Description
Denne oppfinnelse angår en ny fremgangsmåte for fremstilling av derivater av 9-(1,3-dihydroksy-2-propoksymetyl)-guanin.
9-(1,3-dihydroksy-2-propoksymetyl)-guanin (i det nedenstående betegnet DHPG) og dens estere er sterke anti-virusmidler og er blitt fremstilt etter fremgangsmåter beskrevet i US patentskrift nr. 4.355.032 og europeiske patentsøkna-der nr. 49.072, 72.027 og 74.306. Lignende forbindelser med lignende sidekjeder er beskrevet i US patentskrifter nr. 4.347.360 og 4.199.574. Den foreliggende oppfinnelse angår en ny og forbedret fremgangsmåte for fremstilling av DHPG og visse eter- og estermellomprodukter for dennes fremstilling, hvilken fremgangsmåte har færre trinn enn de for tiden benyttede fremgangsmåter og gir færre uønskede biprodukter.
Mellomprodukter for fremstilling av DHPG og dens etere og estere er tidligere blitt fremstilt ved omsetning av epiklorhydrin med benzylalkohol for dannelse av en symmetrisk substituert 1,3-dibenzylglyserol, som deretter er blitt klor-metylert. Klormetylforbindelsen er så blitt overført til acetat- eller formiatesteren, som deretter er blitt omsatt med guanin og derved addert til guaninets 9-stilling, se US patentskrift nr. 4.355.032.
I US patentskrift nr. 4.347.360 er det beskrevet en alternativ syntese, i henhold til hvilken 1,3-diklor-2-propa-nol omsettes med natriumbenzylat, hvorved klormetoksyderivatet dannes, hvilket omsettes videre med en purinbase.
En tredje fremgangsmåte er beskrevet i europeisk patentsøknad nr. 74.306, i henhold til hvilken sidekjeden fremstilles ved at det startes med glyserolformal, en blanding av 1,3-dioksan-5-ol og 1,3-dioksolan-4-metanol. Glyserolfor-malet acyleres, og blandingen separeres. Den acylerte forbindelse omsettes så med eddiksyreanhydrid i nærvær av ZnCl2, hvorved det fås en forbindelse som kan omsettes med et purin.
Disse fremgangsmåter er alle flertrinnssynteser som medfører kompliserte separasjons- og renseteknikker og ikke gjør bruk av de mellomprodukter som anvendes ved den foreliggende nye fremgangsmåte.
Det ville være en fordel å ha til rådighet en fremgangsmåte som er enkel (krever færre trinn), gjør bruk av let-tilgjengelige utgangsmaterialer, ikke gir uønskede isomerer og tillater enkel substituering av de som sluttprodukter oppnådde estere. Den nye fremgangsmåte medfører alle disse fordeler.
Spesielt kan DHPG og en rekke 1,3-di-estere og 1,3-di-etere av DHPG lett fremstilles ved hjelp av den nye fremgangsmåte.
Med oppfinnelsen tilveiebringes det således en fremgangsmåte for fremstilling av derivater av 9-(1,3-dihydroksy-2-propoksymetyl)-guanin med den generelle formel I:
hvor R<1> og R3 er valgt blant hydrogen, C1.4-alkyl, C4_20-alkyl-C0-, fenyl-CO-, 1-adamantanoyl, fenyl eller fenyl-C^-alkyl, hvor alle alkyl- og fenylgrupper eventuelt kan være substituert med halogenatomer, C^-alkylgrupper og fenylgrupper, og R<5> er hydrogen, C4.20-alkyl-CO- eller fenyl-CO-, hvor alkylgrup-pen, henholdsvis fenylgruppen kan være substituert med halogenatomer, C1.4-alkylgrupper og fenylgrupper. Fremgangsmåten er særpreget ved at : (a) en guaninforbindelse med den generelle formel II:
hvor R5 er som angitt for R<5> ovenfor eller er (alkyl )3Si, omsettes, eventuelt i nærvær av et heksa-C1.4-alkyl-disilazan, med en forbindelse med den generelle formel A:
hvor R1 og R<3> er som ovenfor angitt, og R<4> er <C>1.4-alkyl som eventuelt kan være substituert med halogenatomer, C1_4-alkylgrupper og fenylgrupper, hvoretter eventuelt (b) den resulterende blanding behandles med en Cx_4-alkylalkohol og/eller (c) den resulterende blanding - dersom R<1> og R3 ikke er hydrogen - behandles med ammoniumhydroksid og vann.
Slik de her benyttes, har de følgende betegnelser og uttrykk de nedenfor angitte betydninger, med mindre annet er angitt.
Med uttrykket "eventuelt substituert" menes substituering av en alkyl- eller fenylgruppe med halogenatomer, C^-alkylgrupper og fenylgrupper.
Uttrykket "eventuelt substituert guanin" refererer seg til et guaninmolekyl som eventuelt er substituert med acyl- og/eller (alkyl )3Si-grupper på aminogruppen i 2-stillingen og på nitrogenet i 9-stillingen.
"Acyl" er her definert som eventuelt substituert alkyl-C(O) med 4-20 karbonatomer i alkylkjeden, eventuelt substituert fenyl-C(O), eller eventuelt substituert alkylfenyl-C(0) hvor den eventuelle substitusjon er på fenylringen. Eksempler er n-heksanoyl, n-heptanoyl, palmitoyl, sterakanoyl, arachidoyl, pivaloyl, 1-metyl-l-cykloheksan-karboksoyl, 2-ok-tyl-dekanoyl, benzoyl, p-klorbenzoyl, toluoyl, fenylacetyl, 3-fenylpropanoyl og 4-fenylbutanoyl.
"Pivalinsyre" er 2,2-dimetylpropionsyre.
"1-adamantanoyl" betyr resten med den nedenfor angitte struktur.
Med "hydrogendonator" menes enhver substans som kan tjene som en kilde for aktivt hydrogen. Noen eksempler på hydrogendonatorer er hydrogengass, cykloheksen, 1,4-cyklohek-sadien o.l. I noen tilfeller kan katalysatorer brukes i forbindelse med hydrogendonatorer, f.eks. edelmetallene, som f.eks. platina, palladium og rhodium, brukt som finfordelt metall eller som metall båret på en hvilken som helst vanlig katalysatorbærer, f.eks. karbon, aluminiumoksyd eller sili-siumoksyd.
Med "Lewis-syre" menes enhver syre som ikke har et uttrekkbart proton. Eksempler er Be(CH3)2, BF3, B(0-alkyl)3, A1(CH3), A1(C1)3, Fe<3+>, Mg<2>+, Ca<2+>, Cu<+>, TiCl4, Hg<2+>, C02+, Fe2+, Zn<2+>, o.l. En omfattende liste over Lewis-syrer kan finnes i Chem. Rev., 1 (1975) på side 2.
Med betegnelsen "protisk syre" menes alle Brønsted-syrer med en pKa lavere ann 2,0, fortrinnsvis lavere enn 1,0, som har et uttrekkbart proton. Eksempler på protiske syrer er mineralsyrer, som f.eks. salpetersyre, svovelsyre, fosforsyre, og hydrogenhalogenider som f.eks. saltsyre; organiske syrer, som f.eks. trifluoreddiksyre; organiske sulfonsyrer som f.eks. fenylsulfonsyre og, spesielt, para-toluensulfonsyre; samt sure harpikser som f.eks. "Amberlyst" (Rohm and Haas).
Med betegnelsen "beskyttende grupper" menes alle grupper som er i stand til å beskytte funksjonelle grupper i en forbindelse og lar seg fjerne ved hydrolyse eller hydroge-nering. Eksempler på beskyttende grupper som er anvendelige ved fremgangsmåten ifølge oppfinnelsen, er sterisk hindrede grupper (hvor "sterisk hindret" refererer seg til en spesiell atomgruppe som hindrer eller hemmer en forventet kjemisk reaksjon pga. sitt volum), benzyl eller eventuelt substituert benzyl, eller alkylgrupper, og sterisk hindrede silylgrupper med den generelle formel R3Si(R2)-.
Fremgangsmåten ifølge oppfinnelsen er vist i Reak-sjonsskjemaer I og II.

Forbindelsen med formel A kan overføres til en 2-halogenalkyl-l,3-diacylglyserol ved bruk av HX hvor X er et fluor-, klor-, brom- eller jodatom i metylenklorid.
Beskyttet guanin med formel H fremstilt f.eks. etter metodene til Tshido et al., Bull. Chem. Soc. Japan, 37, 1389
(1956) omsettes med en forbindelse med formel A. R<5> i formel A er acyl eller hydrogen, og dersom den er acyl, danner denne gruppe sammen med guaninets nitrogen, som R<5> beskytter, fortrinnsvis et monoamid av en lavere alkansyre, f.eks. acetamid eller propionsyreamid. Reaksjonen utføres ved høyere temperatur under vakuum, i nærvær av en syrekatalysator, f.eks. para-toluensulfonsyre. DHPG-derivatet med formel J dannes derved, hvor R<1> og R3 er som ovenfor angitt. Dersom ester- eller eter-derivatet er direkte anvendelig, f.eks. dipivaloyl- DHPG eller
di-adamantanoyl-DHPG, er ingen ytterligere reaksjoner nødven-dig, dersom R<5> er hydrogen. Dersom R5 er acyl, kan esteren eller eteren av DHPG fremstilles ved å omsette forbindelsen med formel J med f.eks. metanolisk ammoniakk. Dersom DHPG er ønsket, kan formel J avbeskyttes (fjerning av R<1>, R3 og R<5>) under dannelse av DHPG ved å omsette forbindelsen med en blanding av ammoniumhydroksid og vann.
I reaksjonsskjemaet er symbolene som ovenfor angitt.
Alternativt kan forbindelsen med formel A omsettes direkte med et trialkylsilylbeskyttet guanin til DHPG-esteren eller -eteren. Guanin eller et eventuelt substituert guaninde-rivat oppvarmes i et aprotisk hydrokarbonløsningsmiddel, f.eks. toluen eller xylener o.l., med mer enn 3 molekviva-lenter heksa-C1.4-alkyl-disilazan, f.eks. heksametyldisilazan, heksaetyldisilazan o.l., og en katalysator. Katalysatoren vel-ges fra gruppen trialkylsilylsulfater, klorsilaner, ammoniumsulfat og pyridin. Et volum av flyktige bestanddeler lik volu-met av hydrokarbonoppløsningsmiddel fjernes ved vakuumdestil-lasjon fra den resulterende løsning. Så tilsettes forbindelsen med formel A, og blandingen oppvarmes ved en høyere temperatur enn omgivelsestemperaturen, inntil TLC-analyse av blandingen viser et ønsket forbruk av forbindelsen med formel A og dannelse av DHPG-esteren eller -eteren (formel J). Behandling av reaksjonsblandingen på dette tidspunkt for å fjerne alle trimetylsilylgrupper, f.eks. ved å oppvarme med lavere alkyl-alkoholer, f.eks. metanol, etanol eller isopropanol, gir DHPG-esteren eller -eteren, hvilken kan renses etter standardmeto-der. Dersom R<5> er acyl, kan R<5> fjernes med f.eks. metanolisk ammoniakk og gi DHPG-esteren eller -eteren. Dersom DHPG ønskes, kan forbindelsen med formel J avbeskyttes (R<1>, R<3> og R<5>) ved omsetning med en blanding av ammoniumhydroksid og vann.
Forbindelsene med den generelle formel A, som anvendes som utgangsmaterialer ved den foreliggende fremgangsmåte, er nye forbindelser. Fremstillingen av disse er vist nedenfor i Reaksjonsskjerna III. I reaksjonsskjemaet er R1, R<3>, R<4> og R<6> som ovenfor angitt, mens R7 er R<4>C(0)OCH2 eller R6OCH2, hvor R4 og R<6> er som ovenfor angitt.
I reaksjonstrinn A omsettes et dialkyloksymetan med formel F med en anhydrid med formel G i nærvær av en protisk syre som katalysator. De to reaktanter foreligger i et molfor-hold på ca. 1:1, i nærvær av en katalytisk mengde av en protisk syre, f.eks. en organisk sulfonsyre, f.eks. para-toluensulfonsyre, eller en katalytisk mengde av en mineralsyre, som f.eks. saltsyre eller svovelsyre. Reaksjonen kan utføres i et aprotisk hydrokarbonoppløsningsmiddel, som f.eks. benzen, eller uten oppløsningsmiddel, vanligvis ved oppløsningens til-bakeløpstemperatur. Oppløsningen omsettes i 1-12 timer, idet ca. 6 timer vil være en typisk reaksjonstid. Se fremgangsmåten ifølge J. Rm. Chem. Soc, 76, side 5161 (1954).
Det foretrekkes at anhydridet og dialkyloksymetanet er symmetriske, fordi antallet mulige produkter reduseres sterkt når symmetriske reagenser benyttes. Assymetriske anhydrider eller asymmetriske alkyloksymetaner kan imidlertid selvfølgelig også omsettes med hverandre. I visse tilfeller vil asymmetriske anhydrider selektivt gi ett av de mulige pro dukter.
1,3-dialkanoylglyserolen med formel E kan fremstilles på én av to måter. Behandling av glyserol med litt mer enn 2 ekvivalenter av et syreklorid eller et syreanhydrid i apro-tiske oppløsningsmidler i nærvær av organiske baser, f.eks. trialkylaminer, og en nukleofil katalysator, f.eks. pyridin, gir 1,3-dialkanoylglyserolen med formel E.
Alternativt er disse forbindelser tilgjengelige fra dihydroksyaceton ved bruk av fremgangsmåten ifølge J. Org. Chem., 35, 2082, (1970). Behandling av dihydroksyaceton i pyridin med en ekvivalent mengde av det tilsvarende fettsyre-klorid, etterfulgt av reduksjon av den sentrale ketogruppe med borhydrid i tetrahydrofuran gir 1,3-dialkanoylglyserolen med formel E.
I Reaksjonstrinn B omsettes forbindelsen med formel D med forbindelsen med formel E i nærvær av en protisk syre som katalysator, f.eks. en organisk sulfonsyre som para-toluensulfonsyre, eller en katalytisk mengde av en mineralsyre, f-eks. saltsyre eller svovelsyre, hvorved det fås en forbindelse med formel C. Forbindelsene med formel D og formel E blandes, uten bruk av oppløsningsmiddel, med en katalytisk mengde av en protisk syre som katalysator. Det foretrekkes å bruke et overskudd av forbindelsen med formel D, idet overskuddet virker som et oppløsningsmiddel for reaksjonen. Reaktantene kan blandes sammen uten at det er nødvendig å tilføre varme utenfra, selv om varmetilførsel vil påskynde reaksjonen. Reaksjonen i reaksj onstr inn B kan være eksoterm, og når temperaturen faller til omgivelsestemperaturen, er reaksjonen av-sluttet. Reaksjonen tar mellom 1 og 6 timer, og den er gjerne ferdig etter ca. 2 timer.
Forbindelsen med formel C vil variere avhengig av ut-gangsmaterialene. Reaksjonstrinn C kan utelates dersom produk-tet fra reaksjonstrinn B er en forbindelse med formel A. Ellers omsettes forbindelsene videre i reaksjonstrinn C.
I reaksjonstrinn C omsettes blandingen av forbindelser med formel C videre i nærvær av et alkylanhydrid og en Lewis-syre som katalysator. Det foretrekkes at anhydridet som benyttes i denne reaksjon er et symmetrisk anhydrid for å re-dusere antallet mulige produkter, selv om dette ikke er et absolutt krav til reaksjonen.
Den katalyserende syre er fortrinnsvis en ikke-protisk Lewis-syre, f.eks. bortrifluorid eller aluminiumtri-klorid. Reaksjonstemperaturen øker fra omgivelsestemperaturen til ca. 50°C, hvoretter man begynner å regulere temperaturen. Reaksjonstemperaturen holdes vanligvis mellom 45 og 65°C, fortrinnsvis mellom 50 og 60°C.
Reaksjonsforløpet følges gjerne ved tynnsjiktkro-matografi (TLC) for å få en indikasjon om hvor langt reaksjonen har gått. Etter mellom 60 og 180 minutter vil reaksjonen normalt være fullført. Reaksjonsblandingen ekstraheres med et organisk oppløsningsmiddel som er blitt vasket med natriumkar bonat og vann. Det organiske sjikt blir deretter tørret, og forbindelsen med formel A fås ved avdampning av oppløsnings-midlet.
Nedenfor beskrives i eksempler I-VI fremstillingen av noen mellomprodukter.
Eksempel I
1, 3- di-( 1- adamantanoyl)- glyserol.
I en kolbe under nitrogen oppløses 1,053 g glyserol i 4 ml pyridin og 6 ml metylenklorid, og oppløsningen kjøles til ca. -10°C. Deretter tilsettes 5,00 g (1-adamantanoyl)-klorid (Aldrich Chemical Co.) i én porsjon, og blandingen omrøres i ytterligere ca. 1/2 time under kjøling og deretter i ytterligere to timer ved romtemperatur. Det resulterende faste stoff frafiltreres og vaskes med metylenklorid. De sammen-slåtte organiske sjikt vaskes to ganger med fortynnet vandig HC1 og deretter én gang med vann og tørres over magnesiumsulfat. 1,3-di-(1-adamantanoyl)-glyserol isoleres som en olje. Utbytte 69%.

1,3-di-alkylglyserolene kan fremstilles etter fremgangsmåten beskrevet i US patentskrift nr. 4.355.032.
Eksempel II
1, 3- di- isopropyl- 2- propanoyloksymetylglyserol.
154 g 1,3-di-isopropylglyserol, 616 ml metoksymetyl-propionat og 9,98 g para-toluensulfonsyre ble blandet i en 2-liters rundkolbe utstyrt med magnetrører og et innvendig montert termometer. Temperaturen steg 6°C i løpet av 10 min. Temperaturøkningen ble etterfulgt av et jevnt temperaturfall tilbake til romtemperatur. En TLC-kontroll (25% etylacetat/75% heksan, rf = 0,69, kiselgel) etter 45 min reaksjon viste et nesten fullstendig forbruk av den benyttede alkohol.
Reaksjonsblandingen ble tilført til en skilletrakt med 500 ml heksan som en organisk fase, etterfulgt av vask av det organiske sjikt med 500 ml vann og to gangers vask med 500 ml mettet vandig natriumbikarbonat og en sluttvask med 500 ml vann. Det organiske sjikt ble så tørret over vannfritt magnesiumsulfat og inndampet i en roterende inndamper. 1,3-di-isopropyl-2-propanoyloksymetylglyserol ble isolert.

Eksempel III
1, 3- dipropanovl- 2- propanovloksymetvlglvserol.
339 g propionsyreanhydrid og 185 g av den urene olje som ble fremstilt i eksempel II (1,3-di-isopropyl-2-propanoyloksymetylglyserol), ble tilsatt til en 2 liters, 3-halset rundkolbe som var utstyrt med magnetrører, tilbakeløpskjøler, innvendig montert termometer og nitrogeninntak, og som var rigget til slik at man kunne varme eller kjøle raskt. 17,9 g bortrifluorideterat ble så tilsatt i én porsjon. Reaksjonstemperaturen steg øyeblikkelig fra ca. 20°C (omgivelsestempera-tur) til ca. 50°C, hvoretter man kjølte. Reaksjonstemperaturen ble omhyggelig holdt mellom 50°C og 60°C ved oppvarming eller kjøling etter behov. Reaksjonsforløpet ble fulgt ved TLC-kontroll (25% etylacetat/75% heksan, rf = 0,31, kiselgel) av reaksjonsblandingen.
Etter 100 min ble reaksjonsblandingen tilsatt til en skilletrakt med 500 ml toluen. Det organiske sjikt ble vasket med 500 ml mettet vandig natriumkarbonat og to ganger med 500 ml vann. Det organiske sjikt ble så tørret over vannfritt natriumsulfat, filtrert og inndampet i en roterende inndamper under et vakuum på 66 cm Hg. En uren svart olje ble oppnådd og deretter renset ved to gjennomløp gjennom en tynnfilmdestil-lasjonsapparatur. Den rensede olje var 1,3-dipropanoyl-2-propanoyloksymetylglyserol.

Eksempel IV
1, 3- dibenzyl- 2- acetoksymetylglvserol.
75 g di-benzylglyserol, 5,25 g paratoluensulfonsyre, 300 ml heksan og 300 ml metoksymetylacetat ble tilsatt til en 2 liters rundkolbe med magnetrøring, innvendig termometer og tørkerør. Reaksjonsblandingen ble holdt ved 60-65°C i 90 min, hvoretter det ble foretatt TLC-analyse (20% etylacetat/80%
heksan, rf = 0,31, kiselgel) etterfulgt av kjøling, overføring til en skilletrakt, vask én gang med 700 ml H20 og én gang med 700 ml mettet vandig natriumkarbonatløsning. 150 ml CH2C12 ble så tilsatt, hvoretter det ble foretatt en sluttvask med 700 ml H20. Det organiske sjikt ble tørret over natriumsulfat, filtrert og inndampet i høyvakuum, hvorved man fikk 1,3-dibenzyl-2-acetoksymetylglyserol som en klar olje.

Eksempel V
1, 3- dipivalovl- 2- metoksvmetvlglyserol.
50,45 g 1,3-dipivaloylglyserol ble oppløst i 200 ml metoksymetylacetat. Etter tilsetning av 2,5 g p-toluensulfon-syremonohydrat ble blandingen omrørt i 1 time. Deretter ble
den fortynnet med 200 ml heksan og vasket to ganger med 200 ml mettet vandig natriumkarbonat og én gang med vandige NaCl-løs-ninger, tørret over vannfritt magnesiumsulfat og konsentrert i vakuum, hvilket ga 1,3-dipivaloyl-2-metoksymetylglyserol som
en fargeløs olje.

Ved på tilsvarende måte å bruke de tilsvarende 1,3-di-substituerte glyseroler fra eksempel I ble den følgende forbindelse fremstilt: 1, 3-di-(1-adamantanoyl)-2-metoksymetylglyserol. Proton-NMR, CDC13, 1,6-2,1 (kompleks, 30H);
3,4 (s, 3H); 4,1 (kompleks, 1H); 4,2 (kompleks 4H), 4,8 (s, 2H).
Eksempel VI
1, 3- dipivalovl- 2- acetoksymetvlqlyserol.
50 g 1,3-dipivaloyl-2-metoksymetylglyserol, 150 ml metylenklorid og 17 ml eddiksyreanhydrid ble kjølt til ca.
-5°C. Deretter ble 0,3 ml bortrifluorideterat tilsatt, og blandingen ble omrørt i ytterligere én time ved ca. 0°C. Den
fikk så oppvarmes til 25°C i løpet av ytterligere 1/2 time. Etter dette tidsrom ble det tilsatt 100 ml mettet vandig Na2C03 til blandingen etterfulgt av 100 ml CH2C12. Etter kraftig om-røring i 5 min ble det organiske sjikt skilt fra og vasket én gang med 100 ml H20 og én gang med vandig NaCl, tørret over vannfritt magnesiumsulfat og konsentrert i vakuum. 1,3-dipivaloyl-2-acetoksymetylglyserol ble oppnådd som en svakt farget olje.

På tilsvarende måte ble 1,3-di-(1-adamantanoyl)-2-acetoksymetylglyserol fremstilt.

Det etterfølgende eksempel illustrerer fremgangsmåten ifølge oppfinnelsen.
Eksempel.
Fremstilling av 9-( 1. 3- dipivalovl- 2- propoksymetyl)- guanin og 9-( 1, 3- dibenzyloksy- 2- propoksymetyl)- guanin.
8,00 g guanin, 40 ml heksametyldisilazan, 80 ml xylen og 0,64 g ammoniumsulfat ble kokt med tilbakeløp i ca. 20
timer. 80 ml flyktige bestanddeler ble så destillert ut av den resulterende klare løsning og kastet. 23,1 g 1,3-dipivaloyl-2-acetoksymetylglyserol ble deretter tilsatt til den gjenværende oppløsning, og blandingen ble kokt med tilbakeløp i ca. 20
timer. Etter avkjøling ble de flyktige bestanddeler fjernet i vakuum. Residuet ble kokt med tilbakeløpskjøling sammen med 40 ml isopropanol, hvoretter flyktige bestanddeler ble avdestil-lert i vakuum. Ved kromatografering av residuet over kiselgel ble 9-(l,3-dipivaloyl-2-propoksymetyl)-guanin (1,3-dipivaloyl-DHPG) oppnådd. Utbytte 66-70%.

Ved anvendelse av 1,3-dibenzyl-2-acetoksymetyl-glyserol som utgangsmateriale ble på tilsvarende måte 9-(1,3-dibenzyloksy-2-propoksymetyl)-guanin (1,3-dibenzyl-DHPG) fremstilt. Utbytte 66-70%.

Claims (1)
- Fremgangsmåte for fremstilling av derivater av 9-(1,3-dihydroksy-2-propoksymetyl)-guanin med den generelle formel I:hvor R<1> og R<3> er valgt blant hydrogen, C^-alkyl, C4_20-alkyl-C0-, fenyl-CO-, 1-adamantanoyl, fenyl eller fenyl-C1.4-alkyl, hvor alle alkyl- og fenylgrupper eventuelt kan være substituert med halogenatomer, C1.4-alkylgrupper og fenylgrupper, og R<5> er hydrogen, C4.20-alkyl-CO- eller fenyl-CO-, hvor alkylgrup-pen, henholdsvis fenylgruppen kan være substituert med halogenatomer, C1.4-alkylgrupper og fenylgrupper, karakterisert ved at (a) en guaninforbindelse med den generelle formel II:hvor hver R<5> uavhengig av den andre er som angitt for R<5> ovenfor eller er (alkyl)3Si, omsettes, eventuelt i nærvær av et heksa-C1.4-alkyl-disilazan, med en forbindelse med den generelle formel A:hvor R<1> og R<3> er som ovenfor angitt, og R<4> er C1.4-alkyl som eventuelt kan være substituert med halogenatomer, C1.4-alkylgrupper og fenylgrupper, hvoretter eventuelt (b) den resulterende blanding behandles med en C^-alkylalkohol og/eller (c) den resulterende blanding - hvis R<1> og R<3> ikke er hydrogen - behandles med ammoniumhydroksid og vann.
