Description
본 발명은 헤르페스비리데 (Herpesviridae), 인간 파필로마 바이러스, 박테리아 감염 및 진균 감염을 치료하기 위해 사용될 수 있는 화합물에 관한 것이다.
헤르페스비리데는 인간 및 동물에서 다수의 질환을 유발하는 거대 DNA 바이러스 패밀리이다. 인간에서 질환을 유발하는 가장 일반적인 헤르페스비리데는 수두 대상 포진 바이러스 (Varicella Zoster virus), 엡스타인-바 바이러스 (Epstein-Barr virus), 사이토메갈로바이러스 (Cytomegalovirus), 헤르페스 심플렉스 바이러스 1 (Herpes Simplex virus 1) 및 헤르페스 심플렉스 바이러스 2이다.

사이토메갈로바이러스는 헤르페스비리데 바이러스 과에 속하는 또다른 구성원이다. 인간 사이토메갈로바이러스 (HCMV 또는 CMV 또는 인간 헤르페스바이러스-5 (HHV-5))는 침샘과 관련있는 바이러스이며, 전형적으로 건강한 개체에서는 간과되지만, HIV 환자와 같이 면역약화된 장기 이식자 및 신생아에 대해서는 생명을 위협할 수 있다.

스타필로코커스 아우레우스 (Staphylococcus aureus) ATCC 6538
칸디다 알비칸스 (Candida albicans) ATCC 10231
화합물 2, 3 및 4를 다이메틸 설폭사이드 (DMSO)에 10 mg/ml 농도로 희석하였으며, 검사를 위해 추가적으로 TSB로 2배 희석하였다.
방법
1일
접종원 표준화 (standardisation of inoculum)
밤새 배양한 (o/n) 순수한 배양물에서, 적어도 콜로니 3-4개를 취하여, 튜브 안 4 ml 식염수에 전부 현탁하였다. 현탁액을 혼합하였다.
접종원의 탁도를 백그라운드로서 검정선이 있는 백색 종이를 사용해 0.5의 표준 탁도액과 시각적으로 비교하여, 표준 탁도액과 일치하도록 조정하였다.
맥파랜드 0.5 현탁액을 검사할 종에 따라 다음과 같이 희석하였다:
그람-음성: McFarl. 0.5 10 ㎕을 브로스 10 ml에 첨가
그람-양성: McFarl. 0.5 50 ㎕을 브로스 10 ml에 첨가
현탁액은 15분 이내에 접종에 사용하였다.
접종 및 배양
둥근 바닥형의 5 ml 팔콘 튜브에, 접종 현탁물 500 ㎕를 2배 희석한 항미생물제 500 ㎕와 함께, 눈금이 매겨진 파이펫을 사용해, 접종하였다.
튜브를 밀봉하고, 37℃에서 18-22시간 동안 배양하였다. 이는 배양 배지 소실을 방지하고, 교차 오염을 방지하기 위해 행하였다.
맥파랜드 0.5는 대략 108 CFU/ml이다. 결과의 해석은 정해진 접종원에 기초하므로, 접종원의 표준화가 필수적이다.
각각의 튜브에는, 박테리아 접종 후 약 5x105- 1x106 CFU/ml 및 효모 접종 후 5x103 - 1x104 CFU/ml이, 수용되었다.
2일
접종 현탁액의 순도를 체크하였다.
양성 대조군 튜브 3개에서 증식을 체크하였다.
가시적인 증식이 없는 항미생물제의 최소 농도를 최소 저해 농도 (MIC)로 기록하였다.
결과는 아래 표 1에 나타낸다.
요컨대, 검사 화합물들 모두 칸디다 알비칸스에 대한 강력한 항진균 활성뿐만 아니라 스타필로코커스 아우레우스와 살모넬라 엔테리카 세로바르 티피무리움에 대한 항박테리아 활성을 나타내었다.

* 진하게 나타낸 숫자는 박테리아/효모 세포의 최종 타이터를 검출하기 위한 시딩 (seeding)으로부터 유래된 것이다. 즉, 진한 숫자는 살균성 농도 (bactericide concentration)를 의미하고, 진하게 표시되지 않은 숫자는 MIC 범위에서 화합물의 정균성 농도 (bacteristatic concentration)를 의미함.

실시예 2
본 발명의 화합물의 항바이러스 효과 및 세포독성을 여러가지 포유류 세포주에서 화합물 2, 3 및 4를 검사함으로써 조사하였다.
재료 및 방법
세포 및 바이러스
헤르페스 심플렉스 바이러스-1 (HSV 1) 및 백시니아 바이러스 (VV) 2종을 검사하였으며, 사용 세포주는 HeLa, BSC 40 및 Vero 세포였다.
HeLa, Vero 및 BSC 40 세포들은, 5% CO2 인큐베이터에 넣어, 10% 소 태아 혈청 (FBS) 및 1% 페니실린/스트렙토마이신 (Gibco)이 첨가된 둘베코의 변형된 이글 배지 (DMEM) 내에서 37℃에서 배양하였다.
백시니아 바이러스의 웨스턴 리저브 균주 (WR-VV)는 BSC40 세포에서 증폭시켰으며, 타이트레이션한 후 -80℃에서 보관하였다.
헤르페스 심플렉스 바이러스 1 (HSV-1)은 Vero 세포에서 증폭시켰으며, 타이트레이션한 후 -80℃에서 보관하였다.
화합물 및 시약
화합물 2, 3 및 4 (분말 형태)를 앱솔루트 에탄올에 용해하여, 0.2 g/ml 농도의 원액을 수득하였다. 분액으로 나누어 -20℃에서 보관하였다. 실험하기 전에, 신선한 연속 희석을 무혈청성 배양 배지 (DMEM)에서 수행하였다.
세포독성
화합물들의 세포독성 효과를 HeLa, BSC40 및 Vero 세포에서 검사하였다. 세포를 12웰 플레이트에 접종하였으며, 다음날 플레이트는 80% 컨플루언스에 도달하였다. 화합물 2, 3 및 4를 다양한 농도로 세포에 적용하였다. 전-처리 30분 전, 배양 배지를 세포에 동일 농도의 화합물 2, 3 및 4의 존재 하에 첨가하였다. 세포를 매일 모니터링하였으며, 처리 후 48시간 또는 72시간에 고정 및 염색한 다음 사진을 촬영하였다. 샘플들 모두 20분간 실온에서 4% 포름알데하이드/H2O에서 고정한 다음 크리스탈 바이올렛으로 실온에서 30분간 염색하였다.
바이러스 감염 및 플라크 분석
컨플루언트에 도달한 신선한 세포에, WR-VV 및 SV-1 (약 200 내지 300 PFU/웰, 6웰 플레이트 또는 12웰 플레이트)을 1시간 동안 37℃에서 감염시켰다. 세포를 헹구고, 1% 아가로스가 함유된 배양 배지에서 배양하였으며, 감염 후 2일째 고정하였다.
다른 예로, 세포를 무혈청성 둘베코의 변형된 이글 배지 (DMEM) 또는 여러가지 농도의 화합물이 함유된 DMEM으로 감염 전 전처리하였으며, 화합물 2 및 3은 세포를 상기와 같이 고정할 때까지 실험 내내 배양물 내에 유지시켰다. 일부 실험은, 아가를 첨가하지 않고 액체 플라크 분석을 수행하였다.
결과
여러가지 포유류 세포주에서의 화합물의 세포독성
화합물 2, 3 및 4의 항바이러스 효과를 검사하기 위해, 화합물이 세포에 무독성이고, 세포 생존성에 영향을 미치지 않도록, 적절한 화합물의 희석도를 선택하였다. HeLa, BSC40 및 Vero 세포 등의 여러가지 포유류 세포주들에서 다양한 화합물 농도를 검사하였다.
밤새 배양한 세포에, 무혈청성 배지 내에서 30분간 화합물을 고 농도에서부터 저농도까지 전-처리하였다. 그런 후, 실험 내내 배지를 화합물 2, 3 및 4를 포함하는 정상 배양 배지로 교체하였다. 세포를 최대 72시간 매일 모니터링하였으며, 이후 고정, 염색 및 사진 촬영을 수행하였다. 도 4, 5 및 6에 나타낸 바와 같이, 실험 결과, 화합물 3종 모두, 농도 25 - 100 ㎍/ml에서 세포 허용적인 것으로 확인되었다. 도 4 및 5 각각 C로 표시된 열과 도 6은 Vero 세포에 대한 처리 결과를 나타낸 것으로, 이 Vero 세포는 HSV-1으로 감염되지 않았는데, 이는 화합물 2, 3 및 4로 처리된 것이었다.
HSV-1 증폭 및 바이러스 플라크 형성.
화합물 2 및 3의 저해 효과를 Vero 세포에서 모니터링하였다. 세포에 여러가지 농도로 화합물 2 및 3을 전처리한 다음, 완충제 또는 HSV-1 함유 완충제를 처리하였다. 감염 2일 후, 세포를 고정 및 염색하고, 사진을 촬영하였다. 화합물이 HSV-1 바이러스 증폭 또는 플라크 형성을 저해할 수 있는지를 확인하기 위해, 액체 중에 플라크 분석을 수행하여 증폭을 확인하고 (도 4 및 5에서 A열), 아가 중에서 플라크 분석을 수행하여 플라크 수를 측정하였다 (도 4 및 5에서 B열). 도 4 및 5에 각각 나타낸 바와 같이, 강력한 저해 효과는 화합물 2 및 3에서 관찰되었다. 이들 2종의 화합물은 HSV-1 증폭뿐만 아니라 Vero 세포에서 플라크 수를 농도 의존적인 방식으로 효과적으로 감소시켰다.
화합물 2 및 3는 백시니아 바이러스 플라크 형성에 대해서는 저해 효과를 나타내지 않았다.
화합물 2 및 3의 HSV-1 바이러스에 대한 특이성을 검사하기 위해, 또 다른 DNA 바이러스인 백시니아 바이러스에서 화합물들을 또한 검사하였다. BSC 40 세포는 무처리 상태로 두거나 또는 화합물을 50 mg/ml 농도로 전처리한 다음 백시니아 바이러스를 감염시켜 실험 내내 계속 모니터링하였다. 도 7에 나타낸 바와 같이, 화합물 2 또는 3은 백시니아 바이러스 (VV) 플라크 형성을 저해하지 않았으며, 이는 HSV-1 감염에 대한 화합물 2 및 3의 저해 효과가 매우 특이적이라는 것을 의미한다.
전술한 실험들로부터, 검사한 모든 세포들이 화합물 2, 3 및 4에 대해 0 - 100 ㎍/ml 농도에서 허용적일 수 있다는 결론을 도출할 수 있었다. 화합물 2 및 3 둘다, 증폭 및 플라크 형성 측면에서 HSV-1 감염된 Vero 세포에 대해 강력한 항바이러스 효과를 나타내었다. 화합물 50 ㎍/ml의 존재 하에서는, 감염성이 50%까지 감소하였다. 화합물 100 ㎍/ml의 존재 하에서는, 감염성이 80%까지 감소하였다. 검사 화합물들 모두 백시니아 바이러스에 대해서는 활성을 나타내지 않았다.

- 헤르페스 바이러스 (Herpes virus) 및/또는 인간 파필로마 바이러스 (Human Papilloma virus); 또는 헤르페스 심플렉스 바이러스 (Herpes Simplex virus) 또는 사이토메갈로바이러스 (Cytomegalovirus); 또는 헤르페스 심플렉스 바이러스 1(Herpes Simplex virus 1) 또는 헤르페스 심플렉스 바이러스 2(Herpes Simplex virus 2)을 치료하는데 사용하기 위한, 제1항 내지 제8항 중 어느 한 항에 따른 화합물.
- 제1항 내지 제4항 중 어느 한 항에 따른 화합물의 제조 방법으로서,
i) 카르바크롤 (carvacrol)을 R2CH2COCl과 반응시켜, R2가 염소 또는 브롬을 포함하는 할로겐인 식: (식 VII)을 가진 화합물을 제조하는 단계;
ii) 식: (식 VIII)을 가진 화합물을 식: (식 VII)을 가진 화합물과 반응시켜, 식:(식 IX)을 가진 화합물을 제조하는 단계로서,
