Claims (12)

  1. (1) 하기 (a)와 (b)로 이루어진 DNA 단편, 즉 (a) 폴리펩티드 코우딩 지역의 5'말단에 위치할 경우 메신저 RNA의 전사를 조절할 수 있고, (i)이 DNA 단편을 함유한 숙주 유기체와 접촉하고 있는 배지속의 메탄올 존재, (ii)이 DNA 단편을 함유한 숙주 유기체와 접촉하고 있는 배지속에서의 메탄올 이외의 비-이화 물질 억제 탄소원 존재 및 (iii)이 DNA 단편을 함유한 숙주 유기체를 이화물질 억제 탄소 및 에너지원위에서 배양한 후 그 숙주 유기체와 접촉하고 있는 배지속의 탄소원 기아현상중에서 선택되는 한 가지 조건에 대하여 반응하는 조절지역 및, (b) B형 간염 표면 항원 또는 그 일부를 암호화하는 폴리펩티드 코우딩 지역으로 이루어진 DNA 단편, 또는 (2) 그와같은 DNA 단편과 아울러 박테리아 플라스미드 DNA, 선택성 효모 표식 유전자 및 효모 자율복제 서열로 이루어진 플라스미드로 인하여 형질 전환이 된 효모 균주를 메탄올, 이화물질 비(非)-억제 탄소원, 또는 그 혼합물, 또는 적어도 하나의 이화물질 억제 탄소 및 에너지원으로 이루어진 영향 배지속에서 배양하고, 상기 탄소 및 에너지원이 이화물질 억제 탄소 및 에너지원일 경우에는 그 배양 단계의 생성물에게 탄소원 기아현상의 조건을 가해줌을 특징으로 하는 B형간염 표면항원의 제조방법.
  2. 제1항에 있어서, 상기 DNA 단편이 도면 제2도의 제한지도에 나와 있는 것과 같음을 특징으로 하는 조절지역을 갖고 있는 방법.
  3. 제1항 또는 제2항에 있어서, 상기 DNA 단편에 상기 폴리펩티드 코우딩 지역에 의해서 암호화된 메신저 RNA의 전사 종결 및 폴리아데닐화를 조절할 수 있는, 폴리펩티드 코우딩 지역 아래쪽 DNA 3' 서열이 부착되어 있음을 특징으로 하는 방법.
  4. 제1항 내지 제3항 중 어느 한 항에 있어서, 상기 DNA 단편에 박테리아 플라스미드 DNA, 박테리오파아지 DNA, 효모 플라스미드 DNA, 효모 염색체 DNA 또는 그중 둘 이상으로 부터 유래된 하나 이상의 추가적인 DNA 서열이 부착되어 있음을 특징으로 하는 방법.
  5. 제4항에 있어서, 상기 효모 염색체 DNA가 자율 복제 DNA 서열 및 표식 유전자로 이루어짐을 특징으로 하는 방법.
  6. 제5항에 있어서, 상기 자율복제 서열이 PARS1 또는 PARS2인 방법.
  7. 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 DNA 단편에 첫번째 삽입 DNA 단편, 선택성 표식 유전자 및 두번째 삽입 DNA 단편으로 이루어진 연속배열 DNA가 부착되어 있으며 ; 이에 상기 첫번째 및 두번째 삽입 DNA 단편들은 각각 그 길이가 적어도 약 200뉴클레오티드에 상당하고 또 피치아속에 속한 종의 게놈 DNA 부분과 상동하는 뉴클레오티드 서열을 갖고 있으며 ; 상기 DNA 단편과 상기 표식 유전자는 상기 첫번째 삽입 DNA 단편의 3' 말단과 상기 두번째 삽입 DNA 단편의 5' 말단 사이에 위치하며 ; 상기 첫번째 및 두번째 삽입 DNA 단편들은 피치아의 게놈속에 배향되는 것과 같이 서로에 대하여 배향됨을 특징으로 하는 방법.
  8. 제1항 내지 제7항 중 어느 한 항에 있어서, 상기 폴리펩티드 코우딩 지역이, 필수적으로 아래애 나와 있는 약 700 염기쌍 단편, 또는 그의 아(sub)단위, 또는 그의 변이체나 기능상의 등가물로 이루어져 있는 방법

  9. 제1항 내지 제8항 중 어느 한 항에 있어서, 상기 B형 간염 표면항원을 분리하고 정제함을 특징으로 하는 방법.
  10. (a) 폴리펩티드 코우딩 지역의 5' 말단에 위치할 경우 메신저 RNA의 전사를 조절할 수 있으며, (i) 이 DNA 단편을 함유한 숙주 유기체와 접촉하고 있는 배지속의 메탄올 존재 ; (ii)이 DNA 단편을 함유한 숙주 유기체와 접촉하고 있는 배지속에서의 메탄올 이외의 비-이화물질 억제 탄소원 존재 및 (iii) 이 DNA 단편을 함유한 숙주 유기체를 이화물질 억제 탄소 및 에너지원 위에서 배양한 후 그 숙주 유기체와 접촉하고 있는 배지속의 탄소원 기아현상중에서 선택되는 적어도 한가지 조건에 대하여 반응하는 조절지역에다 (b) B형간염 표면항원 또는 그 일부를 암호화하는 폴리펩티드 코우딩 지역에 부착시킴을 특징으로 하는 신규 DNA 단편의 제조 방법.
  11. 제10항에 있어서, 상기 조절지역이 도면 제2도의 제한지도에 나와 있는 것과 같음을 특징으로 하는 방법.
  12. 제10항 또는 제11항에 있어서, 상기 폴리펩티드 코우딩 지역에 의해서 암호화된 메신저 RNA의 전사 종결 및 폴리아데닐화를 조절할 수 있는 DNA 3' 서열을 폴리펩티드 코우딩 지역 아래쪽 DNA에 부착시킴을 특징으로 하는 방법.
