Description
Foreliggende oppfinnelse angår fremstilling av nye, farmakologisk aktive, substituerte imidazolylfenyl-amidiner
som er H2-reseptor-blokkerende midler som hemmer mavesyre-sekresjon, og som er nyttige anti-sårmidler.
Eftersom de ordinære antihistamin.-midler er ineffektive
når det gjelder å blokkere den stimulerende virkning histamin har på mavesyresekresjonen, er søking efter histamin-antagonister som kan påvirke denne respons, intensivert. Denne virkningen til histamin, kjent som en kraftig agonist for mavesyre-
sekresjon, formidles gjennom H2~reseptorer (Black et al,
Nature 236, 385, 1972) og hemmes ikke av de klassiske anti-histaminer, H-^-reseptor-blokkere (Ash og Schild, Brit.
J. Pharmacol., 27, 427, 1966). Undersøkelsene innen dette felt kulminerte med syntesen av den klasse forbindelser (G.J. Durant et al, J. Med. Chem. 20, 901, 1977) som burimamid, den første klinisk effektive H2-reseptor-antagonist, er et typisk eksempel på. Selv om burimamid farmakologisk var tilstrekkelig selektivt, syntes det ikke å være tilstrekkelig tilgjengelig for organismen ved oral administrering. Metiamid, en senere vurdert H2~antagonist, viste seg å være kraftigere enn burimamid, og oralt virksomt i mennesker. Imidlertid kunne metiamid ikke anvendes terapeutisk på grunn av dets giftige bivirkninger (agranulocytose). Cimetidin, en forbindelse beslektet med metiamid, som har en cyanoguanidino-gruppe istedenfor en tioureido-gruppe, viste seg å være en like effektiv H2~antagonist som metiamid, men uten de toksiske bivirkningene til metiamid. Cimetidin er i det senere anvendt terapeutisk som et antisårmiddel. Imidlertid er halveringstiden relativt kort, og administrering av flere daglige doser på 200-300 mg er nødvendig. Denne ulempe har tilskyndet videre forskning innen området H2-reseptor-blokkere,
med det som mål å finne forbindelser med lenger virkningstid og/eller med kraftigere virkning. Nylig er to nye H2~reseptor-antagonister beskrevet [ranitidin (AH 19065) og tiotidin (ICI 125.225)] som har kjemiske trekk analoge med cimetidin,
dvs. en lineær metyltioetyl-sidekjede som bærer nøytrale polare grupper. Sammenlignet med cimetidin, består den vesentlige kjemiske forskjellen hos disse to forbindelsene i at imidazol-ringen er erstattet av henholdsvis en aminoalkyl-furan-ring
og en 2-guanidino-tiazol-ring. Både ranitidin (Bradshaw et al, Brit. J. Pharmacol. 66, 464 P, 1979) og tiotidin (T.O. Yellin, Life Sei., 25, 2001-9, 1979) er angitt å være kraftigere enn cimetidin enten som H2~reseptor-antagonister ved in vitro-undersøkelser eller som mavesyresekresjons-inhibitorer in vivo.
Vi har nu funnet at ved å erstatte den lineære sidekjede
som finnes både i de klassiske såvel som i de to nyere H2~reseptor-antagonister, med en fenylring som bærer eventuelt substituerte amidino-grupper, oppnås nye forbindelser med kraftig P^-blokkerende virkning, som hemmer utskillelse av mavesyre.
Ved siden av å være nyttige som midler for å hemme mavesyresekresjonen, er I^-reseptor-antagonistene terapeutisk virksomme ved behandling av betennelser (Yoshioka T, Monafo W.W, Ayvazian et al (Am. J. Surg. 136, (1981)] og kardiovaskulære forstyrrelser [Jerome P., Trzechiakowski og R. Levi (J. Pharmacol. Exp. Ther. 214, 629-634, (1980)]. Ved noen tilstander ville
det også være nyttig med en kombinasjon av H^- og H^-antagonist-aktivitet [Barry L. Tepperman, Eguene D. Jacobson et al (Life Sciences, vol. 24, 2301-2308 (1979))]. Noen av forbindelsene fremstilt ifølge oppfinnelsen oppviser begge disse reseptor-påvirkningene.
I henhold til oppfinnelsen fremstilles forbindelser med
den generelle formel I
hvor R, og R3, som kan være like eller forskjellige, betyr et hydrogenatom eller en lavere alkylgruppe, og
R2 betyr en lineær eller forgrenet C^_g alkyl-, C2_5 alkenyl-eller C.,. alkynyl-gruppe, en cyanogruppe, en hydroksylgruppe, en substituert eller usubstituert cykloalkyl- eller C2_g-cykloalkyl-(C^_2)-alkylgruppe, hvor substituenten er en C^_2 alkylgruppe, en norbornyl-gruppe, en substituert eller usubstituert benzyl- eller fenylgruppe hvor substituenten er halogen, metyl, metoksy eller metylendioksy-grupper, eller en 5-leddet heterocyklisk C^_2~alkyl- eller heterocyklisk gruppe som kan inneholde 1 eller 2 heteroatomer valgt fra oksygen og nitrogen,
og ugiftige syreaddisjonssalter derav.
"Ugiftige" betegner her salter som er dannet med syrer (enten uorganiske eller organiske) hvis anioniske del er fysiologisk godtagbar i de doser saltene blir administrert; salter dannet med saltsyre, maleinsyre, fumarsyre, metansulfonsyre, sitronsyre, vinsyre og eddiksyre.
Det skal forstås at for enkelhets skyld refereres det
i beskrivelsen enten til forbindelsene som baser eller til de tilsvarende salter.
Det skal forstås at selv om dobbeltbindingen i amidin-radikalet og i imidazol-ringen er inntegnet i en spesiell stilling, er forskjellige andre tautomere former mulige, og foreliggende oppfinnelse omfatter fremstilling også av de tautomere formene.
Når R, R^ og/eller Rj i forbindelsene med formel I betyr lavere alkyl, kan disse være alkylgrupper inneholdende fra 1 til 3 karbonatomer, f.eks. metyl.
I forbindelsene med formel (I) kan amidin-radikalet
være i orto-, meta- eller para-stilling i benzenringen. Foretrukne forbindelser fremstilt i henhold til
oppfinnelsen er de hvor amidin-radikalet er i para-stilling i benzenringen, R, R^ og er hydrogenatomer og R2 er hydroksy, metyl i etyl, n-propyl, isopropyl, n-butyl, isobutyl, sek.butyl, t.butyl, neopentyl, heksyl, heptyl, oktyl,
allyl, a-metylallyl, prenyl, propargyl, a-metyl-propargyl,
cyano, norbornyl, benzyl, cyklopropyl, cyklopropylmetyl, dimetylcyklopropylmetyl, mentyl, cykloheksyl, cykloheksylmetyl, fenyl, p-klorfenyl, p-metylfenyl, p-metoksyfenyl, metylendioksyfenyl eller 2-furylmetyl og de ugiftige syreaddisjonssaltene derav (som definert ovenfor). ,
Forbindelsene med den generelle formel I kan f.eks. fremstilles ved følgende fremgangsmåter.
A. Et iraidazolylfenylamin med formel II
hvor R er som angitt ovenfor, omsettes med et reaktivt derivat av karboksamidet med formel III
hvor R^ og R2 er som angitt ovenfor, X betyr anionet av en uorganisk syre så som klorid eller fluorborat, og A betyr en benzoyloksygruppe, klor eller en etoksygruppe.
Om ønsket kan forbindelsen med formel
III også omsettes i baseform. Omsetningen utføres vanligvis ved en temperatur mellom 0 og 100°C, fortrinnsvis mellom 20 og 60°C. Omsetningen utføres fordelaktig i nærvær av et inert organisk oppløsningsmiddel. Egnede oppløsningsmidler omfatter f.eks. en alkohol med 1 til 3 karbonatomer, så som metanol eller etanol, halogenerte hydrokarboner så som diklormetan, dioksan eller aceton.
Forbindelsene med formel III anvendt som utgangsmateriale ved fremgangsmåten ovenfor, kan fremstilles ved vanlige metoder ved at et karboksamid med formel IV hvor og R2 er som angitt ovenfor, omsettes med benzoylklorid, trietyloksonium-fluorborat, etylklorformiat, fosfor-oksyklorid eller fosforpentaklorid.
B. Et N-(imidazolylfenyl)imidat med formel V
hvor R og R^ er som angitt ovenfor, og Y betyr en metyl- eller etyl-gruppe, omsettes med et amin med formel VI
hvor R2 er som angitt ovenfor.
Omsetningen utføres hensiktsmessig i nærvær av et inert, organisk oppløsningsmiddel. Egnede oppløsningsmidler omfatter f.eks. alkoholer, så som metanol, etanol eller dioksan. Imidlertid kan omsetningen utføres i fravær av et oppløsnings-middel. Overskudd av aminet med formel VI kan anvendes som oppløsningsmiddel. Omsetningen utføres vanligvis ved en temperatur fra 20 til 80°C. Om ønsket kan omsetningen ovenfor utføres i ett trinn ved at et amin med formel II omsettes med en forbindelse med formel VII
hvor R^ og Y er som angitt ovenfor, i nærvær av en uorganisk syre, f.eks. svovelsyre, som katalysator. Omsetningen utføres ved en temperatur mellom 60 og 120°C. Efter noen få timer settes det passende amin med formel VI til reaksjonsblandingen.
Forbindelsen med den generelle formel V anvendt som utgangsmateriale, kan fremstilles ved metoder kjent fra litteraturen, f.eks. ved at et amin med formel II omsettes med en forbindelse med formel VII uten oppløsningsmiddel, idet alkoholen som
dannes under reaksjonen avdestilleres.
C. Et N,N-disubstituert kafboksamid-dialkylacetal med formel VIII
hvor R^, R2, R^ og Y er som angitt ovenfor, omsettes med et amin med formel II. Omsetningen utføres ved en temperatur mellom 20 og 80°C, og alkoholen som dannes under reaksjonen avdestilleres. D. En N,N<1->disubstituert amidin-forbindelse med formel IX hvor R og R^ er som angitt ovenfor, og B betyr en cyano-, acetyl-, karbetoksy- eller karbamoylgruppe, omsettes med et amin med formel (VI). Omsetningen utføres hensiktsmessig i nærvær av vann eller et inert, organisk oppløsningsmiddel som f.eks. en alkohol, så som metanol eller etanol, formamid, dioksan eller acetonitril. Omsetningen kan f.eks. utføres ved en temperatur fra 10 til 50°C, fortrinnsvis ved romtemperatur. Forbindelsene med formel IX anvendt som utgangsmateriale ved omsetningen ovenfor kan fremstilles ved fremgangsmåter som i og for seg er kjent fra litteraturen, f.eks. ved at et amin med formel II omsettes med et N-substituert etyl-imidat med formel X
hvor R^ og B er som angitt ovenfor, eller, når B i forbindelsen med formel IX betyr en cyanogruppe, kan omsetningen eventuelt
utføres i ett enkelt trinn ved at et amin med formel II omsettes med cyanamid i nærvær av en forbindelse med formel VII hvor Y og er som angitt ovenfor.
Omsetningen utføres vanligvis i nærvær av et egnet inert, organisk oppløsningsmiddel, som f.eks.»en lavere alkohol, etere, etylacetat, acetonitril eller dioksan, eller uten oppløsningsmiddel, ved en temperatur fra 20 til 80°C. Forbindelsen med formel X kan fremstilles ved vanlige metoder.
E. Et N,N'-disubstituert sulfinylamidin med formel XI
hvor R og R., er som angitt ovenfor, omsettes med en svak organisk syre, f.eks. maursyre, eddiksyre eller propionsyre.
N,N'-disubstituert sulfinylamidin med formel XI kan fremstilles ved vanlige metoder, f.eks. ved at det tilsvarende tiourinstoff oksyderes ved hjelp av hydrogenperoksyd i nærvær av lavere alkohol ved temperaturer mellom 20 og 50°C.
F. 1-(N-alkyliminoformyl)-imidazol-mellomproduktet med formel

hvor R2 er som angitt ovenfor, som ikke er isolert og er oppnådd in situ ved omsetning av et isonitril med formelen C ^. N-R2
med imidazol i nærvær av AgCl i henhold til T. Saegusa et al (Tetrah. Letters 1286 (1974)), omsettes med et amin med formel II, eller et amin med formel II omsettes direkte med et isonitril med formelen C f. N-R2 i nærvær av CuCl. Omsetningen kan utføres uten oppløsningsmiddel, eller i nærvær av et inert, organisk oppløsningsmiddel, som f.eks. etanol eller dioksan.
Omsetningen utføres ved temperaturer mellom 100 og 150°C.
Forbindelsene med formel I fremstilt i henhold til fremgangsmåtene A til F, kan derefter omdannes til de ugiftige syreaddisjonssaltene derav med uorganiske eller organiske syrer som angitt ovenfor, f.eks. ved vanlige metoder, som f.eks. ved at forbindelsen i baseform omsettes med en oppløsning av den tilsvarende syre i et egnet oppløsningsmiddel.
Særlig foretrukne syrer omfatter f.eks. saltsyre, maleinsyre, fumarsyre og metansulfonsyre. De oppnådde salter er normalt oppløselige i vann. Som angitt ovenfor er de nye forbindelsene med den generelle formel I og de fysiologisk godtagbare syre-addis jonssalter derav I^-reseptor-blokkerende midler som hemmer mavesyre-sekresjon. Forbindelsene med den generelle formel I hvor amidin-radikalet er i para-stilling i benzen-ringen,
R, R^ og R^ er hydrogenatomer og R2 er hydroksy, metyl, etyl, n-propyl, isopropyl, n-butyl, isobutyl, sek.butyl, t-butyl, neopentyl, heksyl, heptyl, oktyl, allyl, a-metylallyl, prenyl, propargyl, a-metylpropargyl, cyano, norbornyl, benzyl., cyklopropyl, cyklopropylmetyl, dimetylcyklopropylmetyl, mentyl, cykloheksyl, cykloheksylmetyl, fenyl, p-klorfenyl, p-metylfenyl, p-metoksyfenyl, metylendioksyfenyl eller 2-furylmetyl, og de fysiologisk godtagbare syreaddisjonssalt derav har generelt en bedre virkning og er derfor foretrukne forbindelser som antisårmidler for behandling av sykdommer i mave-tarm-kanalen.

Antagonistaktiviteten på histamin H2-reseptorer vises
enten in vitro eller in vivo ved at forbindelsene hemmer H2-avhengige biologiske virkninger, omfattende henholdsvis histamin-fremkalt positiv kronotropisk virkning og histamin-fremkalt mavesyresekresjon.
Den positive kronotropiske virkning er undersøkt på
isolert marsvin-forkammer suspendert i et organbad (50 ml) inneholdende oksygenert (02: 95% - C02: 5%) Krebs-Henseleit oppløsning (pH 7,4) holdt ved 32 C. Det myokardiale preparat påsatt 1 g isometrisk spenning, får stabiliseres i 60 minutter,
de myokardiale sammentrekninger opptegnes gjennom en isometrisk arm forbundet til en deformasjonsmåler og øyeblikkshastigheten overvåkes med et kardiotachymeter og en varmeskrivende ut-skriver. Efter at kontroll-respons (tachykardi) overfor histamin (10 ^ g/ml) er målt tre ganger med 30 minutters mellomrom, blir prøveforbindelsene satt til badet til den ønskede endelige konsentrasjon, og forkammeret utsettes igjen for histamin-påvirkning. Den kronotropiske respons oppnådd i nærvær av antagonisten sammenlignes derefter med kontroll-responsen for histamin, og prosent reduksjon i den histamin-H2_ fremkalte respons beregnes. Den gjennomsnittlige effektive konsentrasjon (EC5Q) for H2~antagonistene beregnes også ved standard metoder i henhold til Dr. Waud, Analysis of dose-

Noen av forbindelsene med formel I er funnet også å
være effektive for å hemme histamin-fremkalt krampe som hemmes av klassiske H^-antihistaminer, så som difenydramin og pyrilamin, i isolert marsvin-ileum. Evnen prøveforbindelsene har til å hemme histamin-fremkalt mavesyresekresjon er undersøkt efter intravenøs eller intraduodenal administrering til mave-perfuserte rotter, i henhold til Gosh og Shild (Br. J. Pharmacol. Chemother. 13, 5_4, 1958).
Dyrene blir bragt i narkose (uretan, 1 g/kg i.p.) ved konstant temperatur, og et polyetylenrør (PE 50) innføres og festes i spiserøret og i pylorus-antral-området. Efter at maven er vasket for å fjerne gjenværende mat, blir perfusjon av maven utført først med saltoppløsning, 0,5 ml»min ^ (37°C), ved hjelp av en Jobling peristaltisk pumpe. Efter perfusering i 30 minutter ble mave-perfusatet oppsamlet i prøver hvert 30. minutt og titrert på syreinnholdet, uttrykt som pekv. av IN NaOH. Når syre-kontrollverdien er konstant, begynner man intravenøs perfusjon med histamin (1 mg/kg/time), og denne opprettholdes gjennom hele forsøksperioden. Efter at syre-utskillelsen har nådd et jevnt høyere nivå administreres økende doser av prøveforbindeIsene intravenøst for å oppnå en dose-respons-funksjon. ED5q kle derefter beregnet ved standard metoder. Forbindelsene undersøkt ved denne metode viste meget kraftig antisekretorisk virkning ved intravenøs administrering av 100 Mg/kg eller mindre, idet resultatene er angitt i den følgende tabell II:
Den aktutte toksisitet av særlig foretrukne forbindelser med den generelle formel I ble bestemt tilnærmet ved oral administrering av en enkelt dose til grupper på 5 sveitsiske mus som var fastet 18 timer før forsøket. Beregning av antall dødsfall ble foretatt 14 dager' efter administreringen. Resultatene er angitt i den følgende tabell hvor verdiene er beregnet for forbindelsene i baseform.
Det kan fremstilles farmasøytiske preparater som inne-holder som aktiv bestanddel minst én forbindelse med formel I som angitt ovenfor eller et fysiologisk godtagbart syre-addis jonssalt derav, sammen med en farmasøytisk godtagbar bærer eller eksipiens. For farmasøytisk administrering innføres forbindelsene med formel I eller de fysiologisk godtagbare salter derav i konvensjonell farmasøytiske preparater, enten i fast eller flytende form. Preparatene kan f.eks. være i en form som er egnet for oral eller parenteral administrering. Foretrukne former omfatter f.eks. kapsler, tabletter,
belagte tabletter og ampuller.
Den aktive bestanddelen kan blandes i eksipienser eller bærere som vanligvis anvendes for farmasøytiske preparater,
som f.eks. talk, gummi arabicum, laktose, gelatin, magnesium-stearat, maisstivelse, vandige eller ikke-vandige bærere. Preparatene fremstilles fordelaktig i enhetsdoser, idet hver enhetsdose tilpasses slik at den gir en bestemt dose av den aktive bestanddel. Hver enhetsdose kan hensiktsmessig inneholde 10 til 500 mg og særlig foretrukket 20 til 150 mg.
De følgende eksempler illustrerer noen av forbindelsene fremstilt ifølge oppfinnelsen.
Eksempel 1
5- metyl- 4-( 3- aminofenyl)- 1- H- imidazol
5-metyl-4-(3-nitrofenyl)-1-H-imidazol [R. Morgenstern
et al, Pharmazie 30, 103 (1975)] (84 g) oppløst i metanol ble hydrogenert i nærvær av Pd/C, 5% (0,8 g) ved atmosfærisk trykk og romtemperatur. Da den beregnede mengde hydrogen var tatt opp ble katalysatoren frafiltrert og oppløsningen inndampet til tørrhet for å gi 62,5 g av den ønskede forbindelse.

Eksempel 2
(a) 5- metyl- 4-( 4- nitrofenyl)- 1- H- imidazol
5-metyl-4-fenyl-l-H-imidazol i form av nitratsaltet
[R. Morgenstern et al. Pharmazie 30, 103 (1975)] (50 g) ble satt porsjonsvis til 96% H2S04 (100 ml). Den således erholdte tykke oppløsningen ble oppvarmet ved 100°C i 2 timer, fortynnet med vann (1000 ml) og igjen oppvarmet til 70°C i 20 timer. Efter avkjøling ble oppløsningen nøytralisert med 30% NaOH og 17% Na^O^ som vandige oppløsninger. Det faste bunnfall (en blanding av o- og p-nitro-derivatet) ble frafiltrert og omdannet til nitratsaltet ved hjelp av 30% salpetersyre.
Den således erholdte blanding av saltene av de to o- og p-nitro-isomerene ble omkrystallisert fra vann for å gi den rene para-isomeren. Nøytralisering av en vandig oppløsning av para-nitro-derivatet ga 30 g 5-metyl-4-(4-nitrofenyl)-1-H-imidazol som base, tilstrekkelig ren til å bli anvendt i neste trinn. Sm.p. 190-191°C.

Til en oppløsning av N-metylformamid (2,36 g) i vannfri dietyleter (10 ml) ble satt langsomt ved 0°C en oppløsning av benzoylklorid (5,62 g) i vannfri dietyleter (30 ml). Efter 1 times omrøring ble det hvite, faste stoff som var dannet, frafiltrert og øyeblikkelig satt til en suspensjon av 4-(4-aminofenyl)-1-H-imidazol (1,59 g) i dioksan (50 ml). Blandingen ble omrørt natten over og derefter inndampet til tørrhet. Residuet ble tatt opp i vann, oppløsningen ble gjort basisk med 10% NaOH til pH 10 og det utfelte produkt ble ekstrahert med etylacetat. Den organiske oppløsning ble tørret (MgS04) og inndampet til tørrhet for å gi 0,75 g urenset amidin som ble omkrysallisert fra aceton. Sm.p. 189-190°C.

Blandingen ble omrørt natten over og derefter inndampet til tørrhet. Fluorboratsaltet av N-metyl-N"-[4-(imidazol-4-yl)-fenyl]-formamidin ble oppløst i vann og oppløsningen gjort basisk med 10% NaOH til pH 10.
Produktet som utkrystalliserte, ble oppsamlet ved filtrering
for å gi 1,4 g av tittel-formamidinet. Sm.p. 189-190°C.

N-fenyl-N1 -[4-(imidazol-4-yl]-formamidin utkrystalliserte
som acetatsaltet som ble filtrert for å gi 4,6 g av tittelproduktet. Sm.p. 123-124°C.

N- metyl- N'-[ 4-( imidazol- 4- yl)- fenyl]- formamidin ( Forbindelse 5)
En blanding av 4-(4-aminofenyl)-1-H-imidazol (1,59 g)
og trietyl-ortoformiat (2,22 g) ble oppvarmet i 30 minutter ved" 100°C i nærvær av katalytiske mengder 96% H2S04- En opp-løsning av metylamin 33% i etanol (9,5 g) ble derefter satt
dråpevis til den avkjølte reaksjonsblandingen. Oppløsningen ble omrørt ved romtemperatur i 20 timer og derefter inndampet til tørrhet. Det erholdte residuum ble oppløst i 10% saltsyre, og den sure oppløsningen ble vasket med etylacetat og derefter gjort basisk med 10% NaOH til pH 10. Det utfelte produkt ble ekstrahert med etylacetat for å gi, efter avdampning av oppløsningsmidlet, 0,45 g av Forbindelse 5. Sm.p. 188-189°C.

En oppløsning av 4-(4-aminofenyl-l-H-imidazol (2,3 g)
og metylisotiocyanat (1,15 g) i 30 ml etanol ble tilbakeløps-behandlet i 0,5 time og inndampet til tørrhet. Den således erholdte oljeaktige base ble renset ved at den ble overført til det tilsvarende hydrokloridsalt ved at gassformig hydroklorid ble boblet gjennom i isopropanol. Det ønskede tiourinstoff ble erholdt som et hvitt, fast stoff (2 g). Sm.p. 210-211°C. Analyse: C^H^ClN.S

av 30 minutter til en omrørt, isavkjølt oppløsning av N-metyl-N'-[4-(imidazol-4-yl)-fenyl]tiourinstoff (1,16 g) i 5 ml metanol. Efter 2 timers omrøring ble blandingen inndampet til tørrhet,
og det således oppnådde residuum ble krystallisert fra etanol for å gi 0,95 g av den ønskede forbindelse.

c) N- metyl- N'-[ 4-( imidazol- 4- yl)- fenyl]- formamidin ( Forbindelse 5)
N-metyl-N'-[4-(imidazol-4-y1)-fenyl]-formamidin-sulfinsyre (0,9 g) ble oppløst i 15 ml iseddik og tilbakeløpsbehandlet i 1 time. Den konsentrerte oppløsning ble gjort basisk med 10% NaOH til pH 10 og ekstrahert med etylacetat. Den organiske oppløsning ble inndampet til tørrhet for å gi 0,3 g N-metyl-N * -[4-(imidazol-4-yl)-fenyl]rformamidin. Sm.p. 187-189°C.
Eksempel 11
N- t. butyl- N'-[ 4-( imidazol- 4- yl)- fenyl]- formamidin ( Forbindelse 21)
En blanding av 4-(p-aminofenyl)-1-H-imidazol (2,3 g), t-butyl-isonitril (1,25 g), AgCl (0,2 g) og imidazol (0,11)
ble oppvarmet ved 100°C i 13 timer. Den tykke reaksjonsblandingen ble behandlet med 10% HC1 og etylacetat. Den sure oppløsningen ble regulert til pH 6,5 med 10% NaOH, behandlet med trekull, filtrert og gjort fullstendig basisk. Det utskilte lag ble ekstrahert inn i etylacetat. Efter tørring ble den organiske oppløsning inndampet for å gi 0,38 g av tittel-forbindelsen. Fumaratsalt. Sm.p. 192-194°C.

N- cykloheksyl- N'-[ 4-( imidazol- 4- y1)- fenyl]- formamidin
( Forbindelse 29)
En blanding av 4-(p-aminofenyl)-1-H-imidazol (1,59 g), cykloheksyl-isonitril (1,2 g) og CuCl (0,99 g) ble oppvarmet ved 150°C i 3 timer. Den tykke reaksjonsblandingen ble ekstrahert i metanol. Metanoloppløsningen ble efter behandling med trekull, inndampet til tørrhet og ekstrahert inn i etylacetat og 10% saltsyre. Den sure oppløsningen ble gjort basisk med 17% Na2C03 til pH 7,5 og ekstrahert i etylacetat.
Den organiske oppløsningen ble inndampet til tørrhet. Residuet. ga efter omkrystallisering fra aceton 0,25 g av tittelproduktet. Sm.p. 220-221°C.
Fumaratsalt, sm.p. 139-141°C.
Claims (2)
1. Analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser med den generelle formel I
hvor R, R^ og R^, som kan være like eller forskjellige, betyr et hydrogenatom eller en lavere alkylgruppe, og
R2 betyr en lineær eller forgrenet C^_g-alkyl-, C2_^-alkenyl-eller C-^^-alkynyl-gruppe, en cyanogruppe, en hydroksylgruppe,
en substituert eller usubstituert cykloalkyl- eller Cj , — oQ-cykloalkyl-(C1_2)-alkyl-gruppe, hvor substituenten er en C1_3~alkylgruppe, en norbornyl-gruppe, en substituert eller usubstituert benzyl- eller fenylgruppe hvor substituenten er halogen, metyl, metoksy eller metylendioksy-grupper, eller en 5-leddet heterocyklisk C^_2~alkyl- eller heterocyklisk gruppe som kan inneholde 1 eller 2 heteroatomer valgt fra oksygen og nitrogen,
og ugiftige syreaddisjonssalter derav,
karakterisert ved at a) en forbindelse med formel II
hvor R er som angitt ovenfor, omsettes med en forbindelse med formel III .hvor og R2 er som angitt ovenfor, X~ betyr anionet av en uorganisk syre så som klorid eller fluorborat, og A betyr en benzoyloksygruppe, klor eller en etoksygruppe, b) en forbindelse med formel V
hvor R og er som angitt ovenfor, og Y betyr en metyl- eller etylgruppe, omsettes med et amin med formel VI
hvor R2 er som angitt ovenfor, c) en forbindelse med formel VIII
hvor R^, R2, R3 og Y er som angitt ovénfor, omsettes med et amin med formel II, d) en forbindelse med formel IX
hvor R og R^ er som angitt ovenfor, og B betyr en cyano-, acetyl-, karbetoksy- eller karbamylgruppe, omsettes med et amin med formel VI, e) en forbindelse med formel XI

for fremstilling av N-etyl-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, N-isopropyl-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, N-allyl-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, N-n-propyl-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, N-sek.butyl-N•-[4-(imidazol-4-yl)-fenyl]-formamidin, N-isobutyl-N'-[4-(imidazbl-4-yl)-fenyl]-formamidin, N-prenyl-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, N-cyklopropylmetyl-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, N-cyklopropyl-N[4-(imidazol-4-yl)-fenyl]-formamidin, N-(a-metylallyl)-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, N-neopentyl-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, eller N-(2,2-dimetyl)-cyklopropylmetyl-N'-[4-(imidazol-4-yl)-fenyl]-formamidin, karakterisert ved at man anvender tilsvarende substituerte utgangsmaterialer.
