Abstract

Способ получения гетерологичного полипептида в эукариотических микроорганизмах, предусматривающий конструирование рекомбинантной плазмидной ДНК, содержащей промотор, последовательность, кодирующую сигнальный пептид и структурный ген, трансформацию полученной рекомбинантной ДНК штаммов -реципиентов, культивирование трансформантов, выделение и очистку целевого продукта. Конструируют рекомбинантную плазмидную ДНК, содержащую промотор BARI Saccharomyces cerevisiae или промотор алкогольдегидрогеназы I S.cerevisiae, часть последовательности гена BARI, кодирующей сигнальный пептид, и место отщепления сигнального пептида КЕХ2 от гетерологичного полипептида, которое представляет собой Arg-Arg или Lys-Lys.
