Farmaceutisch bruikbare dihydropyridinyldicarboxylaatamiden en esters.
De onderhavige uitvinding heeft betrekking op de heterocyclische koolstofverbindingen van de 1,4-dihydropyridine-klasse met een 3-carboxylaat of carboxamidogroep verbonden met een arylpiperazinyl-alkyleengroep. Deze verbindingen bezitten biologische eigenschappen.
5 De laatste tien jaar is een groot aantal publicaties verschenen dat betrekking heeft op verbindingen van de 4-aryl-l,4-dihy-dropyridine-reeks, welke verbindingen calciumantagonist-eigenschappen bezitten en bruikbaar zijn in de behandeling van cardiovasculaire ziekten. De calcium-blokkerende werkingen blijken vaatverwijding te bevorde-10 ren, waardoor deze verbindingen bruikbaar zijn voor de behandeling van angina en hypertensie. Een typisch voorbeeld van deze structuren vormt nifedipine met formule 1 van het formuleblad; chemisch heet deze verbinding 4-(2'-nitrofenyl)-2,6-dimethyl-3,5-dicarbomethoxy-l,4-dihydropyri-dine. Nifedipine en enkele verwante 4-aryl-l,4-dihydropyridines vormen 15 het onderwerp van het op 23 december 1969 gepubliceerde Amerikaanse octrooischrift 3.485.847. Vele latere octrooien zijn verleend, die 1,4-dihydropyridines betreffen waarin andere substituentgroepen op de verscheidene ringposities van de dihydropyridinegroep zijn opgenomen via uiteenlopende chemische verbindingsgroepen.
20 Gebruikmakend van medicinale chemische technieken, was één doel van de onderhavige uitvinding om een therapeutisch middel te ontwikkelen, dat α^-adrenergische blokkerende eigenschappen combineert met de calcium-blokkerende werking in één enkele molecuulstructuur. De biologische reden voor de combinatie van werkingen ligt in de gedachte 25 dat een dergelijk middel een sterke en efficiënte behandeling van vaso-spastische circulatie-afwijkingen zou verschaffen.
De stand van de techniek, die relatie heeft met de reeks verbindingen volgens de onderhavige uitvinding, kan worden gegenerali- 2 4 5 6 seerd met structuurformule 2 van het formuleblad waarin R , R , R en R 30 elk van een aantal substituentgroepen kunnen zijn die eerder in de veelomvattende dihydropyridine-literatuur zijn gedefinieerd; maar waarbij specifieke aandacht wordt geschonken aan de definitie voor de aan de 3- $δΰ ^ δ -v : 2 positie van de 1,4-dihydropyridinering gebonden substituentstructuur.
Tot op heden is niet eerder een aryl- of hetaryl-piperazinylalkylgroep in een 1,4-dihydropyridineringverbinding opgenomen via een carboxylaat-amide of esterfunctionaliteit in de 3-positie van de ring. De meest rele-5 vante literatuur wordt gevormd door de Amerikaanse octrooischriften 3.905.970 en 3.974.275 van Bossert et al., die resp. gepubliceerd zijn op 16 september 1975 en 10 augustus 1976. De in deze octrooischriften beschreven verbindingen hebben als de 3-substituent-zijketen de groep met formule 3, die in formule 2 is getoond, de groep met formule 4.
10 Deze zelfde 3-substituent-zijketen met formule 4 werd ook beschreven in het Amerikaanse octrooischrift 4.393.070 van Sato et al., dat in juli 1983 is gepubliceerd.
De op 21 september 1983 gepubliceerde Europese octrooiaanvrage 88.903 toont 1,4-dihydropyridineverbindingen waarbij de estergroep 15 van het 3-carboxylaatgedeeite formule 5 heeft, waarin n gelijk is aan 0 -5; Z staat voor aryl of hetaryl; en R staat voor lager alkyl, alkoxy-carbonyl, of alkanoylamino. De nieuwheid van deze anti-hypertensieve middelen is gebaseerd op Z, doordat ”de invoering van de aromatische ring of aromatische heterocyclische ring op de α-positie van het cyclische 20 aminoalkylestergedeelte in de zijketen een grotere en aanzienlijk langere effectiviteit veroorzaakt."
Iets minder relevant is de Europese octrooiaanvrage 63.365, die op 27 oktober 1982 is gepubliceerd en 1,4-dihydropyridines toont met een 3-carboxylaatestergroep welke een piperidinering met for-25 mule 6 omvat, waarin n gelijk is aan 0-3, en R staat voor arylalkyl of acyl.
Bovendien zijn 1,4-dihydropyridineverbindingen beschreven die een arylpiperazinesysteem bezitten, dat door een alkyl of alkoxy-alkyl-keten is verbonden met de 2-positie van de dihydropyridinering. Ari-30 tomi et al., in Chem. Pharm. Buil. 29 (11), 3163 - 71 (1980) toont verbindingen met de groep volgens formule 7 aan ringpositie 2, in welke groep n gelijk is aan 2 en R staat voor alkyl, aryl of arylalkyl. De verbinding met formule 8 wordt in de vorm van een voorbeeld expliciet beschreven.
35 De op 22 september 1982 gepubliceerde Europese octrooi aanvrage 60.674 beschrijft anti-ischemische en antihypertensieve middelen 8 5 0 1 0 4 8 * 1 3 met formule 9, waarin R een ethyleen- of propyleenketen voorstelt; en 5 R staat voor -alkyl, aryl, arylalkyl en dergelijke. Deze verbindingen kunnen gemakkelijk structureel worden onderscheiden van de verbindingen volgens de onderhavige uitvinding als gevolg van de ringpositie 5 en de functionele verbindingsgroep. Verbindingen volgens de onderhavige uitvinding bevatten namelijk een α-blokkerend aryl- of hetarylpiperazine-gedeelte dat door een alkyl, alkoxyalkyl, of alkylaminoalkylketen is verbonden met een carboxylaat of carboxamidefunctie aan ringpositie 3.
Al de bovengenoemde verbindingen ontlenen hun therapeu-10 tische bruikbaarheid, volgens de heersende theorie van hun biologisch mechanisme, aan hun inherente vermogen om als calciumkanaal-blokkerings-middelen te werken. In essentie kunnen de verbindingen volgens de uitvinding van de verbindingen volgens de stand van de techniek worden onderscheiden op basis van zowel de molecuulstructuur als op basis van de bio-15 logische werking. De verbindingen volgens de uitvinding bezitten zowel calciumkanaal- als α-adrenergisch blokkerende eigenschappen, waardoor de bruikbaarheid van deze verbindingen in de behandeling van hypertensie en ischemische afwijkingen wordt vergroot. De onderhavige verbindingen blijken ook bruikbare werkingen te bezitten voor de inhibitie van bepaal-20 de functies van bloedplaatjes. Er is niets in de stand van de techniek dat de verbindingen volgens de onderhavige uitvinding anticipeert of suggereert.
De onderhavige uitvinding omvat de verbindingen met formule 10, alsmede de zuuradditiezouten van deze stoffen. In struc- 25 tuurformule 10 hebben de symbolen R^, R4, R~*, R^, X, Y en Z de volgende 2 6 betekenissen. R en R worden onafhankelijk van elkaar gekozen uit lage alkyl, hydroxyalkyl, alkoxyalkyl, alkylaminoalkyl, of dialkylaminoalkyl en kunnen gelijk of verschillend zijn. Lage alkyl betekent tot C^; alkoxyalkyl duidt op een C ^ -alkyleenketen en een via een zuurstofatoom 30 verbonden C^^-alkylgroep; op dezelfde wijze duiden alkylaminoalkyl en dialkylaminoalkyl op lage alkylgroepen en een C^.-alkyleenketen, verbon- 4 den via een secundaire (-NH-) of tertiaire ( > N-) aminogroep. R staat voor cycloalkyl met 5-7 koolstof atomen, bicycloalkenyl met 7-9 koolstof atomen, hetaryl, zoals furanyl, indolyl, methylthiopyridyl, thiênyl, 35 benzoxadiazolyl, benzothiadiazolyl, en dergelijke; aryl betekent fenyl, naftyl, of gesubstitueerd-fenyl, waarbij de substituenten aceetamino, sni·τ λ - λ • * 4 lage alkyl, lage alkoxy, cyano, halogeen, hydroxyl, nitro, trifluorome- thyl, trifluoromethylsulfonyl, en methylsulfonyl en dergelijke omvatten.
5 2 R is R of de groep met formule 3. X staat voor 0 of NH; Y staat voor een lage (C, _) alkyleenketen, alkoxyalkyleen, of alkylaminoalkyleen-1 -b 5 keten; en Z staat voor fenyl, pyridinyl, of pyrimidinyl, al dan niet gesubstitueerd door één of meerdere substituentgroepen, gekozen uit lage alkyl, lage alkoxy, cyano, halogeen, en trifluoromethyl.
Geprefereerde verbindingen volgens de onderhavige uit- 6 vinding hebben de structuur met formule 10, waarin R2 en R lage alkyl 4 5 10 zijn, R nitrofenyl is, R lage alkyl of de groep met formule 3 voorstelt, X staat voor O of NH, Y een ^-alkyleenketen is, en Z gesubstitueerd fenyl voorstelt. De meest geprefereerde verbindingen volgens de on- 2
derhavige uitvinding hebben de structuur volgens formule 10, waarin R
6 4 5 en R methyl zijn, R 2- of 3-nitrofenyl voorstelt, R methyl of de 15 groep met formule 3 is, X staat voor O, Y een propyleenketen voorstelt en Z een 2-gesubstitueerde fenylring, bij voorkeur O-methoxyfenyl is.
De verbindingen volgens de onderhavige uitvinding kunnen als optische isomeren voorkomen en zowel de racemische mengsels van deze isomeren als de individuele optische isomeren zelf vallen binnen de 20 omvang van de onderhavige uitvinding. De racemische mengsels kunnen in de individuele isomeren worden gescheiden via welbekende technieken, zoals de scheiding van de diastereomere zouten, die met optisch actieve zuren worden gevormd, gevolgd door terugomzetting in de optisch actieve basen.
Zoals vermeld heeft de onderhavige uitvinding ook be-25 trekking op de farmaceutisch aanvaardbare niet-toxische zouten van deze basische verbindingen. Dergelijke zouten omvatten de zouten die afgeleid zijn van organische en anorganische zuren zoals, zonder beperking, waterstof chloride , waterstofbromide, fosforzuur, zwavelzuur, methaansulfonzuur, azijnzuur, wijnsteenzuur, melkzuur, barnsteenzuur, citroenzuur, 30 malelnezuur, sorbinezuur, aconietzuur, salicylzuur, ftaalzuur, embonzuur, enanthzuur, en dergelijke.
De verbindingen volgens de onderhavige uitvinding kunnen volgens de volgende werkwijzen worden bereid, waarbij variaties worden toegepast van de Hantzsch synthesereactie, toegepast op de juiste uit-35 gangsmaterialen.

5 bruik van een gemodificeerd Hantzsch-proces voor de bereiding van de verbindingen met formule 10 volgens de nog volgende reactieschema's.
Het algemeen reactieproces en veel van de vereiste tussenprodukten zijn eerder beschreven in het Amerikaanse octrooischrift 4.414.213, waarvan 5 de inhoud door verwijzing hier opgenomen moet worden geacht.
Algemene werkeijzen voor de bereiding van verbindingen met formule 10 zijn op het formuleblad in het algemeenschema weergegeven.

In het algemeenschema hebben R en X de betekenissen 2 6 als gedefinieerd voor formule 10. R' kan zowel R als R van formule 10 IQ voorstellen; R" kan zowel R^ als de groep met formule 15 voorstellen.
De bereiding van de verbindingen met formule 10 volgens de werkwijze die in het algemeen schema is getoond, omvat in het algemeen het samen verwarmen van tussenprodukten van het formule 13-type en het formule 14-type alleen of in tegenwoordigheid van een grote verscheidenheid van reac-15 tie-inerte organische oplosmiddelen. Geschikte oplosmiddelen omvatten, zonder beperking, benzeen, tolueen, tetrahydrof uran, dibutylether, butanol, hexanol, methanol, dimethoxyethaan, ethyleenglycol, ethanol, propanol, enz. Geschikte reactietemperaturen lopen van ongeveer 60 tot 150°C. Gewoonlijk is geen katalysator of condensatiemiddel vereist. De enamine-20 esters of amides met formule 14 worden uit tussenprodukten met formule 12 gevormd door Hantzsch-type reactiecondities (NH^OAc/alcohol). De tussenprodukten met formule 14 (die hierna zullen worden gedefinieerd) worden gewoonlijk niet geïsoleerd maar laten hun direct reageren met verbindingen met formule 13. De acylcinnamaatverbindingen met formule 13 worden in 25 het algemeen bereid onder toepassing van bekende Knoevenagel-condensatie-reactiecondities. In het algemeen werden geschikt gesubstitueerde aldehydes en 1,3-dicarbonylverbindingen gecondenseerd onder vorming van verbindingen met formule 13.
Geprefereerde variaties van het algemene schema worden 30 getoond in de verdere op het formuleblad weergegeven reactieschema's.
In de reactieschema's 1-3 hebben R^,R,R,R^,X, Y en Z de betekenissen zoals gedefinieerd voor formule 10.
Volgens schema 1 wordt een 1,3-dicarbonyl-tussenprodukt met formules 17 of 24, gevormd door behandeling van hetzij een chloro-35 alkanol met formule 16 of een arylpiperazinylalkanol met formule 23 met hetzij Meldrum's zuur (zie The Merck Index, 10de editie, 5635, blz. 828 350 f ü ♦ * 6 (1983)) als in schema 4, hetzij met diketeen, onderworpen aan gemodificeerde Hantzsch-condensatiecondities (ammoniumacetaat/ethanol) gevolgd door een reactie met het cinnamaat-tussenprodukt met formule 19 om hetzij een gewenst produkt met formule 10 te verkrijgen zoals in schema 1B, 5 hetzij de chloroalkylester-verbinding met formule 20, getoond in schema IA te verkrijgen. Het tussenprodukt met formule 20 kan in reactie worden gebracht met hetzij een eenvoudig arylpiperazine met formule 21 hetzij een arylpiperazinylalkylalcohol of amine met formule 22, waarbij verschillende uitvoeringsvormen van 3-carboxylaatester-verbindingen met 10 formule 10 worden verkregen zoals getoond is in schema IA. De dihydro-piridineprodukten met formule 10, die met de in schema 1 weergegeven methode worden verkregen, worden na chromatografie in matige opbrengsten geïsoleerd.
Syntheseschema 2 toont de reactie-sequentie, die gebruikt 15 wordt om de 3-carboxamide-subklasse van produkten met formule 10 te verkrijgen. In schema 2 wordt een aminoalkylpiperazine met formule 25, bereid uit het overeenkomstige arylpiperazine met formule 21 en een chloroalkyl-ftaalimide via de welbekende Gabriel-synthese, behandeld met diketeen of een geschikt alkanoylacetylhalogenide onder vorming van het acetozuurami- 20 de-tussenprodukt met formule 26, dat omgezet wordt in het kaneelzuurami- 4 de met formule 27, onder toepassing van Knoevenagel-condities (R CHO, pipe-idine en azijnzuur in benzeen). Toepassing van de algemene reactie op de tussenprodukten met de formules 27 en 28 levert, zoals getoond, het gewenste produkt met formule 10 waarin X = NH.
25 Schema 3 toont een route die gebruikt wordt voor de synthese van symmetrische dihydropyridines, d.w.z. de 3- en 5-carboxylaat-groepen zijn identiek. Zoals getoond ondergaat het tussenprodukt met formule 29 (bereid uit de verbindingen met formule 24 onder Hantzsch-reac- 4 tiecondities) een condensatie met een geschikt R CHO waarbij het gewenste 30 produkt met formule 10 wordt verkregen.
Additionele reactie-tussenprodukten, die in de bereiding van de verbindingen volgens de onderhavige uitvinding kunnen worden toegepast, kunnen worden bereid volgens de syntheseschema's 4 en 5 of door modificaties daarvan, die aan deskundigen in de chemie duidelijk zul-35 len zijn. Voor meer details over schema 4 wordt verwezen naar Y. Oikawa, et al., J. Org. Chem. 43_, 2087 (1978).

In schema 5 kan R~* ook staan voor de groep met formule 15. Bovendien, wanneer het structuurgedeelte dat aangeduid is als "alkyl" in formule 31, een standaard 0- of N-beschermende groep in organische syntheses zou zijn, zou zijn verwijdering in het algemeen aanleiding geven 2 6 5 tot tussenprodukten waarin R of R alkanol of alkylamino zijn zoals bovenstaand is gedefinieerd. In het algemeen wordt de verbinding met formule 31 bereid door een koude (0°C) oplossing van het gemakkelijk verkrijgbare tussenprodukt met formule 30 te behandelen met lithiumdilsopropyl-amide, gevolgd door introductie van een alkalimetaalzout van het X-alkyΙ-ΙΟ reagens. Voorbeelden van dit reagens zouden natriummethoxyde, natrium-ethoxyde, 2-fenylethylethoxyde, natriumfenoxyde, lithiummethylamide, lithiumdimethylamide, lithiummethylfenylamide en dergelijke zijn. Het reac-tiemedium is een inerte organische vloeistof, bij voorkeur tetrahydro-furan of tetrahydrofuran-DMSO. Isolatie en zuivering van de verbinding 15 met de formule 31 wordt door chromatografie over silicagel gerealiseerd.
Gevonden is dat de verbindingen volgens de uitvinding verschillende bruikbare farmacologische eigenschappen bezitten.
De evaluatie van deze farmacologische eigenschappen werd uitgevoerd door middel van zowel biologische tests in vitro als in vivo. Het in vitro-20 onderzoek omvatte calciumactiviteit in verschillende gladde spiersystemen zoals ratten dorsal-aorta, poortader en trachea; en a-bindings-affiniteiten, bepaald in ratte-hart en hersenen. In het algemeen bezaten de voorkeursverbindingen volgens de onderhavige uitvinding een calciumtoe-treding-blokkerende werkzaamheid met sterkten die die van de vergelij-25 kingsverbinding nifedipine benaderden. De α-bindingsactiviteit was veel groter voor de verbindingen volgens de uitvinding dan voor nifedipine, waarbij de meeste leden van de serie verbindingen volgens de uitvinding één of twee grootteorden sterker waren.
Dosis-verschuivingsonderzoeken, betrekking hebbend op 30 de respons op fenylefrine in ganglion-geblokkeerde, geanesthetiseerde ratten, liet zien, dat α-adrenoceptor-blokkering het farmacologische resultaat van de α-binding was. De methodiek is beschreven door Deitchman et al., in J. Pharmacol. Methods, 3, 311 - 321 (1980).
Het in vivo-onderzoek omvatte vaatverwijdingsresulta-35 ten in de ganglion-geblokkeerde, angiotensine-II ondersteunde rat en een antihypertensief onderzoek in hetzij spontaan hypertensieve ratten (SHR) ” c 010 4 6 * v 8 hetzij DOCA zoutratten. In het algemeen verliep de vaatverwijdende activiteit evenwijdig aan de calcium-blokkerende werkzaamheid bij leden van de reeks verbindingen volgens de uitvinding. De gegevens van het antihypertensieve onderzoek lieten zien dat goede calcium- en a-adrener-5 gische blokkerende werkzaamheid in samenwerking de meest dramatische antihypertensieve effecten gaven. Een meest geprefereerde verbinding, de verbinding BMY 20064 met formule 32 van het formuleblad, werd oraal toegediend aan zowel normotensieve als spontaan hypertensieve, bij bewustzijn verkerende ratten. Doses van 1 en 10 mg/kg veroorzaakten maxi-10 male gemiddelde slagaderlijke bloeddrukverlagingen van 25 resp. 53 mmHg, in de normotensieve ratgroep en 34 resp. 100 mmHg in de spontaan hypertensieve ratgroep. Deze gemiddelde slagaderlijke bloeddrukresponsies, die in beide groepen werden waargenomen, hadden een snelle start (minder dan 10 minuten) en de significant gemiddelde slagaderlijke bloeddruk 15 hield meer dan 4 uur aan na de hogere dosis van BMY 20064.
Bovendien werden BMY 20064 en nifedipine onderzocht in geanesthetiseerde beagle-honden teneinde .hun vergelijkende acute hemo-dynamische effecten vast te stellen. De twee verbindingen vertoonden vergelijkbare sterkte en hemodynamische profielen in dit bijzondere hemo-20 dynamische model.
De specifieke tests die voor het evalueren van de verbindingen volgens de uitvinding werden toegepast, werden volgens de volgende procedures of met kleine modificaties uitgevoerd. De demonstratie dat de onderhavige verbindingen een specifieke blokkerende werking op 25 calcium-ionkanalen bezitten, volgde uit in vitro-onderzoeken die bestonden uit het hangen van cavia ileale longitudinale gladde spierstroken in baden, die Tyrodes-oplossing bevatten, die op 37°C werd gehouden en belucht werd met 95% 0^ - 5% CO^. De weefsels werden gedurende 60 minuten geëquilibreerd voordat alle experimenten werden begonnen. Eén enkele 30 respons op carbachol werd verkregen en in alle experimenten als controle-maximum gebruikt. Tussen achtereenvolgende doses dóór, werden de weefsels opnieuw geëquilibreerd en elke 15 minuten gewassen met Tyrodes-oplossing. Om het effect van de verbindingen te bestuderen, werden de weefsels gedurende 10 minuten aan de antagonist blootgesteld voordat car-35 bachol werd toegevoegd. Voor alle experimenten werd in elk weefsel slechts één antagonist in elke concentratie getest. De resultaten zijn *·· - · j , r-\ 'U 'U >i ij

9 uitgedrukt als molaire concentraties antagonist die 50% inhibitie van de spierrespons geven. Aangezien calcium-antagonisme in het algemeen inhibitie geeft van excitatie-contractiekoppeling in vasculaire gladde spiren, roepen middelen van dit type gewoonlijk vaatverwijding op. Testen van ge-5 selecteerde verbindingen volgens de uitvinding in het ganglion-geblokkeer-de, angiotensine II-ondersteunde ratmodel (Deitchman, et al., J. Pharmacol. Methods, 3, 311 - 321 (1980)) toonde vaatverwijding aan met de daarmee gepaard gaande verlaging van de bloeddruk.
De geselecteerde verbindingen volgens de onderhavige 1.0 uitvinding zijn bovendien in vitro en in vivo onderzocht in laboratoriumproeven, die ontwikkeld werden om het vermogen van een geneesmiddel om hartweefsel te beschermen tegen schade als gevolg van ischemie, te voorspellen. Deze tests maken gebruik van het bekende verband tussen een progressieve uitputting van hoog energetisch fosfaat en het begin van le-15 thale celbeschadiging in ischemisch myocardium. Resultaten van deze onderzoeken tonen aan dat de geselecteerde verbindingen een sterke anti-ischemie-werking bezitten.
Tenslotte vertonen verbindingen volgens de onderhavige uitvinding een sterke inhibitie van verschillende aspecten van bloedplaat-20 jesfuncties. Deze veranderingen zijn niet gerelateerd aan de calcium-blokkerende werking van de verbindingen. Dit blijkt uit tabel A, die de vergelijkende werkingen van BMY 20064 met twee als referentie gebruikte calciumtoetreding blokkerende middelen, nifedipine en verapamil, laat zien.

De bovenstaande discussie van de biologische activeiten samenvattend, bezitten de verbindingen volgens de uitvinding cardiovasculaire eigenschappen, die bijzonder geschikt zijn voor hun toepassing in 5 hypertensie en ischemie. Derhalve heeft een ander aspect van de onderhavige uitvinding betrekking op een werkwijze voor het brengen van verbetering in hypertensie of ischemie in een zoogdier dat aan een dergelijke behandeling behoefte heeft, welke werkwijze een systemische toediening omvat aan een dergelijk zoogdier van een effectieve dosis van een verbinding 10 met formule 10 of een farmaceutisch aanvaardbaar zuuradditiezout daarvan. Op basis van dierproeven is een effectieve orale dosis naar verwachting ongeveer 1 tot 20 mg/kg en een effectieve parenterale dosis naar verwachting lager, in het bereik van ongeveer 0,05 tot 1 mg/kg lichaamsgewicht .
15 Voor klinische toepassingen moeten de dosering en do- seringsvoorschriften in elk geval echter zorgvuldig worden ingesteld, waarbij gebruik wordt gemaakt van een verstandige professionele beoordeling en rekening wordt gehouden met de leeftijd, het gewicht en de conditie van de ontvanger, de toedieningsroute en de aard en ernst van de 20 ziekte. In het algemeen zullen de verbindingen volgens de onderhavige uitvinding op dezelfde wijze worden toegediend als het ter vergelijking dienende geneesmiddel nifedipine en zal de dagelijkse orale dosis ongeveer 5 tot ongeveer 50 mg, bij voorkeur 10 tot 20 mg omvatten, toegediend één tot drie keer per dag. In sommige gevallen kan een voldoende thera-25 peutisch effect worden verkregen bij lagere doseringen, terwijl in andere gevallen grotere doseringen vereist zullen zijn.
De hierin gebruikte term systemische toediening duidt op orale, rectale, en parenterale (d.w.z. intramusculaire, intraveneuze en subcutane) routes. In het algemeen zal worden gevonden dat wanneer een 30 verbinding volgens de onderhavige uitvinding oraal wordt toegediend, hetgeen· de voorkeur heeft, een grotere hoeveelheid reactief middel nodig is om hetzelfde effect te produceren als een kleinere, parenteraal toegediende hoeveelheid. Volgens goed klinisch gebruik heeft het de voorkeur dat de verbindingen volgens de uitvinding worden toegediend in een concen-35 tratieniveau, dat effectieve antihypertensieve en/of anti-ischemische effecten zal geven zonder schadelijke of ongewenste neveneffecten te ver- «5010··· 11 oorzaken.
Therapeutisch gezien worden de verbindingen volgens de uitvinding in het algemeen toegediend in de vorm van farmaceutische samenstellingen, die een effectieve antihypertensieve en/of anti-ischemi-5 sche hoeveelheid van een verbinding met formule 10 of een farmaceutisch aanvaardbaar zuuradditie zout daarvan, alsmede een farmaceutisch aanvaardbare drager omvatten. Farmaceutische samenstellingen voor het uitvoeren van een dergelijke behandeling zullen een overwegende of ondergeschikte hoeveelheid, bijvoorbeeld 95 tot 0,5% van tenminste één verbinding volgens 10 de onderhavige uitvinding bevatten in combinatie met de farmaceutische drager, waarbij de drager één of meerdere vaste, semi-vaste, of vloeibare verdunningsmiddelen, vulstoffen, en formuleringshulpmiddelen omvatten, die niet-toxiscn, inert en farmaceutisch aanvaardbaar zijn. Dergelijke farmaceutische samenstellingen hebben bij voorkeur de vorm van doserings-15 eenheden; d.w.z. fysisch discrete eenheden die een voorafbepaalde hoeveelheid van het geneesmiddel bevatten, overeenkomend met een fractie of veelvoud van de dosis die berekend is voor het produceren van de gewenste therapeutische respons. De doseringseenheden kunnen 1, 2, 3, 4 of meerdere enkelvoudige doses bevatten of als alternatief de helft, een derde, 20 of een vierde van een enkele dosis bevatten. Een enkelvoudige dosis bevat bij voorkeur een hoeveelheid die voldoende is om het gewenste therapeutische effect bij toediening te geven bij één toepassing van één of meerdere doseringseenheden volgens het voorafbepaalde doseringsschema gewoonlijk een volledige, halve, derde of kwart van de dagelijkse dosering 25 eenmaal, tweemaal, driemaal of viermaal per dag toegediend. Andere therapeutische middelen kunnen ook aanwezig zijn. Farmaceutische samenstel-lingen die ongeveer 1 tot 50 mg van het actieve ingrediënt per doserings-eeriheid verschaffen, hebben de voorkeur en worden conventioneel bereid als tabletten, lozenges, capsules, poeders, waterige of olieachtige sus-30 pensies, siropen, elixers en waterige oplossingen, Geprefereerde orale samenstellingen hebben de vorm van tabletten of capsules en kunnen conventionele excipiëntia bevatten, zoals bindmiddelen (bijvoorbeeld siroop, acacia, gelatine, sorbitol, tragecanth, of polyvinylpyrrolidon), vulstoffen (b.v. lactose, suiker, maïszetmeel, calciumfosfaat, sorbitol, of gly-35 cine), smeermiddelen (b.v. magnesiumstearaat, talk, polyethyleenglycol of silica), desintegratiemiddelen (b.v. zetmeel) en bevochtigingsmiddelen Λ -j» « J! f * i 0 · ‘ Λ +· 12 (b'.v. natriumlaurylsulfaat). Oplossingen of suspensies van een verbinding met formule 10 met conventionele farmaceutische dragers worden gebruikt voor parenterale samenstellingen zoals een waterige oplossing voor intraveneuze injectie of een olieachtige suspensie voor intramusculaire 5 injectie. Dergelijke samenstellingen met de gewenste helderheid, stabiliteit en vermogen om aan parenteraal gebruik te worden aangepast, worden verkregen door 0,1 tot 10 gew.% van de actieve verbinding op te lossen in water of een drager bestaande uit een polyhydrische alifatische alcohol, zoals glycerine, propyleenglycol en polyethyleenglycolen of meng-10 seis daarvan. De polyethyleenglycolen bestaan uit een mengsel van niet-vluchtige, gewoonlijk vloeibare polyethyleenglycolen die in zowel water als organische vloeistoffen oplosbaar zijn en molecuulgewichten hebben van ongeveer 200 tot 1500.
De verbindingen die de uitvinding vormen alsmede hun 15 bereidingsmethoden zullen vollediger duidelijk worden uit een lezing van de volgende voorbeelden die slechts ter toelichting worden gegeven en niet zo mogen worden opgevat dat ze de uitvinding in omvang beperken. Alle temperaturen zijn, tenzij anders is aangegeven, uitgedrukt in graden Celsius.
20 De kernmagnetische resonantie (NMR) spectraaleigen- schappen verwijzen naar chemische verschuivingen ( S }, uitgedrukt in delen per miljoen (ppm) versus tetramethylsilaan (TMS) als referentie-standaard. Het relatieve oppervlak dat voor de verscheidene verschuivingen in de proton NMR spectraalgegevens wordt vermeld, correspondeert 25 met het aantal waterstofatomen van een bepaald functioneel type in het molecuul. De IR bepalingen werden verkregen met kaliumbromide (KBr) als verdunningsmiddel. De elementairanalyses zijn in gewichtsper-centages uitgedrukt.

2- Chloroethyl-acetoacetaat
Een oplossing van 75 g (0,403 mol) 5-(1-hydroxyethyl-ideen)-2,2-dimethyl-l,3-dioxalaan-4,6-dion (zie Y. Oikawa, et al., J.
5 Org. Chem., 43, 2087 - 2088 (1978)) en 200 ml 2-chloorethanol werd gedurende 5 uur op 125°C verwarmd. Na de donkere oplossing tot kamertemperatuur te hebben afgekoeld, werd de overmaat 2-chloorethanol onder verminderde druk verwijderd en werd het resterende residu gedestilleerd, waarbij 49,4 g (74%) produkt werd verkregen als een heldere vloeistof met een 10 kookpunt van 80 - 85°C/0,4 mm.

3- Chloropropylacetoacetaat
Men behandelde 47,3 g (0,50 mol) 3-chloropropanol en een katalytische hoeveelheid triëthylamine bij 65°C druppelsgewijze met 15 42 g (0,50 mol) diketeen. Nadat de toevoeging voltooid was, werd het reactiemengsel gedurende nog één uur bij 65°C geroerd. Destillatie van het residu gaf 72,9 g (82%) produkt als een heldere vloeistof met een kookpunt van 78 - 85°C bij 150 mm.
Verdere tussenprodukten met formule 12 konden worden 20 bereid door modificaties van bovenstaande voorbeelden, die voor deskundigen op het gebied van organische chemische synthese geen probleem vormen.
B. Tussenprodukten met formule 13 Voorbeeld III
Methyl-2-[(3-nitrofenyl)methyleenl-3-oxabutanoaat 25 Een oplossing van 151 g (1,00 mol) 3-nitrobenzaldehyde, 116 g (1,00 mol) methylacetoacetaat, 10 ml ijsazijn, 4 ml piperidine, en 400 ml benzeen werd gedurende 2 uur onder terugvloeikoeling gekookt, in welke tijd 21 ml water via een Dean-Stark val werd verwijderd. De donkergele oplossing werd tot kamertemperatuur afgekoeld waarbij stolling 30 plaatsvond. Filtratie, gevolgd door wassen met ether, leverde 180 g produkt als een gele vaste stof. Een extra hoeveelheid produkt van 23 g werd verkregen uit het filtraat, waarbij men in totaal 203 g (82%) produkt verkreeg met een smeltpunt van 145 - 146°C (literatuur smeltpunt 158°C; zie Meyer, et al., Arzneim.-Forsch/Drug Research, 31, 407 (1981)).

Deze verbinding werd op molaire schaal bereid volgens * ^ A és ? ;* o 14 de methode die bovenstaand in voorbeeld III is beschreven, waarbij de gebruikte methylester werd vervangen door ethylacetoacetaat. Herkristal-lisatie uit ethanol gaf 182 g (69%) produkt als een gele vaste stof met een smeltpunt van 103 - 106°C (literatuur smeltpunt 110°C; zie Ruhemann, 5 J. Chem. Soc., 83, 717 (1903)).

C. Tussenprodukten met formule 23 Voorbeeld XLII
4-(2-Methoxyfenyl)-1-piperazinepropanol
De synthese voor dit tussenprodukt en andere tussenproduk-25 ten met formule 23 is ontleend aan Wu, et al., J. Med. Chem., 12, 876 (1969). Een mengsel van 10,0 g (52,1 mmol) l-(2-methoxyfenyl)piperazine, 4,25 g (45,0 mmol) 3-chloropropanol, 6,21 g (45 mmol) microscopisch verpoederd kaliumcarbonaat en 75 ml acetonitrile werd gedurende 23 uur onder terugvloeikoeling gekookt. Na afkoeling tot kamertemperatuur werd 30 200 ml water toegevoegd en het resulterende mengsel werd met methyleen- chloride geëxtraheerd. De gecombineerde organische porties werden met water en zoutoplossing gewassen, en daarna boven magnesiumsulfaat gedroogd. Filtratie en verwijdering van vluchtige stoffen onder verminderde druk, gaf 11,7 g ruw alcoholprodukt. Herkristallisatie uit acetonitrile gaf 9,4 35 g (72%) produkt als witte vaste stof met een smeltpunt van 94-95eC.

Voorbeeld XLIII
2- Chloroethyl-methyl-l,4-dihydro-2,6-dimethyl-4-(3-nitrofenyl) -3,5-pyri-dinedicarboxylaat
Men voegde 3,85 g (50,0 mmol) ammoniumacetaat toe aan een 5 oplossing van acetoacetaat, zoals bovenstaand in voorbeeld I bereid (8,25 g, 50,0 mmol) en 50 ml absolute ethanol, en kookte gedurende 1 uur onder terugvloeikoeling onder stikstof. Het bovenstaand in voorbeeld III bereide cinnamaat-tussenprodukt (12,5 g, 50,0 mmol) werd vervolgens toegevoegd en de resulterende gele oplossing werd nog eens 12 uur onder terug-10; vloeikoeling gekookt. Na afkoeling tot kamertemperatuur werd het oplosmiddel onder verminderde druk verwijderd en werd het residu herkristalli-seerd uit ethanol, waarbij het produkt werd verkregen als een gele vaste stof met een smeltpunt van 129 - 131°C (literatuur smeltpunt 130°C; zie Iwanami, et al., Chem. Pharm. Buil., 27, 1426 (1979)).

Volgens de bovenstaand in voorbeeld XLIII beschreven methode, waarbij echter de in de voorbeelden II en III bereide tussenprodukten 20 werden gebruikt, werd het in de titel vermelde tussenprodukt bereid in een 99%'s opbrengst op een 50 mmol-schaal. Het produkt werd verkregen als een gele vaste stof met een smeltpunt van 125 - 130°C.

Een oplossing van het boven in voorbeeld XLIII bereide tussenprodukt met formule 20 (1,98 g, 5,00 mmol), 1-(2-methoxyfenyl)-piperazine (1,05 g; 5,5 mmol), triëthylamine (0,61 g, 6,04 mmol), en 30 25 ml tetrahydrofuran werd gedurende 36 uur onder stikstof onder terug vloeikoeling gekookt. Na verwijdering van de vluchtige stoffen onder verminderde druk, werd het residu opgelost in 2-propanol, werd 50 ml 10%’s waterig HC1 (V : V) toegevoegd en werd de oplossing gedurende 15 minuten op een stoombad verwarmd. Na extractie met methyleenchloride, werden de 35 gecombineerde organische porties gewassen met water en zoutoplossing, gedroogd boven magnesiumsulfaat, gefiltreerd en onder verminderde druk ge-

Een oplossing vein het dihydropyridine-tussenprodukt met formule 20, dat bovenstaand in voorbeeld XLIV was bereid (8,65 g, 20 mmol), 35 1-(2-methoxyfenyl)piperazine (4,1 g, 20 mmol), 6 g triëthylamine, en een katalytische hoeveelheid kaliumjodide in 50 ml acetonitrile werd geduren- > -¾ v4' · >v - 18 de 48 uur onder terugvloeikoeling gekookt. Na verwijdering van vluchtige stoffen onder verminderde druk, werd het residu fijngewreven in acetonitrile, waarbij 3,5 g (17%) produkt werd verkregen als een gele vaste stof met eensmeltpunt van 70 - 75°C.

Methyl-1,4-dihydro-5- [ [ [2- (4- (2-methoxyfenyl) -1-piperazinyl) 3 ethyl] amino] -carbonyl-2,6-dimethyl-4-(3-nitrofenyl)-3-pyridinecarboxylaat 15 Diketeen (1,85 g, 22,0 mmol) werd langzaam druppelsgewijze toegevoegd aan een 0°C oplossing van 1-(2-aminoethyl)-4-(2-methoxyfenyl)-piperazine (4,7 g, 22 mmol; zie Muil, et al., J. Med. Pharm. Chem., 5, 944 (1962) voor de bereiding) in 20 ml absolute ethanol. Nadat de toevoeging voltooid was, liet men de oplossing opwarmen tot kamertemperatuur 20 en werd gedurende 30 minuten geroerd. Concentratie onder verminderde druk gaf 5,5 g van het ruwe acetozuuramide-tussenprodukt als een heldere olie die zonder verdere zuivering werd gebruikt.
De Knovenagel-condensatie werd gerealiseerd door de oplossing van 3-nitrobenzaldehyde (2,25 g, 15 mmol), het acetozuuramide-tussenpro-25 dukt, 5 druppels ijsazijn, en 3 druppels piperidine in 25 ml benzeen verscheidene uren onder terugvloeikoeling te koken. Na opwerking en flash-chromatografie (5% methanol : chloroform) werd 4,35 g (44%) produkt verkregen als een gele olie.
Een deel van deze gele olie (3,5 g, 7,7 mmol), methyl 3-amino-30 crotonaat (3,9 g, 7,8 mmol) en 40 ml 2-propanol werd gedurende een nacht (18 uu!r) onder terugvloeikoeling gekookt. Concentratie onder verminderde druk gaf 3,5 g van een gele gom. Flash-chromatografie (2% methanol ; chloroform; 3% methanol : chloroform; en daarna 4 % methanol : chloroform) gaf 0,73 g (17%) produkt als een geel schuim met een smeltpunt van 83 -35 88°C.

Deze verbinding werd op soortgelijke wijze bereid als beschre-10 ven is in voorbeeld XLVIII, maar onder toepassing van een aminopropyl- piperazine-uitgangsprodukt, dat beschreven is in Wu, et al., J. Med. Chem., 12, 876 (1969). Het gewenste produkt werd geïsoleerd als een geel schuim met een smeltpunt van 70 - 80°C.

Bis-[3-[4-(2-methoxyfenyl)-1-piperazinyl]propyl]-1,4-dihydro-2,6-dimethyl- 4-(3-nitrofenyl)-3,5-pyridinedicarboxylaat-dihydrochloride
Aan een smelt van het piperazine-tussenprodukt met formule 23, bovenstaand in voorbeeld XLII bereid (5,00 g, 20,0 mmol) in een 105°C 25 oliebad, werd langzaam diketeen (1,68 g, 20,0 mmol) toegevoegd. Nadat de toevoeging voltooid was, werden 50 ml absolute ethanol, 1,20 g (16 mmol) ammoniumacetaat en 1,58 g (10,4 mmol) 3-nitrobenzaldehyde toegevoegd, en de resulterende oplossing werd gedurende 17 uur onder terugvloeikoe-ling gekookt. Na afkoeling tot kamertemperatuur werd het reactiemengsel 30 onder verminderde druk geconcentreerd, waarbij 8,5 g van een donkergele olie werd verkregen. De olie werd opgenomen in methyleenchloride en gewassen met 2 porties 10%'s waterig HCl (V : V) en de organische laag werd geconcentreerd waarbij een gele vaste stof werd verkregen. Her-kristallisatie uit ethanol-ethylether gaf 2,95 g (35%) produkt als een ge-35 le vaste stof met een smeltpunt van 155 - 175°C (ontleding).

4- [4- [2-Methoxyfenyl) -l-piperazinyl] -butyl] -methyl-1,4-dihydro-2,6-dime-thyl-4-(3Lnitrofenyl)-3,5-pyridinedicarboxylaat-dihydrochloride 10 Diketeen (4,6 g, 0,055 mol) werd bij 100°C druppelsgewij ze toegevoegd aan 13,2 g (0,05 mol) 4-(2-methoxyfenyD-l-piperazinebuta-nol (Brits octrooischrift 803.403 van 22 oktober 1958). De smelt werd gedurende 20 minuten op 100°C verwarmd. De smelt werd opgelost in 200 ml ethanol en 3,5 g (0,05 moil) ammoniumacetaat werd toegevoegd. De oplossing 15 werd gedurende 2 uur onder terugvloeikoeling gekookt. Men voegde 8,4 g (0,05 mol) methyl m-nitro-2-acetylcinnamaat toe en bleef gedurende 17 uur onder terugvloeikoeling koken. De oplossing werd onder verminderde druk geconcentreerd. Het residu werd gezuiverd door flash-chromatografie (2% MeOH in CHCl^ op silicagel) . Het produkt werd met ethanolisch HC1 20 omgezet in het hydrochloride waarbij 1,55 g (4,7%) produkt werd verkregen met een smeltpunt van 110 - 130°C.

Enkele verdere verbindingen met formule 10, anders dan die welke bovenstaand zijn gedefinieerd, hebben eveneens bruikbare cardiovasculaire eigenschappen. Zij bezitten bijvoorbeeld calcium-toetreding en α-adrenergische blokkering alsmede antihypertensieve werkzaamheid. De beteke-5 nis van deze bevindingen is reeds adequaat bovenstaand besproken voor de verbindingen met formule 10. De verdere verbindingen worden bereid zoals is toegelicht in de onderstaande voorbeelden LXXVII en LXXVIII en beantwoorden aan formule 10, waarin de symbolen R^, R~, R^, R^, X en Z dezelfde betekenissen hebben als bovenstaand is gedefinieerd. De defi-10 nitie voor Y wordt echter uitgebreid doordat hij alkyleenketens met 2 -5 koolstofatomen en alkyleenketens die een zwavelatoom in de keten bevatten, omvat. De omvang van de onderhavige uitvinding strekt zich derhalve uit tot alle verbindingen met formule 10 alsmede farmaceutisch aanvaard- 2 4 bare zuuradditiezouten en/of solvaten daarvan, waarin de symbolen R , R , 5 6 2 6 15 R , R , X, Y en Z de volgende betekenissen hebben. R en R worden onafhankelijk gekozen uit lage alkyl, hydroxyalkyl, alkoxyalkyl, alkylamino-alkyl, of dialkylaminoalkyl en kunnen gelijk of verschillend zijn. Lage alkyl betekent ; alkoxyalkyl duidt op een ^-alkyleenketen en een via een zuurstofatoom daarmee verbonden ^-alkylgroep; evenzo duiden 20 alkylaminoalkyl en dialkylaminoalkyl op lage alkylgroepen en een C^_4-alkyleenketen, met elkaar verbonden via een secundaire (-NH-) of een v, 4 tertiaire ( /N-) aminogroep. Deze verdere verbindingen met formule 10 kunnen worden bereid door de syntheseprocessen welke in de schema's 1-3 zijn getoond, aan te passen. In het bijzonder kunnen de verbindingen met formule 10 worden verkregen door gebruik te maken van de eerdervermelde methoden 5 voor de verbindingen met formule 10 of een voor de hand liggende modificatie van deze procedures. Dit wordt aan de hand van de volgende voorbeelden geïllustreerd.
Voorbeeld LXXVII
[2- [ [2-[4-(2-Methoxyfenyl)-l-piperazinyl]-ethyl]ethio] ethyl]-methyl-1,4-10 dihydro-2,6-dimethyl-4-(3-nitrofenyl)-3,5-pyridine-dicarboxylaat
Men voegde bij 80°C druppelsgewijze 8,4 g (100 mmol) diketeen toe aan 12,2 g (100 mmol) 2,2-thiodiëthanol dat een katalytische hoeveelheid triëthylamine bevatte. Het mengsel werd gedurende 15 minuten op 80°C verwarmd. Het mengsel werd opgelost in 200 ml ethanol en er 15 werd 7,7 g (100 mmol) ammoniumacetaat toegevoegd. De oplossing werd daarna gedurende een uur onder terugvloeikoeling gekookt. Men voegde 24,9 g (100 mmol) methyl-2-t(3-nitrofenyl)methyleen]-3-oxobutanoaat (voorbeeld III) toe en bleef gedurende 18 uur onder terugvloeikoeling koken.
De oplossing werd onder verminderde druk geconcentreerd en het residu 20 werd gezuiverd door flash-chromatografie (2% MeOH in methyleenchloride op silicagel). Men verkreeg 5 g (21,8%) van een synthetisch tussenprodukt waarvan de structuur een modificatie van de verbinding met formule 20 was (Cl vervangen door OH).
Een oplossing van 4,0 g (9,2 mmol) van dit alkyleenthio-25 alkanol-tussenprodukt en 1,1 g (9,2 mmol) thionylchloride in 100 ml chloroform werd gedurende 1,25 uur onder terugvloeikoeling gekookt. De oplossing werd onder verminderde druk geconcentreerd. Het residu werd opgelost in 300 ml acetonitrile en gecombineerd met 3,5 g (18 mmol) o-methoxyfenylpiperazine; 2,5 g (18 mmol) microscopisch verpoederd ka-30 liumcarbonaat; en een natriumjodidekristal. Het mengsel w erd gedurende 4 dagen onder terugvloeikoeling gekookt en onder verminderde druk geconcentreerd. Het residu werd door flash-chromatografie gezuiverd (0,5% methanol en 1 : 4 ethylacetaat-methyleenchloride op silicagel) waarbij 2,2 g (39,3%) produkt werd verkregen met een smeltpunt van 52 - 63°C.

Men voegde bij 85°C langzaam 3,0 g (36 mmol) diketeen 5 toe aan een smelt van 10,0 g (36 mmol) l-(5-hydroxypentyl)-4-(2-metho-xyfenyl)piperazine (bereid volgens het Britse octrooischrift 803.403 van 22 oktober 1958). Nadat de toevoeging voltooid was, werden 100 ml absolute ethanol en 2,8 g (36 mmol) ammoniumacetaat toegevoegd. Na 2 uur koken onder terugvloeikoeling, werd 9,0 g (36 mmol) methyl-2-[(3-nitrofenyl)me-1Q thyleen] -3-oxobutanoaat (bereid in voorbeeld III) toegevoegd en werd nog 17 uur onder terugvloeikoeling gekookt. De oplossing werd onder verminderde druk geconcentreerd. Het residu werd gezuiverd door flash-chro-matografie (1,5% methanol en methyleenchloride). Men verkreeg 5,2 g (23,7%) produkt met een smeltpunt van 52 - 62°C.

Verbinding met formule 10, alsmede de farmaceutisch aan vaardbare zuuradditiezouten daarvan, waarin 6 R“ en R onafhankelijk van elkaar gekozen worden uit lage alkyl, hydroxyalkyl, alkoxyalkyl, alkylaminoalkyl, of dialkylamino-5 alkyl,* 4 R staat voor cycloalkyl met 5-7 koolstofatomen, bi-cycloalkenyl met 7-9 koolstofatomen, hetaryl (omvattende furanyl, indolyl, methylthiopyridyl, thiënyl, benzoxadiazolyl en benzothiadiazo-lyl); aryl (omvattende fenyl, naftyl of gesubstitueerd fenyl met substi-10 tuenten die onafhankelijk gekozen zijn uit acetoamino, lage alkyl, lage alkoxy, cyano, halogeen, hydroxyl, nitro, trifluoromethyl, trifluorme- thylsulfonyl, of methylsulfonyl); 5 2 R staat voor R of de groep met formule 3; X staat voor 0 of NH; 15. staat voor een C^^-alkyleenketen, alkyleenoxyalky- leen, alkyleenthioalkyleen, of alkyleenaminoalkyleenketen; en Z staat voor fenyl, pyridinyl, of pyrimidinyl, waarbij deze fenyl-, pyridinyl·* of pyrimidinylgroepen al dan niet gesubstitueerd kunnen zijn door een of meerdere substituentgroepen, gekozen uit lage al-20 kyl, lage alkoxy, cyano, halogeen, en trifluoromethyl.
