Description
Foreliggende oppfinnelse omhandler analogifremgangsmåter ved fremstilling av nye quinolinforbindelser med meget høy antibakteriell aktivitet.
Forbindelsene fremstilt ifølge denne oppfinnelse er guinolinderivater representert av følgende generelle formel Ia:
hvor R er hydrogen eller en metylgruppe og Y er hydrogen eller C1-C4~alkyl, eller et farmasøytisk akseptabelt salt derav.
Saltene av forbindelsene ifølge formel Ia kan være ethvert salt dannet fra forbindelsene av formel Ia med farmasøytisk akseptable syrer eller baser. Saltene av forbindelsene fremstilt ifølge oppfinnelsen er salter avledet fra uorganiske syrer såsom saltsyre eller fosforsyre; de fra organiske syrer såsom eddiksyre, melkesyre, oksalsyre, ravsyre, metahsulfonsyre, eplesyre, maleinsyre, malonsyre eller glukonsyre; de fra sure aminosyrer såsom asparaginsyre eller glutaminsyre; metall ( for eks. natrium kalium, kalsium, magnesium, sink eller sølv) salter; de fra organiske baser såsom dimetylamin, trietylamin, dicyklo-heksylamin eller benzylamin og de fra basiske aminosyrer såsom lysin eller arginin.
Forbindelsene av formel Ia og salter av disse forbindelser vil følgelig generelt refereres til heri som forbindelsene fremstilt ifølge oppfinnelsen.
Forbindelsene fremstilt ifølge oppfinnelsen innebefatter de som har asymmetriske karbonatomer og piperazinringen som har asymmetriske karbonatomer ved 7-posisjon og derfor fore-ligger i optisk aktive former. Således innebefatter de D-isomere, L-isomere eller blandinger derav.
Enkelte av forbindelsene ifølge oppfinnelsen har to asymmetriske karbonatomer på piperazinringen ved 7-posisjon og kan følgelig eksistere som stereo-isomere med forskjellige kon-figurasjoner (cis eller trans form). Disse stereo-isomere og deres blandinger er også innebefattet i forbindelsene fremstilt ifølge oppfinnelsen.
Tidligere teknikk angående farmasøytisk effektive forbindelser i dette omdrådet vil bli omtalt nedenfor.
Japansk offentliggjort patent publikasjon no. 174367/1983
(et sammendrag som er beskrevet i Derwent World Patent Index, tilgangsnummer 83-8232272) omtaler at forbindelser representert ved den generelle formel (10)
hvori R representerer et hydrogenatom eller en lavere alkyl gruppe, har antibakteriell aktivitet. Imidlertid har forbindelsene ifølge foreliggende oppfinnelse høyere antibakteriell aktiviet enn de ovenfor angitte kjente forbindelser.
Syd Afrikansk utlagt Patentbeskrivelse no. 8509369 omtaler følgende generelle formel (11)
Beskrivelsen angir ikke en cyklopropylgruppe for R som gruppen er i formel (11).
Europeisk utlagt patentbeskrivelse nr.172651 angir forbindelser representert av følgende generelle formel (12);
Beskrivelsen angir ikke en piperazinyl gruppe som gruppen Z i formel (12).
US Patent No, 4,556,658 beskriver forbindelser representert av følgende generelle formel (13)
De har imidlertid ikke en amino gruppe ved 5-posisjonen i quinolin ringen som klart fremgår av formel (13).
Det er et mål for foreliggende oppfinnelse og fremskaffe nye quinolinderivater av formel Ia med høy antibakteriell aktivitet mot både gram positive bakterier og gram negative bakterier, og farmasøytisk akseptable salter av disse.
Forbindelsene fremstilt ifølge oppfinnelsen viser utmerket antibakteriell aktivitet og et bredt antibakterielt spektrum i in vitro tester. Videre viser disse forbindelser en utmerket infeksjonsbeskyttende effekt in vivo på topiske eller systemiske infeksjoner forårsaket av Gram-positive og Gramnegative bakterier.
Forbindelsene ifølge oppfinnelsen har også utmerket anti-mycoplasma aktivitet.
Fremgangsmåtene for fremstilling av forbindelsene ifølge oppfinnelsen er som angitt i krav l's karakteriserende del, og vil bli beskrevet nedenfor.
A. Substitusionsreaksion av piperazinderivater
Forbindelsene kan ifølge oppfinnelsen fremstilles ved å omsette en forbindelse av formel (Ila)
hvor X er halogen og Y er som angitt ovenfor, med et piperazinderivat av formel (Illa)
hvor R er hydrogen eller en metylgruppe, og R<1> er H eller en beskyttelsesgruppe, hvorpå eventuelle beskyttelsesgrupper avspaltes for å oppnå forbindelsene av formel (I).
Eksempler på halogneatomer for X i fomel Ila er fluor, klor eller bromatomer.
Denne reaksjon kan utføres ved å omrøre utgangsforbindelsene Ila og Illa ved 10 - 180°C i 10 min til 24 timer i et inert oppløsningsmiddel. Eksempler på inerte oppløsnings-midler innebefatter alkoholer såsom metanol, etere såsom dioksan, tetrahydrofuran og 1,2-dimetoksyetan, aromatiske hydrokarboner såsom benzen, toluen, xylen, acetonitril, dimetylformamid, dimetylsulfoksyd, pyridin og vann.
Generelt utføres ovenfor angitte reaksjon i nærvær av en syreakseptor ved å bruke utgangsforbindelsen av formel Illa i en mengde ekvivalent til eller i lite overskudd med hensyn til utgangsforbindelsen Ila. Om ønsket kan utgangsforbindelsen Illa være brukt i overskudd for å bevirke at det senere virker som syreakseptor. Eksempler poå syreakseptor er natriumhydrogenkarboat, natriumkarbonat, kaliumkarbonat, trietylamin, pyridin og picolin.
Utgangsforbindelsen Illa brukt i denne reaksjon kan om mulig, bli brukt i en beskyttet form og etter reaksjonen fjernes dens beskyttende gruppe på vanlig måte. Den beskyttende gruppe kan være enhver beskyttende gruppe som ikke ødeleg-ger strukturen til forbindelsen ifølge oppfinnelsen dannet ved reaksjonen. Grupper som normalt brukes som beskyttelsesgrupper for aminogruppen i området peptidkjemi, aminosuk-kere, nukleinsyrer eller beta-laktamforbindelser kan bli brukt i denne oppfinnelsen.
De aminobeskyttende grupper spaltes av ved solvolyse (innebefattende hydrolyse) eller reduksjon avhengig av
egenskapene til de beskyttende grupper.
Spesielle eksempler på grupper som er istand til å bli eli-minert ved solvolyse innebefatter acylgrupper såsom formyl, acetyl og trifluoracetyl; substituerte eller usub-stituerte alkoksykarbonyl grupper såsom etoksykarbonyl, t-butoksykarbonyl, benzyloksykarbonyl, p-metoksybenzyloksy-karbonyl og beta-(p-toluensulfonyl)-etoksykarbonyl; en trityl gruppe, en trimetylsilyl gruppe, en o-nitrofenylsul-fenyl gruppe, en difenylfosfinyl gruppe og en tetrahydro-pyranyl gruppe.
Denne reaksjon utføres i et oppløsningsmiddel ved 0 - 150°C i nærvær eller fravær av en katalysator såsom en syre eller base.
Eksempler på syrer er uorganiske syrer såsom saltsyre, hydrobromsyre, svovelsyre og fosforsyre; organiske syrer såsom eddiksyre, trifluoreddiksyre, maursyre og toluen-sulfonsyre; Lewis-syrer såsom bor-tribromid og aluminium-klorid. Eksempler på baser er alkaliske metallhydroksyder såsom natriumhydroksyd og barium-hydroksyd, alkalimetall-karbonater såsom natriumkarbonat og kaliumkarbonat; alkaliske metallalkoksyder såsom natriummetoksyd og natrium-etoksyd og natriumacetat. Vanligvis blir vann brukt som oppløsningsmiddel. Avhengig av egenskapen til forbindelsen kan et annet oppløsningsmiddel såsom etanol, dioksan, etylenglykoldimetyleter, benzen eller eddiksyre, eller et blandet oppløsningsmiddel såsom et oppløsningsmiddel med vann, bli brukt.
Eksempler på beskyttende grupper som kan elimineres ved reduksjon innbefatter arylsulfonyl grupper såsom p-touluen-sulfonyl, en metylgruppe substituert med fenyl eller benzyloksy, såsom benzyl, trityl eller benzyloksymetyl, arylmetoksykarbonylgrupper, såsom benzyloksykarbonyl- og p-metoksybenzyloksykarbonyl-, og halogenetoksykarbonylgrupper såsom beta, beta, beta-trikloretoksykarbonyl og beta-iodo-
etoksykarbonylgrupper.
Reaksjonen bruker forskjellige reaksjonsbetingelser avhengig av egenskapen til beskyttelsesgruppen som skal elimineres. For eks. utføres den ved å behandle forbindelsen med en hydrogenstrøm i et inert oppløsningsmiddel ved 10-60°C i nærvær av en katalysator såsom platina, palladium eller Raney-nikkel eller behandling med metallisk natrium i flytende ammoniakk vanligvis ved -50 - -20°C eller ved å behandle den med et metall såsom sink i eddiksyre eller i en alkohol såsom metanol. Eksempler på oppløsningsmiddel ved katalytisk reduksjon kan innbefatte etylenglykol, di-metyleter, dioksan, dimetylformamid, etanol, etylacetat og eddiksyre.
Utgangsforbindelsen Ila kan fremstilles ved metodene beskrevet i referanseeksempel 1 og 2 eller metoder vesentlig i samsvar med disse.
B. Aminerinqsreaksi on
Forbindelsene kan ifølge oppfinnelsen fremstilles ved å omsette en forbindelse av formel (IVa)
hvor X er halogen og R og Y er som angitt ovenfor, med ammoniakk.
Denne reaksjon kan utføres ved å føre sammen utgangsforbindelsen (IVa) med ammoniakk i 1 - 50 timer ved en temperatur på 50-150°C i et inert løsningsmiddel f. eks. en alkohol såsom etanol, pyridin, dimetylformamid eller vann fortrinns-vis i et forseglet rør.
Denne reaksjon utføres i nærvær av en syreakseptor ved å bruke ammoniakk i en mengde ekvivalent til eller i lite overskudd av utgangsforbindelsen (IVa). Passende brukes ammoniakk i overskudd for å gjøre at det virker også som syreakseptor. Et salt såsom ammoniumacetat ble brukt for å virke istedet for ammoniakk i denne sammenheng.
Utgangsforbindelsen (IVa) brukt i denne reaksjon kan, om mulig bli brukt i en form beskyttet med en slik beskytte1-sesgruppe som beskrevet ovenfor med hensyn til reaksjon A og efter reaksjonen elimineres beskyttelsesgruppen på vanlig måte. C. Avspalting av den 5- amino beskyttende gruppe.
Forbindelsen kan ifølge oppfinnelsen fremstilles ved å fjerne de beskyttende grupper i en forbindelse av formel
(Va)
hvor W er en for aminer vanlig benyttet beskyttelsesgruppe, og R og Y er som angitt ovenfor.
Eksempler på den aminobeskyttende gruppe W i formel (Va) innbefatter acylgrupper såsom formyl, acetyl og trifluoracetyl; alkoksykarbonylgrupper såsom etoksykarbonyl og benzyloksykarbonyl; substituerte metylgrupper såsom benzyl og benzhydryl og en benzyloksygruppe.
Denne reaksjonen kan utøføres på samme måte som beskrevet tidligere med hensyn til eliminering av den aminobeskyttende gruppe i reaksjon A.
Utgangsforbindelsen (Va) brukt i denne reaksjonen, kan om mulig bli brukt i en form beskyttet med en slik beskyttelsesgruppe som beskrevet med hensyn til reaksjon A, og der-etter eller samtidig med foreliggende reaksjon elimineres beskyttelsesgruppen på vanlig måte.
Utgangsforbindelsen (Va) er en ny forbindelse og kan fremstilles ved fremgangsmåtene beskrevet i referanseeksempel 3-5 nedenfor eller ved metoder i hovedsak i samsvar med disse. Når forbindelsene ifølge oppfinnelsen erholdt ved overfor angitte fremgangsmåter er estere, kan de konverteres til forbindelser av formel Ia ved å hydrolysere ester-enheten på vanlig måte. Om krevet kan forbindelsene av formel l esterifiseres på vanlig måte for å danne estere av forbindelsene ifølge formel Ia.
Farmasøytisk akseptable salter av forbindelsen av formel Ia kan fremstilles ved å behandle forbindelsene av formel Ia med syrer eller ved å behandle forbindelsene med baser eller metallsalter. Syrer passende for saltdannelse inneholder for eks. saltsyre, fosforsyre, eddiksyre, melkesyre, oksalsyre, ravsyre, metansulfonsyre, maleinsyre, malonsyre, glukonsyre, asparaginsyre og glutaminsyre. Baser eller metallsalter passende for saltdannelser innebefatter for eksempel metall -hydroksyder såsom natriumhydroksyd og kaliumhydroksyd, metallkarbonater såsom natriumkarbonat og kaliumkarbonat, sinkklorid, sinksulfat, sinknitrat og sølvnitrat.
Forbindelsene fremstilt ifølge oppfinnelsen som angitt ovenfor, isoleres og renses på vanlig måte og, avhengig av isolering og opprenskingsbetingelsene, kan de erholdes i form av et salt eller en fri syre. De kan konverteres i hverandre for å danne forbindelsene ifølge oppfinnelsen i de ønskede former.
Stereoisomere (cis og trans form) av forbindelsen fremstilt ifølge oppfinnelsen kan isoleres ved konvensjonelle metoder såsom fraksjonær krystallisering eller kromatografi. Det er mulig å fremstille forbindelser ifølge oppfinnelsen med en cis eller trans konfigurasjon ved reaksjonen A beskrevet ovenfor ved å bruke utgangsforbindelsene (Illa) med en cis eller trans konfigurasjon. De optisk aktive isomere av forbindelsen fremstilt ifølge oppfinnelsen kan separeres ved kjente metoder.
Forbindelsene (Ia) således erholdt, deres estere og salter av disse er alle nye forbindelser. Spesielt er forbindelsene (Ia) og deres salter verdifulle som antibakterielle midler siden de har meget høy antibakteriell aktivitet. Forbindelsene (Ia) og deres salter kan brukes som medisiner for mennesker og dyr.
Forbindelsene (Ib) ifølge oppfinnelsen, og som er angitt i krav 2, er verdifulle som utgangsmaterialer for forbindelser (Ia) .
Når forbindelsene fremstilt ifølge oppfinnelsen brukes som antibakterielle midler for mennesker, er det anbefalt at de tilføres i en dose på 5 mg til 5 g pr. dag én eller flere ganger om dagen, selv om dosen kan varieres avhengig av alder, kroppsvekt og symptom hos pasienten, tilføringsrute etc.. Forbindelsen kan tilføres oralt eller parenteralt.
Forbindelsen fremstilt ifølge oppfinnelsen kan tilføres i deres som erholdte pulverform, men de tilføres vanligvis i form av et farmasøytisk preparat sammen med farmasøytisk akseptable tilsettningsmidler. Spesielle eksempler på farmasøytiske preparater er tabletter, oppløsninger, kapsler, granuler, fine granuler, pelleter, pulvere, eliksirer, injeksjoner og salver. Disse farmasøytiske preparater fremstilles ved i og for seg kjente metoder. Tilsettningsmidler for oal tilførsel er de som vanligvis er brukt i området ved dannelse av farmasytiske preparater og reagerer ikke med forbindelsen ifølge oppfinnelsen såsom stivelse, manitol, krystallinsk cellulose, CMC-Na, vann, etanol osv. Tilsettningsmidler for injeksjoner er de som vanligvis er brukt angående injeksjoner såsom vann, isotonnatriumkloridoppløsning, glukoseoppløsning og transfusjonsoppløsning.
Ovenfor angitte flytende preparater og salver kan også bli brukt for lokal behandling i oto-rhino-laryngologi eller oftalmologi.
De følgende eksempler illustrerer dannelsen av forbindelsene ifølge oppfinnelsen mer spesielt.
Referanseeksempel 1
l-cyklopropyl-5,6,7,8-tetrafluor-1,4-dihydro-4-oksoquinolin-3-karboksylsyre;
Tallene til reaksjonstrinnene beskrevet nedenfor tilsvarer tallene i ovenfor angitte skjema. (1) En blanding av den kjente forbindelsen etyl pentafluor-benzoylacetat (J. Org. Chem., 35, 930 (1970)) 25 g, etylortoformat 20 g og eddiksyre anhydrid 23 g ble kokt under tilbakeløp i 2 timer, reaksjonblandingen ble inndampet til tørrhet under redusert trykk. Residiet ble oppløst i dietyleter og tillatt og reagere med cyklopropylamin 5,1 g for å gi etyl 2-pentafluorbenozyl-3-cyklopropylamioacrylat 28 g, smp 89°C. (2) Ovenfor angitte forbindelse 28 g, ble oppløst i tørr tetrahydrofuran og tillatt og reagere med 60% natrium hyd-rid 3,85 g ved romtemperatur for gi etyl 1-cyklopropyl-5,6,7,8-tetrafluor-1,4-dihydro-4-oksoguinolin-3-karboksylsyre 18,4 g, smp. 170-171°C. (3) Ovenfor angitte forbindelse 10 g ble hydrolysert ved å koke den under tilbakeløp i en blanding av is-eddiksyre 60 ml, vann 500 ml og konsentrert svovelsyre i 30 min. for å gi l-cyklopropyl-5,6,7,8-tetrafluor-1,4-dihydro-4-okso-quinolin-3-karboksylsyre 8,7 g, smp. 181-182°C. Referanseeksempel 2 5-amino-l-cyklopropyl-6,7,8-trifluor-1,4-dihydro-4-okso-guinolin-3-karboksylsyre;
Tall av reaksjonstrinn beskrevet nedenfor tilsvarer tallene i overfor angitte skjema. (1) En blanding av etyl l-cyklopropyl-5,6,7,8-tetrafluor-1,4-dihydro-4-okosquinolin-3-karboksylat 28,2 g fremstilt i referanse eksempel 1 (2), benzylamin 9,8ml, vannfritt kalium karbonat 23,6 g og acetonitril, 140 ml ble oppvarmet ved 100-110°C i 1 time for å gi etyl 5-benzylamino-l-cyklopropyl-6,7,8-trifluor-1,4-dihydro-4-oksoquinolin-3-kar-boskylate 21,4 g som ble rekrystallisert fra etanol, smp. 134-135°C. (2) Ovenfor angitte forbindelse 20 g ble oppløst i eddiksyre 100 ml og etanol 150 ml og hydrolysert i nærvær av 5% palladium-karbon 0,5 g for å gi etyl 5-amino-l-cyklopropyl-6,7,8-trifluor-1,4-dihydro-4-oksoquinolin-3-karboksylat 14,1 g som ble rekrystallisert fra kloroform-etanol, smp. 236-237°C. (3) En blanding av ovenfor angitte forbindelse 12,6 g, eddiksyre 80 ml vann 50 ml og konsentrert svovelsyre 9 ml ble oppvarmet til 100-110°C i 40 min for å gi 5-aminol-cyklopropyl-6,7,8-trifluor-1,4-dihydro-4-oksoquinolin-3-karboksylsyre og hydroklorid som ble rekrystallisert fra kloroform-etanol, smp. 294-295°C. EKSEMPEL 1 5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl) - 1,4-dihydro-4-oksoquinolin-3-karboksylsyre og dens hydro-klorider
Tallene på reaksjontrinnene beskrevet nedenfor, tilsvarer tallene i det ovenfor nevnte skjema.
(1) En blanding av 5-amino-l-cyklopropyl-6,7,8-trifluor-i f4-dihydro-4-oksoquinolin-3-karboksylsyre 1,25 g, 2-metylpiperazin 2,0 g, og pyridin 13 ml ble oppvarmet ved 105-110°C i 1. time under omrøring. Reaksjonblandingen ble inndampet til tørrhet under redusert trykk og vann ble tilsatt residiet. Blandingen ble ekstrahert med kloroform og ekstraktet ble tørket. Etter inndamping av kloroform ble etanol tilsatt residiet. De resulterende krystaller ble filtrert og rekkrystallisert fra kloroform etanol for å gi 5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl)
-1,4-dihydro-4-oksoquinolin-3-karboksylsyre 1,4 g, smp. 251-253°C. (2) 700 mg av ovenfor angitte forbindelse 700 mg ble oppløst i 20% saltsyre 7 ml og inndampet til tørrhet under redusert trykk, etanol ble tilsatt resideiet og de resulterende krystaller ble filtrert og reksrystallisert fra vann-etanol for å gi 5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl)-1,4-dihydro-4-oksoquinolin-3-karboksylsyre hydroklorid 425 mg, spm. >300°C. EKSEMPEL 2 5-amino-l-cyklopropyl-6,8-difluor-7-(cis-3,5-dimetyl-l piperazinyl)-1,4-dihydro-4-oksoquinolin-3-karboksylsyre;
På samme måte som beskrevet i Eksempel 1 (1), ble en blanding av 5-amino-l-cyklopropyl-6,7,8-trifluor-1,4-dihydro-4-oksoquinolin-3-karboksylsyre, cis-2,6-dimetyl-piperazin, og dimetylfromamid omrørt ved romtemperatur i 24 timer for å gi 5-amino-a-cyklopropyl-6,8-difluor-7-(cis-3,5-dimetyl-l-piperaz iny1)-1,4-dihydro-4-oksoquinolin-3-karboksylsyre, smp. 258-260°C.
EKSEMPEL 3
5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl)-l,4-dihydro-4-oksoquinolin-3-karboksylsyre og dens hydro-klorider;
Tallene på reaksjonstrinnene beskrevet nedenfor tilsvarer tallene i det ovenfor viste skjema. (1) På samme måte som beskrevet i Eksempel 1 (1) ble det, foruten å bruke l-acetyl-2-metylpiperazin istedet for 2-metylpiperazin, oppnådd 7-(4-acetyl-3-metyl-l-piperazinyl)-5-amino-l-cyklopropyol-l,4-dihydro-4-oksoquinolin-3-karboksylsyre, smp. 258-260°C. (2) En blanding av den ovenfor nevnte fremstiling, 20% vandig natrium hydroksyd, og etanol ble kokt under til-bakeløp i 12 timer. Reaksjonsblandingen ble behandlet med aktivert karbon og tilpasset en pH på 1 til 2 med 10 hydrokloridsyre. Tallene på reaksjonstrinnene beskrevet ovenfor tilsvarer tallene i det ovenfor viste skjema. (1) På samme måte som beskrevet i Eksempel 1 (1), ble det, foruten å bruke 5-amino-l-cyklopropyl-6,7,8-trifluor-1,4 - dihydro-4-oksoquinolin-3-karboksyat istedet for 5-amino-l-cyklopropyl-6,7,8-trifluor-1,4-dihydro-4-oksoquinolin-3-karboksylsyre, fremstilt etyl 5-amino-l-cyklopropyl-6,8-di-f luor-7^-(3-metyl-l-piperazinyl) 1, 4-dihydro-4-oksoquinolin3-karboksylat. smp. 132-134°C. (2) Ovenfor nevnte forbindelse ble behandlet på samme måte som beskrevet i Eksempel 7 (2) for å gi 5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl)-1,4,dihydro-4-ok-soquinolin-3-karboksylsyre og hydroklorid, smp. > 300°C. Referanseeksempel 3 l-cyklopropyl-5,6,8-trifluor-7-(3-metyl-l-piperazinyl)-1,4-dihydro-4-oksoquinolin-3-karboksylsyre;
En blanding av l-cyklopropyl-5,6,7,8-tetrafluor-1,4-dihydro-4- oksoguinolin-3-karboksylsyre 910 mg, 2-metylpiperazin 320 mg, og pyridin 10 ml ble omrørt ved 80°C i 1 time. Efter å ha fordampet reaksjonsblandingen under redusert trykk, ble det gjenværende oppløst i fortynnet vandig ammoniakk og behandlet med aktivert karbon. Filtratet ble fordampet under redusert trykk og tilpasset pH på 7 til 8. De resulterende krystaller ble filtert, vasket med vann ...ag tørket for å gi l-cyklopropyl-5, 6,8-trif luor-7- (3-metyl-l-piperazinyl) 1,4-dihydro-4-oksoquinolin-3-karboksylsyre 810 mg, smp. 235-237°C.
EKSEMPEL 5
5- amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl)
-1,4-dihydro-4-oksoquinolin-3-karboksylsyre;
En blanding av l-cyklopropyl-5,6,8-trifluor-7-(3-metyl-l-piperazinyl) -1,4-dihydro-4-oksoquinolin-3-karboksylsyre 150 mg og 2 0% vandig ammoniakk 15 ml ble oppvarmet ved 100°C i 48 timer i et forseglet rør. Reaksjonsblandingen ble fordampet til tørrhet under redusert trykk, og vann ble tilsatt det gjenværende. Blandingen ble ekstrahert med kloroform. Efter tørking, ble ekstraktanten fordampet og det gjenværende ble rekrystallisert fra kloroform-etanol for å gi 5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl) -1, 4-dihydro-4-oksoquinolin-3-karboksylsyre 93 mg, smp. 251-253°C.

På samme måte som beskrevet i Eksempel 1 (1) ble etyl-1-cyklopropyl-5-etoksykarbonylamino-6,7,8-trifluor-7-1,4-dihydro-4-oksoquinolin-3-karboksylat, smp. 189-190°C tillatt å reagere med 2-metylpiperazin for å gi etyl 1-cyklopropyl-5-etoksykarbonylamino-6,8-difluor-7-(3-metyl-l-piperazinyl)-1,4-dihydro-4-oksoquinolin-3-karboksylat.
EKSEMPEL 6
5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl)
-1,4-dihydro-4-oksoquinolin-3-karboksylsyre hydroklorid;
En blanding av etyl—l-cyklopropyl-S-etoksykarbonylamino-e^-dif luor-7- (3-metyl-l-piperazinyl) -1,4-dihydro-4-okso-quinolin-3-karboksylat, 20 % vandig natrium-hydroksyd, og etanol ble kokt under tilbakeløp i 12 timer. Reaksjonsblandingen ble behandlet med aktivert karbon og tilpasset en pH på 1 til 2 med 10% hydrokloridsyre. Efter avkjøling ble de resulterende krystaller samlet ved filtrering og rekrystallisert fra vann-etanol for å gi 5-amino-l-cyklopropyl-6,8-di fluor-7-(3-metyl-l-p iperaz iny1)-1,4-dihydro-4-oksoquino-lin-3-karboksylsyre hydroklorid, smp. > 300°C.
EKSEMPEL 7
Etyl 5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl) -l,4-dihydro-4-oksoquinolin-3-karboksylat;
5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperaz inyl) -1,4-dihydro-4-oksoquinolin-3-karboksylsyre ble suspendert i absolutt etanol. Konsentrert svovelsyre ble tilsatt til opplsøningen og blandingen ble kokt under tilbakeløp i 10 timer under omrøring. Efter fordamping av etanol, ble kloroform og 20 % vandig natrium hydroksyd tilsatt til det gjenværende av oppløsningen og blandingen ble tilpasset en pH 9. Det organiske lag ble separert og kloroform ble fordampet under redusert trykk. De resulterende krystaller ble oppsamlet ved filtrering for å gi etyl 5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl)-1,4-dihydro-4-oksoquinolin-3-karboksylat, smp. 132-134°C.
De kjemoterapeutiske aktiviteter av forbindelsen ifølge oppfinnelsen er vist i eksemplene 8-11 nedenfor. De under-søkte forbindelser omfatter: Forbindelse 1: 5-amino-l-cyklopropyl-6,8-difluor-7-(3-metyl-l-piperazinyl)-1,4-dihydro-4-oksoquinolin-3-karboksylsyre,

5-amino-l-etyl-6,8-difluor-7-(l-piperazinyl)-1,4-dihydro-4-oksoquinolin-3-karboksylsyre.
EKSEMPEL 8
Den antibakterielle aktivitet in vitro er vist i tabell 1. Tallene i tabellen viser minimale inhibitoriske konsentrasjoner (MIC) (pg/ml), utregnet for fri base. Minimal inhibitorisk konsentrasjon ble bestemt ved 2 ganger agar-fortynningsmetoden som ble anbefalt av Japan Society of Chemotherapy (Chemotherapy, 29(1), 76(1981) ), ved å bruke Muller-Hinton agar. En løkke av en overnattningskultur av testorganismen i Muller-Hinton buljong ble inokulert i 10 ml droge inneholdende agar lag i petri skåler. Bakterieller inokulater inneholdt ca. IO<6> kolonidannende enheter Bakteriell vekst ble observert etter 20 timers inkubering ved 37°C. MIC ble definert som laveste drogekonsentrasjon som forhindret synlig bakteriell vekst.
EKSEMPEL 9
In vivo effektivitet mot systemiske infeksjoner i mus er vist i tabell 2.
Forbindelsene ble hver oppløst i deionisert vann. Hver av oppløsningene ble tilført oralt til musene infisert med hver av forsøksorgansimene under betingelsene vist nedenfor og den midlere effektive dose (ED50) ble regnet ut ved prøveanalyse. Tallene i tabellen viser ED50 (mg/kg) verdien, utregnet for fri base.
Eksperiment betingelser:
Mus. Hanmus (ddy-S) vekt ca. 20 gram.
Infeksjon:
Staphylococcus aureus 50774
Intravenøs infeksjon med 5 x IO<8> celler pr. mus suspendert i fysiologisk saltvann.
Streptococcus pneumoniae 1 Neuteld
Intraperetoneal infeksjon med 3 x 10<3> celler pr. mus suspendert i hjerne-hjerte infusjonsbuljong.
Streptococcus pvo<g>enes A65
Intraperetoneal infeksjon med 3 x IO<7> celler pr. mus suspendert i hjerne-hjerte infusjonsbuljong.
Pseudomonas aeruginosa 12
Intraperetoneal infeksjon med 5 x IO<3> celler pr. mus suspendert i tryptosoyabuljong med 4 % mucin.
Tilførsel:
Fire ganger, umiddelbart 6, 24 og 30 timer etter infeksjon i tilfelle med Streptococcus pnemoniae 1. To ganger umiddelbart og 6 timer etter infeksjon i tilfelle med andre organismer.
Observasj on:
I 14 dager i tilfelle med Staphylococcus aureus 50774 og Streptocuccuc pneumoniae 1 Neufeld. I 7 dager i tilfelle med andre organismer.
EKSEMPEL 10
Anti-Mykoplasma aktiviteten til forbindelse 1 er vist i tabell 3. Tallene i tabellen viser minimum inhibitorisk konsentrasjon (MIC) ( g/ml), utregnet for fri base. Minimal inhibitorisk konsentrasjon ble bestemt ved to ganger agar-fortynningsmetoden ved å bruke Chanok agar. Tre av en kultur av forsøksorganismer i Chanok buljong ble inokulert i 10 ml droge-inneholdene agar lag i petri-skåler. Vekst av Mycoplasma ble observert etter inkubering ved 37°C under betigelsene vist nedenfor. MIC ble definert som laveste droge-konsentrasjon som forhindret vekst av Mycoplasma mikroskopisk.

Mycoplasma pneumoniae:
I 7 dager aerobisk.
Mycoplama arginini og Acholeplasma Laidlawii; I 2 dager aerobisk.

I 3 dager aerobisk.

EKSEMPEL 11
Antibakteriell aktivitet av forbindelse 1 mot Campvlobacter nejuni er vist i tabell 4. Tallene i tabellen viser minimal inhibitorisk konsentrasjon (MIC) ( g/ml), utregnet for fri base. Minimal inhibitorisk konsentrasjon ble bestemt ved 2 ganger agar-fortynningsmetoden ved å bruke blod-inneholdene Mueller-Hinton agar. En løkke av en kultur av forsøks-organismer i Mueller-Hinton buljong ble inokulert i 10 ml drogeinneholdende agar lag i petri-skåler. Bakteriell vekst ble observert etter 48 timers inkubering ved 37°C mikroaero-bisk. MIC ble definert som laveste drogekonsetrasjon som forhindret synlig bakteriell vekst.
EKSEMPEL 12 (Akutt toksisitet)
En oppløsning inneholdende hver av forbindelsene 1, 2 og 3 ifølge oppfinnelsen i forskjellige konsentrasjoner ble gitt oralt til han-mus (ddY) ved en dose på 0,1 ml pr. 10 gram kroppsvekt. Antal døde mus ble tellet etter 7 dager og verdien til median lethal dose (LD5O, mg/kg) ble regnet ut i samsvar med Behrens-Kaerber metoden. Resultatene er vist i tabell 5.
Fra resultatene vist i tabell 3, observeres at forbindelsene 1, 2 og 3 ifølge oppfinnelsen har lav oral toksisitet.
Det ble foretatt ytterligere sammenligningsforsøk angående in vitro antibakteriell aktivitet og antimykoplasma-aktivitet mellom forbindelser angitt i NO patent 162.236 (Ref. 1), EP patent 140.116 (Ref. 2), EP patent 167.763 (Ref. 3), EP patent 178.388 (Ref. 4) og. forbindelsene fremstilt ifølge foreliggende oppfinnelse. Resultatene er angitt i tabell I og II.

1. Analogifremgangsmåte ved fremstilling av terapeutisk aktive quinolinderivater av formel (Ia)

hvor R er hydrogen eller en metylgruppe og Y er hydrogen eller C1-C4-alkyl,
eller et farmasøytisk akseptabelt salt derav, karakterisert ved(i) å omsette en forbindelse av formel (Ila)
hvor X er halogen og Y er som angitt ovenfor,
med et piperazin-derivat av formel (Illa)
hvor R er hydrogen eller en metylgruppe;
hvor R<1> er H eller en beskyttelsesgruppe,
hvoretter eventuelle beskyttelsesgrupper avspaltes for å oppnå forbindselser med formel (Ia)
JO (ii) å omsette en forbindelse av formel (IVa)
hvor X er halogen og R og Y er som angitt ovenfor,
med ammoniakk; eller (iii) å fjerne de beskyttende grupper i en forbindelse av
formel (Va)
hvor W er en for aminer vanlig benyttet beskyttelsesgruppe, og R og Y er som angitt ovenfor;
