Abstract

본 발명은 폴리프럭탄(레반)을 생성하는 DNA 서열, 및 이들 DNA 서열을 포함하는 플라스미드 뿐만 아닐, 이들 플라스미드를 사용하며 폴리프럭탄(레반)을 발현하는 유전자전환 식물을 제조하기 위한 방법에 관한 것을기재하고 있다.

Description

폴리프럭탄(레반)을 생성하는 DNA 서열, 이들 서열을 포함하는 플라스미드 및 유전자전환 식물의 제조 방법(DNA SEQUENCES WHICH LEAD TO THE FORMATION OF POLYFRUCTANS (LEVANS), PLASMIDS CONTAINING THESE SEQUENCES AS WELL AS A PROCESS FOR PREPARING TRANSGENIC PLANTS)

제1도는 p35-CW-LEV 플라스미드의 구조를 나타내다. 이는 세 개의 단편 A, B 및 C를 포함한다. 단편 A는 꽃양배추 모자이크 바이러스(CaMV)의 35s 프로모터를 함유한다(뉴클레오티드 6906-7437). 단편 B는 에르위니아 아밀로보라로부터 얻은 레반 슈크라아제의 서열(뉴클레오티드 689-2122)을 함유한다(서열 번호 1). 단편 C는 Ti-플라스미드, pTi ACH 5의 유전자 3의 폴리아데닐화 신호를 함유한다.(뉴글레오티드 11749-11939).
제2도는 p35s-CY-LEV 플라스미드의 구조를 나타낸다. 이는 세 개의 단편 A, B 및 C를 포함한다. 단편 A는 꽃양배추 모자이크 바이러스(CaMV)의 35s 프로모터를 함유한다(뉴클레오티드 6906-7437). 단편 B는 에르위니아 아밀로보라로부터 얻은 레반 슈크라아제의 서열(뉴클레오티드 864-2122)을 함유한다(서얼 번호 1). 단편 C는 Ti-플라스미드, pTi ACH 5의 유전자 3의 폴리아데닐화 신호를 함유한다.
제3도는 p33-CW-LEV 플라스미드의 구조를 나타낸다. 이는 세 개의 단편 A, B 및 C를 포함한다. 단편 A는 파타틴 유전자 B33의 프로모터 영역의 DraI-DraI-단편(위치-1512에서 위치+14)을 함유한다. 단편 B는 에르위니아 아밀로보라로 부터 얻은 레반 슈크라아제의 서열(뉴클레오티드 689-2122)을 함유한다(서열 번호 1). 단편 C는 Ti-플라스미드, pTi ACH 5, T-DNA의 유전자 3의 폴리아데닐화 신호(뉴클레오티드 l1749-l1939)를 함유한다.

Claims (12)

  1. 레반 슈크라아제의 서열이 하기의 뉴클레오티드 서열(서열 번호 1)을 가지며, 에르위니아 아밀로보라(Erwinia amylovora)종의 그램-음성 박테리아로부터 유래하는, 식물 게놈 중에 통합됨으로써 잎 및 괴경에서 폴리프럭탄(레반)을 발현시킬 수 있는 것을 특징으로 하는, 레반 슈크라아제의 서열이 있는 DNA 서열.
  2. 잎 및 괴경에서 플리프럭탄(레반)을 발현하는 식물의 제조를 위한, 제1항에 따른 DNA 서열을 포함하는 플라스미드.
  3. 플라스미드 p35s-CW-LEV(DSM 7186).
  4. 플라스미드 p35s-CY-LEV(DSM 7187).
  5. 플라스미드 p33-CW-LEV(DSM 7188).
  6. (a) (i) 식물 내에서 활성이고 의도된 표적 조직 또는 표적 세포 내에서 RNA를 생성하는 프로모터, (ii) 제1항에 따른 레반 슈크라아제의 DNA서열, 및 (iii) 식물 세포 내에서 전사 종결 뿐만 아니라 RNA의 3′-말단에 폴리 A잔기틀 부착시키는 3′-비-번역 서열의 부분적 서열들을 갖는 DNA 서열을 제조하는 단게, (b) 제2항 내지 5항 중 어느 한 항에 따른 플라스미드를 사용하여 식물 세포로부터 얻은 재조합 이중 가닥 DNA분자의 식물 게놈 내에 상기 DNA 서열을 전달 및 통합하는 단계, 및 (c) 형질전환된 식물 세로로부터 온전한 식물 전체를 재생하는 단계로 이루어진, 잎 및 괴경에서 폴리프럭탄(레반)을 발현하는 유전자전환 식물의 제조 방법.
  7. 제6항에 있어서, 잎 및 괴경에서의 폴리프럭판(레반)슈크라아제 발현이 엽록체, 백색체, 미토콘드리아, 세포질, 세포간 공극 및(또는) 액포 네에서 폴리프럭탄(레반)슈크라아제 활성이 나타나도록 하는 것을 특징으로 하는 방법.
  8. 제6항 또는 7항에 따른 방법으로 얻은 식물.
  9. 제8항에 있어서, 농작물인 식물.
  10. 제8항 또는 9항에 있어서, 옥수수, 벼, 밀, 보리, 사탕무우, 사탕수수, 담배 또는 감자 식물인 식물.
  11. 잎 및 괴경에서 폴리프럭탄(레반)을 발현하는 식물의 제조를 위한, 제1항에 따른 DNA 서열의 용도.
  12. 잎 및 괴겨8에서 폴릭프럭탄(레반)을 발현하는 식물의 제조를 위한, 제2항 내지 5항 중 어느 한 항에 따른 플라스미드의 용도.
