発現では逆にOKD48−Lucの活性が上昇した(図5E)。これらの結果は、OKD48−LucがNrf2により誘導され、Keap1による分解/安定化の制御を受けるという想定した実施モデルを支持する。
このようなルシフェラーゼを融合したレポーターに加えて、GFPを融合したレポーター(OKD48−Venus)も作製し、培養細胞でのチェックを行った(図7)。Luc型のレポーターと一致して、顕微鏡観察においてOKD48−VenusはASNやDEM処理に伴って蛍光を発した(図7A)。また、同様の結果が細胞溶解液(Lysate)を用いた蛍光測定(図7B)やGFP抗体を用いたウエスタンブロット(図7C)によっても確認された。
[OKD48マウスの作製及びin vivoでの酸化ストレスのモニタリング]
In vivoで酸化ストレスをモニターするために、3×ARE機動のNrf2(1−433)−Luc発現遺伝子を有するOKD48−トランスジェニックマウスを作製した。in vivoで酸化ストレス下の細胞を効果的に用いることが可能であることを確認するために、種々の組織や器官で酸化ストレスを誘導するASNをトランスジェニックマウスに腹腔内注射し(12.5mg/kg)、6時間後、開腹下で発光シグナルを観察した。ASNが注射されたトランスジェニックマウスは、肝臓で強い発光を示した(図8)。他の組織や器官でもいくらか発光が検出された(例えば、胃,及び腎臓;データを示さず)。これに対して、PBSを注射された対照トランスジェニックマウスは、ほとんど発光を示さなかった。この結果は、作製したトランスジェニックマウスが、動物個体での酸化ストレスの検出に利用可能であることを示す。
実施例2
[マウスOKD48の作製とその特性]
実施例1と同様の手法で作製したマウスOKD48−Luc(3xARE+TK basal promoter+mouse Nrf2(1−426)+Luc)を導入したHeLa細胞を、酸化ストレス物質(DEM又はASN)で処理し、発光強度をルシフェラーゼアッセイ法で測定した。その結果、mouse Nrf2(1−426)−Lucが、DEM及びASNの両方で、mouse Nrf2(1−93)及びmouse Nrf2(full)と比べて最も高いS/N比を示した(図9)。

Claims (16)
- Nrf2タンパク質において少なくともNeh2ドメイン配列を含み、かつ、Neh1ドメイン配列又はNeh1−Neh3ドメイン配列を実質的に欠失させるか又は機能欠損させてなる部分タンパク質をコードする核酸配列と、該核酸配列の上流に配置されたストレス誘導性プロモーター配列と、該核酸配列の下流に配置された、検出可能シグナルを発することが可能なタンパク質をコードする核酸配列とを含む、酸化ストレスインジケーター発現用核酸構築物。
- 部分タンパク質が、Nrf2タンパク質からNeh1ドメイン配列又はNeh1−Neh3ドメイン配列を実質的に欠失させてなるアミノ酸配列を含む、請求項1に記載の核酸構築物。
- Nrf2タンパク質が哺乳動物由来Nrf2である、請求項1又は2に記載の核酸構築物。
- 哺乳動物由来Nrf2がヒトNrf2又はマウスNrf2である、請求項3に記載の核酸構築物。
- 部分タンパク質が、(a)配列番号1(ヒト)又は配列番号2(マウス)のNrf2タンパク質のアミノ酸配列中のアミノ酸1位~93位からなるNeh2ドメイン配列、(b)該Neh2ドメイン配列と80%以上の同一性を有するアミノ酸配列、或いは、(c)該Neh2ドメイン配列において1若しくは数個のアミノ酸の欠失、置換又は付加を含むアミノ酸配列、を含むものである、請求項1~4のいずれか1項に記載の核酸構築物。
- 部分タンパク質が、(d)配列番号1(ヒト)又は配列番号2(マウス)のNrf2タンパク質のアミノ酸配列中のそれぞれアミノ酸1位~433位又はアミノ酸1位~425位からなる、Neh2−Neh6ドメイン配列を含むアミノ酸配列、(e)該Neh2−Neh6ドメイン配列を含むアミノ酸配列と80%以上の同一性を有するアミノ酸配列、或いは、(f)該Neh2−Neh6ドメイン配列を含むアミノ酸配列において1若しくは数個のアミノ酸の欠失、置換又は付加を含むアミノ酸配列、を含むものである、請求項1~4のいずれか1項に記載の核酸構築物。
- ストレス誘導性プロモーターが、抗酸化剤応答配列(ARE)、又はAREとプロモーターの組み合わせからなる、請求項1~6のいずれか1項に記載の核酸構築物。
- AREが複数回リピートからなる、請求項7に記載の核酸構築物。
- 前記核酸構築物が、その3’末端にタグコード配列を含む、請求項1~8のいずれか1項に記載の核酸構築物。
- タグコード配列がFlagタグコード配列である、請求項9に記載の核酸構築物。
- 請求項1~10のいずれか1項に記載の核酸構築物を含むベクター。
- 請求項1~10のいずれか1項に記載の核酸構築物又は請求項11に記載のベクターを含む細胞。

- 非ヒト動物が小胞体ストレスインジケーター発現用核酸構築物又は低酸素ストレスインジケーター発現用核酸構築物をさらに含む、請求項13に記載の非ヒト動物。
- 請求項12に記載の細胞又は請求項13又は14に記載の非ヒト動物において、酸化ストレスを提供した際に増大する検出可能シグナルの強度を測定することを含む、酸化ストレスを測定するための方法。
- 請求項12に記載の細胞又は請求項13又は14に記載の非ヒト動物に、候補薬剤の存在下で、ある特定の酸化ストレスを提供し、該候補薬剤非存在下の対照と比べて検出可能シグナルの強度が減少するときに該候補薬剤が酸化ストレス抑制能を有すると判定することを含む、酸化ストレス抑制剤をスクリーニングする方法。
