Abstract
1. Устройство для доставки конструкции киРНК через дермальный барьер, содержащеемикроиглу и множество наноструктур, изготовленных на поверхности микроиглы, где наноструктуры образуют заранее определенный узор;и конструкцию киРНК, сообщающуюся по потоку текучей среды с микроиглой.2. Устройство по п.1, в котором узор дополнительно включает микроструктуры, причем наноструктуры обладают меньшим размером в сечении, чем микроструктуры.3. Устройство по п.2, дополнительно содержащее вторые наноструктуры с размером в сечении меньше размера в сечении микроструктур и больше размера в сечении первых наноструктур.4. Медицинское устройство по п.1 или 2, в котором по меньшей мере участок наноструктур обладает размером в сечении меньше примерно 500 и больше примерно 5 нанометров.5. Медицинское устройство по п.1 или 2, в котором по меньшей мере участок наноструктур обладает промежутком между центрами от примерно 50 нанометров до примерно 1 микрометра.6. Медицинское устройство по п.1 или 2, в котором по меньшей мере участок наноструктур обладает высотой от примерно 10 нанометров до примерно 20 микрометров.7. Медицинское устройство по п.1 или 2, в котором по меньшей мере на некотором участке наноструктуры обладают отношением размеров от примерно 0,15 до примерно 30.8. Claims (23)
1. Устройство для доставки конструкции киРНК через дермальный барьер, содержащее
микроиглу и множество наноструктур, изготовленных на поверхности микроиглы, где наноструктуры образуют заранее определенный узор;
и конструкцию киРНК, сообщающуюся по потоку текучей среды с микроиглой.
2. Устройство по п.1, в котором узор дополнительно включает микроструктуры, причем наноструктуры обладают меньшим размером в сечении, чем микроструктуры.
3. Устройство по п.2, дополнительно содержащее вторые наноструктуры с размером в сечении меньше размера в сечении микроструктур и больше размера в сечении первых наноструктур.
4. Медицинское устройство по п.1 или 2, в котором по меньшей мере участок наноструктур обладает размером в сечении меньше примерно 500 и больше примерно 5 нанометров.
5. Медицинское устройство по п.1 или 2, в котором по меньшей мере участок наноструктур обладает промежутком между центрами от примерно 50 нанометров до примерно 1 микрометра.
6. Медицинское устройство по п.1 или 2, в котором по меньшей мере участок наноструктур обладает высотой от примерно 10 нанометров до примерно 20 микрометров.
7. Медицинское устройство по п.1 или 2, в котором по меньшей мере на некотором участке наноструктуры обладают отношением размеров от примерно 0,15 до примерно 30.

10. Устройство по п.1 или 2, киРНК-препарат содержит дополнительные нуклеотиды или аналоги нуклеотидов.
11. Устройство по п.1 или 2, в котором киРНК-препарат содержит нуклеотидную последовательность, идентичную участку гена-мишени.
12. Устройство по п.1 или 2, в котором киРНК-препарат содержит вставки, делеции или одноточечные мутации по сравнению с участком гена-мишени.
13. Устройство по п.1 или 2, в котором киРНК-препарат представляет собой одноцепочечную киРНК.
14. Устройство по п.1 или 2, в котором киРНК содержит лиганд, связанный с киРНК, например модификатор терапевтического средства, диагностическое соединение, "репортерную" группу, перекрестно-сшитый препарат, нуклеотидное основание, липофильную молекулу или белок.
15. Устройство по п.1 или 2, в котором киРНК-препарат внедрен в носитель для доставки, например, в липосому.
16. Устройство по п.1 или 2, в котором конструкция киРНК содержит вектор, например вирусный вектор.
17. Способ доставки конструкции киРНК через дермальный барьер, где способ включает:
проникновение через роговой слой микроиглы, микроигла содержит множество наноструктур, сформированных на поверхности микроиглы и скомпонованных в узор, конструкция киРНК сообщается по потоку текучей среды с микроиглой, конструкция киРНК транспортируется через роговой слой после проникновения в роговой слой микроиглы.
18. Способ по п.17, в котором конструкция киРНК транспортируется через роговой слой внутри круглого канала или канала микроиглы.
19. Способ по п.17 или 18, в котором конструкция киРНК транспортируется через роговой слой при стабильной концентрации в течение некоторого периода времени.
20. Способ формирования устройства для доставки конструкции РНК через дермальный барьер, где способ включает:
изготовление массива микроигл;
изготовление узора из наноструктур на поверхности по меньшей мере одной из микроигл;
присоединение конструкции киРНК к микроиглам, чтобы конструкция киРНК сообщалась по потоку текучей среды с микроиглами.
21. Способ по п.20, способ дополнительно включает удержание конструкции киРНК внутри емкости, которая сообщается по потоку текучей среды с массивом микроигл.
22. Способ по п.21, в котором емкость присоединена к подложке, на которой расположен массив микроигл.
