Claims (16)
- 다음의 특성을 갖는 플루라나제 타입의 신규의 디브랜칭 효소를 함유하는 디브랜칭효소 생성물 :a) 그것은 적당한 영양배지속에서, 이하에 바실루스 아시도 풀루리티쿠스(Bacillus Acidopullulyticus)라고 명명된 분류군에 속하는 균주를 배양하므로서 생성되는 발효즙으로 부터 얻어지며,b) 그것은 바실루스균주 NCIB 11607로 부터 유도된 디 브랜칭효소와 기본적으로 동일한 효소화학적 특성을 갖고 있으며, 면역학적으로도 동일하거나 부분적으로 동일한 특성을 갖고 이으며,c) 그것의 최적활성은, 아세트산염완충액(0.05M)속에서 pH 4-5로, 30분간 배양시켜서 측정했을때, 적어도 60℃정도에서 나타나며,d) 그것의 최적 pH는, 아세트산염완충액(0.05M)속에서, 60℃에서 30분간 배양시켜서 구한바에 의하면 3.5내지 5.5범위이내이며,e) 그것은 pH 5에서 포도당용액(중등으로 30퍼센트의 건조고체)속에서 측정했을 때 60℃에서 72시간후에도 적어도 50%의 잔류활성을 갖고 있음.
- 특허청구의 범위 1에 있어서의 디브랜칭효소 생성물로서 디브랜칭효소의 활성범위가 그람당 10내지 350,000풀루라나제 단위 이내인 것을 특징으로 하는 디브랜칭 효소 생성물.
- 특허청구 범위 2의 디브랜칭효소생성문로서 그 활성범위가 그람당 100내지 15,000풀루라나제 단위 이내의 것을 특징으로 하는 디브랜칭 효소 생성물.
- 디브랜칭효소를 생성시키는 바실루스 아시도풀루리티쿠스에 속하는 균주를 적당한 영양배지속에서 배양하므로서 만들어지는 디브랜칭 효소생성물과 그 디 브랜칭효소를 영양배지로 부터 회수하는 방법.
- 특허청구 범위 1의 디 브랜칭효소 생성물을 제조하는 방법에 있어, 탄소와 질소원 및 무기염을 함유하는 적당한 영양배지 속에서, 바실루스아시도풀루리티쿠스 분류군에 속하고 디브랜 칭효소를 생성시키는간균주를 배양하고 이어서 그 디브랜칭 효소생성물을 회수하는 것으로 되어 있는 것을 특징으로 하는 디브랜칭효소생성물의 제조방법.
- 특허청구 범위 제5항에 있어서, NCIB 11607, NCIB 11610 및 이들의 변종과 돌연변이체로 이루어진 군으로 부터 바실루스 아시도풀루리티쿠스가 선택되는 것을 특징으로 하는 제조방법.
- 특허청구범위 제6항에 있어서, NCIB 11638과 NCIB 11647로 구성된 돌연변위체군으로 부터 균주가 선택되는 것을 특징으로 하는 제조방법.
- 특허청구범위 제5항에 있어서, 바실루스아시도풀루리티쿠스균주는 NCIB 11611 또는 그 변종이나 돌연변위체인 것을 특징으로 하는 제조방법.
- 특허청구범위 제5항에 있어서, 바실루스아시도풀루리티쿠스균주는 NCIB 11636 또는 그 변종이나 돌연변이체인 것을 특징으로 하는 제조방법.
- 특허청구범위 제5항에 있어서, 바실루스 아시도 풀루리티쿠스균주는 NCIB 11637 또는 그 변종이나 돌연변이체인 것을 특징으로 하는 제조방법.
- 특허청구범위 제5항에 있어서, 바실루스 아시도 풀루리티쿠스균주는 NCIB 11639 또는 그 변종이나 돌연변이체인 것을 특징으로 하는 제조방법.
- 특허청구범위 제4항의 신규의 디브랜칭효소의 유효량과, 글루코아밀라아제와 베타-아밀라아제로 구성되는 균으로 부터 선택된 당화효소로된 효소계의 존재하에서, 녹말이나 녹말가루 분해물을 당화시키는 것을 특징으로 하는 녹말에서 텍스트로오스 및 또는 말토오스를 함유하는 시럽을 제조하는 방법.
- 특허청구범위 제12항에 있어서, 최저 30중량 퍼센트의 건조고체를 함유하는 녹말가수분해물을 당화시키는 과정을 더욱 포함하는 제조방법.
- 특허청구범위 제12항에 있어서, pH 범위 3.5내지 5.5와 온도 55℃내지 65℃이내에서 당화가 행해지는 것을 특징으로하는 제조방법.
- 특허청구범위 제12항에 있어서, 글루코아밀라아제와 베타-아밀라아제의 투입량은 건조고체 1그람당 각기 0.05내지 0.5 AG 단위와 0.005내지 0.3베타-아밀라아제 단위이내인 것을 특징으로 하는 제조방법.
- 특허청구범위 제15항에 있어서, 디브랜칭효소의 투입량은, 건조고체 1그람당 0.005내지 풀루라나제 단위 이내인 것을 특징으로 하는 제조방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
