1274021 九、發明說明: 【發明所屬之技術領域】 本發明係關於包含塑膠材料之無機添加物容器 之用途。 【先前技術】 塑膠容器廣泛地用於藥品包裝,然而自已習知 之技術得知塑膠容器之特性會造成某些應用上之 限制。特別係由聚乙烯或聚丙烯以及其共聚物/混合 # 物所製成之容器,為了抑制此類容器對某些特定化 學藥劑之反應,數種方法於是乎被應用於:避免使 用會增加鏈狀分子之能動性的塑化劑、使用較高密 度聚合物或聚烯混合物(如聚丙烯/聚丙烯酸酯)、 增加外壁厚度、將容器包起(如鋁箔)或封住(如 氟化處理、矽橡膠)。
美國專利公告第4,123,417號(Finberg,1978) 主張藉由具有非晶體乙烯一丙烯共聚物(有一定程 度之結晶性且有特定之乙烯成分)之低密度聚乙烯 混合物,將可提升低密度聚乙烯之強度。 美國專利公告第 4,546,882 號(1^1161&1.,1985) 主張一用於含油產品之多層包裝,其中包含由尼龍 或乙烯-乙烯醇共聚物所製成之隔油層。 4 1274021 美國專利公告第 5,5 0 0,2 6 1 號 (T a k e i e t al.,1996) 主張一抗油容器包含具有特定玻璃轉移 溫度(glass transition temperature )之混合樹月旨合 成物。
美國專利公告第 6,800,363號(Suetal.,2004) 主張一種存在於食用油中不會扭曲之薄膜,係利用 一聚烯多層薄膜,上述薄膜具有吸油孔狀粒子(碳 酸鈣、二氧化矽、非晶體矽、矽酸鋁鈉、活性碳) 所形成之表層以及金屬處理層。 美國專利公告第 6,815,506 號 (Takashima et a 1.,2 0 0 4 )主張一抗油熱塑性彈性化合物,其中包 含丙烯樹脂、不飽和族一壓克力樹脂以及用於樹脂 化合物之無機填充物(最好為二氧化矽)。
此外還有其他添加物可用以增強塑膠之性 質,主要係色素與紫外線穩定劑(有機與無機色 素、染料、二苯曱酮、受阻胺等)。如此可滿足某 些需求,如高溫穩定性、不會因光線和天氣而褪 色,其中最普遍之藥物係二氧化鈦。二氧化鈦係一 惰性物質,具由於其吸收紫外線與非活性(不與藥 物配方相互作用)之特性而廣為悉知。 5 1274021 【發明内容】 本發明之目的係提供一容器之用途,上述容器 係由一包含塑膠材料之無機添加物所製成,用以儲 存含油質、脂質和/或蠟質之配方。 為達到本發明之目的,藉由提出一容器之用 途,上述容器係由一包含塑勝材料之無機添加物所 製成,上述容器用以降低此容器與裝於此容器内之 φ 含油質、脂質和/或蠟質配方間之物理/化學作用。 其中所述之物理/化學作用係指上述塑膠材料 吸附 (a d s 〇 r p t i ο η ) 該S己方,且其中所述之無機添 加物包含至少一色素(pigment),此至少一色素可為 二氧化鈦(T i Ο 2 )、表面處理之二氧化鈦或兩者之混 合物。
根據本發明之較佳實施例,其中所述之添加物 存在於塑膠材料中,該添加物佔介於總重量(以塑 膠材料之總重量而言)之0 . 1 %至1 0 %間;更佳地, 該添加物佔介於總重量之0 . 1 %至5 %間;最佳地, 該添加物約佔總重量之2% 。 上述塑膠材料包含聚烯(polyolefin),此外前 述之聚烯可選自一群組包含 聚乙烯 6 1274021 (polyethylene )、聚丙稀(polypropylene )、 乙烯 (ethylene) 與丙婦 (propylene) 之共聚合物或上 述各項之混合物。更佳地,此塑膠材料包含低密度 聚乙烯 (L D P E )。並且,此塑膠材料適用於吹出成 形(extrusion blow molding)0
其中所述之配方包含至少一類固醇賀爾蒙 (s t e r 〇 i d h 〇 r m ο n e ) 溶解或懸浮於油、脂和/或壤 中。且上述之類固醇賀爾蒙係一性賀爾蒙藥劑 (sexual hormone drug),為睪固酮(testosterone ) 更佳,以及該配方更包含至少一親脂性(1 i p o p h i 1 i c ) 或部分親脂性媒介物;以及具有表面張力可降低活 性之化合物或化合物之混合物,且具有足夠數量使 該配方接觸水後而產生乳劑之原處生成(in situ generation)。前述之配方適用於鼻腔的應用(nasal application),特別適用哺乳類動物(m a m m a 1 i a η )。 較佳的低密度聚乙烯舉例如 L u ρ ο 1 e η⑧1840 Η , 此外較佳之配方可為歐洲專利公開第 0 3 0 2 5 7 6 9.5號内所揭露之任一種配方。 出人意外地,上述包含塑膠材料之無機添加物 容器對於儲存含油質、脂質和/或蠟質之配方有極佳 之用途,舉例如含油之類固醇賀爾蒙配方,使用上 7 1274021

在鼻腔治療方面有兩種主要之包裝材料與包 裝型態:玻璃與塑膠,以及重複給藥(multiple-dose) 與單一劑量(u n i t - d 〇 s e )。塑膠材料其優點主要係 其靈活度,可提供廣泛的設計、重量輕、不易碎、 易拿取等優點,故塑膠材料適用於單一劑量之鼻腔 治療藥物,特別如不需要唧筒裝置,也不需額外對 產品配方保護。
基本上此類塑膠容器基本材料係聚乙烯、聚丙 稀以及兩者之聚合物,而此類容器可能之缺點如透 氧率、較差之抗紫外線,由於其非極性之特性,以 及結晶性與摩爾質量之度數,與對特定化學物質較 差之抗性。 聚乙烯與聚丙烯通常對脂肪族碳氫化合物、芳 香族碳氫化合物以及其鹵素衍生物(某些低揮發性 物質,如脂肪、油以及蠟)不具有抗性。關於此不 相容性之問釋,可由塑膠吸收化學物質、化學物質 造成塑膠擴散與鼓起或甚至化學物質分解塑膠等 狀況中看出。 9 1274021
另一方面,碳氫化合物之衍生物(如類固醇賀 爾蒙)廣泛地運用油類當其媒介物,以增加其溶解 度及有效時間。為了避免主要包裝材料造成上述穩 定性問題,這類含油配方(大多為血管注射劑)通 常裝入玻璃瓶中,但此類包裝方式並不適用於所有 的治療方式(如需要用到含油配方之鼻腔治療)。 在考量到重複給藥裝置之狀況,雖然藥瓶本身係玻 璃,但上述裝置總有部件係塑膠製成(如唧筒)。 而在單一劑量裝置方面,考量某些黏性配方必須以 擠壓的方式使用,這類配方大多利用塑膠成形以吹 灌封 (blow-fill-seal) 三合一技術包裝,也不適用 於玻璃包裝。 考量前述因素,測試含類固醇賀爾蒙睪固酮之 配方於不同材質容器中之穩定性,測試結果如下列

Claims (1)
1274021 十、申請專利範圍: 1. 一容器之用途,該容器係由一包含塑膠材料之無 機添加物所製成,該容器用以降低該容器與裝於 該容器内之油質、脂質和/或蠟質配方間之物理/ 化學作用。
