- 【特許請求の範囲】 1.例えば凍結後の製品の氷再結晶を防止又は抑制することにより例えば潜在的 凍結損傷を最小限にするように氷晶成長プロセスを改変して特に再成長における 氷の寸法及び形状特徴を変更して性質を改善する製品改善用製品添加剤としての 、III型AFP HPLC12に実質的に対応するアミノ酸配列をもつポリペ プチド又はタンパク質の使用であって、同量のニシナメタガレイAFPで得られ るよりも高い特異改変活性、特に不凍活性を得るように不凍ペプチドにそれ自体 公知の方法で実施することを特徴とする前記使用。 2.例えば凍結後の製品の氷再結晶を防止又は抑制することにより例えば潜在的 凍結損傷を最小限にするように氷晶成長プロセスを改変して特に再成長における 氷の寸法及び形状特徴を変更して性質を改善した改善製品の製造方法であって、 非改善製品又は非改善製品の製造で通常使用される成分もしくは混合物に、II I型AFP HPLC12のアミノ酸配列に実質的に対応するアミノ酸配列をも つ組換えポリペプチド又はタンパク質を、氷晶形成をより高い程度まで改変する に十分な量、即ち 同量のニシナメタガレイAFPで得られるよりも高い特異不凍活性を達成するに 十分な量で加えることを特徴とする前記方法。 3.例えばより高い程度まで、即ち同量のニシナメタガレイAFPで得られるよ りも高い特異不凍活性を達成するように凍結後の製品の氷再結晶を防止又は抑制 することにより例えば潜在的凍結損傷を最小限にするように氷晶成長プロセスを 改変して特に再成長における氷の寸法及び形状特徴を変更して性質を改善した改 善製品の製造方法であって、非改善製品自体又は非改善製品を製造するために通 常使用される成分もしくは混合物に、III型AFP HPLC12に実質的に 対応するアミノ酸配列をもつ組換えポリペプチド又はタンパク質の発現が可能な 宿主生物を加え、次いで、非改善製品を製造するために通常使用される段階以外 に請求項10から20のいずれか一項に記載の方法を実施することにより、II I型AFP HPLC12をコードする核酸配列を該製品又は成分もしくは混合 物で発現させるような条件下に宿主生物を置くことを特徴とする前記方法。 4.III型AFP HPLC12に実質的に対応するアミノ酸配列をもつ組換 えポリペプチド又はタンパク質が発現又は分泌された後に、AFPポリペプチド 又はタンパク質をモノマー 産物として得るためにプロテアーゼ又は薬品を加える必要がない請求項3に記載 の方法。 5.製品が食品又は生物材料である請求項1から4のいずれか一項に記載の方法 又は使用。 6.製品がアイスクリーム等の冷凍用製品である請求項1から5のいずれか一項 に記載の方法又は使用。 7.製品が冷凍生地又は冷凍パン製品である請求項1から6のいずれか一項に記 載の方法又は使用。 8.ポリペプチド又はタンパク質が組換えポリペプチド又はタンパク質である請 求項1から7のいずれか一項に記載の方法又は使用。 9.ポリペプチド又はタンパク質が食品級ポリペプチド又はタンパク質である請 求項1から8のいずれか一項に記載の方法又は使用。 10.例えば凍結後の氷再結晶を防止又は抑制することにより例えば潜在的凍結 損傷を最小限にするように氷晶成長プロセスを改変して特に再成長における氷の 寸法及び形状特徴を変更して改善された性質を示す組換えポリペプチド又はタン パク質、即ち対応量のニシナメタガレイAFPよりも高い特異不凍活性 を示す組換え不凍ペプチド(AFP)の製造方法であって、 1)天然宿主生物には存在せず且つ (a)宿主生物で活性な強力で場合により誘導性のプロモーターと、 (b)下記(c)のAFPコーディング核酸配列の発現中に宿主生物により生産 されるタンパク質を分泌することが可能な任意DNAシグナル配列中に存在して いてもよいATG開始コドン(その場合にはシグナル配列はAFPコーディング 核酸配列と同種でも異種でもよく、ATG開始コドンと同一読み枠に配置される )と、 (c)III型AFPタンパク質HPLC12のアミノ酸配列に実質的に対応す るアミノ酸配列をコードし、ATG開始コドンと同一読み枠に配置された少なく とも1種の核酸配列と、場合により、 (d)AFPコーディング核酸配列の3’末端に結合した停止コドン をこの順序で含むDNA構築物に含まれる少なくとも1種のAFPコーディング 核酸配列の発現を生じるか又は誘導するような条件下で食品級宿主生物を培養す る段階と、場合により 2)それ自体公知の方法で更に処理することにより得られるAFPコーディング 核酸配列から発現される産物を採取する段階からなる前記方法。 11.DNA構築物が宿主の培養中にIII型AFPタンパク質HPLC12の アミノ酸配列に実質的に対応するアミノ酸配列をコードする核酸配列の発現産物 を宿主に分泌させる宿主生物のシグナル配列を含む請求項10に記載の方法。 12.宿主生物が微生物である請求項10又は11に記載の方法。 13.宿主生物が酵母である請求項10から12のいずれか一項に記載の方法。 14.III型AFPタンパク質HPLC12のアミノ酸配列に実質的に対応す るアミノ酸配列をもつモノマーポリペプチドを分泌するような条件下で宿主生物 を培養する請求項10から13のいずれか一項に記載の方法。 15.III型AFPタンパク質HPLC12に実質的に対応するアミノ酸配列 をコードする核酸配列が宿主生物の好適コドンを含む請求項10から14のいず れか一項に記載の方法。 16.DNAシグナル配列がS.cerevisiaeのα接 合因子のDNAプレ配列である請求項10から15のいずれか一項に記載の方法 。 17.DNA構築物がプレ配列とAFPコーディング核酸配列の間にS.cer evisiaeのα接合因子のプロ配列を更に含み、プレ配列とプロ配列とAF Pコーディング核酸配列が同一読み枠に配置される請求項10から16のいずれ か一項に記載の方法。 18.DNA構築物がAFPをコードする核酸配列の前にS.cerevisi aeのインベルターゼシグナル配列を含む請求項10から17のいずれか一項に 記載の方法。 19.プロモーターがSaccharomyces cerevisiaeの誘 導的GAL7プロモーター又は構成的GAPDHプロモーターである請求項10 から18のいずれか一項に記載の方法。 20.例えば凍結後の氷再結晶を防止又は抑制することにより例えば潜在的凍結 損傷を最小限にするように氷晶成長プロセスを改変して特に再成長における氷の 寸法及び形状特徴を変更して改善された性質をもつ食品級組換え宿主生物であっ て、III型AFP HPLC12に実質的に対応するポリペプチド又 はタンパク質及び/又は請求項10から19のいずれか一項に記載の方法により 製造されたAFPを含むか及び/又はこれに囲まれた前記食品級組換え宿主生物 。 21.少なくとも1種のAFPコーディング核酸配列を発現することが可能な食 品級組換え宿主生物であって、前記核酸配列がDNA構築物に含まれ、前記DN A構築物が天然宿主生物には存在せず、前記DNA構築物が、 (a)宿主生物で活性な強力で場合により誘導性のプロモーターと、 (b)下記(c)のAFPコーディング核酸配列の発現中に宿主生物により生産 されるタンパク質を分泌することが可能な任意DNAシグナル配列中に存在して いてもよいATG開始コドン(その場合にはシグナル配列はAFPコーディング 核酸配列と同種でも異種でもよく、ATG開始コドンと同一読み枠に配置される )と、 (c)III型AFPタンパク質HPLC12のアミノ酸配列に実質的に対応す るアミノ酸配列をコードし、ATG開始コドンと同一読み枠に配置された少なく とも1種の核酸配列と、場合により、 (d)AFPコーディング核酸配列の3’末端に結合した停止コドン をこの順序で含む前記食品級組換え宿主生物。 22.請求項10から19のいずれか一項に記載の方法で使用される請求項21 に記載の組換え食品級宿主生物。 23.III型AFP HPLC12に実質的に対応するポリペプチド又はタン パク質及び/又は請求項20から22のいずれか一項に記載の組換え食品級宿主 生物を含む製品であって、好ましくは食品であり、天然に前記ポリペプチド又は タンパク質を前記形態及び量で含む食用生物以外のものである製品。 24.冷凍用製品であり、例えばアイスクリーム、冷凍生地又は冷凍パン製品等 の食品である請求項23に記載の製品。 25.例えば凍結後の氷再結晶を防止又は抑制することにより例えば潜在的凍結 損傷を最小限にするように氷晶成長プロセスを改変して特に再成長における氷の 寸法及び形状特徴を変更して改善された性質を示す実質的に純粋な単離組換え食 品級ポリペプチド又はタンパク質、即ち同量のニシナメタガレイI型AFPより も高いAFP活性を示す組換え不凍ペプチド(AFP)であって、III型AF Pタンパク質HPLC12のアミ ノ酸配列に実質的に対応するアミノ酸配列をもつことを特徴とする前記ポリペプ チド又はタンパク質。 26.例えば凍結後の氷再結晶を防止又は抑制することにより例えば潜在的凍結 損傷を最小限にするように氷晶成長プロセスを改変して特に再成長における氷の 寸法及び形状特徴を変更して改善された性質を示す組換え食品級ポリペプチド又 はタンパク質、即ち請求項10から19のいずれか一項に記載の方法により製造 され、同量のニシナメタガレイI型AFPよりも高いAFP活性を示す組換え不 凍ペプチド(AFP)。
