Description
本發明提供一種具式[Zn(C6H14N2O2)2Cl]+Cl-之鋅-離胺酸錯合物,其具有抗菌及止汗性質,包含該錯合物之個人保健組成物,及製造與使用這些錯合物及組成物的方法。
習知的止汗劑是含有鋁或鋁/鋯鹽類。這些物質作為止汗劑是經由阻塞汗腺孔以阻斷汗液的流出。含有鋁或鋁/鋯鹽類之止汗劑組成物,經過一段時間後,這些鹽類容易呈現聚合現象,產生分子量從約500至約500,000克/莫耳之物質。一般來說,較低分子量之物質具有比高分子量物質為佳的止汗作用。無關任何理論依據,一般相信較小分子量的分子,具較快速且有效的阻塞汗腺孔,因此產生期望的止汗效果。維持相對的小分子量及避免過度聚合作用可增強止汗效果,及降低控制出汗所需止汗劑鹽類的用量。
腋下防臭劑可經由殺死產生臭味的細菌來控制味道。習知止汗劑鹽類在水溶液中呈現酸性,此性質使得它成為有效的殺菌劑,因此提供防臭的效果,但其也會造成皮膚的刺激。

腺孔,以阻斷出汗之適當尺寸、分子量之錯合物,提供除臭/抗菌效果,及較習知止汗劑酸性鹽類對皮膚產生較低的刺激。
本發明提供一種鋅-離胺酸錯合物,有時於本文中稱為ZLC,其由氧化鋅及離胺酸鹽酸鹽混合物中生成。ZLC化學結構是[Zn(C6H14N2O2)2Cl]+Cl-。這個鹽類具有重要的性質(如導電性、水解反應及蛋白質凝聚作用),使得其可與商售之止汗鹽類競爭。
如同現有的鋁或鋁-鋯止汗鹽類,ZLC在流汗情形下形成沉澱物,其可阻塞毛孔並阻斷汗液的流出。在蛋白質存在下,ZLC會凝聚及阻塞汗腺。當水的量增加,溶液變得更稀釋,其並非典型地如同離子錯合物之情形般進入或留存在溶液中,ZLC錯合物會水解,產生相對地不溶性氧化鋅沉澱物,藉此允許進一步阻塞毛孔及/或將氧化鋅控制的沉積在皮膚上。鋅是一種抗菌劑,除了提供一種沉澱物可阻斷汗液由毛孔流出之外,亦可經由減少產生臭味的細菌,提供一種除臭的功效。最後,ZLC可提供於接近中性pH值之調配物中,其較現行使用止汗鹽類或除臭調配物更接近皮膚的pH值。
本發明提供ZLC本身,也提供一將ZLC傳遞至皮膚之個人保健產品,及一種製造及使用ZLC的方法。在一個具體例中,本發明提供一種止汗或除臭組成物,其包含ZLC。由於ZLC之抗菌性質,本發明亦包括其他可施用至皮膚之個人保健組成物,如洗手皂或沐浴乳,其包含ZLC,如任何一個錯合物1及以下等,及/或其前驅物。本發明亦提供一種減少流汗的方法,其包括將組成物施用
於皮膚,及一種殺菌的方法,包括將組成物與細菌接觸。
本發明其他的應用領域,由本文的詳細說明變的顯而易知。可了解的是,本發明的詳細說明及特定具體例,其係顯示本發明較佳的具體例,其僅作為說明用途而不是限制本發明之範圍。
以下說明之較佳具體例,本質上僅是例示,而不是限制本發明之應用或使用。
在第一個具體例,本發明提供一個具分子式[Zn(C6H14N2O2)2Cl]+Cl-之鋅-離胺酸錯合物(有時本文稱為ZLC),例如
1.1 錯合物1,其中錯合物是由氧化鋅及離胺酸鹽酸鹽混合物中生成,如ZnO:離胺酸.HCl莫耳比1:1至1:3,如約1:2。
1.2 錯合物1,或1.1,呈結晶形式。

本發明更提供一種減少出汗的方法,其包括將有效止汗量之任一組成物2及以下等施用至皮膚;減少體味的方法,其包括將有效除臭量之任一組成物2及以下等施用至皮膚;及殺死細菌的方法,其包括將抗菌有效量之ZLC(如任一錯合物1及以下等)與細菌接觸,例如與任一組成物2及以下等接觸。
本發明更提供一種製造含有ZLC(如任一錯合物1及以下等)組成物的方法,其包括在水溶液中將氧化鋅及離胺酸鹽酸鹽組合,且任意地將混合物加入乙醇中,並分離所得之結晶沉澱物。
本發明更提供(i)使用ZLC(如任一錯合物1及以下等)殺死細菌,減少出汗,及/或減少體味;(ii)使用ZLC(如任一錯合物1及以下等)製造殺死細菌,減少出汗,及/或減少體味組成物;及(iii)用ZLC(如任一錯合物1及以下等)來殺死細菌,減少出汗,及/或減少體味。
可了解的,雖然ZLC可主要地以錯合物形式存在,其可能與氧化鋅及離胺酸鹽酸鹽前驅物質間有某程度的平衡,所以真正以錯合物形式及前驅物形式存在的比例,可視調配物確切條件、物質濃度、pH值、水存在與否、其他帶電分子存在與否等因素而異。
ZLC(如任一錯合物1及以下等)可與一適當之基料組合,如棒、滾珠、噴霧劑或氣溶膠以施用至腋下。施用後,ZLC於皮膚上在帶電分子,如蛋白質存在下,ZLC會凝聚產生阻塞以阻斷汗液流出。汗液中的水分更進一步稀釋調配物,造成錯合物分解,產生氧化鋅沉澱,其可減少前揭流汗及味道。類似地,如ZLC以洗手皂或沐浴乳形態提供,在沖洗過程ZLC稀釋,會在皮膚上產生一層薄的氧化鋅沉積,並提供抗菌效果。
本文所用止汗劑這個名詞,是有關可在汗腺孔形成阻塞,減少流汗的任何物質,或在食品藥物管理局CFR 21第350篇分類為止汗劑的任何物質。止汗劑亦可為除臭劑,尤其是在本發明,鋅具有抗菌性質,且可減少皮膚上造成味道的細菌。
組成物可包含ZLC,如任一錯合物1及以下等,及/或其前驅物,如氧化鋅及離胺酸鹽酸鹽。在一具體例中,ZLC是在室溫下,於水溶液中混合前驅物質而製備。在原位生成使其容易配製。前驅物質可被用來代替必須首先生成的ZLC。在另一個具體例,水容許ZLC生成,如任一錯合物1及以下等。此ZLC來自於汗液與組成物接觸後所生成之前驅物。
在特定具體例中,ZLC(如任一錯合物1及以下等)在本發明組成物(如任一組成物2及以下等)的含量,是0.05至40%,以組成物重量計。在特定具體例中,前驅物如氧化鋅及離胺酸鹽酸鹽,其存在之量在結合成ZLC(如任一錯合物1及以下等),是以0.05至10%,以組成物重量計。在這些具體例中,ZLC(如任一錯合物1及以下等)的量可依所期望的用途如抗菌劑或作為止汗劑而不同。在其他具體例,ZLC(如任一錯合物1及以下等)的量是至少0.1,至少0.2,
至少0.3,至少0.4,至少0.5,至少1,至少2,至少3,或至少4至高達10%,以組成物重量計。在其他具體例,ZLC(如任一錯合物1及以下等)的量是少於9,少於8,少於7,少於6,少於5,少於4,少於3,少於2,少於1,少於0.5至0.05%,以組成物重量計。在其他具體例,其量是0.05至5%,0.05至4%,0.05至3%,0.05至2%,0.1至5%,0.1至4%,0.1至3%,0.1至2%,0.5至5%,0.5至4%,0.5至3%或0.5至2%,以組成物重量計。
在一些具體例,組成物中鋅的總量是0.05至10%,以組成物重量計。在其他具體例,鋅的總量是至少0.1,至少0.2,至少0.3,至少0.4,至少0.5或至少1至高達8%,以組成物重量計。在其他具體例,組成物中鋅的總量是少於5,少於4,少於3,少於2,或少於1至0.05%,以組成物重量計。
在特定具體例,組成物是無水的。無水,是水含量少於5%,以重量計,任意地少於4,少於3,少於2,少於1,少於0.5%,少於0.1至0%,以重量計。
當以無水組成物提供時,前驅物如氧化鋅及離胺酸鹽酸鹽,不會顯著地反應生成ZLC,如任一錯合物1及以下等。當與足量的水接觸,其可為汗液形式,而後前驅物會反應生成ZLC,如任一錯合物1及以下等。ZLC,如任一錯合物1及以下等,當導入汗管會與蛋白質發生凝聚作用,及/或與水及/或汗水發生水解,產生沉澱物以阻塞汗管。
在特定具體例,ZLC,如任一錯合物1及以下等,具有導電度大於8,000,任意地大於9,000,大於10,000或大於12,000μS/cm。
組成物可為任亦形式之組成物。在特定具體例,組成物是期望含有抗菌劑,以施用至皮膚的任何組成物。這些組成物的實例是包括,但不限於,個人保健組成物、止汗劑、除臭劑、沐浴乳、沐浴膠、條狀肥皂、洗髮精、護髮乳、化妝品。
載劑係指組成物中除了ZLC(如任一錯合物1及以下等)或氧化鋅與胺基酸氫鹵酸鹽以外之所有物質。載劑的含量是,加入ZLC(如任一錯合物1及以下等),或氧化鋅與胺基酸氫鹵酸鹽後,達到100%重量。
對止汗/除臭組成物,載劑可為任何可用於止汗劑/除臭劑之載劑。載劑可為條狀、凝膠、滾珠,或氣溶膠之形式。對條狀調配物,載劑可包含油類及/或聚矽氧及凝膠劑。調配物的實例可參見US2011/0076309A1,其作為參考文獻併入本文中。
本發明組成物止汗/除臭調配物可包含任意成分,如溶劑;水溶性醇類,如C2-8醇類包括乙醇;二醇類,包括丙二醇、二丙烯甘醇、三伸丙二醇及其混合物;甘油酯類,包括單、雙及三甘油酯;中至長鏈有機酸、醇類及酯類;界面活性劑,包括乳化劑及分散劑;胺基酸類,包括甘胺酸;結構劑,包括增稠劑及凝膠劑,如聚合物、矽酸鹽及二氧化矽;潤滑劑;芳香劑;及著色劑(包括染料及色素)。如果需要,可包括除了ZLC(如任一錯合物1及以下等)外之止汗劑及/或除臭劑,可包括如氣味減低劑如硫沉澱劑,如葡萄酸銅、葡萄酸鋅、檸檬酸鋅等。
止汗組成物可配製成局部止汗及/或除臭調配物,以適合施用至皮膚,例示如條狀、凝膠、乳膏、滾珠、軟固體、粉末、液體、乳液、懸浮液、分散劑或噴霧劑。組成物可包括單相或可為
多相系統,如包括極性相與油相之系統,任意地以安定的乳液型式。組成物可為液體、半固體或固體。止汗及/或除臭調配物可以任何適當的容器提供,如氣溶膠罐、管或具多孔蓋子之容器、滾珠容器、瓶子、末端開口的容器等。
本組成物可施用至皮膚以減少出汗。在特定具體例,是施用至腋下。同時,可將組成物與細菌接觸以殺死細菌。例如,在一具體例,將胺基酸或胺基酸氫鹵酸鹽與氧化鋅結合,增加鋅離子的可用率,其可殺死細菌並減少流汗。
本發明提供(i)一種控制流汗的方法,其包含將本文所涵蓋或特別描述之任何具體例之調配物,如任一組成物1及以下等,以一止汗有效量施用至皮膚;及(ii)一種控制因流汗產生味道的方法,其包含將本文所涵蓋或特別描述之任何具體例之調配物,如任一組成物1及以下等,以一除臭有效量施用至皮膚。
除非特別說明,在本發明說明書所示組成物成分之所有百分比,是以整個組成物或調配物之100%重量計,除非其他特別確認,在本發明組成物及調配物所使用之成分,以化妝品級可接受的成分較佳。“化妝品級可接受的”係該調配物適合局部施用製人類皮膚。化妝品級可接受的賦形劑,例如,適合於外用之賦形劑,其是將本發明調配物以預計之用量及濃度外用,,及包括如美國食品藥物管理局”一般認可為安全”之賦形劑。
在本發明所描述或主張申請之組成物或調配物,有關其成分,是在此技術中所常用的。對習於此技術者是顯而易知的,在一些實例中,各成分可能會與其他成分反應,所以真正組成物其最後調配物,可能與所列出成分不完全一致。因此,要了解的是本
發明擴展至所列成分組合後之產品。
由詳細說明及所附圖示使得本發明更充分了解,其中圖1描述實驗室合成ZLC粉末之X光粉末繞射圖(PXRD)(頂圖)及ZLC單晶之X光粉末繞射圖(底圖)。
ZnO+2(Lysine.HCl)-›[Zn(Lysine)2Cl]+Cl-.2H2O(ZLC)
莫耳比2:1之ZnO:離胺酸.HCl懸浮液在室溫下攪拌約12小時。將混合液離心,取1毫升上清液移轉入NMR管。而後NMR管置入一封閉試管,以乙醇充滿讓結晶成長。一週後生成一些無色立體結晶。ZLC結晶之晶體結構以單晶X-光繞射測定。ZLC具有分子量464.44克/莫耳,如式C12H32N4O6Cl2Zn之實驗式。在這個錯合物,Zn陽離子在赤道面被二個離胺酸配位基(其具有來自NH2基之二個氮原子及來自羧基的氧原子)配位。此結構其頂點被氯原子佔據,呈現扭曲的方形錐體幾何結構。此結構產生一個正陽離子部分,並與氯陰離子結合形成離子性的鹽類。
實驗室擴大合成純的ZLC粉末:2莫耳離胺酸HCl溶於1000毫升去離子水,在室溫下攪拌,將1莫耳固體ZnO緩慢加入離胺酸HCl溶液並攪拌,在室溫下持續攪拌過夜(約12小時)。懸浮溶液在高速下離心15分鐘,將上清液緩慢到入乙醇中,立即生成沉澱。得
到1克粉末約需5-8毫升乙醇。將含有粉末之乙醇溶劑過濾,可得到灰白色的粉末。將粉末置於50℃之烘箱乾燥,可得到產率88%之產物。利用PXRD確認ZLC粉末的純度與ZLC結晶比較(圖1)。在這個實施例,利用乙醇作為反溶劑將ZLC結晶。可使用任何反溶劑,任意地,溶液可噴霧乾燥。
水解反應:製備185毫克/毫升ZLC溶液,而後稀釋數倍,在37℃之烘箱熟化5小時,以進行混濁度研究。當溶液稀釋時生成白色沉澱,使用濁度計測量溶液混濁度,結果以濁度單位(NTU)計。表一顯示熟化前後,pH與混濁度之比較,其顯示稀釋及熟化會增加混濁度。
以離心方式收集經8倍、16倍及32倍稀釋溶液生成之沉澱物,經PXRD確認為ZnO晶體。在上清液,長成之單晶以X光繞射顯示是離胺酸單鹽酸鹽2水合物(離胺酸.HCl.2H2O)。這些結果顯示ZLC錯合物在稀釋過程中會分離,而後產生氧化鋅沉澱。

在一個腋下用產品,ZnO+離胺酸HCl混合物,在汗液存在下會生成ZLC,其可進入汗管並生成ZnO塞子。
凝聚作用:另一個ZLC阻斷汗液流出的機制,是有關在蛋白質存在下ZLC的凝聚作用。在這個研究中以牛血清白蛋白(BSA)作為蛋白質。製備不同pH值之對照溶液(去離子水)及3個1%BSA水溶液,如表2所示。
將ZLC粉末加入前述樣品,以研究ZLC與BSA間之交互作用,並測定ZLC是否具收斂性質,例如,是否可生成沉澱以作為止汗劑。將混合液置入37℃之烘箱5小時後,測定溶液之混濁度及pH值,結果如表3所示。
因此,在汗管(pH=5-7)ZLC會水解成不溶的ZnO,物理性的阻斷汗管。除此,ZLC在汗液中,也具有凝聚蛋白質(如BSA)的能力,促進在汗管形成”塞子”。
