Description
Foreliggende oppfinnele vedrører en analogifremgangsmåte ved fremstilling av terapeutisk aktive fenylhydrazoner med formelen I,
hvori:
R<1>, R<2> og R3 uavhengig av hverandre betyr et hydrogen- eller halogenatom, en Ci-C^-alkylgruppe, en hydroksyl- eller C^ C^-alkoksygruppe eller acetoksyresten, RA betyr et hydrogenatom,
en hydroksylgruppe eller en Cj-Cg-alkylrest-, R<5> betyr et hydrogenatom eller en Ci-C^-alkylgruppe eller R<4> og R<5> under dannelse av en rir g i fellesskap betyr en -C(CH3)2-A-C(CH3)2-gruppe (med A i betydningen -CH2CH2-, -CH(CH3)-, -CH2C(0)- eller
-CH2CHOH-) eller en - (CH2) 3C (CH3) 2-, en -OCH2CH2C(CH3)2-, en - C(CH3)2CH(CH3)CH2C(CH3)2- eller en -NHC (0) CH2C (CH3) 2-gruppe, hvorved RA og R<5> tilsammen danner en ring av nevnte type, eller R<*> er en forgrenet CA-C6-alkoksy-alkylrest, når R<1> til R<3> er hydrogen, R<6> betyr et hydrogenatom, metyl-, etyl- eller cyklopropylresten, m og n er forskjellig fra hverandre og lik 0 eller 1, X betyr et hydrogenatom, en nitro-, metoksy- eller nitrilgruppe, sulfonsyreresten eller restene -CONR<7>OR<7>, - ;C02R<7>, -PO(OR<8>)2, <->S(0)n R<8> (med n=0 eller 2), -S02-NR<9>R<10> eller - CONR9R1<0>, hvori R<7> står for hydrogen, en Cj-Ca-alkylgruppe, og R<8 >står for en Ci-Cs-alkylgruppe, R<9> og R<10> uavhengig av hverandre er hydrogen eller en C-L-C^-alkylgruppe eller tilsammen danner en piperidin-, piperazin-, morfolin- eller tiomorfolinring, forutsatt at RA og R<5> tilsammen danner en ring av ovennevnte type, eller R<3> og R<5> avvekslende er i-propyl eller iso- eller tert-butyl, når X er et hydrogenatom, en nitrogruppe eller metoksygruppe, eller når X er karbonyl og R<1> er hydroksyl, og 4 5. ;R og R ^tilsammen danner en ring av den angitte type når X er CN, og at R ikke er hydroksyl hvis m=0, såvel som av deres ;fysiologisk aksepterbare salter. ;Det er kjent at stilbenderivater (sammenlign DE-OS 2 854 354, DE-OS 3 534 564 og US 4 588 750), ved hvilke polyenstruk-turen i substansene med vitamin-A-type foreligger bundet i aromatiske ringer, har farmakologiske virkninger ved lokal- og systemisk terapi av neoplasier, akne, psoriasis og andre dermatologiske lidelser. Virkningen av disse forbindelsene er imidlertid ikke alltid tilfredsstillende (sammenlign G.L. Peck i: The . JRe ti no ids, vol. II, 391-409, Ed.: M.B. Sporn et al., Academic Press N.Y. (1984), eller R. Marks et al., Med. J. Australia 146, 374-377 (1987), eller CE. Orfanos et al., Drugs 34, 459-503 (1987)). ;Til grunn for oppfinnelsen lå den oppgaven å utvikle forbindelser med bedre virkningsspektrum. ;Overraskende ble det nå funnet at de nye fenylhydrazonene og deres fysiologisk aksepterbare salter får et bedre virkespektrum. ;Blant forbindelsene med formelen I foretrekkes de - dersom R1 og R2 er hydrogen - hvori R3 og R<5> står for en fortrinnsvis forgrenet alkylrest, eller R<4> og R<5> tilsammen danner en ring. ;Betyr R<1>, R2 eller R3 halogen, så foretrekkes fluor og klor. ;Foretrukket er videre forbindelser med formelen I, hvori R6 står for et hydrogenatom eller en metylgruppe, så vel som forbindelser med formelen I, hvori X tilkommer betydningen av en -C02R<7>-, en -CONR7OR<7->, -S(0)nR<8->, S02NR9R<10-> eller CONR9R10-gruppe, hvorved R<7> i betydning av hydrogen og R<8> i betydning av en metyl- eller etylgruppe blir foretrukket ganske spesielt. ;De nye forbindelsene med formelen I inneholder delvis kirale sentra og forekommer vanligvis som diastereomer-blandinger, henholdsvis racemater. De således oppnådde diastereomerene lar seg adskille og isolere i ren form, eksempelvis ved løselighetsforskjeller eller ved kolonne-kromatografi. Ut i fra enantiomerparene kan man ved kjente metoder oppnå enhetlige enantiomerer. Både disse så vel som deres blandinger (racemater) omfattes av den foreliggende oppfinnelse. Som terapautiske eller kosmetiske midler kan man anvende både de enhetlige diastereomerene, henholdsvis enantiomerene som også deres blandinger. ;Noen av forbindelsene fremstilt ifølge oppfinnelsen har et surt hydrogenatom, og kan derfor overføres med baser på vanlig måte i et fysiologisk akspterbart, godt vannløselig salt. Egnede salter er eksempelvis ammonium-, alkalimetall-, spesielt av natrium, kalium og litium, eller jordalkalimetall-salter, .-spesielt av kalsium eller magnesium, så ^vel som salter med egnede organiske baser, som med lavere alkylaminer, f.eks. metylamin, etylamin eller cykloheksylamin, eller med substi-tuerte lavere alkylaminer, spesielt hydroksysubstituerte alkylaminer, som dietanolamin, trietanolamin eller tris-(hydroksymetyl)-aminometan så vel som med piperidin eller morfolin. ;Ifølge foreliggende oppfinnelse fremstilles ovenfor angitte forbindelser med formelen I, ved kondensasjon av ;a) karbonylforbindelser med formelen Ila ;hvori R1-!*6 har den ovenfor angitte betydning, med fenylhydraziner med formelen Illa hvori X har den ovenfor angitte betydning, eller b) karbonylforbindelser med formelen Ilb hvori R<6> og X har, den ovenfor angitte betydning, med fenylhydraziner med formelen Illb
hvori R^R5 har den ovenfor angitte betydning, og eventuelt omsetter reaksjonsproduktet med en fysiologisk akseptabel syre.
Omsetningen foregår på i og for seg kjent måte (sammenlign f.eks. "Methoden der Organischen Chemie" utgiver Eugen MMu"ller, bind VII, 1, side 461-466 Thieme Verlag, Stuttgart 1954 og bind VII, 2b, side 1954-1957, Thieme Verlag, Stuttgart 1976 og bind X, 2, side 410-414, Thieme Verlag, Stuttgart 1967), eventuelt i nærvær av et løsnings- eller fortynnings-middel, eventuelt under tilsetning av en katalysator og eventuelt under anvendelse av et vannbindende middel ved tempera-turer mellom 10°C og blandingens kokepunkt, hvorved man omsetter fortrinnsvis ekvimolare mengder av reaktantene II og III eller en komponent eventuelt i overskudd på inntil 15 mol-
Til de foretrukne løsnings- og fortynningsmidlene hører hydrokarboner som heptan, cykloheksan, toluen eller xylen, dessuten lavere alifatiske alkoholer, som metanol, etanol og isopropanol, men også cykloheksanol så vel som etylenglykol, deres mono- og dialkyletere, glycerol, videre etere som dietyleter, diisopropyleter og metyl-tert.-butyleter eller tetrahydrofuran og dioksan. Enn videre kan nevnes eddiksyre, amider som dimetylformamid eller N-metylpyrrolidon, videre pyridin, sulfolan og vann eller tilsvarende blandinger.
Som reaksjonsakseleratorer kommer mineralsyrer på tale, som saltsyre eller svovelsyre, fortrinnsvis karboksylsyrer som eddiksyre eller trifluoreddiksyre så vel som deres alkali-salter. Men også baser som pyridin eller morfolin kan tjene som katalysator.
Som vannbindende middel anvendes uorganiske salter som vannfritt natriumkarbonat eller magnesiumsulfat eller også molekylsikter; eventuelt tar man det dannede reaksjonsvannet ut av omløp ved arbeid i lipofile medier.
Reaksjonene gjennomføres under trykk eller ikke under trykk.
Utgangsforbindelsene med formelen II er delvis kjente
(f.eks. DE-OS 3 602 473, DE-OS 3 434 942, DE-OS 3 434 944)
eller de kan fremstilles etter vanlige metoder for fremstilling av arylalkylketoner, eksempelvis ved Friedel-Crafts acylering (H.O. House: "Modern Synthetic Reactions", andre utgave, W.A. Benjamin Inc. Menlo Park, CA; 1972, side 797 ff og litteratur sitert deri), eller ved oksydasjon av de tilsvarende alkylbenzenene (H.O. House, l.c, s. 288 f og der angitt litteratur), så vel som til fremstilling av benz-aldehyder, eksempelvis ved aromatformylering ifølge Vilsmeier (konferer De. Meheas, Bull. Soc. Chem. Fr. 1989-1999 (1962) og litteratur sitert der) eller ved reduksjon av de tilsvarende benzoylhalogenidene (konferer Fuson i: Patai, "The Chemistry of the Carbonyl Group", vol. 1, side 211-232, Interscience Publ., N.Y. 1966 eller Wheeler i Patai, "The Chemistry of Acyl Halides", s. 231-251, Interscience Publ. N.Y. 1972) eller benzonitril (konferer J. March: "Advanced Organic Chemistry", andre utgave, McGraw-Hill Kogakusha LTD., Tokyo, 1977, s. 835-836 og litteratur sitert der).
Utgangsforbindelsene med formelen III er kjente (se "Methoden der Organischen Chemie" utgiver Eugen Mu"ller, bind X, 2, s. 169-315, Thieme Verlag, Stuttgart 1967), eller de lar seg fremstille ifølge vanlig kjente fremgangsmåter for fremstilling' av arylhydraziner.
De ifølge oppfinnelsen oppnåelige forbindelsene og deres fysiologisk aksepterbare salter kan anvendes på grunn av sine farmakologiske egenskaper ved lokal- og systemisk terapi og profylakse av precanceroser og carcinomer i huden, slimhudene og indre organer, så vel som ved lokal- og systemisk terapi av akne, psoriasis og andre dermatologiske sykdommer som gir seg utslag i patologisk forandret forhorning, spesielt iktyose, Dariers-sykdom, utslett, leukoplaki, men også mot vitiligo, eksemer Æg vorter, videre mot tørre øyne og andre korneo-patier, så vel som til behandling av reumatiske sykdommer, spesielt av betennelsesmessig eller degenerativ art, som angriper leddene, musklene, senene og andre deler av bevegelsesapparater. Et foretrukket indikasjonsområde er ved siden av terapien av dermatologiske sykdommer og hudskader forårsaket av sollysets påvirkning eller iatrogent betingede, f.eks. ved kortikosteroidindusert atrofi, den profylaktiske behandlingen av precanceroser og tumorer.
Den farmakologiske virkningen kan påvises eksempelvis i de etterfølgende testmodellene: de ifølge oppfinnelsen oppnåelige forbindelsene opphever på trakealvev hos hamstere in vitro den keratiniseringen som opptrer etter vitamin-A-mangel. Keratiniseringen hører til den tidlige fasen i karcinogenesen, som blir inhibert i en lignende teknikk in vivo etter initiering ved kjemiske forbindelser, ved ener-getisk stråling eller etter viral celletransformasjon ved forbindelsene ifølge oppfinnelsen med formelen (I). Denne metodikken kan finnes i Cancer Res. 36, 964-972 (1972) eller i Nature 250, 64-66 (1974) og Nature 253, 47-50 (1975).
I tillegg hertil inhiberes veksthastigheten for bestemte maligne forandrede celler ved de forbindelsene som kan fremstilles ifølge oppfinnelsen. Denne metodikken kan finnes i J. Nati. Cancer Inst. 60, 1035-1041 (1978), Experimental Cell Research 117, 15-22 (1978) og Proe. Acad. Sei. USA 77, 2937-2940 (1980).
Den antiartritiske virkningen av forbindelsene som kan fremstilles ifølge oppfinnelsen, kan bestemmes på vanlig måte ved dyreeksperimenter i adjuvans-artritis- eller streptokokk-celleveggindusert artritis-modellen. Den dermatologiske aktiviteten, eksempelvis for behandling av akne, kan påvises blandt annet ved den komedolytiske aktiviteten og evnen til å redusere antallet cyster i rhino-mus-modellen.
Denne metoden er beskrevet av L.H. Kligman et al., i The Journal of Investigative Dermatology 73, 354-358 (1978).
Som ytterligere mål for den dermatologiske aktiviteten kan tjene reduksjonen av talgkjertlene og den dermed følgende reduserte talgproduksjonen på hamsterens sideorgan. Denne metodikken er beskrevet av E.C. Gomez i J. Am. Dermatol. 6, 746-750 (1982).
Dessuten kan den reversjonen av hudskader som kan oppnås ved de forbindelsene som kan fremstilles ifølge oppfinnelsen, og som ble utløst ved UV-lys, bestemmes i dyremodeller. Denne metodikken er beskrevet av L.H. Kligman et al., i Connect. Tissue Res. 12, 139-150 (1984) og i Journal of the American Academy of Dermatology 15, 779-785 (1986). FARMAKOLOGISKE DATA
Fremstilling av utgangsforbindelsene
Eksempel A
1,4-Dimetoksy-2-formyl-5,6,7,8-tetrahydro-5,5,8,8-tetra-metylnaftalin.
92,9 g (0,37 mol) 1,4-Dimetoksy-5,6,7,8-tetrahydro-5,5,8,8-tetrametylnaftalin og 52,5 g (0,37 mol) heksa-metylentetramin ble oppvarmet i 350 ml trifluoreddiksyre i 2 timer under tilbakeløp. Reaksjonsløsningen ble inn-konsentrert i vakuum, resten helt over i is, nøytralisert med fast natriumkarbonat og ekstrahert med eter. Ekstraktet som ble vasket med vann og tørket over magnesiumsulfat, ga etter avdamping av løsningsmiddelet en oljeaktig rest. Rensing ved hjelp av flash-kromatografi ga 45 g av tittelforbindelsen som ble omkrystallisert fra heptan. Smeltepunkt 55-57°C.
Eksempel B
1,1,2,3,3-Pentametyl-2,3-dihydro-5(1H)-indenyl-cyklo-proylketon.
Til en under avkjøling fremstilt suspensjon av 125 g (0,94 mol) vannfritt aluminiumklorid i 185 ml tørr metylenklorid ble tilsatt dråpevis 12 5 g (1,2 mol) cyklopropylkar-boksylsyreklorid og deretter ved 0-5°C 158 g (0,84 mol) 1,1,2,3,3-pentametyl-2,3-dihydro-(1H)-indan, løst i 230 ml tørr metylenklorid. Reaksjonsløsningen ble omrørt over natten ved 5°C, ble helt over i is og ekstrahert med metylenklorid. Ekstraktene ble vasket nøytrale med natriumkarbonatløsning og vann, tørket over natriumsulfat og inndampet. Den oljeaktige resten leverte etter destillasjon 159 g av tittelforbindelsen som fargeløs olje, K<p>0)4: 120-125°C, n<22>: 1,5425.
Eksempel C
3,5-Di-tert.-butyl-fenylhydrazin.
22 g (0,107 mol) 3,5-Di-tert.-butylanilin ble i 20 ml vann ved 80°C tilsatt dråpevis med 5,7 g (50 mmol) hydroksylamin-O-sulfonsyre; etter avkjøling ble bunnfallet suget -av og kokt ut med toluen. Resten ga etter behandling med 2N natronlut, ekstraksjon med metylenklorid og avdamping av løsningsmiddelet en olje, som ble kromatografert på silikagel i systemet n-heptan/eddiksyreester (10:1). Etter fraskilling av det ikke-omsatte utgangs-anilinet ble tittelforbindelsen eluert i systemet n-heptan/eddiksyreester (10:3). Man oppnådde 3,5 g av en olje, som fra eter med eterisk saltsyre dannet et fargeløst salt, smeltepunkt: 196-199°C.
Fremstilling av sluttproduktene
Eksempel 1
1,4-Dimetoksy-5,6,7,8-tetrahydro-5,5,8,8-tetrametyl-naftyl-2-aldehyd-N-(4-karboksyfenyl)-hydrazon.
8,3 g (30 mmol) 1,4-Dimetoksy-2-formyl-5,6,7,8-tetrahydro-5,5,8,8-tetrametyl-naftalin (eksempel A) og 4,6 g (30 mmol) fenylhydrazin-4-karboksylsyre ble rørt inn i 7 5 ml tetrahydrofuran under tilbakeløp. Etter avsluttet omsetning ble løsningsmiddelet dampet av. Omkrystallisering av resten fra metanol ga 8,5 g av tittelforbindelsen, smeltepunkt: 238-245°C.
Eksempel 2
1,1,2,3,3-Pentametyl-2,3-dihydro-5(1H)-indenyl-cyklopropyl-keton-N-(4-karboksyfenyl)-hydrazon.
7,7 g (30 mmol) 1,1,2,3,3-Pentametyl-2,3-dihydro-5(1H)-indenyl-cyklopropylketon (eksmpel B) og 4,6 g (30 mmol) fenylhydrazin-4-karboksylsyre ble rørt inn i en blanding "av 75 ml tetrahydrofuran, 10 ml etanol og 2 ml
iseddik under tilbakeløp. Etter avsluttet omsetning ble løsningen dampet inn, og resten omkrystallisert fra etanol. Man oppnådde 3,9 g av tittelforbindelsen, smeltepunkt: 227-231°C.
Eksempel 3
1,2,3,4-Tetrahydro-l,1,4,4-tetrametyl-naftalenyl-6-aldehyd-N-(4-karboksyfenyl)-hydrazon. 5.^0 g (23 mmol) 1,2,3,4-Tetrahydro-l,1,4, 4^tetrametyl-naftalenyl-6-aldehyd og 3,8 g (25 mmol) fenylhydrazin-4-karbokylsyre ble rørt inn i 50 ml tetrahydrofuran i 1 time under tilbakeløp. Etter avkjøling ble reaksjonsløsningen helt over i heptan, bunnfallet ble frafiltrert og omkrystallisert fra metanol. Man fikk 5,4 g av tittelforbindelsen, smeltepunkt: 260°C.
Eksempel 4
1,2,3,4-Tetrahydro-l,1,4,4-tetrametyl-naftalenyl-6-metyl-keton-N-(4-karboksyfenyl)-hydrazon.
5,0 g (22 mmol) 1, 2<> 3 , 4-Tetrahydro-l, 1, 4 , 4-^tetrametyl-naftalenyl-6-metylketon og 3,8 g (25 mmol) fenylhydrazin-4-karboksylsyre ble omsatt tilsvarende eksempel 3 og ga etter omkrystallisering fra etanol 3,6 g av tittelforbindelsen,
smeltepunkt: 2 50"C.
Eksempel 5
4-Metoksykarbonyl-benzaldehyd-N-(3,5-di-tert.-butylfenyl)-hydrazon.
3,5 g (15 mmol) 3,5-Di-tert.-butylfenylhydrazin og 2,6 g (15 mmol) 4-formyl-benzosyremetylester ble rørt inn i 150 ml tetrahydrofuran/etanol (1:1) i 1 time under tilbakeløp. Etter avdamping av løsningsmiddelet ble resten digerert kokende varmt med etanol, avsuget etter avkjøling og ettervasket med etanol. Etter tørking i vakuum ved romtempera-tur fikk man 3,2 g av tittelforbindelsen, smeltepunkt: 215-220°C.
Eksempel 6
4-Karboksybenzaldenyd-N-(3,5-di-tert.-butylfenyl)-hydrazon.
2,2 g av esteren fremstilt i eksempel 5 ble oppvarmet til tilbakeløp i 70 ml etanol/dimetylsulfoksyd/vann (3:3:1) med 3 g kaliumhydroksyd i 1 time. Reaksjonsblandingen ble helt over i 140 ml isvann, innstilt på pH 2 med 2N saltsyre, og bunnfallet avsuget. Etter omkrystallisering fra metanol fikk man 1,2 g av tittelforbindelsen, smeltepunkt: 2 08-21-3t°C.
På samme måten som i eksemplene 1-6 ble fremstilt de eksemplene som er oppført i den etterfølgende tabellen:
Beskrivelse til eksempel 4 6
1,2,3,4-Tetrahydro-l,1,4,4-tetrametyl-naftalenyl-6-metyl-keton-N-(metylsulfonylfenyl)-hydrazon (anti-isomer).
4,7 g (20 mmol) 1,2,3,4-Tetrahydro-l,1,4,4-tetrametyl-naftalenyl-6-metylketon og 4,0 g (20 mmol) 4-metylsulfonyl-fenylhydrazin ble oppvarmet under tilbakeløp i 60 ml etanol. Etter avsluttet omsetning ble bunnfallet suget av, ettervasket med etanol og tørket ved 50°C i vakuum. Man fikk 1 g av tittelforbindelsen, smeltepunkt:^ 211-214°C.
Eksempel 4 6a
1,2,3,4-Tetrahydro-l,1,4,4-tetrametylnaftalenyl-6-metyl-keton-N-(4-metylsulfonylfenyl)-hydrazon (syn.-isomer).
Moderluten fra den foregående eksempelforbindelsen ble inndampet til tørrhet, og resten kromatografert på silikagel i systemet heptan/eddiksyreetylester (99:1 til 80:20). Etter inndamping av det tilsvarende eluatet, fikk man 0,2 g av tittelforbindelsen, smeltepunkt 216-218°C.
Claims (1)
- Analogifremgangsmåte ved fremstilling av terapeutisk aktive fenylhydrazoner med formelen I,hvori:R<1>, R<2> og R<3> uavhengig av hverandre betyr et hydrogen- eller halogenatom, en Ci-C^-alkylgruppe, en hydroksyl- eller C^.-CA-alkoksygruppe eller acetoksyresten,R<4> betyr et hydrogenatom, en hydroksylgruppe eller en C^ C^-alkylrest-,R<5> betyr et hydrogenatom eller en Ci-C^-alkylgruppe eller R<4> og R<5> under dannelse av en ring i fellesskap betyr en -C(CH3)2-A-C(CH3)2-gruppe (med A i betydningen -CH2CH2-, - CH(CH3)-, -CH2C(0)- eller -CH2CHOH-) eller en - (CH2)3C(CH3)2-, en -OCH2CH2C (CH3) 2-, en - C(CH3)2CH(CH3)CH2C(CH3)2- eller en -NHC(O) CH2C(CH3)2-gruppe, hvorved R4 og R<5> tilsammen danner en ring av nevnte type, eller R4 er en forgrenet CA-C6-alkoksy-alkylrest, når R<1> til R<3> er hydrogen, R<6> betyr et hydrogenatom, metyl-, etyl- eller cyklopropylresten, m og n er forskjellig fra hverandre og lik 0 eller 1, X betyr et hydrogenatom, en nitro-, metoksy- eller nitrilgruppe, sulfonsyreresten eller restene -CONR<7>OR<7>, - C02R<7>, -PO(OR<8>)2, <->S(0)n R<8> (med n=0 eller 2), -S02-NR<9>R<10 >eller -CONR9R<10>, hvori R7 står for hydrogen, en C^-C,-alkylgruppe, Rd står for en Cx-Cs-alkylgruppe, R<9> og R<10 >uavhengig av hverandre er hydrogen eller en Ci-C^-alkylgruppe eller tilsammen danner en piperidin-, piperazin-, morfolin- eller tiomorfolinring, forutsatt at R4 og R<5 >tilsammen danner en ring av ovennevnte type, eller R3 og R<5 >avvekslende er i-propyl eller iso- eller tert-butyl, når X er et hydrogenatom, en nitrogruppe eller metoksygruppe, eller når X er karbonyl og R er hydroksyl, og R og R tilsammen danner en ring av den angitte type når X er CN, og at R<4 >ikke er hydroksyl hvis m=0, såvel som av deres fysiologisk aksepterbare salter,karakterisert ved at man omsetter a) karbonylforbindelser med formelen Ila hvori RI til R6 har den betydningen som er angitt ovenfor, med fenylhydrazin med formelen Illa hvori X har den betydningen som er angitt ovenfor, eller b) karbonylforbindelser med formelen IIb hvori R<6> og X har de nevnte betydningene, med fenylhydraziner med formelen Illb hvori R<1> til R<5> har de nevnte betydningene, og eventuelt omsetter reaksjonsproduktet med en fysiologisk aksepterbar syre.
